The strong selection pressure exerted by intensive use of glyphosate in cultivated areas has selected populations of the Rubiaceae weed species Borreria latifolia (Aubl.) K.Shum. (broadleaf buttonweed), Galianthe chodatiana (Standl.) E.L. Cabral (galiante) and Richardia brasiliensis Gomes (Brazilian pusley) with differential sensitivity to this herbicide in the South region of Brazil. The control of these weeds with herbicides is troublesome and signals the need to incorporate management practices of ruderal flora and crops, more sustainable and that results in more efficient control for long-term. Therefore, it is very important to expand the information about their biology and management. This study aimed: (a) select efficient methods to overcome dormancy of B. latifolia and G. chodatiana and determine how they influence the kinetics of seeds germination; (b) analyze the effects of temperature, irradiance, pH, aluminum and salinity on seed germination and initial growth of the B. latifolia, G. chodatiana e R. brasiliensis seedlings; (c) evaluate tolerance to glyphosate levels in biotypes of B. latifolia, G. chodatiana e R. brasiliensis through dose-response curves and compare two methods to evaluate herbicidal control; (d) and evaluated the effectiveness of alternative herbicides in pre-emergence and in early and late post-emergence of the three species. The treatment with KNO3 2%/3h + gibberellic acid 400 ppm resulted in higher percentage of G. chodatiana seed germination. This treatment and also the dry heat (60°C/30 min) + KNO3 2%/3h were more effective in overcoming dormancy of B. latifolia. G. chodatiana and R. brasiliensis tolerate lower temperatures during the germination process, while B. latifolia tolerate higher temperatures. B. latifolia and R. brasiliensis are positive photoblastic while G. chodatiana is indifferent to the photoperiod. B. latifolia shows higher germination and early development in pH 3, while G. chodatiana and R. brasiliensis prefer pH range between 5 and 7. B. latifolia and G. chodatiana were more tolerant to the aluminum during the germination process than R. brasiliensis. Low salt levels were sufficient to reduce the seed germination of the three species. Some biotypes of B. latifolia and R. brasiliensis showed medium-high glyphosate tolerance, not being controlled by higher doses than recommended. The G. chodatiana specie was not controlled with the highest dose used, showing a high glyphosate tolerance. The sulfentrazone, s-metolachlor and saflufenacil herbicides sprayed in pre-emergence showed high efficacy both on B. latifolia and R. brasiliensis, while chlorimuron-ethyl and diclosulan were effective only on R. brasiliensis. In early post-emergence the fomesafen, lactofem and flumioxazin herbicides efficiently controlled plants of all species, while bentazon showed high efficacy only on B. latifolia. Noteworthy the susceptibility of the G. chodatiana specie for applications in early post-emergence, because the control effectiveness and the number of effective herbicides are reduced with increasing the plant age. Many treatments with tank mix or sequencial applications with glyphosate, were effective in controlling B. latifolia and R. brasiliensis plants in advanced stage of development.

This work presents the development and modification of techniques to reduce the effects of load variation and mains frequency deviation in repetitive controllers applied to active power filters. To minimize the effects of aperiodic signals resulting from the connection or disconnection of non-linear loads is developed a technique which recognizes linear and nonlinear loads, and operates to reset the controller only when the error due to the transition of considerable value, and the transition is from non-linear to linear load. An algorithm to adapt the gain of the repetitive controller, based on a sigmoid function adaptation, in order to minimize the effects caused by random noise in the measurement system is also used. This work also analyzes the effects of frequency variation and presents the main methods to cope with this situation. Some solutions are the change in the number of samples per period and the variation of the sampling rate. The first has the advantage of using linear design techniques and results in a time invariant system. The second method changes the sampling frequency and leads to a time variant system that demands a difficult analysis of stability. The proposed algorithms were tested using the methods of truncation of the number of samples and the method of changing the sampling rate of the system to compensate possible frequency variations of the grid. Experimental results are presented to validate the proposal.

A qualidade dos alimentos é um conceito que tem vindo a despertar o interesse das indústrias e dos consumidores, daí a preocupação em conservar os produtos sob condições adequadas, evitando alterações físicas e químicas que ponham em causa a integridade dos alimentos, bem como as suas propriedades nutricionais e sensoriais. Deste modo, foi realizado um estudo para avaliar os efeitos do armazenamento sob certas condições de temperatura e diferentes tipos de embalagens sobre as propriedades dos miolos de nozes, avelãs e amêndoas com pele, embora no caso das amêndoas também se tenha estudoa do uma amostra sem pele. As amostras utilizadas eram originárias de Portugal, mas também de outros países, Chile, Espanha, Turquia, Roménia e Estados Unidos. O armazenamento das nozes durou 90 dias, e as condições testadas foram: temperatura ambiente; estufa a 30 e 50 °C, refrigeração (2 °C) e congelamento(15 °C). Os dois tipos de embalagem de plástico utilizados foram o polietileno de baixa densidade (PEBD) e polietileno de baixa densidade linear (PEBDL), comparando com as amostras armazenadas sem qualquer embalagem. No caso das amêndoas, uma das amostras apresentava-se sem pelicula. Foram avaliadas várias características dos frutos: a atividade de água (medida por um higrómetro), o teor de humidade (secagem em estufa até massa constante), a cor (com um colorímetro) e a textura (utilizando-se um texturómetro). A partir dos resultados obtidos concluiu-se que as amostras de diferentes origens são diferentes no que diz respeito ao teor de humidade, atividade de água e cor, independentemente das condições de armazenamento e tipo de embalagem. Os resultaram mostraram que as condições de armazenagem que melhor preservam as características estudadas dos frutos secos foram os de baixa temperatura, sendo observadas maiores alterações nos frutos armazenados a temperaturas elevadas, nomeadamente na cor. Assim verificou-se que os processos de refrigeração e congelação têm uma influência mínima nas características dos frutos, particularmente na humidade, na actividade da água, na dureza e friabilidade. Os resultados indicaram também que os frutos apresentam valores de aw inferiores a 0,6, para todas condições testadas, garantindo alguma estabilidade a nível das estabilidade microbiana e enzimática. A presença da pele no miolo de amêndoa, para todos os tratamentos, tem uma influência significativa na textura e cor. Finalmente, em relação ao tipo de embalagem, foi observado que o uso de sacos de plástico testados não parece contribuir para a melhoria das características dos produtos em relação às amostras não embaladas.

The routine analysis for quantization of organic acids and sugars are generally slow methods that involve the use and preparation of several reagents, require trained professional, the availability of special equipment and is expensive. In this context, it has been increasing investment in research whose purpose is the development of substitutive methods to reference, which are faster, cheap and simple, and infrared spectroscopy have been highlighted in this regard. The present study developed multivariate calibration models for the simultaneous and quantitative determination of ascorbic acid, citric, malic and tartaric and sugars sucrose, glucose and fructose, and soluble solids in juices and fruit nectars and classification models for ACP. We used methods of spectroscopy in the near infrared (Near Infrared, NIR) in association with the method regression of partial least squares (PLS). Were used 42 samples between juices and fruit nectars commercially available in local shops. For the construction of the models were performed with reference analysis using high-performance liquid chromatography (HPLC) and refractometry for the analysis of soluble solids. Subsequently, the acquisition of the spectra was done in triplicate, in the spectral range 12500 to 4000 cm-1. The best models were applied to the quantification of analytes in study on natural juices and juice samples produced in the Paraná Southwest Region. The juices used in the application of the models also underwent physical and chemical analysis. Validation of chromatographic methodology has shown satisfactory results, since the external calibration curve obtained R-square value (R2) above 0.98 and coefficient of variation (%CV) for intermediate precision and repeatability below 8.83%. Through the Principal Component Analysis (PCA) was possible to separate samples of juices into two major groups, grape and apple and tangerine and orange, while for nectars groups separated guava and grape, and pineapple and apple. Different validation methods, and pre-processes that were used separately and in combination, were obtained with multivariate calibration models with average forecast square error (RMSEP) and cross validation (RMSECV) errors below 1.33 and 1.53 g.100 mL-1, respectively and R2 above 0.771, except for malic acid. The physicochemical analysis enabled the characterization of drinks, including the pH working range (variation of 2.83 to 5.79) and acidity within the parameters Regulation for each flavor. Regression models have demonstrated the possibility of determining both ascorbic acids, citric, malic and tartaric with successfully, besides sucrose, glucose and fructose by means of only a spectrum, suggesting that the models are economically viable for quality control and product standardization in the fruit juice and nectars processing industry.

A ocratoxina A e a citrinina são micotoxinas encontrados naturalmente em alimentos e rações animais. A avaliação do risco do consumo de alimentos contaminados por esses compostos deve ser estimada a partir de dados confiáveis que reflitam a verdadeira concentração destas toxinas em diferentes alimentos ou insumos, especialmente se ingeridos com freqüência. Isto gera a necessidade de métodos analíticos precisos que sejam rápidos sem desconsiderar as etapas clássicas de avaliação de traços, amostragem representativa, extração, limpeza, concentração, separação de formas químicas, detecção, confirmação de identidade e quantificação. Para as micotoxinas em geral as técnicas cromatográficas são as mais aplicadas e relatadas em vista da diversidade estrutural destes compostos. Neste trabalho foi realizada uma otimização de extração utilizando o método QuEChERS modificado em comparação com os métodos Soares e Rodrigues Amaya (1989), Tanaka (2001) e Ultrassom (Palma et. al., 2007) empregando diferentes técnicas cromatográficas com detectores distintos para análise destas micotoxinas simultaneamente. Foram analisadas 38 amostras de arroz cultivadas e armazenadas em campos experimentais de Cachoeirinha na região Sul do Brasil. O uso dos sistemas CCD, HPLC-DAD e LC-MS proporcionou especificidade, precisão e sensibilidade, de modo que os limites de detecção e quantificação, obtivessem valores inferiores ao limite máximo estabelecido por órgãos reguladores internacionais (5 µg Kg-1 para ocratoxina A). Os limites de detecção encontrados para citrinina e ocratoxina A em camada delgada foram 4,7 e 6 vezes maior que para cromatografia líquida de alta eficiência acoplada a detector de arranjo de diodos, 14 e 300 vezes maior que cromatografia líquida acoplada a detector de massas. Os limites de quantificação das duas micotoxinas ficaram dentro do exigido pela legislação européia para OTA de 5 µg Kg-1 para HPLC-DAD e LC-MS. Na cromatografia de camada delgada esse valor ficou 4 vezes acima do estabelecido para ocratoxina A. A ocorrência de ocratoxina A e citrinina foi verificada em 16 % das amostras estando os teores variando entre 3 e 560 µg Kg-1, sugerindo possível exposição crônica a estas micotoxinas caso as amostras sejam consumidas.

Requirements specification has long been recognized as critical activity in software development processes because of its impact on project risks when poorly performed. A large amount of studies addresses theoretical aspects, propositions of techniques, and recommended practices for Requirements Engineering (RE). To be successful, RE have to ensure that the specified requirements are complete and correct what means that all intents of the stakeholders in a given business context are covered by the requirements and that no unnecessary requirement was introduced. However, the accurate capture the business intents of the stakeholders remains a challenge and it is a major factor of software project failures. This master’s dissertation presents a novel method referred to as “Problem-Based SRS” aiming at improving the quality of the Software Requirements Specification (SRS) in the sense that the stated requirements provide suitable answers to real customer ́s businesses issues. In this approach, the knowledge about the software requirements is constructed from the knowledge about the customer ́s problems. Problem-Based SRS consists in an organization of activities and outcome objects through a process that contains five main steps. It aims at supporting the software requirements engineering team to systematically analyze the business context and specify the software requirements, taking also into account a first glance and vision of the software. The quality aspects of the specifications are evaluated using traceability techniques and axiomatic design principles. The cases studies conducted and presented in this document point out that the proposed method can contribute significantly to improve the software requirements specification.

Marine algae are one of the major sources of biologic compounds. In extracellular matrix of these organisms there are sulfated polysaccharides that functions as structural components and provides protection against dehydration. The fraction 1.0 (F1.0) rich in sulfated galactans obtained from red seaweed Hypnea musciformis was physicochemical characterized and evaluated for pharmacologic activity through antioxidant activity, cytotoxic action on erythrocytes, anticoagulant, stimulatory action under antithrombotic heparan sulfate synthesis and their effects on cell proliferation and cycle cell progression. The main components of F1.0 were carbohydrates (49.70 ± 0.10%) and sulfate (44.59 ± 0.015%), presenting phenolic compounds (4.79 ± 0.016%) and low protein contamination (0.92 ± 0.001%). Fraction 1.0 showed polidisperse profile and signs in infrared analysis in 1262, 1074 and 930, 900 and 850 attributed to sulfate esters S=O bond, presence of a 3,6- anidrogalactose C-O bond, non-sulfated β-D-galactose and a C-O-SO4 bond in galactose C4, respectively. The fraction rich in sulfated galactans exhibited strong antioxidant action under lipid peroxidation assay with IC50 of 0.003 mg/mL. Besides the inhibition of hemolysis induced by H2O2 in erythrocytes treated with F1.0, this fraction did not promote significant cytotoxity under erythrocytes membranes. F1.0 exhibited low anticoagulant activity causing moderate direct inhibition of enzimatic activity of thrombin. This fraction promoted stimulation around of 4.6 times on this synthesis of heparan sulfate (HS) by rabbit aortic endothelial cells (RAEC) in culture when was compared with non treated cells. The fraction of this algae displayed antiproliferative action under RAEC cells causing incresing on cell number on S fase, blocking the cycle cell progression. Thus F1.0 presented cytostatic and no cytotoxic action under this cell lineage. These results suggest that F1.0 from H. musciformis have antioxidant potential which is a great effect for a compound used as food and in food industry which could be an alternative to food industry to prevent quality decay of lipid containing food due to lipid peroxidation. These polysaccharides prevent the lipid peroxidation once the fraction in study exhibited strong inhibitory action of this process. Furthermore that F1.0 present strong antithrombotic action promoting the stimulation of antithrombotic HS synthesis by endothelial cells, being important for thrombosis preventing, by its inhibitory action under reactive oxygen species (ROS) in some in vitro methods, being involved in promotion of hypercoagulability state.

Atualmente, os “novos turistas” são utilizadores experientes das novas tecnologias e estão predispostos e motivados para praticar um tipo de turismo diferente, especialmente no que diz respeito ao planeamento das suas viagens para obter novas experiências. O presente estudo tem como objetivo analisar, através de uma revisão da literatura, um potencial segmento de turismo designado de Turismo Místico, com o propósito de criar um website de oferta de experiências turísticas em Portugal com carater inovador e sentimental. O Website foi desenvolvido e testado numa fase beta através de um questionário adaptado do Modelo WebQual de Loiacono. Os resultados obtidos, além de permitirem a avaliação da perceção da qualidade do Website, também indicam formas de melhorar ou adaptar o produto final aos objetivos planeados. A principal conclusão destaca o fator entretenimento como o mais importante na perceção da qualidade do Website e na intenção decisiva de revisita do mesmo. A era do marketing digital criou um consumidor mais proativo, o prosumidor, que participa no processo de desenvolvimento de produtos e serviços.

This work aims to obtain a low-cost virtual sensor to estimate the quality of LPG. For the acquisition of data from a distillation tower, software HYSYS ® was used to simulate chemical processes. These data will be used for training and validation of an Artificial Neural Network (ANN). This network will aim to estimate from available simulated variables such as temperature, pressure and discharge flow of a distillation tower, the mole fraction of pentane present in LPG. Thus, allowing a better control of product quality

Since the beginning of the National Program for Production and Use of Biodiesel in Brazil, in 2004, different raw materials were evaluated for biodiesel production, trying to combine the agricultural diversity of the country to the desire to reduce production coasts. To determine the chemical composition of biodiesel produced from common vegetables oils, international methods have been used widely in Brazil. However, for analyzing biodiesel samples produced from some alternative raw materials analytical problems have been detected. That was the case of biodiesel from castor oil. Due the need to overcome these problems, new methodologies were developed using different chromatographic columns, standards and quantitative methods. The priority was simplifying the equipment configuration, realizing faster analyses, reducing the costs and facilitating the routine of biodiesel research and production laboratories. For quantifying free glycerin, the ethylene glycol was used in instead of 1,2,4-butanetriol, without loss of quality results. The ethylene glycol is a cheaper and easier standard. For methanol analyses the headspace was not used and the cost of the equipment used was lower. A detailed determination of the esters helped the deeper knowledge of the biodiesel composition. The report of the experiments and conclusions of the research that resulted in the development of alternative methods for quality control of the composition of the biodiesel produced in Brazil, a country with considerable variability of species in agriculture, are the goals of this thesis and are reported in the following pages
