As broncopneumonias são afecções importantes na pecuária mundial, representando uma das principais causas de mortalidade de bezerros nos primeiros meses de vida. As medidas preventivas e terapêuticas adotadas geralmente são baseadas em resultados de estudos internacionais, não se conhecendo as bactérias implicadas nos quadros pneumônicos em animais criados no Brasil. Aliado a isso, no primeiro mês de vida, os bezerros demonstram imaturidade do sistema imune, o que tem sido pouco estudado em quadros pneumônicos. Desta maneira, objetivou-se estudar as broncopneumonias em bezerros neonatos, identificando bactérias do trato respiratório posterior de bezerros sadios e com pneumonias naturalmente adquiridas, bem como analisar citologicamente a resposta pulmonar frente a estes patógenos. Para isso amostras de lavado do trato respiratório foram colhidas por traqueocentese durante o primeiro mês de vida dos animais. Verificou-se que não houve diferença na microbiota traqueobrônquica de bezerros sadios em relação aos doentes, discordando dos relatos da literatura internacional, sendo constituída principalmente por: Staphylococcus sp., Bacillus sp., Streptococcus sp., Pseudomonas aeruginosa e enterobactérias, permitindo inferir que as medidas profiláticas e terapêuticas adotadas internacionalmente possam não ser tão efetivas para as criações brasileiras. Observou-se também que bezerros neonatos têm uma proporção aproximada de 1:1 de macrófagos e neutrófilos na região traqueobrônquica quando saudáveis, atingindo uma relação aproximada de 1:3 durante os quadros de broncopneumonias, sendo estes perfis provavelmente característicos da idade, período conhecido pela imaturidade do sistema imune e agravado por fatores de manejo que favoreçam uma maior inalação de agentes bacterianos.

Foram estudados 33 surtos de pneumonia em bovinos jovens na área de influência do Laboratório Regional de Diagnóstico (LRD) da Faculdade de Veterinária da Universidade Federal de Pelotas (UFPel) entre os anos de 2000 e 2011. Foram diagnosticados 18 surtos de pneumonia (54,54%) em bovinos de raças leiteiras, Holandês ou Jersey e 13 surtos (39,39%) em gado de corte ou cruzas de gado de corte. A morbidade variou entre 0,06%-100% e a mortalidade foi de 0,06%-34,6%. A doença ocorreu igualmente em todas as estações do ano e foi mais frequente em bezerros de 1-3 meses totalizando 13 surtos. Sete surtos ocorreram em bovinos entre 4-6 meses, sete entre 7-12 meses e seis surtos ocorreram em bezerros de 1-29 dias. Pneumonia enzoótica pela infecção pelo vírus sincicial respiratório bovino (BRSV) com lesões histológicas de broncopneumonia, pneumonia intersticial e presença de células sinciciais foi mais frequente em bovinos de raças de corte com dez surtos (58,8%); seis surtos dessa enfermidade ocorreram em raças de leite (35,2%). O diagnóstico foi confirmado por imuno-histoquímica em sete casos. Os sinais clínicos da maioria dos casos de pneumonia observados caracterizaram-se por dispneia, emagrecimento, apatia, tremores, bruxismo, desidratação, respiração ruidosa, tosse, corrimento nasal seroso ou mucopurulento, decúbito e morte. As lesões macroscópicas caracterizaram-se por presença de áreas de consolidação vermelho-escuras, edema e enfisema nas regiões crânio-ventrais dos lobos pulmonares cardíaco e apical ou pneumonia intersticial com distribuição difusa, edema e enfisema. Histologicamente, as lesões pulmonares eram variáveis. Broncopneumonia necrossupurativa difusa acentuada com hiperplasia de pneumócitos tipo II e edema intersticial e alveolar foi observada em 15 casos. Os resultados deste trabalho demonstram que as pneumonias são importantes causas de perdas econômicas em bovinos jovens na região de influência do LRD. Deve ser destacado que a pneumonia enzoótica devido a infecção pelo BRSV é importante tanto em bovinos de corte como de leite independente da forma de criação.

Os cachorros-do-mato (Cerdocyon thous Linnaeus, 1766) são animais amplamente encontrados em países da América do Sul. Apesar de não ser uma espécie ameaçada de extinção, é possível que muitas populações sofram impactos decorrentes de atropelamentos de indivíduos nas rodovias do país, pois, trata-se de uma das espécies de carnívoros com elevada ocorrência de mortes deste tipo. Foram avaliados 32 sincrânios de C. thous, oriundos de vida livre, armazenados na coleção osteológica do Laboratório de Anatomia Veterinária da Universidade Vila Velha (Vila Velha/ES), a fim de diagnosticar doenças que acometeram estes indivíduos enquanto vivos. As lesões macroscópicas identificadas foram: apinhamento dentário, ausência dentária, cálculo dentário, dentina terciária, desgaste, escurecimento dentário, exposição de polpa, fenestração óssea, fratura dentária, fratura de esmalte, giroversão, pigmentação e reabsorção da crista alveolar. Os achados mais comumente observados foram: ausência dentária, desgaste dentário e fratura dentária. Ausências dentárias anteriores à morte, alterações ósseas, cálculos dentários, apinhamento e giroversão aparentemente não causaram quaisquer prejuízos aos animais enquanto vivos.

A intoxicação experimental por Trema micrantha em cinco ovinos é descrita. Quatro ovinos apresentaram sinais clínicos respiratórios acentuados e morte após ingestão da terceira dose de folhas da planta. As manifestações clínicas mais frequentes nesses casos foram taquipneia, dispneia, retração ritmada das narinas, mucosas cianóticas, corrimento nasal mucoso, hipertermia, aumento de volume abdominal bilateral e na região parotídea, incluindo crepitação à palpação, membros lateralmente afastados ao caminhar e decúbito esternal. T. micrantha mostrou-se tóxica para ovinos na dose de 20-50g/kg de peso animal. Na necropsia desses ovinos foram observados, além das mucosas cianóticas, enfisema subcutâneo em região cervical ventral ou porção mediastinal dorsal, pulmões não colabados, pesados, com impressão das costelas na superfície e conteúdo espumoso vermelho em traqueia e brônquios, além de múltiplas petéquias subpleurais. Na avaliação histológica predominaram alterações pulmonares, com espessamento de septos alveolares por proliferação difusa de pneumócitos tipo II, conferindo aspecto adenomatoso a algumas áreas. Os pneumócitos apresentavam núcleo volumoso, hipercromático, ora bizarro ou eram multinucleados, com nucléolos evidentes e, em algumas áreas, os pneumócitos estavam descamados para a luz alveolar, ora formando sincícios. Havia também proliferação do epitélio bronquiolar, com formação de mais de uma camada celular, núcleos hipercromáticos e volumosos e redução na quantidade de cílios. As alterações proliferativas dos pneumócitos e do epitélio bronquiolar foram evidenciadas pela imunomarcação anti-citoqueratina e anti-Ki-67 e, para a diferenciação entre pneumócitos e macrófagos alveolares, foi empregada imuno-histoquímica anti-mieloide/histiócitos (MAC387). Um ovino apresentou quadro clinico-patológico de insuficiência hepática aguda, com necrose hepatocelular acentuada após ingestão de 25g/kg de T. micrantha. A intoxicação experimental por T. micrantha em ovinos resultou em quadro clinico-patológico, predominantemente, respiratório na maioria dos casos, confirmando os resultados observados em casos de intoxicação espontânea.

Nasua nasua é um animal onívoro, encontrado em todo Brasil. A reprodução da espécie ocorre somente uma vez ao ano, na primavera. As fêmeas são matriarcas e amamentam suas crias até os 5 meses de idade, vivem com seus filhotes em bandos de até 30 indivíduos. Para descrição morfológica da glândula mamaria do Nasua nasua foram utilizados seis animais provenientes do Criatório Cientifico (Cecrimpas), Unifeob. Autorizado pelo Ibama (Proc.02027.002322/98-99). Para análise macroscópica um animal foi injetado com látex neoprene, sendo a artéria femoral injetada com látex de cor vermelha e a veia jugular de cor azul. Os demais animais foram fixados em solução aquosa a 10% de formaldeído. Para análise microscópica, fragmentos glandulares foram coletados e submetidos ao processo rotineiro, embebido em parafina e corados com Hematoxilina e Eosina, Picrossírius e Azul de Toluidina. Macroscopicamente foram evidenciados três pares de glândulas mamárias, sendo dois pares posicionados na região abdominal e um par na região inguinal. Microscopicamente, notou-se epitélio de revestimento externo das papilas mamárias, epitélio pavimentoso estratificado queratinizado, o qual seguia por toda glândula de forma irregular. Na entrada do óstio, o epitélio da epiderme era modificado ocorrendo uma transição de epitélio pavimentoso estratificado para um epitélio cúbico no ducto papilar. O parênquima glandular era caracteristicamente túbulo alveolar com células secretoras, evidenciado principalmente no animal lactente. Os resultados macroscópicos e microscópicos assemelham-se aos já descritos nas cadelas (Canis familiaris) e os do Procyon cancrivorus pertencente à mesma família do quati, Família Procyonidae.

Bronchoalveolar lavage (BAL) is a procedure that retrieves cells and other elements from the lungs for evaluation, which helps in the diagnosis of pulmonary diseases. The aim of this study was to perform this procedure for cellular analysis of BAL fluid alterations during experimental infection with Aelurostrongylus abstrusus in cats. Fourteen cats were individually inoculated with 800 third stage larvae of A. abstrusus and five non-infected cats lined as a control group. The BAL procedure was performed through the use of an endotracheal tube on the nineteen cats with a mean age of 18 months, on 0, 30, 60, 90, 120, 180 and 270 days after infection. Absolute cell counts in the infected cats revealed that alveolar macrophages and eosinophils were the predominant cells following infection. This study shows that the technique allows us to retrieve cells and first stage larvae what provides information about the inflammatory process caused by aelurostrongylosis.

Descrevem-se os dados epidemiológicos, os sinais clínicos e as lesões de quatro surtos da doença do edema e enfisema pulmonar agudo em bovinos (EEPAB) nos estados de Santa Catarina e Paraná e sua reprodução experimental. A doença espontânea ocorreu após transferência de bovinos de pastagem madura e seca para outra jovem e viçosa. Todos os bovinos afetados eram vacas das raças holandês e pardo suíço. Os principais sinais clínicos foram dispneia e respiração abdominal dificultosa com o pescoço estendido e a boca aberta. Apresentaram, também, enfisema subcutâneo, queda na produção de leite e recuperação lenta ou morte. Os achados de necropsia foram restritos ao pulmão o qual tinha coloração vermelho escuro, não colabado, de aspecto brilhante e hipercriptante com enfisema interlobular acentuado. As lesões histológicas no pulmão consistiam principalmente de enfisema alveolar e interlobular intercalado por áreas de congestão e edema, degeneração hialina da parede dos alvéolos e infiltrado de macrófagos e eosinófilos, moderado, difuso. A reprodução experimental da doença foi realizada em um bovino, com administração de 0,7mg/kg de peso corporal de L-triptofano por via oral em dose única. O animal morreu no sétimo dia de experimento. Os sinais clínicos e lesões foram idênticos aos observados na doença espontânea.

This study looks at negotiation of belonging and understandings of home among a generation of young Kurdish adults who were born in Iraq, Iran, and Turkey and who reached adulthood in Finland. The young Kurds taking part in the study belong to the generation of migrants who moved to Finland in their childhood and early teenage years from the region of Kurdistan and elsewhere in the Middle East, then grew to adulthood in Finland. In theoretical terms, the study draws broadly from three approaches: transnationalism, intersectionality, and narrativity. Transnationalism refers to individuals’ cross-border ties and interaction extending beyond nationstates’ borders. Young people of migrant background, it has been suggested, are raised in a transnational space that entails cross-border contacts, ties, and visits to the societies of departure. How identities and feelings of belonging become formed in relation to the transnational space is approached with an intersectional frame, for examination of individuals’ positionings in terms of their intersecting attributes of gender, age/generation, and ethnicity, among others. Focus on the narrative approach allows untangling how individuals make sense of their place in the social world and how they narrate their belonging in terms of various mechanisms of inclusion and exclusion, including institutional arrangements and discursive categorisation schemes. The empirical data for this qualitative study come from 25 semi-structured thematic interviews that were conducted with 23 young Kurdish adults living in Turku and Helsinki between 2009 and 2011. The interviewees were aged between 19 and 28 years at the time of interviewing. Interview themes involved topics such as school and working life, family relations and language-learning, political activism and citizenship, transnational ties and attachments, belonging and identification, and plans for the future and aspirations. Furthermore, data were collected from observations during political demonstrations and meetings, along with cultural get-togethers. The data were analysed via thematic analysis. The findings from the study suggest that young Kurds express a strong sense of ‘Kurdishness’ that is based partially on knowing the Kurdish language and is informed by a sense of cultural continuity in the diaspora setting. Collective Kurdish identity narratives, particularly related to the consciousness of being a marginalised ‘other’ in the context of the Middle East, are resonant in young interviewees’ narrations of ‘Kurdishness’. Thus, a sense of ‘Kurdishness’ is drawn from lived experiences indexed to a particular politico-historical context of the Kurdish diaspora movements but also from the current situation of Kurdish minorities in the Middle East. On the other hand, young Kurds construct a sense of belonging in terms of the discursive constructions of ‘Finnishness’ and ‘otherness’ in the Finnish context. The racialised boundaries of ‘Finnishness’ are echoed in young Kurds’ narrations and position them as the ‘other’ – namely, the ‘immigrant’, ‘refugee’, or ‘foreigner’ – on the basis of embodied signifiers (specifically, their darker complexions). This study also indicates that young Kurds navigate between gendered expectations and norms at home and outside the home environment. They negotiate their positionings through linguistic repertoires – for instance, through mastery of the Finnish language – and by adjusting their behaviour in light of the context. This suggests that young Kurds adopt various forms of agency to display and enact their belonging in a transnational diaspora space. Young Kurds’ narrations display both territorially-bounded and non-territorially-bounded elements with regard to the relationship between identity and locality. ‘Home’ is located in Finland, and the future and aspirations are planned in relation to it. In contrast, the region of Kurdistan is viewed as ‘homeland’ and as the place of origins and roots, where temporary stays and visits are a possibility. The emotional attachments are forged in relation to the country (Finland) and not so much relative to ‘Finnishness’, which the interviewees considered an exclusionary identity category. Furthermore, identification with one’s immediate place of residence (city) or, in some cases, with a religious identity as ‘Muslim’ provides a more flexible venue for identification than does identifying oneself with the (Finnish) nation.

Chronic lung diseases, specifically bronchopulmonary dysplasia (BPD), are still causing mortality and morbidity amongst newborn infants. High protease activity has been suggested to have a deleterious role in oxygen-induced lung injuries. Cathepsin K (CatK) is a potent protease found in fetal lungs, degrading collagen and elastin. We hypothesized that CatK may be an important modulator of chronic lung injury in newborn infants and neonatal mice. First we measured CatK protein levels in repeated tracheal aspirate fluid samples from 13 intubated preterm infants during the first two weeks of life. The amount of CatK at 9-13 days was low in infants developing chronic lung disease. Consequently, we studied CatK mRNA expression in oxygen-exposed wild-type (WT) rats at postnatal day (PN) 14 and found decreased pulmonary mRNA expression of CatK in whole lung samples. Thereafter we demonstrated that CatK deficiency modifies lung development by accelerating the thinning of alveolar walls in newborn mice. In hyperoxia-exposed newborn mice CatK deficiency resulted in increased number of pulmonary foam cells, macrophages and amount of reduced glutathione in lung homogenates indicating intensified pulmonary oxidative stress and worse pulmonary outcome due to CatK deficiency. Conversely, transgenic overexpression of CatK caused slight enlargement of distal airspaces with increased alveolar chord length in room air in neonatal mice. While hyperoxic exposure inhibited alveolarization and resulted in enlarged airspaces in wild-type mice, these changes were significantly milder in CatK overexpressing mice at PN7. Finally, we showed that the expression of macrophage scavenger receptor 2 (MSR2) mRNA was down-regulated in oxygen-exposed CatK-deficient mice analyzed by microarray analysis. Our results demonstrate that CatK seems to participate in normal lung development and its expression is altered during pulmonary injury. In the presence of pulmonary risk factors, like high oxygen exposure, low amount of CatK may contribute to aggravated lung injury while sustained or slightly elevated amount of CatK may even protect the newborn lungs from excessive injury. Besides collagen degrading and antifibrotic function of CatK in the lungs, it is obvious that CatK may affect macrophage activity and modify oxidative stress response. In conclusion, pulmonary proteases, specifically CatK, have distinct roles in lung homeostasis and injury development, and although suggested, broad range inhibition of proteases may not be beneficial in newborn lung injury.

Pulmonary dysfunction represents the most important cause of death in patients with paracoccidioidomycosis (PBM). In order to investigate the functional changes of the lungs in the early stages of PBM, a model of benign disease was developed by intratracheal challenge of 12-week old isogenic Wistar rats with 1 x 106 yeast forms of Paracoccidioides brasiliensis. Animals were studied 30 and 60 days after infection, when fully developed granulomas were demonstrable in the lungs. Measurements of airway resistance, lung elastance and tissue hysteresis were made during sinusoidal deformations (100 breaths/min, tidal volume = 2 ml) with direct measurement of alveolar pressure using the alveolar capsule technique. Infection caused a significant increase in hysteresis (infected: 1.69, N = 13; control: 1.13, N = 12, P = 0.024, ANOVA), with no alterations in airway resistance or lung elastance. Histopathological analysis revealed the presence of fully developed granulomas located in the axial compartment of the lung interstitial space. These results suggest that alterations of tissue mechanics represent an early event in experimental PBM

Since the most characteristic feature of paraquat poisoning is lung damage, a prospective controlled study was performed on excised rat lungs in order to estimate the intensity of lesion after different doses. Twenty-five male, 2-3-month-old non-SPF Wistar rats, divided into 5 groups, received paraquat dichloride in a single intraperitoneal injection (0, 1, 5, 25, or 50 mg/kg body weight) 24 h before the experiment. Static pressure-volume (PV) curves were performed in air- and saline-filled lungs; an estimator of surface tension and tissue works was computed by integrating the area of both curves and reported as work/ml of volume displacement. Paraquat induced a dose-dependent increase of inspiratory surface tension work that reached a significant two-fold order of magnitude for 25 and 50 mg/kg body weight (P<0.05, ANOVA), sparing lung tissue. This kind of lesion was probably due to functional abnormalities of the surfactant system, as was shown by the increase in the hysteresis of the paraquat groups at the highest doses. Hence, paraquat poisoning provides a suitable model of acute lung injury with alveolar instability that can be easily used in experimental protocols of mechanical ventilation

Ketamine is believed to reduce airway and pulmonary tissue resistance. The aim of the present study was to determine the effects of ketamine on the resistive, elastic and viscoelastic/inhomogeneous mechanical properties of the respiratory system, lungs and chest wall, and to relate the mechanical data to findings from histological lung analysis in normal animals. Fifteen adult male Wistar rats were assigned randomly to two groups: control (N = 7) and ketamine (N = 8). All animals were sedated (diazepam, 5 mg, ip) and anesthetized with pentobarbital sodium (20 mg/kg, ip) or ketamine (30 mg/kg, ip). The rats were paralyzed and ventilated mechanically. Ketamine increased lung viscoelastic/inhomogeneous pressure (26%) compared to the control group. Dynamic and static elastances were similar in both groups, but the difference was greater in the ketamine than in the control group. Lung morphometry demonstrated dilation of alveolar ducts and increased areas of alveolar collapse in the ketamine group. In conclusion, ketamine did not act at the airway level but acted at the lung periphery increasing mechanical inhomogeneities possibly resulting from dilation of distal airways and alveolar collapse.

Matrix metalloproteinases (MMP) are considered to be key initiators of collagen degradation, thus contributing to bone resorption in inflammatory diseases. We determined whether subantimicrobial doses of doxycycline (DX) (<=10 mg kg-1 day-1), a known MMP inhibitor, could inhibit bone resorption in an experimental periodontitis model. Thirty male Wistar rats (180-200 g) were subjected to placement of a nylon thread ligature around the maxillary molars and sacrificed after 7 days. Alveolar bone loss (ABL) was measured macroscopically in one hemiarcade and the contralateral hemiarcade was processed for histopathologic analysis. Groups of six animals each were treated with DX (2.5, 5 or 10 mg kg-1 day-1, sc, 7 days) and compared to nontreated (NT) rats. NT rats displayed significant ABL, severe mononuclear cell influx and increase in osteoclast numbers, which were significantly reduced by 5 or 10 mg kg-1 day-1 DX. These data show that DX inhibits inflammatory bone resorption in a manner that is independent of its antimicrobial properties.

Toxic cyanobacteria in drinking water supplies can cause serious public health problems. In the present study we analyzed the time course of changes in lung histology in young and adult male Swiss mice injected intraperitoneally (ip) with a cyanobacterial extract containing the hepatotoxic microcystins. Microcystins are cyclical heptapeptides quantified by ELISA method. Ninety mice were divided into two groups. Group C received an injection of saline (300 µl, ip) and group Ci received a sublethal dose of microcystins (48.2 µg/kg, ip). Mice of the Ci group were further divided into young (4 weeks old) and adult (12 weeks old) animals. At 2 and 8 h and at 1, 2, 3, and 4 days after the injection of the toxic cyanobacterial extract, the mice were anesthetized and the trachea was occluded at end-expiration. The lungs were removed en bloc, fixed, sectioned, and stained with hematoxylin-eosin. The percentage of the area of alveolar collapse and the number of polymorphonuclear (PMN) and mononuclear cell infiltrations were determined by point counting. Alveolar collapse increased from C to all Ci groups (123 to 262%) independently of time, reaching a maximum value earlier in young than in adult animals. The amount of PMN cells increased with time of the lesion (52 to 161%). The inflammatory response also reached the highest level earlier in young than in adult mice. After 2 days, PMN levels remained unchanged in adult mice, while in young mice the maximum number was observed at day 1 and was similar at days 2, 3, and 4. We conclude that the toxins and/or other cyanobacterial compounds probably exert these effects by reaching the lung through the blood stream after ip injection.

The aim of the present study was to compare the efficacy of a novel phosphodiesterase 4 and 5 inhibitor, LASSBio596, with that of dexamethasone in a murine model of chronic asthma. Lung mechanics (airway resistance, viscoelastic pressure, and static elastance), histology, and airway and lung parenchyma remodeling (quantitative analysis of collagen and elastic fiber) were analyzed. Thirty-three BALB/c mice were randomly assigned to four groups. In the asthma group (N = 9), mice were immunized with 10 µg ovalbumin (OVA, ip) on 7 alternate days, and after day 40 they were challenged with three intratracheal instillations of 20 µg OVA at 3-day intervals. Control mice (N = 8) received saline under the same protocol. In the dexamethasone (N = 8) and LASSBio596 (N = 8) groups, the animals of the asthma group were treated with 1 mg/kg dexamethasone disodium phosphate (0.1 mL, ip) or 10 mg/kg LASSBio596 dissolved in dimethyl sulfoxide (0.2 mL, ip) 24 h before the first intratracheal instillation of OVA, for 8 days. Airway resistance, viscoelastic pressure and static elastance increased significantly in the asthma group (77, 56, and 76%, respectively) compared to the control group. The asthma group presented more intense alveolar collapse, bronchoconstriction, and eosinophil and neutrophil infiltration than the control group. Both LASSBio596 and dexamethasone inhibited the changes in lung mechanics, tissue cellularity, bronchoconstriction, as well as airway and lung parenchyma remodeling. In conclusion, LASSBio596 at a dose of 10 mg/kg effectively prevented lung mechanical and morphometrical changes and had the potential to block fibroproliferation in a BALB/c mouse model of asthma.
