As 4-quinolonas e as acridonas são duas importantes famílias de compostos heterocíclicos azotados naturais que apresentam uma variedade de importantes aplicações biológicas. As 4-quinolonas têm sido objecto de extensos estudos devido às suas potenciais aplicações como fortes agentes citotóxicos, antimitóticos e anti-plaquetários e como protectores cardiovasculares, mas o seu uso é principalmente como antibióticos de largo espectro. As acridonas são conhecidas por apresentarem importante actividade antiviral, antiparasitária, contra a leishmania e a malária, e anticancerígena. A variedade de importantes aplicações biológicas das 4-quinolonas e das acridonas e a contínua procura pela comunidade científica de novas substâncias com actividades biológicas atractivas destaca estes compostos como alvos interessantes para a preparação de novos derivados e/ou para o desenvolvimento de novos métodos de síntese destas duas famílias de compostos. No primeiro capítulo desta dissertação descreve-se a síntese dos compostos de partida que tiveram de ser previamente preparados para o desenvolvimento das novas rotas de síntese apresentadas nos capítulos seguintes. A 4-cloro-3- formilquinolina foi obtida através da reacção de Vilsmeier-Haack da 2’-aminoacetofenona enquanto que a 3-formilquinolin-4(1H)-ona foi facilmente preparada por hidrólise ácida da anterior. Foi necessário proteger o grupo amina da quinolin-4(1H)-ona para evitar reacções secundárias e foram descritas as reacções de protecção com o grupo metilo, etoxicarbonilo e ptoluenossulfonilo. Os 2,2-dióxidos de 1,3-di-hidrobenzo[c]tiofeno, necessários para o estudo das reacções de Diels-Alder, não se encontram disponíveis comercialmente e a sua síntese é também descrita. No segundo capítulo reporta-se uma nova e eficiente rota de síntese de (Z)- e (E)-3-estirilquinolin-4(1H)-onas a partir da reacção de Wittig da 4-cloro-3- formilquinolina e de 3-formilquinolin-4(1H)-onas com benzilidenotrifenilfosforanos. As (Z)-3-estirilquinolin-4(1H)-onas foram obtidas com elevada diastereoselectividade a partir da reacção das 3-formilquinolin- 4(1H)-onas N-protegidas enquanto que as (E)-3-estirilquinolin-4(1H)-onas foram preparadas através da reacção de Wittig da 4-cloro-3-formilquinolina seguida da hidrólise ácida das respectivas (Z)- e (E)-4-cloro-3-estirilquinolinas obtidas. Ambas as rotas sintéticas são eficientes, independentemente dos substituintes dos benzilidenotrifenilfosforanos. No terceiro capítulo, descreve-se um novo método de síntese de novas benzo[b]acridonas a partir da reacção de Diels-Alder de 3-formilquinolin-4(1H)- onas N-protegidas, que actuam como dienófilos, com dienos altamente reactivos, os orto-benzoquinodimetanos, preparados in situ através da extrusão térmica do dióxido de enxofre dos respectivos 2,2-dióxidos de 1,3-dihidrobenzo[ c]tiofeno. A reacção de cicloadição das 3-formilquinolin-4(1H)-onas N-protegidas com orto-benzoquinodimetanos origina as benzo[b]-1,6,6a,12atetra- hidroacridin-12(7H)-onas esperadas, que são o resultado da referida cicloadição seguida de desformilação in situ, e mostrou ser eficiente apenas quando o grupo amina está derivatizado com um grupo sacador de electrões. A desidrogenação destas benzo[b]-1,6,6a,12a-tetra-hidroacridin-12(7H)-onas em dimetilsulfóxido utilizando uma quantidade catalítica de iodo foi também descrita e originou como produto principal as benzo[b]acridin-12(7H)-onas N-desprotegidas. Todos os compostos novos sintetizados foram caracterizados por diversas técnicas analíticas, especialmente por estudos espectroscópicos de ressonância magnética nuclear (RMN), incluindo espectros de 1H e 13C, bidimensionais de correlação espectroscópica homonuclear e heteronuclear e de efeito nuclear de Overhauser (NOESY). Foram também efectuados, sempre que possível, espectros de massa (EM) e análises elementares ou espectros de massa de alta resolução (EMAR) para todos os compostos novos sintetizados. O tautomerismo da 3-formil- e 3-estirilquinolin-4(1H)-onas e as isomerizações (E) (Z) e rotacional das 3-estirilquinolin-4(1H)-onas e das 4-cloro-3-estirilquinolinas foram estudados através de ressonância magnética nuclear experimental (RMN 1H e 13C) e teórica [B3LYP/6-311++G(d,p)].

The solid-fluid transition properties of the n - 6 Lennard-Jones system are studied by means of extensive free energy calculations. Different values of the parameter n which regulates the steepness of the short-range repulsive interaction are investigated. Furthermore, the free energies of the n < 12 systems are calculated using the n = 12 system as a reference. The method relies on a generalization of the multiple histogram method that combines independent canonical ensemble simulations performed with different Hamiltonians and computes the free energy difference between them. The phase behavior of the fullerene C60 solid is studied by performing NPT simulations using atomistic models which treat each carbon in the molecule as a separate interaction site with additional bond charges. In particular, the transition from an orientationally frozen phase at low temperatures to one where the molecules are freely rotating at higher temperatures is studied as a function of applied pressure. The adsorption of molecular hydrogen in the zeolite NaA is investigated by means of grand-canonical Monte Carlo, in a wide range of temperatures and imposed gas pressures, and results are compared with available experimental data. A potential model is used that comprises three main interactions: van der Waals, Coulomb and induced polarization by the permanent electric field in the zeolite.

Background: The present study investigated whether prochlorperazine affects vestibular-ocular reflex (VOR) and vestibulo-perceptual function. Methods: We studied 12 healthy naïve subjects 3 hours after a single dose of oral prochlorperazine 5mg in a randomised, placebo-controlled, double-blind, cross-over study in healthy young subjects. Two rotational tests in yaw were used: 1) a Threshold task investigating perceptual motion detection and nystagmic thresholds (acceleration steps of 0.5deg/s/s) and 2) Suprathreshold responses to velocity steps of 90deg/s in which vestibulo-ocular (VO) and vestibulo-perceptual (VP) time constants of decay, as well as VOR gain, were measured. Results: Prochlorperazine had no effect upon any measure of nystagmic or perceptual vestibular function compared to placebo. This lack of effects on vestibular-mediated motion perception suggests that the drug is likely to act more as an antiemetic than as an anti-vertiginous agent.

Kinematic redundancy occurs when a manipulator possesses more degrees of freedom than those required to execute a given task. Several kinematic techniques for redundant manipulators control the gripper through the pseudo-inverse of the Jacobian, but lead to a kind of chaotic inner motion with unpredictable arm configurations. Such algorithms are not easy to adapt to optimization schemes and, moreover, often there are multiple optimization objectives that can conflict between them. Unlike single optimization, where one attempts to find the best solution, in multi-objective optimization there is no single solution that is optimum with respect to all indices. Therefore, trajectory planning of redundant robots remains an important area of research and more efficient optimization algorithms are needed. This paper presents a new technique to solve the inverse kinematics of redundant manipulators, using a multi-objective genetic algorithm. This scheme combines the closed-loop pseudo-inverse method with a multi-objective genetic algorithm to control the joint positions. Simulations for manipulators with three or four rotational joints, considering the optimization of two objectives in a workspace without and with obstacles are developed. The results reveal that it is possible to choose several solutions from the Pareto optimal front according to the importance of each individual objective.

With the recent advances in technology and miniaturization of devices such as GPS or IMU, Unmanned Aerial Vehicles became a feasible platform for a Remote Sensing applications. The use of UAVs compared to the conventional aerial platforms provides a set of advantages such as higher spatial resolution of the derived products. UAV - based imagery obtained by a user grade cameras introduces a set of problems which have to be solved, e. g. rotational or angular differences or unknown or insufficiently precise IO and EO camera parameters. In this work, UAV - based imagery of RGB and CIR type was processed using two different workflows based on PhotoScan and VisualSfM software solutions resulting in the DSM and orthophoto products. Feature detection and matching parameters influence on the result quality as well as a processing time was examined and the optimal parameter setup was presented. Products of the both workflows were compared in terms of a quality and a spatial accuracy. Both workflows were compared by presenting the processing times and quality of the results. Finally, the obtained products were used in order to demonstrate vegetation classification. Contribution of the IHS transformations was examined with respect to the classification accuracy.

This present study aimed to investigate the fatigue life of unused (new) endodontic instruments made of NiTi with control memory by Coltene™ and subjected to the multi curvature of a mandibular first molar root canal. Additionally, the instrument‟s structural behaviour was analysed through non-linear finite element analysis (FEA). The fatigue life of twelve Hyflex™ CM files was assessed while were forced to adopt a stance with multiple radius of curvature, similar to the ones usually found in a mandibular first molar root canal; nine of them were subjected to Pecking motion, a relative movement of axial type. To achieve this, it was designed an experimental setup with the aim of timing the instruments until fracture while worked inside a stainless steel mandibular first molar model with relative axial motion to simulate the pecking motion. Additionally, the model‟s root canal multi-curvature was confirmed by radiography. The non-linear finite element analysis was conducted using the computer aided design software package SolidWorks™ Simulation, in order to define the imposed displacement required by the FEA, it was necessary to model an endodontic instrument with simplified geometry using SolidWorks™ and subsequently analyse the geometry of the root canal CAD model. The experimental results shown that the instruments subjected to pecking motion displayed higher fatigue life values and higher lengths of fractured tips than those with only rotational relative movement. The finite element non-linear analyses shown, for identical conditions, maximum values for the first principal stress lower than the yield strength of the material and those were located in similar positions to the instrument‟s fracture location determined by the experimental testing results.

A detailed theoretical investigation of the large amplitude motions in the S, excited electronic state of formic acid (HCOOH) was done. This study focussed on the the S, «- So electronic band system of formic acid (HCOOH). The torsion and wagging large amplitude motions of the S, were considered in detail. The potential surfaces were simulated using RHF/UHF ab-initio calculations for the two electronic states. The energy levels were evaluated by the variational method using free rotor basis functions for the torsional coordinates and harmonic oscillator basis functions for the wagging coordinates. The simulated spectrum was compared to the slit-jet-cooled fluorescence excitation spectrum allowing for the assignment of several vibronic bands. A rotational analysis of certain bands predicted that the individual bands are a mixture of rotational a, b and c-type components.The electronically allowed transition results in the c-type or Franck-Condon band and the electronically forbidden, but vibronically allowed transition creates the a/b-type or Herzberg-Teller components. The inversion splitting between these two band types differs for each band. The analysis was able to predict the ratio of the a, b and c-type components of each band.

A fluorescence excitation spectrum of formic acid monomer (HCOOH) , has been recorded in the 278-246 nm region and has been attributed to an n >7r* electron promotion in the anti conformer. The S^< S^ electronic origins of the HCOOH/HCOOD/DCOOH/DCOOD isotopomers were assigned to weak bands observed at 37431.5/37461.5/37445.5/37479.3 cm'''. From a band contour analysis of the 0°^ band of HCOOH, the rotational constants for the excited state were estimated: A'=1.8619, B'=0.4073, and C'=0.3730 cm'\ Four vibrational modes, 1/3(0=0), j/^(0-C=0) , J/g(C-H^^^) and i/,(0-H^yJ were observed in the spectrum. The activity of the antisymmetric aldehyde wagging and hydroxyl torsional modes in forming progressions is central to the analysis, leading to the conclusion that the two hydrogens are distorted from the molecular plane, 0-C=0, in the upper S. state. Ab initio calculations were performed at the 6-3 IG* SCF level using the Gaussian 86 system of programs to aid in the vibrational assignments. The computations show that the potential surface which describes the low frequency OH torsion (twisting motion) and the CH wagging (molecular inversion) motions is complex in the S^ excited electronic state. The OH and CH bonds were calculated to be twisted with respect to the 0-C=0 molecular frame by 63.66 and 4 5.76 degrees, respectively. The calculations predicted the existence of the second (syn) rotamer which is 338 cm'^ above the equilibrium configuration with OH and CH angles displaced from the plane by 47.91 and 41.32 degrees.

We investigated the likelihood that hypokinesia/bradykinesia coexist with druginduced dyskinesias (DID) in patients with Parkinson's disease (PD). The influence of dyskinesias on rapid alternating movements (RAM) was investigated in ten dyskinetic patients (DPD). Their motor performance was compared to that of ten age/gendermatched non-dyskinetic patients (NDPD) and ten healthy control subjects. Whole-body magnitude (WBM) and fast pronation-supination at the wrist were recorded using 6- degrees of freedom magnetic motion tracker and forearm rotational sensors, respectively. Subjects were asked to pronate-supinate their dominant hand for 10s. Pre- and postmeasures were taken in a neutral position for 20s. RANGE (measure of hypokinesia), DURATION (measure of bradykinesia). VELOCITY (measure of bradykinesia) and IRREGULARITY (measure of fluctuations in movement amplitude) were used to assess RAM performance. Results showed that DPD patients had greater WBM than NDPD and control groups during rest and RAM performance. There were no differences in performance between NDPD and DPD groups for RANGE, DURATION and VELOCITY, despite significant longer disease duration for the DPD group (DPD = 15.5 ± 6.2 years versus NDPD = 6.6 ± 2.6 years). However, both the NDPD and DPD groups showed lower RANGE, longer DURATION, and reduced VELOCITY compared to controls,, suggesting the presence of bradykinesia and hypokinesia. In the case of IRREGULARITY, DPD patients showed clear fluctuations in movement amplitude compared to the NDPD and control groups. However, the lack of correlation between WBM and IRREGULARITY within the DPD group (Spearman's rank order, Rho - 0.31, p > 0.05), suggests that DID was not the primary cause of the fluctuating movementamplitude observed in that group. In conclusion, these findings suggest that DID may coexists with bradykinesia and hypokinesia, but that they are not inevitably accompanied with worsening motor performance.

An energy theory is formulated for the rotational energy levels in a p-complex Rydberg state of an asymmetric top molecule of symmetry C2v. The effective Hamiltonian used consists of the usual rigid rotor Hamiltonian augmented with terms representing electronic spin and orbital angular momentum effects. Criteria for assigning symmetry species to the rotational energy levels, following Houganfs scheme that uses the full molecular group,are established and given in the form of a table. This is particularly suitable when eigenvectors are calculated on a digital computer. Also, an intensity theory for transitions to the Rydberg p-complex singlet states is presented and selection rules in terms of symmetry species of energy states are established. Finally, applications to HpO and DpO are given.

L’étoile Wolf-Rayet WR 46 est connue pour sa variabilité complexe sur des échelles de temps relativement courtes de quelques heures et sur des échelles de temps plus longues de plusieurs mois. Des décalages périodiques mais intermittents en vitesse radiale ont déjà été observés dans ses raies d’émission optiques. Plusieurs périodes photométriques ont aussi été mesurées dans le passé. Des pulsations non-radiales, une modulation liée à la rotation rapide, ou encore la présence d’un compagnon de faible masse dont la présence reste à confirmer ont été proposées pour expliquer le comportement de l’étoile sur des échelles de temps de quelques heures. Dans un effort pour dévoiler sa vraie nature, nous avons observé WR 46 avec le satellite FUSE sur plusieurs cycles de variabilité à court terme. Nous avons trouvé des variations sur une échelle de temps d’environ 7,5 heures dans le continu ultraviolet lointain, dans l’aile bleue de la composante d’absorption du profil P Cygni du doublet de O vi 1032, 1038, ainsi que dans la composante d’absorption du profil P Cygni de S vi 933, 944. Nous avons également récupéré des données archivées de cette étoile obtenues avec le satellite XMM-Newton. La courbe de lumière en rayons X montre des variations sur une échelle de temps similaire aux courbes de lumière du continu ultraviolet et ultraviolet lointain, et le spectre rayons X de WR 46 est très mou avec un pic d’émission à des énergies plus faibles que 1 keV. Nous discutons des différentes contraintes sur la nature de la variabilité de cette étoile que ces nouvelles observations aident à poser. Parmi les scénarios suggérés, nous concluons que celui des pulsations non-radiales est le plus probable, bien que nous soyons encore loin d’une compréhension détaillée de WR 46.

Une série de dimères composés de thiophène-aniline encombrée stériquement a été synthétisée. Les différents processus de désactivation de l’état singulet excité ont été étudiés par UV-visible, fluorescence, phosphorescence, photolyse par impulsion laser et calculs théoriques. Les graphiques de Stern-Volmer obtenus à partir des expériences de désactivation des états singulet et triplet ont démontré l’efficacité de l’azométhine à désactiver les fluorophores. Les calculs semi-empiriques AM1 examinant l’effet des substituants encombrés ont démontrés que les groupements tert-butyls sur l’aniline ont moins d’influence sur la barrière de rotation N-aryl que les substitutions alkyles en ont sur la rotation de thiophène-C. Les calculs Rehm-Weller basés sur les potentiels d’oxydation et de réduction ont montré que l’autodésactivation de l’état excité des azométhines se fait par transfert d’électron photoinduit menant à une éradication complète de la fluorescence. Des complexes métalliques contenant des ligands azométhines ont aussi été préparés. Le ligand est composé d’une unité hydroxyquinoline lié à un cycle thiophène. Les données photophysiques de ces complexes indiquent un déplacement bathochromique aussi bien en absorbance qu’en fluorescence. Des dispositifs de détection d’ion métallique ont été préparés et un exemple à partir d’une solution de cuivre a montré un déplacement bathochromique.

Nous présentons les résultats de trois campagnes d'observation d'un mois chacune dans le cadre de l'étude de la collision des vents dans les systèmes binaires Wolf-Rayet + OB. Ce travail se concentre sur l'étude des objets de l'hémisphère sud n'ayant jamais encore fait l'objet d'études poussées dans ce contexte. À cela, nous avons ajouté l'objet archétype pour ce type de systèmes : WR 140 (WC7pd + O5.5fc) qui a effectué son dernier passage périastre en janvier 2009. Les deux premières campagnes (spectroscopiques), ont permis une mise à jour des éléments orbitaux ainsi qu'une estimation de la géométrie de la zone de collision des vents et d'autres paramètres fondamentaux des étoiles pour 6 systèmes binaires : WR 12 (WN8h), 21 (WN5o+O7V), 30 (WC6+O7.5V), 31 (WN4o+O8), 47 (WN6o+O5) et 140. Une période non-orbitale courte (probablement reliée à la rotation) a également été mesurée pour un des objets : WR 69 (WC9d+OB), avec une période orbitale bien plus grande. La troisième campagne (photométrique) a révélé une variabilité étonnamment faible dans un échantillon de 20 étoiles WC8/9. Cela supporte l'idée que les pulsations ne sont pas courantes dans ce type d'étoiles et qu'il est peu probable que celles-ci soient le mécanisme dominant de formation de poussière, suggérant, par défaut, le rôle prédominant de la collision des vents.

Le flou de mouvement de haute qualité est un effet de plus en plus important en rendu interactif. Avec l'augmentation constante en qualité des ressources et en fidélité des scènes vient un désir semblable pour des effets lenticulaires plus détaillés et réalistes. Cependant, même dans le contexte du rendu hors-ligne, le flou de mouvement est souvent approximé à l'aide d'un post-traitement. Les algorithmes de post-traitement pour le flou de mouvement ont fait des pas de géant au niveau de la qualité visuelle, générant des résultats plausibles tout en conservant un niveau de performance interactif. Néanmoins, des artefacts persistent en présence, par exemple, de mouvements superposés ou de motifs de mouvement à très large ou très fine échelle, ainsi qu'en présence de mouvement à la fois linéaire et rotationnel. De plus, des mouvements d'amplitude importante ont tendance à causer des artefacts évidents aux bordures d'objets ou d'image. Ce mémoire présente une technique qui résout ces artefacts avec un échantillonnage plus robuste et un système de filtrage qui échantillonne selon deux directions qui sont dynamiquement et automatiquement sélectionnées pour donner l'image la plus précise possible. Ces modifications entraînent un coût en performance somme toute mineur comparativement aux implantations existantes: nous pouvons générer un flou de mouvement plausible et temporellement cohérent pour plusieurs séquences d'animation complexes, le tout en moins de 2ms à une résolution de 1280 x 720. De plus, notre filtre est conçu pour s'intégrer facilement avec des filtres post-traitement d'anticrénelage.
