Vegetables are critical for human health as they are a source of multiple vitamins including vitamin E (VTE). In plants, the synthesis of VTE compounds, tocopherol and tocotrienol, derives from precursors of the shikimate and methylerythritol phosphate pathways. Quantitative trait loci (QTL) for alpha-tocopherol content in ripe fruit have previously been determined in an Solanum pennellii tomato introgression line population. In this work, variations of tocopherol isoforms (alpha, beta, gamma, and delta) in ripe fruits of these lines were studied. In parallel all tomato genes structurally associated with VTE biosynthesis were identified and mapped. Previously identified VTE QTL on chromosomes 6 and 9 were confirmed whilst novel ones were identified on chromosomes 7 and 8. Integrated analysis at the metabolic, genetic and genomic levels allowed us to propose 16 candidate loci putatively affecting tocopherol content in tomato. A comparative analysis revealed polymorphisms at nucleotide and amino acid levels between Solanum lycopersicum and S. pennellii candidate alleles. Moreover, evolutionary analyses showed the presence of codons evolving under both neutral and positive selection, which may explain the phenotypic differences between species. These data represent an important step in understanding the genetic determinants of VTE natural variation in tomato fruit and as such in the ability to improve the content of this important nutriceutical.

We identified a 4-year-old Brazilian boy from a family of Japanese descent and history of consanguinity, who suffered from severe recurrent pneumonia. He carries factor H (FH) deficiency associated with reduced levels of component C9 and low serum levels of C3 and factor B. His mother also presented low levels of these proteins and factor I, while his father and sister had only lower levels of FH. Western blot assays confirmed the complete absence of FH and FHL-1 polypeptides in this patient. Sequencing of the proband`s FH cDNA revealed a homozygous G453A substitution, encoding an Arg(127)His change. His mother, father and sister are heterozygous for this substitution. Despite the absence of FH in the plasma, this protein was detected in the patient`s fibroblasts, suggesting that Arg(127) may be important for FH secretion. Low concentrations of C9 were detected in the proband serum but no mutations in the patient`s C9 gene or promoter have been identified, suggesting that this is a consequence of uncontrolled complement activation and high C9 consumption.

Freshwater fish are an important source of protein, but they also contain other highly nutritive components such as fats. Polyunsaturated fatty acids (PUFAs) are essential for normal growth, development and reproduction of vertebrates. The antioxidant role of vitamin E in cell membranes prevents fatty acid and cholesterol oxidation, thereby promoting PUFA and subcellular particle stabilization. The effects of vitamin E supplementation on the quality of Nile tilapia (Oreochromis niloticus) carcass were investigated. The experiments were carried out in an experimental laboratory over 106 d. After sex reversal, 400 early juvenile O. niloticus were tested in a completely randomized experiment with 5 treatments (4 repetitions each), consisting of vitamin E monophosphate supplementation at 0, 50, 100, 150 or 200 mg/kg of a base diet. Treatment diets contained equal amounts of protein and energy. Tilapias supplemented with vitamin E contained arachidonic acid (20:4 omega-6; AA) which participates in inflammatory response. Nile tilapia carcasses that received vitamin E at 100 and 150 mg/kg diet had improved carcass quality by increasing the PUFA:SFA ratio and had the highest levels of polyunsaturated fatty acids from the omega-3 (linolenic acid; 18:3 omega-3) and omega-6 (linoleic acid; 18:2 omega-6) series. O experimento foi conduzido em câmara climática para determinar os efeitos das vitaminas D3 e C sobre o desempenho, temperatura corporal e qualidade dos ovos de poedeiras em três temperaturas: termoneutra (24,8º a 27,0º C), estresse calórico cíclico (26,2º C por 16h e 32,1º C por 8h) e estresse calórico constante (30,0º a 32,0º C). Foram utilizadas 144 galinhas brancas com 31 semanas num delineamento inteiramente ao acaso em arranjo fatorial 3x3x3: vitamina D3 (2500, 3000 e 3500 UI/kg), vitamina C (0, 200 e 400 ppm) e temperatura ambiente (termoneutra, estresse calórico cíclico e constante). As características avaliadas foram: consumo de ração (CR), produção de ovos (PO), peso dos ovos (PE), massa de ovos (MO), conversão alimentar (CA), porcentagens de albúmem (AL) e de gema (GE), unidade Haugh (UH), índice de gema (IG), porcentagem de casca (PC), espessura de casca (EC), gravidade específica dos ovos (GO), temperaturas dorsal (TD) e retal (TR). A vitamina D3 interferiu nas características PC, EC, GO e TD; a vitamina C no IG, PC e GO. A temperatura ambiente não afetou somente a UH. Concluiu-se que níveis mais altos de vitamina D3 e níveis de 200 ou 400 ppm de vitamina C na dieta podem melhorar a qualidade da casca e que o estresse calórico prejudicou as principais características avaliadas.

Racional A retirada total ou parcial do estômago resulta em conseqüências nutricionais, agudas ou crônicas, perfeitamente prognosticáveis, mas nem sempre ponderadas na terapia pós-operatória. Objetivo - Rever as participações mecânicas e químicas do estômago no aproveitamento do nutriente dietético, e as conseqüências nutricionais da gastrectomia. Resultados - A deficiência energética, com conseqüente perda de peso, acompanha inversamente o volume gástrico remanescente e o tempo pós-operatório; tem a anorexia e diarréia (má absorção) como principais causas, sendo a primeira decorrente de fatores emocionais ou de mediadores químicos de ação hipotalâmica. A diarréia pode ser decorrente da maior motilidade ou do supercrescimento bacteriano intestinais, com o agravante da insuficiência pancreática exócrina e maior esvaziamento da vesícula biliar. A má absorção traz conseqüências não apenas energética-protéica com a perda fecal de gordura e nitrogênio, como também vitamínico-mineral pelo menor aproveitamento da vitamina D e cálcio dietéticos. A anemia verificada no gastrectomizado é conseqüente à diminuição da produção de HCl (e menor solubilização do ferro) e do fator intrínseco (com menor absorção da vitamina B12). Conclusão - Perda de peso e anemia são os sinais de desnutrição mais comumente observados nestes pacientes, em intensidade e duração variáveis dependentes do tipo de cirurgia e do tempo e tratamento nutricional pós-operatório, sendo recomendável o tratamento dietético supervisionado.

In order to know which stage of fruit development is better for acerola industrialization, we studied the PME specific activity, pectin content and vitamin C at various stages of development. The acerola fruits were classified according to colour and weight in five stages: immature green (2.62-3.21 g), green (4.04-4.83 g), mature green/yellow (5.03-5.88 g), pale-red (6.16-6.77 g) and ripe mature (6.92-8.37 g). The results showed that the highest content of pectin and vitamin C occurred at the immature green stage, 4.51 +/- 0.1% yield, 2424 mg/100 g of pulp and decreased as fruit ripened, 2.99 +/- 0.03% yield, 957 +/- 0.0 mg/100 g of pulp, respectively. However, at the same stages, the values of PME specific activity were lowest, 0.61 +/- 0.01 and 0.55 +/- 0.0 units g(-1)/g of pulp, respectively. The highest value of PME activity was 2.08 +/- 0.01 units g(-1)/g of pulp in the green stage. (C) 2001 Elsevier B.V. Ltd. All rights reserved.

Cotton (Gossypium hirsutum var. Latifolium) was grown in nutrient media, at two K levels: 58.5 mg/K and 11.7 mg/K. Potassium deficiency (11.7 mg K/g of K) was imposed upon cotton plants at different stages of plant development. A sequence of increasing sensitivity to K deficiency among cotton plant parts was observed: leaves < bolls < roots < stems. When K deficiency symptoms are clearly visible in the leaves, all the other plant parts are already affected. Bolls are a very important component in K partitioning within the cotton plant, but K is required most by the bur itself and is not translocated to seeds or fibers. Cotton could overcome a 30 day deficiency late in the season without significant losses in lint and seed cotton yields.

Water temperature alterations can determine harmful physiological modifications in fish, which should be prepared to cope with this, and nutrition strategies seem to be essential. This study evaluated the effects of different levels of vitamin C and lipids on physiological responses of Nile tilapia, Oreochromis niloticus, submitted to temperature stress. There were two phases: Phase I - preparing fish to store vitamin C and lipid at appropriate temperature, and Phase II - evaluating the contributions these reserves make to fish physiology under low-temperature stress. The experiment used a 3 x 2 factorial design with three vitamin C levels (300, 600, and 1200 mg/kg diet) and two lipid levels (8.0 and 12.0%), plus absence of nutrient test and a diet of 6.0% lipids and 125.0 mg/kg vitamin C. In Phase I, 192 fish were kept at 26.0 +/- 1.0 C for 112 d, and in Phase II, 48 fish were kept at 18.0 +/- 0.5 C for 32 d and at 15.0 +/- 0.5 C for 11 d. Fish fed C0L0 diet showed lower erythrocytes values in both phases; higher vitamin C supplement determined higher red blood cell (RBC) number and higher hematocrit (Htc) (Phase II); Htc was significantly lower in Phase II; after temperature stress, fish fed C0L0 diet had higher mean corpuscular volume, lower hemoglobin corpuscular concentration, and significantly lower vitamin C concentration in the liver; and higher supplementation determined a higher concentration in the liver (Phases I and II). Higher plasmatic cortisol concentration was seen in fish fed C0L0 diet. In conclusion, our results show that the absence of vitamin C in diets impairs RBC formation and does not enable fish to cope with stress; excess vitamin C is efficient in mitigating stress and 600 mg/kg diet is economic and physiologically sufficient to prepare fish for coping with low-temperature stress. Lipid supplementation does not determine alterations in stress biochemical parameters.

The effect of dietary vitamin (VS) and mineral (MS) mix withdrawal between 42 and 49 d of age on broiler chicken performance was evaluated. The diets were formulated based on corn and soybean meal, and the experiments were conducted in floor pens using wood shavings as litter. Trial I evaluated withdrawal of VS and MS mix at 42, 45, and 49 d. The results of this trial showed that VS and MS mix withdrawal at 42 d of age did not impair feed intake (FI) or weight gain (WG), but feed conversion (FC) was improved (P = 0.043) when both mixes were maintained in the diets until 49 d of age. Trial 2 assessed the withdrawal of VS or MS mix at 42 d of age. FI and WG were not affected by withdrawal, but FC was poorer (P = 0.035) for the broilers fed the diet without VS. The relative liver weight was reduced by VS or MS mix withdrawal (P = 0.014 and P = 0.001, respectively). Carcass, breast, and leg yields were not affected by VS or MS mix withdrawals in either trial. The findings of the study suggested that VS mix withdrawal during the final period of broiler chicken growth was more deleterious than the withdrawal of MS mix, because it affected FC ratio. VS or MS withdrawal during this period did not affect carcass yield.
