The mechanism of action and properties of a solid-phase ligand library made of hexapeptides (combinatorial peptide ligand libraries or CPLL, for capturing the "hidden proteome", i.e. the low- and very low-abundance proteins Constituting the vast majority of species in any proteome. as applied to plant tissues, are reviewed here. Plant tissues are notoriously recalcitrant to protein extraction and to proteome analysis, Firstly, rigid plant cell walls need to be mechanically disrupted to release the cell content and, in addition to their poor protein yield, plant tissues are rich in proteases and oxidative enzymes, contain phenolic Compounds, starches, oils, pigments and secondary metabolites that massively contaminate protein extracts. In addition, complex matrices of polysaccharides, including large amount of anionic pectins, are present. All these species compete with the binding of proteins to the CPLL beads, impeding proper capture and identification I detection of low-abundance species. When properly pre-treated, plant tissue extracts are amenable to capture by the CPLL beads revealing thus many new species among them low-abundance proteins. Examples are given on the treatment of leaf proteins, of corn seed extracts and of exudate proteins (latex from Hevea brasiliensis). In all cases, the detection of unique gene products via CPLL Capture is at least twice that of control, untreated sample. (c) 2008 Elsevier B.V. All rights reserved.

The new ingenane diterpene, 5-deoxy-13-hydroxyingenol, was isolated from the alcohol preserved fresh latex of the stems of Mabea excelsa and characterized from its semi-synthetic triacetate. This is the first instance of an ingenane diterpene obtained from species other than those of Euphorbia and Elaeophorbia. This diterpene occurred in the latex in the form of an inseparable mixture of six aliphatic mono-esters of the tertiary C-13 hydroxy group.

This paper introduces PSOPT, an open source optimal control solver written in C++. PSOPT uses pseudospectral and local discretizations, sparse nonlinear programming, automatic differentiation, and it incorporates automatic scaling and mesh refinement facilities. The software is able to solve complex optimal control problems including multiple phases, delayed differential equations, nonlinear path constraints, interior point constraints, integral constraints, and free initial and/or final times. The software does not require any non-free platform to run, not even the operating system, as it is able to run under Linux. Additionally, the software generates plots as well as LATEX code so that its results can easily be included in publications. An illustrative example is provided.

This paper investigates the acquisition of syntax in L2 grammars. We tested adult L2 speakers of Spanish (English L1) on the feature specification of T(ense), which is different in English and Spanish in so-called subject-to-subject raising structures. We present experimental results with the verb parecer “to seem/to appear” in different tenses, with and without experiencers, and with Tense Phrase (TP), verb phrase (vP) and Adjectival Phrase (AP) complements. The results show that advanced L2 learners can perform just like native Spanish speakers regarding grammatical knowledge in this domain, although the subtle differences between both languages are not explicitly taught. We argue that these results support Full Access approaches to Universal Grammar (UG) in L2 acquisition, by providing evidence that uninterpretable syntactic features can be learned in adult L2, even when such features are not directly instantiated in the same grammatical domain in the L1 grammar.

Different theoretical accounts of second language (L2) acquisition differ with respect to whether or not advanced learners are predicted to show native like processing for features not instantiated in the native language (L1). We examined how native speakers of English, a language with number but not gender agreement, process number and gender agreement in Spanish. We compare agreement within a determiner phrase (órgano muy complejo “[DP organ-MASC-SG very complex-MASC-SG]”) and across a verb phrase (cuadro es auténtico “painting-MASC-SG [VP is authentic-MASC-SG]”) in order to investigate whether native like processing is limited to local domains (e.g. within the phrase), in line with Clahsen and Felser (2006). We also examine whether morphological differences in how the L1 and L2 realize a shared feature impact processing by comparing number agreement between nouns and adjectives, where only Spanish instantiates agreement, and between demonstratives and nouns, where English also instantiates agreement. Similar to Spanish natives, advanced learners showed a P600 for both number and gender violations overall, in line with the Full Transfer/Full Access Hypothesis (Schwartz and Sprouse, 1996), which predicts that learners can show native-like processing for novel features. Results also show that learners can establish syntactic dependencies outside of local domains, as suggested by the presence of a P600 for both within and across phrase violations. Moreover, similar to native speakers, learners were impacted by the structural distance (number of intervening phrases) between the agreeing elements, as suggested by the more positive waveforms for within than across-phrase agreement overall. These results are consistent with the proposal that learners are sensitive to hierarchical structure.

Previous studies showed that intercellular communication by gap junctions has a role in bone formation. The main connexin involved in the development, differentiation, and regulation of bone tissue is connexin (Cx) 43. In addition, Cx46 is also expressed, mostly localized within the trans-Golgi region. Alterations in the expression pattern and aberrant location of these connexins are associated with oncogenesis, demonstrating a deficient gap junctional intercellular communication (GJIC) capacity in neoplastic tissues. In this study, we evaluated normal and neoplastic bone tissues regarding the expression of Cx43 and Cx46 by immunofluorescence, gene expression of these connexins by real-time PCR, and their correlation with cell proliferation index and deposition of collagen. Fourteen neoplastic bone lesions, including 13 osteosarcomas and I multilobular tumor of bone, were studied. The mRNA levels of Cx43 were similar between normal and neoplastic bone tissue. In normal bone tissue, the Cx43 protein was found mainly in the intercellular membranes. However, in all bone tumors studied here, the Cx43 was present in both cell membranes and also aberrantly in the cytoplasm. Regarding only tumor samples, we determined a possible inverse correlation between Cx43 expression and cellular proliferation, although a positive correlation between Cx43 expression and collagen deposition was also noted. In contrast, Cx46 had lower levels of expression in neoplastic bone tissues when compared with normal bone and was found retained in the perinuclear region. Even though there are differences between these two connexins regarding expression in neoplastic versus normal tissues, we concluded that there are differences regarding the subcellular location of these connexins in normal and neoplastic dog bone tissues and suggest a possible correlation between these findings and some aspects of cellular proliferation and possibly differentiation.

The deep crustal structure of the Parana Basin of southern Brazil is investigated by analyzing P- and PP-wave receiver functions at 17 Brazilian Lithosphere Seismic Project stations within the basin. The study area can be described as a typical Paleozoic intracratonic basin that hosts one of the largest Large Igneous Province of the world and makes a unique setting for investigating models of basin subsidence and their interaction with mantle plumes. Our study consists of (1) an analysis of the Moho interaction phases in the receiver functions to obtain the thickness and bulk Vp/Vs ratio of the basin`s underlying crust and (2) a joint inversion with Rayleigh-wave dispersion velocities from an independent tomographic study to delineate the detailed S-wave velocity variation with depth. The results of our analysis reveal that Moho depths and bulk Vp/Vs ratios (including sediments) vary between 41 and 48 km and between 1.70 and 1.76, respectively, with the largest values roughly coinciding with the basin`s axis, and that S-wave velocities in the lower crust are generally below 3.8 km/s. Select sites within the basin, however, show lower crustal S-wave velocities slightly above 3.9 km/s suggestive of underplated mafic material. We show that these observations are consistent with a fragmented cratonic root under the Parana basin that defined a zone of weakness for the initial Paleozoic subsidence of the basin and which allowed localized mafic underplating of the crust along the suture zones by Cenozoic magmatism.

Knowing the best 1D model of the crustal and upper mantle structure is useful not only for routine hypocenter determination, but also for linearized joint inversions of hypocenters and 3D crustal structure, where a good choice of the initial model can be very important. Here, we tested the combination of a simple GA inversion with the widely used HYPO71 program to find the best three-layer model (upper crust, lower crust, and upper mantle) by minimizing the overall P- and S-arrival residuals, using local and regional earthquakes in two areas of the Brazilian shield. Results from the Tocantins Province (Central Brazil) and the southern border of the Sao Francisco craton (SE Brazil) indicated an average crustal thickness of 38 and 43 km, respectively, consistent with previous estimates from receiver functions and seismic refraction lines. The GA + HYPO71 inversion produced correct Vp/Vs ratios (1.73 and 1.71, respectively), as expected from Wadati diagrams. Tests with synthetic data showed that the method is robust for the crustal thickness, Pn velocity, and Vp/Vs ratio when using events with distance up to about 400 km, despite the small number of events available (7 and 22, respectively). The velocities of the upper and lower crusts, however, are less well constrained. Interestingly, in the Tocantins Province, the GA + HYPO71 inversion showed a secondary solution (local minimum) for the average crustal thickness, besides the global minimum solution, which was caused by the existence of two distinct domains in the Central Brazil with very different crustal thicknesses. We report on the measurements of the quantum Hall effect states in double quantum well structures at the filling factors v = 4N + 1 and 4N + 3, where N is the Landau index number, in the presence of the in-plane magnetic field. The quantum Hall states at these filling factors vanish and reappear several times. Repeated reentrance of the transport gap occurs due to the periodic vanishing of the tunneling amplitude in the presence of the in-plane field. When the gap vanishes, the transport becomes anisotropic. The anisotropy persist at half-odd filling factors, when bilayer quantum Hall states are recovered with increase of the tilt angle. Cationic supported bilayers on latex are useful to isolate and immobilize oppositely charged proteins as a monomolecular layer over a range of low protein concentrations and particle number densities. Cholera toxin (CT) from Vibrio cholerae, an 87 kDa AB(5) hexameric protein and bovine serum albumin (BSA) self-assembled on dioctadecyldimethylammonium bromide (DODAB) supported bilayers with high affinity yielding highly organized and monodisperse particulates at 5 x 10(9) particles/mL, over a range of low protein concentrations (0-0.025 mg/mL BSA or CT). Protein association onto the bilayer-covered polystyrene sulfate (PSS) was determined from adsorption isotherms, dynamic light scattering for size distributions and zeta-potential analysis revealing a monomolecular, thin and highly organized protein layer surrounding each particle with potential for biospecific recognition such as antigen-antibody, receptor-ligand, hybridization of oligonucleotide sequences, all of them important in immunodiagnosis, selective biomolecular chromatographic separations, microarrays design and others.

The interaction between cationic bilayer fragments and a model oligonucleotide was investigated by differential scanning calorimetry, turbidimetry, determination of excimer to monomer ratio of 2-(10-(1-pyrene)-decanoyl)-phosphatidyl-choline in bilayer fragment dispersions and dynamic light scattering for sizing and zeta-potential analysis. Salt (Na(2)HPO(4)), mononucleotide (2`-deoxyadenosine-5`-monophosphate) or poly (dA) oligonucleotide (3`-AAA AAA AAA A-5`) affected structure and stability of dioctadecyldimethylammonium bromide bilayer fragments. Oligonucleotide and salt increased bilayer packing due to bilayer fragment fusion. Mononucleotide did not reduce colloid stability or did not cause bilayer fragment fusion. Charge neutralization of bilayer fragments by poly (dA) at 1:10 poly (dA):dioctadecyldimethylammonium bromide molar ratio caused extensive aggregation, maximal size and zero of zeta-potential for the assemblies. Above charge neutralization, assemblies recovered colloid stability due to charge overcompensation. For bilayer fragments/poly (dA), the nonmonotonic behavior of colloid stability as a function of poly (dA) concentration was unique for the oligonucleotide and was not observed for Na(2)HPO(4) or 2`-deoxyadenosine-5`-monophosphate. For the first time, such interactions between cationic bilayer fragments and mono- or oligonucleotide were described in the literature. This work is aimed at studying the adsorption mechanism of short chain 20-mer pyrimidinic homoss-DNA (oligodeoxyribonucleotide, ODN: polyC(20) and polyT(20)) onto CNT by reflectometry. To analyze the experimental data, the effective-medium theory using the Bruggemann approximation represents a Suitable optical model to account for the surface properties (roughness, thickness, and optical constants) and the size of the adsorbate. Systematic information about the involved interactions is obtained by changing the physicochemical properties of the system. Hydrophobic and electrostatic interactions are evaluated by comparing the adsorption oil hydrophobic CNT and oil hydrophilic silica and by Modulating the ionic Strength With and without Mg(2+). The ODN adsorption process oil CNT is driven by hydrophobic interactions only when the electrostatic repulsion is Suppressed. The adsorption mode results in ODN molecules in a side-on orientation with the bases (nonpolar region) toward the surface. This unfavorable orientation is partially reverse by adding Mg(2+). On the other hand, the adsorption oil silica is dominated by the strong repulsive electrostatic interaction that is screened at high ionic strength or mediated by Mg(2+). The cation-mediated process induces the interaction of the phosphate backbone (polar region) with the surface, leaving the bases free for hybridization. Although the general adsorption behavior of the pyrimidine bases is the same, polyC(20) presents higher affinity for the CNT Surface due to its acid-base properties.

Esta dissertação faz uma análise prévia da predicação complexa no modelo da Gramática Tradicional; aponta, em seguida, o enfoque das estruturas predicativas complexas à luz da Gramática Gerativa modelo de Princípios & Parâmetros (Chomsky, 1981, 1986). Ressalta que as estruturas sintáticas são projetadas a partir do léxico e correspondem a várias construções de predicação complexa. Mostra que os predicados complexos formados por APs e DPs predicativos apresentam características sintáticas e semânticas diferentes. Focaliza que essas construções superficialmente semelhantes são, em essência, diferentes, porque cada uma delas possui um processo próprio de estruturação interna, do qual decorre um sentido específico. Aponta, essencialmente, dois tipos de estruturas complexas: predicados secundários e miniorações complementos. Os predicados secundários se dividem, ainda, em predicados secundários orientados para o sujeito de [DP IP], predicados secundários orientados para o objeto de [DP VP] e predicados resultativos de [DP VP]. Os predicados secundários de [DP IP] e de [DP VP] e os predicados resultativos de [DP VP] constituem as Small Clauses adjuntas de predicados secundários. Apresenta que é possível encontrar evidências que as estruturas predicativas complexas do PB estão amalgamadas na concepção de Small Clauses nominais. Postula que as estruturas predicativas complexas do PB e as estruturas de Small Clauses nominais são em essência semelhantes. Tais estruturas possuem um mesmo processo de estruturação interna, que são analisadas conforme as configurações das representações sintáticas de cada sentença.

Este estudo esteve envolvido com o desenvolvimento de um modelo para avaliação de valor de supermercado, sob a perspectiva do consumidor. O conceito de valor empregado neste trabalho parte da idéiade que um comportamento somente deve ser levado adiante caso o prazer decorrente seja superior à dor relacionada à obtenção desse prazer. Em Marketing, isto quer dizer que o consumo de determinado bem ou serviço exige do indivíduo uma certa quantidade de sacrifício (sacrifício percebido) para gozar dos benefícios associados (qualidade percebida). Em outras palavras, o consumidor precisa levar em consideração uma série de fatores numa situação de n avaliação e escolha. Para tanto, foram feitas duas consultas a clientes de supermercado do sexo feminino, pertencentes às classes A e B, na cidade de Campo Grande (MS), sendo uma qualitativa (n = 25) e outra quantitativa (n = 200). Os resultados indicam que valor percebido (VP) é uma função linear de qualidade percebida (QP), mas não de sacrifício percebido (SP). Portanto, duas das quatro hipóteses do estudo não foram confirmadas. Cerca de três quartos das mulheres preferem fazer ·uma compra pesada por mês e várias avulsas", aliás comportamento mais evidente nas faixa etárias mais elevadas. Para metade das entrevistadas, fazer compra em supermercado dá mais prazer do que trabalho. A maioria das mulheres que não trabalha fora sente prazer em fazer compra, ao contrário das mulheres que trabalham fora que acham isso uma obrigação. Por último, as mulheres que não trabalham fora, as de idade mais elevada e aquelas que consideram a compra um prazer, valorizam melhor o seu supermercado. O estudotambém permitiu verificar as características determinantes do supermercado que o cliente leva em consideração. Os atributos mais valorizados pelas entrevistadas foram substituiçao de produtos estragados e/ou vencidos e limpeza da loja. No entanto, elas usaram exposiçêo e organizaçao dos produtos, limpeza da loja e qualidade dos produtos hortifrutícolas para perceber valor. Os resultados dos testes de hipóteses parecem indicar que não é necessário o uso de variáveis de baixo nível de percepção (atributos de qualidade e de custo) para medir VP de supermercado. Os três atributos que permaneceram no modelo para explicar VP, através de QP (limpeza da loja, exposiçêo e organizaçao dos produtos e qualidade dos produtos hortifrutlcolas), o fazem moderadamente (R2 = 24,7%). Por outro lado, na avaliação global que as entrevistadas fizeram das lojas, QP (agora como variável independente) apresenta um coeficiente de explicação R 2 = 37,4%, resultadosuperior aoverificado quando a relação se deu com atributos específicos. Entretanto, se os clientes percebem valor apenas se valendo de QP (avaliação global), a formação da percepção de qualidade é feita usando sinais específicos associados ao produto "supermercado", sinais estes que incluem os três atributos que permaneceram no modelo. Assim, um modelo que inclui variáveis de percepção mais elevada (QP e SP) para inferir VP permite responder à seguinte questão: qual é a avaliação global que os clientes fazem de seu supermercado? Por outro lado, um modelo que julga VP a partir de atributos especificos (variáveis de baixo nível de percepção) deve responder à seguinte indagação: como se dá a percepção de valor do cliente em relação ao seu supermercado? Outros resultados são discutidos e também são apresentadas sugestões para futuras pesquisas e as principais implicações acadêmicas e empresariais .

A resposta ao broncodilatador (BD) é tradicionalmente medida pela variação do volume expiratório forçado no primeiro segundo (VEF1). Pacientes com doença pulmonar obstrutiva crônica (DPOC) apresentam pouca ou nenhuma resposta neste parâmetro. Outros parâmetros funcionais, medidos por pletismografia, podem ser úteis na avaliação da reversibilidade da obstrução em pacientes com DPOC. Com os objetivos de avaliar a freqüência de resposta ao BD no VEF1 em uma amostra de pacientes com DPOC; correlacioná-la com dados clínicos e avaliar a freqüência de resposta na capacidade vital forçada (CVF), capacidade vital lenta (CVL), capacidade inspiratória (CI), volume residual (VR), resistência das vias aéreas (RVA) e condutância específica das vias aéreas (GVA/VP), foi proposto este estudo. Sessenta e quatro pacientes com DPOC foram submetidos a pletismografia de corpo inteiro e medida a reversibilidade da broncoconstrição após 400μg de fenoterol. Dos 64 pacientes avaliados, 31,3% tiveram resposta no VEF1. A freqüência de pacientes com resposta ao BD, quando medida pelos outros parâmetros em estudo, foi de 50% na CVF, 46,9% no VR, 46,8% na GVA/VP, 37,1% na CVL, 34,4% na CI e 30,6% na RVA. Pacientes sem resposta no VEF1, apresentaram a seguinte freqüência de resposta: 47,5% no VR, 38,1% na GVA/VP, 35% na CI, 31,8% na CVF, 23,8% na CVL e 21,4% na RVA. Concluiu-se que volumes pulmonares estáticos e resistência / condutância das vias aéreas, quando incluídos na avaliação da resposta ao BD, além do VEF1, permitem avaliar com maior amplitude o número de pacientes com resposta funcional à prova farmacodinâmica. Estes resultados estão de acordo com a impressão clínica de que muitos pacientes com DPOC, mesmo sem melhora no VEF1 após BD, apresentam melhora clínica.
