Signaling events controlled by calcineurin promote cardiac hypertrophy, but the degree to which such pathways are required to transduce the effects of various hypertrophic stimuli remains uncertain. In particular, the administration of immunosuppressive drugs that inhibit calcineurin has inconsistent effects in blocking cardiac hypertrophy in various animal models. As an alternative approach to inhibiting calcineurin in the hearts of intact animals, transgenic mice were engineered to overexpress a human cDNA encoding the calcineurin-binding protein, myocyte-enriched calcineurin-interacting protein-1 (hMCIP1) under control of the cardiac-specific, α-myosin heavy chain promoter (α-MHC). In unstressed mice, forced expression of hMCIP1 resulted in a 5–10% decline in cardiac mass relative to wild-type littermates, but otherwise produced no apparent structural or functional abnormalities. However, cardiac-specific expression of hMCIP1 inhibited cardiac hypertrophy, reinduction of fetal gene expression, and progression to dilated cardiomyopathy that otherwise result from expression of a constitutively active form of calcineurin. Expression of the hMCIP1 transgene also inhibited hypertrophic responses to β-adrenergic receptor stimulation or exercise training. These results demonstrate that levels of hMCIP1 producing no apparent deleterious effects in cells of the normal heart are sufficient to inhibit several forms of cardiac hypertrophy, and suggest an important role for calcineurin signaling in diverse forms of cardiac hypertrophy. The future development of measures to increase expression or activity of MCIP proteins selectively within the heart may have clinical value for prevention of heart failure.

There is strong converging evidence that the intermediate and medial part of the hyperstriatum ventrale of the chick brain is a memory store for information acquired through the learning process of imprinting. Neurons in this memory system come, through imprinting, to respond selectively to the imprinting stimulus (IS) neurons and so possess the properties of a memory trace. Therefore, the responses of the intermediate and medial part of the hyperstriatum ventrale neurons to a visual imprinting stimulus were determined before, during, and after training. Of the total recorded population, the proportions of IS neurons shortly after each of two 1-h training sessions were significantly higher (approximately 2 times) than the pretraining proportion. However, ≈4.5 h later this proportion had fallen significantly and did not differ significantly from the pretraining proportion. Nevertheless, ≈21.5 h after the end of training, the proportion of IS neurons was at its highest (approximately 3 times the pretraining level). No significant fluctuations occurred in the proportions of neurons responding to the alternative stimulus. In addition, nonmonotonic changes were found commonly in the activity of 230 of the neurons tracked individually from before training to shortly after the end of training. Thus the pattern of change in responsiveness both at the population level and at the level of individual neurons was highly nonmonotonic. Such a pattern of change is not consistent with simple models of memory based on synaptic strengthening to asymptote. A model is proposed that accounts for the changes in the population responses to the imprinting stimulus in terms of changes in the responses of individual neurons.

The Escherichia coli DEAD (Asp-Glu-Ala-Asp) box protein DbpA is a putative RNA helicase and established RNA-dependent ATPase and is the only member of the DEAD box protein family for which a specific RNA substrate, bacterial 23S rRNA, has been identified. We have investigated the nature of this specificity in depth and have localized by deletion mutagenesis and PCR a single region of 93 bases (bases 2496-2588) in 23S rRNA that is both necessary and sufficient for complete activation of ATPase activity of DbpA. This target region forms part of the peptidyltransferase center and includes many bases involved in interaction with the 3' terminal adenosines of both A- and P-site tRNAs. Deletion of stem loops within the 93-base segment abolished ATPase activation. Similarly, point mutations that disrupt base pairing within stem structures ablated stimulation of ATPase activity. These data are consistent with roles for DbpA either in establishing and/or maintaining the correct three-dimensional structure of the peptidyltransferase center in 23S rRNA during ribosome assembly or in the peptidyltransferase reaction.

Tema 02: Teorías de intervención en el patrimonio histórico: del monumento arquitectónico al patrimonio urbano. Se trata del texto desarrollado del tema que traza un recorrido por las principales actutudes y teorías de intervención en el patrimonio arquitectónico. Por un lado tres momentos clave. Primero: el modo clasicista (ss. XV-XVIII) de imposición sobre las preexistencias. Segundo el siglo XIX con las dos posiciones extremas de 'no restaurar' (J, Ruskin) y la de 'reconstruir idealmente' (V.-le-Duc). Tercero: la formulación de la teoría del restauro scientifico (C. Boitto). Se termina el tema con la creación del patrimonio 'urbano' (C. Sitte y G. Giovannoni) y una reflexión en torno al 'consagración' del patrimonio arquitectónico y urbano.

El presente tema desarrolla las diferentes actitudes de la arquitectura ante la intervención en el patrimonio, para lo que se sigue el esquema propuesto por Solà-Morales (1982) y se refuerza con el discurso de F. Choay (1992). Se trata de un recorrido cronológico donde se distinguen tres etapas a partir del Renacimiento, con un periodo previo donde no se tenía conciencia de diferenciar en la actuación el tiempo pasado del tiempo presente. Se comienza con un periodo previo de invasión y superposición de estratos correspondiente a las edades antigua y media y ya se prosigue con los tres estadios. Un primero que abarca los ss. XV-XVIII, donde las teorías de arquitectura inspiradas en el mundo clásico (Alberti y Palladio), con su discurso de seguridad totalizadora, hace que los lenguajes del sistema clasicista oculten todo legado anterior. Un segundo estadio, que tiene lugar en el s. XIX, al tiempo que se construye la disciplina de la Historia del Arte, de idealización del pasado, en el que se postulan dos planteamientos opuestos: quien sostiene que los monumentos son intocables (Ruskin) y no procede restauración alguna, y quien sostiene que, al contrario, los monumentos deben presentarse con el esplendor que debieron tener en origen (Viollet). Y un tercer estadio, conocido como Restauro Scientifico (Boito), ya del s. XX, que trata de armonizar ambas posturas mediante una teoría de intervención mínima y por contraste (donde se sumará la reversibilidad), la cual será la base de las teorías actuales de intervención en el patrimonio: la autenticidad del monumento radicará en el proceso histórico. Por último, se aborda la cuestión del patrimonio urbano (Giovannoni), construida a lo largo del s. XX, por el que los centros históricos o partes urbanas también se consideran monumentos.

This layer is a georeferenced raster image of the historic paper map entitled: The peninsula of India : from the Kistnah River to Cape Comorin : exhibiting the partition of the territories of the late Tippoo Sultaun, according to the treaty of 1792, made by the Marquis Cornwallis, and that of 1799, made by the Marquis Wellesley by J. Rennell ; writing b W. Harrison. It was published by J. Rennell : sold by G. Nicol in 1800. Scale [ca. 1:2,300,000]. The image inside the map neatline is georeferenced to the surface of the earth and fit to the Kalianpur 1975 India Zone III projected coordinate system. All map collar and inset information is also available as part of the raster image, including any inset maps, profiles, statistical tables, directories, text, illustrations, index maps, legends, or other information associated with the principal map. This map shows features such as drainage, cities and other human settlements, territorial, administrative and colonial boundaries, shoreline features, and more. Relief shown by hachures.This layer is part of a selection of digitally scanned and georeferenced historic maps from the Harvard Map Collection. These maps typically portray both natural and manmade features. The selection represents a range of originators, ground condition dates, scales, and map purposes.

As paisagens dunares são sistemas de elevado dinamismo, devido à proximidade do mar e à extrema mobilidade do substrato arenoso e prevê-se que venham a ser severamente afetadas pelas alterações ambientais globais. As dunas são depósitos de areia criados por processos eólicos e apresentam uma vegetação muito característica. Estes depósitos de areia em conjunto com a vegetação formam uma barreira essencial ao avanço do mar durante as marés altas de águas vivas e tempestades. Em Portugal, a degradação dos ecossistemas costeiros é muito preocupante. O problema das espécies exóticas invasoras agravou-se, aumentando a pressão sobre as plantas nativas. Embora este problema não seja o único motivo da degradação dos ecossistemas costeiros, este trabalho pretende divulgar as plantas dunares da zona costeira de Matosinhos, sensibilizar para a proteção e conservação das dunas e alertar para o facto de diversas plantas invasoras rapidamente colonizarem espaços abertos, pondo em causa e estabilidade dos ecossistemas costeiros. O conhecimento detalhado destes ecossistemas permitirá a aplicação de processos de vigilância e monitorização bem como o restauro ecológico de áreas dunares degradadas.

Esta investigação visa o estudo e enquadramento em termos históricos, tipológicos e artísticos das pinturas de Bento Coelho da Silveira (1620-1708), expostas na igreja de S. Bartolomeu da Charneca em Lisboa. Uma vez que sabemos que não foram realizadas especificamente para a igreja de S. Bartolomeu da Charneca em Lisboa onde, na atualidade se encontram, procurar-se-á averiguar qual a origem destas obras; quem foi o seu encomendador e ainda aferir a pertinência e a coerência do programa iconográfico que veiculam. Caracterizar-se-á a pintura deste artista barroco, em concreto as nove telas de temática mariana que se encontram na igreja supracitada. É ainda objeto deste trabalho a comparação com outras obras do pintor, analisando o cânone, os atributos, os adereços e as composições, com particular relevo nas atitudes e nas características expressivas, bem como no estado de alma dos personagens todos eles evidenciando uma atitude melancólica. Pretende-se, finalmente, tecer considerações acerca do seu estado de conservação e, eventualmente, efetuar propostas de conservação e restauro, caso seja necessário.

Com a presente dissertação temos por objectivo problematizar, através da iconografia, as representações de suicidas míticos na cerâmica grega (séculos VII-IV a.C.). O corpus da pesquisa é constituído por 40 vasos com imagens de onze figuras mitológicas: Ájax, Alceste, Antígona, Ariadne, Cassandra, Fedra, Ifigénia, Macária, Medeia, Orestes e Políxena. Das 40 imagens analisadas, três são do séc. VII a.C., cinco do séc. VI a.C., onze do séc. V a.C. e 21 do séc. IV a.C. A dissertação é dividida em onze capítulos, um para cada figura mitológica, nos quais debateremos as fontes, a evolução do mito e a iconografia associada.
