The layer-by-layer (LbL) deposition method was used to build up alternating layers (five) of different polyelectrolyte solutions (alginate, zein-carvacrol nanocapsules, chitosan and chitosan-carvacrol emulsions) on an aminolysed/charged polyethylene terephthalate (A/C PET) film. These nanolaminated films were characterised by contact angle measurements and through the determination of water vapour (WVTR) and oxygen (O2TR) transmission rates. The effect of active nanolaminated films against the Alternaria sp. and Rhizopus stolonifer was also evaluated. This procedure allowed developing optically transparent nanolaminated films with tuneable water vapour and gas properties and antifungal activity. The water and oxygen transmission rate values for the multilayer films were lower than those previously reported for the neat alginate or chitosan films. The presence of carvacrol and zein nanocapsules significantly decreased the water transmission rate (up to 40 %) of the nanolaminated films. However, the O2TR behaved differently and was only improved (up to 45 %) when carvacrol was encapsulated, i.e. nanolaminated films prepared by alternating alginate with nanocapsules of zein-carvacrol layers showed better oxygen barrier properties than those prepared as an emulsion of chitosan and carvacrol. These films containing zein-carvacrol nanocapsules also showed the highest antifungal activity (30 %), which did not significantly differ from those obtained with the highest amount of carvacrol, probably due to the controlled release of the active agent (carvacrol) from the zein-carvacrol nanocapsules. Thus, this work shows that nanolaminated films prepared with alternating layers of alginate and zein-carvacrol nanocapsules can be considered to improve the shelf-life of foodstuffs.

Este artigo relata uma intervenção pedagógica levada a cabo junto de alunos do 4º ano de escolaridade do Ensino Básico, em Portugal, com dois objetivos: (1) desenvolver competências literácitas, especificamente ao nível da leitura e compreensão leitora; e (2) construir o conhecimento histórico, recorrendo a obras de literatura infanto-juvenil que recriam eventos históricos. O referencial teórico baseou-se quer no conceito de Content Area Literacy (MOSS, 2005; BROZO, 2010), quer no Programa de Leitura Fundamentado na Literatura (YOOP; YOOP, 2006).

Muitos textos da literatura infantil, pela sua relação com propósitos formativos, têm privilegiado determinados pontos de vista fortemente compatíveis com as comunidades socioculturais e com os valores ideológicos aí dominantes. Tal facto não impediu, porém, que alguns textos, para além da afirmação de determinados mundos possíveis, contenham igualmente uma não negligenciável dimensão emancipatória. O artigo apresenta e analisa seis álbuns narrativos para a infância, publicados no espaço ibérico, alguns dos quais traduções de relevantes obras estrangeiras (A princesa espertalhona, Babette Cole, 2004; O livro dos porquinhos, Anthony Browne, 2006), onde se interrogam os modos convencionais de retratar o género, possibilitando ao leitor pensar outros modos de considerar a construção social e cultural do género, afastados de uma concepção patriarcal e tradicional. Bacteriophages (phages), natural enemies of bacteria, can encode enzymes able to degrade polymeric substances. These substances can be found in the bacterial cell surface, such as polysaccharides, or are produced by bacteria when they are living in biofilm communities, the most common bacterial lifestyle. Consequently, phages with depolymerase activity have a facilitated access to the host receptors, by degrading the capsular polysaccharides, and are believed to have a better performance against bacterial biofilms, since the degradation of extracellular polymeric substances by depolymerases might facilitate the access of phages to the cells within different biofilm layers. Since the diversity of phage depolymerases is not yet fully explored, this is the first review gathering information about all the depolymerases encoded by fully sequenced phages. Overall, in this study, 160 putative depolymerases, including sialidases, levanases, xylosidases, dextranases, hyaluronidases, peptidases as well as pectate/pectin lyases, were found in 143 phages (43 Myoviridae, 47 Siphoviridae, 37 Podoviridae, and 16 unclassified) infecting 24 genera of bacteria. We further provide information about the main applications of phage depolymerases, which can comprise areas as diverse as medical, chemical, or food-processing industry.

Here, we evaluate the diagnostic and prognostic role of liquid-based cytology (LBC) in different body lesions, including thyroid, lung, effusions and malignant breast lesions. LBC has gained consensus after being applied to both non-gynecologic and fine-needle aspiration cytology. Although some remain sceptical regarding the diagnostic efficacy of LBC, mainly when used alone, in recent years, good results have been obtained as long as it showed a high diagnostic accuracy. Here, we discuss the additional possibility of storing material for the application of ancillary techniques (immunocytochemistry–molecular analysis) with several diagnostic and prognostic advantages, which may pave the way for the challenging evaluation of both monitoring responses to treatment and resistance to targeted therapies in thyroid, lung, breast carcinoma or malignant effusions. Furthermore, it provides the use of several molecular spots as specific targets for personalized therapy.

Objetivos : Constatando que a depressão é comum em idosos institucionalizados, associando-se à solidão, à ansiedade e à afetividade, pretendemos descrever a evolução da depressão durante dois anos e verificar que fatores se associam a essa evolução. Métodos : Em um estudo de coorte prospectivo em dois momentos (2011 e 2013), avaliamos 83 idosos institucionalizados, com idade no primeiro momento entre os 60 e os 100 anos, sendo 79,5% mulheres, 86,7% sem companheiro(a), e 72,3% com algum grau de escolaridade. Usamos a Escala Geriátrica da Depressão (GDS), a Escala de Solidão (UCLA-L), o Inventário Geriátrico de Ansiedade (GAI) e a Lista de Afetos Positivos e Negativos (PANAS). Resultados: Verificamos que 59,0% mantiveram a depressão e 10,8% desenvolveram depressão. Os idosos com depressão tiveram significativamente piores resultados na UCLA, GAI e PANAS, e os não depressivos tiveram afetos positivos mais altos. Quanto à evolução da depressão, os idosos que mantiveram depressão tiveram inicialmente pontuações elevadas no GDS, GAI, UCLA e na subescala PANAS negativo e pontuações baixas na subescala PANAS positivo. Esses idosos apresentaram associadamente um agravamento dos sentimentos de solidão, dos sintomas ansiosos e do afeto negativo ao longo dos dois anos. Os que desenvolveram depressão tiveram, no primeiro momento, pontuações elevadas na UCLA. Conclusões: Os sintomas de depressão com ou sem solidão no momento inicial, o agravamento da solidão, a ansiedade, o afeto negativo e o baixo afeto positivo poderão ser fatores de risco para a manutenção da depressão. A solidão poderá ainda ser um fator de risco para o desenvolvimento de depressão.

Activation of the dorsomedial nucleus of the hypothalamus (DMH) by galanin (GAL) induces behavioural hyperalgesia. Since DMH neurones do not project directly to the spinal cord, we hypothesized that the medullary dorsal reticular nucleus (DRt), a pronociceptive region projecting to the spinal dorsal horn (SDH) and/or the serotoninergic raphe-spinal pathway acting on the spinal 5-HT3 receptor (5HT3R) could relay descending nociceptive facilitation induced by GAL in the DMH. Heat-evoked paw-withdrawal latency (PWL) and activity of SDH neurones were assessed in monoarthritic (ARTH) and control (SHAM) animals after pharmacological manipulations of the DMH, DRt and spinal cord. The results showed that GAL in the DMH and glutamate in the DRt lead to behavioural hyperalgesia in both SHAM and ARTH animals, which is accompanied particularly by an increase in heat-evoked responses of wide-dynamic range neurons, a group of nociceptive SDH neurones. Facilitation of pain behaviour induced by GAL in the DMH was reversed by lidocaine in the DRt and by ondansetron, a 5HT3R antagonist, in the spinal cord. However, the hyperalgesia induced by glutamate in the DRt was not blocked by spinal ondansetron. In addition, in ARTH but not SHAM animals PWL was increased after lidocaine in the DRt and ondansetron in the spinal cord. Our data demonstrate that GAL in the DMH activates two independent descending facilitatory pathways: (i) one relays in the DRt and (ii) the other one involves 5-HT neurones acting on spinal 5HT3Rs. In experimental ARTH, the tonic pain-facilitatory action is increased in both of these descending pathways.

OBJETIVO: Comparar a mortalidade por doenças do coração e pela doença coronária (DC) nas faixas etárias dos 45 aos 64 anos, no período 1984-87, em capitais brasileiras com outros países. MÉTODOS: Foram utilizadas as estatísticas oficiais de mortalidade de 8 capitais com boa qualidade de informação: Belém, Recife, Salvador, Belo Horizonte, Rio de Janeiro, São Paulo, Curitiba e Porto Alegre e analisadas as doenças do coração [DC (410-414), mais insuficiência cardíaca (428), mais hipertensão (401-404)] e, isoladamente, a DC. A população foi a determinada nos Censos Demográficos de 1980 e 1991. RESULTADOS: A comparação com outros países mostrou que: a) as taxas de mortalidade por doenças do coração situam algumas cidades brasileiras na posição intermediária (Hungria, Rio de Janeiro, Finlândia, Porto Alegre e Polônia) e entre as mulheres no topo (Rio de Janeiro, Curitiba, Hungria, Porto Alegre, Inglaterra e Gales); b) DC apresentou o mesmo padrão (intermediário alto) das doenças do coração entre os homens (Finlândia, Hungria, Inglaterra e Gales, Porto Alegre e Rio de Janeiro). Entre as mulheres, repetiu-se a situação das doenças do coração (Rio de Janeiro, Curitiba, Hungria, Porto Alegre, Inglaterra e Gales). CONCLUSÃO: A comparação com outros países das taxas de mortalidade ajustadas por idade na faixa etária dos 45-64 anos no período 1984-87 mostrou que as cidades brasileiras estudadas têm altas taxas de mortalidade para as doenças do coração, principalmente entre as mulheres, em valores tão ou mais elevados do que os da Europa e dos Estados Unidos.

The metabolism of methanogenic archaea is inhibited by 2-bromoethanesulfonate (BES). Methane production is blocked because BES is an analog of methyl-coenzyme M and competes with this key molecule in the last step of methanogenesis. For this reason, BES is commonly used in several studies to avoid growth of acetoclastic and hydrogenotrophic methanogens [1]. Despite its effectiveness as methanogenic inhibitor, BES was found to alter microbial communities’ structure, to inhibit the metabolism of non-methanogenic microorganisms and to stimulate homoacetogenic metabolism [2,3]. Even though sulfonates have been reported as electron acceptors for sulfate- and sulfite-reducing bacteria (SRB), only one study described the reduction of BES by complex microbial communities [4]. In this work, a sulfate-reducing bacterium belonging to Desulfovibrio genus (98 % identity at the 16S rRNA gene level with Desulfovibrio aminophilus) was isolated from anaerobic sludge after several successive transfers in anaerobic medium containing BES as sole substrate. Sulfate was not supplemented to the anaerobic growth medium. This microorganism was able to grow under the following conditions: on BES plus H2/CO2 in bicarbonate buffered medium; on BES without H2/CO2 in bicarbonate buffered medium; and on BES in phosphate buffered medium. The main products of BES utilization were sulfide and acetate, the former was produced by the reduction of sulfur from the sulfonate moiety of BES and the latter likely originated from the carbon backbone of the BES molecule. BES was found, in this study, to represent not only an alternative electron acceptor but also to serve as electron donor, and sole carbon and energy source, supporting growth of a Desulfovibrio sp. obtained in pure culture. This is the first study that reports growth of SRB with BES as electron donor and electron acceptor, showing that the methanogenic inhibitor is a substrate for anaerobic growth.

[Excerpt] Under anaerobic conditions long chain fatty acids (LCFA) can be converted to methane by syntrophic bacteria and methanogenic archaea. LCFA degradation was also reported in the presence of alternative hydrogenotrophic partners, such as sulfate-reducing bacteria (SRB) and iron-reducing bacteria (IRB), which generally show higher affinity for H2 than methanogens and are more resistant to LCFA [1,2,3]. Their presence in a microbial culture degrading LCFA can be advantageous to reduce LCFA toxicity towards methanogens, although high concentrations of external electron acceptor (EEA) can lead to outcompetition of methanogens and cease methane production. In this work, we tested the effect of adding sub-stoichiometric concentrations of sulfate and iron(III) to methanogenic communities degrading LCFA. (...)
