This dissertation: 1) determines the factor(s) responsible for spawning induction in NematosteJla vectensis; 2) isolates, describes, and documents the source of jelly from egg masses of N. vectensis; and 3) describes N. vectensis' early development. Namatostella vectensis were maintained on a 7-day mussel feeding/water change regime over 159 days. Within 36 hours of mussel feeding/water change. 69.1% of females and 78.5% of males spawned reliably. Through manipulation of feeding, water change, oxygen and nitrogenous waste concentrations, spawning induction was found to be triggered by the oxygen concentration associated with water change, and not by feeding. Ammonia, anemones' major waste product, inhibited this induction in a concentration-dependent manner. Female N. vectensis release eggs in a persistent jellied egg mass which is unique among the Actiniaria. The major component of this egg mass jelly was a positive periodic acid-Schiffs staining, 39.5-40.5 kD glycoprotein. Antibodies developed in rabbits against this glycoprotein bound to jelly of intact egg masses and to granules (~ 2.8 IJm in diameter) present in female anemone mesenteries and their associated filaments. Antibodies did not label male tissues. Nematostella vecfensis embryos underwent first karyokinesis -60 minutes following the addition of sperm to eggs. Second nuclear division took place, followed by first cleavage, 90-120 minutes later. Each of the 4 blastomeres that resulted from first cleavage contained a single nucleus. Arrangement of these blastomeres ranged from radial to pseudospiral. Embryonic development was both asynchronous and holoblastic. Following formation of the 4-cell stage, 71% of embryos proceeded to cleave again to form an 8-cell stage. In each of the remaining 29% of embryos, a fusion of from 2-4 blastomeres resulted in 4 possible patterns which had no affect on either cleavage interval timing or subsequent development. The fusion event was not due to ooplasmic segregation. Blastomeres isolated from 4-celled embryos were regulative and developed into normal planula larvae and juvenile anemones that were 1/4 the size of those that developed from intact 4-celled embryos. Embryos exhibiting the fusion phenomenon were examined at the fine structural level. The fusion phenomenon resulted in formation of a secondary syncytium and was not a mere compaction of blastomeres.

This report presents oceanographic data supporting the detailed chemical studies in the VERTEX Particle Interceptor Trap (PIT) experiment off the central California coast. Prior to the deployment of the PITs, an oceanographic survey of the intended study area was made on R/V CAYUSE from 17 to 21 August 1980. During this cruise, twenty CTD stations (Fig. 1) were occupied in a grid centered about the PIT site selected earlier based on archival oceanographic data. During the second leg of the VERTEX experiment from 25 August to 3 September, CTO profiles were taken as time permitted. In addition, a short survey near the PITs was made on 2 September. The intent of the pre-deployment cruise was to obtain data characterizing the vertical and horizontal variability of physical and chemical properties and to map the geostrophic flow field. Toward this end, vertical profiles of salinity, temperature and dissolved oxygen were made using a Plessey 9040 CTOa profiler. The status of the gulf menhaden, Brevoortia patronus, fishery was assessed with purseseine landing data from 1946 to 1992 and port sampling data from 1964 to 1992. These data were analyzed to determine growth rates, biological reference points for fishing mortality from yield per recruit and maximum spawning potential analyses, spawner-recruit relationships, and maximum sustainable yield (MSY). Virtual population approaches were used to obtain point estimates of stock size, recruits to age I, spawning stock size, and fishing mortality rates. Exploitation rates ranged between 14% and 45% for age-1 fish, between 30% and 72% for age-2 fish, and between 36% and 71% for age-3 fish. Biological reference points from yield per recruit (FO. I: 0.7-0.9 yr-1) and maximum spawning potential (F20: 1.62.9 yr-l and F30: 1.0-2.1 yr-1) were obtained for comparison with recent estimates of F (0.4-0.8 yr-l). Parameters from Ricker-type spawner-recruit relations were estimated, although considerable unexplained variability remained. Estimates of long-term MSY from fits of the generalized production model ranged between 664,000 metric tons (t) and 897,000 t. Declines in landings since 1988 have raised concerns about the status of the gulf menhaden stock. However, gulf menhaden are short lived and highly fecund. Thus, variation in recruitment to age 1 largely mediated by environmental conditions influences fishing success over the next two years (as age-1 and age-2 fish). Comparisons of recent estimates of fishing mortality to biological reference points do not suggest overfishing. (PDF file contains 26 pages.)

Há 25 anos, era promulgada a Lei nº 8.069, de 1990, que instituiu o Estatuto da Criança e do Adolescente (ECA). A lei, inovadora em diversos aspectos, constitui importante marco jurídico, por criar mecanismos que possibilitam a efetivação dos direitos fundamentais de crianças e adolescentes conferidos pela Constituição Cidadã.

The role of Na+ fluxes through voltage-gated sodium channels in the regulation of sperm cell function remains poorly understood. Previously, we reported that several genes encoding voltage-gated Na+ channels were expressed in human testis and mature spermatozoa. In this study, we analyzed the presence and function of the TTX-resistant VGSC a subunit Na(v)1.8 in human capacitated sperm cells. Using an RT-PCR assay, we found that the mRNA of the gene SCN10A, that encode Na-v1.8, was abundantly and specifically expressed in human testis and ejaculated spermatozoa. The Na-v1.8 protein was detected in capacitated sperm cells using three different specific antibodies against this channel. Positive immunoreactivity was mainly located in the neck and the principal piece of the flagellum. The presence of Na-v1.8 in sperm cells was confirmed by Western blot. Functional studies demonstrated that the increases in progressive motility produced by veratridine, a voltage-gated sodium channel activator, were reduced in sperm cells preincubated with TTX (10 mu M), the Na-v1.8 antagonist A-803467, or a specific Na-v1.8 antibody. Veratridine elicited similar percentage increases in progressive motility in sperm cells maintained in Ca2+-containing or Ca2+-free solution and did not induce hyperactivation or the acrosome reaction. Veratridine caused a rise in sperm intracellular Na+, [Na+](i), and the sustained phase of the response was inhibited in the presence of A-803467. These results verify that the Na+ channel Na-v1.8 is present in human sperm cells and demonstrate that this channel participates in the regulation of sperm function.

This report contains the frrst observations made for the Modis Optical Characterization Experiment (MOCE). Data presented here were obtained on the R/V DeSteiguer between 28 August and 8 October along the central California coast and in Monterey Bay. Three types of data are reported here: high spectral resolution radiometry at three depths for seven stations; salinity, temperature, fluorescence and beam attenuation profiles at the same stations; and total suspended matter and suspended organic carbon and nitrogen.
