Visual correspondence is a key computer vision task that aims at identifying projections of the same 3D point into images taken either from different viewpoints or at different time instances. This task has been the subject of intense research activities in the last years in scenarios such as object recognition, motion detection, stereo vision, pattern matching, image registration. The approaches proposed in literature typically aim at improving the state of the art by increasing the reliability, the accuracy or the computational efficiency of visual correspondence algorithms. The research work carried out during the Ph.D. course and presented in this dissertation deals with three specific visual correspondence problems: fast pattern matching, stereo correspondence and robust image matching. The dissertation presents original contributions to the theory of visual correspondence, as well as applications dealing with 3D reconstruction and multi-view video surveillance.

The quality of astronomical sites is the first step to be considered to have the best performances from the telescopes. In particular, the efficiency of large telescopes in UV, IR, radio etc. is critically dependent on atmospheric transparency. It is well known that the random optical effects induced on the light propagation by turbulent atmosphere also limit telescope’s performances. Nowadays, clear appears the importance to correlate the main atmospheric physical parameters with the optical quality reachable by large aperture telescopes. The sky quality evaluation improved with the introduction of new techniques, new instrumentations and with the understanding of the link between the meteorological (or synoptical parameters and the observational conditions thanks to the application of the theories of electromagnetic waves propagation in turbulent medias: what we actually call astroclimatology. At the present the site campaigns are evolved and are performed using the classical scheme of optical seeing properties, meteorological parameters, sky transparency, sky darkness and cloudiness. New concept are added and are related to the geophysical properties such as seismicity, microseismicity, local variability of the climate, atmospheric conditions related to the ground optical turbulence and ground wind regimes, aerosol presence, use of satellite data. The purpose of this project is to provide reliable methods to analyze the atmospheric properties that affect ground-based optical astronomical observations and to correlate them with the main atmospheric parameters generating turbulence and affecting the photometric accuracy. The first part of the research concerns the analysis and interpretation of longand short-time scale meteorological data at two of the most important astronomical sites located in very different environments: the Paranal Observatory in the Atacama Desert (Chile), and the Observatorio del Roque de Los Muchachos(ORM) located in La Palma (Canary Islands, Spain). The optical properties of airborne dust at ORM have been investigated collecting outdoor data using a ground-based dust monitor. Because of its dryness, Paranal is a suitable observatory for near-IR observations, thus the extinction properties in the spectral range 1.00-2.30 um have been investigated using an empirical method. Furthermore, this PhD research has been developed using several turbulence profilers in the selection of the site for the European Extremely Large Telescope(E-ELT). During the campaigns the properties of the turbulence at different heights at Paranal and in the sites located in northern Chile and Argentina have been studied. This given the possibility to characterize the surface layer turbulence at Paranal and its connection with local meteorological conditions.

The purpose of this Thesis is to develop a robust and powerful method to classify galaxies from large surveys, in order to establish and confirm the connections between the principal observational parameters of the galaxies (spectral features, colours, morphological indices), and help unveil the evolution of these parameters from $z \sim 1$ to the local Universe. Within the framework of zCOSMOS-bright survey, and making use of its large database of objects ($\sim 10\,000$ galaxies in the redshift range $0 < z \lesssim 1.2$) and its great reliability in redshift and spectral properties determinations, first we adopt and extend the \emph{classification cube method}, as developed by Mignoli et al. (2009), to exploit the bimodal properties of galaxies (spectral, photometric and morphologic) separately, and then combining together these three subclassifications. We use this classification method as a test for a newly devised statistical classification, based on Principal Component Analysis and Unsupervised Fuzzy Partition clustering method (PCA+UFP), which is able to define the galaxy population exploiting their natural global bimodality, considering simultaneously up to 8 different properties. The PCA+UFP analysis is a very powerful and robust tool to probe the nature and the evolution of galaxies in a survey. It allows to define with less uncertainties the classification of galaxies, adding the flexibility to be adapted to different parameters: being a fuzzy classification it avoids the problems due to a hard classification, such as the classification cube presented in the first part of the article. The PCA+UFP method can be easily applied to different datasets: it does not rely on the nature of the data and for this reason it can be successfully employed with others observables (magnitudes, colours) or derived properties (masses, luminosities, SFRs, etc.). The agreement between the two classification cluster definitions is very high. ``Early'' and ``late'' type galaxies are well defined by the spectral, photometric and morphological properties, both considering them in a separate way and then combining the classifications (classification cube) and treating them as a whole (PCA+UFP cluster analysis). Differences arise in the definition of outliers: the classification cube is much more sensitive to single measurement errors or misclassifications in one property than the PCA+UFP cluster analysis, in which errors are ``averaged out'' during the process. This method allowed us to behold the \emph{downsizing} effect taking place in the PC spaces: the migration between the blue cloud towards the red clump happens at higher redshifts for galaxies of larger mass. The determination of $M_{\mathrm{cross}}$ the transition mass is in significant agreement with others values in literature.

This PhD Thesis is part of a long-term wide research project, carried out by the "Osservatorio Astronomico di Bologna (INAF-OABO)", that has as primary goal the comprehension and reconstruction of formation mechanism of galaxies and their evolution history. There is now substantial evidence, both from theoretical and observational point of view, in favor of the hypothesis that the halo of our Galaxy has been at least partially, built up by the progressive accretion of small fragments, similar in nature to the present day dwarf galaxies of the Local Group. In this context, the photometric and spectroscopic study of systems which populate the halo of our Galaxy (i.e. dwarf spheroidal galaxy, tidal streams, massive globular cluster, etc) permits to discover, not only the origin and behaviour of these systems, but also the structure of our Galactic halo, combined with its formation history. In fact, the study of the population of these objects and also of their chemical compositions, age, metallicities and velocity dispersion, permit us not only an improvement in the understanding of the mechanisms that govern the Galactic formation, but also a valid indirect test for cosmological model itself. Specifically, in this Thesis we provided a complete characterization of the tidal Stream of the Sagittarius dwarf spheroidal galaxy, that is the most striking example of the process of tidal disruption and accretion of a dwarf satellite in to our Galaxy. Using Red Clump stars, extracted from the catalogue of the Sloan Digital Sky Survey (SDSS) we obtained an estimate of the distance, the depth along the line of sight and of the number density for each detected portion of the Stream (and more in general for each detected structure along our line of sight). Moreover comparing the relative number (i.e. the ratio) of Blue Horizontal Branch stars and Red Clump stars (the two features are tracers of different age/different metallicity populations) in the main body of the galaxy and in the Stream, in order to verify the presence of an age-metallicity gradient along the Stream. We also report the detection of a population of Red Clump stars probably associated with the recently discovered Bootes III stellar system. Finally, we also present the results of a survey of radial velocities over a wide region, extending from r ~ 10' out to r ~ 80' within the massive star cluster Omega Centauri. The survey was performed with FLAMES@VLT, to study the velocity dispersion profile in the outer regions of this stellar system. All the results presented in this Thesis, have already been published in refeered journals.

In dieser Arbeit wurden durch Verwendung eines stereodifferenzierenden Kohlenhydrat-Auxiliars chirale Stickstoffheterocyclen und enantiomerenreine Piperidin-Alkaloide synthetisiert. Alkaloide mit einer Piperidin-Grundstruktur sind in der Natur weit verbreitet und weisen vielfältige biologische Aktivitäten auf. Zusammen mit synthetischen Derivaten sind sie daher von großem Interesse für die Wirkstoffforschung. Mit dem aus D-Arabinose zugänglichen 2,3,4-Tri-O-pivaloyl-D-arabinosylamin wurden mit hoher Stereoselektivität N-Glycosyl-dehydropiperidinone aufgebaut, die vielfältig modifizierbare Ausgangsverbindungen zur Synthese unterschiedlich substituierter Stickstoffheterocyclen darstellen. In einer Vielzahl vor allem metallorganischer Reaktionen waren regio- und stereoselektive Derivatisierungen an allen Positionen der N-glycosidisch gebundenen Dehydropiperidinone möglich. Durchgeführt wurden z. B. die Addition aktivierter Cuprate, elektrophile Substitutionen, Reduktionen, Iod-Magnesium-Austausch sowie palladium- und kupferkatalysierte Kupplungen. Die Kombination dieser Methoden führte zu mehrfach substituierten Piperidinen. In einer Ringschlussmetathese wurde zudem ein Zugang zu bicyclischen Heterocyclen geschaffen. Das Kohlenhydrat-Auxiliar steuert den stereochemischen Verlauf der Bildung der Dehydropiperidinone und der daran durchgeführten Funktionalisierungen. Die Konfigurationen der neu gebildeten Stereozentren wurden mittels Röntgenstrukturanalysen und NMR-Spektroskopie sowie durch die Überführung der Piperidin-Derivate in Alkaloide mit bekanntem Drehwert ermittelt. Die Stickstoffheterocyclen können nach Entfernen der Enamin-Doppelbindung durch milde Acidolyse vom Kohlenhydrat-Auxiliar abgespalten werden, wodurch man die enantiomerenreinen Alkaloide erhält.

The Gaia space mission is a major project for the European astronomical community. As challenging as it is, the processing and analysis of the huge data-flow incoming from Gaia is the subject of thorough study and preparatory work by the DPAC (Data Processing and Analysis Consortium), in charge of all aspects of the Gaia data reduction. This PhD Thesis was carried out in the framework of the DPAC, within the team based in Bologna. The task of the Bologna team is to define the calibration model and to build a grid of spectro-photometric standard stars (SPSS) suitable for the absolute flux calibration of the Gaia G-band photometry and the BP/RP spectrophotometry. Such a flux calibration can be performed by repeatedly observing each SPSS during the life-time of the Gaia mission and by comparing the observed Gaia spectra to the spectra obtained by our ground-based observations. Due to both the different observing sites involved and the huge amount of frames expected (≃100000), it is essential to maintain the maximum homogeneity in data quality, acquisition and treatment, and a particular care has to be used to test the capabilities of each telescope/instrument combination (through the “instrument familiarization plan”), to devise methods to keep under control, and eventually to correct for, the typical instrumental effects that can affect the high precision required for the Gaia SPSS grid (a few % with respect to Vega). I contributed to the ground-based survey of Gaia SPSS in many respects: with the observations, the instrument familiarization plan, the data reduction and analysis activities (both photometry and spectroscopy), and to the maintenance of the data archives. However, the field I was personally responsible for was photometry and in particular relative photometry for the production of short-term light curves. In this context I defined and tested a semi-automated pipeline which allows for the pre-reduction of imaging SPSS data and the production of aperture photometry catalogues ready to be used for further analysis. A series of semi-automated quality control criteria are included in the pipeline at various levels, from pre-reduction, to aperture photometry, to light curves production and analysis.

From the discoveries of Pasteur, stereochemistry has played an increasingly important role in the chemical sciences. In particular conformational study of molecules with axial chirality is object of intense research. Through Dynamic-NMR analysis and simulation of the spectra, the energy rotational barriers value of conformers are obtained. When this barrier is high sufficiently, atropisomeric stable compounds can be reached. They can be separated and used in stereo-synthesis and in packing processes. 3,4-bis-aryl maleimides, in which the aromatic groups are sufficiently bulky, generate atropisomeric stable configurations, that can be isolated at room temperature. The assignment of absolute configurations is performed through ECD analysis and comparison with computational calculations. The biological activities of maleimide derivatives widen the field of atropisomers application also in biological systems.

This Thesis focuses on the X-ray study of the inner regions of Active Galactic Nuclei, in particular on the formation of high velocity winds by the accretion disk itself. Constraining AGN winds physical parameters is of paramount importance both for understanding the physics of the accretion/ejection flow onto supermassive black holes, and for quantifying the amount of feedback between the SMBH and its environment across the cosmic time. The sources selected for the present study are BAL, mini-BAL, and NAL QSOs, known to host high-velocity winds associated to the AGN nuclear regions. Observationally, a three-fold strategy has been adopted: - substantial samples of distant sources have been analyzed through spectral, photometric, and statistical techniques, to gain insights into their mean properties as a population; - a moderately sized sample of bright sources has been studied through detailed X-ray spectral analysis, to give a first flavor of the general spectral properties of these sources, also from a temporally resolved point of view; - the best nearby candidate has been thoroughly studied using the most sophisticated spectral analysis techniques applied to a large dataset with a high S/N ratio, to understand the details of the physics of its accretion/ejection flow. There are three main channels through which this Thesis has been developed: - [Archival Studies]: the XMM-Newton public archival data has been extensively used to analyze both a large sample of distant BAL QSOs, and several individual bright sources, either BAL, mini-BAL, or NAL QSOs. - [New Observational Campaign]: I proposed and was awarded with new X-ray pointings of the mini-BAL QSOs PG 1126-041 and PG 1351+640 during the XMM-Newton AO-7 and AO-8. These produced the biggest X-ray observational campaign ever made on a mini-BAL QSO (PG 1126-041), including the longest exposure so far. Thanks to the exceptional dataset, a whealth of informations have been obtained on both the intrinsic continuum and on the complex reprocessing media that happen to be in the inner regions of this AGN. Furthermore, the temporally resolved X-ray spectral analysis field has been finally opened for mini-BAL QSOs. - [Theoretical Studies]: some issues about the connection between theories and observations of AGN accretion disk winds have been investigated, through theoretical arguments and synthetic absorption line profiles studies.

Kurzzusammenfassung Elaeocarpacae-Alkaloide: flexible Synthesen optisch aktiver (-) Elaeokanin C Schlüsselbausteine Im Tier- und Pflanzenreich sind Alkaloide weit verbreitet und werden von der Biogenese her als Produkte des Aminosäure-Stoffwechsels angesehen. Die Elaeocarpacae-Alkaloide zählen zu den Indolizidinen, welche durch ein Azabicyclo-[4.3.0]-nonan Grundgerüst charakterisiert sind und erstmals Ende der 60er Jahre des letzten Jahrhunderts aus den Blättern der in Neu Guinea beheimateten Ölbaumgewächse isoliert wurden. Für verschiedene Vertreter dieses Alkaloid-Typs wurden sowohl racemische als auch asymmetrische Totalsynthesen entwickelt. Während für das (+) Elaeokanin C bereits Totalsynthesen existieren, gibt es für das (-) Elaeokanin C bis heute keine asymmetrische Synthese. Als Fernziel der vorliegenden Arbeit wurde die erste Totalsynthese von (-) Elaeokanin C ausgewählt. Der Syntheseplan sieht zunächst den diastereoselektiven Aufbau eines optisch aktiven Schlüsselbausteins mit Naturstoff-Stereotriade im Sinne einer konvergenten ex-chiral-pool Synthese vor. Im Rahmen dieser Arbeit konnte dies durch die Aza-Claisen-Umlagerung realisiert werden. Ausgehend von diesem Schlüsselbaustein wurden verschiedene Synthesewege verfolgt um sowohl das Substitutionsmuster der Seitenkette als auch das des Piperidinsegments vielfältig variieren zu können. Die Einführung der Seitenkette erwies sich durch vielfältige Nachbargruppeneffekte wie die unerwünschte 5-exo-trig Cyclisierung zu einem Pyrrolizidin Derivat als große Hürde. Eine geänderte Synthesestrategie mit einem schrittweisen Aufbau der Kette lieferte schließlich den Baustein, aus dem nun in wenigen Stufen das (-) Elaeokanin C sowie vielfältige Analoga herzustellen sein sollten.

Weak lensing experiments such as the future ESA-accepted mission Euclid aim to measure cosmological parameters with unprecedented accuracy. It is important to assess the precision that can be obtained in these measurements by applying analysis software on mock images that contain many sources of noise present in the real data. In this Thesis, we show a method to perform simulations of observations, that produce realistic images of the sky according to characteristics of the instrument and of the survey. We then use these images to test the performances of the Euclid mission. In particular, we concentrate on the precision of the photometric redshift measurements, which are key data to perform cosmic shear tomography. We calculate the fraction of the total observed sample that must be discarded to reach the required level of precision, that is equal to 0.05(1+z) for a galaxy with measured redshift z, with different ancillary ground-based observations. The results highlight the importance of u-band observations, especially to discriminate between low (z < 0.5) and high (z ~ 3) redshifts, and the need for good observing sites, with seeing FWHM < 1. arcsec. We then construct an optimal filter to detect galaxy clusters through photometric catalogues of galaxies, and we test it on the COSMOS field, obtaining 27 lensing-confirmed detections. Applying this algorithm on mock Euclid data, we verify the possibility to detect clusters with mass above 10^14.2 solar masses with a low rate of false detections.

Studio di un esercizio dell'entrata alle parallele asimmetriche in ginnastica artistica effettuato mediante le tecniche della biomeccanica, in particolare la rilevazione per mezzo della stereo-fotogrammetria e l'analisi dei dati attraverso SMART Tracker e SMART Analyzer allo scopo di introdurre un nuovo movimento di entrata alle parallele di maggiore difficoltà.

Our view of Globular Clusters has deeply changed in the last decade. Modern spectroscopic and photometric data have conclusively established that globulars are neither coeval nor monometallic, reopening the issue of the formation of such systems. Their formation is now schematized as a two-step process, during which the polluted matter from the more massive stars of a first generation gives birth, in the cluster innermost regions, to a second generation of stars with the characteristic signature of fully CNO-processed matter. To date, star-to-star variations in abundances of the light elements (C, N, O, Na) have been observed in stars of all evolutionary phases in all properly studied Galactic globular clusters. Multiple or broad evolutionary sequences have also been observed in nearly all the clusters that have been observed with good signal-to-noise in the appropriate photometric bands. The body of evidence suggests that spreads in light-element abundances can be fairly well traced by photometric indices including near ultraviolet passbands, as CNO abundance variations affect mainly wavelengths shorter than ~400 nm owing to the rise of some NH and CN molecular absorption bands. Here, we exploit this property of near ultraviolet photometry to trace internal chemical variations and combined it with low resolution spectroscopy aimed to derive carbon and nitrogen abundances in order to maximize the information on the multiple populations. This approach has been proven to be very effective in (i) detecting multiple population, (ii) characterizing their global properties (i.e., relative fraction of stars, location in the color-magnitude diagram, spatial distribution, and trends with cluster parameters) and (iii) precisely tagging their chemical properties (i.e., extension of the C-N anticorrelation, bimodalities in the N content).

Numerosi incidenti verificatisi negli ultimi dieci anni in campo chimico e petrolchimico sono dovuti all’innesco di sostanze infiammabili rilasciate accidentalmente: per questo motivo gli scenari incidentali legati ad incendi esterni rivestono oggigiorno un interesse crescente, in particolar modo nell’industria di processo, in quanto possono essere causa di ingenti danni sia ai lavoratori ed alla popolazione, sia alle strutture. Gli incendi, come mostrato da alcuni studi, sono uno dei più frequenti scenari incidentali nell’industria di processo, secondi solo alla perdita di contenimento di sostanze pericolose. Questi eventi primari possono, a loro volta, determinare eventi secondari, con conseguenze catastrofiche dovute alla propagazione delle fiamme ad apparecchiature e tubazioni non direttamente coinvolte nell’incidente primario; tale fenomeno prende il nome di effetto domino. La necessità di ridurre le probabilità di effetto domino rende la mitigazione delle conseguenze un aspetto fondamentale nella progettazione dell’impianto. A questo scopo si impiegano i materiali per la protezione passiva da fuoco (Passive Fire Protection o PFP); essi sono sistemi isolanti impiegati per proteggere efficacemente apparecchiature e tubazioni industriali da scenari di incendio esterno. L’applicazione dei materiali per PFP limita l’incremento di temperatura degli elementi protetti; questo scopo viene raggiunto tramite l’impiego di differenti tipologie di prodotti e materiali. Tuttavia l’applicazione dei suddetti materiali fireproofing non può prescindere da una caratterizzazione delle proprietà termiche, in particolar modo della conducibilità termica, in condizioni che simulino l’esposizione a fuoco. Nel presente elaborato di tesi si è scelto di analizzare tre materiali coibenti, tutti appartenenti, pur con diversità di composizione e struttura, alla classe dei materiali inorganici fibrosi: Fibercon Silica Needled Blanket 1200, Pyrogel®XT, Rockwool Marine Firebatt 100. I tre materiali sono costituiti da una fase solida inorganica, differente per ciascuno di essi e da una fase gassosa, preponderante come frazione volumetrica. I materiali inorganici fibrosi rivestono una notevole importanza rispetto ad altri materiali fireproofing in quanto possono resistere a temperature estremamente elevate, talvolta superiori a 1000 °C, senza particolari modifiche chimico-fisiche. Questo vantaggio, unito alla versatilità ed alla semplicità di applicazione, li rende leader a livello europeo nei materiali isolanti, con una fetta di mercato pari circa al 60%. Nonostante l’impiego dei suddetti materiali sia ormai una realtà consolidata nell’industria di processo, allo stato attuale sono disponibili pochi studi relativi alle loro proprietà termiche, in particolare in condizioni di fuoco. L’analisi sperimentale svolta ha consentito di identificare e modellare il comportamento termico di tali materiali in caso di esposizione a fuoco, impiegando nei test, a pressione atmosferica, un campo di temperatura compreso tra 20°C e 700°C, di interesse per applicazioni fireproofing. Per lo studio delle caratteristiche e la valutazione delle proprietà termiche dei tre materiali è stata impiegata principalmente la tecnica Transient Plane Source (TPS), che ha consentito la determinazione non solo della conducibilità termica, ma anche della diffusività termica e della capacità termica volumetrica, seppure con un grado di accuratezza inferiore. I test sono stati svolti su scala di laboratorio, creando un set-up sperimentale che integrasse opportunamente lo strumento Hot Disk Thermal Constants Analyzer TPS 1500 con una fornace a camera ed un sistema di acquisizione dati. Sono state realizzate alcune prove preliminari a temperatura ambiente sui tre materiali in esame, per individuare i parametri operativi (dimensione sensori, tempi di acquisizione, etc.) maggiormente idonei alla misura della conducibilità termica. Le informazioni acquisite sono state utilizzate per lo sviluppo di adeguati protocolli sperimentali e per effettuare prove ad alta temperatura. Ulteriori significative informazioni circa la morfologia, la porosità e la densità dei tre materiali sono state ottenute attraverso stereo-microscopia e picnometria a liquido. La porosità, o grado di vuoto, assume nei tre materiali un ruolo fondamentale, in quanto presenta valori compresi tra 85% e 95%, mentre la frazione solida ne costituisce la restante parte. Inoltre i risultati sperimentali hanno consentito di valutare, con prove a temperatura ambiente, l’isotropia rispetto alla trasmissione del calore per la classe di materiali coibenti analizzati, l’effetto della temperatura e della variazione del grado di vuoto (nel caso di materiali che durante l’applicazione possano essere soggetti a fenomeni di “schiacciamento”, ovvero riduzione del grado di vuoto) sulla conducibilità termica effettiva dei tre materiali analizzati. Analoghi risultati, seppure con grado di accuratezza lievemente inferiore, sono stati ottenuti per la diffusività termica e la capacità termica volumetrica. Poiché è nota la densità apparente di ciascun materiale si è scelto di calcolarne anche il calore specifico in funzione della temperatura, di cui si è proposto una correlazione empirica. I risultati sperimentali, concordi per i tre materiali in esame, hanno mostrato un incremento della conducibilità termica con la temperatura, da valori largamente inferiori a 0,1 W/(m∙K) a temperatura ambiente, fino a 0,3÷0,4 W/(m∙K) a 700°C. La sostanziale similitudine delle proprietà termiche tra i tre materiali, appartenenti alla medesima categoria di materiali isolanti, è stata riscontrata anche per la diffusività termica, la capacità termica volumetrica ed il calore specifico. Queste considerazioni hanno giustificato l’applicazione a tutti i tre materiali in esame dei medesimi modelli per descrivere la conducibilità termica effettiva, ritenuta, tra le proprietà fisiche determinate sperimentalmente, la più significativa nel caso di esposizione a fuoco. Lo sviluppo di un modello per la conducibilità termica effettiva si è reso necessario in quanto i risultati sperimentali ottenuti tramite la tecnica Transient Plane Source non forniscono alcuna informazione sui contributi offerti da ciascun meccanismo di scambio termico al termine complessivo e, pertanto, non consentono una facile generalizzazione della proprietà in funzione delle condizioni di impiego del materiale. La conducibilità termica dei materiali coibenti fibrosi e in generale dei materiali bi-fasici tiene infatti conto in un unico valore di vari contributi dipendenti dai diversi meccanismi di scambio termico presenti: conduzione nella fase gassosa e nel solido, irraggiamento nelle superfici delle cavità del solido e, talvolta, convezione; inoltre essa dipende fortemente dalla temperatura e dalla porosità. Pertanto, a partire dal confronto con i risultati sperimentali, tra cui densità e grado di vuoto, l’obiettivo centrale della seconda fase del progetto è stata la scelta, tra i numerosi modelli a disposizione in letteratura per materiali bi-fasici, di cui si è presentata una rassegna, dei più adatti a descrivere la conducibilità termica effettiva nei materiali in esame e nell’intervallo di temperatura di interesse, fornendo al contempo un significato fisico ai contributi apportati al termine complessivo. Inizialmente la scelta è ricaduta su cinque modelli, chiamati comunemente “modelli strutturali di base” (Serie, Parallelo, Maxwell-Eucken 1, Maxwell-Eucken 2, Effective Medium Theory) [1] per la loro semplicità e versatilità di applicazione. Tali modelli, puramente teorici, hanno mostrato al raffronto con i risultati sperimentali numerosi limiti, in particolar modo nella previsione del termine di irraggiamento, ovvero per temperature superiori a 400°C. Pertanto si è deciso di adottare un approccio semi-empirico: è stato applicato il modello di Krischer [2], ovvero una media pesata su un parametro empirico (f, da determinare) dei modelli Serie e Parallelo, precedentemente applicati. Anch’esso si è rivelato non idoneo alla descrizione dei materiali isolanti fibrosi in esame, per ragioni analoghe. Cercando di impiegare modelli caratterizzati da forte fondamento fisico e grado di complessità limitato, la scelta è caduta sui due recenti modelli, proposti rispettivamente da Karamanos, Papadopoulos, Anastasellos [3] e Daryabeigi, Cunnington, Knutson [4] [5]. Entrambi presentavano il vantaggio di essere stati utilizzati con successo per materiali isolanti fibrosi. Inizialmente i due modelli sono stati applicati con i valori dei parametri e le correlazioni proposte dagli Autori. Visti gli incoraggianti risultati, a questo primo approccio è seguita l’ottimizzazione dei parametri e l’applicazione di correlazioni maggiormente idonee ai materiali in esame, che ha mostrato l’efficacia dei modelli proposti da Karamanos, Papadopoulos, Anastasellos e Daryabeigi, Cunnington, Knutson per i tre materiali analizzati. Pertanto l’obiettivo finale del lavoro è stato raggiunto con successo in quanto sono stati applicati modelli di conducibilità termica con forte fondamento fisico e grado di complessità limitato che, con buon accordo ai risultati sperimentali ottenuti, consentono di ricavare equazioni predittive per la stima del comportamento, durante l’esposizione a fuoco, dei materiali fireproofing in esame. Weltweit existiert keine zum Tierversuch alternative Methode, um adsorbierte Pertussis-Impfstoffe auf restliche Toxin-Aktivität hin zu untersuchen. Der im Europäischen Arzneibuch vorgeschriebene Tierversuch besitzt nach Erfahrungen der Industrie, internationaler Prüfbehörden sowie des Paul-Ehrlich-Institutes eine schlechte Aussagekraft. Er ist wenig standardisierbar und weist häufig ein zweifelhaftes Ergebnis auf, so dass Wiederholungen und damit einhergehend ein hoher Verbrauch an Versuchstieren unumgänglich sind. Enthält der Impfstoff Reste von nicht-inaktiviertem Pertussis-Toxin (PTx), muss mit schweren und schwersten Nebenwirkungen bei den Impflingen gerechnet werden. In dieser Arbeit wurde ein In vitro-Nachweis für aktives PTx entwickelt. rnAngeregt durch Publikationen, wonach Pertussis-Toxin humane Monozyten aktiviert, wurde zunächst versucht, diesen Effekt zum Toxin-Nachweis auszunutzen. Die vorliegende Arbeit zeigt jedoch eindeutig, dass Pertussis-Toxin selbst nicht zur Stimulation humaner Monozyten führt. Vielmehr konnte nachgewiesen werden, dass die Aktivierung dieser Immunzellen auf Kontaminationen durch Lipopolysaccharide zurückzuführen ist. Damit wurden die Aussagen in den oben erwähnten Veröffentlichungen widerlegt. Dieses Ergebnis wurde bereits zur Publikation eingereicht.rnNunmehr wurden verschiedene Ansätze zum Nachweis von Pertussis-Toxin entwickelt, welche seine enzymatischen Aktivitäten als NAD-Glycohydrolase und ADP-Ribosyltransferase ausnutzen. Zunächst wurde versucht, die Hydrolyse von NAD zu ADP-Ribose und Nicotinamid photometrisch nachzuweisen. Wegen unbefriedigender Sensitivität wurde dieses Verfahren zu einem fluorometrischen Nachweis weiterentwickelt. Verwendet wurde hier fluorogenes etheno-NAD, welches von Pertussis-Toxin als Substrat akzeptiert wird. Letzteres Prinzip ist zum In vitro-Nachweis von Pertussis-Toxin geeignet, wird jedoch durch das in Impfstoffen häufig verwendete Adsorbens Aluminiumhydroxid gestört. Deshalb wurde dieser Ansatz aufgegeben und ein neuer Weg verfolgt, welcher am Energiestoffwechsel von humanen Zellen ansetzt. Eine Konsequenz des Angriffs von Pertussis-Toxin auf seine Zielzellen im Respirationstrakt besteht – nach komplexen Reaktionen des Signaltransduktionsweges – im Absenken des ATP-Gehaltes. Als menschliche Surrogat-Zellen wurden frisch isolierte periphere mononukleäre Zellen (PBMCs) sowie die permanente Lymphozyten-Zelllinie Jurkat eingesetzt und deren ATP-Gehalt mittels Luziferin-Luziferase-Lumineszenz gemessen. Der Test wird nicht durch Lipopolysaccharid gestört und auch Aluminiumhydroxid übt erst nach mehreren Stunden Inkubation einen interferierenden Einfluss aus. Ebenso konnte aktives Pertussis-Toxin mit Hilfe kryokonservierter PBMCs detektiert werden, auch in orientierenden Versuchen mit komplexen Impfstoffen. Der Pertussis-ATP-Test kommt der In vivo-Situation in der Zelle sehr nahe, weil beide Untereinheiten des Toxins in einem Test überprüft werden. Demnach soll er Bestandteil einer geplanten internationalen Studie zu alternativen Pertussis-Toxin-Testungen sein.
