The growth and the metabolism of Bifidobacterium adolescentis MB 239 fermenting GOS, lactose, galactose, and glucose were investigated. An unstructerd unsegregated model for growth of B. adolescentis MB 239 in batch cultures was developed and kinetic parameters were calculated with a Matlab algorithm. Galactose was the best carbon source; lactose and GOS led to lower growth rate and cellular yield, but glucose was the poorest carbon source. Lactate, acetate and ethanol yields allowed calculation of the carbon fluxes toward fermentation products. Similar distribution between 3- and 2-carbon products was observed on all the carbohydrates (45 and 55%, respectively), but ethanol production was higher on glucose than on GOS, lactose and galactose, in decreasing order. Based on the stoichiometry of the fructose 6-phosphate shunt and on the carbon distribution among the products, ATP yield was calculated on the different carbohydrates. ATP yield was the highest on galactose, while it was 5, 8, and 25% lower on lactose, GOS, and glucose, respectively. Therefore, a correspondance among ethanol production, low ATP yields, and low biomass production was established demonstrating that carbohydrate preferences may result from different sorting of carbon fluxes through the fermentative pathway. During GOS fermentation, stringent selectivity based on the degree of polymerization was exhibited, since lactose and the trisaccharide were first to be consumed, and a delay was observed until longer oligosaccharides were utilized. Throughout the growth on both lactose and GOS, galactose accumulated in the cultural broth, suggesting that β-(1-4) galactosides can be hydrolysed before they are taken up. The physiology of Bifidobacterium adolescentis MB 239 toward xylooligosaccharides (XOS) was also studied and our attention was focused on an extracellular glycosyl-hydrolase (β-Xylosidase) expressed by a culture of B. adolescentis grown on XOS as sole carbon source. The extracellular enzyme was purified from the the supernatant, which was dialyzed and concentrated by ultrafiltration. A two steps purification protocol was developed: the sample was loaded on a Mono-Q anion exchange chromatography and then, the active fractions were pooled and β-Xylosidase was purified by gel filtration chromatography on a Superdex-75. The enzyme was characterized in many aspects. β- Xylosidase was an homo-tetramer of 160 kDa as native molecular mass; it was a termostable enzyme with an optimum of temperature at 53 °C and an optimum of pH of 6.0. The kinetics parameter were calculated: km = 4.36 mM, Vmax = 0.93 mM/min. The substrate specificity with different di-, oligo- and polysaccharides was tested. The reactions were carried out overnight at pH 7 and at the optimum of temperature and the carbohydrates hydrolysis were analyzed by thin layer chromatography (TLC). Only glycosyl-hydrolase activities on XOS and on xylan were detected, whereas sucrose, lactose, cellobiose, maltose and raffinose were not hydrolyzed. It’s clearly shown that β-Xylosidase activity was higher than the Xylanase one. These studies on the carbohydrate preference of a strain of Bifidobacterium underlined the importance of the affinity between probiotics and prebiotics. On the basis of this concept, together with Barilla G&R f.lli SpA, we studied the possibility to develop a functional food containing a synbiotic. Three probiotic strains Lactobacillus plantarum BAR 10, Streptococcus thermophilus BAR 20, and Bifidobacterium lactis BAR 30 were studied to assess their suitability for utilization in synbiotic products on the basis of antioxidative activity, glutathione production, acid and bile tolerance, carbohydrates fermentation and viability in food matrices. Bile and human gastric juice resistance was tested in vitro to estimate the transit tolerance in the upper gastrointestinal tract. B. lactis and L. plantarum were more acid tolerant than S. thermophilus. All the strains resisted to bile. The growth kinetics on 13 prebiotic carbohydrates were determined. Galactooligosaccharides and fructo-oligosaccharides were successfully utilized by all the strains and could be considered the most appropriate prebiotics to be used in effective synbiotic formulations. The vitality of the three strains inoculated in different food matrices and maintained at room temperature was studied. The best survival of Lactobacillus plantarum BAR 10, Streptococcus thermophilus BAR 20, and Bifidobacterium lactis BAR 30 was found in food chocolate matrices. Then an in vivo clinical trial was carried out for 20 healthy volunteers. The increase in faecal bifidobacteria and lactobacilli populations and the efficacy of the pre-prototype was promising for the future develop of potential commercial products.

[ES]Entre mediados del siglo VIII y mediados del siglo XII, el espacio comprendido entre el Sistema Central y el río Tajo en su parte extremeña ejerció como confín territorial de al- Andalus. A lo largo de todo este tiempo se sucedieron distintas fases en las que la frontera fluctuó según vinieran los aires cristianos del norte o islámicos del sur. Ello provocaría que estos territorios actuaran con relativa autonomía en muchos momentos de su historia andalusí y que se convirtieran en zonas donde confluían gentes de toda condición social y religiosa. En el presente trabajo tratamos de vislumbrar las dinámicas territoriales y sociales que se dieron en aquella región hasta finales del siglo XI, cuando se produjeron las conquistas cristianas de las ciudades islámicas más importantes de la frontera. [EN] The geographical area between the Spanish Central System and the part of the Tagus passing through Extremadura served as the boundary of the al-Andalus from the 8th to the 12th century. The boundary had different phases that fluctuated according to the influence exercised by the northern Christians or southern Muslims. This allowed these territories to have relative autonomy during its Andalusian history and turned them into zones where people from different social and religious background converged. The aim of this paper is to analyse the territorial and social dynamics of this area until the end of the 11th century, when the Christians conquered the most important Islamic towns of the borderland.

This study aims at analysing Brian O'Nolans literary production in the light of a reconsideration of the role played by his two most famous pseudonyms ,Flann Brien and Myles na Gopaleen, behind which he was active both as a novelist and as a journalist. We tried to establish a new kind of relationship between them and their empirical author following recent cultural and scientific surveys in the field of Humour Studies, Psychology, and Sociology: taking as a starting point the appreciation of the comic attitude in nature and in cultural history, we progressed through a short history of laughter and derision, followed by an overview on humour theories. After having established such a frame, we considered an integration of scientific studies in the field of laughter and humour as a base for our study scheme, in order to come to a definition of the comic author as a recognised, powerful and authoritative social figure who acts as a critic of conventions. The history of laughter and comic we briefly summarized, based on the one related by the French scholar Georges Minois in his work (Minois 2004), has been taken into account in the view that humorous attitude is one of manâs characteristic traits always present and witnessed throughout the ages, though subject in most cases to repression by cultural and political conservative power. This sort of Super-Ego notwithstanding, or perhaps because of that, comic impulse proved irreducible exactly in its influence on the current cultural debates. Basing mainly on Robert R. Provineâs (Provine 2001), Fabio Ceccarelliâs (Ceccarelli 1988), Arthur Koestlerâs (Koestler 1975) and Peter L. Bergerâs (Berger 1995) scientific essays on the actual occurrence of laughter and smile in complex social situations, we underlined the many evidences for how the use of comic, humour and wit (in a Freudian sense) could be best comprehended if seen as a common mind process designed for the improvement of knowledge, in which we traced a strict relation with the play-element the Dutch historian Huizinga highlighted in his famous essay, Homo Ludens (Huizinga 1955). We considered comic and humour/wit as different sides of the same coin, and showed how the demonstrations scientists provided on this particular subject are not conclusive, given that the mental processes could not still be irrefutably shown to be separated as regards graduations in comic expression and reception: in fact, different outputs in expressions might lead back to one and the same production process, following the general âEconomy Ruleâ of evolution; man is the only animal who lies, meaning with this that one feeling is not necessarily biuniquely associated with one and the same outward display, so human expressions are not validation proofs for feelings. Considering societies, we found that in nature they are all organized in more or less the same way, that is, in élites who govern over a community who, in turn, recognizes them as legitimate delegates for that task; we inferred from this the epistemological possibility for the existence of an added ruling figure alongside those political and religious: this figure being the comic, who is the person in charge of expressing true feelings towards given subjects of contention. Any community owns one, and his very peculiar status is validated by the fact that his place is within the community, living in it and speaking to it, but at the same time is outside it in the sense that his action focuses mainly on shedding light on ideas and objects placed out-side the boundaries of social convention: taboos, fears, sacred objects and finally culture are the favourite targets of the comic personâs arrow. This is the reason for the word a(rche)typical as applied to the comic figure in society: atypical in a sense, because unconventional and disrespectful of traditions, critical and never at ease with unblinkered respect of canons; archetypical, because the âvillage foolâ, buffoon, jester or anyone in any kind of society who plays such roles, is an archetype in the Jungian sense, i.e. a personification of an irreducible side of human nature that everybody instinctively knows: a beginner of a tradition, the perfect type, what is most conventional of all and therefore the exact opposite of an atypical. There is an intrinsic necessity, we think, of such figures in societies, just like politicians and priests, who should play an elitist role in order to guide and rule not for their own benefit but for the good of the community. We are not naïve and do know that actual owners of power always tend to keep it indefinitely: the âsocial comicâ as a role of power has nonetheless the distinctive feature of being the only job whose tension is not towards stability. It has got in itself the rewarding permission of contradiction, for the very reason we exposed before that the comic must cast an eye both inside and outside society and his vision may be perforce not consistent, then it is satisfactory for the popularity that gives amongst readers and audience. Finally, the difference between governors, priests and comic figures is the seriousness of the first two (fundamentally monologic) and the merry contradiction of the third (essentially dialogic). MPs, mayors, bishops and pastors should always console, comfort and soothe popular mood in respect of the public convention; the comic has the opposite task of provoking, urging and irritating, accomplishing at the same time a sort of control of the soothing powers of society, keepers of the righteousness. In this view, the comic person assumes a paramount importance in the counterbalancing of power administration, whether in form of acting in public places or in written pieces which could circulate for private reading. At this point comes into question our Irish writer Brian O'Nolan(1911-1966), real name that stood behind the more famous masks of Flann O'Brien, novelist, author of At Swim-Two-Birds (1939), The Hard Life (1961), The Dalkey Archive (1964) and, posthumously, The Third Policeman (1967); and of Myles na Gopaleen, journalist, keeper for more than 25 years of the Cruiskeen Lawn column on The Irish Times (1940-1966), and author of the famous book-parody in Irish An Béal Bocht (1941), later translated in English as The Poor Mouth (1973). Brian O'Nolan, professional senior civil servant of the Republic, has never seen recognized his authorship in literary studies, since all of them concentrated on his alter egos Flann, Myles and some others he used for minor contributions. So far as we are concerned, we think this is the first study which places the real name in the title, this way acknowledging him an unity of intents that no-one before did. And this choice in titling is not a mere mark of distinction for the sake of it, but also a wilful sign of how his opus should now be reconsidered. In effect, the aim of this study is exactly that of demonstrating how the empirical author Brian O'Nolan was the real Deus in machina, the master of puppets who skilfully directed all of his identities in planned directions, so as to completely fulfil the role of the comic figure we explained before. Flann O'Brien and Myles na Gopaleen were personae and not persons, but the impression one gets from the critical studies on them is the exact opposite. Literary consideration, that came only after O'Nolans death, began with Anne Clissmannâs work, Flann O'Brien: A Critical Introduction to His Writings (Clissmann 1975), while the most recent book is Keith Donohueâs The Irish Anatomist: A Study of Flann O'Brien (Donohue 2002); passing through M.Keith Bookerâs Flann O'Brien, Bakhtin and Menippean Satire (Booker 1995), Keith Hopperâs Flann O'Brien: A Portrait of the Artist as a Young Post-Modernist (Hopper 1995) and Monique Gallagherâs Flann O'Brien, Myles et les autres (Gallagher 1998). There have also been a couple of biographies, which incidentally somehow try to explain critical points his literary production, while many critical studies do the same on the opposite side, trying to found critical points of view on the authorâs restless life and habits. At this stage, we attempted to merge into O'Nolan's corpus the journalistic articles he wrote, more than 4,200, for roughly two million words in the 26-year-old running of the column. To justify this, we appealed to several considerations about the figure O'Nolan used as writer: Myles na Gopaleen (later simplified in na Gopaleen), who was the equivalent of the street artist or storyteller, speaking to his imaginary public and trying to involve it in his stories, quarrels and debates of all kinds. First of all, he relied much on language for the reactions he would obtain, playing on, and with, words so as to ironically unmask untrue relationships between words and things. Secondly, he pushed to the limit the convention of addressing to spectators and listeners usually employed in live performing, stretching its role in the written discourse to come to a greater effect of involvement of readers. Lastly, he profited much from what we labelled his âspecific weightâ, i.e. the potential influence in society given by his recognised authority in determined matters, a position from which he could launch deeper attacks on conventional beliefs, so complying with the duty of a comic we hypothesised before: that of criticising society even in threat of losing the benefits the post guarantees. That seemingly masochistic tendency has its rationale. Every representative has many privileges on the assumption that he, or she, has great responsibilities in administrating. The higher those responsibilities are, the higher is the reward but also the severer is the punishment for the misfits done while in charge. But we all know that not everybody accepts the rules and many try to use their power for their personal benefit and do not want to undergo lawâs penalties. The comic, showing in this case more civic sense than others, helped very much in this by the non-accessibility to the use of public force, finds in the role of the scapegoat the right accomplishment of his task, accepting the punishment when his breaking of the conventions is too stark to be forgiven. As Ceccarelli demonstrated, the role of the object of laughter (comic, ridicule) has its very own positive side: there is freedom of expression for the person, and at the same time integration in the society, even though at low levels. Then the banishment of a âsocialâ comic can never get to total extirpation from society, revealing how the scope of the comic lies on an entirely fictional layer, bearing no relation with facts, nor real consequences in terms of physical health. Myles na Gopaleen, mastering these three characteristics we postulated in the highest way, can be considered an author worth noting; and the oeuvre he wrote, the whole collection of Cruiskeen Lawn articles, is rightfully a novel because respects the canons of it especially regarding the authorial figure and his relationship with the readers. In addition, his work can be studied even if we cannot conduct our research on the whole of it, this proceeding being justified exactly because of the resemblances to the real figure of the storyteller: its âchaptersâ âthe daily articlesâ had a format that even the distracted reader could follow, even one who did not read each and every article before. So we can critically consider also a good part of them, as collected in the seven volumes published so far, with the addition of some others outside the collections, because completeness in this case is not at all a guarantee of a better precision in the assessment; on the contrary: examination of the totality of articles might let us consider him as a person and not a persona. Once cleared these points, we proceeded further in considering tout court the works of Brian O'Nolan as the works of a unique author, rather than complicating the references with many names which are none other than well-wrought sides of the same personality. By putting O'Nolan as the correct object of our research, empirical author of the works of the personae Flann O'Brien and Myles na Gopaleen, there comes out a clearer literary landscape: the comic author Brian O'Nolan, self-conscious of his paramount role in society as both a guide and a scourge, in a word as an a(rche)typical, intentionally chose to differentiate his personalities so as to create different perspectives in different fields of knowledge by using, in addition, different means of communication: novels and journalism. We finally compared the newly assessed author Brian O'Nolan with other great Irish comic writers in English, such as James Joyce (the one everybody named as the master in the field), Samuel Beckett, and Jonathan Swift. This comparison showed once more how O'Nolan is in no way inferior to these authors who, greatly celebrated by critics, have nonetheless failed to achieve that great public recognition OâNolan received alias Myles, awarded by the daily audience he reached and influenced with his Cruiskeen Lawn column. For this reason, we believe him to be representative of the comic figureâs function as a social regulator and as a builder of solidarity, such as that Raymond Williams spoke of in his work (Williams 1982), with in mind the aim of building a âculture in commonâ. There is no way for a âculture in commonâ to be acquired if we do not accept the fact that even the most functional society rests on conventions, and in a world more and more âconnectedâ we need someone to help everybody negotiate with different cultures and persons. The comic gives us a worldly perspective which is at the same time comfortable and distressing but in the end not harmful as the one furnished by politicians could be: he lets us peep into parallel worlds without moving too far from our armchair and, as a consequence, is the one who does his best for the improvement of our understanding of things.

The vast majority of known proteins have not yet been experimentally characterized and little is known about their function. The design and implementation of computational tools can provide insight into the function of proteins based on their sequence, their structure, their evolutionary history and their association with other proteins. Knowledge of the three-dimensional (3D) structure of a protein can lead to a deep understanding of its mode of action and interaction, but currently the structures of <1% of sequences have been experimentally solved. For this reason, it became urgent to develop new methods that are able to computationally extract relevant information from protein sequence and structure. The starting point of my work has been the study of the properties of contacts between protein residues, since they constrain protein folding and characterize different protein structures. Prediction of residue contacts in proteins is an interesting problem whose solution may be useful in protein folding recognition and de novo design. The prediction of these contacts requires the study of the protein inter-residue distances related to the specific type of amino acid pair that are encoded in the so-called contact map. An interesting new way of analyzing those structures came out when network studies were introduced, with pivotal papers demonstrating that protein contact networks also exhibit small-world behavior. In order to highlight constraints for the prediction of protein contact maps and for applications in the field of protein structure prediction and/or reconstruction from experimentally determined contact maps, I studied to which extent the characteristic path length and clustering coefficient of the protein contacts network are values that reveal characteristic features of protein contact maps. Provided that residue contacts are known for a protein sequence, the major features of its 3D structure could be deduced by combining this knowledge with correctly predicted motifs of secondary structure. In the second part of my work I focused on a particular protein structural motif, the coiled-coil, known to mediate a variety of fundamental biological interactions. Coiled-coils are found in a variety of structural forms and in a wide range of proteins including, for example, small units such as leucine zippers that drive the dimerization of many transcription factors or more complex structures such as the family of viral proteins responsible for virus-host membrane fusion. The coiled-coil structural motif is estimated to account for 5-10% of the protein sequences in the various genomes. Given their biological importance, in my work I introduced a Hidden Markov Model (HMM) that exploits the evolutionary information derived from multiple sequence alignments, to predict coiled-coil regions and to discriminate coiled-coil sequences. The results indicate that the new HMM outperforms all the existing programs and can be adopted for the coiled-coil prediction and for large-scale genome annotation. Genome annotation is a key issue in modern computational biology, being the starting point towards the understanding of the complex processes involved in biological networks. The rapid growth in the number of protein sequences and structures available poses new fundamental problems that still deserve an interpretation. Nevertheless, these data are at the basis of the design of new strategies for tackling problems such as the prediction of protein structure and function. Experimental determination of the functions of all these proteins would be a hugely time-consuming and costly task and, in most instances, has not been carried out. As an example, currently, approximately only 20% of annotated proteins in the Homo sapiens genome have been experimentally characterized. A commonly adopted procedure for annotating protein sequences relies on the "inheritance through homology" based on the notion that similar sequences share similar functions and structures. This procedure consists in the assignment of sequences to a specific group of functionally related sequences which had been grouped through clustering techniques. The clustering procedure is based on suitable similarity rules, since predicting protein structure and function from sequence largely depends on the value of sequence identity. However, additional levels of complexity are due to multi-domain proteins, to proteins that share common domains but that do not necessarily share the same function, to the finding that different combinations of shared domains can lead to different biological roles. In the last part of this study I developed and validate a system that contributes to sequence annotation by taking advantage of a validated transfer through inheritance procedure of the molecular functions and of the structural templates. After a cross-genome comparison with the BLAST program, clusters were built on the basis of two stringent constraints on sequence identity and coverage of the alignment. The adopted measure explicity answers to the problem of multi-domain proteins annotation and allows a fine grain division of the whole set of proteomes used, that ensures cluster homogeneity in terms of sequence length. A high level of coverage of structure templates on the length of protein sequences within clusters ensures that multi-domain proteins when present can be templates for sequences of similar length. This annotation procedure includes the possibility of reliably transferring statistically validated functions and structures to sequences considering information available in the present data bases of molecular functions and structures.

Das Ziel dieser Arbeit war die Entwicklung neuartiger organischer Materialien, die eine Absorption im NIR-Bereich besitzen. Zu diesem Zweck wurden höher konjugierte Carbinole und Tetrakis(stilbenyl)methane synthetisiert und aus den Carbinolen mit Trifloressigsäure Carbokationen-Salze hergestellt. Die Darstellung der Oligomeren erfolgte durch eine konvergente Synthesestrategie, bei der die meisten der synthetisierten Vorstufen in mehreren Folgereaktionen eingesetzt werden konnten. Als Schlüsselschritt der Synthesen wurde fast ausschließlich die Horner-Reaktion angewendet. Die Vorteile lagen dabei in der leichten Zugänglichkeit der Edukte, in hohen Produktausbeuten und vor allem in der hohen trans-Selektivität der Reaktion. Die effektiven Konjugationslängen (EKL) der Carbinole und Carbokationen wurden mit Hilfe der Fitfunktion berechnet. Mit zunehmender Ausdehnung des p-Systems wird der HOMO-LUMO Abstand des energieärmsten p-p*- Übergangs geringer und die Absorption zum sichtbaren Bereich hin bathochrom (langwellig) verschoben. Die Substitution mit einem Dialkylamino-Rest bewirkt ebenfalls eine bathochrome Verschiebung der Absorptionsbande.Umgekehrt verhält es sich bei den Carbokationen. Hier absorbieren die dialkylaminosubstituierten Verbindungen bei kürzerer Wellenlänge als die alkoxysubstituierten, da bei der Herstellung der Kationen aus den Carbinolen ,zuerst der Sickstoff als elektronenreichste Stelle protoniert wird. Dabei hat die Menge an zugesetzter Trifluoressigsäure einer entscheidende Einfluß auf die Bildung der Carbokationen aus den Carbinolen (s. Tabele 4-3). Während bei den alkoxysubstituierten Carbinolen das Gleichgewicht zwischen Carbinol und Carbokation schon nach Zusatz von ca 4 % Trifluoressigsäure erreicht ist, benötigt man bei den dialkylaminosubstituierten Carbinolen eine wesentlich höhere Säurekonzentration (10 %) zur vollständigen Protonierung (n = 4). Beiden Systemen ist gemein, daß nach Überschreiten der benötigten Gleichgewichtskonzentration an Trifluoressigsäure, eine Erhöhung der Säuremenge einen Abbau des Chromophos zur Folge hat. Offensichtlich wird dann die Doppelbindung protoniert, ein Vorgang der allerdings reversibel ist und keine Isomerisierungen beinhaltet.E/Z-Isomerisierungen finden allerdings durch Bestrahlungen mit Licht der Wellenlänge l = 366 nm statt. Bei dem Tetrakis(stilbenyl)methan 25a liegt das photostationäre Gleichgewicht bei ca 1/9 (Z/E), bei dem Tris(stilbenyl)methanol 19a liegt es bei 1/7 (Z/E).

Zusammenfassung:In der vorliegenden Arbeit konnte gezeigt werden, dass durch die Anwendung von Cyclodextrinen in der Polymerchemie ein neuer Weg gefunden wurde, Homo- und Copolymerisationen von hydrophoben, fluorophilen sowie ionischen Monomeren in wässrigem Medium durchzuführen, die bislang nur durch den Einsatz von Emulgatoren oder in organischen Lösemittelgemischen möglich waren. Standardmonomere wie z.B. Styrol konnten erfolgreich aus wässriger Phase in Gegenwart von Cyclodextrin polymerisiert werden; unter Anwendung des Zulauf-Verfahrens wurden stabile Latices mit monodisperser Teilchengrössenverteilung erhalten. Durch Verwendung von Cyclodextrinen als Löslichkeitsvermittler konnten fluorierte Methacrylate in wässriger Lösung in fast quantitativer Ausbeute homo- bzw. mit Styrol copolymerisiert werden, während bei Abwesenheit von Cyclodextrin keine bzw. nur geringe Umsätze erzielt wurden. Weiterhin ließen sich auch sehr unterschiedliche Monomerpaare wie 1H,1H,2H,2H-Perfluordecylacrylat mit Stearylmethacrylat in wässriger Phase unter Verwendung von Cyclodextrin copolymerisieren. Ein ebenfalls gegensätzliches Monomerenpaar, das sich als Cyclodextrin-Komplex in hohen Ausbeuten und zu hohen Molekulargewichten copolymerisieren ließ, war Natrium-4-(acrylamido)-phenyldiazosulfonat und Styrol, dessen Copolymerisation bislang nur in org. Lösemittelgemischen möglich war. In der Synthese leitfähiger Polymere konnte ein neuer, umweltfreundlicher Weg zur Herstellung von Polyheterocyclen durch oxidative Polymerisation der komplexierten Monomere in Wasser, wie z.B. 3,4-Ethylendioxythiophen oder Pyrrol, die ihre Anwendung zur Herstellung leitfähiger Polymere finden, erarbeitet werden.Zur Variation der Polymereigenschaften wurde auch erstmalig die neue Substanzklasse von 2H-Benzo[5,6][1,4]dioxino[2,3-c]pyrrol-Derivaten synthetisiert.

Photosensible Polymere haben bis heute vielfältigeAnwendungsmöglichkeiten erlangt, wie z. B. als Photoresistsund als Grundlage optischer Datenspeicher. In dieser Arbeitwurden photosensible mesoionische Pyrimidinium-olate unterEinsatz von verschiedenen neuen Methoden an Polymereangebunden und Filme dieser Polymere untersucht.Es wurde dazu zunächst eine Serie von niedermolekularenmesoionischen Modellverbindungen hergestellt, um derenVerhalten bei der Photoreaktion zum Bis(ß-lactam) zuanalysieren und die Eigenschaften mit semiempirischberechneten Werten zu vergleichen. Durch die Auswahlverschiedener Substituenten konnte dieAbsorptionswellenlänge des HOMO-LUMO Überganges in einemBereich zwischen 320 und 490 nm verändert werden.Zur Anbindung mesoionischer Pyrimidinium-olate an Polymerewurden verschiedene Methoden eingesetzt, um möglichst großeVariationen der Substituenten zu ermöglichen. Es wurdendabei sowohl auf Methacrylaten basierende Homo- undCopolymere mit der mesoionischen Funktion in derSeitenkette, als auch Polymere mit der mesoionischenFunktion in der Hauptkette durch eine polymeranalogeUmsetzung von polymeren Amidinen hergestellt.Am Beispiel von mesoionischem2-[2-(Isopropenylcarbonyloxy)ethylthio]-1-methyl-6-oxo-3-phenyl-5-propyl-1,6-dihydropyrimidin-3-ium-4-olatwurde auch eine Polymerisation in Wasser als Lösungsmitteldurchgeführt. Dazu wurde der 2:1 Komplex aus methyliertemß-Cyclodextrin (Methylierungsgrad 1,8) und dem Monomer miteinem wasserlöslichen Redoxinitiator polymerisiert. Dasausgefallene Produkt enthielt neben einem Oligomeranteil dasPolymer mit einem Molekulargewicht von 3.000 g/mol und einerniedrigen Polydispersität von 1,3. Außerdem wurde auch einCopolymer aus dem Komplex und komplexiertemMethylmethacrylat hergestellt. Es zeigte ein von dem in DMFohne Cyclodextrin hergestellten Copolymer abweichendesmassenabhängiges Einbauverhältnis.Mit der dielektrischen Spektroskopie konnten verschiedeneArrhenius aktivierte Rotationsprozesse gefunden werden,weiterhin gelang der Nachweis des Einflusses derPhotoreaktion auf den Brechungsindex und Geschwindigkeit desHochtemperaturprozesses. Zur genauen Betrachtung von Änderungen von Brechungsindexund Schichtdicke bei der Bestrahlung von Polymerfilmen wurdedie Wellenleitermodenspektroskopie eingesetzt. Während dieSeitenkettenpolymere auf Methacrylatbasis bei derBestrahlung eine deutliche Verringerung der Brechungsindicesund eine Erhöhung der Filmdicke zeigten, konnte bei demHauptkettenpolymer ein anisotropes Verhalten gefundenwerden. Zudem ist es auch gelungen, holographische Gitter inPolymere aus mesoionischen Pyrimidinium-olateneinzuschreiben. Die Bildung von Oberflächenwellen konntedabei mit der Atomkraftmikroskopie nachgewiesen werden.Weiterhin wurde auch die Kinetik der Photoreaktionuntersucht. Es konnte dabei gezeigt werden, dass einedeutlicher Beschleunigung sowohl durch eineTemperaturerhöhung über die Glasübergangstemperatur hinausals auch durch den Einsatz von flüssigkristallinenComonomeren, die zu einer weicheren Umgebung derphotoreaktiven Gruppen im Polymer führten, erzielt werdenkonnte.

Es wurden neue funktionalisierte Carbazole und anellierte Benzo[a]carbazole als potentielle pharmakologische Therapeutika durch 1,6-pi-Elektrocyclisierung auf photo-chemischem, thermischem und sonochemischem Weg synthetisiert und die Synthesemethoden der 1,6-pi-Elektrocyclisierung sowie der 2,3-Divinylindole und der 2-Aryl-3-vinylindole als entsprechende Ausgangsprodukte validiert und evaluiert. Es gelang weder das nach den Woodward-Hoffmann-Regeln erwartete primäre Cyclisierungsprodukt mit Indolochinodimethanstruktur noch die Existenz des in einer photochemischen Abfangreaktion daraus resultierenden Cycloprodukts NMR-spektroskopisch nachzuweisen, um den stereochemischen Verlauf der Cyclisierung vorherzusagen. Ergebnisse der quantenchemischen Berechnungen der Eduktmoleküle (AO-Koeffizienten der MO's, HOMO/LUMO-Energien) sowie der Übergangszustandsgeometrien der Cyclisierungen decken sich mit den experimentellen Daten. Divinyl- und 2-Aryl-3-vinylindole sind als Systeme mit Hexatriensymmetrie aufzufassen, deren Cyclisierungsverhalten sich mit den Woodward-Hoffmann-Regeln beschreiben läßt. Im Vergleich der verschiedenen 1,6-pi-Elektrocyclisierungsmethoden zeigte sich, daß die photochemische Variante eine elegante Synthesemethode darstellt, um funktionalisierte Carbazole und Benzo[a]carbazole mit unterschiedlichen pharmakologischen Aktivitäten unter schonenden Reaktionsbedingungen mit den vergleichbar höchsten Ausbeuten zu erhalten. Demgegenüber lieferten die Ultraschallreaktionen keine nachweisbaren Cyclisierungsprodukte. Die thermische Cyclisierung führte zur Gruppe der 1,2-Dihydrocarbazole. Sie bildeten sich in einer Folgereaktion durch [1,5s]-H-Verschiebung aus dem primär entstandenen Woodward-Hoffmann-Cyclisierungsprodukt. In abschließenden DNA-Bindestudien mit verschiedenen Testsystemen zeigte keine der synthetisierten Testsubstanzen DNA-Bindungsaktivität.

Es wird eine Reihe von dimeren Seltenerdhydriden der Zusammensetzung [Ln(C5Me4NCMe2R)(THF)(H)]2 mit Ln= Y,Tb,Er,Lu und R= Me,Et auf ihre Eigenschaften als Polymerisationsinitiatoren untersucht.Die kontrollierte Polymerisation von Styrol ist nach der Bildung der Monoinsertionsprodukte von 1-Olefinen möglich. Dabei ist die Menge des im System verbleibenden THF von entscheidender Bedeutung. Die Insertion des ersten Styrolmoleküls verläuft selektiv sekundär.Es wird eine Korrelation der Polymerisationsgeschwindigkeit mit dem Ionenradius des verwendeten Metalls beobachtet.Die Erzeugung von Blockcopolymeren aus Styrol und tert.-Butylacrylat sowie anderen polaren Monomeren ist auf diesem Weg möglich und wird untersucht. Während die Länge des Polystyrolblocks eingestellt werden kann, verläuft die Polymerisation des Acrylats unkontrolliert. Es bilden sich Blockcopolymere mit einem Blocklängenverhältnis von etwa 1:1sowie Homopoly(tert.-Butylacrylat).Darüber hinaus wird das Verhalten der genannten Hydride sowie mehrerer anderer Seltenerdkompexe (u. a. mit Schiff-Base-Liganden) in der Polymerisation von rac.-beta-Butyrolacton untersucht. Die Hydride polymerisieren rac.-beta-Butyrolacton zu Polymeren mit Molekulargewichten von 10000-20000 g/mol. Gelbildung und Umesterungsreaktionentreten auf. Unter Kühlung ist eine bessere Kontrolle der Reaktion möglich. Die erhaltenen Polymere weisen eine syndiotaktisch angereicherte Mikrostruktur auf.Die Synthese der Schiff-Base-Komplexe erfolgt durch Umsetzung von R(-)-2,2'-Bis(6-tert.-butyl-4-ethylsalicylidenimin)-1,1'-binaphthyl ('LigH2') bzw. (1R,2R)-(-)-1,2-Cyclohexandiamino-N,N'-bis(3,5-di-tert.-butylsalicyliden)('(R,R)-Jacobsen-Ligand') mit Y(N(SiHMe2)2)3(THF)2. Die resultierenden Verbindungen sowie Y(CH2(SiMe3))3(THF)2 sind Initiatoren für die Polymerisation von rac.-beta-Butyrolacton und Lactid. Es wird keine kontrollierte Polymerisation beobachtet. Mit dem Schiff-Base-KomplexLigY(N(SiHMe2)2)(THF) wird bei der Polymerisation von rac.-beta-Butyrolacton eine syndiotaktische Anreicherung von bis zu 75 % s-Diaden erreicht.
Structure and dynamics of supramolecular assemblies studied by advanced solid-state NMR spectroscopy

Ziel der vorliegenden Arbeit ist die Aufklärung von Struktur und Dynamik komplexer supramolekularer Systeme mittels Festkörper NMR Spektroskopie. Die Untersuchung von pi-pi Wechselwirkungen, welche einen entscheidenden Einfluss auf die strukturellen und dynamischen Eigenschaften supra- molekularer Systeme haben, hilft dabei, die Selbst- organisationsprozesse dieser komplexen Materialien besser zu verstehen. Mit dipolaren 1H-1H and 1H-13C Wiedereinkopplungs NMR Methoden unter schnellem MAS können sowohl 1H chemische Verschiebungen als auch dipolare 1H-1H und 1H-13C Kopplungen untersucht werden, ohne dass eine Isotopenmarkierung erforderlich ist. So erhält man detaillierte Informationen über die Struktur und die Beweglichkeit einzelner Molekül- segmente. In Verbindung mit sogenannten nucleus independent chemical shift (NICS) maps (berechnet mit ab-initio Methoden) lassen sich Abstände von Protonen relativ zu pi-Elektronensystemen bestimmen und so Strukturvorschläge ableiten. Mit Hilfe von homo- und heteronuklearen dipolaren Rotationsseitenbandenmustern könnenaußerdem Ordnungs- parameter für verschiedene Molekülsegmente bestimmt werden. Die auf diese Weise gewonnenen Informationen über die strukturellen und dynamischen Eigenschaften supramolekularer Systeme tragen dazu bei, strukturbestimmende Molekül- einheiten und Hauptordnungsphänomene zu identifizieren sowie lokale Wechselwirkungen zu quantifizieren, um so den Vorgang der Selbstorganisation besser zu verstehen.

Aim of this thesis was to further extend the applicability of the Fourier-transform (FT) rheology technique especially for non-linear mechanical characterisation of polymeric materials on the one hand and to investigated the influence of the degree of branching on the linear and non-linear relaxation behaviour of polymeric materials on the other hand. The latter was achieved by employing in particular FT-rheology and other rheological techniques to variously branched polymer melts and solutions. For these purposes, narrowly distributed linear and star-shaped polystyrene and polybutadiene homo-polymers with varying molecular weights were anionically synthesised using both high-vacuum and inert atmosphere techniques. Furthermore, differently entangled solutions of linear and star-shaped polystyrenes in di-sec-octyl phthalate (DOP) were prepared. The several linear polystyrene solutions were measured under large amplitude oscillatory shear (LAOS) conditions and the non-linear torque response was analysed in the Fourier space. Experimental results were compared with numerical predictions performed by Dr. B. Debbaut using a multi-mode differential viscoelastic fluid model obeying the Giesekus constitutive equation. Apart from the analysis of the relative intensities of the harmonics, a detailed examination of the phase information content was developed. Further on, FT-rheology allowed to distinguish polystyrene melts and solutions due to their different topologies where other rheological measurements failed. Significant differences occurred under LAOS conditions as particularly reflected in the phase difference of the third harmonic, ¶3, which could be related to shear thinning and shear thickening behaviour.
