Avaliou-se o crescimento micelial in vitro das linhagens ABI-05/03, ABI-06/04, ABI-04/02, ABI-06/05 e ABI-01/01 de Agaricus bisporus em meios de cultura sólidos à base de composto. As avaliações foram realizadas por meio de medições de quatro diâmetros das colônias, a cada 48h, durante 12 dias de incubação, no escuro, a 20 e 25ºC. O delineamento experimental foi o de blocos casualizados, com uso do teste de Tukey para a comparação das médias. Co m base nos resultados obtidos, verificou-se que: o crescimento micelial de A. bisporus é influenciado pela temperatura de incubação; a temperatura de 25ºC foi mais favorável para o crescimento micelial de todas as linhagens de A. bisporus; na temperatura de 20ºC, o melhor crescimento foi obtido com as linhagens ABI-06/05 e ABI-01/01; na temperatura de 25ºC, a linhagem ABI-01/01 apresentou crescimento significativamente maior que todas as demais.

A seca de ponteiros é uma doença que vem acarretando danos severos em plantas de eucalipto, causando cancros ao longo do ramo principal. O objetivo deste trabalho foi avaliar o efeito de fungicidas e extratos vegetais no controle de Botryosphaeria ribis. O teste in vitro dos fungicidas foi inteiramente casualizado com oito tratamentos: carbendazim, clorotalonil, difenoconazole, picoxystrobin + ciproconazole, ciproconazole, azoxystrobin, picoxystrobin e testemunha, quatro doses: 1 µg/mL, 10 µg/mL, 100 µg/mL e 1000 µg/mL; com cinco repetições. Após homogeneização do meio foram vertidas para as placas, repicado um disco de meio de cultura contendo o patógeno para estas placas e mantidas em BOD a 25°C por cinco dias. A avaliação foi feita através de medição diária do crescimento radial do micélio em centímetros. O delineamento experimental do controle químico a campo foi em fatorial 5x3, com cinco fungicidas e três métodos de aplicação (poda, pincelado e pulverizado), com quatro repetições. Foram feitas quatro aplicações, com intervalo de quinze dias. Os tratamentos foram: 1. azoxystrobin, 2. carbendazim, 3. clorotalonil + tiofanato-metílico, 4. difenoconazole e 5. testemunha. Simultaneamente foram feitas avaliações seguindo uma escala de notas de 1 a 6. O delineamento experimental do teste in vitro dos extratos vegetais foi em fatorial 5x4, com três repetições. Os tratamentos avaliados foram os extratos de: mil folhas, melão de são caetano, eucalipto , álcool e testemunha, nas concentrações de 5, 10, 15 e 20%. Os extratos e o álcool foram misturados ao meio de cultura previamente autoclavado, sendo estes colocados em placas de Petri. A avaliação foi feita através de medição diária do crescimento radial do micélio em centímetros. No teste com mudas o delineamento experimental foi em fatorial 2 x 2 x 4, sendo utilizado no experimento de duas procedências, dois modos de tratamento (preventivo e convencional) e quatro produtos para o controle (extrato de melão de são caetano, extrato de Corymbia citriodora, álcool e água). A inoculação do patógeno foi feita no caule das mudas. Foram feitas aplicações dos produtos e avaliações semanais. A avaliação foi feita através da contagem de plantas doentes. O ingrediente ativo carbendazin foi o que mostrou os melhores resultados in vitro diferindo estatísticamente dos outros tratamentos pelo teste Tukey a 1% de probabilidade; seguido pelo clorotalonil e difenoconazole. Todos os ingredientes ativos avaliados mostraram-se superiores a testemunha. A campo, azoxystrobin foi superior aos demais tratamentos e não houve diferença significativa entre os métodos de aplicação. In vitro, os extratos de mil folhas, melão e eucalipto não diferiram entre si pelo teste de Tukey a 5% de probabilidade. O álcool proporcionou a maior inibição do crescimento micelial e diferiu estatisticamente dos outros tratamentos utilizados. As concentrações de 10, 15 e 20% dos extratos não diferiram entre si, mas foram superiores a concentração de 5%. No teste em mudas, a aplicação preventiva mostrou-se superior a aplicação curativa, sendo que o álcool e o extrato de C. citriodora não diferiram entre si, mas foram superiores ao extrato de melão-de-são-caetano. Todos os produtos foram superiores a testemunha.

Botryosphaeria rhodina and Trichoderma harzianum Rifai were grown on botryosphaeran (an exopolysaccharide (EPS) of the beta-1,3; 1,6-D-Glucan type produced by B. rhodina) as sole carbon source with the objective of producing beta-glucanases of the beta-type. Conditions for beta-1,3-glucanase production by T harzianum were examined by a statistical response surface method, and showed maximal enzyme production at 5 days growth in media containing 1.5 g/1 of EPS. Good agreement was obtained between the experimental values of beta-1, 3-glucanase activity and the corresponding values predicted by the mathernatical model. The crude beta-1,3-glucanase preparations were active towards a number of different beta-1,3-glucans and beta-glucosides. The mycelium of B. rhodina also proved to be a good substrate for beta-1,3-glucanase production by both fungal species. (c) 2005 Published by Elsevier Ltd.
Genetic structure of populations of Rhizoctonia solani anastomosis group-1 IA from soybean in Brazil

The Basidiomycete fungus Rhizoctonia solani anastomosis group (AG)-1 IA is a major pathogen of soybean in Brazil, where the average yield losses have reached 30 to 60% in some states in Northern Brazil. No information is currently available concerning levels of genetic diversity and population structure for this pathogen in Brazil. A total of 232 isolates of R. solani AG1 IA were collected from five soybean fields in the most important soybean production areas in central-western, northern, and northeastern Brazil. These isolates were genotyped using 10 microsatellite loci. Most of the multilocus genotypes (MLGTs) were site-specific, with few MLGTs shared among populations. Significant population subdivision was evident. High levels of admixture were observed for populations from Mato Grosso and Tocantins. After removing admixed genotypes, three out of five field populations (Maranhao, Mato Grosso, and Tocantins), were in Hardy-Weinberg (HW) equilibrium, consistent with sexual recombination. HW and gametic disequilibrium were found for the remaining soybean-infecting populations. The findings of low genotypic diversity, departures from HW equilibrium, gametic disequilibrium, and high degree of population subdivision in these R. solani AG-1 IA populations from Brazil are consistent with predominantly asexual reproduction, short-distance dispersal of vegetative propagules (mycelium or sclerotia), and limited long-distance dispersal, possibly via contaminated seed. None of the soybean-infecting populations showed a reduction in population size (bottleneck effect). We detected asymmetric historical migration among the soybean-infecting populations, which could explain the observed levels of subdivision.

Leaf-cutting ants belonging to the tribe Attini are major herbivores and important agriculture pests in the neotropics, these ants being thought to feed on the sap which exudes from the plant material which they cut and also on the mycelium of a symbiotic fungus that grows on plant material inside their nests in what is called 'the fungus garden'. However, we have found that the survival of Atta sexdens worker ants on leaves, on mycelium. of the ants' symbiotic fungus, Leucoagaricus gongylophorus, or on plant polysaccharides was the same as that of starved A. sexdens, while, conversely, significantly longer survival was achieved by ants fed on the fungus garden material or on some of the products (especially glucose) of the hydrolysis of plant polysaccharides. We found that the fungus garden contained glucose at a higher concentration than that found in leaves or fungal mycelium, and that this glucose was consumed by the ant to the extent that it was probably responsible for up to 50% of the nutritional needs of the workers. The fungus garden contained polysaccharide degrading enzymes (pectinase, amylase, xylanase and cellulase) in proportions similar to that observed in laboratory cultures of L. gongylophorus. It thus appears that A. sexdens workers obtain a significant part of their nutrients from plant polysaccharide hydrolysis products produced by the action of extracellular enzymes released by L. gongylophorus. In this paper we discuss the symbiotic nutrition strategy of A. sexdens workers and brood and the role played by plant polysaccharides in the nutrition of attine ants. (C) 2003 Elsevier B.V. Ltd. The gene encoding glycogen synthase in Neurospora crassa (gsn) is transcriptionally down-regulated when mycelium is exposed to a heat shock from 30 to 45 degrees C. The gsn promoter has one stress response element (STRE) motif that is specifically bound by heat shock activated nuclear proteins. In this work, we used biochemical approaches together with mass spectrometric analysis to identify the proteins that bind to the STRE motif and could participate in the gsn transcription regulation during heat shock. Crude nuclear extract of heat-shocked mycelium was prepared and fractionated by affinity chromatography. The fractions exhibiting DNA-binding activity were identified by electrophoretic mobility shift assay (EMSA) using as probe a DNA fragment containing the STRE motif DNA-protein binding activity was confirmed by Southwestern analysis. The molecular mass (MM) of proteins was estimated by fractionating the crude nuclear extract by SDS-PAGE followed by EMSA analysis of the proteins corresponding to different MM intervals. Binding activity was detected at the 30-50 MM kDa interval. Fractionation of the crude nuclear proteins by IEF followed by EMSA analysis led to the identification of two active fractions belonging to the pIs intervals 3.54-4.08 and 6.77-7.31. The proteins comprising the MM and pI intervals previously identified were excised from a 2-DE gel, and subjected to mass spectrometric analysis (MALDI-TOF/TOF) after tryptic digestion. The proteins were identified by search against the MIPS and MIT N. crassa databases and five promising candidates were identified. Their structural characteristics and putative roles in the gsn transcription regulation are discussed.

Both P-i-repressible acid phosphatases, IIb (mycelial) and IIc (extracellular), synthesized by Neurospora crassa and purified to apparent homogeneity by 7.5% PAGE, are monomers, are inhibited by 2 mM ZnCl2 and are nonspecifically stimulated by salts. However, the IIc form is activated by p-nitrophenylphosphate (in a negative cooperativity effect with a K-0.5 of 2.5 mM) whereas form IIb shows Michaelis kinetics, with a K-m of 0.5 mM. Thus, since both enzymatic forms may be expressed by the same gene (pho-3), it is possible that post-translational modifications lead to the excretion of an enzymatic form with altered Michaelis kinetics compared with the enzymatic form retained by the mycelium.
