A simple and fast method to determine Ca, K and Mg in a single aliquot of medicinal plants by HR-CS FAAS is proposed. The secondary lines for Ca (239.856 nm) and K (404.414 nm), and the alternate line measured at wing of the secondary line for Mg at (202.588 nm) allowed calibration within the 20 - 500 mg.L-1 Ca and K, and 1.0 - 80 mg.L-1 Mg. Twenty samples and three plant certified materials were analyzed. Results were in agreement at a 95% confidence level with reference values. Limits of detection were 2.4 mg.L-1 Ca, 1.9 mg.L-1 K and 0.3 mg.L-1 Mg. The RSD (n=12) were ≤ 5.1% and recoveries were between 83 and 108% for all analytes.

A fast method was optimized and validated in order to quantify amphetamine-type stimulants (amphetamine, AMP; methamphetamine, MAMP; fenproporex, FPX; 3,4-methylenedioxymethamphetamine, MDMA; and 3,4-methylenedioxyamphetamine, MDA) in human hair samples. The method was based in an initial procedure of decontamination of hair samples (50 mg) with dichloromethane, followed by alkaline hydrolysis and extraction of the amphetamines using hollow-fiber liquid-phase micro extraction (HF-LPME) in the three-phase mode. Gas chromatography-mass spectrometry (GC-MS) was used for identification and quantification of the analytes. The LoQs obtained for all amphetamines (around 0.05 ng/mg) were below the cut-off value (0.2 ng/mg) established by the Society of Hair Testing (SoHT). The method showed to be simple and precise. The intra-day and inter-day precisions were within 10.6% and 11.4%, respectively, with the use of only two deuteratecl internal standards (AMP-d5 and MDMA-d5). By using the weighted least squares linear regression (1/x(2)), the accuracy of the method was satisfied in the lower concentration levels (accuracy values better than 87%). Hair samples collected from six volunteers who reported regular use of amphetamines were submitted to the developed method. Drug detection was observed in all samples of the volunteers. The reproductive performance of cattle may be influenced by several factors, but mineral imbalances are crucial in terms of direct effects on reproduction. Several studies have shown that elements such as calcium, copper, iron, magnesium, selenium, and zinc are essential for reproduction and can prevent oxidative stress. However, toxic elements such as lead, nickel, and arsenic can have adverse effects on reproduction. In this paper, we applied a simple and fast method of multi-element analysis to bovine semen samples from Zebu and European classes used in reproduction programs and artificial insemination. Samples were analyzed by inductively coupled plasma spectrometry (ICP-MS) using aqueous medium calibration and the samples were diluted in a proportion of 1:50 in a solution containing 0.01% (vol/vol) Triton X-100 and 0.5% (vol/vol) nitric acid. Rhodium, iridium, and yttrium were used as the internal standards for ICP-MS analysis. To develop a reliable method of tracing the class of bovine semen, we used data mining techniques that make it possible to classify unknown samples after checking the differentiation of known-class samples. Based on the determination of 15 elements in 41 samples of bovine semen, 3 machine-learning tools for classification were applied to determine cattle class. Our results demonstrate the potential of support vector machine (SVM), multilayer perceptron (MLP), and random forest (RF) chemometric tools to identify cattle class. Moreover, the selection tools made it possible to reduce the number of chemical elements needed from 15 to just 8.

Arrabidaea chica (crajiru) is an important Amazonian plant. Its extracts are used as red pigments, antimicrobial agents and astringents. Three different varieties of this species are cultivated in the Amazon region. In this work, direct infusions of A. chica extracts from these three varieties were analyzed via electrospray ionization mass spectrometry (ESI(+)-MS) fingerprinting. Derived data from the spectra were classified by using a multivariate method (PLS-DA, partial least squares-discriminant analysis). The direct method that is herein presented relies on extraction of dry, powdered leaves with acidified methanol/water solution with no further sample preparation. The resulting supernatants were analyzed by direct infusion ESI(+)-MS, which provides characteristic fingerprints of the sample composition. 3-Deoxyanthocyanidins are important substances in A. chica, their ions were used as markers in the PLS-DA data treatment. PLS-DA was able to differentiate the three varieties. ESI(+)-MS fingerprinting works as a simple and fast method to differentiate varieties of A. chica.

In this report it was designed an innovative satellite-based monitoring approach applied on the Iraqi Marshlands to survey the extent and distribution of marshland re-flooding and assess the development of wetland vegetation cover. The study, conducted in collaboration with MEEO Srl , makes use of images collected from the sensor (A)ATSR onboard ESA ENVISAT Satellite to collect data at multi-temporal scales and an analysis was adopted to observe the evolution of marshland re-flooding. The methodology uses a multi-temporal pixel-based approach based on classification maps produced by the classification tool SOIL MAPPER ®. The catalogue of the classification maps is available as web service through the Service Support Environment Portal (SSE, supported by ESA). The inundation of the Iraqi marshlands, which has been continuous since April 2003, is characterized by a high degree of variability, ad-hoc interventions and uncertainty. Given the security constraints and vastness of the Iraqi marshlands, as well as cost-effectiveness considerations, satellite remote sensing was the only viable tool to observe the changes taking place on a continuous basis. The proposed system (ALCS – AATSR LAND CLASSIFICATION SYSTEM) avoids the direct use of the (A)ATSR images and foresees the application of LULCC evolution models directly to „stock‟ of classified maps. This approach is made possible by the availability of a 13 year classified image database, conceived and implemented in the CARD project (http://earth.esa.int/rtd/Projects/#CARD).The approach here presented evolves toward an innovative, efficient and fast method to exploit the potentiality of multi-temporal LULCC analysis of (A)ATSR images. The two main objectives of this work are both linked to a sort of assessment: the first is to assessing the ability of modeling with the web-application ALCS using image-based AATSR classified with SOIL MAPPER ® and the second is to evaluate the magnitude, the character and the extension of wetland rehabilitation.

The present study was performed to validate a spatial working memory task using pharmacological manipulations. The water escape T-maze, which combines the advantages of the Morris water maze and the T-maze while minimizes the disadvantages, was used. Scopolamine, a drug that affects cognitive function in spatial working memory tasks, significantly decreased the rat performance in the present delayed alternation task. Since glutamate neurotransmission plays an important role in the maintaining of working memory, we evaluated the effect of ionotropic and metabotropic glutamatergic receptors antagonists, administered alone or in combination, on rat behaviour. As the acquisition and performance of memory tasks has been linked to the expression of the immediately early gene cFos, a marker of neuronal activation, we also investigated the neurochemical correlates of the water escape T-maze after pharmacological treatment with glutamatergic antagonists, in various brain areas. Moreover, we focused our attention on the involvement of perirhinal cortex glutamatergic neurotransmission in the acquisition and/or consolidation of this particular task. The perirhinal cortex has strong and reciprocal connections with both specific cortical sensory areas and some memory-related structures, including the hippocampal formation and amygdala. For its peculiar position, perirhinal cortex has been recently regarded as a key region in working memory processes, in particular in providing temporary maintenance of information. The effect of perirhinal cortex lesions with ibotenic acid on the acquisition and consolidation of the water escape T-maze task was evaluated. In conclusion, our data suggest that the water escape T-maze could be considered a valid, simple and quite fast method to assess spatial working memory, sensible to pharmacological manipulations. Following execution of the task, we observed cFos expression in several brain regions. Furthermore, in accordance to literature, our results suggest that glutamatergic neurotransmission plays an important role in the acquisition and consolidation of working memory processes.

Tethered bilayer lipid membranes provide an efficient, stable and versatile platform for the investigation of integrated membrane proteins. However, the incorporation of large proteins, as well as of proteins with a large submembrane part is still a very critical issue and therefore, further optimisation of the system is necessary. The central element of a tBLM is a lipid bilayer. Its proximal leaflet is, at least to some extend, covalently attached to a solid support via a spacer group. The anchor lipid consists of three distinct parts, a lipid headgroup, a spacer group and an anchor. All parts together influence the final bilayer properties. In the frame of this work, the synthesis of new thiolipids for tBLMs on gold has been investigated. The aim was to obtain molecules with longer spacers in order to increase the submembrane space. The systems obtained have been characterized using SPR and EIS. The results obtained during this study are multiple. First, the synthesis of a previously synthesized architecture was successfully scaled up in an industrial lab using a new synthetic approach. The synthesis of large amounts is now feasible. Then, the synthesis of the new thiolipids was carried out taking into account the following requirements: the increase of the submembrane space by having longer ethyleneglycol spacers, the attachment of the molecules to a gold substrate via a thiol bond, and the tunability of the lateral mobility by changing the lipid headgroup. Three different synthetic strategies have been investigated. The polymeric approach did not prove to be successful, merely because of the broad molecular weight distribution. The synthesis of heterofunctionally protected oligoethyleneglycols allowed to obtain ethyleneglycol moieties with 6 and 8 units, but the tedious purification steps gave very low yields. Finally, the block by block synthesis using ethyleneglycol precursors proved to be an efficient and fast method to synthesize the target molecules. Indeed, these were obtained with very high yields, and the separation was very efficient. A whole family of new compounds was obtained, having 6, 8 and 14 ethyleneglycol units and with mono- or diphytanyl lipid headgroups. This new pathway is a very promising synthetic strategy that can be used further in the development of new compounds of the tether system. The formation of bilayers was investigated for the different thiolipids mainly by using EIS. The electrical properties of a bilayer define the quality of the membrane and allow the study of the functionality of proteins embedded in such a system. Despite multiple trials to improve the system using self assembly, Langmuir Blodgett transfer, and detergent mixed vesicles, the new polymer thiolipids did not show as high electrical properties as tBLMs reported in the literature. Nevertheless, it was possible to show that a bilayer could be obtained for the different spacer lengths. These bilayers could be formed using self assembly for the first monolayer, and two different methods for bilayer formation, namely vesicle fusion and solvent exchange. We could furthermore show functional incorporation of the ion carrier valinomycin: the selective transport of K+ ions could be demonstrated. For DPHL, it was even possible to show the functional incorporation of the ion channel gramicidin. The influence of the spacer length is translated into an increase of the spacer capacitance, which could correspond to an increase in the capacity of charge accumulation in the submembrane space. The different systems need to be further optimised to improve the electrical properties of the bilayer. Moreover, the incorporation of larger proteins, and proteins bearing submembrane parts needs to be investigated.

In der vorliegenden Arbeit wurden Materialien und Aufbauten für Hybrid Solarzellen entwickelt und erforscht. rnDer Vergleich zweier bekannter Lochleitermaterialien für Solarzellen in einfachen Blend-Systemen brachte sowohl Einsicht zur unterschiedlichen Eignung der Materialien für optoelektronische Bauelemente als auch neue Erkenntnisse in Bereichen der Langzeitstabilität und Luftempfindlichkeit beider Materialien.rnWeiterhin wurde eine Methode entwickelt, um Hybrid Solarzelle auf möglichst unkomplizierte Weise aus kostengünstigen Materialien darzustellen. Die „Eintopf“-Synthese ermöglicht die unkomplizierte Darstellung eines funktionalen Hybridmaterials für die optoelektronische Anwendung. Mithilfe eines neu entwickelten amphiphilen Blockcopolymers, das als funktionelles Templat eingesetzt wurde, konnten mit einem TiO2-Precursor in einem Sol-Gel Ansatz verschiedene selbstorganisierte Morphologien des Hybridmaterials erhalten werden. Verschiedene Morphologien wurden auf ihre Eignung in Hybrid Solarzellen untersucht. Ob und warum die Morphologie des Hybridsystems die Effizienz der Solarzelle beeinflusst, konnte verdeutlicht werden. Mit der Weiterentwicklung der „Eintopf“-Synthese, durch den Austausch des TiO2-Precursors, konnte die Solarzelleneffizienz von 0.15 auf 0.4 % gesteigert werden. Weiterhin konnte die Übertragbarkeit des Systems durch den erfolgreichen Austausch des Halbleiters TiO¬2 mit ZnO bewiesen werden.rn

The increase in aquaculture operations worldwide has provided new opportunities for the transmission of aquatic viruses. The occurrence of viral diseases remains a significant limiting factor in aquaculture production and for the sustainability. The ability to identify quickly the presence/absence of a pathogenic organism in fish would have significant advantages for the aquaculture systems. Several molecular methods have found successful application in fish pathology both for confirmatory diagnosis of overt diseases and for detection of asymptomatic infections. However, a lot of different variants occur among fish host species and virus strains and consequently specific methods need to be developed and optimized for each pathogen and often also for each host species. The first chapter of this PhD thesis presents a complete description of the major viruses that infect fish and provides a relevant information regarding the most common methods and emerging technologies for the molecular diagnosis of viral diseases of fish. The development and application of a real time PCR assay for the detection and quantification of lymphocystivirus was described in the second chapter. It showed to be highly sensitive, specific, reproducible and versatile for the detection and quantitation of lymphocystivirus. The use of this technique can find multiple application such as asymptomatic carrier detection or pathogenesis studies of different LCDV strains. The third chapter, a multiplex RT-PCR (mRT-PCR) assay was developed for the simultaneous detection of viral haemorrhagic septicaemia (VHS), infectious haematopoietic necrosis (IHN), infectious pancreatic necrosis (IPN) and sleeping disease (SD) in a single assay. This method was able to efficiently detect the viral RNA in tissue samples, showing the presence of single infections and co-infections in rainbow trout samples. The mRT-PCR method was revealed to be an accurate and fast method to support traditional diagnostic techniques in the diagnosis of major viral diseases of rainbow trout.

Ziel dieser Arbeit war der Aufbau und Einsatz des Atmosphärischen chemischen Ionisations-Massenspektrometers AIMS für boden- und flugzeuggetragene Messungen von salpetriger Säure (HONO). Für das Massenspektrometer wurden eine mit Gleichspannung betriebene Gasentladungsionenquelle und ein spezielles Druckregelventil entwickelt. Während der Instrumentenvergleichskampagne FIONA (Formal Intercomparisons of Observations of Nitrous Acid) an einer Atmosphären-Simulationskammer in Valencia (Spanien) wurde AIMS für HONO kalibriert und erstmals eingesetzt. In verschiedenen Experimenten wurden HONO-Mischungsverhältnisse zwischen 100 pmol/mol und 25 nmol/mol erzeugt und mit AIMS interferenzfrei gemessen. Innerhalb der Messunsicherheit von ±20% stimmen die massenspektrometrischen Messungen gut mit den Methoden der Differenziellen Optischen Absorptions-Spektrometrie und der Long Path Absorption Photometrie überein. Die Massenspektrometrie kann somit zum schnellen und sensitiven Nachweis von HONO in verschmutzter Stadtluft und in Abgasfahnen genutzt werden.rnErste flugzeuggetragene Messungen von HONO mit AIMS wurden 2011 bei der Messkampagne CONCERT (Contrail and Cirrus Experiment) auf dem DLR Forschungsflugzeug Falcon durchgeführt. Hierbei konnte eine Nachweisgrenze von < 10 pmol/mol (3σ, 1s) erreicht werden. Bei Verfolgungsflügen wurden im jungen Abgasstrahl von Passagierflugzeugen molare HONO zu Stickoxid-Verhältnisse (HONO/NO) von 2.0 bis 2.5% gemessen. HONO wird im Triebwerk durch die Reaktion von NO mit OH gebildet. Ein gemessener abnehmender Trend der HONO/NO Verhältnisse mit zunehmendem Stickoxid-Emissionsindex wurde bestätigt und weist auf eine OH Limitierung im jungen Abgasstrahl hin.rnNeben den massenspektrometrischen Messungen wurden Flugzeugmessungen der Partikelsonde Forward Scattering Spectrometer Probe FSSP-300 in jungen Kondensstreifen ausgewertet und analysiert. Aus den gemessenen Partikelgrößenverteilungen wurden Extinktions- und optische Tiefe-Verteilungen abgeleitet und für die Untersuchung verschiedener wissenschaftlicher Fragestellungen, z.B. bezüglich der Partikelform in jungen Kondensstreifen und ihrer Klimawirkung, zur Verfügung gestellt. Im Rahmen dieser Arbeit wurde der Einfluss des Flugzeug- und Triebwerktyps auf mikrophysikalische und optische Eigenschaften von Kondensstreifen untersucht. Unter ähnlichen meteorologischen Bedingungen bezüglich Feuchte, Temperatur und stabiler thermischer Schichtung wurden 2 Minuten alte Kondensstreifen der Passagierflugzeuge vom Typ A319-111, A340-311 und A380-841 verglichen. Im Rahmen der Messunsicherheit wurde keine Änderung des Effektivdurchmessers der Partikelgrößenverteilungen gefunden. Hingegen nehmen mit zunehmendem Flugzeuggewicht die Partikelanzahldichte (162 bis 235 cm-3), die Extinktion (2.1 bis 3.2 km-1), die Absinktiefe des Kondensstreifens (120 bis 290 m) und somit die optische Tiefe der Kondensstreifen (0.25 bis 0.94) zu. Der gemessene Trend wurde durch Vergleich mit zwei unabhängigen Kondensstreifen-Modellen bestätigt. Mit den Messungen wurde eine lineare Abhängigkeit der totalen Extinktion (Extinktion mal Querschnittsfläche des Kondensstreifens) vom Treibstoffverbrauch pro Flugstrecke gefunden und bestätigt.

Sex determination in forensic practice is performed mostly on sexually dimorphic bones, including pelvic bones such as the os sacrum. Postmortem CT scan provides an easy and fast method for depicting and measuring bone structures prior to elaborate autopsy preparations. To develop a simple and objective method for sex determination in postmortem CT, metric data were evaluated from CT images of the pelvic-associated os sacrum of 95 corpses (49 men and 46 women) from the Canton of Bern, Switzerland. Discriminant function analysis of the data showed that the best accuracy in determining sex was 76.8% and 78.9% with two different observers. It is concluded that measuring the os sacrumin postmortem CT for sex determination has moderate accuracy and should only be applied in combination with other methods.

BACKGROUND Streptococcus spp. and other Gram-positive, catalase-negative cocci (PNC) form a large group of microorganisms which can be found in the milk of cows with intramammary infection. The most frequently observed PNC mastitis pathogens (major pathogens) are Streptococcus uberis, Strep. dysgalactiae, and Strep. agalactiae. The remaining PNC include a few minor pathogens and a large nonpathogenic group. Improved methods are needed for the accurate identification and differentiation of PNC. A total of 151 PNC were collected from cows with intramammary infection and conclusively identified by 16S rRNA sequencing as reference method. Nine phenotypic microbiological tests (alpha-hemolysis, CAMP reaction, esculin hydrolysis, growth on kanamycin esculin azide agar and on sodium chloride agar, inulin fermentation, hippurate hydrolysis, leucine aminopeptidase and pyrrolidonyl peptidase activity), multiplex PCR for the three major pathogens (target genes for Strep. uberis, Strep. dysgalactiae and Strep. agalactiae: pauA, 16S rRNA, and sklA3, respectively), and mass spectroscopy using the matrix-assisted laser desorption ionization-time of flight (MALDI-TOF MS) were evaluated for the diagnosis and discrimination of the three clinically most relevant PNC. RESULTS The probability that a strain of Strep. uberis, Strep. dysgalactiae and Strep. agalactiae was correctly identified by combining the results of the 9 phenotypic tests was 92%, 90%, and 100%, respectively. Applying the multiplex PCR, all strains of the three major pathogens were correctly identified and no false positive results occurred. Correct identification was observed for all strains of Strep. uberis and Strep. agalactiae using MALDI-TOF MS. In the case of Strep. dysgalactiae, some variability was observed at the subspecies level, but all strains were allocated to one single cluster. CONCLUSIONS The results of the present study show that reliable identification of the clinically most relevant PNC (Strep. uberis, Strep. agalactiae and Strep. dysgalactiae) can be obtained by use of a combination of colony morphology, hemolysis type and catalase reaction, and a multiplex PCR with specific primers restricted to these 3 pathogens. The MALDI-TOF MS is a fast method that shows promising results, although identification of Strep. dysgalactiae at the subspecies level is not yet satisfactory.

Models of DNA sequence evolution and methods for estimating evolutionary distances are needed for studying the rate and pattern of molecular evolution and for inferring the evolutionary relationships of organisms or genes. In this dissertation, several new models and methods are developed.^ The rate variation among nucleotide sites: To obtain unbiased estimates of evolutionary distances, the rate heterogeneity among nucleotide sites of a gene should be considered. Commonly, it is assumed that the substitution rate varies among sites according to a gamma distribution (gamma model) or, more generally, an invariant+gamma model which includes some invariable sites. A maximum likelihood (ML) approach was developed for estimating the shape parameter of the gamma distribution $(\alpha)$ and/or the proportion of invariable sites $(\theta).$ Computer simulation showed that (1) under the gamma model, $\alpha$ can be well estimated from 3 or 4 sequences if the sequence length is long; and (2) the distance estimate is unbiased and robust against violations of the assumptions of the invariant+gamma model.^ However, this ML method requires a huge amount of computational time and is useful only for less than 6 sequences. Therefore, I developed a fast method for estimating $\alpha,$ which is easy to implement and requires no knowledge of tree. A computer program was developed for estimating $\alpha$ and evolutionary distances, which can handle the number of sequences as large as 30.^ Evolutionary distances under the stationary, time-reversible (SR) model: The SR model is a general model of nucleotide substitution, which assumes (i) stationary nucleotide frequencies and (ii) time-reversibility. It can be extended to SRV model which allows rate variation among sites. I developed a method for estimating the distance under the SR or SRV model, as well as the variance-covariance matrix of distances. Computer simulation showed that the SR method is better than a simpler method when the sequence length $L>1,000$ bp and is robust against deviations from time-reversibility. As expected, when the rate varies among sites, the SRV method is much better than the SR method.^ The evolutionary distances under nonstationary nucleotide frequencies: The statistical properties of the paralinear and LogDet distances under nonstationary nucleotide frequencies were studied. First, I developed formulas for correcting the estimation biases of the paralinear and LogDet distances. The performances of these formulas and the formulas for sampling variances were examined by computer simulation. Second, I developed a method for estimating the variance-covariance matrix of the paralinear distance, so that statistical tests of phylogenies can be conducted when the nucleotide frequencies are nonstationary. Third, a new method for testing the molecular clock hypothesis was developed in the nonstationary case. ^

Distributed target tracking in wireless sensor networks (WSN) is an important problem, in which agreement on the target state can be achieved using conventional consensus methods, which take long to converge. We propose distributed particle filtering based on belief propagation (DPF-BP) consensus, a fast method for target tracking. According to our simulations, DPF-BP provides better performance than DPF based on standard belief consensus (DPF-SBC) in terms of disagreement in the network. However, in terms of root-mean square error, it can outperform DPF-SBC only for a specific number of consensus iterations.

La finalidad de esta investigación se enmarca dentro de los estudios sobre hormigones de presas llevados a cabo en el Laboratorio Central de Estructuras y Materiales del CEDEX. En España se han diagnosticado 18 obras afectadas tanto por la reacción álcali-sílice de tipo rápido como por la de tipo lento. Dos de de las obras, fabricadas con áridos graníticos, no presentan signos de deterioro pero en laboratorio se han hallado productos expansivos. En las 16 presas españolas restantes, el hormigón de 10 de ellas estaba fabricado con áridos graníticos, de las cuales, 7 están afectadas por la reacción de tipo lento. Sin embargo, en las clasificaciones internacionales de las rocas potencialmente reactivas se establece habitualmente que las rocas graníticas son inocuas o de reactividad muy baja. En los casos puntuales encontrados en la literatura, la reactividad de este tipo de áridos se encuentra frecuentemente asociada al cuarzo microcristalino que contienen y ocasionalmente se asocia esta reactividad también al cuarzo deformado y/o microfisurado. En la tesis doctoral de Víctor Daniel Lanza Fernández, también realizada en el CEDEX, se han tratado los áridos de reacción rápida, dando una descripción detallada de los componentes que intervienen en este tipo de reacción y presentando un ensayo eficaz para su detección en el laboratorio. La investigación desarrollada en la presente tesis doctoral se ha centrado en los áridos de reacción lenta, mucho menos estudiados. A partir del estudio bibliográfico realizado sobre este tipo de reacción se ha detectado ciertas lagunas, basadas principalmente en la falta de un método de detección en el laboratorio para los áridos de reacción lenta. No se ha encontrado un procedimiento en el estudio petrográfico que sea fiable y cuantificable para este tipo de áridos. El ensayo acelerado de barras de mortero, que para los áridos de reacción rápida supone un método rápido y fiable, falla en la detección de los áridos de reacción lenta al aplicar los límites normalizados. Si bien en publicaciones recientes se ha propuesto la posibilidad de ampliar el tiempo de tratamiento hasta los 90 días, la duración del ensayo es más larga de lo deseado. Para resolver estas lagunas, se han tomado áridos de obras reales afectadas por la reacción álcali-sílice (en algún caso también de las canteras de donde se extrajeron los áridos para la ejecución), con lo que la reactividad de estos áridos queda demostrada por su comportamiento en obra. Al objeto de aumentar la muestra de ensayo y utilizar también áridos inocuos, se han tomado muestras de canteras, tanto en uso como abandonadas. Sobre estos áridos se ha realizado una caracterización completa con los ensayos actualmente disponibles para el estudio de su reactividad con los álcalis del cemento: estudio petrográfico, ensayo acelerado de barras de mortero y Gel-Pat modificado. En la investigación se ha desarrollado una metodología de tinción de geles álcali-sílice para la cuantificación de los mismos en el interior de las barras de mortero. Se ha relacionado el volumen de gel generado en la reacción con la expansión producida, tanto para áridos lentos, como rápidos, estableciendo analogías y diferencias en el comportamiento de ambos. La cuantificación de este tipo de compuestos realizada en las barras de mortero, abre la posibilidad de estudiar la capacidad expansiva que presentan en testigos de hormigón de obras afectadas. Este dato podría ser una herramienta importante para el desarrollo de modelos matemáticos que puedan predecir el futuro comportamiento de las estructuras afectadas por la reacción álcali-sílice. La tinción ha sido asimismo utilizada para establecer un método de detección de áridos reactivos basado en el ensayo de Gel-Pat modificado. La metodología de detección propuesta para estos áridos es cuantitativa y proporciona los resultados en 14 días, suponiendo una ventaja importante sobre los métodos actuales, que permiten detectarlos en 90 días (de forma orientativa en 56 días). This research is part of the studies on durability of concrete dams carried out in the Laboratorio Central de Estructuras y Materiales of CEDEX during the last years. At the present time, 18 public works affected by alkali-silica reaction have been diagnosed in Spain. In 12 of them the concrete was mixed with granitic aggregates, 7 of which were damaged by the slow alkali-silica reaction, 3 by the rapid alkali-silica reaction and in the other 2 cases, although there was no visual evidence of the reaction in field, the laboratory tests showed the presence of expansive products inside the concrete. However, in the international classifications of potentially reactive aggregates granitic rocks are pointed out as innocuous or as low reactivity and usually their reactivity is attributed to the presence of microcrystalline quartz and occasionally with the strained and microcracked quartz. The rapid reactive aggregates were deeply studied in a previous research (Víctor Daniel Lanza’s Thesis) also carried out in CEDEX. In this research, a detailed description of the components involved in the rapid alkali-silica reaction was given. Also, a fast method to detect them in the laboratory was developed. The research of the present PhD Thesis is focused in the slow-reactive aggregates, much less studied. The state of the art has shown some gaps in the knowledge about this reaction, mainly the absence of a reliable method to detect slow-reactive aggregates. On one side, there is no systematic and quantitative petrographic test for these aggregates. On the other side, the accelerated mortar bar test has been proved to be effective to detect rapid reactive aggregates but the standard limits fail in the detection of the slowreactive aggregates. In recent investigations it has been proposed extending the test until 90 days, but this period is considered too long. In this research, aggregates taken from affected structures have been used in the test, so their reactivity has been shown in practice. With the aim of increasing the number of samples aggregates from quarries have been also used. A method to stain alkali-silica gels has been applied to mortar bars, thereby quantifying the volume of gel generated inside. This volume has been related to the expansion of the bars, both for rapid for slow reactive aggregates, establishing similarities and differences. The quantification of the gel volume, applied to concrete cores extracted from affected structures, could be an important tool in mathematical models to predict the future behaviour of the structures affected by the alkali-silica reaction. The staining method has been also used to develop a method to detect slow reactive aggregates, based on the optimised Gel-Pat test, obtaining results in 14 days. This represents a major improvement compared to the actual methods (accelerated mortar bar test) which give an indicative value of reactivity at 56 days, and a final result at 90 days.
