Dietary antioxidants can affect cellular processes relevant to chronic inflammatory diseases such as atherosclerosis. We have used non- standard techniques to quantify effects of the antioxidant soy isoflavones genistein and daidzein on translocation of Nuclear Factor-KB (NF-KB) and nitric oxide (NO) production, which are important in these diseases. Translocation was quantified using confocal immunofluoresecence microscopy and ratiometric image analysis. NO was quantified by an electrochemical method after reduction of its oxidation products in cell culture supernatants. Activation of the RAW 264.7 murine monocyte/macrophage cell line increased the ratio of nuclear to cytoplasmic immunostaining for NF-kB. The increase was exacerbated by pre-treatment with genistein or daidzein. To show that decreases could also be detected, pre-treatment with the pine bark extract Pycnogenol (R) r was examined, and found to reduce translocation. NO production was also increased by activation, but was reduced by pre-treatment with genistein or daidzein. In the EA. hy926 human endothelial cell line, constitutive production was detectable and was increased by thrombin. The confocal and electrochemical methods gave data that agreed with results obtained using the established electromobility shift and Griess assays, but were more sensitive, more convenient, gave more detailed information and avoided the use of radioisotopes.

A simple and highly selective electrochemical method was developed for the single or simultaneous determination of paracetamol (N-acetyl-p-aminophenol, acetaminophen) and caffeine (3,7-dihydro-1,3,7-trimethyl-1H-purine-2,6-dione) in aqueous media (acetate buffer, pH 4.5) on a boron-doped diamond (BDD) electrode using square wave voltammetry (SWV) or differential Pulse voltammetry (DPV). Using DPV with the cathodically pre-treated BDD electrode, a separation of about 550 mV between the peak oxidation potentials Of paracetamol and caffeine present in binary mixtures was obtained. The calibration curves for the simultaneous determination of paracetamol and caffeine showed an excellent linear response, ranging from 5.0 x 10(-7) mol L(-1) to 8.3 x 10(-7) mol L(-1) for both compounds. The detection limits for the simultaneous determination of paracetamol and caffeine were 4.9 x 10(-7) mol L-1 and 3.5 x 10(-8) mol L(-1), respectively. The proposed method Was Successfully applied in the simultaneous determination of paracetamol and caffeine in several pharmaceutical formulations (tablets), with results similar to those obtained using a high-performance liquid chromatography method (at 95% confidence level). In general, among the corrosion inhibitors surfactants are the most commonly used compounds, because they are significantly effective by forming protective films on anodic and cathodic areas. In this study, microemulsions containing he biodegradable saponified coconut oil as surfactant (SME-OCS) was used as green corrosion inhibitors. With this purpose, methanolic extracts of Ixora coccinea Linn (IC) and a polar fraction rich in alkaloids (FA) obtained from Croton cajucara Benth solubilized in the SME-OCS system were examined in the presence of AISI 1020 carbon steel, in saline solution (NaCl 3,5 %). The efficiency of corrosion inhibition of IC and FA were evaluated in the following microemulsions: SME-OCS-IC and SME-OCS-FA. The microemulsion system SME-OCS in the presence and absence of IC and FA was assessed by measurements of weight loss and the electrochemical method of polarization resistance, with variation in the concentration of IC and FA (50 - 400 ppm), showing significant results of corrosion inhibition (83,6 % SME-OCS; 92,2 % SME-OCS-FA; and 95,3 % SME-OCS-IC)

The construction industry is one of the largest consumers of natural raw materials, and concrete is considered today the most used material wide. This accentuated consumption of natural resources has generated concern with the preservation of the environment, and has motivated various studies related to the use of resid ues, which can partially or entirely substitute, with satisfactory performance, some materials such as the aggregate, and in so doing, decrease the impact on the environment caused by the produced residues. Research has been done to better understand and improve the microstructure of concrete, as well as to understand the mechanism of corrosion in reinforced steel. In this context, this work was developed aiming at discovering the influence of the substitution of natural sand by artificial sand, with rega rd to mechanical resistance, microstructure, and durability. To obtain the electrochemical parameters, an adaptation was made to the galvanostatic electrochemical method to study the corrosion in reinforced steel. Concretes of categories 20 MPa and 40 MPa were produced, containing natural sand, and concretes of the same categories were produced with artificial sand substituting the natural sand, and with the addition of sodium nitrate and sodium chloride. Due to the use of rock dust reject (artificial sand), an evaluation was made of its environmental risk. The results indicate that the concretes of category 20 MPa present a better performance than the concrete made with natural sand, thus making it a viable substitute. For the category 40 MPa, the better performance is from the concrete containing natural sand. The adaptation of the galvanostatic electrochemical technique to the study of the corrosion of reinforced steel within concrete proved to be valid for obtaining electrochemical parameters with a high degree of reliability, considering the number of degrees of freedom

This work is directed to the treatment of organic compounds present in produced water from oil using electrochemical technology. The water produced is a residue of the petroleum industry are difficult to treat , since this corresponds to 98 % effluent from the effluent generated in the exploration of oil and contains various compounds such as volatile hydrocarbons (benzene, toluene, ethylbenzene and xylene), polycyclic aromatic hydrocarbons (PAHs), phenols, carboxylic acids and inorganic compounds. There are several types of treatment methodologies that residue being studied, among which are the biological processes, advanced oxidation processes (AOPs), such as electrochemical treatments electrooxidation, electrocoagulation, electrocoagulation and eletroredution. The electrochemical method is a method of little environmental impact because instead of chemical reagents uses electron through reactions of oxide-reducing transforms toxic substances into substances with less environmental impact. Thus, this paper aims to study the electrochemical behavior and elimination of the BTX (benzene, toluene and xylene) using electrode of Ti/Pt. For the experiment an electrochemical batch system consists of a continuous source, anode Ti/Pt was used, applying three densities of current (1 mA/cm2, 2,5 mA/cm2 and 5 mA/cm2). The synthetic wastewater was prepared by a solution of benzene, toluene and xylene with a concentration of 5 ppm, to evaluate the electrochemical behavior by cyclic voltammetry and polarization curves, even before assessing the removal of these compounds in solution by electrochemical oxidation. The behavior of each of the compounds was evaluated by the use of electrochemical techniques indicate that each of the compounds when evaluated by cyclic voltammetry showed partial oxidation behavior via adsorption to the surface of the Ti/Pt electrode. The adsorption of each of the present compounds depends on the solution concentration but there is the strong adsorption of xylene. However, the removal was confirmed by UV-Vis, and analysis of total organic carbon (TOC), which showed a percentage of partial oxidation (19,8 % - 99,1 % TOC removed), confirming the electrochemical behavior already observed in voltammetry and cyclic polarization curves

The electrochemical behaviour of alanine and phenylalanine protected by the toluenesulphonyl (tosyl) and nitrobenzesulphonyl (nosyl) groups was examined in dimethyl formamide. The N-tosyl-protected amino acids are reduced in one cathodic step (-2.4 V/SCE) leading to cleavage of the SN bond after a two-electron transfer process. The N-nosyl-protected amino acids are reduced in three cathodic steps. Cyclic voltammetry and controlled potential electrolysis of N-nosyl amino acids have been used to probe the mechanism of these reductions in an aprotic medium and lead to cleavage of the SN bond in good yields ( > 60%) at -1.10 V/SCE. The comparative study of these compounds is used to comment on the applicability of the electrochemical method of removing protecting groups from amino acids. © 1993.

This report shows an unexpected toxicity decrease during atrazine photoelectrodegradation in the presence of NaCl. Atrazine is a pesticide classified as endocrine disruptor occurring in industrial effluents and agricultural wastewaters. We therefore studied the effects of the degradation method, electrochemical and electrochemical photo-assisted, and of the supporting electrolyte, NaCl and Na2SO4, on the residual toxicity of treated atrazine solutions. We also studied the toxicity of treated atrazine solutions using Results show that at initial concentration of 20 mg L-1, atrazine was completely removed in up to 30 min using 10 mA cm(-2) electrolysis in NaCl medium, regardless of the electrochemical method used. The total organic carbon removal by the photo-assisted method was 82% with NaCl and 95% with Na2SO4. The solution toxicity increased during sole electrochemical treatment in NaCl, as expected. However, the toxicity unexpectedly decreased using the photo-assisted method. This finding is a major discovery because electrochemical treatment with NaCl usually leads to the formation of toxic chlorine-containing organic degradation by-products.

In the present study, thin functional conducting polyaniline (PANI) films, either doped or undoped, patterned or unpatterned, were prepared by different approaches. The properties of the obtained PANI films were investigated in detail by a combination of electrochemistry with several other techniques, such as SPR, QCM, SPFS, diffraction, etc. The sensing applications (especially biosensing applications) of the prepared PANI films were explored. Firstly, the pure PANI films were prepared by the electropolymerisation method and their doping/dedoping properties in acidic conditions were investigated in detail by a combination of electrochemistry with SPR and QCM. Dielectric constants of PANI at different oxidation states were obtained quantitatively. The results obtained here laid a good foundation for the following investigations of PANI films in neutral pH conditions. Next, PANI multilayer films doped by a variety of materials were prepared by the layer-by-layer method in order to explore their biosensing applications, because of the loss of redox activity of pure PANI in neutral pH conditions. The dopants used include not only the traditionally used linear polyelectrolytes, but also, for the first tim, some other novel materials, like modified gold nanoparticles or modified carbon nanotubes. Our results showed that all the used dopants could form stable multilayer films with PANI. All the obtained PANI multilayer films showed good redox activity in a neutral pH environment, which makes them feasible for bioassays. We found that all the prepared PANI multilayer films can electrocatalyze the oxidation of NADH in neutral conditions at a low potential, although their catalytic efficiencies are different. Among them, PANI/carbon nanotube system showed the highest catalytic efficiency toward the oxidation of NADH, which makes it a good candidate as a NADH sensor. Besides, because some of the prepared PANI multilayer systems were end-terminated with –COOH groups (like PANI/Au nanoparticles system), which can be utilized to easily link biomolecules for biosensing applications. Here, we demonstrated, for the first time, to use the prepared PANI multilayer films for the DNA hybridisation detection. The detection event was monitored either by direct electrochemical method, or by enzyme-amplified electrochemical method, or by surface plasmon enhanced fluorescence spectroscopic method. All the methods can effectively differentiate non-complementary DNA from the complementary ones, even at the single-base mismatch level. It should also be noted that, our success in fabricating PANI multilayer films with modified Au nanoparticles or carbon nanotubes also offered another novel method for incorporating such novel materials into (conducting) polymers. Because of the unique electrochemical and optical properties of each component of the obtained PANI multilayer films, they should also find potential applications in many other fields such as microelectronics, or for electrochromic and photovoltaic devices. Finally, patterned PANI films were fabricated by the combination of several patterning techniques, such as the combination of electrocopolymerization with micromolding in capillaries (EP-MIMIC), the combination of microcontact printing with the layer-by-layer technique (µCP-LBL), and the polystyrene (PS) template induced electropolymerisation method. Using the obtained stripe-shaped PANI/PSS film, a redox-switchable polymer grating based on the surface-plasmon-enhanced mode was constructed and its application in the field of biosensing was explored. It was found that the diffraction efficiency (DE) of the grating was very sensitive to the applied potential (i.e. redox state of the film) as well as the pH environment of the dielectric medium. Moreover, the DE could also be effectively tuned by an electrocatalytic event, such as the electrocatalytic oxidation of NADH by the grating film. By using PS colloidal crystal assemblies as templates, well-ordered 3D interconnected macroporous PANI arrays (PANI inverse opals) were fabricated via electropolymerisation method. The quality of the obtained inverse opals was much higher than those reported by chemical synthesis method. By electrochemical method, the structures of the prepared inverse opals can be easily controlled. To explore the possible biosensing applications of PANI inverse opals, efforts were also done toward the fabrication of PANI composite inverse opals. By selecting proper dopants, high quality inverse opals of PANI composites were fabricated for the first time. And the obtained opaline films remained redox-active in neutral pH conditions, pointing to their possible applications for electrobioassays.

The aim of the present thesis, carried out at the Analytical Group of the Faculty of Industrial Chemistry in Bologna, is to develop a new electrochemical method for the determination of the Antioxidant Capacity (AOC). The approach is based on the deposition of a non-conducting polymeric film on the working electrode surface and the following exposition to the radicals OH· produced by H2O2 photolysis. The strongly oxidant action of hydroxyl radicals degrades, causing an increase of the Faradic current, relevant to the redox couple [Ru(NH3)6]2+/3+ monitored by cyclic voltammetry(CV); the presence of an antioxidant compound in solution slows down the radical action, thus protecting the polymeric film and blocking the charge transfer. The parameter adopted for the quantification of the AOC, was the induction time, called also lag phase, which is the time when the degradation of the film starts. Five pure compounds, among most commonly antioxidant, were investigated : Trolox®(an analogue water-soluble of vitamin E), (L)-ascorbic acid, gallic acid, pyrogallol and (-)- epicatechin. The AOC of each antioxidant was expressed by TEAC index (Trolox® Equivalent Antioxidant Capacity), calculated from the ratio between the slope of the calibration curve of the target compound and the slope of the calibration curve of Trolox®. The results from the electrochemical method, have been compared with those obtained from some other standardized methods, widely employed. The assays used for the comparison, have been: ORAC, a spectrofluorimetric method based on the decrease of fluorescein emission after the attack of alkylperoxide radicals, ABTS and DPPH that exploit the decoloration of stable nitrogen radicals when they are reduced in presence of an antioxidant compound and, finally, a potentiometric method based on the response of the redox couple [Fe(CN)6]3-/ [Fe(CN)6]4-. From the results obtained from pure compounds, it has been found that ORAC is the methodology showing the best correlation with the developed electrochemical method, maybe since similar radical species are involved. The comparison between the considered assays, was also extended to the analysis of a real sample of fruit juice. In such a case the TEAC value resulting from the electrochemical method is higher than those from standardized assays.

Membranproteine spielen eine wichtige Rolle bei physiologischen Prozessen wie Signalweiterleitung oder Immunreaktion. Deshalb stehen sie im Fokus der pharmakologischen Wirkstoffentwicklung und es besteht großes Interesse, Membranproteinbasierte Biosensoren zu entwickeln, die sich z.B. als Screening-Plattformen eignen. Allerdings stellt die Handhabung von Membranproteinen wegen ihrer amphiphilen Struktur eine große Herausforderung dar. Membranproteine werden meist in Zellkultur oder in bakteriellen Expressionssystemen synthetisiert. Diese Verfahren liefern aber oft nur eine geringe Ausbeute und erlauben wenig Kontrolle über die Expressionsbedingungen. Als alternativer Ansatz bietet sich stattdessen die in vitro Synthese von Proteinen an, die in einer zellfreien Umgebung stattfindet. Ziel der vorliegenden Arbeit war die Etablierung eines miniaturisierten Analysesystems, das Aktivitätsmessungen an in vitro synthetisierten Ionenkanälen erlaubt. Dafür wurde ein Labon- Chip entwickelt, der elektrochemische und optische Nachweismethoden in parallelen Anätzen ermöglicht. Als amphiphile Umgebung für die Inkorporation von Membranproteinen wurden vier verschieden biomimetische Membranaufbauten hinsichtlich ihrer Dichtigkeit und ihrer Reproduzierbarkeit untersucht. Als Methode fanden insbesondere die Impedanzspektroskpie und die Oberflächenplasmonen-Resonanzspektroskopie Anwendung. Die peptide cushioned Bilyer Lipid Membranes (pcBLM) eignete sich dabei am besten für Untersuchungen an Membranproteinen. Zur Detektion der Ionenkanalaktivität wurde eine neue Messmethode etabliert, die auf der Messung der Impedanz bei fester Frequenz basiert und u.a. eine Aussage über die Änderung des Membranwiderstandes bei Aktivierung erlaubt. Am Beispiel des nicotinischen Acetylcholinrezeptors (nAchR) konnte gezeigt werden, dass sich die Aktivität von Ionenkanälen mit dem entwickelten Chip-System nachweisen ließ. Die Spezifität der Methode konnte durch verschiedene Kontrollen wie die Zugabe eines nicht-aktivierenden Liganden oder Inhibition des Rezeptors nachgewiesen werden. Weiterhin konnte die in vitro Synthese des Ionenkanals a7 nAchR durch Radioaktivmarkierung nachgewiesen werden. Die Inkorporation des Rezeptors in die biomimetischen Membranen wurde mit Immunodetektion und elektrochemischen Methoden untersucht. Es zeigte sich, dass die funktionelle Inkorporation des a7 nAchR davon abhing, welcher biomimetische Membranaufbau verwendet wurde.

Zusammenfassung der Dissertation, Carolin Edinger, April 2015. Im Rahmen der Dissertation ist eine effiziente und zuverlässige Methode zur elektrochemischen Desoxygenierung von aromatischen Carbonsäureamiden entwickelt worden (Schema 1).[1] Unter galvanostatischen Bedingungen eignet sich das optimierte Elektrolytsystem bestehend aus 2%iger methanolischer H2SO4 und geringen Mengen an Additiv 1 in Kombination mit einer Bleikathode hervorragend in dem gewählten geteilten Zellaufbau. Schema 1: Elektrochemische Desoxygenierung aromatischer Carbonsäureamide. Untersuchungen an verschiedensten Amidsubstraten haben gezeigt, dass ein breites Spektrum an Aminen mit dieser Methode zugänglich ist und durch umfangreiche Studien konnten optimale Elektrolyseparameter gefunden werden. Außerdem wurde die Hochskalierung der Ansatzgröße an einem Testsubstrat mit hohen Aminausbeuten von bis zu 73% gewährleistet. Ein besonderes Merkmal der entwickelten Synthese ist neben milden Bedingungen und hoher Selektivität die Verwendung von Ammoniumsalzadditiven. Der positive Effekt dieser Additive auf die Desoxygenierungsreaktion ist vielfältig: Die Wasserstoffentwicklung als unerwünschte Nebenreaktion wird zu negativeren Potentialen verschoben und die Bleikathode wird durch Zurückdrängung der PbSO4-Bildung effektiv vor Korrosion geschützt. Dies konnte durch experimentelle Werte wie die Erhöhung der Produkt- und Stromausbeute durch Additivzusatz während der Elektrolyse hinreichend bestätigt werden. Aber auch zyklovoltammetrische Untersuchungen und Lichtmikroskopaufnahmen der Elektrodenoberfläche bekräftigen eindeutig diese Aussagen.[2,3] Die entwickelte elektrochemische Methode konnte zusätzlich erfolgreich auf Verbindungen übertragen werden, die mit Carbonsäureamiden verwandt sind. So gelang es, aromatische und aliphatische Sulfoxide in sehr guten Ausbeuten selektiv zu den entsprechenden Sulfiden umzusetzen. Zusätzlich konnten bereits bei weiteren, durch klassische Methoden schwer reduzierbare Stoffklassen erste Erfolge erzielt werden. So gelang es, den Grundstein zur Reduktion von Estern und Triphenylphosphinoxid zu legen und erste, vielversprechende Ergebnisse zu erlangen. Da Elektronen als Reduktionsmittel eingesetzt werden und lediglich Wasser als Nebenprodukt gebildet wird, zeichnet sich die entwickelte Desoxygenierungsmethode vor allem durch milde Bedingungen und hohe Selektivität aus. Da weder Reagenzien noch Katalysatoren verwendet werden müssen, werden Abfälle vermieden. Dadurch ist die gefundene Reduktionsmethode nicht nur kostengünstig, sondern erweist sich auch in der Reaktionsführung als vorteilhaft. Literatur: [1] C. Edinger, S. R. Waldvogel, Eur. J. Org. The cyclic enzymatic function of a cytochrome P450, as it catalyzes the oxygen-dependent metabolism of many organic chemicals, requires the delivery of two electrons to the hemeprotein. In general these electrons are transferred from NADPH to the P450 via an FMN- and FAD-containing flavoprotein (NADPH-P450 reductase). The present paper shows that NADPH can be replaced by an electrochemically generated reductant [cobalt(II) sepulchrate trichloride] for the electrocatalytically driven omega-hydroxylation of lauric acid. Results are presented illustrating the use of purified recombinant proteins containing P450 4A1, such as the fusion protein (rFP450 [mRat4A1/mRatOR]L1) or a system reconstituted with purified P450 4A1 plus purified NADPH-P450 reductase. Rates of formation of 12-hydroxydodecanoic acid by the electrochemical method are comparable to those obtained using NADPH as electron donor. These results suggest the practicality of developing electrocatalytically dependent bioreactors containing different P450s as catalysts for the large-scale synthesis of stereo- and regio-selective hydroxylation products of many chemicals.
