The tripeptide glutathione (GSH) is one of the most abundant peptides and the major repository for nonprotein sulfur in both animal and plant cells. It plays a critical role in intracellular oxidative stress management by the reversible formation of glutathione disulfide with the thiol-disulfide pair acting as a redox buffer. The state of charge of the ionizable groups of GSH can influence the redox couple, and hence the pK(a) value of the cysteine residue of GSH is critical to its functioning. Here we report ab initio Car-Parrinello molecular dynamics simulations of glutathione solvated by 200 water molecules, all of which are considered in the simulation. We show that the free-energy landscape for the protonation-deprotonation reaction of the cysteine residue of GSH computed using metadynamics sampling provides shift in the dissociation constant values as compared with the isolated accurate estimates of the pK(a) and correctly predicts the cysteine amino acid.

Threefold symmetric Fe phosphine complexes have been used to model the structural and functional aspects of biological N2 fixation by nitrogenases. Low-valent bridging Fe-S-Fe complexes in the formal oxidation states Fe(II)Fe(II), Fe(II)/Fe(I), and Fe(I)/Fe(I) have been synthesized which display rich spectroscopic and magnetic behavior. A series of cationic tris-phosphine borane (TPB) ligated Fe complexes have been synthesized and been shown to bind a variety of nitrogenous ligands including N2H4, NH3, and NH2-. These complexes are all high spin S = 3/2 and display EPR and magnetic characteristics typical of this spin state. Furthermore, a sequential protonation and reduction sequence of a terminal amide results in loss of NH3 and uptake of N2. These stoichiometric transformations represent the final steps in potential N2 fixation schemes.

Treatment of an anionic FeN2 complex with excess acid also results in the formation of some NH3, suggesting the possibility of a catalytic cycle for the conversion of N2 to NH3 mediated by Fe. Indeed, use of excess acid and reductant results in the formation of seven equivalents of NH3 per Fe center, demonstrating Fe mediated catalytic N2 fixation with acids and protons for the first time. Numerous control experiments indicate that this catalysis is likely being mediated by a molecular species.

A number of other phosphine ligated Fe complexes have also been tested for catalysis and suggest that a hemi-labile Fe-B interaction may be critical for catalysis. Additionally, various conditions for the catalysis have been investigated. These studies further support the assignment of a molecular species and delineate some of the conditions required for catalysis.

Finally, combined spectroscopic studies have been performed on a putative intermediate for catalysis. These studies converge on an assignment of this new species as a hydrazido(2-) complex. Such species have been known on group 6 metals for some time, but this represents the first characterization of this ligand on Fe. Further spectroscopic studies suggest that this species is present in catalytic mixtures, which suggests that the first steps of a distal mechanism for N2 fixation are feasible in this system.

Three separate topics, each stimulated by experiments, are treated theoretically in this dessertation: isotopic effects of ozone, electron transfer at interfaces, and intramolecular directional electron transfer in a supramolecular system.

The strange mass-independent isotope effect for the enrichment of ozone, which has been a puzzle in the literature for some 20 years, and the equally puzzling unconventional strong mass-dependent effect of individual reaction rate constants are studied as different aspects of a symmetry-driven behavior. A statistical (RRKM-based) theory with a hindered-rotor transition state is used. The individual rate constant ratios of recombination reactions at low pressures are calculated using the theory involving (1) small deviation from the statistical density of states for symmetric isotopomers, and (2) weak collisions for deactivation of the vibrationally excited ozone molecules. The weak collision and partitioning among exit channels play major roles in producing the large unconventional isotope effect in "unscrambled" systems. The enrichment studies reflect instead the non-statistical effect in "scrambled" systems. The theoretical results of low-pressure ozone enrichments and individual rate constant ratios obtained from these calculations are consistent with the corresponding experimental results. The isotopic exchange rate constant for the reaction ^(16)O + ^(18)O ^(18)O→+ ^(16)O ^(18)O + ^(18)O provides information on the nature of a variationally determined hindered-rotor transition state using experimental data at 130 K and 300 K. Pressure effects on the recombination rate constant, on the individual rate constant ratios and on the enrichments are also investigated. The theoretical results are consistent with the experimental data. The temperature dependence of the enrichment and rate constant ratios is also discussed, and experimental tests are suggested. The desirability of a more accurate potential energy surface for ozone in the transition state region is also noted.

Electron transfer reactions at semiconductor /liquid interfaces are studied using a tight-binding model for the semiconductors. The slab method and a z-transform method are employed in obtaining the tight-binding electronic structures of semiconductors having surfaces. The maximum electron transfer rate constants at Si/viologen^(2-/+) and InP /Me_(2)Fc^(+/O) interfaces are computed using the tight-binding type calculations for the solid and the extended-Huckel for the coupling to the redox agent at the interface. These electron transfer reactions are also studied using a free electron model for the semiconductor and the redox molecule, where Bardeen's method is adapted to calculate the coupling matrix element between the molecular and semiconductor electronic states. The calculated results for maximum rate constant of the electron transfer from the semiconductor bulk states are compared with the experimentally measured values of Lewis and coworkers, and are in reasonable agreement, without adjusting parameters. In the case of InP /liquid interface, the unusual current vs applied potential behavior is additionally interpreted, in part, by the presence of surface states.

Photoinduced electron transfer reactions in small supramolecular systems, such as 4-aminonaphthalimide compounds, are interesting in that there are, in principle, two alternative pathways (directions) for the electron transfer. The electron transfer, however, is unidirectional, as deduced from pH-dependent fluorescence quenching studies on different compounds. The role of electronic coupling matrix element and the charges in protonation are considered to explain the directionality of the electron transfer and other various results. A related mechanism is proposed to interpret the fluorescence behavior of similar molecules as fluorescent sensors of metal ions.

The electrochemical and electrocatalytic behavior of a series of heteropolytungstate anions in which a tungsten atom in the well known Keggin structure has been replaced by an iron atom is described. All of the iron substituted ions exhibit a one electron reversible couple associated with the Fe3+ center and a pair of two electron waves attributed to electron addition and removal from the tungsten oxo framework. The pH and ionic strength effects upon the various electrochemical processes are discussed and interpreted in terms of a competition between protonation and ion pairing of the highly negatively charged ions.

The anions are efficient catalysts for the electroreduction of hydrogen peroxide. A catalytic mechanism involving a formally Fe(IV) intermediate is proposed. Pulse radiolysis experiments were employed to detect the intermediate and evaluate the rate constants for the reactions in which it is formed and decomposed. A chain mechanism for the catalytic decomposition of hydrogen peroxide in which the Fe center shuttles between the +2, +3, and +4 oxidation states is proposed to explain the non-integral stoichiometry observed for the iron substituted polytungstate catalyzed electroreduction of hydrogen peroxide.

The anions are also efficient electrocatalyst for the electrochemical conversion of nitric oxide to ammonia. The catalyzed reduction does not produce hydroxylamine as an intermediate and appears to depend upon the ability of the multiply reduced heteropolytungstates to deliver several electrons to the bound NO group in a concerted step. A valuable feature of the heteropolytungstates is the ease at which the formal potentials of the several redox couples they exhibit may be shifted by changing the identity of the central heteroatom. Exploitation of this feature provided diagnostic information that was decisive in establishing the mechanism of electrocatalytic reduction.

The iron substituted heteropolytungstates are not degraded by repeated cycling between their oxidized and reduced states. They also show superior activity compared to their unsubstituted analogues, indicating that the Fe center acts as a binding site that facilitates inner-sphere electron transfer processes. The basic electrochemistry of several other transition metal substituted Keggin ions is also described.

The subject of this thesis is electronic coupling in donor-bridge-acceptor systems. In Chapter 2, ET properties of cyanide-bridged dinuclear ruthenium complexes were investigated. The strong interaction between the mixed-valent ruthenium centers leads to intense metal-to-metal charge transfer bands (MMCT). Hush analysis of the MMCT absorption bands yields the electronic-coupling strength between the metal centers (H_(AB)) and the total reorganization energy (λ). Comparison of ET kinetics to calculated rates shows that classical ET models fail to account for the observed kinetics and nuclear tunneling must be considered.

In Chapter 3, ET rates were measured in four ruthenium-modified highpotential iron-sulfur proteins (HiPIP), which were modified at position His50, His81, His42 and His18, respectively. ET kinetics for the His50 and His81 mutants are a factor of 300 different, while the donor-acceptor separation is nearly identical. PATHWAY calculations corroborate these measurements and highlight the importance of structural detail of the intervening protein matrix.

In Chapter 4, the distance dependence of ET through water bridges was measured. Photoinduced ET measurements in aqueous glasses at 77 K show that water is a poor medium for ET. Luminescence decay and quantum yield data were analyzed in the context of a quenching model that accounts for the exponential distance dependence of ET, the distance distribution of donors and acceptors embedded in the glass and the excluded volumes generated by the finite sizes of the donors and acceptors.

In Chapter 5, the pH-dependent excited state dynamics of ruthenium-modified amino acids were measured. The [Ru(bpy)_(3)] ^(2+) chromophore was linked to amino acids via an amide linkage. Protonation of the amide oxygen effectively quenches the excited state. In addition. time-resolved and steady-state luminescence data reveal that nonradiative rates are very sensitive to the protonation state and the structure of the amino acid moiety.

Tryptophan and unnatural tryptophan derivatives are important building blocks for the total synthesis of natural products, as well as the development of new drugs, biological probes, and chiral small molecule catalysts. This thesis describes various catalytic methods for the preparation of tryptophan derivatives as well as their functionalization and use in natural product total synthesis.

Herein, the tandem Friedel–Crafts conjugate addition/asymmetric protonation reaction between 2-substituted indoles and methyl 2-acetamidoacrylate to provide enantioenriched trytophans is reported. This method inspired further work in the area of transition metal catalyzed arylation reactions. We report the development of the coppercatalyzed arylation of tryptamine and tryptophan derivatives. The utility of these transformations is highlighted in the five-step syntheses of the natural products (+)-naseseazine A and B. Further work on the development of a mild and general Larock indolization protocol to access unnatural tryptophans is also discussed.

The activation of Fe-coordinated N2 via the formal addition of hydrogen atom equivalents is explored in this thesis. These reactions may occur in nitrogenase enzymes during the biological conversion of N2 to NH3. To understand these reactions, the N2 reactivity of a series of molecular Fe(N2) platforms is investigated. A trigonal pyramidal, carbon-ligated FeI complex was prepared that displays a similar geometry to that of the resting state 'belt' Fe atoms of nitrogenase. Upon reduction, this species was shown to coordinate N2, concomitant with significant weakening of the C-Fe interaction. This hemilability of the axial ligand may play a critical role in mediating the interconversion of Fe(NxHy) species during N2 conversion to NH3. In fact, a trigonal pyramidal borane-ligated Fe complex was shown to catalyze this transformation, generating up to 8.49 equivalents of NH3. To shed light on the mechanistic details of this reaction, protonation of a borane-ligated Fe(N2) complex was investigated and found to give rise to a mixture of species that contains an iron hydrazido(2-) [Fe(NNH2)] complex. The identification of this species is suggestive of an early N-N bond cleavage event en route to NH3 production, but the highly-reactive nature of this complex frustrated direct attempts to probe this possibility. A structurally-analogous silyl-ligated Fe(N2) complex was found to react productively with hydrogen atom equivalents, giving rise to an isolable Fe(NNH2) species. Spectroscopic and crystallographic studies benefited from the enhanced stability of this complex relative to the borane analogue. One-electron reduction of this species initiates a spontaneous disproportionation reaction with an iron hydrazine [Fe(NH2NH2)] complex as the predominant reaction product. This transformation provides support for an Fe-mediated N2 activation mechanism that proceeds via a late N-N bond cleavage. In hopes of gaining more fundamental insight into these reactions, a series of Fe(CN) complexes were prepared and reacted with hydrogen-atom equivalents. Significant quantities of CH4 and NH3 are generated in these reactions as a result of complete C-N bond activation. A series of Fe(CNHx) were found to be exceptionally stable and may be intermediates in these reactions. The stability of these compounds permitted collection of thermodynamic parameters pertinent to the unique N-H bonds. This data is comparatively discussed with the theoretically-predicted data of the N2-derived Fe(NNHx) species. Exceptionally-weak N-H bond enthalpies are found for many of these compounds, and sheds light on their short-lived nature and tendency to evolve H2. As a whole, these works both establish and provide a means to understand Fe-mediated N2 activation via the addition of hydrogen atom equivalents.

No presente trabalho, pretendeu-se avaliar a alga marinha Sargassum filipendula na sua capacidade de remoção do metal cobre na presença do metal cálcio, de modo a verificar o efeito da presença do cálcio, proveniente do hidróxido de cálcio (cal hidratada), utilizado no tratamento primário de efluentes por precipitação química. Para tanto, foi realizado primeiramente o estudo da cinética de biossorção de cobre e cálcio em regime de batelada, nas concentrações de 50 e 200 g/mL, e em seguida foi estudado o equilíbrio da biossorção de cobre e cálcio, também em regime de batelada, utilizando soluções isoladas e combinadas de cobre e cálcio, em concentrações variadas, com biomassa lavada com água corrente e com HCl 0,1 mol/L, de modo a verificar se houve melhora na biossorção com a protonação da biomassa. Os resultados do estudo cinético da biossorção do cobre e do cálcio mostraram, em todos os casos, que o equilíbrio ocorreu até os 30 minutos iniciais e que os resultados do cobre se ajustaram melhor a um modelo cinético de segunda ordem, enquanto que os resultados do cálcio não se ajustaram a nenhum dos dois modelos propostos. Foi possível verificar ainda uma relação direta entre biossorção de cobre e liberação de elementos alcalinos e alcalino-terrosos, sugerindo o envolvimento de troca-iônica durante o processo. Já com os resultados de estudo do equilíbrio da biossorção dos metais cobre e cálcio, foi possível obter algumas conclusões, dentre as quais podemos destacar a predileção pelo modelo de isotermas de Langmuir e a interferência na biosorção do cobre causada pela presença do cálcio na solução. Nesta etapa, foi possível ainda estabelecer novamente a correlação de permuta entre os metais cobre e alcalinos/alcalino terrosos. Os modelos de pseudo-primeira ordem e segunda ordem foram utilizados para avaliar a cinética de adsorção dos íons metálicos pela biomassa, enquanto que os modelos das isotermas de Langmuir e de Freundlich, foram utilizados para a representação do equilíbrio da biossorção

植物已经演化出多种保护其免受强光抑制和破坏的机制,从而使植物体在自然界能够应付复杂多变的光照环境。虽然人们早就确定Cyt b-559存在于PSII反应中心内,但目前对其性质与功能的认识还不充分。本工作的目的就是研究Cyt b-559天然分子特性,探讨其生理功能和存在的意义。取得了一些有新意的结果: 1、依据PSII反应中心分离纯化的原理,应用更有效的层析介质DEAE-Sephacel,我们设计了快速高效的从菠菜和水稻中分离纯化Cyt b-559的方法和流程,获得了高纯度的样品。它们在非变性胶电泳中具有相同的泳动性。蛋白组分的HPLC结果证明,纯化的Cyt b-559的确由两个亚基组成,α亚基和β亚基的分子量用我们设计的适合于分析小蛋白的Tricine—SDS—PAGE方法准确测定为9.4kDa和4.5kDa。 2、利用HPLC技术分析了纯化的Cyt b-559样品的色素组成,结果表明Cytb-559中含有Chl α而不含类胡萝卜素分子,这一结果通过吸收光谱和共振拉曼光谱的分析得到进一步地证明。通过等电聚焦方法分析了Cyt b-559的等电点,发现其亚基的等电点相差很大,全蛋白的等电点与...更多D1、D2蛋白的等电点也不相同,推测在体内生理pH条件下它们具有相反带电性而在PSII组装中发挥作用。 3、低温荧光光谱的检测结果表明,Cyt b-559的荧光发射峰位在563nm和666nm;首次证明Cyt b-559可以发出荧光和将电子传递给结合在其上的辅助叶绿素,但传递能力比较低故而导致其荧光特性与PSII反应中心的不相同。Cytb-559的紫外荧光光谱表明Trp残基位于其内部的疏水区域,证明Cyt b-559中的芳香族氨基酸可能在其功能的发挥中起一定作用。 4、通过MCD的分析,发现Cyt b-559中血红素的MCD信号在540—580nm和400—440nm波段,而且光谱形状和强度与PSII反应中心的相一致,说明PSII反应中心该范围内的MCD信号中有Cyt b-559的贡献。FTIR光谱的测定结果证明Cyt b-559血红素的配体是组氨酸,其二级结构中α-螺旋占了一半。此外,还比较了Cyt b-559和PSII反应中心的膜脂成分,发现两者有很大的相似性。不同植物来源的Cyt b-559在许多性质上都表现出高度一致,从一个侧面证明Cyt b-559在进化中的保守性。 5、PSll反应中心发生光破坏时,原初电子供体P680己受到严重破坏。我们发现,在光抑制的最初一段时间内,Cyt b-559吸收峰值发生变化:在受体侧光抑制的条件下,其吸收峰值先略有增加而后才下降,而在供体侧光抑制条件下则相反,说明 Cyt b-559对光抑制的发生非常敏感,可能在光抑制早期保护PSll反应中心。 6、纯化的Cyt b-559的组氨酸含量在照光前后没有显著的变化,说明 PSll反应中心内被破坏的组氨酸不属于Cyt b-559。PSll反应中心所含的组氨酸中有些可被DEPC修饰,但我们的实验结果表明DEPC不能修饰Cyt b-559的组氨酸。这可能有利于Cyt b-559保护功能的发挥。 7、我们观察到,在两种光抑制条件下,LP Cyt b-559光还原和 HP Cyt b-559光氧化具有对pH值的依赖性,说明Cyt b-559在光保护中的作用不仅与其高低电势态有关,而且与其质子化程度有联系。CCCP促进HP Cyt -559释放质子,从而维持循环电子传递。DCBQ和 DCMU在很低浓度时都抑制 Cyt b-559光还原,前者不影响Cyt b-559光氧化而后者在CCCP存在时也会抑制Cyt b-559光氧化。 8、Cyt b-559有定位PSll反应中心其它蛋白的锚蛋白的作用。黄化苗转绿实验证明在 HP Cyt b-559的含量增加超过 45%以后放氧活性开始逐渐增加。Cytb-559从低电势态到高电势态的转变是放氧复合物组装到PSll反应中心的关键步骤之一。在植物正常生长时,Cyt b-559与 P680的其它电于供体发生竟争,起到安全阀门的作用。 9、在逆境条件下,Cyt b-559具有保护PSll反应中心免受强光破坏而起到“分于开关”的作用。我们的实验表明,在室温条件下存在通过Cyt b-559的环式电子流,存在从氧化态LP Cyt b-559到还原态HP Cyt b-559的一个循环,其中的氧化还原变化与质子化/去质子化反应相连。通过与其它血红素蛋白的比较,我们推测 Cyt b-559“分子开关”的关键是:光抑制情况下,铁原子与远端His之间的疏水空穴被氧自由基占据后使得铁进入叶琳中央孔中,迫使近端HIS向叶琳平面位移,从而引起 Cyt b-559构象改变,使电势态发生转变。

1,4,10,13,16-Pentaazatricycloheneicosane-9,17-dione (macrocyclic polyamine)-modified polymer-based monolithic column for CEC was prepared by ring opening reaction of epoxide groups from poly(glycidyl methacrylate-co-ethylene dimethacrylate) (GMA-co-EDMA) monolith with macrocyclic polyamine. Conditions such as reaction time and concentration of macrocyclic polyamine for the modification reaction were optimized to generate substantial EOF and enough chromatographic interactions. Anodic EOF was observed in the pH range of 2.0-8.0 studied due to the protonation of macrcyclic polyamine at the surface of the monolith. Morphology of the monolithic column was examined by SEM and the incorporation of macrocyclic polyamine to the poly(GMA-co-EDMA) monolith was characterized by infrared (IR) spectra. Successful separation of inorganic anions, isomeric benzenediols, and benzoic acid derivatives on the monolithic column was achieved for CEC. In addition to hydrophobic interaction, hydrogen bonding and electrostatic interaction played a significant role in the separation process.

An anionic, phosphonate-functionalized polyfluorene, i.e., poly(9,9-bis(3'-phosphatepropyl)fluorene-alt-1,4-phenylene) sodium salt (PFPNa), has been synthesized by copolymerization of phosphonic acid-substituted 2,7-dibromofluorene and phenyldiboronic ester via direct Suzuki polycondensation reaction in DMF/water. Polymer PFPNa is highly soluble and emissive in water with a solubility of 60 mg/mL and a photoluminescence quantum yield of 75%. The absorption and fluorescence spectra of PFPNa are strongly dependent on pH value owing to the partial protonation of phosphate groups and the aggregation of the polymer chains.

Electrodeposition of novel Au/Pd bimetallic nanostructures with dendrimer films as matrices has been reported. The dendrimers exhibited highly open structures arising from protonation of amines and this made them have good penetrability for solvent molecules. The unique properties of dendrimers obviously affected the morphologies and compositions of deposited bimetallic nanostructures compared with those from unmodified surfaces. Field-emitted scanning electron microscopy, energy dispersive spectroscopy, X-ray photoelectron spectroscopy and UV-vis spectroscopy were used to characterize these nanostructures.

Formation and stabilities of four 14-mer intermolecular DNA triplexes, consisting of third strands with repeating sequence CTCT, CCTT, CTT, or TTT, were studied by electrospray ionization Fourier-transform ion cyclotron resonance mass spectrometry (ESI-FTICR-MS) in the gas phase. The gas-phase stabilities of the triplexes were compared with their CD spectra and melting behaviors in solution, and parallel correlation between two phases were obtained. In the presence of 20 mm NH4+ (pH 5.5), the formation of the TTT triplex was not detected in both solution and the gas phase.

The controllable synthesis of nanosized carbon-supported Pd catalysts through a surface replacement reaction (SRR) method is reported in this paper. Depending on the synthesis conditions the Pd can be formed on Co nanoparticles surface in hollow nanospheres or nanoparticles structures. Citrate anion acts as a stabilizer for the nanostructures, and protonation of the third carboxyl anion and hence the nanostructure and size of the resulting catalysts are controlled via the pH of the synthesis solution. Pd hollow nanospheres, containing smaller Pd nanoparticles, supported on carbon are formed under the condition of pH 9 reaction solution. Meanwhile, highly dispersed carbon-supported Pd nanoparticles can be formed with higher pH (pH >= 10). All catalysts prepared through the SRR method show enhanced activities for the HCOOH electro-oxidation reaction compared to catalysts reduced by NaBH4.

Sequentially spectrophotometric titrations by sodium hydroxide of meso-tetraphenylporphyrin derivatives bearing one, two, three, or four p-hydroxyl groups result in new types of spectra. The strong new bands appear in the visible region with splitting or broadening of the Soret band and its significant loss of oscillator strength. To understand the molecular origin of these phenomena, the Resonance Raman (RR) and Fourier Transform Infrared (FTIR) experiments are carried out. The results demonstrate that the charges of the deprotonated para-hydroxy substituted meso-tetraphenylporphyrins are localized on the substituents, not delocalized into the pi system of the porphyrin macrocycles and that the ground states of the macrocycles remain essentially unperturbed. Both the related behavior of diprotonated tetrakis(p-(dimethylamino)phenyl) porphyrin and protonated Schiff base porphyrins show that the new bands considered as hyperporphyrin spectra are due to pi(phenoxide anion) --> pi*(porphyrin) transitions, where pi is an orbital on the phenoxide anion substitutent and pi* is a LUMO on the porphyrin.
