This paper presents a methodology for the study of a molten carbonate fuel cell co-generation system. This system is applied to a dairy industry of medium size that typically demands 2100 kW of electricity, 8500 kg/h of saturated steam (P = 1.08 MPa) and 2725 kW of cold water production. Depending on the associated recuperation equipment, the co-generation system permits the recovery of waste heat, which can be used for the production of steam, hot and cold water, hot and cold air. In this study, a comparison is made between two configurations of fuel cell co-generation systems (FCCS). The plant performance has been evaluated on the basis of fuel utilisation efficiency and each system component evaluated on the basis of second law efficiency. The energy analysis presented shows a fuel utilisation efficiency of about 87% and exergy analysis shows that the irreversibilities in the combustion chamber of the plant are significant. Further, the payback period estimated for the fuel cell investment between US$ 1000 and US$ 1500/k-W is about 3 and 6 years, respectively. (C) 2002 Elsevier B.V. B.V. All rights reserved.

A produção in vitro de embriões (PIV) é uma biotecnologia utilizada para aumentar o potencial reprodutivo de animais geneticamente superiores, os embriões produzidos in vitro são de qualidade inferior aos produzidos in vivo, por isso técnicas tentam melhorar os índices de embriões produzidos in vitro. Uma técnica é o sistema de co-cultivo com células somáticas que removem metabólitos tóxicos e protegem contra o stress oxidativo. As células-tronco mesenquimais derivadas de tecido adiposo (CTA) são células multipotentes que segregam fatores de crescimento e citocinas. As células-tronco foram utilizadas em co-cultivo in vitro de embriões bovinos em diferentes concentrações com o objetivo de melhorar o protocolo de PIVE. CTAs foram submetidas à diferenciação em três linhagens mesenquimais, e foi realizada a imunofenotipagem de marcadores específicos de membrana das CTMs. A taxa de clivagem foi avaliada no segundo dia após a fertilização e taxa de blastocistos no sétimo dia, quando foram armazenados para contagem do número total de células e expressão gênica. Os resultados foram analisados por ANOVA, Teste-t e pós-teste de Fisher, adotando um nível de significância de 5%. O tratamento do co-cultivo com CTAs influenciou significativamente a formação de blastocisto, o número total de células de embriões e a expressão gênica correlacionada a pluripotência e metabolismo de carboidratos. Estes resultados mostraram aumento da taxa de produção e qualidade dos embriões produzidos in vitro em co-cultivo com CTAs em relação ao co-cultivo com células da granulosa. Os resultados deste trabalho indicam também que a presença constante de CTAs em co-cultivo é superior ao condicionamento com CTAs. Os efeitos verificados das CTAs podem ocorrer através de fatores solúveis ou via exossomos secretados pelas CTAs. Estudos futuros são necessários para esclarecer a possível via causadora dos efeitos positivos verificados neste trabalho pelas CTAs em co-cultivo.

Molecular investigations retrieve information on properties often accessible only by expensive and time-consuming experimental techniques, such as those involved in the study of radical-based chain reactions. In this work, different quantum chemical techniques were used to study phenol oxidation by hydroxyl radicals in Advanced Oxidation Processes used for wastewater treatment. The results obtained by applying a DFT-based model showed good agreement with experimental values available, as well as qualitative insights into the mechanism of the overall reaction chain. Solvation models were also tried, but were found to be limited for this reaction system within the considered theoretical level without further parameterization.

Waste management is becoming, year after year, always more important both for the costs associated with it and for the ever increasing volumes of waste generated. The discussion on the fate of organic fraction of municipal solid waste (OFMSW) leads everyday to new solutions. Many alternatives are proposed, ranging from incineration to composting passing through anaerobic digestion. “For Biogas” is a collaborative effort, between C.I.R.S.A. and R.E.S. cooperative, whose main goal is to generate “green” energy from both biowaste and sludge anaerobic co-digestion. Specifically, the project include a pilot plant receiving dewatered sludge from both urban and agro-industrial sewage (DS) and the organic fraction of MSW (in 2/1 ratio) which is digested in absence of oxygen to produce biogas and digestate. Biogas is piped to a co-generation system producing power and heat reused in the digestion process itself, making it independent from the national grid. Digestate undergoes a process of mechanical separation giving a liquid fraction, introduced in the treatment plant, and a solid fraction disposed in landfill (in future it will be further processed to obtain compost). This work analyzed and estimated the impacts generated by the pilot plant in its operative phase. Once the model was been characterized, on the basis of the CML2001 methodology, a comparison is made with the present scenario assumed for OFMSW and DS. Actual scenario treats separately the two fractions: the organic one is sent to a composting plant, while sludge is sent to landfill. Results show that the most significant difference between the two scenarios is in the GWP category as the project "For Biogas" is able to generate “zero emission” power and heat. It also generates a smaller volume of waste for disposal. In conclusion, the analysis evaluated the performance of two alternative methods of management of OFMSW and DS, highlighting that "For Biogas" project is to be preferred to the actual scenario.

Stable isotope composition of atmospheric carbon monoxide: A modelling study.rnrnThis study aims at an improved understanding of the stable carbon and oxygen isotope composition of the carbon monoxide (CO) in the global atmosphere by means of numerical simulations. At first, a new kinetic chemistry tagging technique for the most complete parameterisation of isotope effects has been introduced into the Modular Earth Submodel System (MESSy) framework. Incorporated into the ECHAM/MESSy Atmospheric Chemistry (EMAC) general circulation model, an explicit treatment of the isotope effects on the global scale is now possible. The expanded model system has been applied to simulate the chemical system containing up to five isotopologues of all carbon- and oxygen-bearing species, which ultimately determine the δ13C, δ18O and Δ17O isotopic signatures of atmospheric CO. As model input, a new stable isotope-inclusive emission inventory for the relevant trace gases has been compiled. The uncertainties of the emission estimates and of the resulting simulated mixing and isotope ratios have been analysed. The simulated CO mixing and stable isotope ratios have been compared to in-situ measurements from ground-based observatories and from the civil-aircraft-mounted CARIBIC−1 measurement platform.rnrnThe systematically underestimated 13CO/12CO ratios of earlier, simplified modelling studies can now be partly explained. The EMAC simulations do not support the inferences of those studies, which suggest for CO a reduced input of the highly depleted in 13C methane oxidation source. In particular, a high average yield of 0.94 CO per reacted methane (CH4) molecule is simulated in the troposphere, to a large extent due to the competition between the deposition and convective transport processes affecting the CH4 to CO reaction chain intermediates. None of the other factors, assumed or disregarded in previous studies, however hypothesised to have the potential in enriching tropospheric CO in 13C, were found significant when explicitly simulated. The inaccurate surface emissions, likely underestimated over East Asia, are responsible for roughly half of the discrepancies between the simulated and observed 13CO in the northern hemisphere (NH), whereas the remote southern hemisphere (SH) compositions suggest an underestimated fractionation during the oxidation of CO by the hydroxyl radical (OH). A reanalysis of the kinetic isotope effect (KIE) in this reaction contrasts the conventional assumption of a mere pressure dependence, and instead suggests an additional temperature dependence of the 13C KIE, which is driven by changes in the partitioning of the reaction exit channels. This result is yet to be confirmed in the laboratory.rnrnApart from 13CO, for the first time the atmospheric distribution of the oxygen mass-independent fractionation (MIF) in CO, Δ17O, has been consistently simulated on the global scale with EMAC. The applicability of Δ17O(CO) observations to unravelling changes in the tropospheric CH4-CO-OH system has been scrutinised, as well as the implications of the ozone (O3) input to the CO isotope oxygen budget. The Δ17O(CO) is confirmed to be the principal signal for the CO photochemical age, thus providing a measure for the OH chiefly involved in the sink of CO. The highly mass-independently fractionated O3 oxygen is estimated to comprise around 2% of the overall tropospheric CO source, which has implications for the δ18O, but less likely for the Δ17O CO budgets. Finally, additional sensitivity simulations with EMAC corroborate the nearly equal net effects of the present-day CH4 and CO burdens in removing tropospheric OH, as well as the large turnover and stability of the abundance of the latter. The simulated CO isotopologues nonetheless hint at a likely insufficient OH regeneration in the NH high latitudes and the upper troposphere / lower stratosphere (UTLS).rn

A triple cell co-culture model was recently established by the authors, consisting of either A549 or 16HBE14o- epithelial cells, human blood monocyte-derived macrophages and dendritic cells, which offers the possibility to study the interaction of xenobiotics with those cells. The 16HBE14o- containing co-culture model mimics the airway epithelial barrier, whereas the A549 co-cultures mimic the alveolar type II-like epithelial barrier. The goal of the present work was to establish a new triple cell co-culture model composed of primary alveolar type I-like cells isolated from human lung biopsies (hAEpC) representing a more realistic alveolar epithelial barrier wall, since type I epithelial cells cover >93% of the alveolar surface. Monocultures of A549 and 16HBE14o- were morphologically and functionally compared with the hAEpC using laser scanning microscopy, as well as transmission electron microscopy, and by determining the epithelial integrity. The triple cell co-cultures were characterized using the same methods. It could be shown that the epithelial integrity of hAEpC (mean ± SD, 1180 ± 188 Ω cm(2)) was higher than in A549 (172 ± 59 Ω cm(2)) but similar to 16HBE14o- cells (1469 ± 156 Ω cm(2)). The triple cell co-culture model with hAEpC (1113 ± 30 Ω cm(2)) showed the highest integrity compared to the ones with A549 (93 ± 14 Ω cm(2)) and 16HBE14o- (558 ± 267 Ω cm(2)). The tight junction protein zonula occludens-1 in hAEpC and 16HBE14o- were more regularly expressed but not in A549. The epithelial alveolar model with hAEpC combined with two immune cells (i.e. macrophages and dendritic cells) will offer a novel and more realistic cell co-culture system to study possible cell interactions of inhaled xenobiotics and their toxic potential on the human alveolar type I epithelial wall.

Both climate change and socio-economic development will significantly modify the supply and consumption of water in future. Consequently, regional development has to face aggravation of existing or emergence of new conflicts of interest. In this context, transdisciplinary co-production of knowledge is considered as an important means for coping with these challenges. Accordingly, the MontanAqua project aims at developing strategies for more sustainable water management in the study area Crans-Montana-Sierre (Switzerland) in a transdisciplinary way. It strives for co-producing system, target and transformation knowledge among researchers, policy makers, public administration and civil society organizations. The research process basically consisted of the following steps: First, the current water situation in the study region was investigated. How much water is available? How much water is being used? How are decisions on water distribution and use taken? Second, participatory scenario workshops were conducted in order to identify the stakeholders’ visions of regional development. Third, the water situation in 2050 was simulated by modeling the evolution of water resources and water use and by reflecting on the institutional aspects. These steps laid ground for jointly assessing the consequences of the stakeholders’ visions of development in view of scientific data regarding governance, availability and use of water in the region as well as developing necessary transformation knowledge. During all of these steps researchers have collaborated with stakeholders in the support group RegiEau. The RegiEau group consists of key representatives of owners, managers, users, and pressure groups related to water and landscape: representatives of the communes (mostly the presidents), the canton (administration and parliament), water management associations, agriculture, viticulture, hydropower, tourism, and landscape protection. The aim of the talk is to explore potentials and constraints of scientific modeling of water availability and use within the process of transdisciplinary co-producing strategies for more sustainable water governance.

Notochordal cells (NC) remain in the focus of research for regenerative therapy for the degenerated intervertebral disc (IVD) due to their progenitor status. Recent findings suggested their regenerative action on more mature disc cells, presumably by the secretion of specific factors, which has been described as notochordal cell conditioned medium (NCCM). The aim of this study was to determine NC culture conditions (2D/3D, fetal calf serum, oxygen level) that lead to significant IVD cell activation in an indirect co-culture system under normoxia and hypoxia (2% oxygen).

Biological sensing of small molecules such as NO, O2, and CO is an important area of research; however, little is know about how CO is sensed biologically. The photosynthetic bacterium Rhodospirillum rubrum responds to CO by activating transcription of two operons that encode a CO-oxidizing system. A protein, CooA, has been identified as necessary for this response. CooA is a member of a family of transcriptional regulators similar to the cAMP receptor protein and fumavate nitrate reduction from Escherichia coli. In this study we report the purification of wild-type CooA from its native organism, R. rubrum, to greater than 95% purity. The purified protein is active in sequence-specific DNA binding in the presence of CO, but not in the absence of CO. Gel filtration experiments reveal the protein to be a dimer in the absence of CO. Purified CooA contains 1.6 mol heme per mol of dimer. Upon interacting with CO, the electronic spectrum of CooA is perturbed, indicating the direct binding of CO to the heme of CooA. A hypothesis for the mechanism of the protein’s response to CO is proposed.

S-Nitrosothiols have generated considerable interest due to their ability to act as nitric oxide (NO) donors and due to their possible involvement in bioregulatory systems—e.g., NO transfer reactions. Elucidation of the reaction pathways involved in the modification of the thiol group by S-nitrosothiols is important for understanding the role of S-nitroso compounds in vivo. The modification of glutathione (GSH) in the presence of S-nitrosoglutathione (GSNO) was examined as a model reaction. Incubation of GSNO (1 mM) with GSH at various concentrations (1–10 mM) in phosphate buffer (pH 7.4) yielded oxidized glutathione, nitrite, nitrous oxide, and ammonia as end products. The product yields were dependent on the concentrations of GSH and oxygen. Transient signals corresponding to GSH conjugates, which increased by one mass unit when the reaction was carried out with 15N-labeled GSNO, were identified by electrospray ionization mass spectrometry. When morpholine was present in the reaction system, N-nitrosomorpholine was formed. Increasing concentrations of either phosphate or GSH led to lower yields of N-nitrosomorpholine. The inhibitory effect of phosphate may be due to reaction with the nitrosating agent, nitrous anhydride (N2O3), formed by oxidation of NO. This supports the release of NO during the reaction of GSNO with GSH. The products noted above account quantitatively for virtually all of the GSNO nitrogen consumed during the reaction, and it is now possible to construct a complete set of pathways for the complex transformations arising from GSNO + GSH.

Óleo de soja epoxidado (OSE) é um produto químico há muito tempo utilizado como co-estabilizante e plastificante secundário do poli (cloreto de vinila) (PVC), ou seja, como um material que tem limitações na quantidade máxima que pode ser usada no composto de PVC. A sua aplicação como plastificante primário, ou seja, como o principal elemento plastificante no composto de PVC, e como base para outros plastificantes de fontes renováveis, tem aumentado nos últimos anos, principalmente devido a melhorias de desempenho e à redução do custo do OSE em comparação com plastificantes tradicionais. A reação de epoxidação do óleo de soja é bem conhecida e ocorre em duas fases líquidas, com reações em ambas as fases, e transferência de massa entre as fases. O processo industrial mais utilizado conta com formação in-situ do ácido perfórmico, através da adição gradativa do principal reagente, o peróxido de hidrogênio a uma mistura agitada de ácido fórmico e óleo de soja refinado. Industrialmente, o processo é realizado em batelada, controlando a adição do reagente peróxido de hidrogênio de forma que a geração de calor não ultrapasse a capacidade de resfriamento do sistema. O processo tem um ciclo que pode variar entre 8 e 12 horas para atingir a conversão desejada, fazendo com que a capacidade de produção seja dependente de investimentos relativamente pesados em reatores agitados mecanicamente, que apresentam diversos riscos de segurança. Estudos anteriores não exploram em profundidade algumas potenciais áreas de otimização e redução das limitações dos processos, como a intensificação da transferência de calor, que permite a redução do tempo total de reação. Este trabalho avalia experimentalmente e propõe uma modelagem para a reação de epoxidação do óleo de soja em condições de remoção de calor máxima, o que permite que os reagentes sejam adicionados em sua totalidade no início da reação, simplificando o processo. Um modelo foi ajustado aos dados experimentais. O coeficiente de troca térmica, cuja estimativa teórica pode incorrer em erros significativos, foi calculado a partir de dados empíricos e incluído na modelagem, acrescentando um fator de variabilidade importante em relação aos modelos anteriores. O estudo propõe uma base teórica para potenciais alternativas aos processos adotados atualmente, buscando entender as condições necessárias e viáveis em escala industrial para redução do ciclo da reação, podendo inclusive apoiar potenciais estudos de implementação de um reator contínuo, mais eficiente e seguro, para esse processo.

The knowledge of thermophysical properties of liquid Co-Si alloys is a key requirement for manufacturing of composite materials by infiltration method. Despite this need, the experimental and predicted property data of the Co-Si system are scarce and often inconsistent between the various sources. In the present work the mixing behaviour of Co-Si melts has been analysed through the study of the concentration dependence of various thermodynamic, surface (surface tension and surface composition) and structural properties (concentration fluctuations in the long-wavelength limit and chemical short-range order parameter) in the framework of the Compound Formation Model (CFM) and Quasi Chemical Approximation for regular solutions (QCA). In addition, the surface tension of the Co22·5Si77.5 (in at%) eutectic alloy, that is proposed to be used as the infiltrant, has been measured by the pendant drop method at temperatures ranging from 1593 to 1773 K. The results obtained were discussed with respect to both, temperature and concentration, and subsequently compared with the model predictions and literature data.

Objective: To introduce a new technique for co-registration of Magnetoencephalography (MEG) with magnetic resonance imaging (MRI). We compare the accuracy of a new bite-bar with fixed fiducials to a previous technique whereby fiducial coils were attached proximal to landmarks on the skull. Methods: A bite-bar with fixed fiducial coils is used to determine the position of the head in the MEG co-ordinate system. Co-registration is performed by a surface-matching technique. The advantage of fixing the coils is that the co-ordinate system is not based upon arbitrary and operator dependent fiducial points that are attached to landmarks (e.g. nasion and the preauricular points), but rather on those that are permanently fixed in relation to the skull. Results: As a consequence of minimizing coil movement during digitization, errors in localization of the coils are significantly reduced, as shown by a randomization test. Displacement of the bite-bar caused by removal and repositioning between MEG recordings is minimal (∼0.5 mm), and dipole localization accuracy of a somatosensory mapping paradigm shows a repeatability of ∼5 mm. The overall accuracy of the new procedure is greatly improved compared to the previous technique. Conclusions: The test-retest reliability and accuracy of target localization with the new design is superior to techniques that incorporate anatomical-based fiducial points or coils placed on the circumference of the head. © 2003 International Federation of Clinical Neurophysiology. The objective of this work was to design, construct and commission a new ablative pyrolysis reactor and a high efficiency product collection system. The reactor was to have a nominal throughput of 10 kg/11r of dry biomass and be inherently scalable up to an industrial scale application of 10 tones/hr. The whole process consists of a bladed ablative pyrolysis reactor, two high efficiency cyclones for char removal and a disk and doughnut quench column combined with a wet walled electrostatic precipitator, which is directly mounted on top, for liquids collection. In order to aid design and scale-up calculations, detailed mathematical modelling was undertaken of the reaction system enabling sizes, efficiencies and operating conditions to be determined. Specifically, a modular approach was taken due to the iterative nature of some of the design methodologies, with the output from one module being the input to the next. Separate modules were developed for the determination of the biomass ablation rate, specification of the reactor capacity, cyclone design, quench column design and electrostatic precipitator design. These models enabled a rigorous design protocol to be developed capable of specifying the required reactor and product collection system size for specified biomass throughputs, operating conditions and collection efficiencies. The reactor proved capable of generating an ablation rate of 0.63 mm/s for pine wood at a temperature of 525 'DC with a relative velocity between the heated surface and reacting biomass particle of 12.1 m/s. The reactor achieved a maximum throughput of 2.3 kg/hr, which was the maximum the biomass feeder could supply. The reactor is capable of being operated at a far higher throughput but this would require a new feeder and drive motor to be purchased. Modelling showed that the reactor is capable of achieving a reactor throughput of approximately 30 kg/hr. This is an area that should be considered for the future as the reactor is currently operating well below its theoretical maximum. Calculations show that the current product collection system could operate efficiently up to a maximum feed rate of 10 kg/Fir, provided the inert gas supply was adjusted accordingly to keep the vapour residence time in the electrostatic precipitator above one second. Operation above 10 kg/hr would require some modifications to the product collection system. Eight experimental runs were documented and considered successful, more were attempted but due to equipment failure had to be abandoned. This does not detract from the fact that the reactor and product collection system design was extremely efficient. The maximum total liquid yield was 64.9 % liquid yields on a dry wood fed basis. It is considered that the liquid yield would have been higher had there been sufficient development time to overcome certain operational difficulties and if longer operating runs had been attempted to offset product losses occurring due to the difficulties in collecting all available product from a large scale collection unit. The liquids collection system was highly efficient and modeling determined a liquid collection efficiency of above 99% on a mass basis. This was validated due to the fact that a dry ice/acetone condenser and a cotton wool filter downstream of the collection unit enabled mass measurements of the amount of condensable product exiting the product collection unit. This showed that the collection efficiency was in excess of 99% on a mass basis.
