Epithelial ovarian cancer (EOC) has an innate susceptibility to become chemoresistant. Up to 30% of patients do not respond to conventional chemotherapy [paclitaxel (Taxol®) in combination with carboplatin] and, of those who have an initial response, many patients relapse. Therefore, an understanding of the molecular mechanisms that regulate cellular chemotherapeutic responses in EOC cells has the potential to impact significantly on patient outcome. The mitotic arrest deficiency protein 2 (MAD2), is a centrally important mediator of the cellular response to paclitaxel. MAD2 immunohistochemical analysis was performed on 82 high-grade serous EOC samples. A multivariate Cox regression analysis of nuclear MAD2 IHC intensity adjusting for stage, tumour grade and optimum surgical debulking revealed that low MAD2 IHC staining intensity was significantly associated with reduced progression-free survival (PFS) (p = 0.0003), with a hazard ratio of 4.689. The in vitro analyses of five ovarian cancer cell lines demonstrated that cells with low MAD2 expression were less sensitive to paclitaxel. Furthermore, paclitaxel-induced activation of the spindle assembly checkpoint (SAC) and apoptotic cell death was abrogated in cells transfected with MAD2 siRNA. In silico analysis identified a miR-433 binding domain in the MAD2 3' UTR, which was verified in a series of experiments. Firstly, MAD2 protein expression levels were down-regulated in pre-miR-433 transfected A2780 cells. Secondly, pre-miR-433 suppressed the activity of a reporter construct containing the 3'-UTR of MAD2. Thirdly, blocking miR-433 binding to the MAD2 3' UTR protected MAD2 from miR-433 induced protein down-regulation. Importantly, reduced MAD2 protein expression in pre-miR-433-transfected A2780 cells rendered these cells less sensitive to paclitaxel. In conclusion, loss of MAD2 protein expression results in increased resistance to paclitaxel in EOC cells. Measuring MAD2 IHC staining intensity may predict paclitaxel responses in women presenting with high-grade serous EOC.

Aberrant expression of the MAD2 protein has been linked to chromosomal instability, malignant transformation and chemoresistance. Although reduced MAD2 expression is well recognised in human cancer cell lines, the mechanism(s) underlying its downregulation remain elusive. The objective of this study was to establish the impact of hypoxia on MAD2 expression and to investigate the potential role of aberrant promoter methylation as a possible mechanism of MAD2 downregulation. For this purpose, three ovarian cancer cell lines, displaying differing levels of MAD2, were treated with chromatin modifying drugs, pre and post-hypoxia exposure and a DHPLC analysis of DNA promoter methylation carried out. We show that hypoxia induces downregulation of MAD2 expression, independently of MAD2 promoter methylation. We also show no evidence of MAD2 promoter methylation in breast and prostate cancer cells or in breast cancer clinical material. While our findings provide no evidence for MAD2 promoter methylation, we show a concomitant upregulation of p21 with downregulation of MAD2 in hypoxia. Our in vitro results were also confirmed in an ovarian cancer tissue microarray (TMA), where a reciprocal staining of MAD2 and CAIX was found in 21/60 (35%) of tumours. In summary, MAD2 downregulation may be a crucial mechanism by which hypoxic cells become chemorefractory. This stems from our previous work where we demonstrated that MAD2 downregulation induces cellular senescence, a viable cellular fate, with resultant cellular resistance to paclitaxel. Moreover, MAD2 downregulation could play a central role in the induction of chemoresistance in hypoxia, a key tumour microenvironment associated with chemoresistance.

Purpose:This study documents the frequency of insulin-like growth factor-II (IGF-II) loss of imprinting (LOI) in a series of 87 bladder tissues. E-cadherin (CDH1) immunolocalization was also investigated due to the known redistribution of this adherence protein to the cytoplasm following exogenous exposure to IGF-II.
Experimental Design: Informative IGF-II cases were identified following DNA-PCR amplification and subsequent sequencing of the transcribable ApaI RFLP in exon 9 of IGF-II. Similar approaches using primer-specific cDNA templates identified the imprinting status of IGF-II in these informative cases. CDH1cellular localization was assessed on a tissue microarray platform of 114 urothelial carcinoma of the bladder (UCB) cases (70 pTanoninvasive and 44 pT1laminapropria invasive) using the commercially available Novocastra antibody.
Results: IGF-IILOI was evident in 7 of17 (41%) UCB tumors and 4 of11 (36%) tumor-associated normal urothelial samples.Two of four pT1grade 3 tumors, the subject of much debate concerning their suitability for radical cystectomy, showed LOI at the IGF-II locus. In those tumors showing IGF-II LOI, 4 of 7 (57%) displayed concomitant CDH1cytoplasmic staining. In contrast, only 3 of 10 (30%) IGF-IImaintenance ofimprinting tumorshad concomitant CDH1cytoplasmiclocalization. UCB cell lines displaying cytoplasmic CDH1immunolocalization expressed significantly higher levels of IGF-II (CAL29, HT1376, and RT112) compared with RT4, a cell line displaying crisp membranous CDH1staining. Finally, cytoplasmic CDH1staining was an independent predictor of a shorter time to recurrence independent of tumor grade and stage.
Conclusions: We suggest that CDH1 cytoplasmic immunolocalization as a result of increased IGF-II levels identifies those nonmuscle invasive presentations most likely to recur and therefore might benefit from more radical nonconserving bladder surgery

Schizophrenia is an idiopathic mental disorder with a heritable component and a substantial public health impact. We conducted a multi-stage genome-wide association study (GWAS) for schizophrenia beginning with a Swedish national sample (5,001 cases and 6,243 controls) followed by meta-analysis with previous schizophrenia GWAS (8,832 cases and 12,067 controls) and finally by replication of SNPs in 168 genomic regions in independent samples (7,413 cases, 19,762 controls and 581 parent-offspring trios). We identified 22 loci associated at genome-wide significance; 13 of these are new, and 1 was previously implicated in bipolar disorder. Examination of candidate genes at these loci suggests the involvement of neuronal calcium signaling. We estimate that 8,300 independent, mostly common SNPs (95% credible interval of 6,300-10,200 SNPs) contribute to risk for schizophrenia and that these collectively account for at least 32% of the variance in liability. Common genetic variation has an important role in the etiology of schizophrenia, and larger studies will allow more detailed understanding of this disorder.

Ovarian carcinoma (OC) is the most lethal of the gynecological malignancies, often presenting at an advanced stage. Treatment is hampered by high levels of drug resistance. The taxanes are microtubule stabilizing agents, used as first-line agents in the treatment of OC that exert their apoptotic effects through the spindle assembly checkpoint. BUB1-related protein kinase (BUBR1) and mitotic arrest deficient 2 (MAD2), essential spindle assembly checkpoint components, play a key role in response to taxanes. BUBR1, MAD2, and Ki-67 were assessed on an OC tissue microarray platform representing 72 OC tumors of varying histologic subtypes. Sixty-one of these patients received paclitaxel and platinum agents combined; 11 received platinum alone. Overall survival was available for all 72 patients, whereas recurrence-free survival (RFS) was available for 66 patients. Increased BUBR1 expression was seen in serous carcinomas, compared with other histologies (P = .03). Increased BUBR1 was significantly associated with tumors of advanced stage (P = .05). Increased MAD2 and BUBR1 expression also correlated with increased cellular proliferation (P < .0002 and P = .02, respectively). Reduced MAD2 nuclear intensity was associated with a shorter RFS (P = .03), in ovarian tumors of differing histologic subtype (n = 66). In this subgroup, for those women who received paclitaxel and platinum agents combined (n = 57), reduced MAD2 intensity also identified women with a shorter RFS (P < .007). For the entire cohort of patients, irrespective of histologic subtype or treatment, MAD2 nuclear intensity retained independent significance in a multivariate model, with tumors showing reduced nuclear MAD2 intensity identifying patients with a poorer RFS (P = .05).

Paclitaxel is a microtubule inhibitory chemotherapeutic drug that is increasingly used for the treatment of solid tumours. In vitro studies have demonstrated that attenuating the spindle assemble checkpoint (SAC) alters the post-mitotic responses to paclitaxel. Furthermore, the aberrant expression of a number of the SAC proteins, MAD2, BUBR1, and Aurora A kinase, are associated with poor patient prognosis. We have identified a microRNA, miR-433, that regulates the expression of MAD2. Overexpression of miR-433 in Hela cells induced downregulation of MAD2 mRNA and protein expression. We have also shown that Hela cells overexpressing miR-433 and treated with paclitaxel are no longer capable of cyclin B stabilisation, and thus have lost the ability to activate the SAC in response to paclitaxel. In addition, cell viability assays showed that Hela cells overexpressing miR-433 and treated with paclitaxel have an attenuated response to paclitaxel compared with microRNA scrambled controls. We have characterised the levels of miR-433, MAD2 gene expression and MAD2 protein levels in a cohort of ovarian cancer cell lines. Cell viability assays on this cohort revealed that responsiveness to paclitaxel is associated with high MAD2 protein expression and lower miR-433 expression. We hypothesise that the expression of miR-433 when deregulated in cancer leads to altered MAD2 expression and a compromised SAC, a key feature underlying drug resistance to paclitaxel. In a pilot study of paired human breast tumour and normal breast tissue samples we have shown that expression levels of miR-433 are elevated in cancer tissue. Targeting this microRNA in cancer may improve the efficacy of paclitaxel in treating breast cancer and ovarian cancer.

A produção de uva de mesa “Dona Maria”, casta 100% nacional criada e obtida nos anos 50 na antiga Estação Agronómica Nacional, em Oeiras, pelo engenheiro-agrónomo Leão Ferreira de Almeida, vem decrescendo ano após ano como resultado do desinteresse e abandono por parte dos agricultores. É uma uva muito doce, com bagos grandes e apreciada pelos portugueses que sentem cada vez mais dificuldades em encontrá-la no mercado. Este estudo de caso tem como objetivos verificar o porquê de tão pouca produção desta uva por parte dos agricultores e sugerir através de ferramentas e estratégias de marketing formas de fazer com que a uva seja mais conhecida e consumida pelo mercado português. No intuito de valorizar e consumir o que é nacional, a uva “Dona Maria” é uma excelente opção para o consumo em fresco ou na forma de passa, em cantinas de escolas e universidades. Os investimentos em novas tecnologias de produção e investigação por parte dos produtores também são uma boa opção para rentabilizar a casta e fazer com que ela não desapareça do mercado, o que seria desastroso na perspectiva cultural e económica; ### Abstract: The production of the table grape “Dona Maria”, a 100% national Portuguese grape variety, created and produced in the 1950s in the former National Agricultural Station (EAN) in Oeiras, Portugal, by Leão Ferreira de Almeida, has been decreasing year after year as a result of lack of interest and abandonment on behalf of producers. It is a very sweet grape with big berries and is highly appreciated by the Portuguese, who find it ever more difficult to find it in the marketplace. This case study´s objectives were to investigate the shortage of production by growers, and to suggest through marketing strategies and tools, ways to make this grape better known and more consumed by the Portuguese market. Within the scope of highlighting and promoting this national product, “Dona Maria” grape is an excellent option either as a fresh consumable or raisin in school and university restaurants. Investment in new production technologies and research are also a good choice and direction to promote the variety and avoid its disappearance from the market, which would be disastrous both culturally and economically.

Trabalho que investiga a recepção dos Kinder - und Hausmärchen em Portugal, fruto duma aturada e minuciosa pesquisa documental. Com efeito, Teresa Cortez passa em revista os livros e publicações periódicas enquadrados entre 1837 — data do aparecimento das primeiras traduções de contos dos KHM, no periódico lisboeta Biblioteca Familiar e Recreativa — e 1910, data a que a autora estendeu a sua pesquisa. Cada incidência da presença dos contos dos Grimm em Portugal é cuidadosamente investigada, contextualisada e analisada, Assim, à medida que vemos surgir, primeiro esparsas em revistas destinadas a um público infantil, e sem indicação de autoria, traduções (do francês) dos KHM, vamo-nos inteirando sobre o epírito e motivações do seu aparecimento, quer partindo do estudo comparativo com o texto de origem, quer de um exame da publicação de chegada, alargando-se a pesquisa aos seus editores e autores. Todo o processo de recepção dos contos de Grimm vai sendo examinado pela autora à luz dos movimentos das últimas décadas do séc. XIX e primeira do séc. XX, nomeadamente o nacionalismo neo-romântico e o positivismo.

Como qualquer profissional, cumpre-nos, no papel de formadores, o dever de procurar, a cada momento, uma melhoria da nossa atuação e da formação que facultamos. Enquanto formador de professores, esta assunção foi um dos motivos que conduziu à elaboração de uma investigação centrada no desenvolvimento profissional do professor focando, em particular, algumas das dimensões que considero centrais no seu conhecimento profissional (crenças, objetivos, conhecimentos – mathematical knowledge for teaching – e tipos de comunicação matemática promovidos), tendo para isso sido desenvolvidos dois modelos de análise complementares (Modelo Cognitivo e de Representação do Desenvolvimento Profissional – MRDP). Neste texto, a partir da análise da prática, analisam-se e discutem-se as “evoluções” das relações entre as dimensões em análise, recorrendo aos dois modelos. Inicio com algumas considerações sobre cada uma das dimensões em análise e, com recurso a um exemplo da prática, ilustro a aplicação do MRDP. A utilização destes modelos permite obter uma mais ampla compreensão da prática, dos fatores que a influenciam e como a influenciam, revelando-se o MRDP um potente instrumento que permite uma mais clara representação visual das alterações e persistências que se verificam na prática ao longo do tempo, sendo estas (in)alterações encaradas em termos da reflexividade do professor sobre a sua prática. Um outro aspeto relevante relacionase com as potencialidades do recurso a ambos os modelos para a/na formação de professores e o papel que podem assumir nela.

A transmissão de agentes patogénicos (bactérias, vírus e parasitas) através do consumo de géneros alimentícios pode desencadear um problema gravíssimo de saúde pública. Esta preocupação crescente levou o Homem a criar regras e aplicar medidas rigorosas que impedissem a contaminação dos alimentos e a propagação de doenças infecto-contagiosas que provocam muitos prejuízos à sociedade atual. De acordo com a Retificação ao Regulamento (CE) n.º 852/2004 do Parlamento Europeu e do Conselho de 29 de abril, verificou-se a necessidade de implementar um programa de pré-requisitos (PPR) (1.ª Fase) baseado na metodologia do Sistema HACCP no Mercado Municipal de Tavira, com o intuito de valorizar estes locais de comércio a retalho de diversos grupos de géneros alimentícios, onde existem inúmeros riscos de contaminação. Foi elaborada uma checklist e realizada uma avaliação minuciosa das condições de construção do edifício, a disposição da estrutura física, o modo de funcionamento dos equipamentos e o fluxo das operações integradas no respetivo mercado municipal, desde a zona de receção da mercadoria até aos locais de exposição e venda dos alimentos aos consumidores. A cascata de operações para a implementação do Sistema HACCP divide-se em três fases, contudo, para esta dissertação, apenas a primeira fase (programa de pré-requisitos - PPR) foi implementada no respetivo mercado municipal, a saber: - PPR n.º 1: Instalações e equipamentos; - PPR n.º 2: Plano de higienização; - PPR n.º 3: Controlo de pragas; - PPR n.º 4: Abastecimento de água; - PPR n.º 5: Recolha de resíduos; - PPR n.º 6: Material em contacto com géneros alimentícios; - PPR n.º 7: Higiene pessoal; - PPR n.º 8: Formação do pessoal. A implementação do PPR tem sido um processo moroso e ainda se encontra na fase de implementação. Periodicamente, os procedimentos, registos e documentos são verificados por forma evitar a repetição dos problemas.
