O paracetamol (PAR) é um dos medicamentos de venda livre mais utilizado em todo o mundo. Entretanto, doses elevadas do PAR produzem toxicidade hepática e/ou renal. No intuito de minimizar a toxicidade do PAR e obter melhor atividade analgésica e anti-inflamatória, um estudo prévio realizou modificações na estrutura química do PAR por modelagem molecular, dando origem ao ortobenzamol (OBZ) – análogo do PAR. Assim, o OBZ foi sintetizado e avaliado em modelos de nocicepção e inflamação em animais. O estudo demonstrou atividade analgésica central do OBZ, com potência superior ao PAR. Além disso, nos testes de inflamação, essa droga apresentou inibição significativa no processo inflamatório. Entretanto, para que o OBZ possa ser considerado uma alternativa terapêutica nova e importante para o tratamento da dor e/ou da inflamação é necessário determinar sua toxicidade. Assim, este estudo objetivou avaliar a toxicidade in vitro e in vivo do OBZ e, compará-la com a do PAR. Para isso, a neurotoxicidade foi avaliada in vitro em culturas primárias de neurônios corticais, através de ensaios de viabilidade celular, determinação dos níveis de glutationa total e reduzida, assim como a possível capacidade neuroprotetora frente ao estresse oxidativo. Foram realizados estudos in vivo em camundongos, iniciados pela determinação da dose efetiva mediana (DE50) do PAR, a fim de compará-la com a do OBZ nos modelos de toxicidade estudados. Determinou-se o estresse oxidativo hepático e cerebral pela análise dos níveis de peroxidação lipídica e nitritos. A possível disfunção hepática e renal foi determinada, por meio da análise dos níveis plasmáticos das enzimas aspartato aminotransferase (AST), de alanina aminotransferase (ALT), gama glutamiltransferase (GGT) e, da creatinina no sangue. Avaliaram-se alterações nos parâmetros clínicos através do hemograma, leucograma e plaquetograma e, realizou-se a determinação da toxicidade aguda. Os resultados obtidos neste estudo demonstraram que o ortobenzamol é mais seguro que o paracetamol. Registrou-se ao ortobenzamol ausência de neurotoxicidade, menor potencial hepatotóxico e hematotóxico, ausência de nefrotoxicidade e, ainda, foi classificado como um xenobiótico de baixa toxicidade após a avaliação da toxicidade aguda. Portanto, o ortobenzamol pode ser considerado como uma futura alternativa terapêutica segura ao paracetamol, no tratamento da dor e inflamação.

O conhecimento da dinâmica das alterações nos parâmetros hematológicos e na cinética das proteínas de fase aguda em animais saudáveis nas primeiras semanas de vida é essencial para a interpretação correta dessas avaliações em situações de morbidez e para diferenciar animais sadios e enfermos de forma confiável. Com o intuito de avaliar a cinética desses parâmetros no primeiro mês de vida de bezerros de corte sadios, filhos de vacas primíparas ou pluríparas, amostras de sangue foram coletadas antes da ingestão de colostro e 1, 2, 7, 15 e 30 dias após o nascimento. Os parâmetros eritrocitários foram influenciados pelo número de partos das vacas e o leucograma mostrou alterações características de influência do cortisol fetal liberado por ocasião do nascimento. O teor sérico de proteína total aumentou significativamente após a ingestão do colostro. As concentrações de ceruloplasmina, haptoglobina e proteínas de pesos moleculares 33 kDa e 23 kDa aumentaram significativamente no primeiro dia de vida, seja pela resposta ao nascimento ou pela ingestão do colostro, enquanto os teores de transferrina, albumina e α1-glicoproteína ácida mantiveram-se relativamente estáveis nos primeiros dias de vida, aumentando gradualmente até os 30 dias de idade.

The leukocyte count interpretation on the blood helps on understanding about the possible dysfunction showed by the animal. In general, an abnormal leukogram allows the identification of pathological process. A lot of events lead to an increase in the number of neutrophils, among then, inflammatory process, infection and stress. Dependent on the stress mediator that the animal is submited, the leukocyte alteration will be different. Is important to know how the stress acts on the animal organism, changing the hematological parameters, in order to avoid mistakes on diseases‟ diagnosis. It‟s known that after a stress situation, it‟s expected that stress leukogram appears on the hemogram of the animal. However there are different kinds of stress with different responses. It‟s believed that when the animal is under an acute stress situation, a clinical picture of physiologic leukocytosis occurs which is mediated by the epinephrine. This is characterized by neutrophilia, lymphocytosis, monocytosis and eosinofilia. The physiologic leukocytosis is a transient alteration in the leukogram that occurs within minutes of the stimulus and due to fleeting effect of catecolamines it resolves within 20 to 30 minutes. However, if the animal is under chronicle stress, as solitude, it „s expected to find the stress leukogram on the leukogram, that can be noted especially in dogs. The stress leukogram is characterized by leukocytosis, neutrophilia, lymphopenia, monocytosis and eosinopenia. This kind of stress is induced by the glucocorticoids. It´s important to understand that stress leukogram takes time to occur. The variation on time depends on the animal species, although, in general, the glucocorticoides have a peak effects among 4 to 8 hours, which can last from 24 hours to 2 or 3 days

The aim of this study was to determine the influence of climatic conditions on the hematological profile of Saanen and mixed-breed (1/2 Saanen x 1/2 Anglo-Nubian) goats, as well as to define reference values for these animals bred in Ceara, Brazil. Thirty goats were utilized and blood samples were collected monthly during the rainy (February to May) and dry (August to November) periods to obtain an erythrogram a leukogram. The averange were compared by the t-Student test and Mann Whitney test, with parametric and non-parametric distribution of the data, respectively, where p<0.05 was considered significant. A study was carried out of simple Pearson correlations of the hematological parameters with environmental and physiological variables. The number of red blood cells (RBCs) was higher in the Saanen goats and in the rainy period, while the hematocrit was higher in the dry period (p<0.05). The leukocytes and lymphocytes were higher in the 1/2S1/2 AN goats in the two periods (p<0.05). In the two genotypes, the leukocytes and lymphocytes were higher in the dry season and the segmented neutrophils higher in the rainy season (p<0.05). The other parameters did not differ (p>0.05). The RBCs and segmented neutrophils displayed a negative correlation with air temperature, but positive with relative humidity and rectal temperature (p<0.05). The hematocrit positively correlated with air temperature and respiratory rate (p<0.05). The leukocytes and lymphocytes showed a negative correlation with rectal temperature (p<0.05). It was therefore concluded that Saanen females are more affected by climatic variations and that the rainy season has a greater negative impact on hematological parameters. The values obtained could serve as a reference for these genotypes in Ceara.

Stress is one of the major obstacles in frog culture and can be caused by factors such as inappropriate farming systems; inadequate management among other situations. The objective of the present study was to assess the hemogram, erythrogram and leukogram of bullfrogs (L. catesbeianus) when exposed to stress caused by different types of management: density and handling (manipulation), developed in the laboratory and repeated in the field for the appropriate comparisons in a experimental period of 30 days. The density experiment was conducted with four treatments: 70 animals m(-2) (D70); 100 animals m(-2) (D100), Control; 150 animals m(-2) (D150) and 200 animals m(-2) (D200), with 10, 14, 21 and 28 animals/box in the laboratory, respectively. Each treatment was performed with three simultaneous replicates. The handling experiment was conducted with three treatments: Treatment Without Handling (WH); Treatment with Partial Handling (PH) every 15 days and Treatment with Total Handling (TH) every 15 days. Each treatment was performed with four simultaneous replications. The methodology of the blood analysis followed international recommendations. In the present study we could observe that the animals of the field experiment did not reflect the same stress response observed in the laboratory in both experiment, which demonstrated the plasticity of these animals.

Hyperadrenocorticism is a relatively common endocrinopathy in dogs, which is associated to an excessive production or administration of cortisol. The most affected breeds are Poodles, Teckels, Boxers, Boston Terriers and Beagles. The clinical signs most commonly observed are polyuria, polydipsia, polyfagia, panting, distended abdomen, endocrine alopecia, muscular weakness and lethargy. Laboratorial abnormalities include stress leukogram, increase in alkaline phosphatase and alanine aminotransferase activities, hypercholesterolemia, lipemia, hyperglycemia and hyposthenuria. The preferred essay to evaluate adrenal gland function is the low-dose dexamethasone suppression test, whereas the most used treatments include mitotane and trilostane. The objective of this paper is to review hyperadrenocorticism in dogs, because this disease is relatively common in small animal clinics and has many long-term complications.

To establish reference values and to assess the influence of age on the leukograms of healthy Holstein calves, blood samples were obtained from 300 animals. These samples were distributed equally (n=20) among 15 experimental groups according to age: birth to 8 hours, 9 to 16 hours, 17 to 24 hours, 2 days, 3 days, 4 days, 5 days, 6 to 7 days, 8 to 9 days, 10 to 11 days, 12 to 13 days, 14 to 15 days, 16 to 20 days, 21 to 25 days and 26 to 30 days of age. The maximum numbers of leukocytes (9,305.0/mL), segmented neutrophils (6,551.2/mL) and total neutrophils (6,678.3/mL) were noted within the first 8 hours of life, while band neutrophils peaked in number (133.3/mL) between 9 and 16 hours after birth. Meanwhile, the maximum total lymphocyte (4,992.1/µL) and typical lymphocyte (4,686.1/µL) counts occurred between 21 and 25 days, whereas atypical lymphocytes (388.5/µL) reached their maximum number between 26 and 30 days, demonstrating an inversion of the neutrophil:lymphocyte ratio over time. Thus, the influence of age on the leukocyte count of the evaluated calves was verified. The release of endogenous corticosteroids during labor or at birth may contribute to this variation in leukograms with age.
