Unique Alterations of an Ultraconserved Non-Coding Element in the 3 ' UTR of ZIC2 in Holoprosencephaly

Coding region alterations of ZIC2 are the second most common type of mutation in holoprosencephaly (HPE). Here we use several complementary bioinformatic approaches to identify ultraconserved cis-regulatory sequences potentially driving the expression of human ZIC2. We demonstrate that an 804 bp element in the 3' untranslated region (3'UTR) is highly conserved across the evolutionary history of vertebrates from fish to humans. Furthermore, we show that while genetic variation of this element is unexpectedly common among holoprosencephaly subjects (6/528 or >1%), it is not present in control individuals. Two of six proband-unique variants are de novo, supporting their pathogenic involvement in HPE outcomes. These findings support a general recommendation that the identification and analysis of key ultraconserved elements should be incorporated into the genetic risk assessment of holoprosencephaly cases.

Clinical findings in patients with GLI2 mutations - phenotypic variability

Mutations in the human GLI2 gene were first reported in association with defective anterior pituitary formation, panhypopituitarism, and forebrain anomalies represented by typical holoprosencephaly (HPE) and holoprosencephaly-like (HPE-L) phenotypes and postaxial polydactyly. Subsequently, anophthalmia plus orbital anomalies, heminasal aplasia, branchial arch anomalies and polydactyly have also been incorporated into the general phenotype. Here we described six Brazilian patients with phenotypic manifestations that range from isolated cleft lip/palate with polydactyly, branchial arch anomalies to semi-lobar holoprosencephaly. Novel sequence variants were found in the GLI2 gene in patients with marked involvement of the temporomandibular joint (TMJ), a new clinical finding observed with mutations of this gene. Clinical, molecular and genetic aspects are discussed.

Zic2 regulates the kinetics of neurulation

Mutation in human ZIC2, a zinc finger protein homologous to Drosophila odd-paired, causes holoprosencephaly (HPE), which is a common, severe malformation of the brain in humans. However, the pathogenesis is largely unknown. Here we show that reduced expression (knockdown) of mouse Zic2 causes neurulation delay, resulting in HPE and spina bifida. Differentiation of the most dorsal neural plate, which gives rise to both roof plate and neural crest cells, also was delayed as indicated by the expression lag of a roof plate marker, Wnt3a. In addition the development of neural crest derivatives such as dorsal root ganglion was impaired. These results suggest that the Zic2 expression level is crucial for the timing of neurulation. Because the Zic2 knockdown mouse is the first mutant with HPE and spina bifida to survive to the perinatal period, the mouse will promote analyses of not only the neurulation but also the pathogenesis of human HPE.

Häpeän ilmeneminen äänen välityksellä Yleisradion kuunnelmassa Puhdistus (2011)

Pro gradu -tutkielmani käsittelee Yleisradion Radioteatterin kuunnelmaa Puhdistus, joka esitettiin syksyllä 2011. Tutkin feministisen affektitutkimuksen ja (audiovisuaalisen) äänitutkimuksen keinoin sitä, miten kuunnelman äänet ilmentävät Puhdistuksen tarinassa keskeisenä esiintyvää häpeäaffektia kuuntelijalle. Tutkin myös häpeään kietoutuneita tunteita ja affekteja, jotka ilmentävät häpeää tai peittävät sen. Jaan tutkimani äänet hahmojen ruumiiden tuottamiin ääniin, joihin kuuluu myös puhe, ja efekteihin. Efektejä ovat kaikki äänet, jotka eivät ole lähtöisin ihmisruumiista. Olen jakanut nämä edelleen tarinatilan ääniin eli diegeettisiin ääniin, tarinatilan ulkopuolisiin eli ei-diegeettisiin ääniin ja niihin ääniin, joita ei voi määritellä tarinatilaan kuuluviksi tai kuulumattomiksi. Puhdistuksen toisen päähenkilön Zaran ruumis näyttäytyy raskaasti hengittävänä ja nyyhkyttävänä, tuoreesti ja hallitsemattomasti häpeävänä. Toisen päähenkilön Aliiden ruumis on puolestaan omaksunut häpeäaffektin kymmeniä vuosia aiemmin, mistä johtuen hänen häpeänsä on peittynyt ylpeyden ja inhon kokemusten sekä hallitun puheäänen alle. Kumpikin päähenkilö kokee lisäksi häpeästä kumpuavaa pelkoa ja vihaa. Aliiden ja Zaran ruumiit affektoituvat paitsi häpäisijöidensä vaikutuksesta, myös kohdatessaan toisensa. Päähenkilöiden häpeää ilmentävät myös kuunnelman ääniefektit. Niistä keskeisimpiä ovat terävä kirskunta ja Aliiden hahmoon kietoutuva kärpäsensurina. Nämä molemmat luovat tulkintani mukaan ahdistavaa ja kaoottista äänimaailmaa.

Progettazione di sistema di distribuzione a cascata di ingranaggi a bassa emissione sonora per motore ad alte prestazioni

Il progetto è stato svolto in collaborazione con HPE COXA, azienda modenese dedicata alla progettazione e simulazione di elementi meccanici per il settore automotive. La finalità del progetto di tesi presentato è quella di progettare un sistema di distribuzione motore a cascata di ingranaggi dedicato ed adattato ad un motore termico V8, 4000cc già esistente, così da sostituire l’attuale sistema di distribuzione a catena e rocchetti. La peculiarità del meccanismo da progettare è la bassa emissione sonora, la quale deve essere ottenuta mediante modifiche al profilo della dentatura e ad un corretto dimensionamento del sistema. Per riuscire a soddisfare la richiesta, si utilizzano software di progettazione e simulazione (KissSoft e KissSys) che consentono di studiare la fase di ingranamento delle ruote permettendo di comprenderne il comportamento. Lo studio approfondisce poi tematiche riguardanti la natura delle modifiche effettuate, la resistenza strutturale, la distribuzione delle forze e delle tensioni e la progettazione di un sistema di ancoraggio e lubrificazione che permetta il corretto funzionamento. Il lavoro sarà completato poi con due appendici, le quali approfondiscono ancora più nel dettaglio alcune delle caratteristiche delle modifiche del profilo di dentature elicoidali, le quali non sono state prese in considerazione per il progetto della distribuzione.
