A very short-strong hydrogen bond (<2 Å, >20kcal/mol) is found in the monoanion of certain dicarboxylic acids derived from maleic and dialkylmalonic acids. Certain aromatic diamines that are known as proton sponge have exceptionally high basicity (pKa) and are only monoprotonated with strong acids like percloric acid. The closed proximity between the two basic centers provokes a strong steric interaction that is relieved upon protonation. Similar effects are found in dicarboxylic acids (hydrogen maleate and hydrogen dialkylmalonates) that present a very short distance between the two oxygens and a short-strong hydrogen bond.

Phytochemical investigation of the flowers of Wedelia paludosa afforded two diterpenes, ent-kaur-16-en-19-oic acid and ent-kaur-9(11),16-dien-19-oic acid, three acylated triterpenes, 3beta-O-hexa-decanoylolean-12-en-28-oic acid, 3beta-O-pentadecanoylolean-12-en-28-oic acid and 3beta-O-tetradecanoylolean-12-en-28-oic acid, and three steroids, stigmasterol, 3beta-O-beta-D-glycopyranosyl sitosterol and 3beta-O-beta-D-glycopyrano-sylstigmasterol. The substances were identified by their spectral data.

Crude extract of Tibouchina granulosa, Rhododendron simsii and Phaseolus vulgaris L. were prepared and used as alternative indicators in quantitative analysis teaching in standardization of NaOH solutions and in the determination of acetic acid contents in vinegar. Effect of using such natural extracts as indicators was very attractive to the students and the quantitative results were compared with conventional indicators with good agreement. Concepts of data statistics can successfully be discussed using the interest revived by the use of natural indicators.

From the leaves of Bauhinia forficata kaempferol and four flavonoid glycosides, 3,7-di-O-alpha-L-rhamnopyranosylkaempferol, 3,7-di-O-alpha-L-rhamnopyranosylquercetin, 3-O-[alpha-L-rhamnopyranosyl-(1->6)-beta-D-glucopyranosyl]-7-O-alpha-L-rhamnopyranosylkaempferol and 3-O-[alpha-L-rhamnopyranosyl-(1->6)-beta-D-glucopyranosyl]-7-O-alpha-L-rhamnopyranosylquercetin were obtained. From the flowers only the flavonoid 7-O-alpha-L-rhamnopyranosylkaempferol was isolated. The structures were established by chemical and spectroscopic methods.

The volatile constituents obtained from a static cryogenic headspace of Caesalpinia echinata Lam. (Leguminosae) showed E-beta-ocimene as the major compound (57.2%), beside other monoterpenes, C6 derivatives like n-hexanal and (E)-2-hexenal and nitrogen compounds such as indole and methyl anthranilate. From the essential oil of the leaves obtained by hydrodistillation in a Clevenger apparatus, (E)-3-hexen-1-ol was identified as the major constituent while phenolic compounds were the most representative class of secondary metabolites.

Aspects of visible spectrophotometry can be presented to students using simple experiments in which the color of the crude extract of Macroptilium lathyroides (L.) Urb. is bleached in the presence of nitrite ions in acidic medium. The dependence of the absorption intensity with time, the reaction completeness and the Beer law can be demonstrated. Quantitative results for mineral water samples "contaminated" with nitrite ions were obtained from a method based on the Griess reaction and a procedure based on the bleaching reaction between the crude extract and NO2- ions. Both the Griess and the bleaching reactions were found to be time dependent. Recoveries of about 100 - 104% were obtained with these procedures. The use of natural dyes attracted students' interest enhancing the teaching process. Experiments performed by the teaching staff suggested that the proposed methodology can be performed in a 4 h class, with relative errors ranging from 0.19 to 1.86% in relation to the Griess method.

In vitro bioassays with leave extracts of Erythrina speciosa showed promising activity against Trypanosoma cruzi. From the flowers of E. speciosa two alkaloids were isolated: erysotrine and erythartine. The leaves furnished one alkaloid, nororientaline. The structures were determined on the basis of spectroscopic evidence. This is the first report about the investigation of alkaloids in flowers and leaves of this species, as well the first report of nororientaline occurrence in this plant.

Considering the attraction of the students' attention by the changes in the colors of vegetable crude extracts caused by the variation of the pH of the medium, the use of these different colors in order to demonstrate principles of spectrophotometric acid-base titrations using the crude extracts as indicators is proposed. The experimental setup consisted of a simple spectrophotometer, a homemade flow cell and a pump to propel the fluids along the system. Students should be stimulated to choose the best wavelength to monitor the changes in color during the titration. Since the pH of the equivalence point depends on the system titrated, the wavelength must be properly chosen to follow these changes, demonstrating the importance of the correct choice of the indicator. When compared with the potentiometric results, errors as low as 2% could be found using Rhododendron simsii (azalea) or Tibouchina granulosa (Glory tree, quaresmeira) as sources of the crude extracts.

The phytochemical investigation of the flowers of Pterogyne nitens (Caesalpinioideae) resulted in the isolation and identification of nine phenolic derivatives, quercetin 3-O-sophoroside, taxifolin, astilbin, ourateacatechin, caffeic, ferulic, sinapic, chlorogenic and gallic acid, besides two guanidine alkaloids, pterogynine, pterogynidine. This is the first time these compounds have been reported in P. nitens flowers. As this is a monospecific genus, these secondary metabolites may have taxonomical significance. Their structures were assigned on the basis of spectroscopic analyses, including two-dimensional NMR techniques.

This work describes the isolation of an active flavonoid fraction and identification of isorhamnetin 3-O-β-D-(6''-acetyl)-galactopyranoside from flowers of B. perennis, and also the evaluation of anticholinesterase (AChE) activity of ethanolic extract from flowers (EEF) and the active fraction. The chemical structure of the flavonoid was defined on the basis of spectroscopic ¹H NMR, IR and UV data. EEF or flavonoid reduces AChE activity in vivo, while flavonoid also reduces AChE activity in vitro, showing a value of 1.49 mM for 50% inhibitory concentration (IC50), suggesting potential use as an insecticide or in the treatment of neurodegenerative diseases such as Alzheimer's disease.

Chemical investigation of the MeOH extract of root barks of P. pyramidallis (Tul.) L. P. Queiroz resulted in the isolation of 3,3'-dimethylellagic acid and 3,3'-dimethylellagic acid-4'-O-β-D-xyloside. Lupeol, β-sitosterol/stigmasterol and the mixture of fatty acid methyl ester derivatives were also obtained. Chromatographic procedures of the MeOH extract of the flowers of this species led to obtain an unusual mixture of fatty alcohols, β-sitosterol/stigmasterol, α-amyrin, β-amyrin and methyl gallate. The structures of the isolates were established by spectral data analysis. This is the first occurrence of 3,3'-dimethylellagic acid, 3,3'-dimethylellagic acid-4'- O-β-D-xyloside and free fatty alcohols in the Fabaceae family.

A "hérnia das crucíferas" é a principal doença na produção de brássicas na Região Metropolitana de Curitiba-PR. Fatores ambientais favoráveis, ausência de cultivares resistentes e de controle químico eficiente, aliados ao plantio sucessivo de espécies da mesma família, têm colaborado para sua disseminação. Com o objetivo de controlar a doença com menor impacto ambiental, o presente estudo verificou o efeito do pré-plantio de plantas medicinais e aromáticas na redução de inóculo de Plasmodiophora brassicae. Dois experimentos foram conduzidos em casa de vegetação no Setor de Ciências Agrárias da Universidade Federal do Paraná, nos períodos de junho a novembro de 2003 e julho a dezembro de 2004. O delineamento foi inteiramente casualizado com nove tratamentos e seis repetições. As plantas utilizadas nos tratamentos foram: menta (Mentha piperita L.); alfavaca (Ocimum basilicum L.); bardana (Arctium minus Hill); calêndula (Calendula officinalis L.); cebolinha (Allium fistulosum L.); salsa (Petroselinum hortense Hoffm) e sálvia (Salvia officinalis L.), e como testemunhas foram utilizados solos sem patógeno e solo infestado, ambos em pousio. Utilizaram-se vasos contendo 3 Kg de solo esterilizado via vapor e como inóculo de P. brassicae foram utilizados 2,5 g de galhas por vaso. A rúcula (Eruca sativa Mill) foi o hospedeiro suscetível utilizado. Após 45 dias do plantio da rúcula foram determinadas a massa aérea fresca, incidência de galhas galhas e o índice de severidade. A maior massa aérea fresca e as menores severidades foram obtidas no experimento de 2003, nos tratamentos com o pré-plantio das medicinais bardana, salsa, menta, alfavaca e cebolinha.

Um estudo da ontogênese das caneluras induzidas em ramos de laranjeiras doces suscetíveis por isolados severos do vírus da tristeza dos citros (Citrus tristeza vírus - CTV) foi feito usando-se como modelo pedúnculos florais e de frutos. O menor calibre destes órgãos permite um melhor acompanhamento do processo. As observações foram feitas em laranjeira cv. Pêra infetada pelo isolado severo Capão Bonito do CTV. Cinco fases do processo de formação de caneluras puderam ser deduzidas pelas análises anatômicas. As primeiras alterações são representadas pelo aparecimento de células adensadas, hipertrofia e hiperplasia no parênquima e câmbio do floema e uma desorganização generalizada desta área. Segue-se uma atividade intensa do câmbio do floema adjacente e sua expansão em direção ao xilema. Esta invasão do xilema resulta na ruptura do anel do xilema pela massa celular do floema constituída de células recém formadas de parede celular delgada. Esta invasão do floema em direção ao xilema inicia um processo de degeneração dos vasos e parênquima do xilema. Finalmente há um colapso completo da região do xilema invadida, que é substituída pela massa do floema, resultando na canelura, notada ao se remover a casca.

Considerando a importância do cultivo de flores tropicais no contexto nacional e internacional, buscou-se realizar um levantamento da ocorrência de fungos e nematóides associados às mesmas, em áreas de cultivos de flores tropicais em São Luís - MA. Foram realizadas visitas periódicas, em intervalos bimestrais, nos locais onde a principal atividade era o cultivo de flores tropicais, para o monitoramento e coleta de plantas ou partes de plantas das espécies: Heliconia spp., Alpinia purpurata e Etlingera elatior com sintomatologia típica de doenças. Realizou-se, ainda, o teste de patogenicidade dos principais fungos detectados como agentes causais das manchas foliares. Os resultados obtidos confirmaram a existência de fungos associados aos cultivos de flores tropicais em São Luís, com destaque para Curvularia eragrostides (78 %), Pestalotiopsis sp. (68 %) e Colletotrichum gloeosporioides (47 %) como agentes causais de manchas foliares em espécies da família Heliconiaceae, e Curvularia eragrostides (75 %), Pestalotiopsis sp. (37 %) em espécies da família Zingiberaceae. Foram registrados oito gêneros de nematóides, tanto na família Heliconiaceae, quanto na Zingiberaceae, destacando-se o gênero Meloidogyne.
