Atividades relacionadas com a extração do petróleo, bem como as atividades derivadas envolvem grandes riscos, que podem ser ambientais ou à saúde humana. Podem ser de forma direta, como os derramamentos, ou indireta, como resíduos gerados pela sua utilização, poluindo ar, corpos d’água e solos. A biorremediação consiste na utilização de microrganismos tais como as bactérias, fungos filamentosos e leveduras para remediar um ambiente contaminado, transformando os compostos em algo pouco tóxico ou sem nenhuma toxicidade. A ciência que estuda os efeitos tóxicos nos organismos e no ambiente se chama toxicologia. Por isso existe uma íntima ligação entre essas duas ciências. O presente projeto tem como objetivo estudar a toxicidade de óleos lubrificantes automotivos (mineral novo, sintético novo e usado), óleos vegetais (novo e usado) e biodiesel durante sua biodegradação em ambiente terrestre. Análises descritivas e comparativas são as bases para o estudo e compreensão da biodegradação dos contaminantes aqui citados. Os testes toxicológicos darão uma idéia de biodegradação dos diferentes contaminantes no solo. Para os testes de toxicidade serão utilizadas sementes de Eruca sativa (rúcula), sementes de Lactuca sativa (alface) e Eisenia andrei (minhoca). Após 180 dias de biodegradação dos compostos pôde-se concluir que o óleo lubrificante sintético é o mais biodegradável e os demais contaminantes necessitam de tempo de biodegradação superior a 6 meses para que sua toxicidade fique baixa.

The hydroponic cultivation of vegetables has increased markedly in recent years, however, little is known about its chemical composition, which is of extreme importance in view of changing food habits of a considerable fraction of the population seeking food quality. For this reason, cultivation of watercress, chicory, rocket and lettuce American (Lucy Brown) and smooth (cv. Regina) were grown in hydroponics, NFT system, aiming to evaluate the yield, nitrate content and chemical composition. The experiment was conducted in a greenhouse at the Universidade Federal de Santa Catarina, Florianopolis city in the Santa Catarina State (Brazil), from may to july 2004 under a completely randomized design with four replications. The spacing used was 25 x 25 cm for the crops of watercress, chicory and lettuce (cv. Lucy Brown and cv. Regina) and 5 x 5 cm to the rocket. The traits were: number of leaves, nitrate content and chemical composition of vegetables. The lettuce cv. Regina and chicory had higher number of leaves per plant. The watercress had lower water content and higher dry mass of shoots. The largest increase in fresh weight was obtained in chicory, lettuce, cv. Regina. Higher levels of lipids, protein, ash, carbohydrates, calories, fiber and nitrate were obtained from the watercress. The rocket had lower values for the variables fresh and dries the whole plant, shoot and root and leaf number per plant. All cultures showed good visual appearance, low calorie and nitrate levels suitable for human consumption.

Foods that provide medical and health benefits or have a role in disease risk prevention are termed functional foods. The functionality of functional foods is derived from bioactive compounds that are extranutritional constituents present in small quantities in food. Bioactive components include a range of chemical compounds with varying structures such as carotenoids, flavonoids, plant sterols, omega-3 fatty acids (n-3), allyl and diallyl sulfides, indoles (benzopyrroles), and phenolic acids. The increasing consumer interest in natural bioactive compounds has brought about a rise in demand for these kinds of compounds and, in parallel, an increasing number of scientific studies have this type of substance as main topic. The principal aim of this PhD research project was the study of different bioactive and toxic compounds in several natural matrices. To achieve this goal, chromatographic, spectroscopic and sensorial analysis were performed. This manuscript reports the main results obtained in the six activities briefly summarized as follows: • SECTION I: the influence of conventional packaging on lipid oxidation of pasta was evaluated in egg spaghetti. • SECTION II: the effect of the storage at different temperatures of virgin olive oil was monitored by peroxide value, fatty acid activity, OSI test and sensory analysis. • SECTION III: the glucosinolate and phenolic content of 37 rocket salad accessions were evaluated, comparing Eruca sativa and Diplotaxis tenuifolia species. Sensory analysis and the influence of the phenolic and glucosinolate composition on sensory attributes of rocket salads has been also studied. • SECTION IV: ten buckwheat honeys were characterised on the basis of their pollen, physicochemical, phenolic and volatile composition. • SECTION V: the polyphenolic fraction, anthocyanins and other polar compounds, the antioxidant capacity and the anty-hyperlipemic action of the aqueous extract of Hibiscus sabdariffa were achieved. • SECTION VI: the optimization of a normal phase high pressure liquid chromatography–fluorescence detection method for the quantitation of flavanols and procyanidins in cocoa powder and chocolate samples was performed.

Oligosaccharide synthesis is an important cryoprotection strategy used by woody plants during winter dormancy. At the onset of autumn, starch stored in the stem and buds is broken down in response to the shorter days and lower temperatures resulting in the buildup of oligosaccharides. Given that the enzyme DSP4 is necessary for diurnal starch degradation in Arabidopsis leaves, this study was designed to address the role of DSP4 in this seasonal process in Castanea sativa Mill. The expression pattern of the CsDSP4 gene in cells of the chestnut stem was found to parallel starch catabolism. In this organ, DSP4 protein levels started to rise at the start of autumn and elevated levels persisted until the onset of spring. In addition, exposure of chestnut plantlets to 4 °C induced the expression of the CsDSP4 gene. In dormant trees or cold-stressed plantlets, the CsDSP4 protein was immunolocalized both in the amyloplast stroma and nucleus of stem cells, whereas in the conditions of vegetative growth, immunofluorescence was only detected in the nucleus. The studies indicate a potential role for DSP4 in starch degradation and cold acclimation following low temperature exposure during activity–dormancy transition.

The European chestnut (Castanea sativa Mill.) is a multipurpose species that has been widely cultivated around the Mediterranean basin since ancient times. New varieties were brought to the Iberian Peninsula during the Roman Empire, which coexist since then with native populations that survived the last glaciation. The relevance of chestnut cultivation has being steadily growing since the Middle Ages, until the rural decline of the past century put a stop to this trend. Forest fires and diseases were also major factors. Chestnut cultivation is gaining momentum again due to its economic (wood, fruits) and ecologic relevance, and represents currently an important asset in many rural areas of Europe. In this Thesis we apply different molecular tools to help improve current management strategies. For this study we have chosen El Bierzo (Castile and Leon, NW Spain), which has a centenary tradition of chestnut cultivation and management, and also presents several unique features from a genetic perspective (next paragraph). Moreover, its nuts are widely appreciated in Spain and abroad for their organoleptic properties. We have focused our experimental work on two major problems faced by breeders and the industry: the lack of a fine-grained genetic characterization and the need for new strategies to control blight disease. To characterize with sufficient detail the genetic diversity and structure of El Bierzo orchards, we analyzed DNA from 169 trees grafted for nut production covering the entire region. We also analyzed 62 nuts from all traditional varieties. El Bierzo constitutes an outstanding scenario to study chestnut genetics and the influence of human management because: (i) it is located at one extreme of the distribution area; (ii) it is a major glacial refuge for the native species; (iii) it has a long tradition of human management (since Roman times, at least); and (iv) its geographical setting ensures an unusual degree of genetic isolation. Thirteen microsatellite markers provided enough informativeness and discrimination power to genotype at the individual level. Together with an unexpected level of genetic variability, we found evidence of genetic structure, with three major gene pools giving rise to the current population. High levels of genetic differentiation between groups supported this organization. Interestingly, genetic structure does not match with spatial boundaries, suggesting that the exchange of material and cultivation practices have strongly influenced natural gene flow. The microsatellite markers selected for this study were also used to classify a set of 62 samples belonging to all traditional varieties. We identified several cases of synonymies and homonymies, evidencing the need to substitute traditional classification systems with new tools for genetic profiling. Management and conservation strategies should also benefit from these tools. The avenue of high-throughput sequencing technologies, combined with the development of bioinformatics tools, have paved the way to study transcriptomes without the need for a reference genome. We took advantage of RNA sequencing and de novo assembly tools to determine the transcriptional landscape of chestnut in response to blight disease. In addition, we have selected a set of candidate genes with high potential for developing resistant varieties via genetic engineering. Our results evidenced a deep transcriptional reprogramming upon fungal infection. The plant hormones ET and JA appear to orchestrate the defensive response. Interestingly, our results also suggest a role for auxins in modulating such response. Many transcription factors were identified in this work that interact with promoters of genes involved in disease resistance. Among these genes, we have conducted a functional characterization of a two major thaumatin-like proteins (TLP) that belongs to the PR5 family. Two genes encoding chestnut cotyledon TLPs have been previously characterized, termed CsTL1 and CsTL2. We substantiate here their protective role against blight disease for the first time, including in silico, in vitro and in vivo evidence. The synergy between TLPs and other antifungal proteins, particularly endo-p-1,3-glucanases, bolsters their interest for future control strategies based on biotechnological approaches.

Ink Disease is considered one of the most important causes of the decline of chestnut orchards. The break in yield of Castanea sativa Mill is caused by two species: Phytophthora cinnamomi and Phytophthora cambivora, being the first one the foremost pathogen of ink disease in Portugal. P. cinnamomi is one of the most aggressive and widespread plant pathogen with nearly 1,000 host species. This oomycete causes enormous economic losses and it is responsible for the decline of many plant species in Europe and worldwide. Up to now no efficient treatments are available to fight these pathogens. Because of the importance of chestnut at economical and ecological levels, especially in Portugal, it becomes essential to explore the molecular mechanisms that determine the interaction between Phytophthora species and host plants through the study of proteins GIP (glucanase inhibitor protein) and NPP1 (necrosis-inducing Phytophthora protein 1) produced by P. cinnamomi during the infection. The technique of RNA interference was used to knockdown the gip gene of P. cinnamomi. Transformants obtained with the silenced gene have been used to infect C. sativa, in order to determine the effect of gene silencing on the plant phenotype. To know more about the function of GIP and NPP1 involved in the mechanism of infection, the ORF’s of gip and npp1 genes have been cloned to the pTOR-eGFP vector for a future observation of P. cinnamomi transformants with fluorescent microscopy and determination of the subcellular localization. Moreover the prediction by bioinformatics tools indicates that both GIP and NPP1 proteins are secreted. The results allow to predict the secretory destination of both GIP and NPP1 proteins and confirm RNAi as a potential alternative biological tool in the control and management of P. cinnamomi. A concentração na castanha de minerais como o cálcio, potássio, o magnésio e outros tem sido estudada tradicionalmente por absorção atómica ou por espectrofotometria de UV-VIS [1-2]. Neste trabalho foi estudada a composição elementar de duas variedades (Longal e Judia) de castanha europeia (Castanea sativa Mill.) com a técnica PIXE (Proton Induced X-ray Emission). Os resultados preliminares mostram que a composição difere de acordo com a variedade, nos elementos maioritários e nos minoritários. Este estudo é parte de um projecto mais amplo, que visa avaliar o efeito do processamento pós-colheita de alimentos com feixe de electrões e raios-X.

Chestnut flowers, lemon balm plants and their decoctions were incorporated into "Serra da Estrela" cheese, to assess their potential to preserve its nutritional properties and provide new foodstuffs. The analyses were carried out after the normal ripening period of 1month and after 6months of storage. The most abundant nutrients were proteins and fats. The most abundant minerals were Ca and Na, while C16:0 and C18:1 were the main fatty acids. Saturated fatty acids were the most abundant, followed by the monounsaturated. Moisture seemed to be lower in the samples with the plants incorporated. The dried plants, when incorporated, seemed to be more efficient as preservers then the decoctions, although these better preserved the proteins. These plants can be regarded as promising natural preservers in foodstuffs cheese, given the preservation of key parameters and the slight impact on the nutritional value.

Molecular characterization represents a valid support for the recovery of germoplasm, also motivated by the interest for the valorization of local productions in order to make their traceability possible. Molecular characterization is also fundamental for the individuation of misnomers in collection fields in which the different varieties are preserved. In particular, microsatellites have been used in this research to investigate the genetic diversity, inside a population and at an individual level, and the correct varietal correspondence. The research is mainly based on the study of European chestnut (Castanea sativa Mill.) cultivars to evaluate the genetic diversity and relationships in Emilia-Romagna region (Italy). A STRUCTURE analysis was carried out at European level with the allelic frequencies of the samples collected in Emilia-Romagna. Variation found at group and subgroup level may reflect a combination of historical migration/selection processes and adaptive factors to different environments between Italian and Spanish regions. In addition, a case study for the valorization of an old local variety and its re-introduction in the cultivation areas was proposed. This research was carried out by a morphological and molecular characterization of the local apple variety 'Rosa Romana'. The conservation of this variety entails the discrimination of different accessions with very similar phenotype that are present in the original cultivation area. The identification of historical trees and most adequate reference plants are fundamental steps for the correct propagation of this old variety and for the development of nursery activities. This will also promote and re-evaluate the exploitation and protection of such ancient Italian apple cultivars. This model could be in future also carried out for chestnut varieties. In conclusion, analysis with molecular markers is of fundamental importance for the protection and the maintenance of local and ancient varieties which allow to increase the allelic variability available for breeding programs.

L’acqua attivata al plasma (PAW), ottenuta esponendo una soluzione d’acqua al plasma freddo, è recentemente emersa come un’alternativa promettente per la decontaminazione degli alimenti freschi in sostituzione all’uso dei tradizionali disinfettanti al cloro. Lo scopo della tesi è stato quello di valutare l’effetto dei lavaggi con PAW su foglie di rucola (Eruca sativa), già testati per la decontaminazione microbica, sulle caratteristiche qualitative del prodotto durante lo stoccaggio. La PAW è stata ottenuta tramite un generatore ad alta tensione collegato ad un elettrodo a corona operante con una scarica di 12 kV a 5 Hz su acqua distillata per 60 secondi. I lavaggi con PAW per 2, 5 e 10 minuti sono stati confrontati con dei lavaggi attuati con acqua distillata non trattata ed una soluzione di ipoclorito di sodio (100 ppm). Durante la conservazione è stata monitorata l’evoluzione della composizione dello spazio di testa delle buste, il contenuto di sostanza secca, il grado di appassimento e le modifiche inerenti al colore delle foglie. I risultati hanno mostrato come i principali parametri qualitativi non siano risultati significativamente modificati a seguito dell’ immersione nella PAW, rispetto al trattamento con acqua distillata o con la soluzione di ipoclorito di sodio. Nel complesso, i risultati sperimentali hanno evidenziato come la PAW sia una tecnologia promettente, nonostante ciò, è necessario condurre ulteriori studi al fine di ottimizzare i parametri di processo.

Three welding procedures used to rebuild worn shafts in sugar cane mills were analysed: two submerged arc welding processes and one flux cored arc welding (FCAW) process. Sliding wear tests were in accordance with ASTM G 77 standard, using rings of welding material, blocks of bronze SAE 67, and oil as lubricant. The worn surfaces of rings and blocks were analysed by scanning electron microscopy to determine the wear mechanisms. High contact pressure, high operating temperature, and low relative speed were applied in sliding wear tests to match the conditions in sugar cane mills. Transferred material and evidence of adhesive junctions were detected. Additionally, hardened fragments produced abrasive grooves on the worn surfaces. The welding deposits that presented strong adhesion on the worn surface showed higher mass loss than the materials that presented more abrasive characteristics. Plastic mechanical properties were measured and related to the mass loss. The tested materials presented similar hardness but different yield stress and hardening coefficient. A relationship between wear, strain hardening coefficient, and yield stress was found. The welding deposit that presented the highest hardening coefficient showed the highest mass loss, with evidence of severe adhesion on the worn surface.
