We examined the effect of two rhesus papillomavirus 1 (RhPV) oncogenes on cytokine-induced signal transduction pathways leading to the possible activation of Ras protein (p21ras) and phosphatidylinositol kinase. p21ras in both the activated (GTP-bound) and inactivated (GDP-bound) states were quantitated. NIH 3T3 cell lines expressing the RhPV 1 E5 gene or epidermal growth factor receptor cDNA had about a sixfold higher ratio of p21ras-bound GTP to p21ras-bound GDP as compared with parental NIH 3T3 cells or a cell line expressing the RhPV 1 E7 gene under normal culture conditions, yet expressed similar levels of p21ras. Quiescent cells had dramatically reduced levels of activated p21ras, except those containing RhPV 1 E7. Levels were restored by stimulation with epidermal growth factor or platelet-derived growth factor. Both epidermal growth factor and platelet-derived growth factor receptor of RhPV 1 E5- and E7-containing cells responded to cytokine stimulation. Endogenous phosphatidylinositol-3′-kinase was up-regulated in NIH 3T3 cells transformed with the E5 genes of RhPV 1 and bovine papillomavirus 1. These results suggest that E5 genes of papillomaviruses play a major role in the regulation of transduction pathways.

The Picornaviridae family consists of positive-strand RNA viruses that are the causative agents of a variety of diseases in humans and animals. Few drugs targeting picornaviruses are available, making the discovery of new antivirals a high priority. Here, we identified and characterized three compounds from a library of kinase inhibitors that block replication of poliovirus, coxsackievirus B3, and encephalomyocarditis virus. The antiviral effect of these compounds is not likely related to their known cellular targets because other inhibitors targeting the same pathways did not inhibit viral replication. Using an in vitro translation-replication system, we showed that these drugs inhibit different stages of the poliovirus life cycle. A4(1) inhibited the formation of a functional replication complex, while E5(1) and E7(2) affected replication after the replication complex had formed. A4(1) demonstrated partial protection from paralysis in a murine model of poliomyelitis. Poliovirus resistant to E7(2) had a single mutation in the 3A protein. This mutation was previously found to confer resistance to enviroxime-like compounds, which target either PI4KIIIβ (major enviroxime-like compounds) or OSBP (minor enviroxime-like compounds), cellular factors involved in lipid metabolism and shown to be important for replication of diverse positive-strand RNA viruses. We classified E7(2) as a minor enviroxime-like compound, because the localization of OSBP changed in the presence of this inhibitor. Interestingly, both E7(2) and major enviroxime-like compound GW5074 interfered with the viral polyprotein processing. Multiple attempts to isolate resistant mutants in the presence of A4(1) or E5(1) were unsuccessful, showing that effective broad-spectrum antivirals could be developed on the basis of these compounds. Studies with these compounds shed light on pathways shared by diverse picornaviruses that could be potential targets for the development of broad-spectrum antiviral drugs.

The effect of adjuvant on induction of human papillomavirus type 16 E7 protein-specific cytotoxic T lymphocytes (CTL) and immunoglobulin G (IgG)(2a) antibody was studied in C57BL/6 J mice immunized with various adjuvants and E7 protein. Quil-A adjuvant, but not complete Freund's adjuvant (CFA) or Algammulin, induced a T-helper 1 (Th1)-type response to E7, which was characterized by CTL activity against a tumour cell line transfected with E7 protein and by E7-specific IgG(2a). All tested adjuvants elicited comparable levels of E7-specific IgG(1). The longest duration and greatest magnitude of CTL response was seen following two immunizations with the highest dose of E7 and Quil-A. Simultaneous immunization with a Th1 and a T helper 2 (Th2)-promoting adjuvant gave a Th1-type response. However, E7 and Quil-A were unable to induce a Th1-type response (as measured by the inability to generate anti-E7 IgG(2a) antibody) in animals with a pre-existing Th2-type response to E7. These results suggest that saponin adjuvants may be suitable for immunotherapy in humans where a Th1-type response is sought, provided that there is no pre existing Th2-type response to the antigen.

Three exceptional modular invariants of SU(4) exist at levels 4, 6 and 8. They can be obtained from appropriate conformal embeddings and the corresponding graphs have self-fusion. From these embeddings, or from their associated modular invariants, we determine the algebras of quantum symmetries, obtain their generators,and, as a by-product, recover the known graphs E4, E6 and E8 describing exceptional quantum subgroups of type SU(4). We also obtain characteristic numbers (quantum cardinalities, dimensions) for each of them and for their associated quantum groupoïds.

We report the prevalence of human papillomavirus type 16 (HPV-16) variants in women with cervical lesions from the Federal District, Central Brazil. We analyzed 34 HPV-16 samples, identifying the sequence variations of E6 and L1 genes and correlating variant frequency with disease status. The most prevalent HPV-16 variant was the European (50%), followed by Asian-American (41.2%), African-1 (5.9%), and African-2 (2.9%). European and non-European variants appeared in equal frequencies among the cytological types of lesions - atypical squamous or glandular cells of undetermined significance, cytological alterations suggesting HPV infection, cervical intraepithelial neoplasias, squamous cell carcinoma, and adenocarcinoma.

Cervical cancer, the second leading cause of cancer mortality in women worldwide, results from infection with a subset of human papillomaviruses (HPV), HPV-16 being the most prevalent type. The available prophylactic vaccines are an effective strategy to prevent this cancer in the long term. However, they only target 70-80% of all cervical cancers and cannot control existing HPV infections and associated lesions. Therapeutic vaccines are thus necessary for women who cannot benefit from prophylactic vaccination. Induction of protective immune responses in the genital mucosa (GM) may be crucial for efficacy of HPV therapeutic vaccines. We report here that mice that received a single subcutaneous (s.c.) vaccination of an adjuvanted long synthetic HPV16 E7(1-98) polypeptide showed induction of 100% tumor protection against s.c. TC-1 tumors and that tumor regression was mainly provided by CD8 T cells. In vivo cytotoxic assay revealed high E7-specific cytolytic T lymphocytes activity in spleen and in genital draining lymph nodes (LN), and E7-specific CD8 T cells could be detected in GM by tetramer staining. More importantly, high-avidity E7-specific INF-gamma secreting CD8 T cells were induced not only in blood, spleen and LN but also in GM of vaccinated mice, thus providing evidence that a parenteral vaccination may be sufficient to provide regression of genital tumors. In addition, there was no correlation between the responses measured in blood with those measured in GM, highlighting the necessity and relevance to determine the immune responses in the mucosa where HPV-tumors reside.

Foram testadas diferentes fontes de nitrogênio na indução e enraizamento de brotações axilares de Chrysanthemum morifolium. Manteve-se constante o teor de nitrogênio total (60 mM) em todos os tratamentos realizados, variando-se apenas as fontes nitrogenadas. O tratamento E1 constituiu-se do meio MS (Murashige & Skoog 1962) completo. Os demais tratamentos foram: E2 = amônio; E3 = nitrato; E4 = nitrato+uréia (1,65 mM); E5 = nitrato+uréia (3,33 mM); E6 = uréia; E7 = uréia+glutamina e E8 = glutamina. Em nenhum tratamento foram adicionados fitorreguladores. No tratamento E1 houve um favorecimento de desenvolvimento de plantas de crisântemo in vitro tanto da parte aérea como de raízes. Porém, a utilização apenas de nitrato como única fonte de nitrogênio (tratamento E3) foi suficiente para sustentar o desenvolvimento das plantas, pois, para a maioria dos parâmetros analisados não foi observada diferença significativa em relação às plantas mantidas no meio básico de MS. A presença de uréia no meio de cultura incrementou o desenvolvimento de raízes de crisântemo, principalmente quando utilizada em adição ao nitrato (tratamentos E4 e E5), proporcionando um sistema radicular bastante denso. Quando utilizadas como única fonte de nitrogênio, tanto a glutamina (tratamento E7) como o íon amônio (tratamento E2) não se mostraram eficientes. O resultado do uso das fontes mencionadas foi um baixo desenvolvimento das plantas, como evidenciado pelos baixos valores de crescimento obtidos em todos os parâmetros analisados.

Ihmisen papilloomavirukset (HPV) on yhdistetty arviolta 28 %:iin virusten aiheuttamista syövistä, joista yksi esiintymispaikka on pään ja kaulan alue. Metylaatio on yksi syövän syntyyn liitetty muutos solujen genomissa. Tämän syventävän työn tutkimuksen aiheena oli metylaatiomuutosten vaikutus E2-proteiinien sitoutumisessa HPV-tyypin 16 genomin onkogeeneina tunnettujen E6- ja E7-geenien alueelle (E2BS). Tarkoituksena oli pystyttää tutkimuslinja, jolla kyseisten sitoutumisalueiden metylaatioasteiden muutokset ja variaatiot voitaisiin havaita HPV-infektoituneissa solulinjoissa. Lähtökohtana käytettiin Chaiwongkot ym. (2012) aiheesta julkaisemaa artikkelia, jonka tulokset pyrittiin toistamaan. Matalan (SiHa) ja korkean (CaSki) asteen HPV-infektoiduista kohdunkaulan levyepiteelisyöpäsolulinjoista eristetty DNA bisulfiittikäsiteltiin, monistettiin PCR:llä ja pyrosekvenoitiin. Saatujen sekvenssien metylaatioasteita verrattiin keskenään sekä aiempiin tutkimuksiin. Tässä syventävien opintojen työssä saatiin pääosin toistettua Chaiwongkotin ym. (2012) saadut tulokset CaSki- ja SiHa-solujen metylaatiosta. CaSki-soluilla E2BS1-alueen metylaatioaste oli keskimäärin 38–42 % (s± 4,1–4,3), E2BS2-alueen 13 % (s± 0,6) ja E2BS3-4alueella 92–99 % (s± 11–42). SiHa-soluilla vastaavat luvut olivat 4-7 %, 2 % ja 5-8 %. CaSki- ja SiHa-soluilla E2BS-alueiden metylaatio oli verrannollinen HPV:n suureen kopiolukuun. Vähäisin merkitys oli E2BS2-alueella, kun taas E2BS1- ja E2BS3-4-alueet olivat vahvemmin metyloituneet. Menetelmässä kohdattuihin ongelmiin kuului E2BS1-alueen pyrosekvenointitulosten jääminen virhemarginaalien ulkopuolelle sekä itse pyrosekvenointimenetelmä.

Le VPH-16 de même que certains VPH, dont le VPH-18, causent le cancer du col utérin. Son intégration dans le génome humain pourrait être un marqueur de progression de l’infection. Les charges virales totale et intégrée sont présentement mesurées en quantifiant par PCR en temps réel les gènes E6 (RT-E6) et E2 (RT-E2-1) du VPH-16. Nous avons évalué l’impact du polymorphisme du gène E2 sur la quantification de l’ADN du VPH-16 dans des spécimens cliniques. Dans un premier temps, le gène E2 de 135 isolats de VPH-16 (123 appartenaient au clade Européen et 12 à des clades non- Européens) fut séquencé. Ensuite, un test de PCR en temps réel ciblant les séquences conservées dans E2 (RT-E2-2) fut développé et optimisé. Cent trente-neuf spécimens (lavages cervicaux et vaginaux) provenant de 74 participantes (58 séropositives pour le VIH, 16 séronégatives pour le VIH) ont été étudiés avec les trois tests E2 (RT-E2-2), E6 (RT-E6) et E2 (RT-E2-1). Les ratios de la quantité d’ADN de VPH-16 mesuré avec RT-E2-2 et RT-E2-1 dans les isolats Européens (médiane, 1.02; intervalle, 0.64-1.80) et Africains 1 (médiane, 0.80; intervalle, 0.53-1.09) sont similaires (P=0.08). Par contre, les ratios mesurés avec les isolats Africains 2 (médiane, 3.23; intervalle, 1.92-3.49) ou Asiatique- Américains (médiane, 3.78; intervalle, 1.47-37) sont nettement supérieurs à ceux obtenus avec les isolats Européens (P<0.02 pour chaque comparaison). Les distributions des quantités de E2 contenues dans les 139 échantillons mesurées avec RT-E2-2 (médiane, 6150) et RT-E2-1 (médiane, 8960) étaient statistiquement différentes (P<0.0001). Nous avons observé que les charges virales totale (odds ratio (OR) OR, 2.16 95% intervalle de confiance (IC) 1.11-4.19), et épisomale du VPH-16 (OR, 2.14 95% IC 1.09-4.19), mais pas la présence de formes intégrées (OR, 3.72 95% IC 1.03-13.4), sont associées aux néoplasies intraepitheliales cervicales de haut grade (CIN-2,3), et ce, en contrôlant pour des facteurs confondants tels que l’âge, le taux de CD4 sanguin, l’infection au VIH, et le polymorphisme de VPH-16. La proportion des échantillons ayant un ratio E6/E2 > 2 pour les femmes sans lésion intraépithéliale (7 de 35) est similaire à celle des femmes avec CIN-2,3 (5 de 11, p=0.24) ou avec CIN- 1 (4 de14, P=0.65). Le polymorphisme du gène E2 est un facteur qui influence la quantification des charges intégrées de VPH-16.

Non-Hodgkin's lymphomas are common tumors of the human immune system, primarily of B cell lineage (NHL-B). Negative growth regulation in the B cell lineage is mediated primarily through the TGF-β/SMAD signaling pathway that regulates a variety of tumor suppressor genes. Ski was originally identified as a transforming oncoprotein, whereas SnoN is an isoform of the Sno protein that shares a large region of homology with Ski. In this study, we show that Ski/SnoN are endogenously over-expressed both in patients' lymphoma cells and NHL-B cell lines. Exogenous TGF-β1 treatment induces down-regulation of Ski and SnoN oncoprotein expression in an NHL-B cell line, implying that Ski and SnoN modulate the TGF-β signaling pathway and are involved in cell growth regulation. Furthermore, we have developed an NHL-B cell line (DB) that has a null mutation in TGF-β receptor type II. In this mutant cell line, Ski/SnoN proteins are not down-regulated in response to TGF-β1 treatment, suggesting that downregulation of Ski and SnoN proteins in NHL-B require an intact functional TGF-β signaling pathway Resting normal B cells do not express Ski until activated by antigens and exogenous cytokines, whereas a low level of SnoN is also present in peripheral blood Go B cells. In contrast, autonomously growing NHL-B cells over-express Ski and SnoN, implying that Ski and SnoN are important cell cycle regulators. To further investigate a possible link between reduction of the Ski protein level and growth inhibition, Ski antisense oligodeoxynucleotides were transfected into NHL-B cells. The Ski protein level was found to decrease to less than 40%, resulting in restoring the effect of TGF-β and leading to cell growth inhibition and G1 cell cycle arrest. Co-immunoprecipitation experiments demonstrated that Ski associates with Smad4 in the nucleus, strongly suggesting that over-expression of the nuclear protein Ski and/or SnoN negatively regulates the TGF-β pathway, possibly by modulating Smad-mediated tumor suppressor gene expression. Together, in NHL-B, the TGF-β/SMAD growth inhibitory pathway is usually intact, but over-expression of the Ski and/or SnoN, which binds to Smad4, abrogates the negative regulatory effects of TGF-β/SMAD in lymphoma cell growth and potentiates the growth potential of neoplastic B cells. ^

The E2F transcription factors play a key role in the regulation of cellular proliferation and terminal differentiation. E2F6 is the most recently identified and the least well understood member of the E2F family. It is only distantly related to the other E2Fs and lacks the sequences responsible for both transactivation and binding to the retinoblastoma protein. Consistent with this finding, E2F6 can behave as a dominant negative inhibitor of the other E2F family members. In this study, we continue to investigate the possible role(s) of E2F6 in vivo. We report the isolation of RYBP, a recently identified member of the mammalian polycomb complex, as an E2F6-interacting protein. Mapping studies indicate that RYBP binds within the known “repression domain” of E2F6. Moreover, we demonstrate that endogenous E2F6 and polycomb group proteins, including RYBP, Ring1, MEL-18, mph1, and the oncoprotein Bmi1, associate with one another. These findings suggest that the biological properties of E2F6 are mediated through its ability to recruit the polycomb transcriptional repressor complex.

In Brazil, there is a high incidence of venomous animals. Among them, scorpions are highlighted by their medical importance, and for being their venom a source of several molecules with biological and pharmacological activity not yet fully understood, including several bioactive peptides. Antimicrobial peptides (AMPs) are components of the immune system in prokaryotes and eukaryotes, used in the first line of defense against microorganisms. In the present study, we characterized the first PAM previously identified through transcriptome of the venom gland of the scorpion Tityus stigmurus, named Stigmurin. The characteristics of Stigmurin were investigated by computational modeling and construction of dendrogram. In vitro tests investigated the antibacterial, antifungal, haemolytic and cytotoxic effects of crude venom and Stigmurin. In addition, the structural characteristics of Stigmurin were investigated by circular dochroism in water, 2, 2 , 2- trifluoethanol (TFE) and sodium dodecyl sulfate (SDS) and the models were refined by molecular dynamics simulations. The results showed that the selected sequence encodes a mature protein of 17 amino acid residues and the dendrogram reveals a case of convergent evolution. The crude venom showed no antimicrobial activity, however, Stigmurin exhibited a broad spectrum of antibacterial activity, with minimal inhibitory concentrations (MIC) ranging from 31.25 and 250 µg/mL for different strains, while the hemolytic activity at these concentrations was low. In cytotoxicity studies, the crude venom was unable to reduce cell viability in VERO E6 cells; in contrast, its activity in SiHa cells was significantly higher, corresponding to IC50 of 3.6 µg/mL. For Stigmurin the concentration sable to decrease cell viability of Vero E6 and SiHa cells in 50% were 275.67 µg/mL and 212.54 µg/mL, respectively. The dichroism spectra revealed the conformational flexibility, with predominating extended and β–sheet structures, as well as a remark able renaturation ability. The results suggest that Stigmurin could be considered as a potential antiinfective drug

Los adenovirus oncolífticos ofrecen un tratamiento muy prometedor para combatir al cáncer. Estos virus se replican pobremente en células de ratón, por lo que para evaluar su eficacia para destruir tumores, usualmente se implantan células tumorales humanas en las cuales se replican los virus, pero con el inconveniente que se deben usar ratones inmunosuprimidos, para evitar que su sistema inmune elimine estas células por ser de diferente especie. Sin embargo, estos ratones no representan un modelo adecuado para evaluar la eficacia terapéutica de dichos adenovirus en ensayos preclfnicos, ya que carecen de mecanismos de defensa propios del sistema inmune que pudiesen coadyuvar en la protección contra el tumor y/o eliminación del adenovirus. Actualmente no existe un modelo de cáncer de cervix en ratones silvestres (inmunocompetentes) en los cuales los tumores se desarrollen a partir de celulas murinas y que los virus oncolfticos se repliquen eficientemente, al igual que en las células humanas, y se permita evaluar su efecto antitumoral. Se ha demostrado que los genes E6 y E7 del HPV-16 facilitan la replicación de los adenovirus. En este trabajo por primera vez se evalu6 1a capacidad de replicación de un adenovirus oncolftico en Ia lfrrea celular murina TC-1 Ia cual posee los genes E6 y E7 del HPV-16. Demostramos que el adenovirus oncolitico Adhz60 indujo en Ia lfnea TC-1 un efecto citopatico evidente, disminuye importantemente su viabilidad celular, indujo la expresión de proteínas virales tempranas y tardas, fue capaz de generar una progenie viral infectiva y de inducir apoptosis. Contribuciones y Conclusiones. Con nuestros resultados demostramos que la linea celular murina TC-1 es permisiva para la replican del virus oncolftico Adhz60. Por lo tanto, las células TC-1 pueden ser usadas como modelo tumoral para evaluar adenovirus oncoliticos en ratones inmunocompetentes.
