Computer vision is a field that uses techniques to acquire, process, analyze and understand images from the real world in order to produce numeric or symbolic information in the form of decisions [1]. This project aims to use computer vision to prepare an app to analyze a Madeira Wine and characterize it (identify its variety) by its color. Dry or sweet wines, young or old wines have a specific color. It uses techniques to compare histograms in order to analyze the images taken from a test sample inside a special container designed for this purpose. The color analysis from a wine sample using an image captured by a smartphone can be difficult. Many factors affect the captured image such as, light conditions, the background of the sample container due to the many positions the photo can be taken (different to capture facing a white wall or facing the floor for example). Using new technologies such as 3D printing it was possible to create a prototype that aims to control the effect of those external factors on the captured image. The results for this experiment are good indicators for future works. Although it’s necessary to do more tests, the first tests had a success rate of 80% to 90% of correct results. This report documents the development of this project and all the techniques and steps required to execute the tests.

The stretch zone width (SZW) data for 15-5PH steel CTOD specimens fractured at -150 degrees C to + 23 degrees C temperature were measured based on focused images and 3D maps obtained by extended depth-of-field reconstruction from light microscopy (LM) image stacks. This LM-based method, with a larger lateral resolution, seems to be as effective for quantitative analysis of SZW as scanning electron microscopy (SEM) or confocal scanning laser microscopy (CSLM), permitting to clearly identify stretch zone boundaries. Despite the worst sharpness of focused images, a robust linear correlation was established to fracture toughness (KC) and SZW data for the 15-5PH steel tested specimens, measured at their center region. The method is an alternative to evaluate the boundaries of stretched zones, at a lower cost of implementation and training, since topographic data from elevation maps can be associated with reconstructed image, which summarizes the original contrast and brightness information. Finally, the extended depth-of-field method is presented here as a valuable tool for failure analysis, as a cheaper alternative to investigate rough surfaces or fracture, compared to scanning electron or confocal light microscopes. The aim of this work is to evaluate the influence of point measurements in images, with subpixel accuracy, and its contribution in the calibration of digital cameras. Also, the effect of subpixel measurements in 3D coordinates of check points in the object space will be evaluated. With this purpose, an algorithm that allows subpixel accuracy was implemented for semi-automatic determination of points of interest, based on Fõrstner operator. Experiments were accomplished with a block of images acquired with the multispectral camera DuncanTech MS3100-CIR. The influence of subpixel measurements in the adjustment by Least Square Method (LSM) was evaluated by the comparison of estimated standard deviation of parameters in both situations, with manual measurement (pixel accuracy) and with subpixel estimation. Additionally, the influence of subpixel measurements in the 3D reconstruction was also analyzed. Based on the obtained results, i.e., on the quantification of the standard deviation reduction in the Inner Orientation Parameters (IOP) and also in the relative error of the 3D reconstruction, it was shown that measurements with subpixel accuracy are relevant for some tasks in Photogrammetry, mainly for those in which the metric quality is of great relevance, as Camera Calibration.

O trabalho aqui apresentado visa o estudo da migração 3D no tempo, em amplitudes verdadeiras, de dados da seção afastamento nulo ("zero-offset"), usando a aproximação de segunda ordem do tempo de trânsito de difração. O processo de migração no tempo usa a função difração determinada, corretamente, através de mesmos parâmetros que controlam a função tempo de trânsito de reflexão, além do auxílio dos raios paraxiais. Os parâmetros que controlam a função tempo de trânsito de reflexão são determinados através de pelo menos nove medidas de tempo de trânsito, obtidas a partir de dados pré-empilhados (tiro comum, ponto médio comum - CMP, etc.). Determinada a função difração, pode-se fazer a migração sem a necessidade de que se conheça o modelo de velocidades, pois a função tempo de trânsito de difração, como definido neste trabalho, não necessita do mesmo. O espalhamento geométrico é removido através do processo de migração e aplicação de uma função peso aos dados da seção afastamento nulo. Juntamente com a função peso, aplicou-se um filtro objetivando a recuperação da fase do sinal, ocorrida como consequência do processo de migração. Devido a limitação computacional, fez-se testes com dados sintéticos, em 2D, objetivando verificar a eficiência do programa. Os resultados obtidos foram satisfatórios, mostrando a eficiência e confiabilidade do processo.

The use of computer-assisted technologies such as CAD - Computed Aided Design, CAM - Computed Aided Manufacturing, CAE - Computed Aided Engineering and CNC - Computed Numerical Control, are priorities in engineering and product designers. However, the dimensional measurement between the virtual and the real product design requires research, and dissemination procedures among its users. This work aims to use these technologies, through analysis and measurement of a CNC milling machine, designed and assembled in the university. Through the use of 3D scanning, and analyzing images of the machined samples, and its original virtual files, it was possible to compare the sizes of these samples in counterposition to the original virtual dimensions, we can state that the distortions between the real and virtual, are within acceptable limits for this type of equipment. As a secondary objective, this work seeks to disseminate and make more accessible the use of these technologies.

Objective: To compare the agreement of multiplanar and rendering modes in the assessment fetal atrioventricular valves (mitral and tricuspid) areas by three-dimensional (3D) ultrasonography using the software spatio-temporal image correlation (STIC). Methods: We conducted a prospective cross-sectional study with normal pregnant women, with single fetuses, between 18-33 weeks. To measure the areas, we used the plan of four-chamber view. In the case of multiplanar, the plane was rotated on the axis "Z" form the heart to position at 9h. For rendering, the green line (region of interest - ROI) was placed from the atria of the heart perpendicular to the crux. The agreement was assessed by a Bland-Altman (limits of agreement) using the relative difference between the measures: ((rendering mode) - (multiplanar mode)) / (average). Results: 328 fetuses were evaluated. We have not identified the occurrence of systematic error between methods: the average relative difference was 1.62% (-2.07% to 5.32%, confidence interval 95%) in the mitral and 1.77% (- 1.08% to 4.62%) in the tricuspid valve. The limits of agreement between methods were -65.26% to 68.51% for the mitral and -49.91% to 53.45% for the tricuspid. Conclusions: There was no systematic error between modes and thus the observed values for the area of fetal atrioventricular valves can be used for comparisons needs to be corrected. However, relatively large variations may be observed when repeating the measurement area by different modes.

Arthropodenhämocyanine und Molluskenhämocyanine, die extrazellulären Atmungsproteine der Arthropoden und Mollusken, unterscheiden sich grundsätzlich im Aufbau, besitzen aber ähnliche aktive Zentren, welche in ihrer oxydierten Form für die Blaufärbung der Hämocyanine verantwortlich sind. Sauerstoff wird im Bindungszentrum zwischen zwei, von sechs Histidinen ligandierten, Kupfer(I)Ionen gebunden. Arthropodenhämocyanine bauen sich artspezifisch aus 1, 2, 4, 6, oder 8 Hexameren mit D3-Symmetrie auf. Die Untereinheiten von je ca. 75 kDa falten sich in drei Domänen unterschiedlicher Funktionen. Der komplexe, hierarchische Zusammenbau der Arthropodenhämocyanine hängt von der Heterogenität der Untereinheiten ab. Die 7 verschieden Sequenzen des 4x6-Hämocyanins von Eurypelma californicum (EcHc) sind biochemisch in der Quartärstruktur lokalisiert. Bislang fehlte noch ein unabhängig erstelltes 3D-Modell der geometrischen Gesamtstruktur welche die hexamere und monomere Topographie eindeutig zeigt. Dessen Erstellung war Gegenstand dieser Arbeit, in Verbindung mit der Zielsetzung, die 3D-Rekonstruktion in den beiden extremen physiologischen Zuständen, mit und ohne gebundenen Sauerstoff, zu erzeugen. Dazu wurden in einer eigens entwickelten Atmosphären-Präparationskammer die Proteine in Lösung schockgefrorenen und mittels Cryo-3D-Elektronenmikroskopie gemessen. Aus den daraus gewonnen Projektionsbildern ließen sich mit der ”Single Particle Analyse“ die 3D-Informationen zurückberechnen. Die 3D-Rekonstruktionen wurden mit der publizierten Röntgenkristallstruktur des hexameren Referenz-Hämocyanins der Languste Panulirus interruptus verifiziert. Die Rekonstruktionen erlaubten die eindeutige Messung diverser in der Literatur diskutierter Parameter der Architektur des 4x6-EcHc und darüber hinaus weiterer geometrischer Parameter, welche hier erstmals veröffentlicht werden. SAXS-Daten sagen extreme Translationen und Rotationen von Teilquartärstrukturen zwischen oxy- und deoxy-EcHc voraus, was von den 3D-Rekonstruktionen der beiden Zustände nicht bestätigt werden konnte: Die 16 Å Rekonstruktion der Deoxyform weicht geometrisch nicht von der 21 Å Rekonstruktion der Oxyform ab. Die Einpassung der publizierten Röntgenstruktur der Untereinheit II des Hämocyanin des Pfeilschwanzkrebses Limulus polyphemus in die Rekonstruktionen unterstützt eine auf der hexameren Hierarchieebene lokalisierte Dynamik der Oxygenierung. Mittels Einpassung modellierter molekularer Strukturen der EcHc-Sequenzen konnte eine erste Vermutung zur Lokalisation der beiden zentralen Linker-Untereinheiten b und c des 4x6-Moleküls gemacht werden: Demnach würde Untereinheit b in den exponierten Hexameren des Moleküls liegen. Aussagen über die Quartärstrukturbindungen auf molekularer Ebene aufgrund der Einpassung modellierter molekularer Daten in die Rekonstruktionen sind als spekulativ einzustufen: a) Die Auflösung der Rekonstruktion ist verbesserungswürdig. b) Es gibt keine adäquate Vorlage für eine verlässliche Strukturvorhersage; die verschiedenen EcHc-Sequenzen liegen nur als Modellierung vor. c) Es wäre eine flexible Einpassung notwendig, um Ungenauigkeiten in den modellierten Strukturen durch Sekundärstrukturanpassung zu minimieren.

Sowohl in der Natur als auch in der Industrie existieren thermisch induzierte Strömungen. Von Interesse für diese Forschungsarbeit sind dabei die Konvektionen im Erdmantel sowie in den Glasschmelzwannen. Der dort stattfindende Materialtransport resultiert aus Unterschieden in der Dichte, der Temperatur und der chemischen Konzentration innerhalb des konvektierenden Materials. Um das Verständnis für die ablaufenden Prozesse zu verbessern, werden von zahlreichen Forschergruppen numerische Modellierungen durchgeführt. Die Verifikation der dafür verwendeten Algorithmen erfolgt meist über die Analyse von Laborexperimenten. Im Vordergrund dieser Forschungsarbeit steht die Entwicklung einer Methode zur Bestimmung der dreidimensionalen Temperaturverteilung für die Untersuchung von thermisch induzierten Strömungen in einem Versuchsbecken. Eine direkte Temperaturmessung im Inneren des Versuchsmaterials bzw. der Glasschmelze beeinflusst allerdings das Strömungsverhalten. Deshalb wird die geodynamisch störungsfrei arbeitende Impedanztomographie verwendet. Die Grundlage dieser Methode bildet der erweiterte Arrhenius-Zusammenhang zwischen Temperatur und spezifischer elektrischer Leitfähigkeit. Während der Laborexperimente wird ein zähflüssiges Polyethylenglykol-Wasser-Gemisch in einem Becken von unten her erhitzt. Die auf diese Weise generierten Strömungen stellen unter Berücksichtigung der Skalierung ein Analogon sowohl zu dem Erdmantel als auch zu den Schmelzwannen dar. Über mehrere Elektroden, die an den Beckenwänden installiert sind, erfolgen die geoelektrischen Messungen. Nach der sich anschließenden dreidimensionalen Inversion der elektrischen Widerstände liegt das Modell mit der Verteilung der spezifischen elektrischen Leitfähigkeit im Inneren des Versuchsbeckens vor. Diese wird mittels der erweiterten Arrhenius-Formel in eine Temperaturverteilung umgerechnet. Zum Nachweis der Eignung dieser Methode für die nichtinvasive Bestimmung der dreidimensionalen Temperaturverteilung wurden mittels mehrerer Thermoelemente an den Beckenwänden zusätzlich direkte Temperaturmessungen durchgeführt und die Werte miteinander verglichen. Im Wesentlichen sind die Innentemperaturen gut rekonstruierbar, wobei die erreichte Messgenauigkeit von der räumlichen und zeitlichen Auflösung der Gleichstromgeoelektrik abhängt.

Procedures for quantitative walking analysis include the assessment of body segment movements within defined gait cycles. Recently, methods to track human body motion using inertial measurement units have been suggested. It is not known if these techniques can be readily transferred to clinical measurement situations. This work investigates the aspects necessary for one inertial measurement unit mounted on the lower back to track orientation, and determine spatio-temporal features of gait outside the confines of a conventional gait laboratory. Apparent limitations of different inertial sensors can be overcome by fusing data using methods such as a Kalman filter. The benefits of optimizing such a filter for the type of motion are unknown. 3D accelerations and 3D angular velocities were collected for 18 healthy subjects while treadmill walking. Optimization of Kalman filter parameters improved pitch and roll angle estimates when compared to angles derived using stereophotogrammetry. A Weighted Fourier Linear Combiner method for estimating 3D orientation angles by constructing an analytical representation of angular velocities and allowing drift free integration is also presented. When tested this method provided accurate estimates of 3D orientation when compared to stereophotogrammetry. Methods to determine spatio-temporal features from lower trunk accelerations generally require knowledge of sensor alignment. A method was developed to estimate the instants of initial and final ground contact from accelerations measured by a waist mounted inertial device without rigorous alignment. A continuous wavelet transform method was used to filter and differentiate the signal and derive estimates of initial and final contact times. The technique was tested with data recorded for both healthy and pathologic (hemiplegia and Parkinson’s disease) subjects and validated using an instrumented mat. The results show that a single inertial measurement unit can assist whole body gait assessment however further investigation is required to understand altered gait timing in some pathological subjects.

For crime scene investigation in cases of homicide, the pattern of bloodstains at the incident site is of critical importance. The morphology of the bloodstain pattern serves to determine the approximate blood source locations, the minimum number of blows and the positioning of the victim. In the present work, the benefits of the three-dimensional bloodstain pattern analysis, including the ballistic approximation of the trajectories of the blood drops, will be demonstrated using two illustrative cases. The crime scenes were documented in 3D, using the non-contact methods digital photogrammetry, tachymetry and laser scanning. Accurate, true-to-scale 3D models of the crime scenes, including the bloodstain pattern and the traces, were created. For the determination of the areas of origin of the bloodstain pattern, the trajectories of up to 200 well-defined bloodstains were analysed in CAD and photogrammetry software. The ballistic determination of the trajectories was performed using ballistics software. The advantages of this method are the short preparation time on site, the non-contact measurement of the bloodstains and the high accuracy of the bloodstain analysis. It should be expected that this method delivers accurate results regarding the number and position of the areas of origin of bloodstains, in particular the vertical component is determined more precisely than using conventional methods. In both cases relevant forensic conclusions regarding the course of events were enabled by the ballistic bloodstain pattern analysis.

The spine is a complex structure that provides motion in three directions: flexion and extension, lateral bending and axial rotation. So far, the investigation of the mechanical and kinematic behavior of the basic unit of the spine, a motion segment, is predominantly a domain of in vitro experiments on spinal loading simulators. Most existing approaches to measure spinal stiffness intraoperatively in an in vivo environment use a distractor. However, these concepts usually assume a planar loading and motion. The objective of our study was to develop and validate an apparatus, that allows to perform intraoperative in vivo measurements to determine both the applied force and the resulting motion in three dimensional space. The proposed setup combines force measurement with an instrumented distractor and motion tracking with an optoelectronic system. As the orientation of the applied force and the three dimensional motion is known, not only force-displacement, but also moment-angle relations could be determined. The validation was performed using three cadaveric lumbar ovine spines. The lateral bending stiffness of two motion segments per specimen was determined with the proposed concept and compared with the stiffness acquired on a spinal loading simulator which was considered to be gold standard. The mean values of the stiffness computed with the proposed concept were within a range of ±15% compared to data obtained with the spinal loading simulator under applied loads of less than 5 Nm.

The measurement of fluid volumes in cases of pericardial effusion is a necessary procedure during autopsy. With the increased use of virtual autopsy methods in forensics, the need for a quick volume measurement method on computed tomography (CT) data arises, especially since methods such as CT angiography can potentially alter the fluid content in the pericardium. We retrospectively selected 15 cases with hemopericardium, which underwent post-mortem imaging and autopsy. Based on CT data, the pericardial blood volume was estimated using segmentation techniques and downsampling of CT datasets. Additionally, a variety of measures (distances, areas and 3D approximations of the effusion) were examined to find a quick and easy way of estimating the effusion volume. Segmentation of CT images as shown in the present study is a feasible method to measure the pericardial fluid amount accurately. Downsampling of a dataset significantly increases the speed of segmentation without losing too much accuracy. Some of the other methods examined might be used to quickly estimate the severity of the effusion volumes.

BACKGROUND: The aim of this study was to determine the performance of a new, 3D-monitor based, objective stereotest in children under the age of four. METHODS: Random-dot circles (diameter 10 cm, crossed, disparity of 0.34 degrees) randomly changing their position were presented on an 3D-monitor while eye movements were monitored by infrared photo-oculography. If > or = 3 consecutive stimuli were seen, a positive response was assumed. One hundred thirty-four normal children aged 2 months to 4 years (average 17+/-15.3 months) were examined. RESULTS: Below the age of 12 months, we were not able to obtain a response to the 3D stimulus. For older children the following rates of positive responses were found: 12-18 months 25%, 18-24 months 10%, 24-30 months 16%, 30-36 months 57%, 36-42 months 100%, and 42-48 months 91%. Multiple linear logistic regression showed a significant influence on stimulus recognition of the explanatory variables age (p<0.00001) and child cooperation (p<0.001), but not of gender (p>0.1). CONCLUSIONS: This 3D-monitor based stereotest allows an objective measurement of random-dot stereopsis in younger children. It might open new ways to screen children for visual abnormalities and to study the development of stereovision. However, the current experimental setting does not allow determining random-dot stereopsis in children younger than 12 months.
