PS I, PS II and light-harvesting complexes (LHC) in oxygen evolving photosynthetic organisms were reviewed. These organisms include cyanobacteria, red algae, brown algae, diatoms, chrysophytes, dinophytes, xanthophytes, crypophytes, green algae and green plants. The diversity of pigment-protein complexes that fuel the conversion of radiant energy to chemical bond energy was highlighted, and the evolutionary relationships among the LHC structural polypeptides and the characteristics of the fluorescence emission of PS I at 77 K was discussed.

Gracilaria lemaneiformis Bory is an economically important alga that is primarily used for agar production. Although tetraspores are ideal seeds for the cultivation of G. lemaneiformis, the most popular culture method is currently based on vegetative fragments, which is labor-intensive and time-consuming. In this study, we optimized the conditions for tetraspore release and evaluated the photosynthetic activities of different colonies formed from the branches of G. lemaneiformis using a PAM (pulse-amplitude-modulated) measuring system. The results showed that variations in temperature and salinityhad significant effects on tetraspore yield. However, variations in the photon flux density (from 15 mu mol m(-2) s(-1) to 480 mu mol m(-2) s(-1)) had no apparent effect on tetraspore yield. Moreover, the PAM-parameters Y(I), Y(II), ETR(I), ETR(II) and F (v)/F (m) of colonies formed from different branches showed the same trend: parameter values of first generation branches > second generation branches > third generation branches. These results suggest that the photosynthetic activities of different colonies of branches changed with the same trend. Furthermore, photosynthesis in G. lemaneiformis was found to be involved in vegetative reproduction and tetraspore formation. Finally, the first generation branches grew slowly, but accumulated organic compounds to form large numbers of tetraspores. Taken together, these results showed that the first generation branches are ideal materials for the release of tetraspores.

The ovary of triploid shrimp Fenneropenaeus chinensis was apparently impaired compared to that of the diploid shrimp at the same age. Therefore triploid shrimp ovary is possible to be taken as a model to understand the mechanism of ovary development of shrimp compared to that of the ovary of diploid shrimp at the same age. In the present study, a suppression subtractive hybridization (SSH) technique was applied to identify differentially expressed genes in the ovary between diploid and triploid shrimp. For the forward library (RNA from the ovary of triploid shrimp as the tester), 54 genes were identified. For the reverse library (RNA from the ovary of diploid shrimp as the tester), 16 genes were identified. The identified genes encoded proteins with multiple functions, including extracellular matrix components, cytoskeleton, cell growth and death, metabolism, genetic information processing, signal transduction/transport or immunity related proteins. Eleven differentially expressed genes were selected to be confirmed in the ovaries of triploid and diploid shrimp by semi-quantitative RT-PCR. Genes encoding spermatogonial stem-cell renewal factor, cytochrome c oxidase subunits I and II, clottable protein, antimicrobial peptide and transposase showed up-regulated expressions in the ovary of triploid shrimp. Genes encoding tubulin, cellular apoptosis susceptibility protein, farnesoic acid O-methyltransferase, thrombospondin and heat shock protein 90 genes showed higher expressions in the ovary of diploid shrimp. The differential expressions of the above genes are suggested to be related to the ovary development of shrimp. It will provide a new clue to uncover the molecular mechanisms underlying the ovarian development in penaeid shrimp. (C) 2010 Elsevier Inc. All rights reserved.

The exploration and study in recent years shows that the upper Paleozoic in the east of Ordos Basin possesses major exploration potential, so it is necessary to have a comprehensive and synthetic research in this area. Following the guideline of T.A. Cross’s high resolution sequences stratigraphy and combined with sedimentology, the strata and sequence in the research area are divided and correlated. This paper emphasizes on the reservoir in this area and its major fruits are: Firstly, form the framework of the high sequence stratigraphy through mainly studying on the data of core, well drilling and field section combining. Then, think that the best reservoir formation mainly appears in the middle or lower part of long arising semi-cycle, and focus on A type and C1 middle sequence cycle that contributes to the development of formation. Next, think the sedimentary source mainly comes from the epimetamorphic crystalline basement in the north of the Ordoes Basin through analyzing depositional background, researching on sandstone petrography, distribution characteristics of the sedimentary system as well as researching on heavy mineral combination characteristics. Fourthly, give priority to Zizhou-Qingjian area for the first time and gain seven lithofacies paleography maps in the No.2 member of Shanxi and the No.8 member of Shihezi through using the method of isochronal lithofacies paleogeography mapping, then bring forth that Sh2 is mainly developed a type I sequence under the margin of ramp lake-basin background. Fifthly, through researching on the characteristics of reservoir petrography and diagenesis, think that all researched areas experienced compaction and cementation, and there is different types of rocks, only little of the primary pores that are made up of litharenite and lithoclastic quartz sandstone is left, while in lithoclastic quartz sandstone, there are still many primary pores Sixthly, through studying on pore types, microstructure, as well as physical property on the key formation, think that the researched area mostly appears typical low pore, low porosity and permeability, which mainly result from sedimentary and diagenesis. Lastly, through researching on classified evaluation in the key formation, productivity analysis, and combing with reservoir distribution, the researched area is divided into three parts, and think that exploration should be emphasized on formation I and II. The characteristics of sandstones distribution in Sh23 member, gas formation distribution and open flow capacity of exploitation well are all consistent properly with the results of reservoir comprehensive evaluation in this thesis proved by the gas production development in 2006.

The Gangxi oil field has reached a stage of high water production. The reservoir parameters, such as reservoir physical characteristics, pore structure, fluid, have obviously changed. This thesis therefore carries out a study of these parameters that control reservoir characteristics, physical and chemical actions that have taken place within the reservoirs due to fluid injection, subsequent variations of reservoir macroscopic physical features, microscopic pore structures, seepages, and formation fluid properties. This study rebuilds a geologic model for this oil field, establishes a log-interpreting model, proposes a methodology for dealing with large pore channels and remnant oil distribution, and offers a basis for effective excavation of potential oil, recovery planning, and improvement of water-injection techniques. To resolve some concurrent key problems in the process of exploration of the Gangxi area, this thesis carries out a multidisciplinary research into reservoir geology, physical geography, reservoir engineering, and oil-water well testing. Taking sandstone and flow unit as objects, this study establishes a fine geologic model by a quantificational or semi-quantificational approach in order to understand the remnant oil distribution and the reservoir potential, and accordingly proposes a plan for further exploration. By rebuilding a geological model and applying reservoir-engineering methods, such as numerical simulation, this thesis studies the oil-water movement patterns and remnant-oil distribution, and further advances a deployment plan for the necessary adjustments and increase of recoverable reserves. Main achievements of this study are as follows: 1. The Minghazhen Formation in the Gangxi area is featured by medium-sinuosity river deposits, manifesting themselves as a transitional type between typical meandering and braided rivers. The main microfacies are products of main and branch channels, levee, inter-channel overflows and crevasse-splay floodplains. The Guantao Group is dominantly braided river deposit, and microfacies are mainly formed in channel bar, braided channel and overbank. Main lithofacies include conglomerate, sandstone, siltstone and shale, with sandstone facies being the principal type of the reservoir. 2. The reservoir flow unit of the Gangxi area can be divided into three types: Type I is a high-quality heterogeneous seepage unit, mainly distributed in main channel; Type II is a moderate-quality semi-heterogeneous seepage unit, mainly distributed in both main and branch channels, and partly seen within inter-channel overflow microfacies; Type III is a low-quality, relatively strong heterogeneous seepage unit, mainly distributed in inter-channel overflow microfacies and channel flanks. 3. Flow units and sedimentary microfacies have exerted relatively strong controls on the flowing of underground oil-water: (1) injection-production is often effective in the float units of Type I and II, whilst in the same group of injection-production wells, impellent velocity depends on flow unit types and injection-production spacing; (2) The injection-production of Type III flow unit between the injection-production wells of Type I and II flow units, however, are little effective; (3) there can form a seepage shield in composite channels between channels, leading to inefficient injection and production. 4. Mainly types of large-scale remnant-oil distribution are as follows: (1) remnant oil reservoir of Type III flow unit; (2) injection-production well group of remnant oil area of Type III flow unit; (3) remnant oil reservoirs that cannot be controlled by well network, including reservoir featured by injection without production, reservoir characterized by production without injection, and oil reservoir at which no well can arrive; (4) remnant oil area where injection-production system is not complete. 5. Utilizing different methods to deal with different sedimentary types, sub-dividing the columns of up to 900 wells into 76 chronostratigraphic units. Four transitional sandstone types are recognized, and contrast modes of different sandstone facies are summarized Analyzing in details the reservoirs of different quality by deciphering densely spaced well patterns, dividing microscopic facies and flow units, analyzing remnant oil distribution and its effect on injection-production pattern, and the heterogeneity. Theory foundation is therefore provided for further excavation of remnant oil. Re-evaluating well-log data. The understanding of water-flood layers and conductive formations in the Gangxi area have been considerably improved, and the original interpretations of 233 wells have changed by means of double checking. Variations of the reservoirs and the fluid and formation pressures after water injection are analyzed and summarized Studies are carried out of close elements of the reservoirs, fine reservoir types, oil-water distribution patterns, as well as factors controlling oil-gas enrichment. A static geological model and a prediction model of important tracts are established. Remaining recoverable reserves are calculated of all the oil wells and oil-sandstones. It is proposed that injection-production patterns of 348 oil-sandstones should be adjusted according to the analysis of adaptability of all kinds of sandstones in the injection-production wells.

大陆溢流玄武岩根据TiO2含量可划分为高钛玄武岩和低钛玄武岩,这两种不同类型的玄武岩往往有不同的空间分布特征,也具有不同的成因指示意义。峨眉山玄武岩也有高、低钛之分,且其空间分布存在明显差异。然而,在以往的峨眉山玄武岩的研究中只对高、低钛玄武岩进行对比研究,而单独对同一类型玄武岩的研究甚少,尤其是对物质来源的研究。在本论文中,作者以峨眉山大火成岩省中部二滩地区高钛玄武岩为研究对象,通过主量元素、微量元素和Sr-Nd-Pb同位素对其地球化学、岩浆过程以及物质来源进行了探讨,得出以下初步认识和结论: ⑴ 二滩高钛玄武岩可划分出Group I和Group II两种岩石类型。Group I中Sr和Zr显示明显负异常,而Group II无Sr和Zr异常。此两种类型岩石的Sr-Nd-Pb同位素地球化学特征也较为不同。 ⑵ 二滩高钛玄武岩为板内构造环境,Group I和Group II显示出碱性或钙碱性岩特征。 ⑶ Group I和Group II具有不同的熔体形成熔融程度,Group I相对低(0~3.5%),Group II相对高(4.0~8.0%)。不同的部分熔融程度导致了控制矿物相的不同。 ⑷ Group I和Group II岩浆分别经历了不同的结晶分异过程。Group I主要显示出斜长石结晶分异趋势,Group II则主要显示出橄榄石结晶分异趋势。 ⑸ 微量不相容元素和Sr-Nd-Pb同位素特征表明,Group I和Group II均源自深部富集地幔,具有不同的物质来源,且与地幔柱有成因上的联系。 以上特征表明,二滩玄武岩与深源地幔柱有成因上的联系。这可能是深部地幔柱(来自核-幔边界或下地幔)上升过程中在不同深度携带了不同物质,这些物质发生部分熔融,从而导致了Group I和Group II的明显差异。其充分说明,二滩高钛玄武岩的物源是不均一的,反映了源区的不均一性特征。

O sistema de avaliação de impacto social da inovação tecnológica agropecuária (Ambitec- Social) consiste de um conjunto de planilhas eletrônicas que integram quatorze indicadores da contribuição de uma dada inovação tecnológica agropecuária para o bem estar social, no âmbito de um estabelecimento rural. Quatro aspectos essenciais de avaliação são considerados: i. Emprego, ii. Renda, iii. Saúde, e iv. Gestão e Administração. Os indicadores são construídos em matrizes de ponderação nas quais dados obtidos em campo, de acordo com o conhecimento do produtor/administrador do estabelecimento, são automaticamente transformados em índices de impacto expressos graficamente. Os resultados da avaliação permitem, ao produtor/administrador, averiguar quais impactos da tecnologia podem estar desconformes com seus objetivos de bem estar social; ao tomador de decisões a indicação de medidas de fomento ou controle da adoção da tecnologia, segundo planos de desenvolvimento local sustentável e finalmente, proporcionam uma unidade de medida objetiva de impacto, auxiliando na qualificação, seleção e transferência de tecnologias agropecuárias.

BACKGROUND:Blood lipid levels including low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL-C), and triglycerides (TG) are highly heritable. Genome-wide association is a promising approach to map genetic loci related to these heritable phenotypes.METHODS:In 1087 Framingham Heart Study Offspring cohort participants (mean age 47 years, 52% women), we conducted genome-wide analyses (Affymetrix 100K GeneChip) for fasting blood lipid traits. Total cholesterol, HDL-C, and TG were measured by standard enzymatic methods and LDL-C was calculated using the Friedewald formula. The long-term averages of up to seven measurements of LDL-C, HDL-C, and TG over a ~30 year span were the primary phenotypes. We used generalized estimating equations (GEE), family-based association tests (FBAT) and variance components linkage to investigate the relationships between SNPs (on autosomes, with minor allele frequency [greater than or equal to]10%, genotypic call rate [greater than or equal to]80%, and Hardy-Weinberg equilibrium p [greater than or equal to] 0.001) and multivariable-adjusted residuals. We pursued a three-stage replication strategy of the GEE association results with 287 SNPs (P < 0.001 in Stage I) tested in Stage II (n ~1450 individuals) and 40 SNPs (P < 0.001 in joint analysis of Stages I and II) tested in Stage III (n~6650 individuals).RESULTS:Long-term averages of LDL-C, HDL-C, and TG were highly heritable (h2 = 0.66, 0.69, 0.58, respectively; each P < 0.0001). Of 70,987 tests for each of the phenotypes, two SNPs had p < 10-5 in GEE results for LDL-C, four for HDL-C, and one for TG. For each multivariable-adjusted phenotype, the number of SNPs with association p < 10-4 ranged from 13 to 18 and with p < 10-3, from 94 to 149. Some results confirmed previously reported associations with candidate genes including variation in the lipoprotein lipase gene (LPL) and HDL-C and TG (rs7007797; P = 0.0005 for HDL-C and 0.002 for TG). The full set of GEE, FBAT and linkage results are posted at the database of Genotype and Phenotype (dbGaP). After three stages of replication, there was no convincing statistical evidence for association (i.e., combined P < 10-5 across all three stages) between any of the tested SNPs and lipid phenotypes.CONCLUSION:Using a 100K genome-wide scan, we have generated a set of putative associations for common sequence variants and lipid phenotypes. Validation of selected hypotheses in additional samples did not identify any new loci underlying variability in blood lipids. Lack of replication may be due to inadequate statistical power to detect modest quantitative trait locus effects (i.e., < 1% of trait variance explained) or reduced genomic coverage of the 100K array. GWAS in FHS using a denser genome-wide genotyping platform and a better-powered replication strategy may identify novel loci underlying blood lipids.

This dissertation illustrates the merits of an interdisciplinary approach to religious conversion by employing Lewis Rambo’s systemic stage model to illumine the process of St. Augustine’s conversion. Previous studies of Augustine’s conversion have commonly explored his narrative of transformation from the perspective of one specific discipline, such as theology, history, or psychology. In doing so, they have necessarily restricted attention to a limited set of questions and problems. By bringing these disciplines into a structured, critical conversation, this study demonstrates how formulating and responding to the interplay among personal, social, cultural, and religious dimensions of Augustine’s conversion process may eventuate in the consideration of issues previously unarticulated and thus unaddressed. Rambo (1993) formulates a model of religious change that consists of what he calls context, crisis, quest, encounter, interaction, commitment, and consequences. Change is explained by drawing upon the research and scholarship of psychologists, sociologists, anthropologists, and religionists, in conjunction with the contributions of theologians. This study unfolds in the following chapters: I. Introduction; II. Literature review of scholarship about conversion, with emphasis on explication of Rambo’s model; III. A description of the case of Augustine, drawn from a close reading of the Confessions; IV. Literature review of scholarship about Augustine’s conversion; V. Interdisciplinary interpretation of Augustine’s conversion; and VI. Implications for scholars of conversion, and for pastoral caregivers, as well as recommendations for future research. This dissertation demonstrates how Augustine’s conversion experience was deeply influenced by 1) psychological distress and crisis; 2) the quest to know himself and the divine; 3) interactions with significant others; 4) participation in Christian communities; 5) philosophical and cultural changes; and 6) the encounter with the divine. As such, this study reveals the value of interpreting Augustine’s conversion as an evolving process constituted in multiple factors that can be differentiated from one another, yet clearly interact with one another. It examines the implications of constructing an interdisciplinary approach to Augustine’s conversion narrative for both the academy and the Christian community, and recommends the use of Rambo’s model in studies of other cases of religious change.

Recent evidence suggests that in addition to their well known stimulatory properties, dendritic cells (DCs) may play a major role in peripheral tolerance. It is still unclear whether a distinct subtype or activation status of DC exists that promotes the differentiation of suppressor rather than effector T cells from naive precursors. In this work, we tested whether the naturally occurring CD4+ CD25+ regulatory T cells (Treg) may control immune responses induced by DCs in vivo. We characterized the immune response induced by adoptive transfer of antigen-pulsed mature DCs into mice depleted or not of CD25+ cells. We found that the development of major histocompatibility complex class I and II-restricted interferon gamma-producing cells was consistently enhanced in the absence of Treg. By contrast, T helper cell (Th)2 priming was down-regulated in the same conditions. This regulation was independent of interleukin 10 production by DCs. Of note, splenic DCs incubated in vitro with Toll-like receptor ligands (lipopolysaccharide or CpG) activated immune responses that remained sensitive to Treg function. Our data further show that mature DCs induced higher cytotoxic activity in CD25-depleted recipients as compared with untreated hosts. We conclude that Treg naturally exert a negative feedback mechanism on Th1-type responses induced by mature DCs in vivo.

BACKGROUND: Incorporation of multiple enrichment biomarkers into prospective clinical trials is an active area of investigation, but the factors that determine clinical trial enrollment following a molecular prescreening program have not been assessed. PATIENTS AND METHODS: Patients with 5-fluorouracil-refractory metastatic colorectal cancer at the MD Anderson Cancer Center were offered screening in the Assessment of Targeted Therapies Against Colorectal Cancer (ATTACC) program to identify eligibility for companion phase I or II clinical trials with a therapy targeted to an aberration detected in the patient, based on testing by immunohistochemistry, targeted gene sequencing panels, and CpG island methylation phenotype assays. RESULTS: Between August 2010 and December 2013, 484 patients were enrolled, 458 (95%) had a biomarker result, and 157 (32%) were enrolled on a clinical trial (92 on biomarker-selected and 65 on nonbiomarker selected). Of the 458 patients with a biomarker result, enrollment on biomarker-selected clinical trials was ninefold higher for predefined ATTACC-companion clinical trials as opposed to nonpredefined biomarker-selected clinical trials, 17.9% versus 2%, P < 0.001. Factors that correlated positively with trial enrollment in multivariate analysis were higher performance status, older age, lack of standard of care therapy, established patient at MD Anderson, and the presence of an eligible biomarker for an ATTACC-companion study. Early molecular screening did result in a higher rate of patients with remaining standard of care therapy enrolling on ATTACC-companion clinical trials, 45.1%, in contrast to nonpredefined clinical trials, 22.7%; odds ratio 3.1, P = 0.002. CONCLUSIONS: Though early molecular prescreening for predefined clinical trials resulted in an increase rate of trial enrollment of nonrefractory patients, the majority of patients enrolled on clinical trials were refractory to standard of care therapy. Within molecular prescreening programs, tailoring screening for preidentified and open clinical trials, temporally linking screening to treatment and optimizing both patient and physician engagement are efforts likely to improve enrollment on biomarker-selected clinical trials. Because only 10% of individuals infected with Mycobacterium tuberculosis will eventually develop disease, antigens that are recognized differently by the immune systems of infected healthy and diseased subjects may constitute potential vaccine candidates. Here, the heparin-binding hemagglutinin adhesin (HBHA) is identified as such an antigen. Lymphocytes from 60% of healthy infected individuals (n=25) produced interferon (IFN)-gamma after stimulation with HBHA, compared with only 4% of patients with active tuberculosis (n=24). In the responders, both CD4(+) and CD8(+) cells secreted HBHA-specific IFN-gamma, and the antigen was presented by both major histocompatibility complex class I and II molecules. In contrast to the reduced ability of patients with tuberculosis to produce HBHA-specific IFN-gamma, most of them (82%) produced anti-HBHA antibodies, compared with 36% of the infected healthy subjects. These observations indicate that HBHA is recognized differently by the immune systems of patients with tuberculosis and infected healthy individuals and might provide a marker for protection against tuberculosis.

La inundación es un factor importante que afecta la implantación y producción de especies forrajeras en ecosistemas de pastizales anegables. Chloris gayana K. y Panicum coloratum L. son dos gramíneas tropicales cuyo uso como forrajera en ambientes templados se ha incrementado recientemente. En esta tesis se realizaron dos experimentos para evaluar la tolerancia de estas especies a eventos de inundación en los estadíos de plántula (implantación, Exp. 1) y de planta adulta (producción, Exp. 2). En el primer experimento plántulas de dos semanas de ambas especies fueron sometidas a tratamientos i) control, ii) sumersión parcial (SP) y iii) sumersión completa (SC) por 14 días, seguidos de un período de recuperación de 12 días. Ambas especies presentaron aerénquima constitutivo en sus raíces, que aumentó considerablemente bajo tratamientos de sumersión. La altura de plántulas sólo difirió de los controles en SC: C. gayana aumentó su altura rápidamente emergiendo del agua mientras P. coloratum no creció, manteniéndose siempre bajo el agua. La respuesta de "escape. exhibida por C. gayana se asoció a una asignación preferencial de biomasa hacia la parte aérea y un marcado alargamiento de las láminas foliares. P. coloratum no mostró cambios en la asignación de biomasa y sus hojas fueron más pequeñas. La biomasa final de C. gayana fue similar bajo SP y SC; mientras que P. coloratum redujo notablemente la biomasa bajo SC incluso al final del experimento, indicando que los daños provocados fueron persistentes. Así, C. gayana parece ser más prometedora para su implantación en pastizales propensos a inundaciones de mayor intensidad. En el segundo experimento se evaluó tolerancia a inundación de plantas adultas en diferentes momentos del ciclo de crecimiento (inundación invernal durante el reposo vegetativo vs. inundación primaveral durante el rebrote). Plantas de 10 meses (mayor a 30 cm altura) fueron sujetas a tratamientos (i) control (ii) inundación invernal por 50 días durante el reposo vegetativo (I), (iii) inundación primaveral por 20 días durante el rebrote (P) y (iv) inundación continua por 70 días (I+P). Luego, se dejaron crecer las plantas por 30 días para evaluar su recuperación. La inundación durante el invierno provocó una reducción del 43-57 por ciento en la biomasa aérea y radical de C. gayana; mientras que P. coloratum no fue afectado. C. gayana mostró una leve mejor tolerancia a la inundación primaveral en comparación a P. coloratum mientras que cuando fue inundada en ambos períodos se detectaron efectos aditivos depresores sobre su biomasa, efecto que en P. coloratum no fue registrado. Al final del período de recuperación las plantas de ambas especies lograron recuperarse completamente, alcanzando una biomasa equivalente a los controles independientemente del tratamiento previo de inundación. Estos resultados sugieren que ambas especies son tolerantes a la inundación en fase adulta, con un compromiso en la reducción de biomasa que no supera el 30 por ciento. Se concluye que ambas especies resultan promisorias para su introducción en los pastizales anegables de la Pampa Deprimida, destacándose C. gayana para sitios donde la intensidad de inundación puede comprometer la sumersión completa en las primeras fases del establecimiento de las plantas.
