Aim: To develop a TaqMan probe-based, highly sensitive and specific quantitative PCR (qPCR) assay for the detection and quantification of Mycoplasma suis in the blood of pigs. Methods and Results: Primers and probes specific to Myc. suis 16S rRNA gene were designed. The qPCR assay`s specificity, detection limit, intra- and inter-assay variability were evaluated and its performance was compared with a Myc. suis conventional PCR assay (cPCR). Blood of two experimentally infected pigs, 40 Indiana pigs, 40 Brazilian sows and 28 peccaries were tested. The assay detected as few as ten copies of Myc. suis plasmids and was 100-fold more sensitive than the cPCR. No cross-reactivity with nontarget pig mycoplasmas was observed. An average of 1.62 x 10(11) and 2.75 x 10(8) target copies ml(-1) of blood were detected in the acutely and chronically infected pigs, respectively. Three (7.5%) pigs and 32 (80.0%) sows were positive while all peccaries were negative for Myc. suis. Conclusion: The developed qPCR assay is highly sensitive and specific for Myc. suis detection and quantification. Significance and Impact of the Study: TaqMan qPCR is an accurate and quick test for detection of Myc. suis infected pigs, which can be used on varied instrumentation platforms.

Genetic diversity and population structure of Plasmodium viva-V parasites call predict the origin and Spread of novel Variants Within a population enabling Population specific malaria control measures. We analyzed the genetic diversity and population Structure of 425 P. vivax isolates from Sri Lanka, Myanmar, and Ethiopia using 12 trinucleotide and tetranucleotide microsatellite markers. All three parasite populations were highly polymorphic with 3-44 alleles per locus. Approximately 65% were multiple-clone infections. Mean genetic diversity (H(E)) was 0.7517 in Ethiopia, 0.8450 in Myanmar, and 0.8610 in Sri Lanka. Significant linkage disequilibrium Was maintained. Population structure showed two clusters (Asian and African) according to geography and ancestry Strong clustering of outbreak isolates from Sri Lanka and Ethiopia was observed. Predictive power of ancestry using two-thirds of the isolates as a model identified 78.2% of isolates accurately as being African or Asian. Microsatellite analysis is a useful tool for mapping short-term outbreaks of malaria and for predicting ancestry.

The present study reports the synthesis of a novel compound with the formula [Ru(2)(aGLA)(4)Cl] according to elemental analyses data, referred to as Ru(2)GLA. The electronic spectra of Ru(2)GLA is typical of a mixed valent diruthenium(II,III) carboxylate. Ru(2)GLA was synthesized with the aim of combining and possibly improving the anti-tumour properties of the two active components ruthenium and gamma-linolenic acid (GLA). The properties of Ru(2)GLA were tested in C6 rat glioma cells by analysing cell number, viability, lipid droplet formation, apoptosis, cell cycle distribution, mitochondrial membrane potential and reactive oxygen species. Ru(2)GLA inhibited cell proliferation in a time and concentration dependent manner. Nile Red staining suggested that Ru(2)GLA enters the cells and ICP-AES elemental analysis found all increase in ruthenium from <0.02 to 425 mg/Kg in treated cells. The sub-G1 apoptotic cell population was increased by Ru(2)GLA (22 +/- 5.2%) when analysed by FACS and this was confirmed by Hoechst staining of nuclei. Mitochondrial membrane potential was decreased in the presence of Ru(2)GLA (44 +/- 2.3%). In contrast, the cells which maintained a high mitochondrial membrane potential had an increase (18 +/- 1.5%) in reactive oxygen species generation. Both decreased mitochondrial membrane potential and increased reactive oxygen species generation may be involved in triggering apoptosis in Ru(2)GLA exposed cells. The EC(50) for Ru(2)GLA decreased with increasing time of exposure from 285 mu M at 24h, 211 mu M at 48 h to 81 mu M at 72 h. In conclusion, Ru(2)GLA is a novel drug with anti proliferative properties in C6 glioma cells and is a potential candidate for novel therapies in gliomas. Copyright (C) 2009 John Wiley & Sons, Ltd.

Several popular Machine Learning techniques are originally designed for the solution of two-class problems. However, several classification problems have more than two classes. One approach to deal with multiclass problems using binary classifiers is to decompose the multiclass problem into multiple binary sub-problems disposed in a binary tree. This approach requires a binary partition of the classes for each node of the tree, which defines the tree structure. This paper presents two algorithms to determine the tree structure taking into account information collected from the used dataset. This approach allows the tree structure to be determined automatically for any multiclass dataset.

Cerium doped yttrium aluminate perovskite YAlO(3) (YAP) powders are pursued as interesting alternatives to bulk crystals for application in scintillating devices. The emissions of these materials in the near-UV and visible spectral regions originate from electric dipole transitions between 4f and 5d energy levels of Ce(3+) and largely depend on the environment occupied by the ion. In search for improved synthesis conditions that can lead to phase pure powders with optimized structural and spectroscopic characteristics, in this work we have employed the polymeric precursor (Pechini) method to prepare crystalline and amorphous YAP:Ce powders doped with 1-10 mol% Ce(3+). Interesting composite materials were also obtained by dispersing some of the YAP:Ce powders in silica xerogels. A comparative structural and spectroscopic study of all the samples was done by XRD, FT-IR, emission, excitation and excited state lifetime measurements. In agreement with previous reports, excitation at 296 nm results in intense emission in the range 315-425 nm with an average decay time of 30 ns. (c) 2010 Elsevier B.V. All rights reserved.

There has been little research on metal concentration levels in urban soils of SA o pound Paulo, a city with 19 million inhabitants with severe pollution problems. In the present study, the concentration of As, Ba, Cr, Cu, Pb, Sb and Zn, were determined by INAA and XRF in surface soil samples from 7 public parks located within SA o pound Paulo city. The results obtained showed that soils of SA o pound Paulo public parks present concentration levels of the studied elements higher than the reference values for soils in SA o pound Paulo State. Traffic related elements such as Cu, Pb and Zn presented high concentrations in parks located near avenues of highly dense traffic and may be associated to vehicular sources.

The authors study the coincidence theory for pairs of maps from the Torus to the Klein bottle. Reidemeister classes and the Nielsen number are computed, and it is shown that any given pair of maps satisfies the Wecken property. The 1-parameter Wecken property is studied and a partial negative answer is derived. That is for all pairs of coincidence free maps a countable family of pairs of maps in the homotopy class is constructed such that no two members may be joined by a coincidence free homotopy.

Thin films of mixtures containing carboxymethylcellulose acetate butyrate (CMCAB) and carbohydrate based surfactant, namely, sorbitan monopalmitate (Span 40) or poly(oxyethylene) sorbitan monopalmitate (Tween 40) were spin-coated onto silicon wafers. The effect of surfactant concentration on resulting film morphology and surface toughness Was Studied by atomic force microscopy (AFM). Upon increasing the concentration of Span 40 in the mixture, films became rougher and more heterogeneous, indicating surface enrichment by Span 40 molecules. In the case of mixtures composed by CMCAB and Tween 40, the increase of Tween 40 in the mixture led to smoother and more homogeneous films, indicating compatibility between both components. Differential scanning calorimetry (DSC) revealed that Span 40 and Tween 40 act as plasticizers for CMCAB, leading to dramatic reduction of glass transition temperature of CMCAB, namely, Delta T(g) = -158 degrees C and Delta T(g)=-179 degrees C. respectively. The catalase mimetic complex Mn(III)-salen chloride (EUK8) was found to be pro-oxidant under low hydrogen peroxide concentrations. The increase in the fluorescence rate of the probe 1,2,3-dihydrorhodamine (DHR) in solution, as well as the carbonyl content of human serum albumin were found to be maximum at H(2)O(2):EUK8 molar ratios ranging from 0 to 2, supporting previous findings regarding the mechanism of EUK8 catalase activity and the formation of highly oxidative Mn(V)-O(2-) species. This pro-oxidant effect is precluded by the presence of glutathione. Cytotoxicity to HeLa cells, as probed by increased rate of oxidation of intracellular DHR, was not observed. Our findings suggest that the combination of H(2)O(2) and EUK8 at specific molar ratios, in the absence of reductants/antioxidants, induces the oxidation of organic molecules. It is shown that the fluorimetric determination of pro-oxidant activity of metal complexes is more sensitive than the colorimetric quantification of protein carbonyl content. The implications of our findings with respect to the somewhat confusing results arising from in vivo studies of EUK8 and other Mn(III) anti-oxidant metal complexes are discussed.

The effects of alkali treatment on the structural characteristics of cotton linters and sisal cellulose samples have been studied. Mercerization results in a decrease in the indices of crystallinity and the degrees of polymerization, and an increase in the alpha-cellulose contents of the samples. The relevance of the structural properties of cellulose to its dissolution is probed by studying the kinetics of cellulose decrystallization, prior to its solubilization in LiCl/N,N-dimethylacetamide (DMAc). Our data show that the decrystallization rate constants and activation parameters are only slightly dependent on the physico-chemical properties of the starting celluloses. This multi-step reaction is accompanied by a small enthalpy and large, negative, entropy of activation. These results are analyzed in terms of the interactions within the biopolymer chains during decrystallization, as well as those between the two ions of the electrolyte and both DMAc and cellulose.

This study investigates variable noun phrase number agreement (VNA) in two second language varieties of Portuguese, spoken in Maputo, Mozambique and in Mindelo, Cape Verde. Quantitative VARBRUL analysis is carried out based on recordings made in Maputo and Mindelo 2007 and 2008. Previous quantitative studies on VNA in varieties of Brazilian Portuguese (Guy, 1981; Lopes, 2001; Andrade, 2003) as well as on VNA in first and second language varieties of Portuguese from São Tomé (Baxter, 2004; Figueiredo, 2008, 2010) indicate contact between Portuguese and African languages as the main origin of this phenomenon. VNA in Brazilian Portuguese is, however, interpreted by Scherre (1988) and Naro & Scherre (1993, 2007) as the result of language internal drift. Varieties of Portuguese from Mozambique and Cape Verde are particularly interesting to contrast in order to investigate influences from African languages on VNA, as in Mozambique Bantu languages are first languages of the vast majority of Portuguese speakers, whereas in Cape Verde, practically all Portuguese speakers are first language speakers of Cape Verdean Creole, whose substrates are West African, and not Bantu, languages. Comparison is also made with previous studies from Brazil and São Tomé. The results of this study comment previously postulated explanations for VNA in Portuguese in various ways. The analysis of the variables onset age and age stratum indicates that VNA in the analyzed varieties is a phenomenon linked to the acquisition of Portuguese as a second language and/or language contact rather than the result of internal drift. The fact that all the compared varieties tend to mark plural on pre-head components contradicts Bantu transfer as an explanation for this pattern, and raises the need to also consider more general explanations based on language contact. The basic structural similarity between the compared varieties suggests the existence of a grammatical restructuring continuum.

In this paper we construct and analyze a growth model with the following three ingredients. (i) Technological progress is embodied. (ii) The production function of a firm is such that the firm makes both technology upgrade as well as capital and labor decisions. (iii) The firm’s production technology is putty-clay. We assume that there are disincentives to the accumulation of capital, resulting in a divergence between the social and the private cost of investment. We solve a single firm’s problem in this environment. Then we determine general equilibrium prices of capital goods of different vintages. Using these prices we aggregate firms’ decisions and construct the theoretical analogues of National Income statistics. This generates a relationship between disincentives and per capita incomes. We analyze this relationship and show the quantitative and qualitative roles of embodiment and putty-clay. We also show how the model is taken to data, quantified and used to determine to what extent income gaps across countries can be attributed to disincentives.

O presente estudo visou verificar o efeito sobre alguns parâmetros da seleção por gradiente de Percoll® e da adição de plasma seminal do sêmen eqüino preservado a +4oC. O primeiro experimento avaliou a taxa de recuperação de espermatozóides após seleção por Percoll® em diferentes protocolos de centrifugação. Foram realizadas 5 coletas de sêmen de um garanhão. Imediatamente após a coleta, o sêmen foi avaliado quanto à motilidade, vigor e concentração. Foram retiradas duas amostras de 4 mL, diluídas em leite desnatado UHT com concentrações de 50 e 100 x 106 espermatozóides por mL cada. Cada uma destas amostras foi dividida em 4 alíquotas de 1 mL, que foram então colocadas sobre Percoll® e submetidas a diferentes tempos e velocidades de centrifugação. V1 - 200 g (5 min) + 800 g (10 min); V2 - 800 g (10 min); V3 - 800 g (15 min); V4 - 800 g (20 min). Após esse processo, o sobrenadante foi desprezado e o pellet de cada alíquota ressuspendido com 0,5 mL de leite UHT. As 8 amostras foram novamente avaliadas para concentração, motilidade e vigor. O segundo experimento estudou o efeito da adição de plasma seminal de diferentes qualidades ao sêmen eqüino selecionado por gradiente de Percoll® e resfriado a +4°C por até 72 horas. Foram utilizados 40 ejaculados de 4 garanhões, sendo dois com boa qualidade de sêmen e dois com baixa qualidade de sêmen. Imediatamente após a coleta, o sêmen foi avaliado quanto à motilidade, vigor e concentração e preparadas cinco frações de 100x106 espermatozóides, diluídas 1:1 (v/v) em EDTA-Glicose. Quatro delas, constituídas por 1mL a 2mL, foram depositadas sobre Percoll®. A fração restante foi centrifugada em tubo de vidro de 10 mL, sob as mesmas condições de tempo e velocidade das demais amostras. Foi realizada centrifugação por 5 minutos em 200 g, seguida de 10 minutos em 800 g. O sobrenadante foi descartado e o pellet ressuspendido com leite UHT desnatado compondo os seguintes tratamentos: Sp: 1,5 mL de leite UHT desnatado sem adição de plasma seminal; Hp: 1,425 mL de leite UHT desnatado acrescido de 75 L de plasma seminal homólogo; Ap: 1,425 mL de leite UHT desnatado acrescido de 75 L de plasma seminal do pool de alta qualidade; Bp: 1,425mL de leite UHT desnatado acrescido de 75 L plasma seminal do pool de baixa qualidade; Cc: foi centrifugada sem seleção por Percoll®, teve seu sobrenadante descartado e ressuspendida com 2 mL de leite UHT; C : uma amostra de sêmen diluído em leite UHT foi mantida como controle no processo de armazenamento. As amostras foram resfriadas a +4°C e examinadas a cada 24h até as 72 horas em relação à motilidade, funcionalidade de membrana (teste hiposmótico) e integridade de membrana (CFDA/PI). A seleção por gradiente descontínuo de Percoll® 90/45% mostrou-se efetiva na recuperação de espermatozóides com motilidades progressiva e total. A adição de 5% de plasma seminal ou a ausência de plasma não influenciaram os valores da motilidade das amostras selecionadas por gradiente de Percoll®. A seleção por Percoll não influenciou na percentagem de células com membrana plasmática funcional. Concentrações superiores a 2% de plasma seminal resultaram em decréscimo do número de células com membrana funcional, enquanto que as amostras sem plasma seminal apresentaram os melhores resultados e o grupo controle, que apresentava a maior percentagem de plasma, os piores. A utilização de Percoll® não separou células com alteração de membrana. A percentagem de células com membrana completamente íntegra foi significativamente maior nas amostras que sofreram processo de centrifugação, independentemente da seleção. O processo de seleção por Percoll® foi efetivo na recuperação de espermatozóides de eqüino com motilidade progressiva, mas não selecionou espermatozóides quanto à funcionalidade nem à integridade de membrana. Concentrações inferiores a 2% de plasma seminal melhoraram a funcionalidade de membrana. A ausência de plasma seminal melhorou os resultados de integridade das membranas plasmática e acrossomal; e a adição de plasma de alta qualidade não melhorou a motilidade de espermatozóides selecionados por Percoll®.
