Autilização de filtros ultravioletas isolados em formulações fotoprotetoras, origina produtos com proteção limitada contra as radiações solares, o que evidencia a necessidade de associar compostos bioativos. Estudos anteriores demonstraram que a rutina, um composto bioativo, interage sinergicamente com filtros solares incorporados em preparações fotoprotetoras. Portanto, este trabalho teve como objetivo avaliar a influência da rutina sobre a estabilidade físico-química e funcional de emulsões fotoprotetoras. 16 formulações foram desenvolvidas, submetidas ao teste de estabilidade preliminar e caracterizadas de acordo com o pH, perfil reológico e eficácia fotoprotetora in vitro . A formulação com o melhor desempenho e a formulação correspondente, sem rutina, foram submetidos ao teste de estabilidade normal. Todas as formulações apresentaram valores de pH compatível com a pele e comportamento reológico semelhante. A formulação F16 e a mesma formulação, sem rutina, foram submetidas ao teste de estabilidade normal e apresentaram valores de pH semelhantes e perfis reológicos que foram mantidos ao longo dos dias de análise. Aatividade antirradicalar foi estável apenas para formulações armazenadas a 5,0 ± 0,5°C. A eficácia fotoprotetora demonstrou resultados semelhantes entre ambas as formulações, que também foi observado em todos os dias de análise. Em conclusão, rutina não influenciou a fotoestabilidade da formulação sob as condições adotadas.

O óleo de café verde (OCV) e formulações cosméticas que contendo 2,5, 5, 10 e 15% de óleo foram avaliados por métodos in vitro quanto às suas atividades antioxidante e antimicrobiana. O OCV e as suas formulações demostraram baixa actividade antioxidante, avaliada pelo método DPPH (42% do OCV foi equivalente a 0,002% de BHT). Não se observou atividade antimicrobiana para o OCV e as suas formulações contra Escherichia coli, Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, Bacilus subtilis, Propionibacterium acnes e Candida albicans, utilizando o método de difusão em poço. Embora o OCV seja utilizado há muitos anos em formulações cosméticas, ainda são necessários mais estudos para apoiar adequadamente a utilidade do óleo de café em produtos para cuidado da saúde da pele e em cosméticos.

O uso de plantas medicinais é uma tendência generalizada na medicina popular. O género Plectranthus pertence à família das Lamiaceae tal como a salva (Salvia), o manjericão (Ocimum) e a hortelã (Mentha), que apresentam uma grande diversidade de usos etnobotânicos. No presente trabalho, foram avaliadas as atividades antimicrobiana e antioxidante de extractos de cinco espécies do género Plectranthus: P. hadiensis (vd), P. madagascarensis, P. neochilus, P. verticillatus e P. barbatus. Os extractos foram obtidos por diversos métodos: infusão, decocção, ultra-sons, micro-ondas e maceração (10% m/v de planta seca) com dois solventes diferentes (água e acetona). O método de infusão permitiu obter a maior quantidade de extrato seco a partir da planta Plectranthus barbatus (22 ± 1,0 – 33 ± 3,0 mg/mL). A actividade antimicrobiana dos extratos obtidos foi determinada usando o método da difusão em poço e foi testada contra três bactérias Gram-positivas e duas bactérias Gram-negativas. Só os extratos acetónicos de P. madagascarensis, P. verticillatus e P. hadiensis (vd) demonstraram ser ativos e exclusivamente em bactérias Gram-positivas. O extrato de P. madagascarensis demonstrou a maior actividade antibacteriana com halos de inibição entre 23 e 36 mm. A atividade antioxidante dos extratos foi determinada qualitativamente usando o método do radical 2,2-difenil-1-picril-hidrazina (DPPH) em ccd obtiveram-se resultados positivos na maioria dos extractos estudados.

A aplicação tópica de antioxidantes provou ser eficaz em proteger a pele contra danos oxidativos. O extracto de Matricaria chamomilla L. tem sido amplamente utilizado em formulações cosméticas. Assim, o objectivo deste estudo foi avaliar o potencial antioxidante do extracto de camomila, bem como a eficácia clínica na hidratação e nas propriedades mecânicas da pele. A actividade antioxidante do extracto de camomila foi avaliada por quimioluminescência (IC50 = 0,14 ug / mL). As formulações foram suplementadas com a α -bisabolol de fração mássica 0,5 cg/g ou com extracto glicólico de Matricaria chamomilla L. de fração mássica 5,0 cg/g ou com 0,01 cg/g de apigenina e aplicados na parte interna do antebraço e na cara de 25 mulheres. A fisiologia da pele foi avaliada antes e após 2 horas (aplicação única) e 2 e 4 semanas de aplicação diária. Depois de uma única aplicação, todas as formulações aumentaram a água do estrato córneo, mas apenas as formulações com α -bisabolol e extracto de camomila reduziram a PTEA . A formulação com o extracto de camomila mostrou o resultado mais marcante na redução PTEA (27%). No entanto, após 2 - e 4- semanas de aplicação, apenas a formulação contendo o extracto aumentou o a água no estrato córneo. O extracto de camomila, foi eficaz na neutralização de radicais livres e, portanto, apresenta potencial em formulações cosméticas com este propósito.

The ferric complexing capacity of four phenolic compounds, occurring in olives and virgin olive oil, namely, oleuropein, hydroxytyrosol, 3,4-dihydroxyphenylethanol-elenolic acid (3,4-DHPEA-EA), and 3,4-dihydroxyphenylethanol-elenolic acid dialdehyde (3,4-DHPEA-EDA), and their stability in the presence of ferric ions were studied. At pH 3.5, all compounds formed a reversible 1:1 complex with ferric ions, but hydroxytyrosol could also form complexes containing > 1 ferric ion per phenol molecule. At pH 5.5, the complexes between ferric ions and 3,4-DHPEA-EA or 3,4-DHPEA-EDA were relatively stable, indicating that the antioxidant activity of 3,4-DHPEA-EA or 3,4-DHPEA-EDA at pH 5.5 is partly due to their metal-chelating activity. At pH 7.4, a complex containing > 1 ferric ion per phenol molecule was formed with hydroxytyrosol. Oleuropein, 3,4-DHPEA-EA, and 3,4-DHPEA-EDA also formed insoluble complexes at this pH. There was no evidence for chelation of Fe(II) by hydroxytyrosol or its derivatives. At all pH values tested, hydroxytyrosol was the most stable compound in the absence of Fe(III) but the most sensitive to the presence of Fe(III).

Olive oil, a typical ingredient of the Mediterranean diet, possesses many beneficial health effects. The biological activities ascribed to olive oil consumption are associated in part to its phenolics constituents, and mainly linked to the direct or indirect antioxidant activity of olive oil phenolics and their metabolites, which are exerted more efficiently in the gastrointestinal (GI) tract, where dietary phenolics are more concentrated when compared to other organs. In this regard, we present a brief overview of the metabolism, biological activities, and anticancer properties of olive oil phenolics in the GI tract. Rocket is a leafy brassicaceous salad crop that encompasses two major genera (Diplotaxis and Eruca) and many different cultivars. Rocket is a rich source of antioxidants and glucosinolates, many of which are produced as secondary products by the plant in response to stress. In this paper we examined the impact of temperature and light stress on several different cultivars of wild and salad rocket. Growth habit of the plants varied in response to stress and with different genotypes, reflecting the wide geographical distribution of the plant and the different environments to which the genera have naturally adapted. Preharvest environmental stress and genotype also had an impact on how well the cultivar was able to resist postharvest senescence, indicating that breeding or selection of senescence-resistant genotypes will be possible in the future. The abundance of key phytonutrients such as carotenoids and glucosinolates are also under genetic control. As genetic resources improve for rocket it will therefore be possible to develop a molecular breeding programme specifically targeted at improving stress resistance and nutritional levels of plant secondary products. Concomitantly, it has been shown in this paper that controlled levels of abiotic stress can potentially improve the levels of chlorophyll, carotenoids and antioxidant activity in this leafy vegetable.

Several bis-malonatooxidovanadium(IV) complexes of the general type [M(2)(H2(O))(n)][VO(mal)(2)(H(2)O)] (where M = Li(1), Na(2), K(3), Cs(4) and NH4(5); n = 3.5, 1, 3, 1 and 1, respectively) were isolated in good yield and high purity. These complexes were fully characterized by various physicochemical techniques (elemental analysis, UV- Vis, IR, EPR, CV, etc.) complexes 1, 2 and 3 were structurally characterized by single crystal X- ray diffraction technique. In vivo antidiabetic properties of bis- malonato complexes 1, 2, 3 and 5 have been studied using Streptozotocin induced diabetic rats. Significant lowering of blood sugar level has been noticed. At the same time these complexes were found to regulate secondary pathophysiological complications like liver damage and lowering of the total antioxidant status (TAS) in diabetic rats. Results of these study are expected to a expand the possibility of designing new oxidovanadium(IV) complexes of O, O chelating ligands with significant antidiabetic properties

The radiation food processing has been demonstrating great effectiveness in the attack of pathogenic agents, while little compromising nutritional value and sensorial properties of foods. The mate (Ilex paraguariensis), widely consumed product in South America, generally in the form of infusions with hot or cold water, calls of chimarrao or terere, it is cited in literature as one of the best sources phenolic compounds. The antioxidants action of these constituent has been related to the protection of the organism against the free radicals, generated in alive, currently responsible for the sprouting of some degenerative illness as cancer, arteriosclerosis, rheumatic arthritis and cardiovascular clutters among others. The objective of that work was to evaluate the action of the processing for gamma radiation in phenolic compounds of terere beverage in the doses of 0, 3, 5, 7 and 10 kGy. The observed results do not demonstrate significant alterations in phenolic compounds of terere beverage processed by gamma radiation. Crown Copyright (C) 2009 Published by Elsevier Ltd. Mate (Ilex paraguariensis) is rich in polyphenolic compounds, which are thought to contribute to the health benefits of tea. Mate tea was administered orally to mice at a dose of 0.5, 1.0 or 2.0 g/kg for 60 d, and changes both in serum lipid concentration and fatty acid composition of liver and kidney were examined. The effects of mate tea on serum and tissue lipid peroxidation were assessed by the evaluation of thiobarbituric acid-reactive substances (TBARS). In tea-consuming mice, both MUFA (18: 1n-9) and PUFA (18: 2n-6 and 20: 4n-6) were increased (P<0.05) in the liver lipid (approximately 90 and 60%, respectively), whereas only MUFA (approximately 20%) were increased in the kidney lipid. The most altered PUFA class was n-6 PUFA, which increased by approximately 60-75 % (P<0.05). This difference in the fatty acid profile in the liver is reflected in the increased PUFA:SFA ratio. Consistent with these results, mice fed with mate tea had much lower TBARS in the liver. No differences (P>0.05) were found in the levels of serum cholesterol, HDL-cholesterol and TAG under the conditions of the present study. These results suggest that treatment with mate tea was able to protect unsaturated fatty acids from oxidation and may have selective protective effects within the body, especially on the liver.

Yerba mate (Ilex paraguariensis) is rich in polyphenols, especially chlorogenic acids. Evidence suggests that dietary polyphenols could play a role in glucose absorption and metabolism. The aim of this study was to evaluate the antidiabetic properties of yerba mate extract in alloxan-induced diabetic Wistar rats. Animals (n = 41) were divided in four groups: nondiabetic control (NDC, n = 10), nondiabetic yerba mate (NDY, n = 10), diabetic control (DC, n = 11), and diabetic yerba mate (NDY, n = 10). The intervention consisted in the administration of yerba mate extract in a 1 g extract/kg body weight dose for 28 days; controls received saline solution only. There were no significant differences in serum glucose, insulin, and hepatic glucose-6-phosphatase activity between the groups that ingested yerba mate extract (NDY and DY) and the controls (NDC and DC). However, the intestinal SGLT1 gene expression was significantly lower in animals that received yerba mate both in upper (p = 0.007) and middle (p < 0.001) small intestine. These results indicate that bioactive compounds present in yerba mate might be capable of interfering in glucose absorption, by decreasing SGLT1 expression.

No evidence for the role of protease-activated receptor-2 (PAR(2)) in human periodontal disease has been demonstrated so far. Thus, we sought to investigate the expression of PAR(2) mRNA in chronic periodontitis, and to examine whether its expression is related to the presence of PAR(2) potential activators. Microbiological and gingival crevicular fluid samples were collected from individuals with chronic periodontitis and control individuals, and the presence of neutrophil serine proteinase 3 (P3) and Porphyromonas gingivalis was evaluated. PAR(2) mRNA expression was higher (p < 0.001) in those with chronic periodontitis compared with control individuals, and it was statistically decreased (p = 0.0006) after periodontal treatment. Furthermore, those with chronic periodontitis presented higher (p < 0.05) levels of IL-1 alpha, IL-6, IL-8, and TNF-alpha, total proteolytic activity, P. gingivalis prevalence, and P3mRNA expression compared with control individuals. We conclude that PAR(2) mRNA expression and its potential activators are elevated in human chronic periodontitis, therefore suggesting that PAR(2) may play a role in periodontal inflammation.

We investigated the toxic effect of indole-3-acetic acid (IAA) combined with horseradish peroxidase (HRP) on Prototheca zopfii from bovine mastitis. P. zopfii isolates were identified and characterized by morpho-physiological parameters; presences of P. zopfii genotype 2 were also investigated. Subsequently, P. zopfii was incubated in the absence (control) or presence of IAA/HRP and examined for: (i) cell viability; (ii) colonies number formation; (iii) antioxidant enzyme activity; and (iv) DNA integrity. Significance of differences was calculated using ANOVA and Tukey`s test (P a parts per thousand currency sign 0.05). As evidenced by Trypan blue exclusion and colony formation in Sabouraud dextrose agar, IAA/HRP addition to the culture reduced respective P. zopfii viability and P. zopfii colony formation in a concentration- and time-dependent manner. IAA/HRP specifically reduced cell viability in 10, 15, 20, 25, and 32% after 4, 6, 8, 10, and 12 h of incubation, respectively, compared with the control at the same time. The number of colony formation was inhibited (45, 82, and 88%) by IAA/HRP after 4, 6, and 9 h of incubation, respectively, compared with the control at the same time. In addition, P. zopfii antioxidant activity increased measurably in the presence of IAA/HRP (6 h); superoxide dismutase, catalase, glutathione reductase, and glutathione peroxidase increased by 90, 120, 150% and 3.4 times, compared with the controls. IAA/HRP did not appear to effect P. zopfii DNA integrity when examined by electrophoresis. In conclusion, IAA/HRP appears to function as a microbicidal mechanism on P. zopfii genotype 2 from bovine mastitis.

The aim of this study was to investigate endothelial venous function, mflammatory markers, and systemic oxidative stress after an oral lipid overload (OLO). We studied 18 healthy adults (9 men; age, 29.2 +/- 0.9 years; body mass index, 22.3 +/- 0.4 kg/m(2)). Blood samples were collected in the fasting state and 3, 4, and 5 hour after the OLO (1000 kcal, 58% fat) for metabolic variables, oxidative stress, inflammatory markers, adiponectin, and resistin. Changes in vein diameter to phenylephrine, acetylcholine, and sodium nitroprusside (dorsal hand vein technique) were measured before and after the OLO. Oral lipid overload increased triglycerides (61 +/- 6 vs 134 +/- 17 mg/dL, P <.001), insulin (7.2 +/- 0.8 vs 10.7 +/- 1.3 mu U/mL, P <.05), and resistin (5.38 +/- 0.5 vs 6.81 +/- 0.7 ng/mL, P <.05) and reduced antioxidant capacity (plasma total antioxidant capacity: 186.7 +/- 56 vs 161.8 +/- 50 U Trolox per microliter plasma, P <.01), vascular reactivity (171.3 +/- 85 vs 894.4 +/- 301 ng/mL, P <.001), and maximum acetylcholine venodilation (105.9% +/- 9% vs 61.0% +/- 7%, P <.05). No changes were observed for sodium nitroprusside. Post-OLO triglycerides were positively correlated with phenylephrine dose (rho = 0.38, P <.05) and resistin (rho = 0.43, P <.01) and negatively correlated with the maximum acetylcholine venodilation (rho = -0.36, P <.05). In conclusion, an OLO impaired venoconstriction responsiveness in healthy subjects, probably because of a reduction in the antioxidant capacity. (C) 2008 Elsevier Inc. All rights reserved.

Oxidative stress has been associated with normal aging and Alzheimer`s disease (AD). However, little is known about oxidative stress in mild cognitive impairment (MCI) patients who present a high risk for developing AD. The aim of this study was to investigate plasma production of the lipid peroxidation marker, malonaldehyde (MDA) and to determine, in erythrocytes, the enzymatic antioxidant activity of catalase, glutathione peroxidase (GPx), glutathione reductase (GR), and glutathione S-transferase (GST) in 33 individuals with MCI, 29 with mild probable AD and 26 healthy aged subjects. GR/GPx activity ratio was calculated to better assess antioxidant defenses. The relationship between oxidative stress and cognitive performance was also evaluated by the Mini Mental State Examination (MMSE). AD patients showed higher MDA levels than both MCI and healthy elderly subjects. MCI subjects also exhibited higher MDA levels compared to controls. Catalase and GPx activity were similar in MCI and healthy individuals but higher in AD. GR activity was lower in MCI and AD patients than in healthy aged subjects. Additionally, GR/GPx ratio was higher in healthy aged subjects, intermediate in MCI and lower in AD patients. No differences in GST activity were detected among the groups. MMSE was negatively associated with MDA levels (r = -0.31, p = 0.028) and positively correlated with GR/GPx ratio in AD patients (r = 0.68, p < 0.001). MDA levels were also negatively correlated to GR/GPx ratio (r = -0.31, p = 0.029) in the AD group. These results suggest that high lipid peroxidation and decreased antioxidant defenses may be present early in cognitive disorders.
