Bliacheriene F, Carmona MJC, Barretti CFM, Haddad CMF, Mouchalwat ES, Bortlotto MRFL, Francisco RPV, Zugaib M - Use of a Minimally Invasive Uncalibrated Cardiac Output Monitor in Patients Undergoing Cesarean Section under Spinal Anesthesia: Report of Four Cases. Background and Objectives: Hemodynamic changes are observed during cesarean section under spinal anesthesia. Non-invasive blood pressure (BP) and heart rate (HR) measurements are performed to diagnose these changes, but they are delayed and inaccurate. Other monitors such as filling pressure and cardiac output (CO) catheters with external calibration are very invasive or inaccurate. The objective of the present study was to report the cardiac output measurements obtained with a minimally invasive uncalibrated monitor (LiDCO rapid) in patients undergoing cesarean section under spinal anesthesia. Case report: After approval by the Ethics Commission, four patients agreed to participate in this study. They underwent cesarean section under spinal anesthesia while at the same time being connected to the LiDCO rapid by a radial artery line. Cardiac output, HR, and BP were recorded at baseline, after spinal anesthesia, after fetal and placental extraction, and after the infusion of oxytocin and metaraminol. We observed a fall in BP with an increase of HR and CO after spinal anesthesia and oxytocin infusion; and an increase in BP with a fall in HR and CO after bolus of the vasopressor. Conclusions: Although this monitor had not been calibrated, it showed a tendency for consistent hemodynamic data in obstetric patients and it may be used as a therapeutic guide or experimental tool.

A nongravimetric acetyl bromide lignin (ABL) method was evaluated to quantify lignin concentration in a variety of plant materials. The traditional approach to lignin quantification required extraction of lignin with acidic dioxane and its isolation from each plant sample to construct a standard curve via spectrophotometric analysis. Lignin concentration was then measured in pre-extracted plant cell walls. However, this presented a methodological complexity because extraction and isolation procedures are lengthy and tedious, particularly if there are many samples involved. This work was targeted to simplify lignin quantification. Our hypothesis was that any lignin, regardless of its botanical origin, could be used to construct a standard curve for the purpose of determining lignin concentration in a variety of plants. To test our hypothesis, lignins were isolated from a range of diverse plants and, along with three commercial lignins, standard curves were built and compared among them. Slopes and intercepts derived from these standard curves were close enough to allow utilization of a mean extinction coefficient in the regression equation to estimate : lignin concentration in any plant, independent of its botanical origin. Lignin quantification by use of a common regression equation obviates the steps of lignin extraction, isolation, and standard curve construction, which substantially expedites the ABL method. Acetyl bromide lignin method is a fast, convenient analytical procedure that may routinely be used to quantify lignin.

Objective. The aim of this study was to assess the effect of cigarette smoke inhalation (CSI) on gene expression in alveolar bone healing sites. Study design. Wistar rats were randomly assigned to the groups: control [animals not exposed to CSI (n = 20)] and test [animals exposed to CSI, starting 3 days before teeth extraction and maintained until killing them (n = 20)]. First mandibular molars were bilaterally extracted, and the expression of alkaline phosphatase, bone morphogenetic protein (BMP) 2 and 7, receptor activator of nuclear factor kappa B ligand, osteoprotegerin, and d2 isoform of vacuolar adenosine triphosphatase V(o) domain were assessed by quantitative polymerase chain reaction in the newly formed tissue in the sockets. Results. Overall, data analysis demonstrated that CSI significantly affected the expression pattern of all of the studied genes except BMP-7. Conclusion. The expression of key genes for bone healing may be affected by CSI in tooth extraction sites. (Oral Surg Oral Med Oral Pathol Oral Radiol Endod 2010;110:447-452)

The objective of this review is to summarize developments in the use of quantitative affinity chromatography to determine equilibrium constants for solute interactions of biological interest. Affinity chromatography is an extremely versatile method for characterizing interactions between dissimilar reactants because the biospecificity incorporated into the design of the affinity matrix ensures applicability of the method regardless of the relative sizes of the two reacting solutes. Adoption of different experimental strategies, such as column chromatography, simple partition equilibrium experiments, solid-phase immunoassay, and biosensor technology, has led to a situation whereby affinity chromatography affords a means of characterizing interactions governed by an extremely broad range of binding affinities-relatively weak interactions (binding constants below 10(3) M-1) through to interactions with binding constants in excess of 10(9) M-1. In addition to its important role in solute separation and purification, affinity chromatography thus also possesses considerable potential for investigating the functional roles of the reactants thereby purified. A new strategy has been developed for the rapid synthesis of peptide para-nitroanilides (pNA). The method involves derivatization of commercially available tritylchloride resin (TCP-resin) with 1,4-phenylenediamine, subsequent coupling with desired amino acids by the standard Fmoc protocol, and oxidation of the intermediate para-aminoanilides (pAA) with Oxone(R). This procedure allows easy assembly of the desired para-aminoanilides (pAA) on standard resin and efficient oxidation and purification of the corresponding para-nitroanilides (pNA). The method allows easy access to any desired peptide para-nitroanilides, which are useful substrates for the characterization and study of proteolytic enzymes.

This review summarizes the development of exclusion chromatography, also termed gel filtration, molecular-sieve chromatography and gel permeation chromatography, for the quantitative characterization of solutes and solute interactions. As well as affording a means of determining molecular mass and molecular mass distribution, the technique offers a convenient way of characterizing solute selfassociation and solute-ligand interactions in terms of reaction stoichiometry and equilibrium constant. The availability of molecular-sieve media with different selective porosities ensures that very little restriction is imposed on the size of solute amenable to study. Furthermore, access to a diverse array of assay procedures for monitoring the column eluate endows analytical exclusion chromatography with far greater flexibility than other techniques from the viewpoint of solute concentration range that can be examined. In addition to its widely recognized prowess as a means of solute separation and purification, exclusion chromatography thus also possesses considerable potential for investigating the functional roles of the purified solutes. With recent advances in molecular biology, it is now possible to use the trace amounts of DNA in faeces to non-invasively sample endangered species for genetic studies. A highly vulnerable population of approximately 100 great bustards (Otis tarda) exists in Morocco necessitating the use of non-invasive protocols to study their genetic structure. Here we report a reliable silica-based method to extract DNA from great bustard faeces. We found that successful extraction and amplification correlated strongly with faeces freshness and composition. We could not extract amplifiable DNA from 30% of our samples as they were dry or contained insect material. However 100% of our fresh faecal samples containing no obvious insect material worked, allowing us to assess the levels of genetic variation among 25 individuals using a 542 bp control region sequence. We were able to extract DNA from four out of five other avian species, demonstrating that faeces represents a suitable source of DNA for population genetics studies in a broad range of species.

A pimenta rosa é o fruto da aroeira (Schinus terebinthifolius Raddi), uma planta nativa do Brasil. A extração e caracterização do seu óleo essencial viabiliza a sua utilização industrial. Este trabalho teve como objetivo avaliar o efeito da adição de óleo essencial de pimenta rosa em queijo Minas Frescal . Foram identificados 95,36% dos constituintes do óleo essencial, sendo o δ-careno-3 o componente majoritário. O óleo essencial de pimenta rosa apresentou características antioxidante (2,53 ± 0,28 μmols de trolox por mL de óleo) e antimicrobiana para o desenvolvimento de Staphylococcus aureus. Foi observado, no teste de difusão em ágar, a formação do halo de inibição de 1,35 ± 0,32 cm, e na análise de concentração inibitória mínima, o valor de 3,13%. O óleo essencial é sensível à degradação no meio em que se encontra, portanto necessita de processos tecnológicos para assegurar sua ação. Para proteger o óleo essencial foi realizada a microencapsulação por meio da secagem por spray dryer. O óleo foi microencapsulado utilizando três formulações com diferentes concentrações de goma arábica, maltodextrina e amido modificado como papel de parede. As microcápsulas obtidas foram analisadas quanto às suas características físico-químicas e morfológicas e foi selecionada uma formulação para posterior adição ao queijo Minas Frescal. A formulação com 5% de amido modificado, 10% de maltodextrina e 5% de goma arábica foi selecionada. Para definição da concentração de óleo essencial que foi utilzada no queijo, foi realizado teste sensorial de ordenação-preferência, visto que a concentração inibitória mínima (3,13%) era muito alta para ser utilizada nos queijos. A formulação com 0,01% de óleo essencial foi a preferida e utilizada para fabricação dos queijos. Foram elaborados e caracterizados os queijos controle (CO) e o queijo com adição das microcápsulas de óleo essencial de pimenta rosa (OEPR). Em relação às características físico-químicas, os queijos CO e OEPR só apresentaram diferença significativa em relação à umidade (P= 0,0021; α=0,05). O queijo OEPR teve boa aceitação sensorial com média de aceitação global de 7,6 ± 0,97, não apresentando diferença significativa, ao nível de 95% de confiança, do queijo controle (7,6 ± 1,18). Para avaliar a capacidade do óleo essencial de pimenta rosa microencapsulado em inibir o desenvolvimento de S. aureus, foram inoculadas nos queijos CO e OEPR concentrações iniciais de 106 UFG.g. Os queijos foram armazenados em BOD à temperatura de 4 ± 1 °C até realização das análises, que ocorreram nos tempos: 0, 3, 6, 9, 12, 15 e 30 dias. Houve redução de 1,53 ciclos Log no queijo adicionado de óleo essencial de pimenta rosa. De acordo com os resultados obtidos, conclui-se que o óleo essencial de pimenta rosa é uma alternativa viável de utilização em queijo Minas Frescal , devido ao seu potencial antioxidante, antimicrobiano e à sua aceitação sensorial.

Humic substances isolated from soil organic matter had been used as stimulators of plant metabolism. Arabidopsis thaliana (L.) Heynh. with only five chromosomes, short cycle and size, is an important model to evaluate the physiological effects of these substances, which are qualitatively and quantitatively influenced by morphogenesis, mineralogy and chemistry of soils. The objective of this study was to evaluate the ambience effects on bioactivity of humic acids. A and B horizons of four typical soils of the North Fluminense were sampled. After isolation and purification, humic acids were applied to plants in increasing concentrations. The number and length of lateral roots and main root length were evaluated and, subsequently, the concentrations of maximum stimulation were determined by dose-response curves and regression equations. The results showed that more stable humic acids isolated from soil in less advanced stages of weathering, high activity clay and high base saturation resulted in better physiological stimulants for Arabidopsis.

The use of iris recognition for human authentication has been spreading in the past years. Daugman has proposed a method for iris recognition, composed by four stages: segmentation, normalization, feature extraction, and matching. In this paper we propose some modifications and extensions to Daugman's method to cope with noisy images. These modifications are proposed after a study of images of CASIA and UBIRIS databases. The major modification is on the computationally demanding segmentation stage, for which we propose a faster and equally accurate template matching approach. The extensions on the algorithm address the important issue of pre-processing that depends on the image database, being mandatory when we have a non infra-red camera, like a typical WebCam. For this scenario, we propose methods for reflection removal and pupil enhancement and isolation. The tests, carried out by our C# application on grayscale CASIA and UBIRIS images show that the template matching segmentation method is more accurate and faster than the previous one, for noisy images. The proposed algorithms are found to be efficient and necessary when we deal with non infra-red images and non uniform illumination.

A Diabetes mellitus tipo 2 (DM2) é uma patologia de etiologia múltipla à qual estão associados vários factores genéticos. A Enzima Conversora da Angiotensina (ECA) tem sido alvo de vários estudos pela sua relação com factores pró-inflamatórios, pró-oxidantes e pró-fibrose, sendo o polimorfismo de Inserção/Delecção o mais estudado. Neste contexto, o objectivo deste estudo é assim verificar a distribuição deste polimorfismo numa amostra de indivíduos de nacionalidade portuguesa e verificar a sua possível associação com a DM2. Para tal, foram analisadas 87 amostras (controlos n =24 e diabéticos n =63) de indivíduos de nacionalidade portuguesa. As amostras foram submetidas a um processo de extracção de ADN, sendo posteriormente amplificadas por Polymerase Chain Reaction e analisadas por eletroforese em gel de agarose a 1%. Observou-se uma prevalência de 8% (n=7) com genótipo I/I, 38% (n=33) com genótipo I/D e 54% (n=47) com genótipo D/D. A amostra em estudo demonstrou assim estar sob o equilíbrio Hardy-Weinberg. Observou-se também uma associação entre níveis mais elevados de glicemia e o genótipo I/I (p=0,019). Na análise da utilização de insulina no controlo dos níveis de glicemia na DM2, observou-se uma maior proporção de indivíduos com genótipo D/D. Este estudo demonstra a importância do investimento da caracterização genética em patologias metabólicas multifactoriais como a DM2.

Contexto: a bronquiolite aguda é a principal patologia a afectar a criança nos primeiros 2 anos de vida, a fisioterapia respiratória é uma intervenção terapêutica utilizada com a intenção de melhorar o curso desta doença mantendo-se a incerteza sobre a sua eficácia. Objectivo: determinar a eficácia e segurança da fisioterapia respiratória em crianças com menos de 2 anos com bronquiolite aguda. Fontes de Informação: Medline (1966 a Agosto 2010), EMBASE(1990 a Agosto 2010), Pedro e Lilacs (1982 a Agosto 2010). Outra fonte de informação incluiu a bibliografia dos estudos obtidos. Selecção de estudos: estudos experimentais comparando a fisioterapia respiratória com cuidados habituais, em crianças com menos de 2 anos e bronquiolite aguda, em ventilação espontânea, em qualquer contexto. Estudos pré-experimentais ou observacionais com os mesmos participantes e intervenções foram admitidos complementarmente aos experimentais. Extracção de dados e análise: um investigador extraiu os dados dos artigos obtidos e avaliou o risco de viés. A eficácia e segurança da fisioterapia respiratória foram determinadas pelos seguintes outcomes: duração do internamento hospitalar ou do evento, variação de scores de severidade clínica, saturação periférica e suplementação de oxigénio, recidivas, recurso a antibióticos e efeitos deletérios ou deterioração clínica reportada. Síntese de dados: 6 estudos experimentais foram admitidos. As suas amostras provinham de criança internadas em hospital. As técnicas de fisioterapia respiratória foram comparadas com cuidados habituais. Nenhum estudo evidenciou melhoria dos outcomes de interesse na comparação entre grupos, excepto avaliações de curta duração da saturação periférica de oxigénio e scores de severidade clínica. 1 estudo reportou uma percentagem significativamente maior no grupo submetido a fisioterapia respiratória de crianças que vomitaram, tiveram uma desestabilização respiratória transitória, e na percepção de stress da criança pelos cuidadores. São relatadas ainda fracturas costais a causa de fisioterapia respiratória. Limitações: o risco de viés era alto em 2 estudos, baixo num estudo e indeterminado nos restantes. Conclusões: aparentemente a fisioterapia respiratória não é eficaz e pode produzir efeitos deletérios importantes, mas a evidência é pobre, carecendo de novos estudos.

This work addresses the treatment by nanofiltration (NF) of solutions containing NaCN and NH(4)Cl at various pH values. The NF experiments are carried out in a Lab-Unit equipped with NF-270 membranes for model solutions that are surrogates of industrial ammoniacal wastewaters generated in the coke-making processes. The applied pressure is 30 bar. The main objective is the separation of the compounds NaCN and NH(4)Cl and the optimization of this separation as a function of the pH. Membrane performance is highly dependent on solution composition and characteristics, namely on the pH. In fact, the rejection coefficients for the binary model solution containing sodium cyanide are always higher than the rejections coefficients for the ammonium chloride model solution. For ternary solutions (cyanide/ammonium/water) it was observed that for pH values lower than 9 the rejection coefficients to ammonium are well above the ones observed for the cyanides, but for pH values higher than 9.5 there is a drastic decrease in the ammonium rejection coefficients with the increase of the pH. These results take into account the changes that occur in solution, namely, the solute species that are predominant, with the increase of the pH.
