Oggigiorno si osserva a livello mondiale un continuo aumento dei consumi di acqua per uso domestico, agricolo ed industriale che dopo l’impiego viene scaricata nei corpi idrici (laghi, fiumi, torrenti, bacini, ecc) con caratteristiche chimico fisiche ed organolettiche completamente alterate, necessitando così di specifici trattamenti di depurazione. Ricerche relative a metodi di controllo della qualità dell’acqua e, soprattutto, a sistemi di purificazione rappresentano pertanto un problema di enorme importanza. I trattamenti tradizionali si sono dimostrati efficienti, ma sono metodi che operano normalmente trasferendo l’inquinante dalla fase acquosa contaminata ad un’altra fase, richiedendo perciò ulteriori processi di depurazione. Recentemente è stata dimostrata l’efficacia di sistemi nano strutturati come TiO2-Fe3O4 ottenuto via sol-gel, nella foto-catalisi di alcuni sistemi organici. Questo lavoro di tesi è rivolto alla sintesi e caratterizzazione di un catalizzatore nanostrutturato composito costituito da un core di Fe3O4 rivestito da un guscio di TiO2 separate da un interstrato inerte di SiO2, da utilizzare nella foto-catalisi di sistemi organici per la depurazione delle acque utilizzando un metodo di sintesi alternativo che prevede un “approccio” di tipo colloidale. Partendo da sospensioni colloidali dei diversi ossidi, presenti in commercio, si è condotta la fase di deposizione layer by layer via spray drying, sfruttando le diverse cariche superficiali dei reagenti. Questo nuovo procedimento permette di abbattere i costi, diminuire i tempi di lavoro ed evitare possibili alterazioni delle proprietà catalitiche della titania, risultando pertanto adatto ad una possibile applicazione su scala industriale. Tale sistema composito consente di coniugare le proprietà foto-catalitiche dell’ossido di titanio con le proprietà magnetiche degli ossidi di ferro permettendo il recupero del catalizzatore a fine processo. Il foto-catalizzatore è stato caratterizzato durante tutte la fasi di preparazione tramite microscopia SEM e TEM, XRF, Acusizer, spettroscopia Raman e misure magnetiche. L’attività foto-calitica è stata valutata con test preliminari utilizzando una molecola target tipo il rosso di metile in fase acquosa. I risultati ottenuti hanno dimostrato che il sistema core-shell presenta inalterate sia le proprietà magnetiche che quelle foto-catalitiche tipiche dei reagenti.

Es wurde ein genomischer DNA-Array der Modellpflanze Arabidopsis thaliana mit einer 13.800 EST-Klone umfassenden cDNA-Bibliothek entwickelt und in der Genexpressionsanalyse der pflanzlichen Pathogenabwehr eingesetzt. Mittels PCR-Amplifikation sind 13.000 PCR-Produkte der cDNA-Fragmente hergestellt worden, mit denen 66 genomische Arabidopsis-Arrays auf Nylon und Polypropylen als Trägermaterial hergestellt werden konnten. Die Validierung mit Fluoreszenz- und Radiaktivhybridisierung sowie der Vergleich von drei Normalisierungsmethoden führte zu reproduzierbaren Ergebnissen bei hohem Korrelationskoeffizienten. Die etablierte DNA-Array-Technologie wurde zur Genexpressionsanalyse der pathogeninduzierten Abwehrmechanismen der Pflanze Arabidopsis thaliana in den ersten 24 Stunden nach Infektion mit dem avirulenten Bakterium Pseudomonas syringae pv. tomato eingesetzt. In einer Auswahl von 75 Genen der Stoffwechselwege Glycolyse, Citrat-Cyclus, Pentosephosphat-Cyclus und Glyoxylatmetabolismus konnte für 25 % der Gene, im Shikimat-, Tryptophan- und Phenylpropanoidsyntheseweg für 60 % der Gene eine erhöhte Transkriptionsrate nachgewiesen werden. Die Ergebnisse dieser Arbeit stimmen mit experimentellen Daten verschiedener unabhängiger Studien zur pflanzlichen Pathogenantwort überein. Darüberhinaus sind erstmals Transkriptionsprofile von bisher auf Transkriptionsebene nicht untersuchten Genen erstellt worden. Diese Ergebnisse bestätigen die transkriptionelle Aktivierung ganzer Stoffwechselwege und gewähren erstmals einen Einblick in die koordinierte differentielle Transkription ganzer Stoffwechselwege während der Pathogenabwehr.

Le Dye – Sensitized Solar Cells (DSSC) sono attualmente considerate tra le alternative più promettenti al fotovoltaico tradizionale. I ridotti costi di produzione e l’elevata versatilità di utilizzo rappresentano i punti di forza di questi dispositivi innovativi. Ad oggi la ricerca è concentrata prevalentemente sull’incremento delle prestazioni delle DSSC, ottenibile solamente attraverso un miglioramento delle funzioni dei singoli componenti e dell’interazione sinergica tra questi. Tra i componenti, ha recentemente assunto particolare interesse il blocking layer (BL), costituito generalmente da un film sottile di TiO2 depositato sulla superficie dell’anodo (FTO) e in grado di ottimizzare i fenomeni all’interfaccia FTO/TiO2/elettrolita. Nel corso di questo lavoro di tesi si è rivolta l’attenzione prevalentemente sulle caratteristiche del BLs (ad esempio proprietà morfologico – strutturali) cercando di mettere in correlazione il processo di deposizione con le caratteristiche finali del film ottenuto. A questo scopo è stato ottimizzato un processo di deposizione dei film via spin coating, a partire da soluzioni acquosa o alcolica di precursore (TiCl4). I film ottenuti sono stati confrontati con quelli depositati tramite un processo di dip coating riportato in letteratura. I BLs sono stati quindi caratterizzati tramite microscopia (SEM – AFM), spettrofotometria (UV.- Vis) e misure elettrochimiche (CV – EIS). I risultati ottenuti hanno messo in evidenza come i rivestimenti ottenuti da soluzione acquosa di precursore, indipendentemente dalla tecnica di deposizione utilizzata (spin coating o dip coating) diano origine a film disomogenei e scarsamente riproducibili, pertanto non idonei per l’applicazione nelle DSSC. Viceversa, i BLs ottenuti via spin coating dalla soluzione alcolica di TiCl4 sono risultati riproducibili, omogenei, e uniformemente distribuiti sulla superficie di FTO. Infine, l’analisi EIS ha in particolare evidenziato un effettivo aumento della resistenza al trasferimento di carica tra elettrodo FTO ed elettrolita in presenza di questi BLs, fenomeno generalmente associato ad un efficace blocking effect.

My research PhD work is focused on the Electrochemically Generated Luminescence (ECL) investigation of several different homogeneous and heterogeneous systems. ECL is a redox induced emission, a process whereby species, generated at electrodes, undergo a high-energy electron transfer reaction to form excited states that emit light. Since its first application, the ECL technique has become a very powerful analytical tool and has widely been used in biosensor transduction. ECL presents an intrinsically low noise and high sensitivity; moreover, the electrochemical generation of the excited state prevents scattering of the light source: for all these characteristics, it is an elective technique for ultrasensitive immunoassay detection. The majority of ECL systems involve species in solution where the emission occurs in the diffusion layer near to the electrode surface. However, over the past few years, an intense research has been focused on the ECL generated from species constrained on the electrode surface. The aim of my work is to study the behavior of ECL-generating molecular systems upon the progressive increase of their spatial constraints, that is, passing from isolated species in solution, to fluorophores embedded within a polymeric film and, finally, to patterned surfaces bearing “one-dimensional” emitting spots. In order to describe these trends, I use different “dimensions” to indicate the different classes of compounds. My thesis was mostly developed in the electrochemistry group of Bologna with the supervision of Prof Francesco Paolucci and Dr Massimo Marcaccio. With their help and also thanks to their long experience in the molecular and supramolecular ECL fields and in the surface investigations using scanning probe microscopy techniques, I was able to obtain the results herein described. Moreover, during my research work, I have established a new collaboration with the group of Nanobiotechnology of Prof. Robert Forster (Dublin City University) where I spent a research period. Prof. Forster has a broad experience in the biomedical field, especially he focuses his research on film surfaces biosensor based on the ECL transduction. This thesis can be divided into three sections described as follows: (i) in the fist section, homogeneous molecular and supramolecular ECL-active systems, either organic or inorganic species (i.e., corannulene, dendrimers and iridium metal complex), are described. Driving force for this kind of studies includes the search for new luminophores that display on one hand higher ECL efficiencies and on the other simple mechanisms for modulating intensity and energy of their emission in view of their effective use in bioconjugation applications. (ii) in the second section, the investigation of some heterogeneous ECL systems is reported. Redox polymers comprising inorganic luminophores were described. In such a context, a new conducting platform, based on carbon nanotubes, was developed aimed to accomplish both the binding of a biological molecule and its electronic wiring to the electrode. This is an essential step for the ECL application in the field of biosensors. (iii) in the third section, different patterns were produced on the electrode surface using a Scanning Electrochemical Microscopy. I developed a new methods for locally functionalizing an inert surface and reacting this surface with a luminescent probe. In this way, I successfully obtained a locally ECL active platform for multi-array application.

In this thesis I present theoretical and experimental results concern- ing the operation and properties of a new kind of Penning trap, the planar trap. It consists of circular electrodes printed on an isolating surface, with an homogeneous magnetic field pointing perpendicular to that surface. The motivation of such geometry is to be found in the construction of an array of planar traps for quantum informa- tional purposes. The open access to radiation of this geometry, and the long coherence times expected for Penning traps, make the planar trap a good candidate for quantum computation. Several proposals for quantum 2-qubit interactions are studied and estimates for their rates are given. An expression for the electrostatic potential is presented, and its fea- tures exposed. A detailed study of the anharmonicity of the potential is given theoretically and is later demonstrated by experiment and numerical simulations, showing good agreement. Size scalability of this trap has been studied by replacing the original planar trap by a trap twice smaller in the experimental setup. This substitution shows no scale effect apart from those expected for the scaling of the parameters of the trap. A smaller lifetime for trapped electrons is seen for this smaller trap, but is clearly matched to a bigger misalignment of the trap’s surface and the magnetic field, due to its more difficult hand manipulation. I also give a hint that this trap may be of help in studying non-linear dynamics for a sextupolarly perturbed Penning trap.

A novel design based on electric field-free open microwell arrays for the automated continuous-flow sorting of single or small clusters of cells is presented. The main feature of the proposed device is the parallel analysis of cell-cell and cell-particle interactions in each microwell of the array. High throughput sample recovery with a fast and separate transfer from the microsites to standard microtiter plates is also possible thanks to the flexible printed circuit board technology which permits to produce cost effective large area arrays featuring geometries compatible with laboratory equipment. The particle isolation is performed via negative dielectrophoretic forces which convey the particles’ into the microwells. Particles such as cells and beads flow in electrically active microchannels on whose substrate the electrodes are patterned. The introduction of particles within the microwells is automatically performed by generating the required feedback signal by a microscope-based optical counting and detection routine. In order to isolate a controlled number of particles we created two particular configurations of the electric field within the structure. The first one permits their isolation whereas the second one creates a net force which repels the particles from the microwell entrance. To increase the parallelism at which the cell-isolation function is implemented, a new technique based on coplanar electrodes to detect particle presence was implemented. A lock-in amplifying scheme was used to monitor the impedance of the channel perturbed by flowing particles in high-conductivity suspension mediums. The impedance measurement module was also combined with the dielectrophoretic focusing stage situated upstream of the measurement stage, to limit the measured signal amplitude dispersion due to the particles position variation within the microchannel. In conclusion, the designed system complies with the initial specifications making it suitable for cellomics and biotechnology applications.

Life Cycle Assessment (LCA) is a chain-oriented tool to evaluate the environment performance of products focussing on the entire life cycle of these products: from the extraction of resources, via manufacturing and use, to the final processing of the disposed products. Through all these stages consumption of resources and pollutant releases to air, water, soil are identified and quantified in Life Cycle Inventory (LCI) analysis. Subsequently to the LCI phase follows the Life Cycle Impact Assessment (LCIA) phase; that has the purpose to convert resource consumptions and pollutant releases in environmental impacts. The LCIA aims to model and to evaluate environmental issues, called impact categories. Several reports emphasises the importance of LCA in the field of ENMs. The ENMs offer enormous potential for the development of new products and application. There are however unanswered questions about the impacts of ENMs on human health and the environment. In the last decade the increasing production, use and consumption of nanoproducts, with a consequent release into the environment, has accentuated the obligation to ensure that potential risks are adequately understood to protect both human health and environment. Due to its holistic and comprehensive assessment, LCA is an essential tool evaluate, understand and manage the environmental and health effects of nanotechnology. The evaluation of health and environmental impacts of nanotechnologies, throughout the whole of their life-cycle by using LCA methodology. This is due to the lack of knowledge in relation to risk assessment. In fact, to date, the knowledge on human and environmental exposure to nanomaterials, such ENPs is limited. This bottleneck is reflected into LCA where characterisation models and consequently characterisation factors for ENPs are missed. The PhD project aims to assess limitations and challenges of the freshwater aquatic ecotoxicity potential evaluation in LCIA phase for ENPs and in particular nanoparticles as n-TiO2.

Al giorno d’oggi, la produzione di energia e di molecole di base per l’industria chimica è completamente dipendente da risorse non rinnovabili come petrolio, carbone e gas naturale ; con tali risorse in via di esaurimento e la sempre crescente domanda di energia e materiali da parte di tutte le economie, risulta obbligatorio sviluppare tecniche per la loro produzione da risorse rinnovabili. Le biomasse, oltre ad essere una fonte rinnovabile, generano minori emissioni di gas serra rispetto ai combustibili fossili, perché la CO2 rilasciata nei processi di utilizzo viene bilanciata da quella consumata nel processo di crescita delle biomasse stesse. Tuttavia, ad oggi, lo sfruttamento di queste fonti risulta ancora sfavorito economicamente a causa di processi industriali non ancora ottimizzati, i cui costi si ripercuotono sul prodotto finale. Le molecole derivanti dagli scarti lignocellulosici possono essere usate come molecole di partenza per industrie chimiche di qualsiasi tipo, da quelle farmaceutiche a quelle plastiche. Queste molecole sono già parzialmente funzionalizzate; ciò significa che la sintesi di prodotti chimici specifici richiede un minor numero di stadi rispetto ai building blocks petroliferi, con conseguente diminuzione di prodotti di scarto e sottoprodotti . Una delle molecole “piattaforma” identificate tra le più importanti per la produzione di diversi prodotti chimici, risulta essere il 5-idrossimetilfurfurale (HMF) , derivante dalla disidratazione di polisaccaridi esosi, da cui si può ottenere tramite ossidazione selettiva, l’acido 2,5-furandicarbossilico (FDCA), potenziale sostituto dell’acido tereftalico nella produzione del PET e molti altri prodotti. Lo scopo di questo lavoro di tesi è stato lo studio della reattività di catalizzatori a base di Pd e Au/Pd utilizzati nella reazione di ossidazione dell’HMF a FDCA. Il lavoro svolto ha avuto come obiettivi principali: • L’ottimizzazione della sintesi di nanoparticelle di Pd e Au/Pd a diverso rapporto molare, e la caratterizzazione delle sospensioni ottenute mediante analisi DLS, XRD e TEM. • La preparazione di catalizzatori supportati su TiO2 e la caratterizzazione dei catalizzatori ottenuti tramite analisi BET, TEM e analisi termiche TGA/DSC. • Lo studio dell’attività catalitica dei catalizzatori preparati nell’ossidazione selettiva in fase liquida del 5-idrossimetilfurfurale (HMF) ad acido 2,5-furandicarbossilico (FDCA) e del meccanismo di reazione.

Questa tesi descrive lo sviluppo di un elettrodo modificato con un polimero isolante per la determinazione indiretta del radicale OH. I polimeri testati sono stati polifenolo, polipirrolo e polipirrolo sovraoossidato ed il primo è risultato quello con le migliori prestazioni. Il film di modificante è stato depositato per elettropolimerizzazione del fenolo in ambiente acido, su un elettrodo di carbone vetroso (GC) ed è risultato isolante e perfettamente adeso al GC, impedendo il trasferimento di carica alle più comuni sonde redox. L’attacco dei radicali OH, generati dalla reazione di Fenton o dalla fotolisi di H2O2, rimuove parzialmente il polimero dal GC, ripristinando parzialmente il comportamento conduttore dell’elettrodo. L’entità della degradazione del film polifenolico è stata valutata sfruttando la corrente relativa alla sonda redox Ru(NH3)63+, che rappresenta il segnale analitico per la determinazione del radicale OH. L’elettrodo è stato impiegato per stimare le prestazioni di foto catalizzatori a base di nanoparticelle di TiO2, ottenendo risultati correlati a quelli ricavati da un metodo HPLC. Inoltre esso è stato usato per sviluppare una nuova procedura per la determinazione della capacità di scavenging verso i radicali OH, che è stata applicata all’analisi di composti puri e campioni reali. I risultati erano confrontabili con quelli determinati con metodiche standardizzate, comunemente impiegate per la determinazione della capacità antiossidante. Inoltre è stato condotto uno studio riguardante la modifica di un elettrodo di platino con un idrossido misto a strati a base di cobalto e alluminio (LDH). In particolare si sono valutati gli effetti di diversi pretrattamenti del Pt sulle caratteristiche e prestazioni elettrocatalitiche del film di LDH nei confronti dell’ossidazione di anilina, fenolo e acido salicilico. Questi composti possono essere impiegati come molecole sonda per la determinazione del radicale OH e rivestono interesse da un punto di vista elettroanalitico perché portano facilmente alla passivazione della superficie di Pt.

Upgrade of biomass to valuable chemicals is a central topic in modern research due to the high availability and low price of this feedstock. For the difficulties in biomass treatment, different pathways are still under investigation. A promising way is in the photodegradation, because it can lead to greener transformation processes with the use of solar light as a renewable resource. The aim of my work was the research of a photocatalyst for the hydrolysis of cellobiose under visible irradiation. Cellobiose was selected because it is a model molecule for biomass depolymerisation studies. Different titania crystalline structures were studied to find the most active phase. Furthermore, to enhance the absorption of this semiconductor in the visible range, noble metal nanoparticles were immobilized on titania. Gold and silver were chosen because they present a Surface Plasmon Resonance band and they are active metals in several photocatalytic reactions. The immobilized catalysts were synthesized following different methods to optimize the synthetic steps and to achieve better performances. For the same purpose the alloying effect between gold and silver nanoparticles was examined.

As the elastic response of cell membranes to mechanical stimuli plays a key role in various cellular processes, novel biophysical strategies to quantify the elasticity of native membranes under physiological conditions at a nanometer scale are gaining interest. In order to investigate the elastic response of apical membranes, elasticity maps of native membrane sheets, isolated from MDCK II (Madine Darby Canine kidney strain II) epithelial cells, were recorded by local indentation with an Atomic Force Microscope (AFM). To exclude the underlying substrate effect on membrane indentation, a highly ordered gold coated porous array with a pore diameter of 1.2 μm was used to support apical membranes. Overlays of fluorescence and AFM images show that intact apical membrane sheets are attached to poly-D-lysine coated porous substrate. Force indentation measurements reveal an extremely soft elastic membrane response if it is indented at the center of the pore in comparison to a hard repulsion on the adjacent rim used to define the exact contact point. A linear dependency of force versus indentation (-dF/dh) up to 100 nm penetration depth enabled us to define an apparent membrane spring constant (kapp) as the slope of a linear fit with a stiffness value of for native apical membrane in PBS. A correlation between fluorescence intensity and kapp is also reported. Time dependent hysteresis observed with native membranes is explained by a viscoelastic solid model of a spring connected to a Kelvin-Voight solid with a time constant of 0.04 s. No hysteresis was reported with chemically fixated membranes. A combined linear and non linear elastic response is suggested to relate the experimental data of force indentation curves to the elastic modulus and the membrane thickness. Membrane bending is the dominant contributor to linear elastic indentation at low loads, whereas stretching is the dominant contributor for non linear elastic response at higher loads. The membrane elastic response was controlled either by stiffening with chemical fixatives or by softening with F-actin disrupters. Overall, the presented setup is ideally suitable to study the interactions of the apical membrane with the underlying cytoskeleton by means of force indentation elasticity maps combined with fluorescence imaging.

Nel primo capitolo viene introdotto lo studio eff�ettuato e descritto un metodo di misure successivo alla caratterizzazione della super�ficie. Nel secondo capitolo vengono descritti i campioni analizzati e, nello speci�fico, la crescita attraverso MaCE dei nanofi�li di silicio. Nel terzo capitolo viene descritto lo strumento AFM utilizzato e la teoria della caratterizzazione alla base dello studio condotto. Nella quarta sezione vengono descritti i risultati ottenuti mentre nelle conclusioni viene tratto il risultato dei valori ottenuti di RMS roughness e roughness exponent.

The thesis analyses the hydrodynamic induced by an array of Wave energy Converters (WECs), under an experimental and numerical point of view. WECs can be considered an innovative solution able to contribute to the green energy supply and –at the same time– to protect the rear coastal area under marine spatial planning considerations. This research activity essentially rises due to this combined concept. The WEC under exam is a floating device belonging to the Wave Activated Bodies (WAB) class. Experimental data were performed at Aalborg University in different scales and layouts, and the performance of the models was analysed under a variety of irregular wave attacks. The numerical simulations performed with the codes MIKE 21 BW and ANSYS-AQWA. Experimental results were also used to calibrate the numerical parameters and/or to directly been compared to numerical results, in order to extend the experimental database. Results of the research activity are summarized in terms of device performance and guidelines for a future wave farm installation. The device length should be “tuned” based on the local climate conditions. The wave transmission behind the devices is pretty high, suggesting that the tested layout should be considered as a module of a wave farm installation. Indications on the minimum inter-distance among the devices are provided. Furthermore, a CALM mooring system leads to lower wave transmission and also larger power production than a spread mooring. The two numerical codes have different potentialities. The hydrodynamics around single and multiple devices is obtained with MIKE 21 BW, while wave loads and motions for a single moored device are derived from ANSYS-AQWA. Combining the experimental and numerical it is suggested –for both coastal protection and energy production– to adopt a staggered layout, which will maximise the devices density and minimize the marine space required for the installation.

Sensors are devices that have shown widespread use, from the detection of gas molecules to the tracking of chemical signals in biological cells. Single walled carbon nanotube (SWCNT) and graphene based electrodes have demonstrated to be an excellent material for the development of electrochemical biosensors as they display remarkable electronic properties and the ability to act as individual nanoelectrodes, display an excellent low-dimensional charge carrier transport, and promote surface electrocatalysis. The present work aims at the preparation and investigation of electrochemically modified SWCNT and graphene-based electrodes for applications in the field of biosensors. We initially studied SWCNT films and focused on their topography and surface composition, electrical and optical properties. Parallel to SWCNTs, graphene films were investigated. Higher resistance values were obtained in comparison with nanotubes films. The electrochemical surface modification of both electrodes was investigated following two routes (i) the electrografting of aryl diazonium salts, and (ii) the electrophylic addition of 1, 3-benzodithiolylium tetrafluoroborate (BDYT). Both the qualitative and quantitative characteristics of the modified electrode surfaces were studied such as the degree of functionalization and their surface composition. The combination of Raman, X-ray photoelectron spectroscopy, atomic force microscopy, electrochemistry and other techniques, has demonstrated that selected precursors could be covalently anchored to the nanotubes and graphene-based electrode surfaces through novel carbon-carbon formation.
