The objective of this study was to evaluate the methodology to establish the hemolytic activity of alternative complement pathway as an indicator of the innate immunity in Brazilian fish pacu (Piaractus mesopotamicus), in addition to verifying the influence of beta-glucan as an immunostimulant. Fish were fed with diets containing 0, 0.1 and 1% beta-glucan, during seven days, and then inoculated with Aeromonas hydrophila. Seven days after the challenge, they were bled for serum extraction. The methodology consisted of a kinetic assay that allows calculating the required time for serum proteins of the complement to promote 50% lysis of a rabbit red blood cell suspension. The method developed in mammals was successfully applied for pacu and determined that the hemolytic activity of the proteins of the complement system (alternative pathway) increased after the pathogen challenge, but was not influenced by the beta-glucan treatment.

Objective: The aim of this study was to investigate the effects of PRP on SAOS-2 cells in terms of cytokine expression, cell activity and oxidative stress. Design: Cell line SAOS-2 (1 x 10(5) cells/mL) were grown in culture medium alpha-MEM with 10% FBS for 24 h and stimulated (or not) with PRP at concentrations of 3, 10 and 20%, LPS (E. coli, 10 g/mL) and IL-1 beta (1 mg/mL) for 24 h. The supernatant was collected and analyzed for the expression of cytokines in a panel array, ALP using a commercial kit and NO2- with Griess reaction method. Also, the cells were analyzed using Western blot for RANKL and slot blotting for nitrotyrosine expression. Result: There were no significant differences amongst the groups in terms of NO2-, protein nitrotyrosine content and RANKL expression. However, all stimuli increased ALP activity and in case of PRP, it was in a dose-dependent manner (p < 0.001). Also, all stimuli induced an increase in cytokines and chemokines expression, but only PRP promoted an increase of component C5, sICAM-1 and RANTES expression. Whilst IL-1 receptor antagonist (IL-1ra) expression was down-regulated by PRP, both LPS and IL-1 beta caused up-regulation of this cytokine. Conclusions: PRP can stimulate osteoblast activity and cytokine/chemokine release, as well as indicate some of the mediators that can (and cannot) be involved in this activation. Background: Cellulose consisting of arrays of linear beta-1,4 linked glucans, is the most abundant carbon-containing polymer present in biomass. Recalcitrance of crystalline cellulose towards enzymatic degradation is widely reported and is the result of intra-and inter-molecular hydrogen bonds within and among the linear glucans. Cellobiohydrolases are enzymes that attack crystalline cellulose. Here we report on two forms of glycosyl hydrolase family 7 cellobiohydrolases common to all Aspergillii that attack Avicel, cotton cellulose and other forms of crystalline cellulose. Results: Cellobiohydrolases Cbh1 and CelD have similar catalytic domains but only Cbh1 contains a carbohydrate-binding domain (CBD) that binds to cellulose. Structural superpositioning of Cbh1 and CelD on the Talaromyces emersonii Cel7A 3-dimensional structure, identifies the typical tunnel-like catalytic active site while Cbh1 shows an additional loop that partially obstructs the substrate-fitting channel. CelD does not have a CBD and shows a four amino acid residue deletion on the tunnel-obstructing loop providing a continuous opening in the absence of a CBD. Cbh1 and CelD are catalytically functional and while specific activity against Avicel is 7.7 and 0.5 U. mg prot-1, respectively specific activity on pNPC is virtually identical. Cbh1 is slightly more stable to thermal inactivation compared to CelD and is much less sensitive to glucose inhibition suggesting that an open tunnel configuration, or absence of a CBD, alters the way the catalytic domain interacts with the substrate. Cbh1 and CelD enzyme mixtures on crystalline cellulosic substrates show a strong combinatorial effort response for mixtures where Cbh1 is present in 2: 1 or 4: 1 molar excess. When CelD was overrepresented the combinatorial effort could only be partially overcome. CelD appears to bind and hydrolyze only loose cellulosic chains while Cbh1 is capable of opening new cellulosic substrate molecules away from the cellulosic fiber. Conclusion: Cellobiohydrolases both with and without a CBD occur in most fungal genomes where both enzymes are secreted, and likely participate in cellulose degradation. The fact that only Cbh1 binds to the substrate and in combination with CelD exhibits strong synergy only when Cbh1 is present in excess, suggests that Cbh1 unties enough chains from cellulose fibers, thus enabling processive access of CelD.

Witzkeite, ideally Na4K4Ca(NO3)(2)(SO4)(4)center dot 2H(2)O, is a new mineral found in the oxidation zone of the guano mining field at Punta de Lobos, Tarapaca region, Chile. It occurs as colorless, tabular crystals up to 140 mu m in length, associated with dittmanite and nitratine. Witzkeite is colorless and transparent, with a white streak and a vitreous luster. It is brittle, with Molts hardness 2 and distinct cleavage on {001}. Measured density is 2.40(2) g/cm(3), calculated density is 2.403 g/cm(3). Witzkeite is biaxial (-) with refractive indexes alpha = 1.470(5), beta = 1.495(5), gamma = 1.510(5), measured 2V = 50-70 degrees. The empirical composition is (electron microprobe, mean of five analyses, H2O, CO2, and N2O5 by gas chromatography; wt%): Na2O 12.83, K2O 22.64, CaO 7.57, FeO 0.44, SO3 39.96, N2O5 12.7, H2O 4.5, total 100.64; CO2 was not detected. The chemical formula, calculated based on 24 O, is: Na3.40K3.95Ca1.11Fe0.05(NO3)(1.93)(SO4)(4.10)(H4.10O1.81). Witzkeite is monoclinic, space group C2/c, with unit-cell parameters: a = 24.902(2), b = 5.3323(4), c = 17.246(1) angstrom, beta = 94.281(7)degrees, V = 2283.6(3) angstrom(3) (Z = 4). The crystal structure was solved using single-crystal X-ray diffraction data and refined to R-1(F) = 0.043. Witzkeite belongs to a new structure type and is noteworthy for the very rare simultaneous presence of sulfate and nitrate groups. The eight strongest X-ray powder-diffraction lines [d in angstrom (I in %) (h k l)] are: 12.38 (100) (2 0 0), 4.13 (19) (6 0 0), 3.10 (24) (8 0 0), 2.99 (7) ((8) over bar 02), 2.85 (6) (8 02), 2.69 (9) ((7) over bar 1 3), 2.48 (12) (10 0 0), and 2.07 (54) (12 0 0). The IR spectrum of witzkeite was collected in the range 390-4000 cm(-1). The spectrum shows the typical bands of SO42- ions (1192, 1154, 1116, 1101, 1084, 993, 634, and 617 cm(-1)) and of NO3- ions (1385, 1354, 830, 716, and 2775 cm(-1)). Moreover, a complex pattern of bands related to H2O is visible (bands at 3565, 3419, 3260, 2405, 2110, 1638, and 499 cm(-1)). The IR spectrum is discussed in detail.

Temporal lobe epilepsy (TLE) is the most common form of partial epilepsy and affects 40% of the patients. Seizures arising from the mesial temporal lobe structures (i.e., amygdala and hippocampus) are common, whereas neocortical seizures are rare. In recent years, many studies aimed to identify the pattern of gene expression of neurotransmitters involved in molecular mechanisms of epilepsy. We used real-time PCR to quantify the expression of GABAA (subunits a1, beta 1, beta 2) and NMDA (subunits NR1, NR2A, and NR2B) receptor genes in amygdalae of 27 patients with TLE and 14 amygdalae from autopsy controls. The NR1 subunit was increased in patients with epilepsy when compared with controls. No differences were found in expression of NMDA subunits NR2A and NR2B or in a1, beta 1, and beta 2 subunits of GABAA receptors. Our results suggest that the NR1 subunit of NMDA receptors is involved in the amygdala hyperexcitability in some of the patients with TLE. (C) 2010 Wiley Periodicals, Inc., Inc.

Carlosbarbosaite, ideally (UO2)(2)Nb2O6(OH)(2)center dot 2H(2)O, is a new mineral which occurs as a late cavity filling in albite in the Jaguaracu pegmatite, Jaguaracu municipality, Minas Gerais, Brazil. The name honours Carlos do Prado Barbosa (1917-2003). Carlosbarbosaite forms long flattened lath-like crystals with a very simple orthorhombic morphology. The crystals are elongated along [001] and flattened on (100); they are up to 120 mu m long and 2-5 mu m thick. The colour is cream to pale yellow, the streak yellowish white and the lustre vitreous. The mineral is transparent (as individual crystals) to translucent (massive). It is not fluorescent under either long-wave or short-wave ultraviolet radiation. Carlosbarbosaite is biaxial(+) with alpha = 1.760(5), beta = 1.775(5), gamma = 1.795(5), 2V(meas) = 70(1)degrees, 2V(calc) = 83 degrees. The orientation is X parallel to a, Y parallel to b, Z parallel to c. Pleochroism is weak, in yellowish green shades, which are most intense in the Z direction. Two samples were analysed. For sample I, the composition is: UO3 54.52, CaO 2.07, Ce2O3 0.33, Nd2O3 0.49, Nb2O5 14.11, Ta2O5 15.25, TiO2 2.20, SiO2 2.14, Fe2O3 1.08, Al2O3 0.73, H2O (calc.) 11.49, total 104.41 wt.%; the empirical formula is (square 0.68Ca0.28Nd0.02Ce0.02)(Sigma=1.00)[U-1.44 square O-0.56(2.88)(H2O)(1.12)](Nb0.80Ta0.52Si0.27Ti0.21Al0.11Fe0.10)(Sigma=2.01) O-4.72(OH)(3.20)(H2O)(2.08). For sample 2, the composition is: UO3 41.83, CaO 2.10, Ce2O3 0.31, Nd2O3 1.12, Nb2O5 14.64, Ta2O5 16.34, TiO2 0.95, SiO2 3.55, Fe2O3 0.89, Al2O3 0.71, H2O (calc.) 14.99, total 97.43 wt.%; the empirical formula is (square 0.67Ca0.27Nd0.05Ce0.01)(Sigma=1.00)[U-1.04 square O-0.96(2.08)(H2O)(1.92)] (Nb0.79Ta0.53Si0.42Ti0.08Al0.10Fe0.08)(Sigma=2.00)O-4.00(OH)(3.96)(H2O)(2.04). The ideal endmember formula is (UO2)(2)Nb2O6(OH)(2)center dot 2H(2)O. Calculated densities are 4.713 g cm(-3) (sample 1) and 4.172 g cm(-3) (sample 2). Infrared spectra show that both (OH) and H2O are present. The strongest eight X-ray powder-diffraction lines [listed as d in angstrom(I)(hkl)] are: 8.405(8)(110), 7.081(10)(200), 4.201(9)(220), 3.333(6)(202), 3.053(8)(022), 2.931(7)(420), 2.803(6)(222) and 2.589(5)(040,402). The crystal structure was solved using single-crystal X-ray diffraction (R = 0.037) which gave the following data: orthorhombic, Cmem, a = 14.150(6), b = 10.395(4), c = 7.529(3) angstrom, V = 1107(1) angstrom(3), Z = 4. The crystal structure contains a single U site with an appreciable deficiency in electron scattering, which is populated by U atoms and vacancies. The U site is surrounded by seven 0 atoms in a pentagonal bipyramidal arrangemet. The Nb site is coordinated by four 0 atoms and two OH groups in an octahedral arrangement. The half-occupied tunnel Ca site is coordinated by four 0 atoms and four H2O groups. Octahedrally coordinated Nb polyhedra share edges and comers to form Nb2O6(OH)(2) double chains, and edge-sharing pentagonal bipyramidal U polyhedra form UO5 chains. The Nb2O6(OH)(2) and UO5 chains share edges to form an open U-Nb-phi framework with tunnels along [001] that contain Ca(H2O)(4) clusters. Carlosbarbosaite is closely related to a family of synthetic U-Nb-O framework tunnel structures, it differs in that is has an (OH)-bearing framework and Ca(H2O)(4) tunnel occupant. The structure of carlosbarbosaite resembles that of holfertite.

Peptides derived from cytosolic, mitochondrial, and nuclear proteins have been detected in extracts of animal tissues and cell lines. To test whether the proteasome is involved in their formation, HEK293T cells were treated with epoxomicin (0.2 or 2 mu M) for 1 h and quantitative peptidomics analysis was performed. Altogether, 147 unique peptides were identified by mass spectrometry sequence analysis. Epoxomicin treatment decreased the levels of the majority of intracellular peptides, consistent with inhibition of the proteasome beta-2 and beta-5 subunits. Treatment with the higher concentration of epoxomicin elevated the levels of some peptides. Most of the elevated peptides resulted from cleavages at acidic residues, suggesting that epoxomicin increased the processing of proteins through the beta-1 subunit. Interestingly, some of the peptides that were elevated by the epoxomicin treatment had hydrophobic residues in P1 cleavage sites. Taken together, these findings suggest that, while the proteasome is the major source of intracellular peptides, other peptide-generating mechanisms exist. Because intracellular peptides are likely to perform intracellular functions, studies using proteasome inhibitors need to be interpreted with caution, as it is possible that the effects of these inhibitors are due to a change in the peptide levels rather than inhibition of protein degradation.

Selection of reference genes is an essential consideration to increase the precision and quality of relative expression analysis by the quantitative RT-PCR method. The stability of eight expressed sequence tags was evaluated to define potential reference genes to study the differential expression of common bean target genes under biotic (incompatible interaction between common bean and fungus Colletotrichum lindemuthianum) and abiotic (drought; salinity; cold temperature) stresses. The efficiency of amplification curves and quantification cycle (C (q)) were determined using LinRegPCR software. The stability of the candidate reference genes was obtained using geNorm and NormFinder software, whereas the normalization of differential expression of target genes [beta-1,3-glucanase 1 (BG1) gene for biotic stress and dehydration responsive element binding (DREB) gene for abiotic stress] was defined by REST software. High stability was obtained for insulin degrading enzyme (IDE), actin-11 (Act11), unknown 1 (Ukn1) and unknown 2 (Ukn2) genes during biotic stress, and for SKP1/ASK-interacting protein 16 (Skip16), Act11, Tubulin beta-8 (beta-Tub8) and Unk1 genes under abiotic stresses. However, IDE and Act11 were indicated as the best combination of reference genes for biotic stress analysis, whereas the Skip16 and Act11 genes were the best combination to study abiotic stress. These genes should be useful in the normalization of gene expression by RT-PCR analysis in common bean, the most important edible legume.

Adipose-derived mesenchymal stem cells (ADMSCs) display immunosuppressive properties, suggesting a promising therapeutic application in several autoimmune diseases, but their role in type 1 diabetes (T1D) remains largely unexplored. The aim of this study was to investigate the immune regulatory properties of allogeneic ADMSC therapy in T cell-mediated autoimmune diabetes in NOD mice. ADMSC treatment reversed the hyperglycemia of early-onset diabetes in 78% of diabetic NOD mice, and this effect was associated with higher serum insulin, amylin, and glucagon-like peptide 1 levels compared with untreated controls. This improved outcome was associated with downregulation of the CD4(+) Th1-biased immune response and expansion of regulatory T cells (Tregs) in the pancreatic lymph nodes. Within the pancreas, inflammatory cell infiltration and interferon-gamma levels were reduced, while insulin, pancreatic duodenal homeobox-1, and active transforming growth factor-beta 1 expression were increased. In vitro, ADMSCs induced the expansion/proliferation of Tregs in a cell contact-dependent manner mediated by programmed death ligand 1. In summary, ADMSC therapy efficiently ameliorates autoimmune diabetes pathogenesis in diabetic NOD mice by attenuating the Th1 immune response concomitant with the expansion/proliferation of Tregs, thereby contributing to the maintenance of functional beta-cells. We discuss the gravitational collapse of a spherically symmetric massive core of a star in which the fluid component is interacting with a growing vacuum energy density. The influence of the variable vacuum in the collapsing core is quantified by a phenomenological beta parameter as predicted by dimensional arguments and the renormalization group approach. For all reasonable values of this free parameter, we find that the vacuum energy density increases the collapsing time, but it cannot prevent the formation of a singular point. However, the nature of the singularity depends on the value of beta. In the radiation case, a trapped surface is formed for beta <= 1/2, whereas for beta >= 1/2, a naked singularity is developed. In general, the critical value is beta = 1-2/3(1 + omega) where omega is the parameter describing the equation of state of the fluid component.

The progression of carcinogenesis entails the detachment of cells, invasion and migration of neoplastic cells. Alterations in epithelial adhesion and basement membrane proteins might mediate the early stages of carcinogenesis. This study investigated the expression of adhesion molecules and the basement membrane protein laminin-5 in actinic cheilitis (AC) and incipient squamous cell carcinoma of the lower lip to understand early photocarcinogenesis. Ln-5 gamma 2 chain as well as alpha 3, beta 1 subunits of alpha 3 beta 1 heterodimer and beta 4 subunit of integrin alpha 6 beta 4 were evaluated by immunohistochemistry in 16 cases of AC and 16 cases of superficially invasive squamous cell carcinoma (SISCC). Most AC cases showed reduced expression of beta 1, beta 4 and alpha 3 integrins, and SISCCs lacked beta 1, beta 4 and alpha 3 integrins in the invasive front. AC cases were negative for the Ln-5 gamma 2 chain. Five cases of SISCC (31%) showed heterogeneous Ln-5 gamma 2 chain expression in the invasive front of the tumor. Integrin beta 1, beta 4 and alpha 3 expression is lost during the early stages of lip carcinogenesis. Expression of Ln-5 gamma 2 in the invasive front in cases and its correlation with tumor progression suggest that it mediates the acquisition of the migrating and invading epithelial cell phenotype. Breast carcinoma, one of the most frequent malignancies in women, is a complex disease in which a number of different factors combine to drive pathogenesis. The biopathological characterization of these tumors is essential to determine their aggressiveness and to find the most appropriate therapy. As in others neoplasms, the deregulation of signal transduction pathways is frequently responsible for conferring selective biological advantages to the tumor. Phosphoinositides play an essential role in diverse cellular functions, their metabolism is highly active, and is tightly controlled. Among the enzymes implicated in this pathway, phospholipase C beta 1 (PLCβ1) is one of the key regulators, both at the cytoplasmic and the nuclear level. The PLCβ1 gene maps onto the short arm of chromosome 20, a region that has been shown to be altered in several solid tumors, including breast cancer. In the present study a FISH approach was used to investigate the genetic alterations of the PLCβ1 gene in various classes of breast cancer which differ in their invasiveness and proliferation status, according to their mitotic index. The overall aim was to find out whether this enzyme could be a suitable prognostic marker for this neoplasm. Our results show that 83% of cases had aneusomies at the 20p12 level, and the most frequent alteration is a gain in this specific locus. Indeed, we found that this amplification is not related to the invasion status since there were no differences in amplified tumor frequencies between in situ and invasive breast cancer. On the contrary, the gain of PLCβ1 was significantly related to the mitotic index (p = 0.001). To verify if the change in genetic dosage influences the expression of PLCβ1 we performed Real Time PCR and Immunohystochemical analysis. Our results confirmed that amplified tumors have higher levels of PLCβ1 mRNA, which is the sum of the two splicing isoforms 1a and 1b. On the other hand, even if protein levels were higher in the majority of cases compared to the nontumoral specimens, there were no significant associations between gain and overexpression. Finally, the significant association between the amplification of PLCβ1 and others important clinicopathological parameters, such as grading and hormonal receptors status, confirmed a correlation of this enzyme with the aggressiveness of breast cancer. This suggests that PLCβ1 has the potential to be a prognostic marker in these tumors. However, further work needs to be carried out to validate these preliminary findings.

Generierung und Transduktion wachstumsnegativer Signale durch den Contactinhibin-RezeptorDie kontaktabhängige Wachstumshemmung, Kontaktinhibition, basierend auf der Interaktion von Contactinhibin (Ci) und seinem Rezeptor (CiR), reguliert das Zellwachstum. Ziel dieser Arbeit war die Aufklärung der Signalweiterleitung über den CiR.Der transformierende Wachstumsfaktor TGF-beta führt in den meisten Zelltypen wie die Kontaktinhibition zum Zellzyklusstopp in der G1-Phase. Um mögliche Interaktionen der beiden Signalkaskaden zu untersuchen, wurden humane Keratinozyten (HaCaT) mit einem dominant-negativen TGF-beta-Rezeptor Typ II stabil transfiziert. Das Ausschalten des TGF-beta-Signalweges resultierte in einer erhöhten Sättigungsdichte und Aufhebung der Kontaktinhibition. Die durch Kontaktinhibition induzierte Zunahme der Expression der TGF-beta-mRNA bestätigte die mögliche Interaktion der beiden Signalwege.In einem weiteren Ansatz sollte die Proteinkinase C (PKC) als möglicher Second Messenger der Kontaktinhibition untersucht werden. Die Herunterregulierung der PKC-Isoformen alpha, delta, epsilon und mu nach Langzeit-Behandlung mit 12-O-Tetradecanoylphorbol-13-acetat führte in humanen Fibroblasten (FH109) zu einer Reduktion der Kontaktinhibition. Nur durch Inkubation mit dem spezifischen Inhibitor der delta-Isoform Rottlerin konnte die Kontaktinhibition vollständig aufgehoben werden. Eine Beteiligung der PKC-delta in die Kontaktinhibition wurde dadurch bestätigt, daß sowohl die Stärke ihrer Proteinexpression als auch ihre intrazelluläre Verteilung über Zell-Zellkontakte reguliert wurde. Außerdem führte die transiente Transfektion von Maus-Fibroblasten, NIH3T3, mit einer dominant-negativen Mutante der PKC-delta zu einem transformierten Phänotyp.

TGF-beta ist ein Schlüsselmolekül zellvermittelter Immuntoleranz. So spielt es neben seiner pleiotropen Rolle in Immunzellen auch bei der Tumorentwicklung eine große Rolle. Das TGF-beta hat bei der Tumorentwicklung eine duale Rolle. So dient es in frühen Phasen als Tumorsuppressor, währenddessen es in späten Phasen der Entwicklung als Tumorpromotor wirkt. Eine strikte Regulation des TGF-beta Signalweges ist daher für ein funktionierendes Immunsystem von essentieller Bedeutung. Die Ubiquitin Ligase Smurf2 ist dabei ein wichtiger negativ Regulator des TGF-beta Signalweges.In der vorliegenden Arbeit konnte eine neue Spleißform des Smurf2 (dE2Smurf2) aus murinen CD4+ T-Zellen isoliert werden, deren Funktion in vitro und in vivo in T-Lymphozyten untersucht worden ist. Für diese Spleißform konnte zudem eine humane Relevanz nachgewiesen werden. Mit Hilfe von Überexpressionen in Cos7 Zellen konnte eine veränderte Lokalisation der Smurf2 Spleißformen (WT und dE2) festgestellt werden. Dabei konnten lysosomale und endosomale Kompartimente bei der Kolokalisation mit dem dE2Smurf2 Konstrukt beobachtet werden. Das Spleißen des Exons2 führte dabei zu Änderungen der Topologie der N-terminalen C2-Domäne, wodurch sich eine veränderte Lokalisation in der Zelle beschreiben ließ. Mit der veränderten intrazellulären Verteilung erfuhr auch die Funktion der dE2Smurf2 Ubiquitin Ligase eine Änderung. So konnte überraschenderweise eine positive Signalinduktion des TGF-beta Signalweges beobachtet werden, was im Gegensatz zum beschriebenen WTSmurf2 stand. Durch eine Überexpression des dE2Smurf2 Proteins in T-Lymphozyten wurde der TGF-beta Signalweg in CD4+ und CD8+ Zellen positiv reguliert, dabei wurde der TGFbetaRII vermehrt exprimiert und gleichzeitig fand eine verstärkte Phosphorylierung der Transkriptionsfaktoren Smad2 und Smad3 nach TGF-beta Stimulation statt. Die transgenen T-Lymphozyten waren somit sensitiver gegenüber TGF-beta. Dies führte zur Hypothese, die durch Western Blot Analyse bestätigt werden konnte, daß das dE2Smurf2 nach Überexpression seine WT-Form bindet und dadurch degradiert. Die Degradation der Ubiquitin Ligase war dabei Smad7 abhängig. Zur Analyse des Einflusses der Ubiquitin Ligase dE2Smurf2 auf die Differenzierung von CD4+ T-Zellen, sowie ihre Rolle bei der T-Zell Proliferation, konnte gezeigt werden, daß durch die höhere Sensitivität gegenüber TGF-beta naive T-Zellen unter Einfluß von TGF-beta und IL6 vermehrt in TH17 Zellen differenzierten. Zudem konnte gezeigt werden, daß die Proliferationsrate transgener naiver CD4+ T-Zellen bei geringen Mengen von TGF-beta starkt vermindert war. Weiterhin konnte gezeigt werden, daß bei einer Differenzierung der naiven CD4+ T-Zellen in TH1 Zellen, diese signifkant weniger das proinflammatorische Zytokin INFγ produzierten.So zeigten in vivo Versuche, daß die transgenen Tiere in der Entwicklung von Kolorektalen Karzinomen protektiert waren. Sowohl im kolitisassiziierten Tumor Modell als auch bei der spontanen Entwicklung von Tumoren im APCmin Modell. Dies konnte zum einen auf eine deutlich verminderte Entzündung (geringere Produktion an Zytokinen durch verminderte Proliferation) des Darms und zum anderen durch eine stärkere Produktion an zytotoxischen Genen, wie Perforin, INFγ und Granzym B erklärt werden. Interessanterweise konnte jedoch im Transfer Kolitis Modell eher eine proinflammatorische Wirkung des dE2Smurf2 Proteins nachgewiesen werden. So wiesen die immundefizienten Mäuse, in denen die transgenen T-Zellen injiziert wurden, eine signifikant stärkere Kolitis auf als die Kontrollen. Dies konnte mit einer Überproduktion an IL17 sezernierenden T-Zellen erklärt werden. Klonierungsexperimente führten zudem zur Identifikation einer bisher nicht beschriebenen nicht kodierenden RNA. Diese zeigte in Kombination mit dem dE2Smurf2 Protein in einer Reportergen Analyse eine Hyperaktivierung des Smad3 Promotors. Diese Daten liefern zum einen ein genaueres Modell über die Regulation des TGF-beta Signalweges sowie wichtige Erkenntnisse zur Pathophysiologie chronisch entzündlicher Darmerkrankung und daraus resultierende Tumorerkrankungen. So entwickelt sich das dE2Smurf2, Teil des TGF-beta Signalweges, als attraktives Zielprotein für die Modulation von chronisch entzündlichen Darmerkrankungen und (kolitisassoziierte) Kolonkarzinomen.

In dieser Arbeit wird eine Einteilung der degenerativen strukturellen Veränderungen der Synovialmembran vorgestellt. Anhand der Kriterien Fibrosierung des Stromas, Rückgang des Gefäßnetzes, Auftreten von Hyalinose und chondroider Metaplasie mit und ohne Nachweis von CPPD Kristallen wurden Präparate der Synovialmembran von 59 Patienten mit Nachweis degenerativer strukturellen Veränderungen in 4 Stadien eingeteilt. rnHyalinose (Stadium 3) konnte in den untersuchten Schnitten nur relativ selten beobachtet werden, so dass am ehesten von einem Vorstadium zur chondroiden Metaplasie auszugehen ist. rnDie Verteilung der Erkrankungsdauer und des Alters in den verschiedenen Stadien lassen darauf schließen, dass höhere Stadien mit höherem Alter und längerer Erkrankungsdauer korrelieren. rnAus der vorhandenen Literatur ergeben sich Hinweise, welche Faktoren zu der Entstehung der strukturellen Veränderungen beitragen können: rnAus dem Netzwerk der Zytokine scheinen TGF-beta und die BMP’s an der Zunahme der Fibrose und an der Entstehung chondroider Metaplasie beteiligt zu sein. Makrophagen scheinen dabei eine wichtige Rolle zu spielen. Dies weist darauf hin, dass entzündliche und strukturelle Veränderungen miteinander vernetzt sind. rnBei der Entstehung der chondroiden Metaplasie kommen zusätzlich mechanische Einflüsse in Form von zyklischen Kompressionen als Einflussfaktor in Frage. rnDie Regulierung der Angiogenese ist noch zu wenig verstanden, um den Gefäßrückgang bei fortgeschrittenen strukturellen Veränderungen zu erklären. Erklärungsansätze sind zum einen zunehmende mechanische Schädigung bei zunehmender Inkongruenz der Gelenkflächen. Zum anderen könnte eine beginnende chondroide Metaplasie mit Expression von Chondromodulin I eine entscheidende Rolle spielen. rnInsgesamt muss man davon ausgehen, dass die zunehmenden strukturellen Veränderungen die Ernährung des Knorpels erschweren. Dabei ist an erster Stelle der Rückgang des Gefäßnetzes zu nennen. Dies erschwert nicht nur die Versorgung mit Nährstoffen, sondern auch den Abtransport von Stoffwechselprodukten. Ab einem gewissen Punkt ist aber auch davon auszugehen, dass die Funktion der Deckzellschicht beeinträchtigt wird. Wenn die Konzentration der Hyaluronsäure in der Synovia dadurch sinkt, kann dies durch eine vermehrte Permeabilität der Synovialmembran zum verstärkten Ausstrom von Wasser aus der Gelenkhöhle führen. Durch ein Ödem des umliegenden Gewebes kann dadurch der Blutfluss im Bereich des Gelenks zusätzlich vermindert werden. rnAuch die zunehmende Fibrosierung der Synovialmembran kann einen Einfluss auf die Permeabilität der Synovialmembran haben. Ob und in welchen Stadien der Veränderungen das einen relevanten Einfluss für die Ernährung der Chondrozyten hat, ist noch unklar.rn
