Ilex paraguariensis A. St. -Hil. is a native species of southern South America. The caffeine content in Ilex paraguariensis leaves has been considered an important quality parameter for maté-derived products. In this work different extraction methods for the determination of these methylxantines are compared. The influence of the extraction conditions on the methylxanthine yields was evaluated. Extraction by decoction with acidic aqueous solution (H2SO4 4N) presented the higher efficiency in the theobromine extraction. The extraction in a Soxhlet with acidic aqueous solution and decoction with acidic aqueous solution showed the highest caffeine yield. For the concomitant theobromine and caffeine quantification, the decoction with acidic aqueous solution is suggested.

Water quality was monitored at the upper course of the Rio das Velhas, a major tributary of the São Francisco basin located in the state of Minas Gerais, over an extension of 108 km from its source up to the limits with the Sabara district. Monitoring was done at 37 different sites over a period of 2 years (2003-2004) for 39 parameters. Multivariate statistical techniques were applied to interpret the large water-quality data set and to establish an optimal long-term monitoring network. Cluster analysis separated the sampling sites into groups of similarity, and also indicated the stations investigated for correlation and recommended to be removed from the monitoring network. Principal component analysis identified four components, which are responsible for the data structure explaining 80% of the total variance of the data. The principal parameters are characterized as due to mining activities and domestic sewage. Significant data reduction was achieved.

Oleanolic and ursolic acids are triterpenes that exist abundantly in the plant kingdom. They show antitumor activity and their cytotoxic activity was already evaluated against a broad range of cell lines. The inhibition of enzymes involved in the metabolism of DNA and the induction of apoptosis are known events that have been related to the antitumour activity of these triterpenes. The antiinflamatory and antioxidant activities also seem to play an important role in their antitumour activity. In this review, it is presented an overview of the importance of the potential antitumour, antiinflamatory and antioxidant activities of these triterpenoid derivatives. Also, we propose a structural-activity relationship to triterpenes containing oleanane or ursane skeleton and their cytotoxic activity.

In this study cellulose acetate butyrate (CAB) and carboxymehtylcellulose acetate butyrate (CMCAB) films adsorbed onto silicon wafers were characterized by means of ellipsometry, atomic force microscopy (AFM), sum frequency generation spectroscopy (SFG) and contact angle measurements. The adsorption behavior of lysozyme (LIS) or bovine serum albumin (BSA) onto CAB and CMCAB films was investigated. The amounts of adsorbed LIS or BSA onto CMCAB films were more pronounced than those onto CAB films due to the presence of carboxymethyl group in the CMCAB structure. Besides, the adsorption of BSA molecules on CMCAB films was more favored than that of LIS molecules. Antimicrobial effect of LIS bound to CAB or CMCAB layers was evaluated using Micrococcus luteus as substrate.

Croton zehntneri, a plant native to northeastern Brazil, is widely used in folk medicine to treat gastrointestinal problems and has rich essential oil content. The essential oil of C. Zehntneri was analyzed by GC-MS, and its inclusion complex with β-cyclodextrin (β-CD) was characterized by both vibrational spectroscopy and differential scanning calorimetry (DSC). Estragol was the major component identified in the essential oil by the study. IR spectra indicated an interaction of β-CD with essential oil from C. zehntneri, a finding corroborated by the stability constant and scanning calorimetry. Microencapsulation within β-CD has the potential to mask sensory attributes and increase aqueous solubility of oils, thereby improving their applicability as drugs.

AbstractThis paper presents a technological innovation that uses a subclass of glycerophospholipids as a booster biocide in antifouling paint. These glycerophospholipid PAF-analogs are economically and environmentally viable compounds because they are synthesized from a metal-free raw material source-soybean lecithin. The synthesis, which involves transesterification followed by an alkylation reaction, produced a mixture of glycerophospholipids that were characterized by mass spectrometry. Evaluation of the antifouling performance with field tests showed that the replacement of ordinary halogenated booster biocide with the synthesized product gave a better efficiency and an exceptional antifouling activity with a significant reduction in the coverage of the fouling macro-organisms.

Field experiments were conducted in the 1995-96 soybean (Glycine max) growing season to evaluate the effects of cultural practices and host genetic resistance on the intensity of soybean stem canker, caused by Diaporthe phaseolorum f.sp. meridionalis (Dpm). Experiments were conducted in a commercial field severely infected in the previous (1994-95) season. In one study, minimum tillage (MT) and no-tillage (NT) cropping systems were investigated for their effects on disease development and on plant yields in cvs. FT-Cristalina (susceptible) and FT-Seriema (moderately resistant). Another study evaluated the effects of plant densities (8, 15, 21 and 36 plants/m) on disease development in cvs. FT-Cristalina, FT-101 (moderately resistant) and FT-104 (resistant). Disease incidence and severity were consistently lower in NT than in MT, and plant yields were increased by 23% and 14% in the NT system for the susceptible and moderately resistant cultivars, respectively, compared to the yields in the MT system. The Gompertz and Logistic models described well the disease progress curves in all situations. For both susceptible and moderately resistant cultivars, disease severity increased proportionately to the increase in plant densities. At the end of the season, 100% of the plants of cv. FT-Cristalina were infected by Dpm, at all plant densities. Disease levels on cv. FT-101 were intermediate while only very low disease levels were recorded on cv. FT-104. There was a consistent negative correlation between stem canker severity and yield. Some practices demonstrated potential for direct application in disease control, and could be combined considering their additive effects.

Two soybean (Glycine max) cultivars were used in this study, Ocepar 4, rated as moderately resistant to Meloidogyne incognita race 3 but susceptible to M. javanica, and 'BR 16', susceptible to both nematodes. The effect of nematodes infection on the uptake and transport of N, P and Ca to the shoot was studied in plants growing in a split root system. The upper half was inoculated with 0, 3,000, 9,000 or 27,000 eggs/plant while the lower half received 15N, 32P or 45Ca. Infected plants showed an increase of root but a decrease of shoot mass with increasing inoculum levels. In general, total endogenous nutrients increased in the roots and tended to decrease in the shoots with increasing inoculum levels. When concentrations were calculated, there was an increase in the three nutrients in the roots, and an increase of Ca but no significant variation of N and P was observed in the shoots. The total amount of 15N in the roots increased at the highest inoculum levels but 32P and 45Ca decreased. In the shoots there was a reduction of 32P and 45Ca. The specific concentrations of the labelled nutrients (abundance or radioactivity/tissue mass) also showed a decrease of 32P and 45Ca in the shoots and roots of infected plants and an increase of 15N in the shoots. Considering that overall nutrient concentrations reflect cumulative nutrient uptake and the data from labelled elements gave information at a specific moment of the infection, thus nematodes do interfere with nutrient uptake and translocation.

The effect of Heterodera glycines on photosynthesis, leaf area and yield of soybean (Glycine max) was studied in two experiments carried out under greenhouse condition. Soybean seeds were sown in 1.5 l (Experiment 1) or 5.0 l (Experiment 2) clay pots filled with a mixture of field soil + sand (1:1) sterilized with methyl bromide. Eight days after sowing, seedlings were thinned to one per pot, and one day later inoculated with 0; 1.200; 3.600; 10.800; 32.400 or 97.200 J2 juveniles of H. glycines. Experiment 1 was carried out during the first 45 days of the inoculation while Experiment 2 was conducted during the whole cycle of the crop. Measurements of photosynthetic rate, stomatic conductance, chlorophyll fluorescence, leaf color, leaf area, and chlorophyll leaf content were taken at ten-day intervals throughout the experiments. Data on fresh root weight, top dry weight, grain yield, number of eggs/gram of roots, and nematode reproduction factor were obtained at the end of the trials. Each treatment was replicated ten times. There was a marked reduction in both photosynthetic rate and chlorophyll content, as well as an evident yellowing of the leaves of the infected plants. Even at the lowest Pi, the effects of H. glycines on the top dry weight or grain yield were quite severe. Despite the parasitism, soybean yield was highly correlated with the integrated leaf area and, accordingly, the use of this parameter was suggested for the design of potential damage prediction models that include physiological aspects of nematode-diseased plants.

Utilizando-se um experimento com microparcelas em condições de campo, a presente pesquisa teve por objetivo estudar os efeitos de duas diferentes doses do nematicida organofosforado sistêmico Terbufos (Counter 50G) sobre as populações dos fitonematóides ectoparasitos Helicotylenchus dihystera, Criconemella ornata e Paratrichodorus sp. em cinco variedades de cana-de-açúcar (Saccharum sp.), verificando-se, ao mesmo tempo, o potencial de reprodução desses nematóides nas mesmas variedades. O produto foi aplicado nas proporções de 60 e 80 kg do produto comercial por hectare (p.c./ha), no momento do plantio. As variedades estudadas foram SP70-1143, RB813804, SP78-4764, CB45-3 e SP79-1011. Com os dois tratamentos nematicidas, as parcelas testemunhas e as cinco variedades, formou-se um desenho experimental do tipo blocos ao acaso, em esquema fatorial, com cinco repetições. As avaliações fundamentaram-se nos níveis populacionais dos nematóides, com a determinação do fator de reprodução (FR) dos parasitos nas variedades, nas parcelas tratadas e não tratadas pelo nematicida. Pelos resultados obtidos concluiu-se que o produto nas duas dosagens empregadas não interferiu significativamente nos índices populacionais dos três nematóides 16 meses após o plantio, exceto para a combinação C. ornata x Terbufos 80 kg/ha, para a variedade SP70-1143. Não foi possível ser determinada a hospedabilidade das variedades estudadas em relação aos três nematóides, devido aos baixos fatores de reprodução (FR), provavelmente relacionados à longa estiagem ocorrente no ano do experimento.

Twenty isolates of four fungal species, agents of "Helminthosporium" diseases in cereals, were collected from different regions: nine Bipolarisoryzae isolated from rice (Oryza sativa), seven B.sorokiniana from wheat (Triticum aestivum), two B. maydis, and two Exserohilumturcicum from maize (Zea mays). The strains were compared by PCR-RFLP and RAPD analysis. Size polymorphism among the isolates in the ITS region comprising the 5.8 S rDNA indicated genetic differences among the isolates, while a UPGMA phenogram constructed after the digestion of this region with restriction enzymes showed inter- and intra-specific polymorphism. The RAPD profiles indicated an expressive level of polymorphism among different species, compared with a low level of polymorphism among isolates of the same species. A UPGMA phenogram grouped the isolates according to the species and their host plant. RAPD profiles did not reveal polymorphism that directly correlated climatic factors with geographic source of the isolates of B. sorokiniana, and B. oryzae. Teleomorphic species revealed high similarity with their correspondent anamorphs.

O efeito de diferentes temperaturas no solo e de diversas palhas sobre o solo na viabilidade dos escleródios de Sclerotinia sclerotiorum foi estudado em estufa. Em três ensaios de campo, estudou-se a solarização do solo associada à presença de palha de milho (Zea mays) sobre o solo na viabilidade de escleródios, durante três meses. Ensaios de campo foram realizados em Piracicaba e em Brasília. Escleródios foram produzidos em meio cenoura+fubá, e incorporados ao solo (ensaios em estufa), ou acondicionados em invólucros, e enterrados no solo a 5, 10 e 30 cm (ensaios no campo). Os tratamentos de solo no campo foram: solarizado (S), não solarizado (NS) e solarizado com adição de palha de milho (PS). Foram feitas avaliações a cada 30 dias, em meio NEON, observando a viabilidade e a presença de contaminantes nos escleródios. O aquecimento do solo em estufa a 50 e 60 ºC com diversas palhas inativou os escleródios, que tiveram maior incidência de contaminantes. No campo, o efeito da solarização do solo foi significativo, inviabilizando os escleródios enterrados a diferentes profundidades: em S após 90 dias, nas três profundidades, e em PS, após 60 dias, a 5 e 10 cm de profundidade. A incidência de escleródios contaminados em solos solarizados foi maior em S, seguido de PS. A maior variabilidade de contaminantes, porém, foi observada em PS. As temperaturas do solo em PS foram maiores quando comparadas aos outros tratamentos na mesma profundidade. Este fator proporcionou a redução do tempo da inativação dos escleródios de 90 dias em S, para 60 dias em PS.

Trichoderma stromaticum é um fungo antagonista a Crinipellis perniciosa, agente causal da vassoura-de-bruxa do cacaueiro (Theobroma cacao). Estromas periteciais de Hypocrea sp. formaram-se sobre a superfície de frutos mumificados e vassouras secas de cacaueiros em áreas onde T. stromaticum tinha sido pulverizado, experimentalmente, no controle biológico de C. perniciosa. Ascósporos obtidos dos estromas de Hypocrea sp. originaram colônias idênticas às de T. stromaticum. DNA genômico de colônias provenientes de ascósporos desta Hypocrea sp., de conídios de T. stromaticum e de Fusarium sp. (controle negativo) foi extraído e amplificado com oito "primers" decâmeros para obtenção de marcadores RAPD. Observaram-se similaridades genéticas de 0,82 a 0,96 entre os isolados ascospóricos e conidiais com base nos marcadores RAPD e de 0,06 a 0,09 entre estes e o isolado de Fusarium sp. O teleomorfo de T. stromaticum é denominado H. stromatica Bezerra, Costa & Bastos sp. nov.

A utilização de tolerância como forma de controle da ferrugem da soja (Glycine max), causada pelo fungo Phakopsora pachyrhizi, pode ser uma alternativa viável, visto que a resistência qualitativa tem-se mostrado instável, em função da variabilidade do fungo. Este trabalho foi realizado na região oeste da Bahia, na safra 2003/04, com objetivo de avaliar a tolerância das principais cultivares de soja utilizadas na região. Foram avaliadas quatro cultivares de ciclo precoce [MG/BR 46 (Conquista), Emgopa 315, BRS Corisco e M-SOY 8411] e quatro cultivares de ciclo tardio (BRS Barreiras, M-SOY 9350, FT 106 e BRS Sambaíba). A tolerância foi quantificada pela diferença de produtividade entre subparcelas tratadas e não tratadas com fungicida. As cultivares MG/BR 46 (Conquista) e M-SOY 8411 apresentaram diferenças de rendimento não significativas, no entanto, essa característica não pôde ser atribuída à tolerância, em função da baixa severidade observada no ensaio. Essa diferença pôde ser atribuída a escape, uma vez que, no momento em que a ferrugem obteve valores mais elevados de severidade, as duas cultivares já estavam na fase final de desenvolvimento. Embora a severidade máxima observada no ensaio tenha sido baixa (30,5% para a cultivar BRS Barreiras), com exceção das cultivares MG/BR 46 (Conquista) e M-SOY 8411, as demais cultivares avaliadas apresentaram diferença significativa entre as subparcelas tratadas e não tratadas com fungicida, mostrando ausência de tolerância das principais cultivares comerciais cultivadas no oeste da Bahia. Neste trabalho, foi observado que as cultivares de ciclo precoce apresentaram reduções de produtividade inferiores às de ciclo tardio.
