Devido às actividades antropogénicas várias substâncias químicas têm sido introduzidas no meio ambiente em concentrações que de outro modo não ocorreriam de forma tão elevada naturalmente. Assim, o conhecimento acerca das características de um químico, tais como, o potencial para se acumular em diferentes níveis tróficos, a sua mobilidade dentro do ecossistema, a toxicidade específica e a bioacumulação, é fundamental para compreender os seus efeitos nos ecossistemas. Esta tese investiga a influência de especiação, na biodisponibilidade do cádmio (Cd) para o isópode Porcellio dilatatus, incluindo os efeitos de especiação do metal: (i) na assimilação do Cd, (ii) no modo como o Cd se distribui internamente no organismo, e (iii) como a sobrevivência e a reprodução são afectadas em isópodes terrestres. Num primeiro ensaio laboratorial avaliou-se a importância da transferência trófica na assimilação do Cd em P. dilatatus. Para tal analisou-se a eficiência de assimilação (EA) do Cd em isópodes, adicionado superficialmente ao alimento (alface) na forma de Cd(NO3)2 e contaminando o meio de crescimento da alface. A hipótese era de que a alface contaminada biologicamente através do cultivo em meio hidropónico contaminado teria uma maior proporção de complexos com proteína ou conjugado na forma de Cd (ex. Cd cisteína). A EA de Cd foi maior entre os isópodes que foram alimentados com o sal (71%, SE = 7%), do que entre os isópodes que se alimentaram de alface contaminada biologicamente (52%, SE = 5%), demonstrando-se assim num teste laboratorial que é provável que a especiação do Cd influencie a taxa de assimilação e acumulação do Cd. Na experiência alimentar que se seguiu, estudou-se em detalhe a especiação do metal comparando as EA do Cd conjugado com cisteína (Cd(Cys)2) e na forma de Cd(NO3)2, com os quais se contaminou gelatina com alface. A utilização de Cd-cisteína, proporcionou uma forma experimental para explorar a biodisponibilidade do Cd complexado dentro do tecido biológico. Como esperado, a EA de Cd em isópodes alimentados com nitrato de Cd (64%, SE = 5%) foi maior do que no caso de isópodes alimentados com o conjugado de cisteína (20%, SE = 3%). De seguida estudou-se a distribuição subcelular das espécies de Cd assimilado através de um processo de fraccionamento. Supunha-se que as diferenças de especiação de Cd reflectiria diferentes estratégias de compartimentalização, com consequências ao nível da detoxificação, armazenamento celular e distribuição subcelular do metal. O “sequestro” na forma de metal biologicamente detoxificado (BDM = proteínas estáveis ao calor - HSP e grânulos ricos em metal - RMG) foi maior nos isópodes alimentados com Cd(NO3)2, sugerindo que são mais eficientes na detoxificação de Cd (22%) do que quando alimentados com Cd(Cys)2 (15%). Foi também demonstrado que os isópodes alimentados com Cd(Cys)2 possuíam níveis de armazenamento de Cd superior nas fracções sensíveis ao metal (MSF = organelos e proteínas desnaturadas pelo calor - HDP) consideradas fracções potencialmente vulneráveis e afectando os isópodes em termos de toxicidade. As diferentes distribuições internas que se seguiram à assimilação e detoxificação das diferentes espécies de Cd foram finalmente avaliadas em termos da sobrevivência e reprodução dos isópodes. O tratamento com Cd(Cys)2 teve maior mortalidade, provavelmente devido à maior disponibilidade de Cd ingerido com implicações ao nível dos processos fisiológicos. Os isópodes alimentados com Cd(NO3)2 armazenaram o Cd nos MRG, como estratégia de detoxificação, sendo mais eficientes a detoxificar o Cd ainda que aumentando a concentração total do metal que se tornou menos tóxico para o isópode. Desta forma, o Cd nos grânulos não estava disponível para os processos fisiológicos e deixou de ser tóxico. Isso poderia estar relacionado com a resistência e tolerância aos metais devido à capacidade dos isópodes compartimentalizarem o Cd no hepatopâncreas, que actua como um mecanismo de detoxificação e contribui para a tolerância a altos níveis de cádmio. Em termos de parâmetros reprodutivos, observou-se uma redução de gestações e duração da gestação na presença de ambas as espécies de metal, mas no caso do Cd(Cys)2 as gravidezes não se concluíram. O número de jovens produzido por fêmeas alimentadas com Cd(NO3)2 foi menor do que no controlo, mas os pesos dos juvenis foram superiores. Finalmente sugere-se assim que esta abordagem seja considerada em estudos do movimento trófico de metais nas cadeias alimentares dado que se espera que a especiação de metais implique diferentes fluxos, dentro de uma dada cadeia trófica.

O presente trabalho de investigação analisa a importância das redes formais e informais na internacionalização da economia do turismo, em particular do sector hoteleiro. Para tal baseia-se numa extensa revisão bibliográfica sobre as teorias que explicam o investimento directo no estrangeiro, assim como a abordagem das redes aplicada à internacionalização e o seu enquadramento no sector do turismo. Com base na revisão de literatura, uma série de hipóteses são sugeridas, as quais são testadas na parte empírica da tese através de uma análise às empresas Portuguesas com investimentos no estrangeiro na área da hotelaria até ao final de 2007 ou com projectos em curso. Esta análise baseia-se principalmente em dados obtidos através de entrevistas-questionário realizadas aos responsáveis das empresas. O inquérito obteve 40% de taxa de resposta, contendo dados relativos às características das empresas e de cada um dos projectos realizados no estrangeiro. Com base nestes resultados são sugeridas uma série de implicações, assim como algumas recomendações para investigações futuras. Adicionalmente, e de forma a investigar mercados com diferentes realidades sócio-culturais, políticas e cultura de negócios, foi analisado o caso de Goa, através de trabalho de campo que envolveu entrevistas informais e semiestruturadas a entidades-chave ligadas ao sector do turismo e aos hoteleiros de unidades de qualidade média-alta. Foi identificado um conjunto de oportunidades e desafios para as empresas Portuguesas. Ao usar uma abordagem qualitativa e quantitativa, esta tese contribui para a compreensão da natureza, determinantes e dimensões do processo de internacionalização do sector do turismo.

This thesis analyses the concept of Political Will, suggests its operationalization and establishes a typological theory that provides the necessary support for the diverse strategies of action of a leader. It claims that political leadership styles articulate a choice of action that results from the Political Will of a leader, which is determined by his intention and his discerned possibilities to act. One main research question guided our research: How does a political leader select and change his leadership style? The most illustrative literature on political leadership is reviewed and the characteristics of democratic governance are analyzed. This is followed by an overview of the most noteworthy theories on the theme and a claim for the need of concept coherence, given the multiplicity of the existent standpoints. After that, we concentrate on leadership styles, with a focus on the local governance context. Human action and intentionality are addressed with particular attention, as well as the motivational drivers for action, in order to advance a conceptualization of Political Will through two dimensions: intention and possibility. This analysis led to a number of relevant propositions: (1) Political Will ‘exists’ when the agent has the intent and the possibility to act; (2) these two dimensions ‘translate’ simultaneously what the agent believes he must do and can do; (3) Intention and possibility reflect diverse but limited worldviews; (4) political leadership styles result from the agent’s Political Will; (5) different combinations of the expected and actual worldviews result in different leadership styles; and (6) political leadership styles can change accordingly to several strategies which allow conformity or reflect reaction to worldviews. We suggested the operationalization of the two dimensions of Political Will through the analytical tool of Grid-group Theory, which provided the identification of the heuristic devices that allowed further comprehension on the subjectivity of the agent’s choice. Four standard property spaces – representing four types of leadership styles – result from a preliminary approach to this process. Afterwards, and because these dimensions operate simultaneously, we advance on the analysis and suggest some plausible heuristical conflicts to happen and describe which consequences, strategies and type migrations are conceivable. An inclusive and more complete set of resulting property spaces renders fourteen different types of leadership styles and sixty different predictable causal paths that result from the expected migration strategies. Case-studies were conducted as plausibility probes designed to provide improvements to our theoretical claims and addressed the cases we selected for research purposes: Portuguese Mayors. The findings from five case studies are discussed and the probable impact and congruence of each with the theoretical claims are assessed. The communalities of the causal mechanisms related to the function of intention and possibility as the dimensions of Political Will and their role in explaining different leadership styles are, finally, addressed. To conclude, we advance some repercussions, mainly in the public policies field of research, and suggest a number of different and necessary paths for further work.

Esta tese apresenta um estudo sobre otimização económica de parques eólicos, com o objetivo de obter um algoritmo para otimização económica de parques eólicos através do custo da energia produzida. No estudo utilizou-se uma abordagem multidisciplinar. Inicialmente, apresentam-se as principais tecnologias e diferentes arquiteturas utilizadas nos parques eólicos. Bem como esquemas de funcionamento e gestão dos parques. São identificadas variáveis necessárias e apresenta-se um modelo dimensionamento para cálculo dos custos da energia produzida, tendo-se dado ênfase às instalações onshore e ligados a rede elétrica de distribuição. É feita uma análise rigorosa das características das topologias dos aerogeradores disponíveis no mercado, e simula-se o funcionamento de um parque eólico para testar a validade dos modelos desenvolvidos. Também é implementado um algoritmo para a obtenção de uma resposta otimizada para o ciclo de vida económico do parque eólico em estudo. A abordagem proposta envolve algoritmos para otimização do custo de produção com multiplas funções objetivas com base na descrição matemática da produção de eletricidade. Foram desenvolvidos modelos de otimização linear, que estabelece a ligação entre o custo económico e a produção de eletricidade, tendo em conta ainda as emissões de CO2 em instrumentos de política energética para energia eólica. São propostas expressões para o cálculo do custo de energia com variáveis não convencionais, nomeadamente, para a produção variável do parque eólico, fator de funcionamento e coeficiente de eficiência geral do sistema. Para as duas últimas, também é analisado o impacto da distribuição do vento predominante no sistema de conversão de energia eólica. Verifica-se que os resultados obtidos pelos algoritmos propostos são similares às obtidas por demais métodos numéricos já publicados na comunidade científica, e que o algoritmo de otimização económica sofre influência significativa dos valores obtidos dos coeficientes em questão. Finalmente, é demonstrado que o algoritmo proposto (LCOEwso) é útil para o dimensionamento e cálculo dos custos de capital e O&M dos parques eólicos com informação incompleta ou em fase de projeto. Nesse sentido, o contributo desta tese vem ser desenvolver uma ferramenta de apoio à tomada de decisão de um gestor, investidor ou ainda agente público em fomentar a implantação de um parque eólico.

During the last century mean global temperatures have been increasing. According to the predictions, the temperature change is expected to exceed 1.5ºC in this century and the warming is likely to continue. Freshwater ecosystems are among the most sensitive mainly due to changes in the hydrologic cycle and consequently changes in several physico-chemical parameters (e.g. pH, dissolved oxygen). Alterations in environmental parameters of freshwater systems are likely to affect distribution, morphology, physiology and richness of a wide range of species leading to important changes in ecosystem biodiversity and function. Moreover, they can also work as co-stressors in environments where organisms have already to cope with chemical contamination (such as pesticides), increasing the environmental risk due to potential interactions. Therefore, the objective of this work was to evaluate the effects of climate change related environmental parameters on the toxicity of pesticides to zebrafish embryos. The following environmental factors were studied: pH (3.0-12.0), dissolved oxygen level (0-8 mg/L) and UV radiation (0-500 mW/m2). The pesticides studied were the carbamate insecticide carbaryl and the benzimidazole fungicide carbendazim. Stressors were firstly tested separately in order to derive concentration- or intensity-response curves to further study the effects of binary combinations (environmental factors x pesticides) by applying mixture models. Characterization of zebrafish embryos response to environmental stress revealed that pH effects were fully established after 24 h of exposure and survival was only affected at pH values below 5 and above 10. Low oxygen levels also affected embryos development at concentrations below 4 mg/L (delay, heart rate decrease and edema), and at concentrations below 0.5 mg/L the survival was drastically reduced. Continuous exposure to UV radiation showed a strong time-dependent impact on embryos survival leading to 100% of mortality after 72 hours of exposure. The toxicity of pesticides carbaryl and carbendazim was characterized at several levels of biological organization including developmental, biochemical and behavioural allowing a mechanistic understanding of the effects and highlighting the usefulness of behavioural responses (locomotion) as a sensitive endpoint in ecotoxicology. Once the individual concentration response relationship of each stressor was established, a combined toxicity study was conducted to evaluate the effects of pH on the toxicity of carbaryl. We have shown that pH can modify the toxicity of the pesticide carbaryl. The conceptual model concentration addition allowed a precise prediction of the toxicity of the jointeffects of acid pH and carbaryl. Nevertheless, for alkaline condition both concepts failed in predicting the effects. Deviations to the model were however easy to explain as high pH values favour the hydrolysis of carbaryl with the consequent formation of the more toxic degradation product 1- naphtol. Although in the present study such explanatory process was easy to establish, for many other combinations the “interactive” nature is not so evident. In the context of the climate change few scenarios predict such increase in the pH of aquatic systems, however this was a first approach focused in the lethal effects only. In a second tier assessment effects at sublethal level would be sought and it is expectable that more subtle pH changes (more realistic in terms of climate changes scenarios) may have an effect at physiological and biochemical levels with possible long term consequences for the population fitness.

Strong water demand for irrigation, energy and drinking water production is responsible for an increasingly regulation of freshwater flow patterns and watersheds. In this context, the construction of dams allows water storage but seriously restricts freshwater flow downstream. Due to scarcity of freshwater resources, reservoir water management often promotes high hydraulic residence. This may cause strong impacts on biological components of aquatic ecosystems, influencing the development of cyanobacteria blooms and aggravating their harmful impacts.

Marine protected areas (MPAs) have been widely proposed for conservation purposes and as a tool for fisheries management. The Arrábida Marine Park is the first MPA in continental Portugal having a management plan, fully implemented since 2009. The main objective of this study was to evaluate the effect of protection measures on rocky reef fish assemblages and target invertebrates through before-after and control-effect (no-take vs. fished areas) underwater visual surveys and analysis of landings trends. Second, we used surveys before, during and after implementation of the management plan to understand fishers‟ preferences for fishing grounds and adaptation to the new rules, and evaluated the reserve effect through analysis of both ecological responses and fishing effort density. Third, we identified the main oceanographic drivers influencing the structure of reef fish assemblages and predicted the community structure for the last 50 years, in light of climatic change. Overall results suggest positive responses in biomass but not yet in numbers of some commercial species, with no effects on non-target species. The reserve effect is reinforced by the increase in landings of commercial species, despite increased fishing effort density in some areas, especially with octopus traps. Fishing grounds are mainly chosen based on the distribution of target species and associated habitats, but distance to port, weather conditions and safety also influence fishers‟ choices. Moreover, different fisheries respond differently to the protection measures, and within each fishery, individual fishers show distinct strategies, with some operating in a broader area whereas others keep preferred territories. Our results also show that wind stress and temperature are the main oceanographic drivers for rocky reef fish assemblages, with tropicalization of assemblages and polewards movements of species over the last 50 years consistent with temperature trends. We believe this study provides significant lessons for marine conservation and management of coastal systems.

Mother-to-child transmission of HIV is a unique setting that allows us to explore both the correlates of protective immunity and the characteristics of transmitted variants. This thesis first describes the levels and functional capacity of breast milk HIV-specific antibodies in 19 women with high plasma viral loads. Neutralizing antibodies (Nabs) were detected in breast milk supernatant (BMS) of 4 of 19 women examined, were of low potency and were not associated with infant infection. The low NAb activity in BMS was reflected in binding antibody levels with HIV envelope specific IgG titers being 2.2 log10 lower in BMS versus plasma. In contrast, non- neutralizing antibodies (nNAbs) capable of antibody dependent cell-mediated cytotoxicity (ADCC) were detected in the BMS from all 19 women. BMS ADCC activity was associated with envelope-specific IgG titers (p = 0.014) and was inversely associated with infant infection risk (p = 0.039). Our data indicate that BMS has limited HIV neutralizing activity, however, BMS ADCC activity is a correlate of transmission that may impact infant infection risk. In the second part of this thesis the neutralization sensitivity of 111 variants of diverse subtypes obtained from mothers and infants was determined against 7 HIVspecific broadly neutralizing monoclonal antibodies (mAbs) (NIH45-46w, VRC01, PGT128, PGT121, PG9 PGT145 and b12). Maternal and infant variants did not differ in their neutralization sensitivity to these mAbs and neither did variants from transmitting versus those from non-transmitting women. However, subtype A viruses were iii significantly more sensitive to neutralization by NIH45-46w and VRC01 (p= 0.0001 in both cases) and PGT145 (p=0.03) compared to non-subtype A viruses. Together, NIH45- 46w and PGT128 neutralization profiles resulted in 100% coverage of the variants tested. These data suggest that the epitopes targeted by these mAbs are present and accessible in both circulating and transmitted variants and that a combination of antibodies would provide maximum coverage against diverse subtypes commonly found in HIV endemic regions. Overall, this data suggest that an antibody based HIV vaccine capable of eliciting antibodies of multiple specificities that can mediate ADCC and/or neutralizing activity can provide protection and conquer the genetic diversity displayed by HIV.

In life cycle impact assessment (LCIA) models, the sorption of the ionic fraction of dissociating organic chemicals is not adequately modeled because conventional non-polar partitioning models are applied. Therefore, high uncertainties are expected when modeling the mobility, as well as the bioavailability for uptake by exposed biota and degradation, of dissociating organic chemicals. Alternative regressions that account for the ionized fraction of a molecule to estimate fate parameters were applied to the USEtox model. The most sensitive model parameters in the estimation of ecotoxicological characterization factors (CFs) of micropollutants were evaluated by Monte Carlo analysis in both the default USEtox model and the alternative approach. Negligible differences of CFs values and 95% confidence limits between the two approaches were estimated for direct emissions to the freshwater compartment; however the default USEtox model overestimates CFs and the 95% confidence limits of basic compounds up to three orders and four orders of magnitude, respectively, relatively to the alternative approach for emissions to the agricultural soil compartment. For three emission scenarios, LCIA results show that the default USEtox model overestimates freshwater ecotoxicity impacts for the emission scenarios to agricultural soil by one order of magnitude, and larger confidence limits were estimated, relatively to the alternative approach.

The characteristics of carbon fibre reinforced laminates had widened their use, from aerospace to domestic appliances. A common characteristic is the need of drilling for assembly purposes. It is known that a drilling process that reduces the drill thrust force can decrease the risk of delamination. In this work, delamination assessment methods based on radiographic data are compared and correlated with mechanical test results (bearing test).

An infinite-horizon discrete time model with multiple size-class structures using a transition matrix is built to assess optimal harvesting schedules in the context of Non-Industrial Private Forest (NIPF) owners. Three model specifications accounting for forest income, financial return on an asset and amenity valuations are considered. Numerical simulations suggest uneven-aged forest management where a rational forest owner adapts her or his forest policy by influencing the regeneration of trees or adjusting consumption dynamics depending on subjective time preference and market return rate dynamics on the financial asset. Moreover she or he does not value significantly non-market benefits captured by amenity valuations relatively to forest income.

The impact of the Board of Directors’ composition on companies’ performance This paper studies the impact that the board of directors’ composition has on companies’ performance in the Italian market. The research has been carried out by using a sample of 10 Italian companies, across different market sectors, over a period of 10 years (2005-2014). The characteristics of the BoD taken into consideration are the following: board size, board diversity (% of female directors), board independence and CEO duality. Results from the sample data collected concluded that these factors have a statistically significant impact on the performance of the companies that have been analysed.

The article considers the ways of organization of databases for the storage of the results obtained during testing. A new variant of the organization of the data to ensure the ability to write to the database different sets of parameters in the form of chronological series. The required set of parameters depends on the modification of the tested technical installation.
