Rapid quantitative detection of Yersinia pestis by lateral-flow immunoassay and up-converting phosphor technology-based biosensor

Up-converting phosphor technology (UPT)-based lateral-flow immunoassay has been developed for quantitative detection of Yersinia pestis rapidly and specifically. In this assay, 400 nm up-converting phosphor particles were used as the reporter. A sandwich immumoassay was employed by using a polyclonal antibody against F1 antigen of Y. pestis immobilized on the nitrocellulose membrane and the same antibody conjugated to the UPT particles. The signal detection of the strips was performed by the UPT-based biosensor that could provide a 980 nm IR laser to excite the phosphor particles, then collect the visible luminescence emitted by the UPT particles and finally convert it to the voltage as a signal. V-T and V-c stand for the multiplied voltage units for the test and the control line, respectively, and the ratio V-T/V-C is directly proportional to the number of Y pestis in a sample. We observed a good linearity between the ratio and log CFU/ml of Y pestis above the detection limit, which was approximately 10(4) CFU/mI. The precision of the intra- and inter-assay was below 15% (coefficient of variation, CV). Cross-reactivity with related Gram-negative enteric bacteria was not found. The UPT-LF immunoassay system presented here takes less than 30 min to perform from the sample treatment to the data analysis. The current paper includes only preliminary data concerning the biomedical aspects of the assay, but is more concentrated on the technical details of establishing a rapid manual assay using a state-of-the-art label chemistry. Cooperative up-conversion luminescence of ytterbium doped yttrium lanthanum oxide transparent ceramic

The up-conversion luminescence of Yb3+-doped yttriurn lanthanum oxide transparent ceramic was investigated. It was ascribed to cooperative luminescence originated from the coupled states of the Yb3+ ion pairs. The proper doping of La2O3 can remove the cooperative luminescence of Yb3+ ion. But excessive La2O3 (at least 10 at.%) the cooperative up-conversion of Yb3+:Y2O3 is obtained again, and the intensity of up-conversion luminescence strengthens with the increase of La2O3 content. An evaluation of mark and recapture techniques for estimating tigerfish biomass in Lake Kariba

The effectiveness of 2 mark and recapture techniques was evaluated using tiger fish, Hydrocynus vittatus. The 2 techniques used were: tagging with a plastic tag and fluorescent spray marking. While the tagging method resulted as the logical method to use within the constraints of the tiger fish study, it cannot be considered completely reliable for the estimation of population size in Lake Kariba.

Presságios literários e herança plástica: Emerson e Thoreau; Richard Serra, Waltercio Caldas e outros

O presente estudo ambiciona examinar as escritas literárias de Ralph Waldo Emerson (1803-1882) e Henry David Thoreau (1817-1862) considerando-as como uma forma de presságio de arte na América, ou seja, como uma escrita literária que parece ter sido herdada no ambiente artístico das obras plásticas contemporâneas de Richard Serra (1939-) e Waltercio Caldas (1946-), entre outras. Tal herança endossa a noção de linguagem ordinária, compreendida como o ponto de acolhimento, ou de uma inquietação, que se faz presente nas circunstâncias da contemporaneidade. Por sua vez, o gesto de endereçamento que envolve estas escrituras expressa a marca de uma indecibilidade acerca da continuidade ou descontinuidade da existência de categorias como literatura, filosofia e artes. Assim, a problematização dessas remissões ganha vulto no presente estudo, por meio de uma abordagem em perspectiva comparada entre Thoreau, Emerson, Waltercio, Serra e outros. E deste modo, a questão que se estabelece nesse panorama diz respeito aos problemas do pensamento, que em âmbito plástico parecem se estender para uma tradição crítica no Novo Mundo

Modified sorting technique to mitigate the collateral mortality of trawled school prawns (Metapenaeus macleayi)

The potential for changes to onboard handling practices in order to improve the fate of juvenile school prawns (Metapenaeus macleayi) discarded during trawling were investigated in two Australian rivers (Clarence and Hunter) by comparing a purpose-built, water-filled sorting tray against a conventional dry tray across various conditions, including the range of typical delays before the start of sorting the catch (2 min vs. 15 min). Juvenile school prawns (n= 5760), caught during 32 and 16 deployments in each river, were caged and sacrificed at four times: immediately (T0), and at 24 (T24), 72 (T72), and 120 (T12 0) hours after having been discarded. In both rivers, most mortalities occurred between T0 and T24 and, after adjusting for control deaths (<12%), were greatest for the 15-min conventional treatment (up to 41% at T120). Mixed-effects logistic models revealed that in addition to the sampling time, method of sorting, and delay in sorting, the weight of the catch, salinity, and percentage cloud cover were significant predictors of mortality. Although trawling caused some mortalities and comparable stress (measured as L -lactate) in all school prawns, use of the water tray lessened the negative impacts of some of the above factors across both the 2-min and 15-min delays in sorting so that the overall discard mortality was reduced by more than a third. When used in conjunction with selective trawls, widespread application of the water tray should help to improve the sustainability of trawling for school prawns.

Use of pop-up satellite archival tag technology to study postrelease survival of and habitat use by estuarine and coastal fishes: an application to striped bass (Morone saxatilis)

Pop-up satellite archival tags (PSATs) have been used to study movements, habitat use, and postrelease survival of large pelagic vertebrates, but the size of these tags has historically precluded their use on smaller coastal species. To evaluate a new generation of smaller PSATs for the study of postrelease survival and habitat use of coastal species, we attached Microwave Telemetry, Inc., X-tags to ten striped bass (Morone saxatilis) 94–112 cm total length (TL) caught on J hooks and circle hooks during the winter recreational fishery in Virginia. Tags collected temperature and depth information every five minutes and detached from the fish after 30 days. Nine of the ten tags released on schedule and eight transmitted 30% to 96% (mean 78.6%) of the archived data. Three tags were physically recovered during or after the transmission period, allowing retrieval of all archived data. All eight striped bass whose tags transmitted data survived for 30 days after release, including two fish that were hooked deeply with J hooks. The eight fish spent more than 90% of their time at depths less than 10 m and in temperatures of 6–9°C, demonstrated no significant diel differences in depth or temperature utilization (P>0.05), and exhibited weak periodicities in vertical movements consistent with daily and tidal cycles.
