The integration of high-resolution archaeological, textual, and environmental data with longer-term, low-resolution data affords greater precision in identifying some of the causal relationships underlying societal change. Regional and microregional case studies about the Byzantine world—in particular, Anatolia, which for several centuries was the heart of that world—reveal many of the difficulties that researchers face when attempting to assess the influence of environmental factors on human society. The Anatolian case challenges a number of assumptions about the impact of climatic factors on socio-political organization and medium-term historical evolution, highlighting the importance of further collaboration between historians, archaeologists, and climate scientists.

The actin nodule is a novel F-actin structure present in platelets during early spreading. However, only limited detail is known regarding nodule organization and function. Here we use electron microscopy, SIM and dSTORM super-resolution, and live-cell TIRF microscopy to characterize the structural organization and signalling pathways associated with nodule formation. Nodules are composed of up to four actin-rich structures linked together by actin bundles. They are enriched in the adhesion-related proteins talin and vinculin, have a central core of tyrosine phosphorylated proteins and are depleted of integrins at the plasma membrane. Nodule formation is dependent on Wiskott-Aldrich syndrome protein (WASp) and the ARP2/3 complex. WASp(-/-) mouse blood displays impaired platelet aggregate formation at arteriolar shear rates. We propose actin nodules are platelet podosome-related structures required for platelet-platelet interaction and their absence contributes to the bleeding diathesis of Wiskott-Aldrich syndrome.

We examined the activation of the p38 mitogen-activated protein kinase (p38-MAPK) pathway by the G protein-coupled receptor agonists, endothelin-1 and phenylephrine in primary cultures of cardiac myocytes from neonatal rat hearts. Both agonists increased the phosphorylation (activation) of p38-MAPK by approximately 12-fold. A p38-MAPK substrate, MAPK-activated protein kinase 2 (MAPKAPK2), was activated approximately fourfold and 10 microM SB203580, a p38-MAPK inhibitor, abolished this activation. Phosphorylation of the MAPKAPK2 substrate, heat shock protein 25/27, was also increased. Using selective inhibitors, activation of the p38-MAPK pathway by endothelin-1 was shown to involve protein kinase C but not Gi/Go nor the extracellularly responsive kinase (ERK) pathway. SB203580 failed to inhibit the morphological changes associated with cardiac myocyte hypertrophy induced by endothelin-1 or phenylephrine between 4 and 24 h. However, it decreased the myofibrillar organization and cell profile at 48 h. In contrast, inhibition of the ERK cascade with PD98059 prevented the increase in myofibrillar organization but not cell profile. These data are not consistent with a role for the p38-MAPK pathway in the immediate induction of the morphological changes of hypertrophy but suggest that it may be necessary over a longer period to maintain the response.

Large freshwater lakes formed in North America and Europe during deglaciation following the Last Glacial Maximum. Rapid drainage of these lakes into the Oceans resulted in abrupt perturbations in climate, including the Younger Dryas and 8.2 kyr cooling events. In the mid-latitudes of the Southern Hemisphere major glacial lakes also formed and drained during deglaciation but little is known about the magnitude, organization and timing of these drainage events and their e ect on regional climate. We use 16 new single-grain optically stimulated luminescence (OSL) dates to de ne three stages of rapid glacial lake drainage in the Lago General Carrera/Lago Buenos Aires and Lago Cohrane/ Pueyrredón basins of Patagonia and provide the rst assessment of the e ects of lake drainage on the Paci c Ocean. Lake drainage occurred between 13 and 8 kyr ago and was initially gradual eastward into the Atlantic, then subsequently reorganized westward into the Paci c as new drainage routes opened up during Patagonian Ice Sheet deglaciation. Coupled ocean-atmosphere model experiments using HadCM3 with an imposed freshwater surface “hosing” to simulate glacial lake drainage suggest that a negative salinity anomaly was advected south around Cape Horn, resulting in brief but signi cant impacts on coastal ocean vertical mixing and regional climate.

There is a widespread assumption that clients’ expectations should be accommodated during a building project. However, there may be conflicting expectations within a client organization and these may change over time in the course of a project. Actor-Network Theory (ANT) is used to study the incorporation of client expectations into the on-going development of a building project. To illustrate this, negotiations over a particular decision, namely the location of a building on one university campus was analysed. Negotiations went through a number of stages, involving a master plan architect, members of the public, campus maps and the Vice Chancellor. An ANT analysis helped to trace diverse actors' interests in a series of discussions and how these interests conflict with each other as one option was chosen over another. The analysis revealed new client interests in each negotiation process. Also, the prioritisation of client interests changed over time. The documentation of diverse and dynamic client interests especially contributes to the understanding of how some client interests fail to be incorporated in decision-making processes

The aim of this study was to evaluate working conditions in the textile industry for different stages of Corporate Social Responsibility (CSR) development, and workers` perception of fatigue and workability. A cross-sectional study was undertaken with 126 workers in the production areas of five Brazilian textile plants. The corporate executive officers and managers of each company provided their personal evaluations of CSR. Companies were divided into 2 groups (higher and lower) of CSR scores. Workers completed questionnaires on fatigue, workability and working conditions. Ergonomic job analysis showed similar results for working conditions, independent of their CSR score. Multivariate analysis models were developed for fatigue and workability, indicating that they are both associated to factors related to working conditions and individual workers` characteristics and life styles. Work organization, (what, how, when, where and for how long the work is done), is also an associated factor for fatigue. This study suggests that workers` opinions should be taken into greater consideration when companies develop their CSR programs, in particular for those relating to working conditions. Relevance to industry: This paper underlines the importance of considering working conditions and workers` opinions of them, work organization and individual workers` characteristics and life styles in order to restore or to maintain workability and to reduce fatigue, independently of how developed a company may be in the field of Corporate Social Responsibility. Objective. To search the literature for circumstances that impede injury and disease prevention and other activities intended to improve the health of the health care worker. Methods. The SciELO database was searched for articles published in 1967-2008. This was supplemented by a PubMed search for the period 1950-2008. The following key words were used to identify articles in English, Portuguese, and Spanish: work, health personnel, occupational, risks, diseases, ergonomics, work ability, quality of life, organization, accidents, work conditions, intervention, and administration. Articles on injury and disease prevention and occupational health in a health care setting in Latin America were selected, along with articles focused on health promotion in the health sector. Results. The following shortcomings were identified: activities lacked a sound theoretical foundation and were not integrated with the health services management; a failure to evaluate the effectiveness of the activity; health surveillance focused solely on a specific disease or injury; management not committed to the proposed activity; miscommunication; inability of workers to participate, or control the work environment; and, programs or efforts that were limited to changing the workers` behaviors. Conclusions. The literature shows that all the barriers identified by this study affect both the health care workers` health as well as their productivity.

The authors present here a summary of their investigations of ultrathin films formed by gold nanoclusters embedded in polymethylmethacrylate polymer. The clusters are formed from the self-organization of subplantated gold ions in the polymer. The source of the low energy ion stream used for the subplantation is a unidirectionally drifting gold plasma created by a magnetically filtered vacuum arc plasma gun. The material properties change according to subplantation dose, including nanocluster sizes and agglomeration state and, consequently also the material electrical behavior and optical activity. They have investigated the composite experimentally and by computer simulation in order to better understand the self-organization and the properties of the material. They present here the results of conductivity measurements and percolation behavior, dynamic TRIM simulations, surface plasmon resonance activity, transmission electron microscopy, small angle x-ray scattering, atomic force microscopy, and scanning tunneling microscopy. Cortical bones, essential for mechanical support and structure in many animals, involve a large number of canals organized in intricate fashion. By using state-of-the art image analysis and computer graphics, the 3D reconstruction of a whole bone (phalange) of a young chicken was obtained and represented in terms of a complex network where each canal was associated to an edge and every confluence of three or more canals yielded a respective node. The representation of the bone canal structure as a complex network has allowed several methods to be applied in order to characterize and analyze the canal system organization and the robustness. First, the distribution of the node degrees (i.e. the number of canals connected to each node) confirmed previous indications that bone canal networks follow a power law, and therefore present some highly connected nodes (hubs). The bone network was also found to be partitioned into communities or modules, i.e. groups of nodes which are more intensely connected to one another than with the rest of the network. We verified that each community exhibited distinct topological properties that are possibly linked with their specific function. In order to better understand the organization of the bone network, its resilience to two types of failures (random attack and cascaded failures) was also quantified comparatively to randomized and regular counterparts. The results indicate that the modular structure improves the robustness of the bone network when compared to a regular network with the same average degree and number of nodes. The effects of disease processes (e. g., osteoporosis) and mutations in genes (e.g., BMP4) that occur at the molecular level can now be investigated at the mesoscopic level by using network based approaches.

Nitrogen uptake and metabolism are essential to microbial growth. Gat1 belongs to a conserved family of zinc finger containing transcriptional regulators known as GATA-factors. These factors activate the transcription of Nitrogen Catabolite Repression (NCR) sensitive genes when preferred nitrogen sources are absent or limiting. Cryptococcus neoformans GAT1 is an ortholog to the Aspergillus nidulans AreA and Candida albicans GAD genes. In an attempt to define the function of this transcriptional regulator in C. neoformans, we generated null mutants (gat1 Delta) of this gene. The gat 1 mutant exhibited impaired growth on all amino acids tested as sole nitrogen sources, with the exception of arginine and proline. Furthermore, the gat1 mutant did not display resistance to rapamycin, an immunosuppressant drug that transiently mimics a low-quality nitrogen source. Gal is not required for C. neoformans survival during macrophage infection or for virulence in a mouse model of cryptococcosis. Microarray analysis allowed the identification of target genes that are regulated by Gat1 in the presence of proline, a poor and non-repressing nitrogen source. Genes involved in ergosterol biosynthesis, iron uptake, cell wall organization and capsule biosynthesis, in addition to NCR-sensitive genes, are Gat1-regulated in C. neoformans. This report describes the outcome of the first visit to Tanzania, within the project "Mini-grids supplied by renewable energy - improving technical and social feasibility". The trip included visits to three different organizations; Ihushi Development Center (IDC) near Mwanza, TIDESO near Bukoba, and Mavuno Project in Karagwe. At IDC, a brief evaluation of the current power system was done and measuring equipment for long term measurements were installed. At all three locations investigations regarding the current and future electricity demand were conducted and connections to people relevant to the study were established. The report is including as well some technical specifications as some observations regarding organization and management of the technical systems. The trip was including only short visits and therefore only brief introductions to the different organizations, based on observations done by the author. A institucionalização de acervos particulares de artistas não é um fenômeno recente, mas esse processo se acelerou nos últimos 30 anos. As transformações sociais decorrentes do estabelecimento de uma ordem econômica globalizada criaram novas regras no universo das organizações culturais. Este trabalho é o resultado de uma pesquisa sobre instituições responsáveis por gerir acervos de artistas contemporâneos realizada com o objetivo de encontrar diretrizes para a constituição de uma iniciativa da mesma ordem: o Ateliê Carlos Vergara. O tema foi examinado com base em entrevistas com cinco artistas plásticos brasileiros e em um levantamento sobre a trajetória de Carlos Vergara e de seu ateliê. Algumas questões relacionadas à organização e à preservação dos acervos, à sustentabilidade das instituições, ao papel dos herdeiros em sua gestão, assim como ao alinhamento dessas iniciativas aos princípios dos artistas, são discutidas neste texto.

Este estudo investigou, na Companhia Vale do Rio Doce (CVRD), como os funcionários perceberam as mudanças ocorridas na empresa, as ações da alta administração e o que ocorreu com os valores na organização após o processo de privatização. O objetivo final da pesquisa foi verificar que valores norteiam as propostas administrativas da Vale após a privatização, e como se manifestam nas ações e na postura dos agentes na sede da empresa. O referencial teórico incluiu mudança organizacional, teoria dos valores e responsabilidade das ações administrativas, para compreender o contexto atual da empresa após a privatização. A pesquisa de campo foi concebida sob o paradigma do construtivismo, em busca de percepções e sentimentos subjacentes e manifestos em entrevistas e questionários a fim de explicitar os valores passados e presentes na organização e possibilidades de uma nova realidade. Uma abordagem fenomenológica complementar favoreceu a inserção da autora no mundo da vida dessa organização, facilitando a fluência do diálogo para a apreensão de manifestações e implicações dos valores das pessoas no processo de transição. As reflexões finais indicam a necessidade de um trabalho organizacional que envolva funcionários e alta administração com o propósito comum de redescoberta do significado próprio da organização. O Programa Vale Viver surge como uma tentativa nessa direção. Os valores "vestir a camisa", "valorizar a prata da casa" e "vencer desafios" acompanham o tempo de vida da cultura da Vale. O valor "vestir a camisa," cujo significado é compartilhamento autêntico, na atualidade é mais uma expectativa a ser retomada do que uma vivência. O valor "vencer desafios" está mais presente na área operacional e o valor "valorizar a prata da casa" aparece mais relacionado a programas como o Banco de OpOliunidades, reavivado pelo Vale Viver. Não significa que o Vale Viver seja a única saída para a retomada da confiança, da elevação da auto-estima e de uma articulação empregados e nova administração na construção de uma base comum de valores, mas é a possibilidade mais visível no momento em que a retomada do espírito da V ALE é fundamental para essa nova etapa de sua vida.

Este trabalho tem o objetivo de analisar. explorar e descrever as características culturais do BANESTES SI A - Banco do Estado do Espírito Santo. que influenciam na sua capacidade de adequar-se à forma de atuação do sistema financeiro privado, concluindo se estas características favorecem ou dificultam sua adaptabilidade. O desenvolvimento da dissertação se dá com a pesquisa bibliográfica referente ao arcabouço teórico da cultura organizacional e da evolução do Sistema Financeiro Nacional, de uma investigação documental para se levantar os dados e descrever o perfil atual do BANESTES e de uma pesquisa de campo. com entrevistas e observações. A pesquisa realizada para a elaboração do trabalho é de natureza qualitativa, do tipo exploratório. descritiva e avaliativa. O modo de investigação é o do estudo de caso histórico-organizacional, com observação participante. Os principais fundamentos que compõem o trabalho são extraídos da pesquisa histórica, das teorias convencionais de administração, da base teórica da cultura organizacional, da organização em aprendizagem e da transformação organizacional. A análise e observações estão descritas ao final da dissertação, mais precisamente no quinto capítulo. A bibliografia compõe um quadro de todos os autores e documentos pesquisados que oferecem suporte à dissertação.

Esta dissertação tem como objetivo principal analisar a forma como os líderes de uma organização que alavanca suas atividades com uma estrutura em rede, transmitem a cultura da empresa central junto às empresas periféricas. Pretende-se investigar também se os líderes mudaram o seu papel em função da mudança da arquitetura organizacional no período considerado por alguns autores como pós-moderno. Como o tema cultura é amplo e complexo, optou-se por enfocar aspectos como a transmissão da missão e objetivos organizacionais, além do estilo de gestão predominante e sua influência no gerenciamento de projetos entre os protagonistas dessas diferentes organizações. Utilizou-se, para tanto, uma base teórico-empírica que buscou definir o conceito de cultura através do estudo do universo simbólico; a questão da liderança e sua relação com estruturas e fontes de poder e controle nas organizações; cultura e liderança no contexto brasileiro; e, por último, o conceito de organizações em rede, explorando também os aspectos de gestão, poder e controle esperado nesse tipo de arquitetura organizacional. Esta pesquisa constitui-se num estudo de caso realizado em uma instituição sem fins lucrativos, que desenvolve projetos sociais com foco em educação. Trata-se de um estudo exploratório, descritivo e explicativo. O método utilizado para a presente pesquisa foi o método qualitativo. Muitos estudos sobre cultura organizacional privilegiam o enfoque qualitativo de pesquisa, uma vez que um grande número de observações não é passível de quantificação. Utilizou-se a técnica de observação participante e de entrevistas semi-estruturadas, além da análise documental, o que possibilitou uma investigação que envolveu a combinação de descrição com interpretação do conteúdo trazido pelos respondentes em relação aos fenômenos observados. Os dados obtidos e a análise realizada frente à fundamentação teórica, indicam que o líder em uma organização em rede transmite a cultura organizacional com o mesmo discurso dominante das organizações com estruturas tradicionais. Acredita-se que este trabalho despertará interesse da comunidade acadêmica, uma vez que os aspectos culturais são normalmente pesquisados dentro da organização e aqui se ressalta a importância de se analisar o contexto no qual a organização está inserida e o papel que o líder assume na gestão das atividades realizadas por terceiros. Estaria o gestor brasileiro disposto a abrir mão do poder e controle que a posição hierárquica lhe confere para trabalhar em uma relação de igualdade com representantes de outras organizações?
