A espécie suína (Sus scropha domesticus) possui relevância nos âmbitos da pesquisa, da xenotransplantação e da produção de carnes. O resfriamento de sêmen é capaz de reduzir o metabolismo celular e possibilitar o armazenamento dos gametas, sendo auxiliar durante práticas de reprodução assistida. Contudo, os espermatozoides suínos são sensíveis ao estresse oxidativo gerado durante o processo de resfriamento. O 2,4 dinitrofenol (DNP) poderia gerar o desacoplamento mitocondrial reduzindo o estresse oxidativo e prolongando indiretamente a viabilidade e capacidade fertilizante de espermatozoides suínos resfriados pela diminuição de espécies reativas de oxigênio. O objetivo do presente estudo foi avaliar os efeitos do desacoplamento mitocondrial induzido pelo DNP e os efeitos desse desacoplamento sobre as de fluidez e integridade de membrana plasmática, funcionalidade de mitocôndria, motilidade espermática, além dos parâmetros de estresse oxidativo de lipoperoxidação e produção de espécies reativas de oxigênio, durante o resfriamento a 17 °C de 24 até 96 horas. Utilizou-se 22 ejaculados expostos ao diluente Betsville Thawing Solution (BTS) (controle) e ao mesmo diluente acrescido das concentrações de 0,01 µM (T1); 0,1 µM (T2); 1,0 µM (T3) e 10 µM (T4) de DNP. Através do teste de Shapiro-Wilk as variáveis que não apresentaram normalidade, tiveram suas médias comparadas pelo teste de KruskalWallis. As análises estatísticas demonstraram que o DNP não se diferiu do controle independente do tempo de armazenamento, para nenhuma das variáveis analisadas. Possivelmente, a falta de ação sobre as mitocôndrias, ou seja, a não promoção do desacoplamento mitocondrial foi o motivo principal para a falta dos efeitos do DNP. Acredita-se que a utilização do DNP em temperaturas mais elevadas por estas promoverem aumento da fluidez de membrana e/ou o aumento das concentrações de DNP poderiam gerar efeitos significativos sobre as mitocôndrias e demais variáveis analisadas. Finalmente, o DNP nas concentrações testadas não se diferiu do controle em todas as variáveis analisadas independente do tempo de armazenamento.

-D-glucosidase (EC 3.2.1.21) is one of the most interesting glycosidases, especially for hydrolysis cellobiose releasing glucose, is last step degradation of cellulose. This function makes the -D-glucosidase is of great interest as a versatile industrial biocatalyst, being critical to various bio-treatment / biorefinery processes, such as bioethanol production. Hen in the report, a -D-glucosidase was extracts from protein extracted of the invertebrate marine Artemia franciscana was purified and characterized with a combination of precipitation with ammonium sulfate (0 - 30%, 30 to 50%, 50 to 80%), the fraction saturated in the range of 30 to 50% (called F-II) was applied in a molecular exclusion chromatography, in Sephacryl S-200, the fractions corresponding to the first peak of activity of -D-glucosidase were gathered and applied in a chromatography of ion exchange in Mono Q; the third peak this protein obtained chromatography, which coincides with the peak of activity of -D-glucosidase was held and applied in a gel filtration chromatography Superose 12 where the first peak protein, which has activity of -D-glucosidase was rechromatography on Superose 12. This enzyme is probably multimerica, consisting of three subunit molecular mass of 52.7 kDa (determined by SDS-PAGE) with native molecular mass of 157 kDa (determined by gel filtration chromatography on Superose 12 under the system FPLC). The enzyme was purified 44.09 times with a recovery of 1.01%. Using up p-nitrophenyl-β-D-glucopiranoside as substrate obtained a Km apparent of 0.229 mM and a Vmax of 1.109 mM.60min-1.mL-1mM. The optimum pH and optimum temperature of catalysis of the synthetic substrate were 5.0 and 45 °C, respectively. The activity of the -D-glucosidase was strongly, inhibited by silver nitrate and N- etylmaleimide, this inhibition indicates the involvement of radical sulfidrila the hydrolysis of synthetic substrate. The -D-glucosidase of Artemia franciscana presented degradativa action on celobiose, lactose and on the synthetic substrate -nitrophenyl-β-D-glucopiranoside indicating potential use of this enzyme in the industry mainly for the production of bioethanol (production of alcohol from the participating cellulose), and production hydrolysate milk (devoid of milk lactose)

Beginnend mit dem Band „Musikpädagogische Forschung 1980 – Einzeluntersuchungen" legt der Arbeitskreis Musikpädagogische Forschung e.V. eine neue Publikationsreihe vor, in der er jährlich über die Arbeit seiner Mitglieder und über wichtige Forschungen im In- und Ausland berichten will. Das Jahrbuch ist damit nicht nur ein Forum der immer zahlreicher werdenden musikpädagogisch forschenden Kollegen in der Bundesrepublik Deutschland, sondern soll auch über die zunehmende Fülle musikpädagogischer und musikdidaktischer Grundlagenforschung in zahlreichen Ländern der Welt informieren. Musikpädagogische Forschung ist nicht an Methoden gebunden, sondern an Fragestellungen orientiert: Die Inhalte des ersten Bandes werden deshalb dokumentieren, daß der Pluralismus der Methoden in den letzten Jahren für viele Kollegen selbstverständlich geworden ist. Musikpädagogische Forschung braucht sich nicht eigens zu legitimieren; wenn ihre Ergebnisse in den letzten Jahren nicht immer den erwünschten Praxisbezug hatten, so ist das nicht nur auf die Jugendlichkeit dieses Forschungsimpulses zurückzuführen, sondern auch auf die Schwierigkeit, fächerübergreifend zu arbeiten. Das Jahrbuch „Musikpädagogische Forschung" wird sich deshalb besonders darum bemühen, Musikpädagogen, Musikwissenschaftler, Musikpsychologen, Musiktherapeuten sowie Vertreter anderer Disziplinen zur Mitarbeit zu bewegen und anzusprechen. Leishmaniasis are endemic diseases wild spread in the New and Old World, caused by the flagelated protozoan Leishmania. In the New World, the distribution of different forms of leishmaniasis is mostly in tropical regions. In the State of Rio Grande do Norte, Northeast Brazil, 85% of the captured sand flies fauna is Lutzomyia longipalpis. The distribution of the sand fly vector in the state overlaps with the disease distribution, where the presence of sand flies is associated with presence of animals shelters. The aim of this study was to analyse the blood meal preference of sand flies vector from the genus Lutzomyia spp. in laboratory conditions, to verify the vector life cicle at different temperatures sets and to identify the main blood meal source in endemic areas for visceral leishmaniasis (VL) at peri-urban regions of Natal. Sand flies samples were collected from the municipalities of São Gonçalo do Amarante and Nísia Floresta where female sand flies were grouped for the colony maintenance in the laboratory and for the analysis of the preferred source of sand fly blood meal in natural environment. The prevalence of blood meal preference and oviposition for the females sand flies was 97% for Cavia porcellus with oviposition of 19 eggs/female; 97% for Eqqus caballus with 19 eggs/female; 98% for human blood with 14 eggs/female; 71.3% for Didelphis albiventris with 8.4 eggs/female; 73% for Gallus gallus with 14 eggs/female; 86% for Canis familiaris with 10.3 eggs/female; 81.4% for Galea spixii with 26 eggs/female; 36% for Callithrix jachus with 15 eggs/female; 42.8% for Monodelphis domestica with 0% of oviposition. Female sand flies did not take a blood meal from Felis catus. Sand flies life cycle ranged from 32-40 days, with 21-50 oviposition rates approximately. This study also showed that at 32°C the life cycle had 31 days, at 28° C it had 50 days and at 22°C it increased to 79 days. Adjusting the temperature to 35°C the eggs did not hatch, thus blocking the life cycle. A total of 1540 sand flies were captured, among them, 1.310 were male and 230 were female. Whereas 86% of the sand flies captured were Lu. longipalpis as compared to 10.5% for Lu. evandroi and, 3.2% for L. lenti and 0.3% for Lu whitmani. The ratio between female and male sandfly was approximately 6 males to 1 female. In Nísia Floresta, 50.7% of the collected females took their blood meal from armadillo, 12.8% from human. Among the female sand flies captured in São Gonçalo do Amarante, 80 of them were tested for the Leishmania KDNA infectivity where 5% of them were infected with Leishmania chagasi. Female Lutzomyia spp. showed to have an opportunistic blood meal characteristic. The behavioral parameters seem to have a higher influence in the oviposition when compared to the level of total proteins detected in the host s bloodstream. A higher Lu. longipalpis life cycle viability was observed at 28°C. The increase of temperature dropped the life cycle time, which means that the life cycle is modified by temperature range, source of blood meal and humidity. Lu longipalpis was the most specie found in the inner and peridomiciliar environment. In Nísia Floresta, armadillos were the main source of blood meal for Lutzomyia spp. At São Gonçalo do Amarante, humans were the main source of blood meal due to CDC nets placed inside their houses

Background: Indoor aeroallergens are the main cause of sensitization in children and represent a risk factor for the development of allergic diseases. Objective: Identify the major indoor aeroallergens most often sensitized to pediatric patients treated at the Allergy Service at the “Dr. José Eleuterio González” University Hospital of Monterrey Methods: We performed an observational and descriptive study where we reviewed reports of positive skin tests to the following common indoor aeroallergens: Dermatophagoides farinae (D. farinae), Dermatophagoides pteronyssinus (D. pteronyssinus), Canis familiaris (C. familiaris), Felis domesticus (F. domesticus), Blattella germanica (B. germanica) and Periplaneta americana (P. americana), found in patients under 16 years with symptoms of allergy, during the period of 2011-2012. Results: We performed 439 skin tests to aeroallergens in pediatric patients. Of these, 57.6% were male and 42.4% were female. Mean age was 6.3 years. The age groups were under 3 years: 17.8%, 3-5 years: 35%, 6-12 years: 36%, and 13-16 years: 11.2%. The main diagnoses were: allergic rhinitis (71.8%), asthma (16.6%), and atopic dermatitis (4.3%). In 57.9% of the cases, they had at least one positive skin test to any aeroallergen. The rate of sensitization to speciic aeroallergens was: D. Pteronyssinus 49.0%, D. farinae 44.6%, B. germanica 13.9%, P. Americana 10.9%, F. domesticus 10.7%, and C. familiaris 5.9%. Conclusion: Indoor aeroallergen sensitization can occur early in life, although it was more frequent in the preschooler and elementary school group. Dust house mites were the most commom cause of allergic sensitization.

Purpose: To investigate the effect of licochalcone A (LA) on the inhibition of cell proliferation and ERK1/2 phosphorylation in bladder carcinoma cell lines. Methods: Cell viability was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide (MTT) assay. Dye-binding method was used to examine the concentration of proteins. Lymphocytes were extracted from mice and after surface staining were subjected to BD fixation and permeabilization for intracellular staining. Flow cytometry was used to measure cellular fluorescence. Results: MTT results revealed a significant decrease in the proliferation of UM-UC-3, J82 and HT-1197 cell lines on treatment with LA. LA also induced reduction in phosphorylation of ERK1/2 in all three carcinoma cell lines. In the mouse model, licochalcone A treatment via intraperitoneal (ip) administration induced a significant decrease in the level of regulatory T cells (Tregs). Comparison of the mouse interferon-α (IFN-α)-treated and LA-treated groups revealed that LA treatment caused enhancement of cytotoxic T lymphocyte (CTL) activity similar to that of IFN-α. Administration of UM-UC-3 cells in C3H/HeN mice resulted in marked reduction in the counts for splenocytes and CD4+ CD25+ Foxp3+ T (regulatory T cells) cell proportion in LA-treated mice compared to untreated control group. Conclusion: Licochalcone A may be of therapeutic importance for the prevention of bladder carcinoma. However, studies are required to ascertain the compound’s usefulness in this regard.

Syftet med detta arbete är att studera vilka olika arbetssätt läraren använder för att introducera, befästa och använda nya begrepp i undervisningen i samhällsorienterande ämnen (SO) samt hur undervisningen uppfattas av elever med språkstörningsdiagnos i en klass i årskurs 4. Utifrån ett specialpedagogiskt perspektiv är fokus att hitta arbetssätt som stöder lärandet för elever med språkstörningsdiagnos. I min studie ingår lärarintervju, lektionsobservationer samt två elevintervjuer. Jag analyserade data utifrån Grundad teori. Analysprocessen var uppbyggd av flera steg och mynnade ut i en egen grundad teoretisk modell med utgångspunkt i mitt syfte och mina frågeställningar. Den teoretiska modellen representeras av sex trappsteg, tydlighet, varierade arbetssätt, repetition, progression, förståelse och förmåga att använda ord och begrepp där det personliga stödet utgör förutsättningen - grunden - för att eleverna med språkstörningsdiagnos ska kunna tillgodogöra sig den klassgemensamma undervisningen. Läraren hade samtalet i helklass som stomme i sin undervisning. Hen använde arbetssätt baserade på forskning: tydliga instruktioner uppdelade i steg, visuellt stöd, elevernas tidigare kunskaper och erfarenheter samt vardagsspråk är utgångspunkt, många tillfällen för repetitioner och uppgifter där eleverna får använda de nya orden och begreppen. Eleverna beskrev att de behövde många repetitioner för att ta till sig nya ord och begrepp. De upplevde de största svårigheterna när många nya begrepp presenterades samtidigt och om de själva skulle söka förklaringar i skriven text. Det bästa sättet att ta till sig ny kunskap var att lyssna, gärna i kombination med bilder av olika slag. De var överens om att Ipad var ett bra verktyg som gav möjlighet att lyssna och tala istället för att läsa och skriva. Studien visar på nödvändigheten av funktionella metoder och uppgifter i klassrummet i kombination med extra anpassningar och personligt stöd utifrån elevens individuella behov.

Increased risk of bleeding after major orthopedic surgery (MOS) has been widely documented in general population. However, this complication has not been studied in elderly patients. The purpose of this study is to determine whether the risk of major bleeding after MOS is higher in elderly patients, compared with those operated at a younger age. Methods: This retrospective cohort study included total hip and total knee arthroplasty patients operated during 5 consecutive years. The main outcome was the occurrence of major bleeding. Patients with other causes of bleeding were excluded. Relative risks (RRs) and confidence intervals (CIs) were calculated, anda multivariate analysis was performed. Results: A total of 1048 patients were included, 56% of patients were hip arthroplasties. At the time of surgery, 553 (53%) patients were older than 70 years. Patients aged >70 years showed an increased risk of major bleeding (RR: 2.42 [95% CI: 1.54-3.81]). For hip arthroplasty, the RR of bleeding was 2.61 (95%CI: 1.50-4.53) and 2.25 (95% CI: 1.03-4.94) for knee arthroplasty. After multivariate analysis, age was found to be independently associated with higher risk of major bleeding. Conclusion: According to European Medicines Agency criteria, patients aged 70 years are at a higher risk of major bleeding after MOS, result of a higher frequency of blood transfusions in this group of patients. Standardized protocols for blood transfusion in these patients are still required.

La presente tesi di Dottorato di Ricerca descrive le attività di ricerca riguardanti diversi ambiti della sicurezza alimentare, della farmacologia e tossicologia veterinaria. Sono presentati i dati relativi alla ricerca di ocratossina A (OTA) in campioni di bile, rene e fegato di polli, salami artigianali e diversi tipi di formaggio; le concentrazioni di diversi metalli pesanti in uova di galline rurali e 5 tipologie di uova industriali; i dati preliminari ottenuti dalle analisi dei primi campioni di uova e penne provenienti da allevamenti amatoriali di pollame, analizzati per la ricerca del fipronil e metaboliti. Sempre per fipronil, e la molecola foxim, sono presentati dati inerenti a studi di potenziale farmaco-resistenza di popolazioni naturali di Dermanyssus gallinae, e dell’attività insetticida di alcuni fitocomposti a base di tannini. Riguardo all’uso di fitoestratti, sono presentati i dati preliminari ottenuti nei primi mesi di una prova sperimentale in galline di razza Livornese allevata con metodo free-range, finalizzata allo studio della attività anti-infiammatoria e immuno-stimolante, nonché alla valutazione della sicurezza della integrazione nella dieta di Boswellia serrata e Salix alba. Sono stati condotti anche studi traslazionali con altri settori scientifici, relativamente la prevalenza sierologica e microbiologica per Salmonella enterica var. enteritidis e var. thiphymurium in allevamenti ornamentali e da auto-consumo di pollame, e di prevalenza e caratterizzazione di potenziali varianti virali e batteriche (incluse zoonosiche) di alcune malattie infettive del pollame ornamentale. In specie avicole ornamentali è riportato anche uno studio di campo sul trattamento off-label con Fenbendazolo e Flubendazolo in infestazioni massive da Cyathostoma bronchialis in oche esotiche ornamentali. A seguito di una collaborazione con l’Università di Firenze, sono presentati anche i risultati ottenuti da uno studio su modelli murini, dell’azione e funzionamento dei recettori Beta3, espressi nelle neoplasie più frequenti che si verificano nelle donne in gravidanza.

The PhD research project was a striking example of the enhancement of milling by-product and alternative protein sources from house cricket (Acheta domesticus), conceived as sustainable and renewable sources, to produce innovative food products. During milling processing of wheat and rye, several by-products with high technological and functional potential, are produced. The use of selected microbial consortia, allowed to obtain a pre-fermented ingredient for use in the bakery. The pre-ferments obtained were characterized by a high technological, functional and nutritional value, also interesting from a nutraceutical point of view. Bakery products obtained by the addition of pre-fermented ingredients were characterized by a greater quantity of aromatic molecules and an increase in SCFA, antioxidant activity, total amino acids and total phenols resulting in positive effect on the functionality. Moreover, the industrial scaling-up of pre-ferment and innovative bakery goods production, developed in this research, underlined the technological applicability of pre-fermented ingredients on a large scale. Moreover, the identification of innovative protein sources, can address the request of new sustainable ingredients able to less impact on the environment and to satisfy the food global demand. To upscale the insect production and ensure food safety of insect-based products, biotechnological formulations based on Acheta domesticus powder were optimized. The use of Yarrowia lipolytica in the biotechnological transformation of cricket powder led to the achievement of a cricket-based food ingredient characterized by a reduced content of chitin and an increase of antimicrobial and health-promoting molecules. The innovative bakery products containing cricket-based hydrolysates from Y. lipolytica possessed specific sensory, qualitative and functional characteristics to the final product. Moreover, the combination of Y. lipolytica hydrolysis and baking showed promising results regarding a reduced allergenicity in cricket-based baked products. Thus, the hydrolysate of cricket powder may represent a versatile and promising ingredient in the production of innovative foods.
