Squamous cell carcinoma (SCC ) is the most common malignancy of the oral cavity (OSCC), with a high mortality rate. Due to this, the discovery of biomarkers that facilitate the understanding of the biological behavior of the tumor and improve treatment is necessary. Urokinase type plasminogen activator (uPA) and its receptor, uPAR, are responsible for the proteolysis of structures of the basement membrana and extracellular matrix, facilitating tumor invasion. This study aims to assess the immuno expression of these proteins in 46 cases of squamous cell carcinoma of the oral tongue (OTSCC). These results were related to the presence of metastasis, clinical TNM staging, locoregional recurrence, outcome of the lesion and histological grading. Immunostaining of each case was evaluated semiquantitatively, in the front of invasion and center of the tumor, in which scores were assigned: 0 (0% of positive cells), 1 (1-10% of positive cells), 2 (11 -50% positive cells) and 3 (more than 50% positive cells). The expression of uPA was observed in 93.5% (n=43) of the cases in the front of invasion, with predominance of score 2 (n=16; 34.8%) and in 67.9% (n=31) of the cases in the center of the tumor, with predominance of score 1 (n=15; 32.6%). Overall, the immunoexpression of uPA was not associated with clinical parameters. Regarding the malignant histological grading, a higher expression of uPA was observed in cases of high-grade malignancy comp ared to low-grade malignancy (p=0.05). Regarding the morphological parameters, increased expression of uPA was observed in the worst mode of invasion (p=0.03 ). The expression of uPAR was observed in 73.9% of cases in the front of invasion, with a predominance of score 1 (n=21; 45.6 %), and in 47.5% (n=21) of the cases in the center of the tumor, with a predominance of score 0 (n=25; 54.4%). Although no statistical differences were observed in relation to lymph node metastasis, clinical TNM staging, outcome, and histological grading, there was a higher expression of uPAR in cases with locoregional recurrence (p=0.04). Regarding the tumor intra -localization, it was observed an increased expression of uPA and uPAR at the front of invasion in relation to the center of the tumor (p<0.001). Regarding the correlation between uPA and uPAR, there was no statistical sign ificance. Based on these results, it is suggested that uPA and uPAR are involved in the progression of CELO, mainly in the deeper region of the tumor.

Angiogenesis and lymphangiogenesis are changes that occur due to gingival inflammation caused by microorganisms present in the biofilm, as well as the migration of immune cells and secretion of mediators in the aggressed site. This study aimed to research angiogenesis and lymphangiogenesis in 90 specimens of clinically healthy, with gingivitis and chronic periodontitis gingival tissue biopsies. The histological sections were evaluated by hematoxylin and eosin and the immunohistochemical technique through immunostaining for CD34 and podoplanin. To evaluate the angiogenic and lymphangiogenic indexes we performed a microvessel counting technique. The results showed that there is a correlation between the indexes (p = 0.030), however, we observed that periodontitis showed less lymphatic vessels than clinically healthy gingival tissue (p = 0.016). Podoplanin showed positive staining in the basal layers of the epithelium, and we observed a relationship between immunostaining intensity and the intensity of inflammatory infiltrate, with more intense staining in the presence of severe inflammatory infiltrate (p = 0.033). For this study, we concluded that there are fewer blood vessels in periodontitis compared with clinically healthy gingiva. The signaling present in the inflammatory process and the actual role of gingival blood and lymphatic vasculature are not fully understood, with further studies on angiogenesis and lymphangiogenesis being suggested.

Brazil is among the largest cashew nut producers of the world. However, the roasting process is still carried out artisanally, especially in the Brazilian semiarid region. In face of this occupational problem, the aim of this study was to perform a physical-chemical characterization of the particulate matter (PM) emitted by the roasting of cashew nuts, as well as to determine the occupational risk and molecular mechanisms associated. The most evident PM characteristics were the prevalence of fine particles, typical biomass burning morphologies such as tar ball and the presence of the elements K, Cl, S, Ca and Fe. In addition, atmospheric modeling analyses suggest that these particles can reach neighboring regions of the emission source. Polycyclic aromatic hydrocarbons (PAHs) with carcinogenic potential, such as benzo[a]pyrene, dibenz[a,h]anthracene, benzo[a]anthracene, benzo[b]fluoranthene, chrysene, benzo[k]fluoranthene, indeno[1,2,3-c,d]pyrene and benzo[j]fluoranthene were the most abundant PAHs found in the two air monitoring campaigns. Among the identified oxy-PAH the benzanthrone (7H-benz[d,e]anthracen-7-one) had the highest concentration and the evaluation of lifetime cancer risk showed an increase of 12 to 37 cases of cancer for every 10,000 exposed people. Chemical analysis of roasted cashew nuts identified the PAHs: phenanthrene, benzo[g,h,i]perylene, pyrene and benzo[a]pyrene, besides the 3-pentadecilfenol allergen (urushiol analogue) as prevalent. Occupational exposure to PAHs was confirmed by the increase of urinary 1-hydroxypyrene levels and genotoxic effects were evidenced by the increase on micronuclei and nuclear bud frequency in exfoliated buccal mucosa cells among the exposed workers. Other biomarkers of effects such as karyorrhexis, pyknotic, karyolytic, condensed chromatin and binucleated cells also have their frequencies increased when compared to an unexposed control group. The investigation of the molecular mechanisms associated with the PM organic extract showed cytotoxicity in human lung cell lines (A549) at concentrations ≥ 4 nM BaPeq. Using non-cytotoxic doses the extract was able to activate proteins involved in the DNA damage response pathway (Chk1 and p53). Moreover, the specific contribution of the four most representative PAHs in the cashew nut roasting sample showed that benzo[a]pyrene was the most efficient to activate Chk1 and p53. Finally, the organic extract was able to increase persistently the mRNA expression involved in the PAHs metabolism (CYP1A1 and CYP1B1), inflammatory response (IL-8 and TNF-α) and cell cycle arrest (CDKN1A) for DNA repair (DDB2). The high PM concentrations and its biological effects associated warn of the serious harmful effects of artisanal cashew nut roasting and urgent actions should be taken to the sustainable development of this activity.

There is a bidirectional association between periodontal disease (PD) and diabetes mellitus, in which diabetes favors the development of PD and PD, if left untreated, can worsen the metabolic control of diabetes. Thus, periodontal disease should be treated to restore periodontal health and reduce the complications of diabetes. Therefore, the objective is assess the effect of full mouth periodontal therapy decontamination (Full Mouth Desinfection - FMD) in diabetic type II patients with chronic periodontitis during 12 months. Thirty-one patients in group one (G1) and 12 in group two (G2) were followed at baseline, 03, 06 09 and 12 months. There following clinical parameters were accessed: probing on bleeding (BOP), visible plaque index (PI), probing depth (PD), clinical attachment level (CAL) and gingival recession (GR). For diabetic patients, there were also made laboratory tests to evaluate blood parameters: fasting glucose and glycated hemoglobin. The results had been analyzed in two ways: all sites in the mouth and another with diseased sites. The Mann-Whitney, Friedman and Wilcoxon tests were used with 5% significance. Intergroup analysis of all sites it is clear that there was no significant difference over time concerning PD, BOP, PI, CAL and RG. However, when evaluating the diseased sites, we observed significant difference for CAL and PD, with higher values in G1. The intragroup analysis for all sites showed a statistically significant reduction at PD, PI and BOP in both groups. Intragroup analysis of periodontal affected sites showed a statistically significant reduction in PD, BOP and CAL in both groups. There was also a statistically significant increase in RG values. There was no significant change concerning glycated hemoglobin and fasting glucose in the G1. Therefore, it can be concluded that there were improvements in periodontal parameters over the 12 months of research, but without changes in glycemic levels of diabetic patients. Thus, periodontal therapy proved effective in maintaining oral health.

CHAPTER II - This study evaluated the effects of two different types of acute aerobic exercise on the osmotic stability of human erythrocyte membrane and on different hematological and biochemical variables that are associated with this membrane property. The study population consisted of 20 healthy and active men. Participants performed single sessions of two types of exercise. The first session consisted of 60 min of moderate-intensity continuous exercise (MICE). The second session, executed a week later, consisted of high-intensity interval exercise (HIIE) until exhaustion. The osmotic stability of the erythrocyte membrane was represented by the inverse of the salt concentration (1/H50) at the midpoint of the sigmoidal curve of dependence between the absorbance of hemoglobin and the NaCl concentration. The values of 1/H50 changed from 2.29 ± 0.1 to 2.33 ± 0.09 after MICE and from 2.30 ± 0.08 to 2.23 ± 0.12 after HIIE. In MICE has occurred an increase in the mean corpuscular volume, probably due to in vivo lysis of older erythrocytes, with preservation of cells that were larger and more resistant to in vitro lysis. The study showed that a single bout of acute exercise affected the erythrocyte osmotic stability, which increased after MICE and decreased after HIIE.

The various types of pig farming, intensive and extensive, expose them to pig parasites but also to those from the environment of the breeding site. In this work was evaluated the exposure of bigs bred in technified farms, SISCAL (intensive breeding system in pens) and not technified (backyard) to leptospira, ticks and rickettsiae. Blood sera were analyzed to determine titers of antibodies anti-Leptospira by SAM technique and antibodies anti-rickettsial by IFA, pigs were inspected for ticks and in their breeding environment and surrounding areas (pastures and riparian vegetation), ticks were collected by the flannel dragging technique. In the farms of pigs 10.4% had anti-Leptospira antibodies, followed by SISCAL (8%) and backyard animals (2.5%). The serovars found were Bratislava, Pomona, serovar, Canicola and Icterohaemorrhagiae. Higher percentage of properties with pigs raised outdoors (SISCAL) had tick infested animals (20%) than those raised in backyard (6.7%), while commercial farms had no infested pigs nor infested breeding place. In both SISCAL and backyard pig breeding properties ticks were observed at the breeding site environment. Tick infestations were detected in areas surrounding pig breeding site in all three husbandry suystems. Ticks found were all Amblyomma scultpum nymphs or adults with the exception of one of Amblyomma parvum adult. In relation to anti-rickettsia serology to five Rickettsia species, 55.2% of pigs from commercial farms reacted to al least one species, backyard pigs reacted to 89.7% and all pigs of SISCAL showed anti-rickettsia titers. Consecutive tick sampling (June 2014 to February 2016) in SISCAL FAZU in Uberaba, showed the establishment A. sculptum ticks maintained by domestic pigs. These observations demonstrate the ability the pigs to maintain populations of A. sculptum at a favorable environment and may indicate a new trend in environmental infestations by this species of tick. Exposure to Leptospira and Rickettsia demonstrated the potential pigs exposure and transmission of important diseases in public health.

Chagas disease, caused by the parasite Trypanosoma cruzi, is the cause of Chronic chagasic cardiomyopathy (CCC). The prospection of innovative therapeutic agents against CCC is a major task. The recombinant form of 21 (rP21), a secreted T. cruzi protein involved in host cell invasion and on progression of chronic inflammatory processes have been studied as a potential novel therapeutic target. Our present work aimed to verify and investigate the impact of rP21 in the formation of blood vessels in vitro and in vivo. First, tEnd cells were treated with different concentrations of rP21 or bacterial extract and viability and cellular adhesion were evaluated by MTT and angiogenesis inhibition by Matrigel tube formation assay and murine model. To verify the proteolytic activity of rP21 on extracellular matrix (ECM) components, fibrinogen, matrigel and fibronectin was incubated with rP21 or not. In addition, we performed proliferation assays and cell cycle analysis. Furthermore, the accumulation and distribution of F-actin was determined by Phalloidin staining using ImageJ software. Finally, tEnd cells were incubated with rP21 and the mRNA levels were analyzed by real-time PCR. Our results showed that rP21 did not alter cell viability and adhesion, but strongly inhibited vessel formation in vitro and in vivo. Tube formation assay showed that angiogenesis inhibition was dependent of the CXCR4-rP21 binding. In addition to these results, we observed that the rP21 was able to inhibit cell proliferation and promoted a significant reduction in the number of 4n cells (G2/M phase). Moreover, we found that rP21 significantly increased F-actin levels and this protein was able to modulate expression of genes related to angiogenesis and actin cytoskeleton. However, rP21 showed no significant activity on the matrix components. In this sense, we conclude that the rP21-endothelial cells (ECs) interaction via CXCR4 promotes inhibition of vessel formation through a cascade of intracellular events, such as inhibition of ECs proliferation and modulation of the expression of molecules associated with angiogenic processes and actin cytoskeleton.

La sincronización de las oscilaciones cerebrales se produce incluso en ausencia de tarea, por eso, el resting state está aportando interesantes vías de estudio de los procesos normales y patológicos. Dada la creciente necesidad por utilizar las medidas derivadas de las señales MEG en resting state como biomarcadores clínicos o en la evaluación de tratamientos, es necesario garantizar su fiabilidad. En este estudio se ha investigado por primera vez la fiabilidad de la las medidas espectrales derivadas de registros MEG explorando la estabilidad en resting state de la potencia de 10 sujetos sanos en tres sesiones con un intervalo test-retest de 7 días. A partir de las señales MEG de cada sujeto y sesión se calculó el espectro de potencia de 1 a 100Hz en cada sensor, y como medida de fiabilidad se utilizó el coeficiente de correlación intraclase (ICC). Para explorar cómo afecta la intensidad de la señal a la estabilidad, se registró la señal de la cámara vacía en cada sesión de registro y se calculó la relación señal/ruido (SNR). La potencia espectral en MEG es muy estable en las bandas de frecuencia α, β y θ, y menos estable en δ y γ-2. Con respecto a la distribución de la estabilidad, la señal capturada en la zona frontal del equipo MEG fue la menos estable a través de todas las bandas de frecuencia. La estabilidad mostró cierta tendencia a disminuir conforme disminuye la SNR; este efecto es parcial, ya que los ritmos cerebrales estables mostraron un alto ICC incluso con baja SNR. En conjunto, estos resultados sugieren que las medidas espectrales en resting state con MEG son suficientemente fiables para ser consideradas en futuros estudios longitudinales sobre cambios en la actividad cerebral.

Os ecossistemas marinhos estão continuamente a ser sobrecarregados com contaminantes derivados de atividades humanas resultando numa dimunuição dos recursos marinhos. A exposição crónica a contaminantes, como metais pesados e poluentes orgânicos persistentes (POPs), pode afetar negativamente o ambiente marinho e, eventualmente, também os seres humanos. Grandes predadores pelágicos, como tubarões, são particularmente afetados pela poluição, principalmente através de processos de bioacumulação e biomagnificação. A fim de resolver o problema acima mencionado, são necessários estudos de avaliação de risco ambiental para prever os padrões de contaminação e evitar efeitos adversos que muitas vezes só são visíveis quando é tarde demais para tomar ações preventivas. Análises de concentração de químicos fornecem-nos informações sobre o nível de contaminação no ambiente; no entanto, podem não ser suficientes para entender como os organismos estão a ser afetados e há uma necessidade de relacionar essas quantificações com parâmetros biológicos. A avaliação de parâmetros bioquímicos, como a atividade enzimática, pode fornecer uma visão mais sensível e precisa sobre os níveis de contaminação. Tubarões como Prionace glauca são predadores de topo e portanto extremamente importantes nos ecossistemas marinhos. A sua grande distribuição, juntamente com o fácil acesso a amostras, fornecidas por barcos de pesca comercial, tornou-as um alvo favorável para utilização em ensaios toxicológicos. Este estudo teve como objetivo avaliar o potencial de P. glauca como uma espécie sentinela para pesquisas de monitorização de poluição, através do desenvolvimento e da aplicação de biomarcadores apropriados. As amostras de tecidos foram recolhidas de vinte tintureiras na costa de Portugal, a bordo de um barco comercial de pesca de espadarte. Níveis de POPs, assim como parâmetros bioquímicos relacionados com destoxificação, stress oxidativo e funções neuronais, foram medidos. A caracterização prévia da atividade das colinesterases no músculo e cérebro de P. glauca foi feita, já que não havia dados disponíveis sobre esta matéria. Esta caracterização foi essencial devido à existência de três classes de ChE conhecidas em peixes, acetilcolinesterase (AChE), butirilcolinesterase (BChE) e propionilcolinesterase (PChE), todas bastante suscetíveis a agentes anticolinérgicos, e outros contaminantes, tornando-as biomarcadores relevantes em estudos de monitorização de poluição. Os resultados obtidos indicaram que o cérebro de P. glauca aparenta possuir ChEs atípicas, revelando propriedades mistas de AChE e BChE e, que o músculo aparentemente possui maioritariamente AChE. A exposição in vitro a chloropyrifos-oxon provocou inibição de ChE das tintureiras em ambos os tecidos, com o cérebro sendo o tecido mais sensível e, por isso, o mais adequado para a detecção de compostos anticolinérgicos no ambiente. Este estudo indica que a actividade de ChE em tintureiras tem potencial para ser usada como um biomarcador sensível e fiável em programas de biomonitorização marinha. O fígado apresentou níveis mais elevados de POP, quando comparado com músculo. Foram encontradas correlações positivas e negativas entre os parâmetros de contaminação e de stresse oxidativo. Este estudo destaca a importância da caracterização de Che antes de a usar como um biomarcador em estudos ecotoxicológicos, e demonstra o grande potencial de P. glauca como espécie modelo e como sentinela de poluição marinha, através do uso de biomarcadores adequados.

Os Oceanos representam o maior sistema de suporte de vida sendo a uma grande fonte de riqueza, oportunidade e abundância. No entanto, a humanidade tem levado este ecossistema ao seu limite com crescentes níveis de poluição e outras pressões antropogénicas. A introdução de espécies não-nativas é reconhecida como uma das maiores ameaças à biodiversidade e a segunda maior causa de extinção das espécies. A macroalga vermelha Asparagopsis armata é uma espécie invasora originária da Austrália e que atualmente apresenta uma ampla distribuição em todo o globo devido à sua estratégia oportunista, ausência de predadores e altas taxas de crescimento. Uma questão emergente está relacionada com a capacidade destas espécies invasoras produzirem grandes quantidades de metabolitos halogenados potencialmente tóxicos. Esta característica pode representar um perigo adicional para o equilíbrio ecológico da comunidade invadida. O presente trabalho teve como objetivo avaliar o potencial ecotoxicológico dos exsudatos de A. armata usando um gastrópode, Gibbula umbilicalis, como organismo modelo. A macroalga recolhida na costa de Peniche (Portugal) foi colocada em tanques no laboratório, durante 12 h, sendo depois o meio recolhido e filtrado para ensaios posteriores com os exsudatos da alga. No ensaio agudo, observou-se a mortalidade de G. umbilicalis que foi exposta a crescentes diluições do exsudato durante 96 h. Adicionalmente, os gastrópodes foram expostos a concentrações não letais do exsudato e analisou-se as respostas bioquímicas recorrendo a biomarcadores relacionados com destoxificação, defesas antioxidantes, danos oxidativos, danos neurotóxicos e metabolismo energético. Os resultados revelaram que os exsudatos de A. armata afetaram significativamente a sobrevivência dos organismos expostos com uma CL50 96h de 5.03% de exsudato da alga. A exposição aos exsudatos da alga também resultou em efeitos bioquímicos e metabólicos ao nível subcelular com resultados significativos na inibição da glutationa-S-transferase (GST), perda de integridade do ADN e níveis crescentes de atividade da lactato desidrogenase (LDH), dando uma indicação dos mecanismos de toxicidade desta alga marinha. Os níveis mais elevados de danos no ADN ocorreram quando a GST apresentou os níveis mais baixos de atividade e esta mesma atividade aumentou quando os danos no ADN diminuíram, em simultâneo com o aumento dos níveis de atividade da LDH, indicando que as necessidades energéticas aumentam devido à necessidade de sintetizar mais enzima. Conclui-se que a A. armata tem capacidade de libertar substâncias tóxicas que podem ter potenciais impactos no ambiente envolvente. Adicionalmente, as respostas bioquímicas estudadas em G. umbilicalis têm potencial para serem usadas como sinais de aviso na determinação dos efeitos provocados pelos compostos libertados por esta macroalga vermelha.

The aim of this study was to determine the haematological value and biochemical blood in baby alpacas with enteric disorder. A total of 30 blood and serum samples were collected from alpacas of 1 month old with diarrhoea and 5 blood samples of clinically healthy baby alpacas (controls). The animals were from communities in the central Andes from Peru. About haematology were determined haematocrit, haemoglobin concentration, red blood count and white blood count that were not significantly different between control animals and animals with diarrhoea. Moreover, biochemical blood parameters as total protein, albumin and calcium decrease significantly (p<0.05). We conclude that our results could be considered as factors in the mortality of baby alpaca by infectious diarrhoea.

Introdução: A neurofibromatose tipo I (NF1), também designada doença de Von Recklinghausen, é causada por uma anormalidade no cromossoma 17, de transmissão autossómica dominante, responsável pela produção deficiente de neurofibromina. Caracteriza-se por displasia nos tecidos mesodérmicos e neuroectodérmicos, e tem uma incidência de um para 2.500-3.300 nascimentos. A presença de manchas café-au-lait, neurofibromas cutâneos e hamartomas da íris são sinais cardinais da doença. Primeiramente descrita por Ruebi em 1945, a patologia vascular é uma complicação subestimada e pouco reconhecida na NF1. A ocorrência de hemorragia fatal ou quase fatal está reportada ocasionalmente nas cavidades pleural, abdominal, retroperitoneu, tecidos moles do tronco e extremidades. Esta hemorragia massiva é causada pela rutura de vasos sanguíneos friáveis, característicos pelos níveis reduzidos de neurofibromina e consequente proliferação endotelial e de músculo liso nas artérias e veias. Uma das consequências clínicas mais sérias descritas na NF1 é a ocorrência de hemorragia severa e dificuldade em alcançar controlo hemostático. Objetivo: Exposição de caso clínico de extenso hematoma cervical e hemotórax espontâneos por rutura troncos venosos braquiocefálico, veia subclávia e junção subclávio-jugular em doente com NF1. Caso clínico: Relata-se caso clínico de mulher de 51 anos, com antecedentes conhecidos de NF1 e hipertensão arterial. Foi admitida no serviço de urgência em choque hipovolémico hemorrágico, no contexto de dor súbita no ombro direito e volumosa tumefação cervical direita. Em angioTC foi objetivado volumoso hematoma, envolvendo a região cervical direita, a região retrofaríngea-prevertebral, escavado supraclavicular direito, mediastino, associando a importante hemotórax direito. Procedeu-se a abordagem supraclavicular com drenagem do hematoma e identificação de fontes hemorrágicas, nomeadamente: tronco venoso braquicefálico, veia subclávia e confluência subclávio-jugular. Foi necessária secção da clavícula para controlo hemorrágico e realização de rafias dos troncos venosos com prolene. Intraoperatoriamente, foi evidente a fragilidade e friabilidade excessiva dos vasos sanguíneos. Após controlo hemorrágico, foi realizada videotoracoscopia para drenagem de hemotórax e evacuação de coágulos, confirmando-se a ausência de hemorragia ativa. No pós-operatório a doente recuperou estabilidade hemodinâmica, sem evidência analítica de queda de hemoglobina. Realizou-se angioTC, onde se confirmou franca melhoria do hematoma cervical direito e retrofaríngeo em critérios quantitativos, e ausência de extravasamento de contraste. Objetivou-se adicionalmente aneurismas acular da artéria vertebral direita, corrigido ulteriormente através de embolização com coils. Conclusão: A NF1 é uma doença genética que raramente se pode associar a hemorragia life- -threatening. A vasculopatia é uma complicação subestimada e pouco reconhecida na NF1. A existência de friabilidade vascular excessiva com consequente hemorragia espontânea na NF1 é rara e pode ser fatal, exigindo um diagnóstico rápido e tratamento atempado.

O osteossarcoma ou sarcoma osteogénico (OS) é o tumor maligno primário mais comum de origem óssea que ocorre em crianças e adultos jovens. Este tumor surge a partir de células mesenquimais e é histologicamente caracterizado pela presença de células fusiformes e formação aberrante de osteóide. A incidência desta neoplasia apresenta uma distribuição bimodal, com um primeiro pico na adolescência e um segundo pico a ocorrer entre a 6ª e a 7ª década de vida. A maioria dos pacientes apresenta doença localizada à data do diagnóstico, e destes, aproximadamente 15 a 20% apresentam metástases detectáveis, em especial, pulmonares. A abordagem terapêutica ao OS engloba quimioterapia pré-operatória (neoadjuvante) seguida de ressecção cirúrgica do tumor primário e de todas as metástases visíveis e de quimioterapia pós-operatória (adjuvante). Historicamente, muitos agentes foram testados e usados no tratamento do OS, mas desde os anos 90 até à actualidade, o esquema de tratamento baseia-se na utilização de três agentes: Metotrexato (MTX), Doxorrubicina (A) e Ciclofosfamida (P), podendo ou não ser incluído um 4º agente, a Ifosfamida (I) ou o Ectopósido (E). Para surtir efeito, o MTX é administrado em alta dose, habitualmente de 12g/m2, podendo ir até 20g/m2, apresentando vários efeitos adversos. Como tal, outros agentes quimioterápicos têm sido testados, assim como outros tipos de tratamento e alvos, com o intuito de melhorar a performance e a sobrevida sem recorrência. A identificação de biomarcadores que permitam o diagnóstico, prognóstico e monitorização da terapia tem sido outra das principais linhas de investigação relacionadas com o OS. O objetivo deste trabalho centra-se na revisão da importância do tratamento neoadjuvante enquanto fator prognóstico do OS, e da monitorização laboratorial na utilização do MTX por forma a evitar efeitos secundários.
