Bothrops jararacussu myotoxin I (BthTx-I; Lys 49) and II (BthTX-II; Asp 49) were purified by ion-exchange chromatography and reverse phase HPLC. In this work we used the isolated perfused rat kidney method to evaluate the renal effects of B. jararacussu myotoxins I (Lys49 PLA(2)) and II (Asp49 PLA(2)) and their possible blockage by indomethacin. BthTX-1 (5 mu g/ml) and BthTX-II (5 mu g/ml) increased perfusion pressure (PP; ct(120) = 110.28+/-3.70 mmHg; BthTX I = 171.28+/-6.30* mmHg; BthTX II = 175.50+/-7.20* mmHg), renal vascular resistance (RVR; ct(120) = 5.49+/-0.54 mmHg/ml.g(-1) min(-1); BthTX I = 8.62+/-0.37* mmHg/ml g(-1) min(-1); BthTX II=8.9+/-0.36* mmHg/ml g(-1) min(-1)), urinary flow (UF; ct(120)= 0.14+/-0.01 ml g(-1) min(-1); BthTX I=0.32+/-0.05* ml g(-1) min(-1); BthTX II=0.37+/-0.01* ml g(-1) min(-1)) and glomerular filtration rate (GFR; ct(120)=0.72+/-0.10 ml g(-1) min(-1); BthTX I=0.85+/-0.13* ml g(-1) min(-1); BthTX II=1.22+/-0.28* ml g(-1) min(-1)). In contrast decreased the percent of sodium tubular transport (%TNa+; ct(120)=79,76+/-0.56; BthTX I=62.23+/-4.12*; BthTX II=70.96+/-2.93*) and percent of potassium tubular transport (%TK+;ct(120)=66.80+/-3.69; BthTX I=55.76+/-5.57*; BthTX II=50.86+/-6.16*). Indomethacin antagonized the vascular, glomerular and tubular effects promoted by BthTX I and it's partially blocked the effects of BthTX II. In this work also evaluated the antibacterial effects of BthTx-I and BthTx-II against Xanthomonas axonopodis. pv. passiflorae (Gram-negative bacteria) and we observed that both PLA2 showed antibacterial activity. Also we observed that proteins Also we observed that proteins chemically modified with 4-bromophenacyl bromide (rho-BPB) decrease significantly the antibacterial effect of both PLA(2). In conclusion, BthTx I and BthTX II caused renal alteration and presented activity antimicrobial. The indomethacin was able to antagonize totally the renal effects induced by BthTx I and partially the effects promoted by BthTx II, suggesting involvement of inflammatory mediators in the renal effects caused by myotoxins. In the other hand, other effects could be independently of the enzymatic activity of the BthTX II and the C-terminal domain could be involved in both effects promoted for PLA(2). Bothrops insularis venom contains a variety of substances presumably responsible for several pharmacological effects. We investigated the biochemical and biological effects of phospholipase A(2) protein isolated from B. insularis venom and the chromatographic profile showed 7 main fractions and the main phospholipase A(2) (PLA(2)) enzymatic activity was detected in fractions IV and V. Fraction IV was submitted to a new chromatographic procedure on ion exchange chromatography, which allowed the elution of 5 main fractions designated as lV-1 to IV-5, from which lV-4 constituted the main fraction. The molecular homogeneity of this fraction was characterized by high-performance liquid chromatography (HPLC) and demonstrated by mass spectrometry (MS), which showed a molecular mass of 13984.20 Da; its N-terminal sequence presented a high amino acid identity (up to 95%) with the PLA(2) of Bothrops jararaca and Bothrops asper. Phospholipase A(2) isolated from B. insularis (Bi PLA(2)) venom (10 mu g/mL) was also studied as to its effect on the renal function of isolated perfused kidneys of Wistar rats (n = 6). Bi PLA(2) increased perfusion pressure (PP), renal vascular resistance (RVR), urinary flow (UF) and glomerular filtration rate (GFR). Sodium (%TNa+) and chloride tubular reabsorption (%TCl-) decreased at 120 min, without alteration in potassium transport. New bone chars for fluoride adsorption from drinking water have been synthetized via metallic doping using aluminum and iron salts. A detailed statistical analysis of the metal doping process using the signal-to-noise ratios from Taguchi's experimental designs and its impact on the fluoride adsorption properties of modified bone chars have been performed. The best conditions, including the proper metallic salt, for metal doping were identified to improve the fluoride uptakes of modified bone chars. Results showed that the fluoride adsorption properties of bone chars can be enhanced up to 600% using aluminum sulfate for the surface modification. This aluminum-based adsorbent showed an adsorption capacity of 31 mg/g, which outperformed the fluoride uptakes reported for several adsorbents. Surface interactions involved in the defluoridation process were established using FTIR, DRX and XPS analysis. Defluoridation using the metal-doped bone chars occurred via an ion exchange process between fluoride ions and the hydroxyl groups on the adsorbent surface, whereas the Al(OH)xFy, FexFy, and CaF2 interactions could play also an important role in the removal process. These metal-doped adsorbents anticipate a promising behavior in water treatment, especially in developing countries where the efficiency – cost tradeoff is crucial for implementing new defluoridation technologies.

O objetivo deste trabalho foi avaliar o efeito da utilização de diferentes manejos alimentares: alimento natural, ração peletizada, extrusada ou farelada, sobre a qualidade da água dos efluentes gerados em uma criação de tilápia do Nilo (Oreochromis niloticus). O experimento foi desenvolvido durante 19 semanas em doze viveiros de 300 m², com renovação contínua de água, povoados com juvenis machos de tilápia do Nilo na densidade de 1,7 peixes m-2. As rações isoproteícas (30% de proteína bruta) e isoenergéticas (3.000kcal de energia digestível) foram fornecidas duas vezes ao dia. Quanto ao tratamento alimento natural, foi utilizado esterco de galinha poedeira. Semanalmente, foram aferidos na água de abastecimento e nos efluentes, temperatura, oxigênio dissolvido, pH, fósforo total, nitrogênio total, clorofila a e material em suspensão. de maneira geral, houve piora na qualidade da água dos efluentes de todos os tratamentos estudados, em comparação a água de abastecimento, evidenciando o impacto ambiental desta atividade produtiva, podendo levar a eutrofização dos corpos d'água receptores.

Soil degradation affects more than 52 million ha of land in counties of the European Union. This problem is particularly serious in Mediterranean areas, where the effects of anthropogenic activities (tillage on slopes, deforestation, and pasture production) add to problems caused by prolonged periods of drought and intense and irregular rainfall. Soil microbiota can be used as an indicator of the soil healthy in degraded areas. This is because soil microbiota participates in the cycle elements and in the organic matter decomposition. All this helps to the young plants establishment and in long term protect the soils against the erosion. During dry periods in the Mediterranean areas, the lack of water entering the soil matrix leads to a loss of soil microbiological activity and it turns into a lower soil production capabilities. Under these conditions, the aim of this study was to evaluate the positive effect on soil biological components produced by an hydro absorbent polymer (Terracottem). The aim of the experiment was to evaluate the impact assessment of an hydropolymer (Terracottem) on the soil biological components. An experimental flowerpot layout was established in June 2015 and 12 variants with different amount of Terracottem were applied as follow: i) 3.0 kg.m3 ; ii) 1.5 kg.m3 and; iii) 0 kg.m3. In all the variants were tested the further additives: a) 1% of glucose, b) 50 kg N.ha-1 of Mineral nitrogen, c) 1% of Glucose + 50 kg N.ha-1 of Mineral nitrogen d) control (no additive). According to natural conditions, humidity have been kept at 15% in all the variants. During four weeks, mineral nitrogen leaching and soil respiration have been measured in each flowerplot. Respiration has been quantified four times every time while moistening containers and alkaline soda lime has been used as a sorbent. The amount of CO 2 increase has been measured with the sorbent. Leaching of mineral nitrogen has been quantified by ion exchange resins (IER). IER pouches have been placed on the bottom of each container, and after completion of the experiment mineral nitrogen leaching has been evaluated by distillation and titration method. Results from respiration have shown statistically significant differences between the variants. According to control, soil with polymers have shown significant difference when comparing respiration with independence of the additive used. CO 2 production in the first week has exceeded the sum of the outputs of the following weeks. Mineral nitrogen leaching measurement has shown statistically significant differences. The lowest leaching has been occurred in control variant, while the highest in variant containing only the addition of mineral nitrogen. Research results may conclude that the biological part of the test soil is not limited by a lack of components, the only thing that suppresses its activity is the lack of moisture. After moistening it leads to a rapid growth of soil activity, without causing the nutrients loss. Besides, Terracottem has affected soil activity neither positively nor negatively, but it considers being a suitable tool for reducing the drought impact in arid and semi-arid areas.

-D-glucosidase (EC 3.2.1.21) is one of the most interesting glycosidases, especially for hydrolysis cellobiose releasing glucose, is last step degradation of cellulose. This function makes the -D-glucosidase is of great interest as a versatile industrial biocatalyst, being critical to various bio-treatment / biorefinery processes, such as bioethanol production. Hen in the report, a -D-glucosidase was extracts from protein extracted of the invertebrate marine Artemia franciscana was purified and characterized with a combination of precipitation with ammonium sulfate (0 - 30%, 30 to 50%, 50 to 80%), the fraction saturated in the range of 30 to 50% (called F-II) was applied in a molecular exclusion chromatography, in Sephacryl S-200, the fractions corresponding to the first peak of activity of -D-glucosidase were gathered and applied in a chromatography of ion exchange in Mono Q; the third peak this protein obtained chromatography, which coincides with the peak of activity of -D-glucosidase was held and applied in a gel filtration chromatography Superose 12 where the first peak protein, which has activity of -D-glucosidase was rechromatography on Superose 12. This enzyme is probably multimerica, consisting of three subunit molecular mass of 52.7 kDa (determined by SDS-PAGE) with native molecular mass of 157 kDa (determined by gel filtration chromatography on Superose 12 under the system FPLC). The enzyme was purified 44.09 times with a recovery of 1.01%. Using up p-nitrophenyl-β-D-glucopiranoside as substrate obtained a Km apparent of 0.229 mM and a Vmax of 1.109 mM.60min-1.mL-1mM. The optimum pH and optimum temperature of catalysis of the synthetic substrate were 5.0 and 45 °C, respectively. The activity of the -D-glucosidase was strongly, inhibited by silver nitrate and N- etylmaleimide, this inhibition indicates the involvement of radical sulfidrila the hydrolysis of synthetic substrate. The -D-glucosidase of Artemia franciscana presented degradativa action on celobiose, lactose and on the synthetic substrate -nitrophenyl-β-D-glucopiranoside indicating potential use of this enzyme in the industry mainly for the production of bioethanol (production of alcohol from the participating cellulose), and production hydrolysate milk (devoid of milk lactose)

Les éléments de terres rares (REEs) sont de plus en plus utilisés dans une multitude d’applications, notamment la fabrication d’aimants, de batteries rechargeables et les écrans de téléviseurs. Ils sont pour la plupart des métaux trivalents peu solubles dans les eaux naturelles. Comme pour les métaux divalents, le risque écologique des REEs est très probablement étroitement lié à leurs spéciations chimiques. Cependant, le comportement du samarium (Sm) dans les matrices environnementales est très peu connu et il n'existe actuellement aucune technique pour évaluer sa spéciation chimique. Dans cette optique, la technique d'échange d'ions (IET) sur la résine Dowex a été optimisée pour mesurer le samarium libre en solution. Les temps d'équilibre ont d'abord été déterminés pour des solutions tamponnées de samarium (Sm 6,7x10-8 M ; MES 1,0 mM M ; pH 6,0) en présence du nitrate de sodium (de 0,01M à 0,5 M). Pour ces diverses forces ioniques, l’équilibre thermodynamique n’est atteint que pour NaNO3 0,5M. Un autre mode d’utilisation de la résine (mode dynamique) a donc été développé pour tenir compte des conditions environnementales et évaluer efficacement le samarium libre. Les impacts des ligands organiques tels le NTA, l’EDTA, le citrate, l’acide malique et l’acide fulvique Suwannee River Standard I (SRFA) ont été étudiés par l’IET en mode dynamique. Une grande corrélation a été trouvée plus entre les taux d’accumulation de samarium sur la résine d’échange pour différents rapports NTA : Sm, EDTA : Sm, SRFA : Sm et le samarium libre. Par contre, aucune corrélation significative n’a été observée pour les ligands citrate et acide malique compte tenu des complexes qu’ils forment avec le samarium et qui s’adsorbent aussi sur la résine Dowex. Les concentrations Sm3+ mesurées par la technique IET ont été fortement corrélées avec celles prédites par le modèle WHAM 7.0 en utilisant la constante de stabilité obtenue par titration de SRFA par extinction de la fluorescence. Par ailleurs, la formation de colloïdes de samarium en fonction du pH influe grandement sur la détermination du samarium libre et doit être prise en compte dans la spéciation du samarium. L'IET assisté par des techniques auxiliaires comme le dosage par extinction de la fluorescence et le SP-ICPMS pourrait être une technique utile pour évaluer les concentrations de Sm biodisponible dans les eaux naturelles.

Protein purification plays a crucial role in biotechnology and biomanufacturing, where downstream unit operations account for 40%-80% of the overall costs. To overcome this issue, companies strive to simplify the separation process by reducing the number of steps and replacing expensive separation devices. In this context, commercially available polybutylene terephthalate (PBT) melt-blown nonwoven membranes have been developed as a novel disposable membrane chromatography support. The PBT nonwoven membrane is able to capture products and reduce contaminants by ion exchange chromatography. The PBT nonwoven membrane was modified by grafting a poly(glycidyl methacrylate) (GMA) layer by either photo-induced graft polymerization or heat induced graft polymerization. The epoxy groups of GMA monomer were subsequently converted into cation and anion exchangers by reaction with either sulfonic acid groups or diethylamine (DEA), respectively. Several parameters of the procedure were studied, especially the effect of (i) % weight gain and (ii) ligand density on the static protein binding capacity. Bovine Serum Albumin (BSA) and human Immunoglobulin G (hIgG) were utilized as model proteins in the anion and cation exchange studies. The performance of ion exchange PBT nonwovens by HIG was evaluated under flow conditions. The anion- and cation- exchange HIG PBT nonwovens were evaluated for their ability to selectively adsorb and elute BSA or hIgG from a mixture of proteins. Cation exchange nonwovens were not able to reach a good protein separation, whereas anion exchange HIG nonwovens were able to absorb and elute BSA with very high value of purity and yield, in only one step of purification.

In most agroecosystems, nitrogen (N) is the most important nutrient limiting plant growth. One management strategy that affects N cycling and N use efficiency (NUE) is conservation agriculture (CA), an agricultural system based on a combination of minimum tillage, crop residue retention and crop rotation. Available results on the optimization of NUE in CA are inconsistent and studies that cover all three components of CA are scarce. Presently, CA is promoted in the Yaqui Valley in Northern Mexico, the country´s major wheat-producing area in which from 1968 to 1995, fertilizer application rates for the cultivation of irrigated durum wheat (Triticum durum L.) at 6 t ha-1 increased from 80 to 250 kg ha-1, demonstrating the high intensification potential in this region. Given major knowledge gaps on N availability in CA this thesis summarizes the current knowledge of N management in CA and provides insights in the effects of tillage practice, residue management and crop rotation on wheat grain quality and N cycling. Major aims of the study were to identify N fertilizer application strategies that improve N use efficiency and reduce N immobilization in CA with the ultimate goal to stabilize cereal yields, maintain grain quality, minimize N losses into the environment and reduce farmers’ input costs. Soil physical and chemical properties in CA were measured and compared with those in conventional systems and permanent beds with residue burning focusing on their relationship to plant N uptake and N cycling in the soil and how they are affected by tillage and N fertilizer timing, method and doses. For N fertilizer management, we analyzed how placement, time and amount of N fertilizer influenced yield and quality parameters of durum and bread wheat in CA systems. Overall, grain quality parameters, in particular grain protein concentration decreased with zero-tillage and increasing amount of residues left on the field compared with conventional systems. The second part of the dissertation provides an overview of applied methodologies to measure NUE and its components. We evaluated the methodology of ion exchange resin cartridges under irrigated, intensive agricultural cropping systems on Vertisols to measure nitrate leaching losses which through drainage channels ultimately end up in the Sea of Cortez where they lead to algae blooming. A throughout analysis of N inputs and outputs was conducted to calculate N balances in three different tillage-straw systems. As fertilizer inputs are high, N balances were positive in all treatments indicating the risk of N leaching or volatilization during or in subsequent cropping seasons and during heavy rain fall in summer. Contrary to common belief, we did not find negative effects of residue burning on soil nutrient status, yield or N uptake. A labeled fertilizer experiment with urea 15N was implemented in micro-plots to measure N fertilizer recovery and the effects of residual fertilizer N in the soil from summer maize on the following winter crop wheat. Obtained N fertilizer recovery rates for maize grain were with an average of 11% very low for all treatments.
