We are witnessing nothing less than a revolution in international policy-thinking, with a shift from imagining that international policy-makers can solve development/ security problems through the export or transfer of policy practices or their imposition through conditionality, to understanding that problems should be grasped as emergent consequences of complex social processes which need to be worked with rather than against. This paper, prepared for the 2014 CEPA conference, focuses therefore less on the politicisation and securitisation of questions of conflict and poverty and more on the depoliticisation of questions of conflict and poverty, especially through frameworks of resilience.

The relationship between population and government in the City of Buenos Aires is analyzed, focusing on the tensions generated by the arrival of new individuals, a local elite with great interests in commerce and in charge of community affairs, and Spanish functionaries that progressively adopted the ideals of the Bourbon Dynasty, despite also being implicated in local logics. It interests us to observe the governors’ perspectives with respect to everyday developments in the city, their preoccupations and interests, and how they would vary the mechanisms to which they resorted in order to organize daily life, in the period defined between 1740 and 1776.

El procedimiento consiste en la. fermentación del zumo de naranja natural mediante dos levaduras, que se adicionan al zumo secuencialmente, dando lugar a dos diferentes fases en las que se separa la producción de productos aromáticos y saborizantes de la formación de etanol. Para la primera fase se utiliza una cepa seleccionada de la levadura Pichia fermentans, mientras que en la segunda fase se utiliza una cepa de la levadura Saccharomyces cerevisiae. Alternativamente, la fermentación se lleva a cabo en una sola fase con la levadura Pichia fermentans sustituyéndose la segunda fase por una incorporación directa del etanol.

Una nueva cepa de Scenedesmus ha sido aislada en la estación experimental "Las Palmerillas-CAJAMAR", siendo identificada como "microorganismo no previamente registrado" por la Universidad de Gottinghem, y depositada en la Culture Collection of Alagae and Protozoa (CCAP) como Scenedesmus almeriensis. La nueva cepa puede ser utilizada para consumo animal y/o humano, y produce elevadas cantidades de carotenoides, especialmente luteína y betacaroteno. Scenedesmus almeriensis crece adecuadamente en un amplio rango de temperaturas de 10°C a 40°C, a un pH de 7.0 a 9.5, y es tolerante a elevadas concentraciones de cobre, de hasta 1 mg/L. La cepa microalgal, cultivada en un fotobiorreactor de 4000 L en el que el cultivo se impulsa mecánicamente, produce luteína en una cantidad de al menos 4 mg por gramo de materia seca. Esta cepa es muy adecuada para la producción de carotenoides de aplicación en el tratamiento de desordenes de la macular ocular.

Una cepa de la microalga marina isochrysis galbana, depositada en la CCAP con el número de depósito CCAP 927/15 es capaz de producir elevadas cantidades de ácidos grasos poliinsaturados, especialmente de ácido eicosapentaenoico (EPA) y de ácido docosahexaenoico (DHA). La cepa microalgal crece adecuadamente a una temperatura de 18 c a 25 c, en un ph de 7 a 9,5, preferentemente a un ph de 7,65 a 8,00. La cepa microalgal, cultivada a 20 c en un fermentador de 5 litros agitado por paletas y con iluminación continua, produce EPA en una cantidad de, al menos, 39,5 mg por gramo de materia seca. Esta cepa es adecuada para su empleo en acuicultura (alimentación de larvas de peces y moluscos) y en la obtención de EPA y/o de un aceite rico en EPA y en DHA necesarios para la nutrición y salud humanas.

Los hongos se caracterizan por su capacidad de producción y secreción de enzimas al medio externo. Las especies del género Mucor son microorganismos importantes en la producción de proteasas. En este trabajo se investigó la producción de estas enzimas con dos cepas (A y B) de Mucor sp utilizando dos medios: Sabouraud-dextrosa y Sabouraud- miel. Se realizaron fermentaciones durante 120 h a temperaturaambiente en un agitador orbital a 110 rpm. Las pruebas cuantitativa y cualitativa indicaron la presencia de actividad proteásica en el medio Saboraud-miel inoculado con la cepa B.

Endophytic fungi, which live within host plant tissues without causing any visible symptom of infection, are important mutualists that mediate plant-herbivore interactions. Thrips tabaci (Lindeman) is one of the key pests of onion, Allium cepa L., an economically important agricultural crop cultivated worldwide. However, information on endophyte colonization of onions, and their impacts on the biology of thrips feeding on them, is lacking. We tested the colonization of onion plants by selected fungal endophyte isolates using two inoculation methods. The effects of inoculated endophytes on T. tabaci infesting onion were also examined. Seven fungal endophytes used in our study were able to colonize onion plants either by the seed or seedling inoculation methods. Seed inoculation resulted in 1.47 times higher mean percentage post-inoculation recovery of all the endophytes tested as compared to seedling inoculation. Fewer thrips were observed on plants inoculated with Clonostachys rosea ICIPE 707, Trichoderma asperellum M2RT4, Trichoderma atroviride ICIPE 710, Trichoderma harzianum 709, Hypocrea lixii F3ST1 and Fusarium sp. ICIPE 712 isolates as compared to those inoculated with Fusarium sp. ICIPE 717 and the control treatments. Onion plants colonized by C. rosea ICIPE 707, T. asperellum M2RT4, T. atroviride ICIPE 710 and H. lixii F3ST1 had significantly lower feeding punctures as compared to the other treatments. Among the isolates tested, the lowest numbers of eggs were laid by T. tabaci on H. lixii F3ST1 and C. rosea ICIPE 707 inoculated plants. These results extend the knowledge on colonization of onions by fungal endophytes and their effects on Thrips tabaci.

In the food industry, Salmonella sp. and other pathogens represent a major threat as potential causes of foodborne diseases. The Salmonella enterica serovar Heidelberg (SH) has a great importance to public health, being currently the most frequent serotype in the official control analyzes carried out in Brazil’s Southern. In this region are concentrated the largest poultry flocks, contributing over 60 % of Brazilian production of chicken meat. To help prevent and minimize infections by pathogenic bacteria, in addition to attend the standards of biosecurity in the field and good hygiene and sanitizing programs in the industry, there is the possibility to control directly on the poultry breeding. Use of organic acids added in the diet of the fowls has an antibacterial action. In the other hand, the use of probiotics has been shown to prevent enteric diseases. This dissertation was divided into two chapters and aimed to evaluate the quantitative and qualitative reduction of SH in broiler chickens. In the first chapter the aim was to collect information on the prevalence rate of SH strain in broiler chickens. This coefficient was 100 %, being a suitable method for determining the frequency of salmonellosis in broiler chickens. In the second chapter five treatments were evaluated (a positive control and four tests), in order to reduce the colonization of SH in broilers, using organic acids and probiotic during production phase. In SH count related to the colonization of the cecum at 28 days of age differences were observed (p<0,05) among treatments. The Treatment 2 showed a count of 41 most probable number of bacterial cells per gram (MPN/g) with lower counts when compared to the positive control (5.106 MPN/g) and treatment 1 (1.354 MPN/g). The evaluation index of infected birds, there was a difference between treatments. In treatment 2, and treatment 4 reduced 58,34 % and 25 % respectively the number of fowls infected with SH. There was no effect of treatments on the performance. Administration of organic acids and probiotic used in the treatment 2 proved to be an effective tool in the SH control in broilers at 28 days of age.

El objetivo de este trabajo fue evaluar en diferentes periodos de almacenamiento la calidad, en términos de viabilidad y concentración de conidias, en formulaciones bioplaguicidas a base de Beauveria bassiana, elaboradas con diferentes materiales inertes. Los materiales inertes (tratamientos) evaluados fueron: harina de trigo, arcilla blanca, arcilla verde, sulfato de calcio, aceite de soya, aceite de maní y el testigo fue B. bassiana en sustrato (no formulado). La concentración del hongo, utilizada para las formulaciones fue 8.5 x 109 conidias por gramo. Se utilizó B. bassiana cepa 114, obtenida del cepario del laboratorio de hongos entomopatógenos de la UNA. La reproducción del hongo se hizo mediante el método de producción semi-industrial, utilizando arroz grano entero como sustrato. La caracterización se hizo mediante características macroscópicas y ritmo de crecimiento del hongo en los medios de cultivo: PDA, SDA y EMA. Luego de elaboradas las formulaciones, fueron almacenadas en condiciones de laboratorio (24 oC -28ºC, HR60%) durante seis meses se determinó la concentración y viabilidad de conidias. Para la evaluación de la concentración de conidias se prepararon diluciones seriadas y el conteo de conidias se realizó en una cámara con rayado Neubauer. La evaluación de viabilidad, se hizo en medio Agar-agua 2%. La evaluación de la concentración de conidias así como la evaluación de la viabilidad se hizo cada 15 días durante los primeros tres meses y luego cada 30 días durante los últimos tres meses. El efecto de los materiales inertes sobre la efectividad biológica de B. bassiana fue evaluado en un bioensayo con la especie Galleria mellonella la que se obtuvo del laboratorio de cría de la UNA, usando una suspensión del hongo a una concentración de 1.6 x 108 por ml. Se realizó Análisis de Varianza y separación de medias según Tukey, por cada fecha de monitoreo. Se concluye que el mejor material para formular B. bassiana es harina de trigo, ya que fue el material inerte que presentó menor disminución en la concentración y viabilidad de conidias durante el periodo evaluado. La mayor velocidad promedio de crecimiento se observó en el medio de cultivo PDA. En general, los materiales inertes no afectaron significativamente la efectividad biológica de Beauveria bassiana en larvas de Galleria mellonella.

As lipases e os biossurfactantes são compostos produzidos por microrganismos através de fermentações em estado sólido (FES) ou sumberso (FSm), os quais são aplicáveis nas indústrias alimentícia e farmacêutica, na bioenergia e na biorremediação, entre outras. O objetivo geral deste trabalho foi otimizar a produção de lipases através de fermentação em estado sólido e fermentação submersa. Os fungos foram selecionados quanto à habilidade de produção de lipases através de FES e FSm e aqueles que apresentaram as maiores atividades lipolíticas foram utilizados na seleção de variáveis significativas e na otimização da produção de lipases nos dois modos de cultivo. Foram empregadas técnicas seqüenciais de planejamento experimental, incluindo planejamentos fracionários, completos e a metodologia de superfície de resposta para a otimização da produção de lipases. As variáveis estudadas na FES foram o pH, o tipo de farelo como fonte de carbono, a fonte de nitrogênio, o indutor, a concentração da fonte de nitrogênio, a concentração do indutor e a cepa do fungo. Na FSm, além das variáveis estudadas na FES, estudaram-se as variáveis concentração inicial de inóculo e agitação. As enzimas produzidas foram caracterizadas quanto à temperatura e pH ótimos e quanto à estabilidade a temperatura e pH. Nas condições otimizadas de produção de lipases, foi avaliada a correlação entre a produção de lipases e bioemulsificantes. Inicialmente foram isolados 28 fungos. Os fungos Aspergillus O- 4 e Aspergillus E-6 foram selecionados como bons produtores de lipases no processo de fermentação em estado sólido e os fungos Penicillium E-3, Trichoderma E-19 e Aspergillus O-8 como bons produtores de lipases através da fermentação submersa. As condições otimizadas para a produção de lipases através de fermentação em estado sólido foram obtidas utilizando-se o fungo Aspergillus O-4, farelo de soja, 2% de nitrato de sódio, 2% de azeite de oliva e pHs inferiores a 5, obtendo-se atividades lipolíticas máximas de 57 U. As condições otimizadas para a produção de lipases na fermentação submersa foram obtidas utilizando-se o fungo Aspergillus O-8, farelo de trigo, 4,5% de extrato de levedura, 2% de óleo de soja e pH 7,15. A máxima atividade obtida durante a etapa de otimização foi 6 U. As lipases obtidas por FES apresentaram atividades máximas a 35ºC e pH 6,0, enquanto que as obtidas por FSm apresentaram ótimos a 37ºC e pH 7,2. A estabilidade térmica das lipases produzidas via FSm foi superior a das lipases obtidas via FES, com atividades residuais de 72% e 26,8% após 1h de exposição a 90ºC e 60ºC, respectivamente. As lipases obtidas via FES foram mais estáveis em pH´s alcalinos, com atividades residuais superiores a 60% após 24 h de exposição, enquanto as lipases produzidas via FSm foram mais estáveis em pH´s ácidos, com 80% de atividade residual na faixa de pH entre 3,5 e 6,5. Na fermentação submersa a correlação entre a produção de lipases e a atividade emulsificante óleo em água (O/A) e água em óleo (A/O) dos extratos foi 95,4% e 86,8%, respectivamente, obtendo-se atividades emulsificantes máximas O/A e A/O de 2,95 UE e 42,7 UE. Embora a maior produção de lipases tenha sido obtida na fermentação em estado sólido, não houve produção concomitante de biossurfactantes. Os extratos da fermentação submersa apresentaram redução da tensão superficial de 50 mN m -1 para 28 mN m -1 e atividade antimicrobiana frente ao microrganismo S. aureus ATCC 25923, com potenciais antimicrobianos de 36 a 43% nos três primeiros dias de fermentação. A fermentação submersa foi a técnica que apresentou os melhores resultados de otimização da produção de lipases, bem como de produção simultânea de biossurfactantes.

Los hongos end?fitos han recibido una creciente atenci?n en los a?os recientes, ya que han demostrado tener un potencial econ?mico en la agronom?a, debido a su posible uso como agentes de control biol?gico. El establecimiento endof?tico de hongos entomopat?genos en cultivos de inter?s, ofrece un campo amplio y prometedor para comprender las relaciones entre plantas y hongos y los mecanismos de defensa que estos ?ltimos pueden inducir en las plantas. El presente estudio busc? inocular y evaluar la colonizaci?n endof?tica de tres especies de hongos entomopat?genos en plantas de fr?jol (Phaseolus vulgaris). Para ello, se emplearon dos cepas de cada especie, una comercial y la otra de la colecci?n de entomopat?genos del Centro Internacional de Agricultura Tropical; cada cepa constituy? un tratamiento individual. A partir de la siembra y desarrollo de las cepas, se generaron in?culos de 108 conidios/mL que fueron inoculados en las plantas de fr?jol Diacol, variedad Calima. Se encontr? que Beauveria bassiana, Metarhizium anisopliae y Lecanicillium lecanii colonizaron individualmente las hojas de fr?jol. Los porcentajes de colonizaci?n promedio para las cepas de la colecci?n del CIAT fueron de 16% (?= 18,84) para B. bassiana, 13% (?= 4,96) en M. anisopliae y 4% (?= 4,64) para L. lecanii. Con respecto a las cepas comerciales, estos valores fueron 4% (?= 4,64) para B. bassiana; 26% (?= 32) para M. anisopliae y 44% (?= 13,02) para L. lecanii. Todas las cepas de hongos inoculadas pudieron ser recuperadas de tejido vegetal, lo que sugiere su posible uso como agentes de control biol?gico al menos para el cultivo del fr?jol.

The incidence of toxic cyanobacterial blooms is one of the important consequences of eutrophication in aquatic ecosystems. It is a very common phenomenon in reservoirs and shrimp ponds in the State of Rio Grande do Norte (RN), Brazil. Cyanobacterias produce toxins which can affect aquatic organisms and men trough the food chain. Aiming to contribute to the studies of cyanobacterias in RN, we propose: a) to evaluate the toxicity of isolated cyanobacterias in important fresh-water environments; and b) to verify the effects of both natural and cultured blooms occurred in reservoirs for human supply and in the cladoceran Ceriodaphnia silvestrii. This study was carried out using samples of natural blooms occurred between March and October of 2004 in Gargalheiras Dam (08º L e 39º W), in July of 2004 in Armando Ribeiro Gonçalves Dam (06o S e 37o W) and in commercial shrimp ponds (Litopenaeus vannamei) located in fresh-water environments. The samples were collected with plankton net (20µm.) for identification, isolation and obtaining of phytoplanktonic biomass for liophilization and later toxicity bioassays. The toxicity of cultured samples and natural blooms was investigated through bioassays in Swiss mice. Quantification of cyanobacteria in samples was conducted following the Ütermol method, with 300mL samples fixed with lugol. The toxicity test with Ceriodaphnia silvestrii followed ABNT, 2001 recommendations, and were accomplished with natural hepatotoxic bloom s samples and cultured samples of both non-toxic and neurotoxic C. raciborskii. In this test, five newborns, aged between 6 and 24 hours, were exposed to different concentrations (0 a 800 mg.L-1) of crude cyanobacterial extracts during 24 and 48 hours. Three replicates were used per treatment. The pH, temperature and dissolved oxygen at the beginning and after 24 and 48hours from the test were measured. We estimated the CL50 through the Trimmed Spearman-Karber method. The blooms were constituted by Microcystis panniformis, M. aeruginosa, Anabaena circinalis, Cylindrospermopsis raciborskii and Planktothrix agardhii, producers of mycrocistin-LR confirmed with HPLC analysis. Samples of hepatotoxic blooms registered toxinogenic potential for C. silvestrii, with CL50-24h value of 47.48 mg.L-1 and CL5048h of 38.15 mg.L-1 for GARG samples in march/2005; CL50-24h of 113,13 mg.L-1 and CL5048h of 88,24 mg.L-1 for ARG July/2004; CL50-24h of 300.39 mg.L-1 and CL50-48h of 149.89 mg.L-1 for GARG October/2005. For cultured samples, values of CL50-24h and CL50-48h for C. raciborskii toxic strains were 228.05 and 120.28 mg.L-1, respectively. There was no mortality of C. silvestrii during the tests with non-toxic C. raciborskii strain. The toxicity test with C. silvestrii presented good sensitivity degree to cyanotoxins. The toxicity of natural hepatotoxic blooms samples (microcystins) and cultured neurotoxic saxitoxins producer samples analyzed in this study give us strong indications of that toxin s influence on the zooplanktonic community structure in tropical aquatic environments. Eleven cyanobacteria strains were isolated, representing 6 species: Anabaenopsis sp., Cylindrospermopsis raciborskii, Chroococcus sp., Microcystis panniformis, Geitlerinema unigranulatum e Planktothrix agardhii. None presented toxicity in Swiss mice. The strains were catalogued and deposited in the Laboratório de Ecologia e Toxicologia de Organismos Aquáticos (LETMA), in UFRN, and will be utilized in ecotoxicológical and ecophysiological studies, aiming to clarify the causes and control of cyanobacterial blooms in aquatic environments in RN. This state s reservoirs must receive broader attention from the authorities, considering the constant blooms occurring in waters used for human consumption

Critical infrastructures are based on complex systems that provide vital services to the nation. The complexities of the interconnected networks, each managed by individual organisations, if not properly secured, could offer vulnerabilities that threaten other organisations’ systems that depend on their services. This thesis argues that the awareness of interdependencies among critical sectors needs to be increased. Managing and securing critical infrastructure is not isolated responsibility of a government or an individual organisation. There is a need for a strong collaboration among critical service providers of public and private organisations in protecting critical information infrastructure. Cyber exercises have been incorporated in national cyber security strategies as part of critical information infrastructure protection. However, organising a cyber exercise involved multi sectors is challenging due to the diversity of participants’ background, working environments and incidents response policies. How well the lessons learned from the cyber exercise and how it can be transferred to the participating organisations is still a looming question. In order to understand the implications of cyber exercises on what participants have learnt and how it benefits participants’ organisation, a Cyber Exercise Post Assessment (CEPA) framework was proposed in this research. The CEPA framework consists of two parts. The first part aims to investigate the lessons learnt by participants from a cyber exercise using the four levels of the Kirkpatrick Training Model to identify their perceptions on reaction, learning, behaviour and results of the exercise. The second part investigates the Organisation Cyber Resilience (OCR) of participating sectors. The framework was used to study the impact of the cyber exercise called X Maya in Malaysia. Data collected through interviews with X Maya 5 participants were coded and categorised based on four levels according to the Kirkpatrick Training Model, while online surveys distributed to ten Critical National Information Infrastructure (CNII) sectors participated in the exercise. The survey used the C-Suite Executive Checklist developed by World Economic Forum in 2012. To ensure the suitability of the tool used to investigate the OCR, a reliability test conducted on the survey items showed high internal consistency results. Finally, individual OCR scores were used to develop the OCR Maturity Model to provide the organisation cyber resilience perspectives of the ten CNII sectors.
