The unlabelled antibody peroxidase-antiperoxidase method was used to study the immunocytochemical properties of Leishmania and Trypanosoma cruzi amastigotes in situ after tissues had been submitted to different fixation procedures. Antisera were obtained from rabbits chronically infected with different strains of T. cruzi or immunized with L. mexicana amazonensis and L. braziliensis guyanensis, and were applied on 5 µm thick sections. T. cruzi antigens were well stained by the three anti-T. cruzi sera and the two anti-heis.hmama.sera at optimum dilution between 1:1,000 and 1:2,000, regardless the parasite strain. Differently, the leishmanial antigens were revealed by Leishmania sera only at low dilutions (between 1:60 -1:160), whereas the anti-T. cruzi sera, at these low dilutions, gave rather weak stainings. Although there is no clear explanation for this immunocytochemical "reverse-monodirectional" cross-reactivity between Leishmania and T. cruzi, the present results show that polyclonal antibodies agains Leishmania species, when used for immunocytochemical detection of these parasites in situ, react more strongly with T. cruzi amastigotes than with the homologous amastigotes.

Resumo: Objetivou-se avaliar a degradação in situ da Jurema preta. Foram estimados os valores da degradação potencial e efetiva da matéria seca, proteína bruta e fibra em detergente neutro. A jurema preta apresenta bom potencial de degradação para matéria seca e fibra em detergente neutro, já para a proteína bruta, parece haver necessidade de ação enzimática pós rúmen para que seja melhor aproveitada. [In situ degradability of jurema preta (Mimosa tenuiflora) in thinned and enriched caatinga rangelands]. Abstract: We aimed with this manuscript to present the evaluation of in situ degradability of Jurema preta in thinned and enriched caatinga. Were estimated the potential and effective degradation of dry matter, crude protein and neutral detergent fiber. The jurema preta shrubs have good potential to dry matter and neutral detergent fiber degradation, but to crude protein, to maximize their harnessing the post rumen digestion is needed.

A utilização dos carvões activados na remoção compostos poluentes dos efluentes urbanos tem demonstrado ser um processo cada vez mais económico e eficaz, sendo por isso muito utilizado no sector de tratamento e purificação de águas de abastecimento. O principal objectivo desta dissertação foi aplicar espectroscopia UV-Vis in situ no processo de adsorção em fase líquida de PPCP’s, nomeadamente cafeína e paracetamol. Nesta nova abordagem de medição e procedimento experimental, tiveram de se ter em conta vários factores e estudar o seu efeito durante o processo de adsorção. Um destes factores é a presença do carvão durante a medição da absorvância em espectroscopia. Deste modo, foram realizadas medições ao longo do tempo em soluções de carvão activado para determinar qual a sua interferência na medição e os motivos de tal comportamento. Verificou-se também que a forma, a quantidade e a agitação do carvão também influenciam os resultados de espectroscopia. Aplicando este novo método de medição, foi necessário adaptar a lei de Lambert-Beer para calcular os valores de concentração das espécies em solução, eliminando o erro na medição, provocado pela presença do carvão. Deste modo, o método de decomposição espectral foi aplicado e verificou-se que apenas as concentrações de carvão calculadas não correspondem aos valores reais em solução, apenas a sua variação demostra a evolução da interferência do adsorvente na medição dos valores de absorvância. No estudo cinético de adsorção dos solutos foi usado um carvão comercial tanto em pó como em formato granular. Durante o processo de adsorção em carvão granular verificou-se que a velocidade de adsorção foi menor, provavelmente devido à maior dificuldade de acesso aos poros, provocada pela complexidade da estrutura. Contudo, registou-se sempre uma remoção completa do soluto neste carvão, uma vez que a quantidade mínima possível nestes ensaios de carvão NOG é de 2,5mg, sendo uma quantidade relativamente superior à necessária para remover o soluto completamente da solução. Num pequeno estudo de adsorção de cafeína em carvões de diferentes origens confirmou-se que a eficiência de remoção do soluto é proporcional ao volume total de poros e deduz-se que a sua velocidade de adsorção está relacionada com a presença de mesoporosidade. Comparando a eficiência de remoção e a velocidade de adsorção dos dois solutos pelo mesmo carvão, observou-se que a adsorção da cafeína é mais rápida. Apesar dos valores de solubilidade e de volume molecular não justificarem de imediato este resultado, a cafeína tem maior afinidade com o carvão comercial, sugerindo a existência de propriedades químicas e texturais do carvão que levam a estes resultados. Por último, efectuou-se um estudo de adsorção de misturas para verificar a hipótese de adsorção competitiva entre os solutos. Após a realização dos ensaios verificou-se que não existe competição significativa entre a cafeína e o paracetamol.

Deep-eutectic solvents (DES) are considered novel renewable and biodegradable solvents, with a cheap and easy synthesis, without waste production. Later it was discovered a new subclass of DES that even can be biocompatible, since their synthesis uses primary metabolites such as amino acids, organic acids and sugars, from organisms. This subclass was named natural deep-eutectic solvents (NADES). Due to their properties it was tried to study the interaction between these solvents and biopolymers, in order to produce functionalized fibers for biomedical applications. In this way, fibers were produced by using the electrospinning technique. However, it was first necessary to study some physical properties of NADES, as well as the influence of water in their properties. It has been concluded that the water has a high influence on NADES properties, which can be seen on the results obtained from the rheology and viscosity studies. The fluid dynamics had changed, as well as the viscosity. Afterwards, it was tested the viability of using a starch blend. First it was tested the dissolution of these biopolymers into NADES, in order to study the viability of their application in electrospinning. However the results obtained were not satisfactory, since the starch polymers studied did not presented any dissolution in any NADES, or even in organic solvents. In this way it was changed the approach, and it was used other biocompatible polymers. Poly(ethylene oxide), poly(vinyl alcohol) and gelatin were the others biopolymers tested for the electrospinning, with NADES. All polymers show good results, since it was possible to obtain fibers. However for gelatin it was used only eutectic mixtures, containing active pharmaceutical ingredients (API’s), instead of NADES. For this case it was used mandelic acid (antimicrobial properties), choline chloride, ibuprofen (anti-inflammatory properties) and menthol (analgesic properties). The polymers and the produced fibers were characterized by scanning electron microscope (SEM), Transmission electron microscopy (TEM) and Fourier transform infrared spectroscopy (FTIR). With the help of these techniques it was possible to conclude that it was possible to encapsulate NADES within the fibers. Rheology it was also study for poly(ethylene oxide) and poly(vinyl alcohol), in a way to understand the influence of polymer concentration, on the electrospinning technique. For the gelatin, among the characterization techniques, it was also performed cytotoxicity and drug release studies. The gelatin membranes did not show any toxicity for the cells, since their viability was maintained. Regarding the controlled release profile experiment no conclusion could be drawn from the experiments, due to the rapid and complete dissolution of the gelatin in the buffer solution. However it was possible to quantify the mixture of choline chloride with mandelic acid, allowing thus to complete, and confirm, the information already obtained for the others characterization technique.

Three different treatments were applied on several specimens of dolomitic and calcitic marble, properly stained with rust to mimic real situations (the stone specimens were exposed to the natural environment for about six months in contact with rusted iron). Thirty six marble specimens, eighteen calcitic and eighteen dolomitic, were characterized before and after treatment and monitored throughout the cleaning tests. The specimens were characterized by SEM-EDS (Scanning Electron Microscopy coupled with Energy Dispersion System), XRD (XRay Diffraction), XRF (X-Ray Fluorescence), FTIR (Fourier Transform Infrared Spectroscopy) and color measurements. It was also made a microscopic and macroscopic analysis of the stone surface along with the tests of short and long term capillary absorption. A series of test trials were conducted in order to understand which concentrations and contact times best suits to this purpose, to confirm what had been written to date in the literature. We sought to develop new methods of treatment application, skipping the usual methods of applying chemical treatments on stone substrates, with the use of cellulose poultice, resorting to the agar, a gel already used in many other areas, being something new in this area, which possesses great applicability in the field of conservation of stone materials. After the application of the best methodology for cleaning, specimens were characterized again in order to understand which treatment was more effective and less harmful, both for the operator and the stone material. Very briefly conclusions were that for a very intense and deep penetration into the stone, a solution of 3.5% of SDT buffered with ammonium carbonate to pH around 7 applied with agar support would be indicated. For rust stains in its initial state, the use of Ammonium citrate at a concentration of 5% buffered with ammonium to pH 7 could be applied more than once until satisfactory results appear.

Introduction Leishmania braziliensis infection induces a large spectrum of lesions that clinically manifest as nodules or papules that progress to ulcers. Although it is already known that T helper cells predominate in the lesions, cytotoxic T cells have also been reported to be present, and their role in leishmaniasis immunopathogenesis is not well known. This study investigated the amounts of CD8+ and granzyme B+ cells in different clinical forms of human cutaneous leishmaniasis (CL). Methods Forty tissue fragments from early (E-CL) and late CL (L-CL) lesions and from disseminated leishmaniasis (DL) - papules and ulcers - were characterized. The inflamed area per fragment was calculated, and the CD8 and granzyme B expression levels in the infiltrates were quantified by counting positive cells in 15 fields. The localization of CD8 and granzyme B was graded subjectively. Results Inflammation was higher in L-CL and DL ulcers. CD8 expression was increased in late ulcerated lesions compared to recent lesions. The increase in CD8+ cells also correlated with the duration of the lesion. Papules had a higher frequency of granzyme B+ cells than E-CL lesions, although the frequency was similar to those for late and DL ulcers. CD8+ cells were mostly found in the papillary dermis. Conclusions CD8+ T and granzyme B+ cells are present in the inflammatory infiltrates of CL and DL and may participate in the immunopathogenesis of Leishmania infection.

This work reports the development of field-effect transistors (FETs), whose channel is based on zinc oxide (ZnO) nanoparticles (NPs). Using screen-printing as the primary deposition technique, different inks were developed, where the semiconducting ink is based on a ZnO NPs dispersion in ethyl cellulose (EC). These inks were used to print electrolyte-gated transistors (EGTs) in a staggered-top gate structure on glass substrates, using a lithium-based polymeric electrolyte. In another approach, FETs with a staggered-bottom gate structure on paper were developed using a sol-gel method to functionalize the paper’s surface with ZnO NPs, using zinc acetate dihydrate (ZnC4H6O4·2H2O) and sodium hydroxide (NaOH) as precursors. In this case, the paper itself was used as dielectric. The various layers of the two devices were characterized using X-ray diffraction (XRD), scanning electron microscopy (SEM), Fourier Transform Infrared spectroscopy (FTIR), thermogravimetric and differential scanning calorimetric analyses (TG-DSC). Electrochemical impedance spectroscopy (EIS) was used in order to evaluate the electric double-layer (EDL) formation, in the case of the EGTs. The ZnO NPs EGTs present electrical modulation for annealing temperatures equal or superior to 300 ºC and in terms of electrical properties they showed On/Off ratios in the order of 103, saturation mobilities (μSat) of 1.49x10-1 cm2(Vs)-1 and transconductance (gm) of 10-5 S. On the other hand, the ZnO NPs FETs on paper exhibited On/Off ratios in the order of 102, μSat of 4.83x10- 3 cm2(Vs)-1and gm around 10-8 S.
