The kinetics of hydrodeoxygenation of waste cooking oil (WCO) is investigated with unsupported CoMoS catalysts. A kinetic model is established and a comprehensive analysis of each reaction pathway is carried out. The results show that hydrodecarbonylation/decarboxylation (HDC) routes are the predominant reaction pathways in the elimination of oxygen, with the rate constant three times as high as that of hydrodeoxygenation (HDO). However, the HDC activity of the CoMoS catalyst deactivates due to gradual loss of sulfur from the catalyst. HDO process is insensitive to the sulfur deficiency. The kinetic modeling shows that direct hydrodecarbonylation of fatty acids dominates the HDC routes and, in the HDO route, fatty acids are transferred to aldehydes/alcohols and then to C-18 hydrocarbons, a final product, and the reduction of acids is the rate limiting step. The HDO route via alcohols is dominant over aldehydes due to a significantly higher reaction rate constant. The difference of C-18/C-17 ratio in unsupported and supported catalysts show that a support with Lewis acid sites may play an important role in the selectivity for the hydrodeoxygenation pathways and promoting the final product quality

A number of Christian churches in South Africa have proclaimed their commitment to reconciliation and the elimination of unjust inequalities. This study analyses how these commitments are being worked out at the micro-level of a congregation. Using an ethnographic approach, I explore how a charismatic congregation in Cape Town has changed from being nearly all-white to being more inclusive. I explore links between individual, cognitive identity change and institutional change; and consider the discourses which justify change, including their emphasis on 'unity in diversity' and 'restitution'. I outline the limitations of change, including the persistence of 'racialised' leadership structures and the discursive privileging of unity over restitution. This allows us to understand how micro-level changes take place, to explore their potentialities and limitations, and to apply these insights to other contexts.

SIGNIFICANCE: Heme degradation, which was described more than 30 years ago, is still very actively explored with many novel discoveries on its role in various disease models every year.

RECENT ADVANCES: The heme oxygenases (HO) are metabolic enzymes that utilize NADPH and oxygen to break apart the heme moiety liberating biliverdin (BV), carbon monoxide (CO), and iron. Heme that is derived from hemoproteins can be toxic to the cells and if not removed immediately, it causes cell apoptosis and local inflammation. Elimination of heme from the milieu enables generation of three products that influences numerous metabolic changes in the cell.

CRITICAL ISSUES: CO has profound effects on mitochondria and cellular respiration and other hemoproteins to which it can bind and affect their function, while BV and bilirubin (BR), the substrate and product of BV, reductase, respectively, are potent antioxidants. Sequestration of iron into ferritin and its recycling in the tissues is a part of the homeodynamic processes that control oxidation-reduction in cellular metabolism. Further, heme is an important component of a number of metabolic enzymes, and, therefore, HO-1 plays an important role in the modulation of cellular bioenergetics.

FUTURE DIRECTIONS: In this review, we describe the cross-talk between heme oxygenase-1 (HO-1) and its products with other metabolic pathways. HO-1, which we have labeled Nike, the goddess who personified victory, dictates triumph over pathophysiologic conditions, including diabetes, ischemia, and cancer.

Aims: Preterm infants are deprived of the normal intra-uterine exposure to maternal melatonin and may benefit from replacement therapy. We conducted a pharmacokinetic study to guide potential therapeutic trials. Methods: Melatonin was administered to 18 preterm infants in doses ranging from 0.04-0.6μgkg-1 over 0.5-6h. Pharmacokinetic profiles were analyzed individually and by population methods. Results: Baseline melatonin was largely undetectable. Infants receiving melatonin at 0.1μgkg-1h-1 for 2h showed a median half-life of 15.82h and median maximum plasma concentration of 203.3pgml-1. On population pharmacokinetics, clearance was 0.045lh-1, volume of distribution 1.098l and elimination half-life 16.91h with gender (P = 0.047) and race (P < 0.0001) as significant covariates. Conclusions: A 2h infusion of 0.1μgkg-1h-1 increased blood melatonin from undetectable to approximately peak adult concentrations. Slow clearance makes replacement of a typical maternal circadian rhythm problematic. The pharmacokinetic profile of melatonin in preterm infants differs from that of adults so dosage of melatonin for preterm infants cannot be extrapolated from adult studies. Data from this study can be used to guide therapeutic clinical trials of melatonin in preterm infants. © 2013 The British Pharmacological Society.

A highly sensitive and specific competitive ELISA on 96-microwell plates was developed for the analysis of the nonsteroidal anti-inflammatory drug diclofenac. Within the water cycle in Europe, this is one of the most frequently detected pharmaceutically active compounds. The LOD at a signal-tonoise ratio (S/N) of 3, and the IC 50, were found to be 6 ng/L and 60 ng/L respectively in tap water. In a comparative study using ELISA and GC-MS, diclofenac levels in wastewater from 21 sewage treatment plants were determined and a good correlation between these methods was found (ELISA vs. GCMS: r = 0.70, slope = 0,90, intercept = 0.37, n = 24). An average degradation rate of -25% can be calculated. Labscale-experiments on the elimination of diclofenac in continuous pilot sewage plants revealed a removal rate of only 5% over a period of 13 weeks. In a further study, the ELISA was applied to a number of extracts of various animal tissues from a range of species, and again a very good relationship between ELISA and LC-ESI/MS data sets was obtained (r = 0.90, p<0.0001; n = 117). The ELISA has proven to be a simple, rapid, reliable and affordable alternative to otherwise costly and advanced techniques for the detection of diclofenac in matrix diverse water samples and tissue extracts after only relatively simple sample preparation. © 2007 American Chemical Society.

The severe combined immunodeficient (SCID) mouse model may be used to evaluate new approaches for the treatment of acute myeloid leukemia (AML). We have previously demonstrated the killing of SCID mouse leukemia initiating cells by in vitro incubation with human GM-CSF fused to Diphtheria toxin (DT-huGM-CSF). In this report, we show that in vivo treatment with DT-huGM-CSF eliminates AML growth in SCID mice. Seven cases of AML were studied. SCID mice were treated intraperitoneally with the maximally tolerated dose of 75 microg/kg/day for 7 days. Antileukemic efficacy was determined at days 40 and 80 after transplantation, by enumerating the percentages of human cells in SCID bone marrow using flow cytometry and short tandem repeat polymerase chain reaction (STR-PCR) analysis. Four out of seven AML cases were sensitive to in vivo treatment with DT-huGM-CSF at both evaluation time points. In three of these cases, elimination of human cells was demonstrated by flow cytometry and STR-PCR. One AML case showed moderate sensitivity for DT-huGM-CSF, and growth of the two remaining AML cases was not influenced by DT-huGM-CSF. Sensitivity was correlated with GM-CSFR expression. Our data show that DT-huGM-CSF can be used in vivo to reduce growth of AML and warrant further development of DT-huGM-CSF for the treatment of human AML.

With advancements in the development of visible light responsive catalysts for H2 production frequently being reported, photocatalytic water splitting has become an attractive method as a potential ‘solar fuel generator’. The development of novel photo reactors which can enhance the potential of such catalyst, however, is rarely reported. This is particularly important as many reactor configurations are mass transport limited, which in term limits the efficiency of more effective photocatalysts in larger scale applications. This paper describes the performance of a novel fluidised photo reactor for the production of H2 over two catalysts under UV-Visible light and natural solar illumination. Catalysts Pt-C3N4 and NaTaO3.La were dispersed in the reactor and the rate of H2 was determined by GC-TCD analysis of the gas headspace. The unit was an annular reactor constructed from stainless steel 316 and quartz glass with a propeller located in the base to control fluidisation of powder catalysts. Reactor properties such as propeller rotational speed were found to enhance the photo activity of the system through the elimination of mass transport limitations and increasing light penetration. The optimum conditions for H2 evolution were found to be a propeller rotational speed of 1035 rpm and 144 W of UV-Visible irradiation, which produced a rate of 89 µmol h-1 g-1 over Pt-C3N4. Solar irradiation was provided by the George Ellery Hale Solar Telescope, located at the California Institute of Technology.

A method is described for the rapid extraction of pectic substances from alcohol insoluble solids (AIS) from material of plant origin, especially fruit. Samples of AIS can be prepared for galacturonic acid assay within 60 min using extraction with 0·5m HCl in a Fibertec-1 system (Tecator) for 30 min. The extraction conditions are carefully standardised and operator error is reduced by the elimination of transfer steps, particularly during filtration. The results obtained for plant-derived alcohol insoluble solids containing from 10% to 33% pectic substances were in close agreement with those obtained by enzymic hydrolysis using a commercially available enzyme preparation (Ultrazyme). The method will have application in the rapid, routine estimation of pectic substances in plant materials. © 1987.

PURPOSE OF REVIEW:

Recent studies underscore the importance of angle-closure glaucoma (ACG) as a cause of world blindness. A major contribution in assessing the true impact of this disease has been an article estimating the number of persons with occludable angles, angle closure, and blindness from ACG in China as 28.2 million, 9.1 million, and 1.7 million, respectively. Although these numbers are based on data from Singapore and Mongolia, which may be applied to China only with caution, they emphasize the blinding potential of ACG, which is three times as likely to be associated with blindness as open-angle glaucoma (OAG).

RECENT FINDINGS:

Recent reports in the Chinese literature on ACG prevalence suffer from definitional problems that would appear to lead to systematic overestimates of ACG prevalence and underestimates of OAG prevalence. Nonetheless, data from studies by Chinese investigators further emphasize the strong association between ACG and blindness, with fully 16% of subjects with ACG blind in one report-a far higher proportion than for OAG in China and elsewhere. The importance of topiramate as a cause of secondary angle closure has recently been understood, in part, because of a series of 19 such cases reported by investigators at the Food and Drug Administration.

SUMMARY:

Angle closure in this setting appears to be caused by uveal effusion and anterior rotation of the ciliary body with resultant closure of the angle. The condition is not always responsive to laser iridectomy, and elimination of the causative agent appears to be critical. Ultrasonic biomicroscopy is a potential new diagnostic modality for ACG, allowing the measurement of novel parameters, such as the angle opening distance (AOD) at 500 microm (AOD 500). The efficacy of such parameters in improving screening for ACG can only be established by prospective studies of potentially at-risk eyes. A number of novel treatments for AC and angle closure have recently been proposed, including cataract extraction, paracentesis, and argon laser iridoplasty. As with proposed new diagnostic modalities, the efficacy of these treatments remains to be demonstrated with prospective studies, ideally organized in a controlled, randomized fashion.

Bounding the tree-width of a Bayesian network can reduce the chance of overfitting, and allows exact inference to be performed efficiently. Several existing algorithms tackle the problem of learning bounded tree-width Bayesian networks by learning from k-trees as super-structures, but they do not scale to large domains and/or large tree-width. We propose a guided search algorithm to find k-trees with maximum Informative scores, which is a measure of quality for the k-tree in yielding good Bayesian networks. The algorithm achieves close to optimal performance compared to exact solutions in small domains, and can discover better networks than existing approximate methods can in large domains. It also provides an optimal elimination order of variables that guarantees small complexity for later runs of exact inference. Comparisons with well-known approaches in terms of learning and inference accuracy illustrate its capabilities.

Schizophrenia is a heritable brain illness with unknown pathogenic mechanisms. Schizophrenia's strongest genetic association at a population level involves variation in the major histocompatibility complex (MHC) locus, but the genes and molecular mechanisms accounting for this have been challenging to identify. Here we show that this association arises in part from many structurally diverse alleles of the complement component 4 (C4) genes. We found that these alleles generated widely varying levels of C4A and C4B expression in the brain, with each common C4 allele associating with schizophrenia in proportion to its tendency to generate greater expression of C4A. Human C4 protein localized to neuronal synapses, dendrites, axons, and cell bodies. In mice, C4 mediated synapse elimination during postnatal development. These results implicate excessive complement activity in the development of schizophrenia and may help explain the reduced numbers of synapses in the brains of individuals with schizophrenia.

Este trabalho focou-se no estudo de técnicas de sub-espaço tendo em vista as aplicações seguintes: eliminação de ruído em séries temporais e extracção de características para problemas de classificação supervisionada. Foram estudadas as vertentes lineares e não-lineares das referidas técnicas tendo como ponto de partida os algoritmos SSA e KPCA. No trabalho apresentam-se propostas para optimizar os algoritmos, bem como uma descrição dos mesmos numa abordagem diferente daquela que é feita na literatura. Em qualquer das vertentes, linear ou não-linear, os métodos são apresentados utilizando uma formulação algébrica consistente. O modelo de subespaço é obtido calculando a decomposição em valores e vectores próprios das matrizes de kernel ou de correlação/covariância calculadas com um conjunto de dados multidimensional. A complexidade das técnicas não lineares de subespaço é discutida, nomeadamente, o problema da pre-imagem e a decomposição em valores e vectores próprios de matrizes de dimensão elevada. Diferentes algoritmos de préimagem são apresentados bem como propostas alternativas para a sua optimização. A decomposição em vectores próprios da matriz de kernel baseada em aproximações low-rank da matriz conduz a um algoritmo mais eficiente- o Greedy KPCA. Os algoritmos são aplicados a sinais artificiais de modo a estudar a influência dos vários parâmetros na sua performance. Para além disso, a exploração destas técnicas é extendida à eliminação de artefactos em séries temporais biomédicas univariáveis, nomeadamente, sinais EEG.

É extensa a bibliografia dedicada a potenciais aplicações de materiais com mudança de fase na regulação térmica e no armazenamento de calor ou de frio. No entanto, a baixa condutividade térmica impõe limitações numa grande diversidade de aplicações com exigências críticas em termos de tempo de resposta curto ou com requisitos de elevada potência em ciclos de carga/descarga de calor latente. Foram desenvolvidos códigos numéricos no sentido de obter soluções precisas para descrever a cinética da transferência de calor com mudança de fase, com base em geometrias representativas, i.e. planar e esférica. Foram igualmente propostas soluções aproximadas, sendo identificados correspondentes critérios de validação em função das propriedades dos materiais de mudança de fase e de outros parâmetros relevantes tais como as escalas de tamanho e de tempo, etc. As referidas soluções permitiram identificar com rigor os fatores determinantes daquelas limitações, quantificar os correspondentes efeitos e estabelecer critérios de qualidade adequados para diferentes tipologias de potenciais aplicações. Os referidos critérios foram sistematizados de acordo com metodologias de seleção propostas por Ashby e co-autores, tendo em vista o melhor desempenho dos materiais em aplicações representativas, designadamente com requisitos ao nível de densidade energética, tempo de resposta, potência de carga/descarga e gama de temperaturas de operação. Nesta sistematização foram incluídos alguns dos compósitos desenvolvidos durante o presente trabalho. A avaliação das limitações acima mencionadas deu origem ao desenvolvimento de materiais compósitos para acumulação de calor ou frio, com acentuada melhoria de resposta térmica, mediante incorporação de uma fase com condutividade térmica muito superior à da matriz. Para este efeito, foram desenvolvidos modelos para otimizar a distribuição espacial da fase condutora, de modo a superar os limites de percolação previstos por modelos clássicos de condução em compósitos com distribuição aleatória, visando melhorias de desempenho térmico com reduzidas frações de fase condutora e garantindo que a densidade energética não é significativamente afetada. Os modelos elaborados correspondem a compósitos de tipo core-shell, baseados em microestruturas celulares da fase de elevada condutividade térmica, impregnadas com o material de mudança de fase propriamente dito. Além de visarem a minimização da fração de fase condutora e correspondentes custos, os modelos de compósitos propostos tiveram em conta a adequação a métodos de processamento versáteis, reprodutíveis, preferencialmente com base na emulsificação de líquidos orgânicos em suspensões aquosas ou outros processos de reduzidas complexidade e com base em materiais de baixo custo (material de mudança de fase e fase condutora). O design da distribuição microestrutural também considerou a possibilidade de orientação preferencial de fases condutoras com elevada anisotropia (p.e. grafite), mediante auto-organização. Outros estágios do projeto foram subordinados a esses objetivos de desenvolvimento de compósitos com resposta térmica otimizada, em conformidade com previsões dos modelos de compósitos de tipo core-shell, acima mencionadas. Neste enquadramento, foram preparados 3 tipos de compósitos com organização celular da fase condutora, com as seguintes características e metodologias: i) compósitos celulares parafina-grafite para acumulação de calor, preparados in-situ por emulsificação de uma suspensão de grafite em parafina fundida; ii) compósitos celulares parafina-Al2O3 para acumulação de calor, preparados por impregnação de parafina em esqueleto cerâmico celular de Al2O3; iii) compósitos celulares para acumulação de frio, obtidos mediante impregnação de matrizes celulares de grafite com solução de colagénio, após preparação prévia das matrizes de grafite celular. Os compósitos com esqueleto cerâmico (ii) requereram o desenvolvimento prévio de um método para o seu processamento, baseado na emulsificação de suspensões de Al2O3 em parafina fundida, com adequados aditivos dispersantes, tensioactivos e consolidantes do esqueleto cerâmico, tornando-o auto-suportável durante as fases posteriores de eliminação da parafina, até à queima a alta temperatura, originando cerâmicos celulares com adequada resistência mecânica. Os compósitos desenvolvidos apresentam melhorias significativos de condutividade térmica, atingindo ganhos superiores a 1 ordem de grandeza com frações de fase condutora inferior a 10 % vol. (4 W m-1 K-1), em virtude da organização core-shell e com o contributo adicional da anisotropia da grafite, mediante orientação preferencial. Foram ainda preparados compósitos de armazenamento de frio (iii), com orientação aleatória da fase condutora, obtidos mediante gelificação de suspensões de partículas de grafite em solução aquosa de colagénio. Apesar da estabilidade microestrutural e de forma, conferida por gelificação, estes compósitos confirmaram a esperada limitação dos compósitos com distribuição aleatória, em confronto com os ganhos alcançados com a organização de tipo core-shell.

Silver nanoparticles (AgNP) have been produced and applied in a variety of products ranging from personal care products to food package containers, clothing and medicine utilities. The antimicrobial function of AgNP makes it very useful to be applied for such purposes. Silver (Ag) is a non-essential metal for organisms, and it has been historically present in the environment at low concentrations. Those concentrations of silver increased in the last century due to the use of Ag in the photographic industry and lately are expected to increase due to the use of AgNPs in consumer products. The presence of AgNP in the aquatic environment may pose a risk for aquatic species, and the effects can vary from lethal to sublethal effects. Moreover, the contact of aquatic organisms with AgNP may not cause immediately the death of individuals but it can be accumulated inside the animals and consequently transferred within the food chain. Considering this, the objective of this work was to study the transfer of silver nanoparticles in comparison to silver ions, which was used as silver nitrate, within an aquatic food chain model. To achieve this goal, this study was divided into four steps: the toxicity assessment of AgNP and AgNO3 to aquatic test-species, the bioaccumulation assessment of AgNP and AgNO3 by Pseudokirchneriella subcapitata and Daphnia magna under different exposure scenarios, and finally the evaluation of the trophic transfer of Ag through an experimental design that included the goldfish Carassius auratus in a model trophic chain in which all the species were exposed to the worse-case scenario. We observed that the bioconcentration of Ag by P. subcapitata is mainly driven by ionic silver, and that algae cannot internalize these AgNPs, but it does internalizes dissolved Ag. Daphnia magna was exposed to AgNP and AgNO3 through different exposure routes: water, food and both water and food. The worse-case scenario for Daphnia Ag bioaccumulation was by the joint exposure of contaminated water and food, showing that Ag body burdens were higher for AgNPs than for AgNO3. Finally, by exposing C. auratus for 10 days through contaminated water and food (supplied as D. magna), with another 7 days of depuration phase, it was concluded that the 10 days of exposure were not enough for fish to reach a plateau on Ag internal concentration, and neither the 7 days of elimination were sufficient to cause total depuration of the accumulated Ag. Moreover, a higher concentration of Ag was found in the intestine of fish when compared with other organs, and the elimination rate constant of AgNP in the intestine was very low. Although a potential for trophic transfer of AgNP cannot be suggested based in the data acquired in this study, there is still a potential environmental risk for aquatic species.

This thesis reports in detail studies of industrial solid wastes valorization as alternative raw materials. All tested wastes are classified as non-hazardous and are generated in the pulp and paper process, including primary sludge, dregs, grits, lime mud and bottom ash (this generated in a process that occurs in parallel to the production of cellulose, whose aim is the production of energy to supply the plant through the combustion of forest biomass in fluidized bed). A detailed general characterization was performed at each waste and according to their characteristics, they were selected some applications in materials with potential use, specifically in Fibercement, Bituminous Mixture for regularization layer and industrial mortars (rendering mortars and cementitious-adhesive). After decided to application each waste was specifically tested to proceed the setting up of formulations containing different content of waste in replacement of the raw conventional material. As an isolated case, the bottom ash was tested not only as an alternative raw material for construction materials, but also it was tested for its use in fluidized bed in which the waste is generated as raw material. Both dregs and bottom ash had undergone special treatment to make possible to obtain a better quality of waste in order do not compromise the final product characteristics and process. The dregs were tested in bituminous mixtures as received and also washed (on the laboratory scale to remove soluble salts) and bottom ash were washed and screened in industrial scale (for removal of soluble salts, especially chlorides and coarse fraction particles elimination - particles larger than 1 mm size). The remaining residues form used in such as received avoiding additional costs. The results indicated potential and some limitations for each application to the use of these wastes as alternative raw material, but in some cases, the benefits in relation to valorization overlap with its limitations in both aspects, environmental and economic.
