Objectives: The aim of this preliminary study was to characterize the plasma lipid profiling of women with preeclampsia. Design and methods: Plasma samples of 8 pregnant women with early-onset preeclampsia and 8 normal pregnant women were evaluated. Lipids were extracted from plasma using the Bligh-Dyer protocol. The extracts were subjected to MALDI-MS. Data matrix was exported for partial least squares discriminant analysis (PLS-DA) and a parameter VIP was employed to reflect the variable importance in the discriminant analysis. The major discriminant variables were selected and underwent to Mann-Whitney U test. Results: A total of 1290 ions were initially identified and twelve m/z signals were highlighted as the most important lipids for the discrimination of patients with preeclampsia. The identification of these differential lipids was carried out through Lipid Database Search. Conclusions: The main classes identified were glycerophosphocholines [GP01], glycerophosphoserines [GP03], glycerophosphoglycerols [GP04], glycosyldiradylglycerols [GL05] and glycerophosphates [GP10]. The effects of small fractions of calcium (x = 0, 0.05, 0.1, 0.15, and 0.20) on the structure and the catalytic properties of La2-xCaxCuO4 peroviskites have been investigated. The samples have been synthesized using the co-precipitation method. Perovskite-type oxides were characterized by XRD, TPR, XPS, XANES, SEM, and TEM. Catalytic tests for the water gas shift reaction (WGSR) were carried out in a tubular reactor at 290 degrees C. All samples showed a well-defined perovskite structure with surface areas between 6 and 18 m(2) g(-1). The partial substitution of La by Ca enhanced the stability of the perovskites and increased their reduction temperature. All catalysts were actives for WGSR, and the best catalytic performance was obtained for the La1.85Ca0.15CuO4 catalyst, but the samples with 5 and 10% of Ca had the best TOF values for reaction. These results can be associated to promoter effect of calcium, the high surface area, and the reducible species Cu-0 and Cu1+. (C) 2011 Elsevier B.V. All rights reserved.

Background: Peroxiredoxins have diverse functions in cellular defense-signaling pathways. 2-Cys-peroxiredoxins (2-Cys-Prx) reduce H2O2 and alkyl-hydroperoxide. This study describes the purification and characterization of a genuine 2-Cys-Prx from Vigna unguiculata (Vu-2-Cys-Prx). Methods: Vu-2-Cys-Prx was purified from leaves by ammonium sulfate fractionation, chitin affinity and ion exchange chromatography. Results: Vu-2-Cys-Prx reduces H2O2 using NADPH and DTT. Vu-2-Cys-Prx is a 44 kDa (SDS-PAGE)/46 kDa (exclusion chromatography) protein that appears as a 22 kDa molecule under reducing conditions, indicating that it is a homodimer linked intermolecularly by disulfide bonds and has a pI range of 4.56-4.72; its NH2-terminal sequence was similar to 2-Cys-Prx from Phaseolus vulgaris (96%) and Populus tricocarpa (96%). Analysis by ESI-Q-TOF MS/MS showed a molecular mass/pI of 28.622 kDa/5.18. Vu-2-Cys-Prx has 8% alpha-helix, 39% beta-sheet, 22% of turns and 31% of unordered forms. Vu-2-Cys-Prx was heat stable, has optimal activity at pH 7.0, and prevented plasmid DNA degradation. Atomic force microscopy shows that Vu-2-Cys-Prx oligomerized in decamers which might be associated with its molecular chaperone activity that prevented denaturation of insulin and citrate synthase. Its cDNA analysis showed that the redox-active Cys(52) residue and the amino acids Pro(45), Thr(49) and Arg(128) are conserved as in other 2-Cys-Prx. General significance: The biochemical and molecular features of Vu-2-Cys-Prx are similar to other members of 2-Cys-Prx family. To date, only one publication reported on the purification of native 2-Cys-Prx from leaves and the subsequent analysis by N-terminal Edman sequencing, which is crucial for construction of stromal recombinant 2-Cys-Prx proteins. Abstract Background Signaling by the vitamin A-derived morphogen retinoic acid (RA) is required at multiple steps of cardiac development. Since conversion of retinaldehyde to RA by retinaldehyde dehydrogenase type II (ALDH1A2, a.k.a RALDH2) is critical for cardiac development, we screened patients with congenital heart disease (CHDs) for genetic variation at the ALDH1A2 locus. Methods One-hundred and thirty-three CHD patients were screened for genetic variation at the ALDH1A2 locus through bi-directional sequencing. In addition, six SNPs (rs2704188, rs1441815, rs3784259, rs1530293, rs1899430) at the same locus were studied using a TDT-based association approach in 101 CHD trios. Observed mutations were modeled through molecular mechanics (MM) simulations using the AMBER 9 package, Sander and Pmemd programs. Sequence conservation of observed mutations was evaluated through phylogenetic tree construction from ungapped alignments containing ALDH8 s, ALDH1Ls, ALDH1 s and ALDH2 s. Trees were generated by the Neighbor Joining method. Variations potentially affecting splicing mechanisms were cloned and functional assays were designed to test splicing alterations using the pSPL3 splicing assay. Results We describe in Tetralogy of Fallot (TOF) the mutations Ala151Ser and Ile157Thr that change non-polar to polar residues at exon 4. Exon 4 encodes part of the highly-conserved tetramerization domain, a structural motif required for ALDH oligomerization. Molecular mechanics simulation studies of the two mutations indicate that they hinder tetramerization. We determined that the SNP rs16939660, previously associated with spina bifida and observed in patients with TOF, does not affect splicing. Moreover, association studies performed with classical models and with the transmission disequilibrium test (TDT) design using single marker genotype, or haplotype information do not show differences between cases and controls. Conclusion In summary, our screen indicates that ALDH1A2 genetic variation is present in TOF patients, suggesting a possible causal role for this gene in rare cases of human CHD, but does not support the hypothesis that variation at the ALDH1A2 locus is a significant modifier of the risk for CHD in humans.

O tratamento das lesões condrais e osteocondrais do joelho em pacientes jovens ainda permanece um desafio para os ortopedistas. As técnicas de reparo atualmente disponíveis no Brasil, como o desbridamento, microfraturas e transplante osteocondral autólogo são insuficientes nos tratamentos de lesões condrais e osteocondrais grandes. O transplante osteocondral homólogo a fresco (TOF) na articulação do joelho vem sendo usado nos Estados Unidos com excelentes resultados. Este artigo tem o intuito de revisar a ciência básica, indicações, técnicas cirúrgicas, possíveis complicações e descrever a técnica de transplante osteocondral homólogo a fresco na articulação do joelho realizada no Instituto de Ortopedia e Traumatologia do Hospital das Clínicas da Universidade de São Paulo.

[EN]Until recently, sample preparation was carried out using traditional techniques, such as liquid–liquid extraction (LLE), that use large volumes of organic solvents. Solid-phase extraction (SPE) uses much less solvent than LLE, although the volume can still be significant. These preparation methods are expensive, time-consuming and environmentally unfriendly. Recently, a great effort has been made to develop new analytical methodologies able to perform direct analyses using miniaturised equipment, thereby achieving high enrichment factors, minimising solvent consumption and reducing waste. These microextraction techniques improve the performance during sample preparation, particularly in complex water environmental samples, such as wastewaters, surface and ground waters, tap waters, sea and river waters. Liquid chromatography coupled to tandem mass spectrometry (LC/MS/MS) and time-of-flight mass spectrometric (TOF/MS) techniques can be used when analysing a broad range of organic micropollutants. Before separating and detecting these compounds in environmental samples, the target analytes must be extracted and pre-concentrated to make them detectable. In this work, we review the most recent applications of microextraction preparation techniques in different water environmental matrices to determine organic micropollutants: solid-phase microextraction SPME, in-tube solid-phase microextraction (IT-SPME), stir bar sorptive extraction (SBSE) and liquid-phase microextraction (LPME). Several groups of compounds are considered organic micropollutants because these are being released continuously into the environment. Many of these compounds are considered emerging contaminants. These analytes are generally compounds that are not covered by the existing regulations and are now detected more frequently in different environmental compartments. Pharmaceuticals, surfactants, personal care products and other chemicals are considered micropollutants. These compounds must be monitored because, although they are detected in low concentrations, they might be harmful toward ecosystems.

The study of electrochemiluminescence (ECL) involves photophysical and electrochemical aspects. Excited states are populated by an electrical stimulus. The most important applications are in the diagnostic field where a number of different biologically-relevant molecules (e.g. proteins and nucleic acids) can be recognized and quantified with a sensitivity and specificity previously not reachable. As a matter of fact the electrochemistry, differently to the classic techniques as fluorescence and chemiluminescence, allows to control the excited state generation spatially and temporally. The two research visits into A. J. Bard electrochemistry laboratories were priceless. Dr. Bard has been one of ECL pioneers, the first to introduce the technique and the one who discovered in 1972 the surprising emission of Ru(bpy)3 2+. I consider necessary to thank by now my supervisors Massimo and Francesco for their help and for giving me the great opportunity to know this unique science man that made me feel enthusiastic. I will never be grateful enough… Considering that the experimental techniques of ECL did not changed significantly in these last years the most convenient research direction has been the developing of materials with new or improved properties. In Chapter I the basics concepts and mechanisms of ECL are introduced so that the successive experiments can be easily understood. In the final paragraph the scopes of the thesis are briefly described. In Chapter II by starting from ECL experimental apparatus of Dr. Bard’s laboratories the design, assembly and preliminary tests of the new Bologna instrument are carefully described. The instrument assembly required to work hard but resulted in the introduction of the new technique in our labs by allowing the continuation of the ECL studies began in Texas. In Chapter III are described the results of electrochemical and ECL studies performed on new synthesized Ru(II) complexes containing tetrazolate based ligands. ECL emission has been investigated in solution and in solid thin films. The effect of the chemical protonation of the tetrazolate ring on ECL emission has been also investigated evidencing the possibility of a catalytic effect (generation of molecular hydrogen) of one of the complexes in organic media. Finally, after a series of preliminary studies on ECL emission in acqueous buffers, the direct interaction with calf thymus DNA of some complexes has been tested by ECL and photoluminescence (PL) titration. In Chapter IV different Ir(III) complexes have been characterized electrochemically and photophysically (ECL and PL). Some complexes were already well-known in literature for their high quantum efficiency whereas the remaining were new synthesized compounds containing tetrazolate based ligands analogous to those investigated in Chapt. III. During the tests on a halogenated complex was unexpectedly evidenced the possibility to follow the kinetics of an electro-induced chemical reaction by using ECL signal. In the last chapter (V) the possibility to use mono-use silicon chips electrodes as ECL analitycal devices is under investigation. The chapter begins by describing the chip structure and materials then a signal reproducibility study and geometry optimization is carried on by using two different complexes. In the following paragraphs is reported in detail the synthesis of an ECL label based on Ru(bpy)3 2+ and the chip functionalization by using a lipoic acid SAM and the same label. After some preliminary characterizations (mass spectroscopy TOF) has been demonstrated that by mean of a simple and fast ECL measurement it’s possible to confirm the presence of the coupling product SAM-label into the chip with a very high sensitivity. No signal was detected from the same system by using photoluminescence.

The Time-Of-Flight (TOF) detector of ALICE is designed to identify charged particles produced in Pb--Pb collisions at the LHC to address the physics of strongly-interacting matter and the Quark-Gluon Plasma (QGP). The detector is based on the Multigap Resistive Plate Chamber (MRPC) technology which guarantees the excellent performance required for a large time-of-flight array. The construction and installation of the apparatus in the experimental site have been completed and the detector is presently fully operative. All the steps which led to the construction of the TOF detector were strictly followed by a set of quality assurance procedures to enable high and uniform performance and eventually the detector has been commissioned with cosmic rays. This work aims at giving a detailed overview of the ALICE TOF detector, also focusing on the tests performed during the construction phase. The first data-taking experience and the first results obtained with cosmic rays during the commissioning phase are presented as well and allow to confirm the readiness state of the TOF detector for LHC collisions.

Das Vorkommen von Häutungshormonen in adulten Insekten, insbesondere solcher, die eine lange Imaginalphase durchlaufen, wirft die Frage nach der Regulation der Ecdysteroidsynthese außerhalb der Prothorakaldrüse auf. Unter diesem Gesichtspunkt kann Gryllus bimaculatus mit einem ausgesprochen langen Adultstadium und rapiden zeitlichen Veränderungen des Ecdysontiters als ein geeignetes Versuchsobjekt angesehen werden.Der vorliegenden Dissertation liegt die Arbeitshypothese zugrunde, daß die Ecdysteroid-Synthese bzw. Sekretion von Adultgeweben in männlichen Imagines der Mittelmeerfeldgrille durch Neuropeptide hormonell reguliert wird. Als Quelle für die Ecdysteroidsynthese wurde auf Grund immunohistochemischer Befunde sowie der Ergebnisse von Sekretionsprofil-Analysen unter anderem die Oenocyten in Betracht gezogen.Zur Überprüfung dieser Hypothese wurde ein in vitro Bioassay entwickelt, der es ermöglichte, die Wirkung von extrahierten Substanzen auf die Hormonsynthese mittels RIA/HPLC zu bestimmen. Aus Köpfen adulter G. bimaculatus ließen sich durch HP-SEC Faktoren isolieren, die die Ecdysteroidsekretion in Oenocyten und Tergiten stimulierten, die aber keinen Einfluß auf die Hormonsekretion von Fettgewebe sowie der Prothorakaldrüsen hatten. Die Wirkung des aufgereinigten Extraktes in Oenocyten war zeit- und dosis-abhängig. Die ecdysiotropen Faktoren besaßen ein Molekulargewicht zwischen 26,5 und 30 kDa. Die Größe der Molmasse der ecdysiotropen Faktoren entsprach somit bei adulten männlichen Grillen etwa dem des Neurohormons PTTH bei Lepidopteren. Dennoch zeigten Antikörper, die gegen PTTH von Bombyx mori gerichtet waren im Western-Blot keine Bindung an Gryllus bimaculatus Kopfextrakte. Die Sekretionsprodukte von Oenocyten, die mit Ecdysiotropinen behandelt waren, wurden durch RP- und NP-HPLC identifiziert. Es konnten zwei zusätzliche Peaks neben einem deutlichen Anstieg von 20-Hydroxyecdyson nachgewiesen werden. Auf Grund identischer Retentionszeiten mit Referenzsubstanzen handelt es sich bei einem Peak vermutlich um Makisteron A.Obwohl die Applikation von Azadirachtin in G. bimaculatus zu einer Senkung des Hämolymph-Ecdysteroidgehalts führte, konnte keine Anreicherung von Ecdysiotropinen erzielt werden.Die die Ecdysteroidsekretion-beeinflussenden Faktoren waren resistent gegen Kochen und Alkylierung aber nicht stabil gegen Reduzierung durch DTT und Behandlung mit Neuramidase. Damit konnte gezeigt werden, daß das Vorhandensein von Disulfidbrücken und Oligosaccharidketten für die biologische Aktivität notwendig ist.Die Aminosäuresequenz-Analyse und der enzymatische Verdau der stimulierenden Faktoren durch Exopeptidasen wiesen auf geschützte N- und C-Termini hin. Ferner wurden einige interne Peptidfragmente von fünf Proteinbanden sequenziert, die keine Homologie zu bereits bekannten regulatorischen Neuropeptiden zeigten. Als einziges bekanntes Protein aus diesem Bereich konnte das â14-3-3-like Proteinâ mit Hilfe MALDI-MS identifiziert werden.Die Stimulierung der Ecdysteroidsekretion in Oenocyten von G. bimaculatus durch Oenocyten-stimulierende Faktoren (OSF) aus Kopfextrakten konnte mittels Signalstoff-Effektoren in vitro nachgeahmt werden. Außerdem wurde mit Hilfe eines RRA nachgewiesen, daß der intrazelluläre cAMP-Spiegel von Oenocyten durch OSF erhöht wird. Daraus kann geschlossen werden, daß cAMP als âSecond Messengerâ an der Wirkung der OSF beteiligt ist. Calcium-Ionen schienen für die Ecdysteroidsekretion notwendig zu sein. Allerdings führte eine artifizielle Erhöhung der intrazellulären Calcium-Konzentration durch das Ionophor Ionomycin zu einer Hemmung der Sekretion. Schließlich wird ein Modell zur Erklärung des Wirkmechanismus von OSF in Gryllus bimaculatus postuliert und diskutiert.

Im Rahmen meiner Arbeit wurden erstmals die Intermediärfilament-Proteine (IF-Proteine) des Sibirischen Störs Acipenser baeri (Strahlenflosser, Knorpelganoid) kloniert und sequenziert. Aus einer cDNA-Bank konnten die Sequenzen von 13 IF-Proteine gewonnen werden. Von insgesamt zehn Keratinen codieren sieben für Typ I-Keratine und drei für Typ II. Zusätzlich konnten noch Desmin, Vimentin und ein Lamin identifiziert werden. Je einem Typ I- (K13) und einem Typ-II-Keratin (K2) fehlen wenige Aminosäuren in der Head-Domäne.Cytoskelett-Präparationen aus Epidermis, Mitteldarm, Magen und Kieme wurden mittels 2D-PAGE aufgetrennt. Durch Einsatz des CKBB-Test und Immunoblots wurden die verschiedenen Typ I und II-Keratine sowie Desmin und Vimentin identifiziert. Die gewebsspezifische Expression der Keratine ermöglichte zumeist ihre Einteilung in 'E' (epidermal) und 'S' ('simple epithelial').Die MALDI-MS-Analyse einer 2D-PAGE-Koelektrophorese von Seitenflosse und Mitteldarm zeigte, daß die 34 vorhandenen Proteinflecke auf nur 13 verschiedene IF-Proteine zurückgehen. Neun dieser Flecke konnten Sequenzen zugewiesen werden. Zusammen mit den verbleibenden vier Proteinflecken ergeben sich für den Stör nunmehr insgesamt 17 bekannte IF-Proteine. Von drei biochemisch identifizierten IS-Keratinen kommt eines nur im Mitteldarm vor und nur einem konnte eine Sequenz zugeordnet werden (K18). Dem einzigen Typ IIS-Keratin konnte keine Sequenz zugeordnet werden, wahrscheinlich handelt es sich um dabei um das K8-Orthologe. Jedem der fünf Typ IE-Proteine konnte eine Sequenz zugeordnet werden (K10 bis K14), ebenso wie dem einzigen identifizierten Typ IIE-Keratin (K2). Von den Typ III-Proteinen wurden Desmin und Vimentin ihren Proteinflecken zugeordnet. Die nicht zugeordnete Sequenz aba-k1 codiert möglicherweise für ein IIE-Keratin, während aba-k15 vermutlich die Sequenz für ein IE-Keratin enthält. Bei den Proteinflecken, denen eine Sequenz zugeordnet werden konnten, kann für Aba-K2 die Zugehörigkeit zum IIE-Typ angenommen werden, während es sich bei Aba-K10 wahrscheinlich um ein IE-Keratin handelt.Durch Datenbankvergleiche und molekulare Stammbäume konnte die Zugehörigkeit der identifizierten Lamin-Sequenz zum B3-Subtyp der Vertebraten gezeigt werden.Die Daten der Biochemie und indirekten Immunfluoreszenzmikroskopie zeigen, daß Keratine in Epithelien und Vimentin in mesenchymalen Geweben vorkommen. Es existieren starke Hinweise, daß im letzten Gewebetyp Keratine auch koexprimiert werden. Desmin kommt in großen Mengen im Magen und im Mitteldarm vor und stellt dort das prominenteste Protein.Mit den gewonnenen Sequenzdaten wurden molekulare Stammbäume und Sequenzidentitäten berechnet. Die daraus resultierenden Konsequenzen für die Verwandtschaftsverhältnisse der verschiedenen IF-Proteine sowie der Wirbeltiere werden diskutiert.

Selektine sind eine Gruppe von Transmembranglycoproteinen, welche als Adhäsionsmoleküle innerhalb des vaskulären Systems Zelladhäsionsprozesse zwischen Leukozyten und Endothelzellen vermitteln. Das Sialyl-Lewisa Epitop und verwandte Kohlenhydratstrukturen wurden als Liganden der E- und P-Selektine identifiziert. Durch die chemische Synthese verwandter Strukturen verspricht man sich, die im Laufe inflammatorischer Prozesse exprimierten Rezeptoren gezielt blockieren zu können und dadurch pathologische Abläufe wie hämatogene Metastasierungen oder Abstoßungsreaktionen zu bekämpfen. Einige Bereiche der Aminosäuresequenz des E-Selektin-Ligand-1 (ESL-1) treten hochkonservativ auch in anderen Selektinliganden wie MG160 oder PSGL-1 auf und wurden deshalb für die N-Glycosylierung mit einem sulfatierten Oligosaccharid ausgewählt (11). -Val665-Glu-Cys-Arg-Asp-Ile-Val-Gly-Asn(Sulfo-Lea)-Leu-Tyr-Glu-Leu-Glu-Ser-Glu-Asp-Ile682- 11 Im ersten Teil der Arbeit wurde eine Strategie ausgearbeitet, das sulfatierte Trisaccharid 60 im Multigrammaßstab zu synthetisieren. Der endogene Ligand 2 wurde an drei Positionen modifiziert: Austausch der α-L-Fucose gegen die biologisch stabilere α-D-Arabinose, Einführung einer Sulfatgruppe anstelle der N-Acetylneuraminsäure sowie Übergang von O- zu N-glykosidischer Verknüpfung. Die hochregioselektive Einführung der Sulfatgruppe gelingt in sehr guten Ausbeuten durch Vorkomplexierung mit Dibutylzinnoxid und anschließende Umsetzung mit Schwefeltrioxid/Trimethylamin. Durch die Verwendung des anomeren Azids als permanente Schutzgruppe kann das Trisaccharid nach schonender Reduktion zum Amin an ein Asparaginsäurederivat angekuppelt und in einer linearen Synthese nach Fmoc-Strategie als N-Glycosylaminosäure in die Synthese eingebracht werden. Das in der Arbeitsgruppe Kunz entwickelte PTMSEL-Ankersystem 20a erlaubt sowohl die problemlose Synthese als auch die Abspaltung vom polymeren Träger unter sehr milden Bedingungen. Nach dem Entfernen der Benzylester und -ether durch Pd(0) – katalysierte Hydrierung können sulfatierte Glycopeptidsequenzen des Typs 11 über NMR-Spektroskopie (korrelierte Spektren) und Massenspektroskopie (ESI, MALDI) identifiziert werden.

The focus of this thesis was the in-situ application of the new analytical technique "GCxGC" in both the marine and continental boundary layer, as well as in the free troposphere. Biogenic and anthropogenic VOCs were analysed and used to characterise local chemistry at the individual measurement sites. The first part of the thesis work was the characterisation of a new set of columns that was to be used later in the field. To simplify the identification, a time-of-flight mass spectrometer (TOF-MS) detector was coupled to the GCxGC. In the field the TOF-MS was substituted by a more robust and tractable flame ionisation detector (FID), which is more suitable for quantitative measurements. During the process, a variety of volatile organic compounds could be assigned to different environmental sources, e.g. plankton sources, eucalyptus forest or urban centers. In-situ measurements of biogenic and anthropogenic VOCs were conducted at the Meteorological Observatory Hohenpeissenberg (MOHP), Germany, applying a thermodesorption-GCxGC-FID system. The measured VOCs were compared to GC-MS measurements routinely conducted at the MOHP as well as to PTR-MS measurements. Furthermore, a compressed ambient air standard was measured from three different gas chromatographic instruments and the results were compared. With few exceptions, the in-situ, as well as the standard measurements, revealed good agreement between the individual instruments. Diurnal cycles were observed, with differing patterns for the biogenic and the anthropogenic compounds. The variability-lifetime relationship of compounds with atmospheric lifetimes from a few hours to a few days in presence of O3 and OH was examined. It revealed a weak but significant influence of chemistry on these short-lived VOCs at the site. The relationship was also used to estimate the average OH radical concentration during the campaign, which was compared to in-situ OH measurements (1.7 x 10^6 molecules/cm^3, 0.071 ppt) for the first time. The OH concentration ranging from 3.5 to 6.5 x 10^5 molecules/cm^3 (0.015 to 0.027 ppt) obtained with this method represents an approximation of the average OH concentration influencing the discussed VOCs from emission to measurement. Based on these findings, the average concentration of the nighttime NO3 radicals was estimated using the same approach and found to range from 2.2 to 5.0 x 10^8 molecules/cm^3 (9.2 to 21.0 ppt). During the MINATROC field campaign, in-situ ambient air measurements with the GCxGC-FID were conducted at Tenerife, Spain. Although the station is mainly situated in the free troposphere, local influences of anthropogenic and biogenic VOCs were observed. Due to a strong dust event originating from Western Africa it was possible to compare the mixing ratios during normal and elevated dust loading in the atmosphere. The mixing ratios during the dust event were found to be lower. However, this could not be attributed to heterogeneous reactions as there was a change in the wind direction from northwesterly to southeasterly during the dust event.

Conjugated polymers have attracted tremendous academical and industrial research interest over the past decades due to the appealing advantages that organic / polymeric materials offer for electronic applications and devices such as organic light emitting diodes (OLED), organic field effect transistors (OFET), organic solar cells (OSC), photodiodes and plastic lasers. The optimization of organic materials for applications in optoelectronic devices requires detailed knowledge of their photophysical properties, for instance energy levels of excited singlet and triplet states, excited state decay mechanisms and charge carrier mobilities. In the present work a variety of different conjugated (co)polymers, mainly polyspirobifluorene- and polyfluorene-type materials, was investigated using time-resolved photoluminescence spectroscopy in the picosecond to second time domain to study their elementary photophysical properties and to get a deeper insight into structure-property relationships. The experiments cover fluorescence spectroscopy using Streak Camera techniques as well as time-delayed gated detection techniques for the investigation of delayed fluorescence and phosphorescence. All measurements were performed on the solid state, i.e. thin polymer films and on diluted solutions. Starting from the elementary photophysical properties of conjugated polymers the experiments were extended to studies of singlet and triplet energy transfer processes in polymer blends, polymer-triplet emitter blends and copolymers. The phenomenon of photonenergy upconversion was investigated in blue light-emitting polymer matrices doped with metallated porphyrin derivatives supposing an bimolecular annihilation upconversion mechanism which could be experimentally verified on a series of copolymers. This mechanism allows for more efficient photonenergy upconversion than previously reported for polyfluorene derivatives. In addition to the above described spectroscopical experiments, amplified spontaneous emission (ASE) in thin film polymer waveguides was studied employing a fully-arylated poly(indenofluorene) as the gain medium. It was found that the material exhibits a very low threshold value for amplification of blue light combined with an excellent oxidative stability, which makes it interesting as active material for organic solid state lasers. Apart from spectroscopical experiments, transient photocurrent measurements on conjugated polymers were performed as well to elucidate the charge carrier mobility in the solid state, which is an important material parameter for device applications. A modified time-of-flight (TOF) technique using a charge carrier generation layer allowed to study hole transport in a series of spirobifluorene copolymers to unravel the structure-mobility relationship by comparison with the homopolymer. Not only the charge carrier mobility could be determined for the series of polymers but also field- and temperature-dependent measurements analyzed in the framework of the Gaussian disorder model showed that results coincide very well with the predictions of the model. Thus, the validity of the disorder concept for charge carrier transport in amorphous glassy materials could be verified for the investigated series of copolymers.

Die Kontrolle der ausgeprägten Aggregationsfähigkeit von alkylsubstituierten Hexa-peri-hexabenzocoronenen (HBC) wurde durch die Reduktion der intermolekularen Wechselwirkungen erreicht. Sterisch anspruchsvolle, verzweigte Alkylketten, mit einem Verzweigungspunkt naher des aromatischen Kerns, wurden in die Corona der aromatischen Scheiben eingebracht und verleihen den Derivaten Schmelzbarkeit ohne thermische Zersetzung. Dies erlaubte eine kostengünstige Verarbeitungstechniken direkt aus der Schmelze wie z.B. Zonenschmelzen, um uniaxial organisierte makroskopische Filme zu erhalten. Abhängig von dem sterischen Anspruch, der durch die Seitenkette erzeugt wird, wurden unterschiedliche molekulare Orientierungen auf Oberflächen erhalten, was eine wichtige Voraussetzung ist, um diskotische Materialien in elektronische Bauteile zu implementieren. Eine weitere Voraussetzung sind hohe Ladungsträgerbeweglichkeiten und Ladungsträgerlebenszeiten in den Halbleitermaterialien, die mit time-resolved pulse-radiolysis microwave conductivity (TR-PRMC) und time-of-flight (TOF) auch für die synthetisieren Materialien bestimmt wurden. Die neuen Materialien zeigten bereits in organischen Solarzellen gute Leistungen. Den Einfluss des Perimeters auf die elektronischen Eigenschaften der polyzyklischen aromatischen Kohlenwasserstoffe (PAKs) wurde theoretisch vorhergesagt und in dieser Arbeit durch die Synthese einer homologe Serie von PAKs experimentell bestätigt. Geht man von der „arm-chair“ Peripherie des HBC sukzessive zu einer partiellen „zickzack“ Peripherie, so findet man eine Abhängigkeit der elektronischen Banden von Symmetrie und Größe des aromatischen Systems. Die spontan ausgebildete Überstruktur dieser Derivate zeigte eine Abhängigkeit von Substitutionsmuster und der Natur der Alkylketten. Zusammenfassend wurden neben der Synthese von neuartigen Materialien für den Einsatz in der organischen Elektronik Synthesen entwickelt, die eine vereinfachte Funktionalisierung von ausgedehnten PAKs ermöglicht. Diese Konzepte erlauben eine Justierung der molekularen und supramolekularen Eigenschaften, eines der wichtigsten Voraussetzungen für den Einsatz von Materialien in elektronischen Bauelementen.

Negli ultimi anni, affianco ai succhi di frutta “classici” è sempre più importante la produzione di “nuovi succhi” che vengono formulati a partire da frutta non convenzionalmente utilizzata per la produzione di succo. Un esempio particolare è il melograno, ricco di sostanze come ellagitannini, antociani e altri derivati dell’acido gallico ed ellagico. Al melograno sono state attribuite differenti attività biologiche tra le quali effetti benefici a livello cardiovascolare ed intestinale, attività di prevenzione del cancro e altre patologie neurodegenarative. Non è ancora chiaro quali siano i costituenti che dimostrano avere tali proprietà. L’alto potere antiossidante degli ellagitannini ha fatto supporre che fossero essi stessi i responsabili di alcune attività benefiche. Tuttavia, è stato dimostrato che la loro biodisponibilità è molto bassa, in particolare, alcuni studi hanno dimostrato che essi vengono trasformati dalla flora batterica intestinale in urolitine, che, in quella forma, possono essere assorbiti nell’intestino. Per questo motivo si pensa che questi composti, formati a partire dai tannini, presenti nel melograno, siano i responsabili dell’attività. Questo lavoro sperimentale si colloca all’interno del progetto finanziato dalla Regione Emilia-Romagna; in particolare fa parte dell’OR4 “componenti alimentari che concorrano al mantenimento di uno stato di benessere fisico e riduca il rischio di contrarre malattie”. In questo lavoro sperimentale sono stati analizzati succhi di melograno ottenuti da arilli, i semi del melograno, di cultivar e provenienze geografiche diverse. Lo scopo è quello di mettere a punto metodiche analitiche strumentali avanzate (HPLC-ESI-TOF-MS) per la determinazione di antociani e altri composti fenolici nei succhi oggetto della sperimentazione e di identificare i composti presenti nei diversi campioni. La possibilità di utilizzare un analizzatore di massa come il TOF, infatti, permetterà di avere un’ottima accuratezza/sicurezza nell’identificazione dei composti fenolici nei campioni in esame. I metodi messi a punto verranno poi utilizzati per la determinazione dei composti fenolici nei diversi succhi di melograno.
