In September 1999 two short-term moorings with cylindrical sediment traps were deployed to collect sinking particles in bottom waters off the Ob and Yenisei river mouths. Samples were studied for their bulk composition, pigments, phytoplankton, microzooplankton, fecal material, amino acids, hexosamines, fatty acids and sterols and compared to suspended matter and surface sediments in order to collect information about the nature and cycling of particulate matter in the water column. Results of all measured components in sinking particles point to an ongoing seasonality in the pelagic system from blooming diatoms in the first phase to a more retention system in the second half of trap deployment. Due to a phytoplankton bloom observed north of the Ob estuary, flux rates were generally higher in the trap deployed off the Ob than off the Yenisei. The Ob trap collected fresh surface-derived particulate matter. Particles from the Yenisei trap were more degraded and resembled deep water suspension. This material may partly have been derived from resuspended sediments.

Lipids, humic acids, and kerogens were isolated from Site 582 sediment cores. The amount of each organic fraction in the samples taken from 5.6 to 694 m sub-bottom is almost unrelated to depth of the sample. A study of the oxidation of organic matter by the alkaline KMnO4 method reveals that distribution of polymethylene chain lengths of the kerogens and humic acids in the marine sediments differ from those in lacustrine sediments. The order of abundance of these chains in the sediments is: kerogens, 52-66% of total methylene chains; humic acids, 25-33%; and lipids, 8-16%. The results suggest that polyunsaturated fatty acids (>=4 double bonds) may be important in the formation of polymethylene chains of kerogens and humic acids in marine sediments.

Concerns about increasing atmospheric CO2 concentrations and global warming have initiated studies on the consequences of multiple-stressor interactions on marine organisms and ecosystems. We present a fully-crossed factorial mesocosm study and assess how warming and acidification affect the abundance, body size, and fatty acid composition of copepods as a measure of nutritional quality. The experimental set-up allowed us to determine whether the effects of warming and acidification act additively, synergistically, or antagonistically on the abundance, body size, and fatty acid content of copepods, a major group of lower level consumers in marine food webs. Copepodite (developmental stages 1-5) and nauplii abundance were antagonistically affected by warming and acidification. Higher temperature decreased copepodite and nauplii abundance, while acidification partially compensated for the temperature effect. The abundance of adult copepods was negatively affected by warming. The prosome length of copepods was significantly reduced by warming, and the interaction of warming and CO2 antagonistically affected prosome length. Fatty acid composition was also significantly affected by warming. The content of saturated fatty acids increased, and the ratios of the polyunsaturated essential fatty acids docosahexaenoic- (DHA) and arachidonic acid (ARA) to total fatty acid content increased with higher temperatures. Additionally, here was a significant additive interaction effect of both parameters on arachidonic acid. Our results indicate that in a future ocean scenario, acidification might partially counteract some observed effects of increased temperature on zooplankton, while adding to others. These may be results of a fertilizing effect on phytoplankton as a copepod food source. In summary, copepod populations will be more strongly affected by warming rather than by acidifying oceans, but ocean acidification effects can modify some temperature impacts

Fatty acid binding proteins (FABPs) exhibit a β-barrel topology, comprising 10 antiparallel β-sheets capped by two short α-helical segments. Previous studies suggested that fatty acid transfer from several FABPs occurs during interaction between the protein and the acceptor membrane, and that the helical domain of the FABPs plays an important role in this process. In this study, we employed a helix-less variant of intestinal FABP (IFABP-HL) and examined the rate and mechanism of transfer of fluorescent anthroyloxy fatty acids (AOFA) from this protein to model membranes in comparison to the wild type (wIFABP). In marked contrast to wIFABP, IFABP-HL does not show significant modification of the AOFA transfer rate as a function of either the concentration or the composition of the acceptor membranes. These results suggest that the transfer of fatty acids from IFABP-HL occurs by an aqueous diffusion-mediated process, i.e., in the absence of the helical domain, effective collisional transfer of fatty acids to membranes does not occur. Binding of wIFABP and IFABP-HL to membranes was directly analyzed by using a cytochrome c competition assay, and it was shown that IFABP-HL was 80% less efficient in preventing cytochrome c from binding to membranes than the native IFABP. Collectively, these results indicate that the α-helical region of IFABP is involved in membrane interactions and thus plays a critical role in the collisional mechanism of fatty acid transfer from IFABP to phospholipid membranes.

Sterol regulatory element-binding protein-1c (SREBP-1c) enhances transcription of genes encoding enzymes of unsaturated fatty acid biosynthesis in liver. SREBP-1c mRNA is known to increase when cells are treated with agonists of liver X receptor (LXR), a nuclear hormone receptor, and to decrease when cells are treated with unsaturated fatty acids, the end products of SREBP-1c action. Here we show that unsaturated fatty acids lower SREBP-1c mRNA levels in part by antagonizing the actions of LXR. In cultured rat hepatoma cells, arachidonic acid and other fatty acids competitively inhibited activation of the endogenous SREBP-1c gene by an LXR ligand. Arachidonate also blocked the activation of a synthetic LXR-dependent promoter in transfected human embryonic kidney-293 cells. In vitro, arachidonate and other unsaturated fatty acids competitively blocked activation of LXR, as reflected by a fluorescence polarization assay that measures ligand-dependent binding of LXR to a peptide derived from a coactivator. These data offer a potential mechanism that partially explains the long-known ability of dietary unsaturated fatty acids to decrease the synthesis and secretion of fatty acids and triglycerides in livers of humans and other animals.

Endogenous ligands of cannabinoid receptors have been discovered recently and include some N-acylethanolamines (NAEs; e.g., N-arachidonoylethanolamine) and some 2-acylglycerols (e.g., sn-2-arachidonoylglycerol). Previously, we found these compounds to be active biologically when administered per os in large quantities to mice. In the present work, piglets were fed diets with and without 20:4n−6 and 22:6n−3 fatty acid precursors of NAEs, in levels similar to those found in porcine milk, during the first 18 days of life, and corresponding brain NAEs were assessed. In piglets fed diets containing 20:4n−6 and 22:6n−3, there were increases in several biologically active NAEs in brain homogenates—20:4n−6 NAE (4-fold), 20:5n−3 NAE (5-fold), and 22:5n−3 and 22:6n−3 NAE (9- to 10-fold). These results support a mechanism we propose for dietary long-chain polyunsaturated fatty acids influences on brain biochemistry with presumed functional sequelae. This paradigm will enable targeted investigations to determine whether and why specific populations such as infants, elderly, or persons suffering from certain clinical conditions may benefit from dietary long-chain polyunsaturated fatty acids.

Peroxisome proliferators induce stearoyl-CoA desaturase activity (EC 1.14.99.5) in liver [Kawashima, Y., Hanioka, N., Matsumura, M. & Kozuka, H. (1983) Biochim. Biophys. Acta 752, 259-264]. We analyzed the changes in stearoyl-CoA desaturase 1 (SCD1) mRNA to further define the molecular mechanism for the induction of stearoyl-CoA desaturase by peroxisome proliferators. SCD1 mRNA was analyzed from the livers of BALB/c mice that had been fed diets supplemented with clofibrate or gemfibrozil. Clofibrate was found to induce liver SCD1 mRNA levels 3-fold within 6 hr to a maximum of 22-fold in 30 hr. Gemfibrozil administration resulted in a similar induction pattern. This induction is primarily due to an increase in transcription of the SCD1 gene, as shown by nuclear run-on transcription assays and DNA deletion analysis of transfected SCD1-chloramphenicol acetyltransferase fusion genes. The cis-linked response element for peroxisome proliferator-activated receptor (PPAR) was localized to an AGGTCA consensus sequence between base pairs -664 to -642 of the SCD1 promoter. Clofibrate-mediated induction of SCD1 mRNA was shown to be independent of polyunsaturated fatty acids, with peroxisome proliferators and arachidonic acid having opposite effects on SCD1 mRNA levels. Additionally, the activation of SCD1 mRNA by clofibrate was inhibited 77% by cycloheximide administration. Levels of liver beta-actin and albumin mRNAs were unchanged by these dietary manipulations. Our data show that hepatic SCD1 gene expression is regulated by PPARs and suggest that peroxisome proliferators and poly-unsaturated fatty acids act through distinct mechanisms.

It has been suggested that transepithelial gradients of short-chain fatty acids (SCFAs; the major anions in the colonic lumen) generate pH gradients across the colonic epithelium. Quantitative confocal microscopy was used to study extracellular pH in mouse distal colon with intact epithelial architecture, by superfusing tissue with carboxy SNARF-1 (a pH-sensitive fluorescent dye). Results demonstrate extracellular pH regulation in two separate microdomains surrounding colonic crypts: the crypt lumen and the subepithelial tissue adjacent to crypt colonocytes. Apical superfusion with (i) a poorly metabolized SCFA (isobutyrate), (ii) an avidly metabolized SCFA (n-butyrate), or (iii) a physiologic mixture of acetate/propionate/n-butyrate produced similar results: alkalinization of the crypt lumen and acidification of subepithelial tissue. Effects were (i) dependent on the presence and orientation of a transepithelial SCFA gradient, (ii) not observed with gluconate substitution, and (iii) required activation of sustained vectorial acid/base transport by SCFAs. Results suggest that the crypt lumen functions as a pH microdomain due to slow mixing with bulk superfusates and that crypts contribute significant buffering capacity to the lumen. In conclusion, physiologic SCFA gradients cause polarized extracellular pH regulation because epithelial architecture and vectorial transport synergize to establish regulated microenvironments.

O objetivo deste trabalho foi avaliar os efeitos da inclusão de caroço de algodão e vitamina E em dietas para bovinos confinados por 83, 104 e 111 dias sobre as características da carcaça, qualidade da carne e características sensoriais da carne in natura e de hambúrgueres. Foram utilizados 54 bovinos da raça Nelore, machos, não castrados, com média de 350 kg ± 30 kg de peso vivo inicial e 24 meses de idade, divididos em três grupos de acordo com o peso vivo inicial e distribuídos em três dietas: dieta sem inclusão de caroço de algodão (C), dieta contendo 30% MS de caroço de algodão (CA) e dieta contendo 30% MS de caroço de algodão e 500 UI de vitamina E/kg de matéria seca da ração (CAE). As dietas empregadas foram compostas de diferentes concentrados, incluindo milho grão seco, polpa cítrica, bagaço de cana cru e farelo de soja, com relação volumoso:concentrado de 14:86 e média de 16% PB nas três dietas. Os animais abatidos aos 83 dias de confinamento eram correspondentes ao grupo de maior peso vivo, já os animais mais leves foram abatidos aos 111 dias de confinamento. O experimento foi em arranjo fatorial 3 x 3, considerando três dietas e três períodos de confinamento, totalizando 9 tratamentos. Os animais que permaneceram em confinamento por mais tempo apresentaram maior rendimento de carcaça (56,33%) e carnes mais macias (3,02 kg). As dietas CA e CAE resultaram em menor força de cisalhamento (3,15 e 3,31 kg, respectivamente). A inclusão de vitamina E se mostrou indiferente para a cor e TBARS, em relação à dieta CA. A inclusão do caroço de algodão em dietas para bovinos não influenciou no teor de ácidos graxos saturados, porém aumentou os teores de alguns ácidos graxos poli-insaturados (AGPI) nas carnes. Por outro lado, houve um aumento linear nos níveis de ácidos graxos saturados (AGS) à medida que aumentaram os dias de confinamento. Em relação às características sensoriais, no teste descritivo, as carnes in natura dos animais alimentados com CA e CAE foram mais macias e suculentas (P < 0,05), porém apresentaram um sabor mais intenso e os provadores treinados detectaram um sabor estranho nestas carnes (P < 0,05). No entanto, para o teste afetivo, apenas foi detectado sabor estranho para os hambúrgueres provenientes da dieta CA, os atributos aroma e aroma estranho não foram influenciados pelas dietas (P > 0,05). Por meio do teste discriminativo, foi observado que os hambúrgueres provenientes de animais alimentados com CA e CAE por 104 e 111 dias de confinamento apresentaram diferença em relação ao sabor quando comparados aos hambúrgueres de animais do grupo C destes mesmos períodos, e que os hambúrgueres da dieta CA não apresentaram diferença de sabor quanto aos dias de confinamento. A inclusão de 30% MS de caroço de algodão mostrou ser uma boa alternativa para melhorar as características físico-químicas da carne e seu perfil de ácidos graxos, entretanto atribui um sabor estranho ao produto final, independente do período de confinamento, sendo perceptível ao consumidor e reduzindo aceitabilidade. Ao longo dos dias de confinamento, a carne dos animais se torna menos saudável, do ponto de vista de composição lipídica, ao consumo humano. A adição de 500 UI de vitamina E em dietas contendo caroço de algodão mostrou-se desnecessária
