Los residuos sólidos como consecuencia directa de las actividades humanas, han representado un reto desde sus inicios, de ahí se empezaron a desarrollar técnicas y prácticas para garantizar una adecuada gestión de los residuos, nuestros primeros ancestros encontraron en los botaderos a cielo abierto la solución más fácil para disponer los desechos de animales y residuos de comida en sitios cercanos a sus viviendas, por ejemplo, los griegos poseen registros de la inauguración del primer relleno municipal en la ciudad de Atenas hace más de 2000 años. La evolución de las culturas, la revolución industrial y el crecimiento poblacional acelerado han modificado no solo la composición sino también la cantidad de los residuos sólidos producidos, haciendo cada día más difícil su adecuada gestión debido a su alto grado de toxicidad, la ausencia de materiales biodegradables y su volumen, traduciendo esto en un gran impacto. El panorama no es muy alentador y los pronósticos emitidos por el Banco Mundial prevén que para el año 2025 los niveles mundiales de residuos sólidos urbanos alcanzaran 2,2 millones de toneladas por año. Basada en el interés por la adecuada gestión de los residuos y la disminución del impacto ambiental de las empresas Coraseo S.A y Seacor S.A, la presente investigación busca seleccionar la metodología y/o tecnología más apropiada teniendo en cuenta las condiciones económicas, logísticas y de infraestructura de la empresa y la comunidad involucrada. En primera instancia se estudió en profundidad el panorama empresarial y posteriormente se seleccionó un grupo de cuatro propuestas para la transformación y aprovechamiento de los residuos sólidos que fueron evaluadas a nivel económico, normativo, medio ambiental. Los datos analizados sugieren entre otras cosas, un robustecimiento del marco legal ambiental colombiano y mundial, una mayor colaboración económica de los organismos internacionales y las entidades bancarias y finalmente una reducción en los costos de las tecnologías disponibles para el aprovechamiento de los residuos sólidos, haría posible el sueño de las sociedades de gestionar efectivamente los residuos sólidos generando el menor impacto ambiental posible y en el caso del grupo empresarial Cordobés lograr innovar y emprender con un proyecto eco eficiente como el que se evaluó.

Changes in fish assemblage structure caused by human activities, such as fishing, can alter trophic relations in fish assemblages. In this context, Marine Protected Areas (MPA) are efficient tools for habitat recovery and ideal environments for evaluating changes on the trophic structure resulting from human activities. The present work targeted fish assemblages from two no-take MPAs from the northern half of South Alentejo and Costa Vicentina Marine Park, established in 2011. Previous works reported positive effects on local fish assemblages after no-take MPA designation, and it is therefore important to further study its impact on local fish assemblages, especially concerning trophic interactions. Local fish assemblages were sampled (summer 2011, winter 2012, summer 2013 and winter 2013) using trammel nets. Diets were characterized and digestive tract contents of the 10 most abundant fish species were compared between the no take MPAs (treatment) and adjacent areas (controls), and changes evaluated as a function of time since protection. Results revealed significant differences between the diets of fish from protected and non protected areas, with crabs being the preferential prey in both protected and control areas but being more ingested outside the no-take areas. However, these differences were evident since the beginning of the study. Fish assemblages from the northern area presented significantly larger niche breadth and significantly increasing with time. This way, the main effects of no-take MPA implementation were directly visible on the niche breadth but did not directly impact the diet composition of the sampled fish assemblages, contributing however to reinforce the already naturally existent differences. This work provides important information regarding the effect of changes in the fish assemblage caused by MPA designation on the trophic ecology of fish.

Crowd induced dynamic loading in large structures, such as gymnasiums or stadiums, is usually modelled as a series of harmonic loads which are defined in terms of their Fourier coefficients. Different values of these Fourier coefficients that were obtained from full scale measurements can be found in codes. Recently, an alternative has been proposed, based on random generation of load time histories that take into account phase lags among individuals inside the crowd. Generally the testing is performed on platforms or structures that can be considered rigid because their natural frequencies are higher than the excitation frequencies associated with crowd loading. In this paper we shall present the testing done on a structure designed to be a gymnasium, which has natural frequencies within that range. In this test the gym slab was instrumented with acceleration sensors and different people jumped on a force plate installed on the floor. Test results have been compared with predictions based on the two abovementioned load modelling alternatives and a new methodology for modelling jumping loads has been proposed in order to reduce the difference between experimental and numerical results at high frequency range.

Periodontitis is an inflammatory disease that causes osteolysis and tooth loss. It is known that the nuclear factor kappa B (NF-κB) signalling pathway plays a key role in the progression of inflammation and osteoclastogenesis in periodontitis. Parthenolide (PTL), a sesquiterpene lactone extracted from the shoots of Tanacetum parthenium, has been shown to possess anti-inflammatory properties in various diseases. In the study reported herein, we investigated the effects of PTL on the inflammatory and osteoclastogenic response of human periodontal ligament-derived cells (hPDLCs) and revealed the signalling pathways in this process. Our results showed that PTL decreased NF-κB activation, I-κB degradation, and ERK activation in hPDLCs. PTL significantly reduced the expression of inflammatory (IL-1β, IL-6, and TNF-α) and osteoclastogenic (RANKL, OPG, and M-CSF) genes in LPS-stimulated hPDLCs. In addition, PTL attenuated hPDLC-induced osteoclastogenic differentiation of macrophages (RAW264.7 cells), as well as reducing gene expression of osteoclast-related markers in RAW264.7 cells in an hPDLC-macrophage coculture model. Taken together, these results demonstrate the anti-inflammatory and antiosteoclastogenic activities of PTL in hPDLCs in vitro. These data offer fundamental evidence supporting the potential use of PTL in periodontitis treatment.

The research project developed a quantitative approach to assess the risk to human health from heavy metals and polycyclic aromatic hydrocarbons in urban stormwater based on traffic and land use factors. The research outcomes are expected to strengthen the scientifically robust management and reuse of urban stormwater. The innovative methodology developed can be applied to evaluate human health risk in relation to toxic chemical pollutants in urban stormwater runoff and for the development of effective risk mitigation strategies.

Biomechanical signals due to human movements during exercise are represented in time-frequency domain using Wigner Distribution Function (WDF). Analysis based on WDF reveals instantaneous spectral and power changes during a rhythmic exercise. Investigations were carried out on 11 healthy subjects who performed 5 cycles of sun salutation, with a body-mounted Inertial Measurement Unit (IMU) as a motion sensor. Variance of Instantaneous Frequency (I.F) and Instantaneous Power (I.P) for performance analysis of the subject is estimated using one-way ANOVA model. Results reveal that joint Time-Frequency analysis of biomechanical signals during motion facilitates a better understanding of grace and consistency during rhythmic exercise.

Three new dibenzocyclooctadiene lignans, schilancifolignans A-C (1-3), together with thirteen known ones, were isolated from the leaves and stems of Schisandru lancifolia. The structures of 1-3 were elucidated by spectroscopic methods, including extensive 1D- and 2D-NMR techniques. Compounds 1-3 were tested for their anti-human immunodeficiency virus-1 activities and showed weak bioactivities.

Human lactoferrin (hLF) is an iron-binding protein with antimicrobial and immunomodulatory activities. hLF cDNA was transferred into grass carp via electroporated sperm. The production of transgenic fish was as high as 55% tinder the best parameters. 2(11) pulses and 20-min incubation. The expression of the transgene was demonstrated by the detection of hLF mRNA by RT-PCR. We also investigated the response of G(0) transgenic grass carp to Aeromonas hydrophila infection. Serum lysozyme activities (P>0.05) and phagocytic activities of kidney cells (P<0.05) were measured in transgenic individuals. The transgenic fish not only cleared A. hydrophila significantly faster than the control carp (P<0.05), but also showed enhanced phagocytic activities. The result shows that hLF has immunomodulatory activities in hLF-transgenic grass carp. The transgenic grass carp exhibited enhanced immunity to A. hydrophila infection. These results reveal that the mechanisms of disease resistance are different between hLF-transgenic plants and hLF-transgenic grass carp. (C) 2004 Elsevier B.V. All rights reserved.

The human coronavirus 229E (HCoV-229E) replicase gene-encoded nonstructural protein 13 (nsp13) contains an N-terminal zinc-binding domain and a C-terminal superfamily 1 helicase domain. A histidine-tagged form of nsp13, which was expressed in insect cells and purified, is reported to unwind efficiently both partial-duplex RNA and DNA of up to several hundred base pairs. Characterization of the nsp13-associated nucleoside triphosphatase (NTPase) activities revealed that all natural ribonucleotides and nucleotides are substrates of nsp13, with ATP, dATP, and GTP being hydrolyzed most efficiently. Using the NTPase active site, HCoV-229E nsp13 also mediates RNA 5'-triphosphatase activity, which may be involved in the capping of viral RNAs.

The human coronavirus 229E replicase gene encodes a protein, p66HEL, that contains a putative zinc finger structure linked to a putative superfamily (SF) 1 helicase. A histidine-tagged form of this protein, HEL, was expressed using baculovirus vectors in insect cells. The purified recombinant protein had in vitro ATPase activity that was strongly stimulated by poly(U), poly(dT), poly(C), and poly(dA), but not by poly(G). The recombinant protein also had both RNA and DNA duplex-unwinding activities with 5'-to-3' polarity. The DNA helicase activity of the enzyme preferentially unwound 5'-oligopyrimidine-tailed, partial-duplex substrates and required a tail length of at least 10 nucleotides for effective unwinding. The combined data suggest that the coronaviral SF1 helicase functionally differs from the previously characterized RNA virus SF2 helicases.

Members of the Burkholderia cepacia complex can secrete proteases, lipases, and hemolysins. We report in this study the identification of a general secretory pathway present in a B. vietnamiensis (formerly genomovar V) clinical isolate, which is required for the efficient secretion of phospholipase C and hemolysin activities. Southern blot hybridization experiments revealed that this general secretion pathway is highly conserved among the different genomovars of the B. cepacia complex and is homologous to a similar system described in B. pseudomallei. We also show that this pathway appears not to be necessary for intracellular survival of B. vietnamiensis within Acanthamoeba polyphaga.

Chemostat culture was used to determine the effects of the antimicrobial agents tetracycline and nystatin on predominant components of the human gut microflora. Their addition to mixed culture systems caused a non-specific, and variable, decrease in microbial populations, although tetracycline allowed an increase in numbers of yeasts. Both had a profound inhibitory effect upon populations seen as important for gut health (bifidobacteria, lactobacilli). However, a tetracycline resistant Lactobacillus was enriched from the experiments. A combination of genotypic and phenotypic characterisations confirmed its identity as Lactobacillus plantarum. This strain exerted powerful inhibitory effects against Candida albicans. Because of its ability to resist the effects of tetracycline, this organism may be useful as a probiotic for the improved management of yeast related conditions such as thrush and irritable bowel syndrome.
