Enterobacter species commonly occur in the environment and are recognized as opportunistic human pathogens in clinical settings. However, with the exception of Enterobacter sakazakii (Cronobacter), Enterobacter species are not normally considered foodborne pathogens. Cronobacter are particularly associated with illness in infants, particularly within the first 3 months after birth. Therefore, although Cronobacter are found in a wide range of fresh and dried food materials, it is their contamination of the infant formula production chain that is the major cause for concern. Cronobacter are noted for their ability to survive during desiccation and their persistence in dried infant food for at least 2 years.

This study investigated and characterised transdermal permeation of bioactive agents from a topically applied Arnica montana tincture. Permeation experiments conducted over 48 h used polydimethylsiloxane (silastic) and human epidermal membranes mounted in Franz-type diffusion cells with a methanol-water (50:50 v/v) receptor fluid. A commercially available tincture of A. montana L. derived from dried Spanish flower heads was a donor solution. Further donor solutions prepared from this stock tincture concentrated the tincture constituents 1, 2 and 10 fold and its sesquiterpene lactones 10 fold. Permeants were assayed using a high-performance liquid chromatography method. Five components permeated through silastic membranes providing peaks with relative retention factors to an internal standard (santonin) of 0.28, 1.18, 1.45, 1.98 and 2.76, respectively. No permeant was detected within 12 h of applying the Arnica tincture onto human epidermal membranes. However, after 12 h, the first two of these components were detected. These were shown by Zimmermann reagent reaction to be sesquiterpene lactones and liquid chromatography/diode array detection/mass spectrometry indicated that these two permeants were 11,13-dihydrohelenalin (DH) analogues (methacrylate and tiglate esters). The same two components were also detected within 3 h of topical application of the 10-fold concentrated tincture and the concentrated sesquiterpene lactone extract.

Ctra. San Vicente. Finca Santa Engracia. 06071 Badajoz, Spain. Worldwide the consumption of fruit and vegetables is increasing due to the dietary guidelines recommended by nutritionist. Because of their high content on phenols, vitamins, mineral and antioxidants, berry fruits are consumed not only in fresh forms but also as processed and derivative products such as juices, yogurts, jellies and dried fruits. As a high consumed red fruit, sweet cherry has been the focus on some studies, mainly regarding bioactive compounds content. “Sweetheart” cherries from São Julião region (Alentejo, Portugal) from two different production campaigns were kept in different storage conditions in order to evaluate both the environmental and storage effect on some physical-chemical properties and phenolics. Cold conditions - Cold (1 ºC, 95% RH) and modified atmosphere - MA (1 ºC, 95% RH with micro-perforated bags of Pplus®, Sidlaw Packaging, Bristol, UK) were tested. In order to establish the appropriate storage conditions, individual phenolic acids and physical-chemical properties were analysed during two consecutive years. Results show a general decrease on phenolic compounds content between cherries from both years. It is also observed that MA conditions do not affect significantly both phenolics and physical-chemical parameters when compared with Cold conditions. Additionally, it is observed similar behaviour on Cold and MA sweet cherries regarding its pH, total soluble solids content (TSS), titratable acidity (TA) and colour and individual phenols during storage time. Concluding, these results show, as expected, changes between cultivars which may be correlated with the environmental conditions on different years. Este trabalho abordou a valorização de um subproduto da indústria de lacticínios (soro de queijo) através da alteração do funcionamento de processos habitualmente utilizados no contexto do tratamento biológico. Foi avaliada a fermentação acidogénica deste subproduto para maximizar a conversão do seu elevado teor de matéria orgânica em ácidos orgânicos voláteis (AOV) que actualmente são produtos com elevada procura, nomeadamente para produção de polihidroxialcanoatos (PHA). Em ensaios descontínuos e semi-contínuos foi caracterizada a produção e composição de AOV a partir de soro de queijo variando a razão food-to-microorganism (F/M) e a concentração de alcalinidade. Recorrendo à modelação dos resultados através de superfícies de resposta, demonstrou-se que condições de F/M = 4 gCQO g-1SSV combinadas com uma adição elevada de alcalinidade (8 g L-1 expresso como CaCO3) resultaram na conversão de 72% da CQO alimentada em AOV. O acetato e o butirato foram os AOV predominantes (60%), mas elevadas razões F/M combinadas com elevadas alcalinidades promoveram o alongamento da cadeia carboxílica, tendo sido produzidos AOV de maior massa molecular (iso-valerato e n-caproato). O processo de fermentação acidogénica foi posteriormente desenvolvido em modo contínuo num reactor MBBR acidogénico operado a longo prazo. Cargas orgânicas entre 30 e 50 gCQO L-1d-1 permitiram obter um grau de acidificação máximo de 68% no efluente fermentado. Foi ainda demonstrado que uma adição dinâmica de alcalinidade (0 – 4,8 g CaCO3 L-1) nestas condições estimulou a produção de AOV de cadeia ímpar (propionato e n-valerato) até 42%. O efluente acidificado no processo anaeróbio foi usado como substrato em reactores SBR operados para selecção de culturas microbianas mistas acumuladoras de PHA, nos quais foi aplicado um regime de alimentação dinâmica em condições aeróbias (“fartura-fome”). Estes sistemas operaram também a longo prazo, e demonstraram ser capazes de remover mais de 96% da CQO alimentada e simultaneamente convertê-la em PHA, até 36% do peso celular seco. A velocidade de remoção de substrato (valor máximo de 1,33 gCQO g-1SSV h-1) foi proporcional ao teor de polímero acumulado, evidenciando o estabelecimento de uma fase de “fome” prolongada que estimulou a selecção de microrganismos com elevada capacidade de acumulação de PHA. Além disso, o teor molar de hidroxivalerato (HV) no copolímero produzido [P(HB-co-HV)] foi directamente proporcional ao teor de AOV de cadeia ímpar (propionato e n-valerato) presente no soro fermentado que serviu de substrato. Uma estratégia de operação do reactor SBR com variação da carga orgânica, aliada ao regime “fartura-fome” estabelecido, permitiu ainda simular a realidade dos processos de tratamento biológico de efluentes, nos quais a composição e concentração inicial de matéria orgânica variam frequentemente. Este modo de operação do sistema estimulou notavelmente o processo de selecção de culturas acumuladoras de PHA tendo resultado num aumento da acumulação de PHA de 7% para 36%. Os resultados demonstraram com sucesso a possibilidade de valorização do soro de queijo através de eco-biotecnologia, contribuindo para uma mudança de paradigma no tratamento convencional de efluentes: ao invés de serem eliminados enquanto poluentes, os componentes orgânicos presentes neste subproduto industrial podem assim ser convertidos em materiais de valor acrescentado.

The use of plants with medicinal purposes is an ancient practice still very common in developing regions, and is rapidly spreading in industrialized countries. This fact is evidenced by the large number of ethnobotanical studies found in the literature referring that these plants are often used as decoctions and infusions. In most studies the reported biological activities are attributed to the presence of phenolic compounds, due to their antioxidant properties, and to polysaccharides, with its anti-tumoral properties. In “Trás-os-Montes” region, some of the most popular infusions used by the popular medicine are prepared with the dried leaves of Fraxinus angustifolia, the dried shoots of Mentha suaveolens, and the dried inflorescences of Pterospartum tridentatum. However, there are no studies about the polysaccharides present in these infusions. Thus, through the structural characterization of the polysaccharides present in the infusions of F. angustifolia, M. suaveolens, and P. tridentatum, the present PhD thesis intends to evaluate the possible relation between polysaccharides and the immunostimulatory activity that these infusions might present. In a preliminary phase, infusions of F. angustifolia were prepared according to the popular tradition, and it was observed that the obtained water soluble material contained approximately 85% of material non-retained in C18 cartridges, with hydrophilic characteristics, with the remaining 15% comprising retained-material with hydrophobic characteristics. It was also shown that the infusions only contained between 2 and 4% of high molecular weight material (HMWM), which comprised approximately 30% of carbohydrate material. Sugar and methylation analysis of the HMWM suggested the presence of pectic polysaccharides, together with type II arabinogalactans, mannans, and xyloglucans. However, the amount of material obtained is to low for the fractionation, and structural analysis of the polysaccharides present. The 4 h decoction, divided in two periods of 2 h, with water renewal, allowed to increase the HMWM yield, relatively to the infusions traditional infusions. It was also observed that the decoction also allowed to increase the HMWM proportion of carbohydrate material, due to an increase in the proportion of uronic acid present, although the neutral sugar residues seemed to be detected in similar proportions. Therefore, in all the experiments subsequently performed, the HMWM used was obtained through the decoction of F. angustifolia dried leaves, M. suaveolens dried shoots, and P. tridentatum dried inflorescences. x After the fractionation, through ethanol precipitation, and anion exchange chromatography, of the polysaccharides from the HMWM obtained by the decoction of the vegetable material of the distinct studied plants, it was observed the presence of high proportions of pectic polysaccharides, containing type I arabinogalactans, together with minor proportions of type II arabinogalactans, mannans, and xyloglucans. The presence of pectic polysaccharides in the extracts from F. angustifolia was also evidenced through endo-polygalacturonase treatment, and ESI-MS and ESI-MS/MS experiments. The detection of linked pentose and uronic acid residues, also seemed to suggest the presence of xylogalacturonan domains in the pectic polysaccharides from F. angustifolia. The extracts from F. angustifolia dried leaves also contained type II arabinogalactans that exhibited a higher structural diversity than those detected in the M. suaveolens, and P. tridentatum extracts, particularly in the substitution degree of the galactan backbone, and in the extension of the (1→5)-Araf side chains. Moreover, for all the plants studied, it was also observed that the type II arabinogalactans, extracted during the 2nd 2h of the extraction process, exhibited a substitution degree of the galactan backbone higher than those extracted during the 1st 2h. The extracts from P. tridentatum dried inflorescences contained higher proportions of mannans, and also of xyloglucans, both presenting a substitution degree higher than those, which were detected in lower proportion in the extracts of F. angustifolia and M. suaveolens. Through ESI-MS and ESI-MS/MS it was possible to evidence that the mannans present in the extracts of P. tridentatum presented acetyl groups on the O-2 of the mannosyl residues. It was also evidenced that the P. tridentatum mannans were more extensively acetylated than the mannans detected in the coffee infusion, LBG, and other non-conventional mannan sources. Moreover, it was detected the presence of oligosaccharides comprising hexose residues linked to non acetylated pentose residues, suggesting the possible presence of arabinose residues in the mannans from P. tridentatum extracts. The immunostimulatory activity of three fractions isolated from the extracts of F. angustifolia, M. suaveolens, and P. tridentatum, was tested and an increase in the NO production by macrophages, without compromising their cellular viability, was observed. The type I, and type II arabinogalactans detected in the extracts from F. angustifolia, and M. suaveolens seem to have contributed for the observed immunostimulatory activity. For the fraction from P. tridentatum, the mannans acetylation, and the presence of type I, and type II arabinogalactans seemed to contribute for the macrophage immunostimulatory activity observed. The possible presence of storage xyloglucans from the inflorescences seeds, also seems to have contributed for the immunostimulatory activity registered when the macrophages were stimulated with higher extract concentrations. The results obtained allow to conclude that the extracts of F. angustifolia dried leaves, M. suaveolens dried shoots, and P. tridentatum dried inflorescences contained high proportions of pectic polysaccharides, exhibiting type I arabinogalactans, together with other polysaccharides, such as type II arabinogalactans, mannans, and xyloglucans. This polysaccharide mixture seems to have contributed to the immunostimulatory activity of fractions isolated from the extracts of the studied plants. Therefore, as the same type of polysaccharides seem to be present in the decoctions and in the infusions, it seems possible that the polysaccharides might contribute for the therapeutic properties frequently associated by the popular tradition to the infusions of these plants.

O estágio realizado teve como premissa principal perceber o comportamento da planta face ao seu crescimento em altura da própria planta e em número em função de diferentes épocas de sementeira, e de condições de cultivo, nomeadamente de tipos de solo. O trabalho realizado inclui todo o processo desde a apanha, preparação das sementes e dos campos, e a sementeira propriamente dita. Foi acompanhado o crescimento das plantas em 3 alturas diferentes, por contagem do número de plantas e do comprimento das plantas aéreas. Igualmente foram realizadas análises da granulometria dos solos. Determinaram-se os metais pesados nos solos e nas plantas, assim como os açúcares após a hidrólise e derivatização das amostras liofilizadas, tanto da raíz como da parte aérea. Foi possível concluir que a planta nasce com maior abundância em solos franco-siltosos do que em solos franco-arenosos, onde o seu crescimento é muito pobre ou mesmo nulo. Os valores de cádmio e chumbo encontram-se a níveis adequados para o consumo humano. Coloca-se a hipótese da existência de arabinoglucoronoxilanas como constituinte da raiz desta planta.

The Mediterranean Diet concept was formulated during the sixties, in association with the food consumption pattern of Mediterranean areas that produced olive oil and shared common health styles. These areas, besides their own cultural and religious differences, share common food habits, such as: - The use of olive oil (supplier of monounsaturated fatty acids and antioxidants); - The abundant use of cereals, mainly under the form of excellent quality bread, flour and pasta (providing fibre and energy); - Large and variegate consumption of fruit (fresh and dried), nuts and vegetables (colourful, rich in fibre, antioxidants and other protective materials); - Abundant use of herbs and spices (rich in antioxidants and other protective materials); - Simple culinary methods, using short cooking times and low temperatures (which enhance the preservation of food nutritional and sensorial characteristics). The Mediterranean Diet reflects a set of characteristics that make it internationally recognized as a health promoter eating pattern, where the relation between monounsaturated and saturated fatty acids is highly advantageous for the former, fibre, vitamins and natural antioxidants intake is high, together with a low consumption of animal protein and salt. The obtained results show contents in protein, lipid and carbohydrates very adequate to the “DRI”; The relation between mono and saturated fatty acids (40:9) should be emphasised, together with the high fibre content. Protective nutrients show remarkable results, with a wide variety of vitamins and minerals, in particular Vitamin A, complex B vitamins, biotin, vit. E, folic acid, iron, manganese and selenium, that are widely recognised as important antioxidants and responsible for the good function of the immune system. In conclusion, tomato soup, consumed traditionally as a poor meal, shows to be a health promoter nutritionally complete recipe.

En alimentation animale, l’utilisation adéquate des aliments nécessite une connaissance précise des valeurs nutritionnelles de leurs composantes, dont celle en acides aminés. Cependant, ces valeurs nutritionnelles dépendent de la teneur en acides aminés essentiels (AAE) totaux et de la digestibilité iléale standardisée (DIS) de ces AAE. Cette dernière varie en fonction de plusieurs facteurs, dont l’origine botanique des graines, les conditions de culture des récoltes, le stockage des aliments, les traitements physico-chimiques appliqués aux grains, les facteurs antinutritionnels (FANs) et les techniques expérimentales utilisées pour le dosage et l’estimation de la digestibilité des AAE. Une approche par méta-analyse a permis d’établir des modèles de prédiction de la valeur nutritionnelle en AAE des ingrédients à partir de leur composition chimique en considérant la protéine brute (PB), les AAE totaux, la teneur en fibre (Acid Detergent Fiber (ADF), Neutral Detergent Fiber (NDF) et Fibre Brute (FB)) et les FANs comme les inhibiteurs de la trypsine. En se référant à l’analyse graphique et statistique, les données ont été réparties en 4 groupes : 1) les tourteaux (tourteau de soja, colza/canola et coton); 2) les légumineuses (féveroles, lupins, pois et soja); 3) les céréales (blé, orge, avoine, sorgho et maïs); 4) les drêches de distilleries (blé et maïs). Ainsi, un modèle général ajusté en fonction du type d’ingrédients a été généré et les facteurs de variation de la digestibilité en AAE ont été identifiés. Pour les tourteaux, la DIS des AAE est réduite par un accroissement de la teneur en NDF, tandis que la DIS des AAE de la féverole, pois et lupin est principalement influencée par la teneur en PB et en FANs. Concernant les graines de soja la DIS des AAE est réduite par une hausse de la teneur en fibre (FB et ADF). Enfin pour les céréales et les sous- produit de céréales telles que les drêches, la PB et les fibres (ADF ou NDF) étaient les meilleurs nutriments pour prédire la DIS des AAE. Ces résultats démontrent que la DIS des AAE peut être prédite avec précision à partir de la composition chimique pour la plupart des ingrédients.

Pseudotype viruses (PVs) are chimeric, replication-deficient virions that mimic wild-type virus entry mechanisms and can be safely employed in neutralisation assays, bypassing the need for high biosafety requirements and performing comparably to established serological assays. However, PV supernatant necessitates -80°C long-term storage and cold-chain maintenance during transport, which limits the scope of dissemination and application throughout resource-limited laboratories. We therefore investigated the effects of lyophilisation on influenza, rabies and Marburg PV stability, with a view to developing a pseudotype virus neutralisation assay (PVNA) based kit suitable for affordable global distribution. Infectivity of each PV was calculated after lyophilisation and immediate reconstitution, as well as subsequent to incubation of freeze-dried pellets at varying temperatures, humidities and timepoints. Integrity of glycoprotein structure following treatment was also assessed by employing lyophilised PVs in downstream PVNAs. In the presence of 0.5M sucrose-PBS cryoprotectant, each freeze-dried pseudotype was stably stored for 4 weeks at up to 37°C and could be neutralised to the same potency as unlyophilised PVs when employed in PVNAs. These results confirm the viability of a freeze-dried PVNA-based kit, which could significantly facilitate low-cost serology for a wide portfolio of emerging infectious viruses.
