Results are presented on the effects of interferon on the intracellular stages of T. cruzi in tissue culture "Vero" cells. Interferon was obtained by infecting monolayers of human amniotic cells with inactivated Newcastle disease virus. Interferon has not affected the cell infection by T. cruzi culture infective stages and neither has it prevented the transformation of amastigote into trypomastigote stages.

The megaesophagus and megacolon endemic in South America are related , to Chagas' disease. These mega conditions are found in patients with chronic Chagas's infection, when the parasite is not demonstrable in the lesions. These are characterized by depopulation of parasympathetic ganglion cells, dilation and hypertrophy of the viscera. In the experiments described here we deminstrate a selective affinity and adherence of Trypanosoma cruzi-immune lymphocytes to myenteric, parasympathetic ganglion cells, leading to neuronolysis. None of these features are observed when non-immune lymphocytes from control rabbits are used, or when the immune lymphocytes are allowed to react with CNS neurons. This demonstration is an indication of the high degree of specificity of the destruction of parasympathetic neurons in Chagas' disease. We postulate that the T. cruzi-immune lymphocyte rejection of parasympathetic neurons, but not of CNS neurons, might be related to recognition of a cross-reacting antigenic determinant secreted only by the target neurons. In favor of this interpretation is the observation of lymphocytic infiltrates and parasympathetic ganglion cell destruction in chronic Chagas' infection in the absence of encephalitis.

Mice inoculated with trypanosoma cruzi display an intense thrombocytopenia which is more severe in animals infected with the Y than CL strain. In animals inoculated with a T. cruzi strain which induces chronic infection (Colombiana), the number of platelets decreases as parasitemia ascends, and then reverts to normal values as soon as the acute infection merges into the chronic phase. The mechanisms involved in the thrombocytopenia are still obscure and are likely to be related to more general phenomena affecting the host rather than to direct damage of platelets or precursor cells by parasitism. Anemia and leukopenia are also present in T. cruzi infected mice.

Uma amostra de T. cruzi foi isolada pela primeira vez de um exemplar do Triatoma costalimai, capturado no município de Mambai, Goiás. Estudos experimentais sobre infectividade e virulência foram conduzidos em triatomíneos, Calomys callosus (Rodentia) e camundongos albinos. Cultivo "In Vitro" da amostra isolada foi obtido com sucesso utilizando-se o meio LIT. As mensurações realizadas em tripomastigotas sanguícolas deram os seguintes resultados (mcg): comprimento total - 16,4 (± 1,1); flagelo livre - 4,9 (± 1,1); largura - 2,8 (± 0,6); distância NA - 4,8 (± 0,6); distância NP - 6,0 (± 0,5) e Indice nuclear 1,3.

Circulating antigens were detected in sera of mice experimentally infected with a high close of Trypanosoma cruzi by reaction with sera from chronically infected mice. The immunodiffusion reaction between homologous acute and chronic sera produced four precipitation lines. By reaction with chronic mouse serum, circulating antingens were detected in sera from heavily infected hamsters, dogs, rabbits and in sera from chagasic patients. A reaction was also found in urine from acutely infected mice and dogs. Trypanosoma cruzi exoantigen was detected in trypanosome culture medium and in the supernatant of infected cell cultures. Attempts to isolate the antigens are described.

Callithrix jacchus marmosets, vaccinated more than once with high doses of the PF strain of Trypanosoma cruzi, showed a certain degree of parasitemia related to the number of parasites injected. Thirty days after vaccination, all animals were alive and showed no apparent morbid symptoms. The relationship between the dose of injected trypanosomes and the observed parasitemias is discussed and analysed as well as the immunologic incompetence of the experimental animals used.

Trypanomastigote forms of Trypanosoma cruzi were derived from tissue culture and incubated with immune and non-immune human sera. All immune sera showed high titers of specific humoral antibodies of the IgM or the IgG type. Agglutination and swelling of parasites were observed after incubation at 37ºC, but many trypomastigotes remained free-swimming in the sera for two to three days. The quantitiy of immune serum capable of lysing a maximum of 10 x 10 [raised to the power of 6] sensitized red cells was not capable of lysing 4 x 10 [raised to the power of 3] tripomastigotes. Typically, the parasites underwent cyclical changes with the formation of clumps of amastigotes and the appearance of epimastigote forms. Multiplication of the parasites was observed in immune sera. Further, the infectivity of the parasites to susceptible mice was not lost. All sera used produced similar general effects on the growth of the parasite. The antibody bound to T. cruzi appeard to enter cells by antigen-receptor mediated endocytosis. The ferritin-conjugated antibody was internalized and delivered to phagolysosomes where they might be completely degraded to amino-acids. This seemed to be a coupled process by which the immunoglobulin is first bound to specific parasite surface receptor and then rapidly endocytosed by the cell.

Neste trabalho estão apresentados resultados de estudos sobre roedores, marsupiais e triatomíneos do norte do municipio de Formosa,Estado de Goiás, e sua importância no ciclo silvestre do T.cruzi. A região estudada esta localizada do ponto de vista geográfico, na "Provincia do Cerrado". Foram coletados 963 roedores, 11 marsupiais e 766 triatomíneos silvestres. O índice de infecção pelo T. cruzi entre os roedores foi de 0,1% e entre os marsupiais 36,3%, enquanto todos os triatomíneos estavam negativos. Face aos aspectos ecológicos estudados, discute-se o papel desempenhado por roedores e marsupiais na manutenção e circulação do T. cruzi em ambiente silvestre. Alguns aspectos epidemiológicos no ambiente doméstico foram também abordados.

Formas de cultura de diferentes cepas do T.cruzi foram submetidas a vários processos de criopreservação. As percentagens de recuperação, avaliadas pela motilidade dos parasitas, foram consideradas como adequadas com algumas das técnicas empregadas, variando entre 60 a 80%. A estabilidade das características biológicas do material criopreservado foi investigada através do estudo das curvas de crescimento e diferenciação em meio acelular, infectividade para celulas de cultura de tecido ("Vero"), diferenciação intracelular em cultura de tecido assim como infectividade e curso da infecção em animais de laboratório. De um modo geral essas características nao foram significativamente alteradas no material congelado e estocado por diferentes períodos de tempo.

O aquecimento de soros chagásicos a 56 grausC por 30 minutos, não só inativa o complemento, como altera a mobilidade iônica das proteínas séricas. Com a inativação, os títulos dos soros por fixação de complemento decrescem sendo a queda do poder fixador relacionada com a diminuição da avidez do complexo-imune, formado com o antígeno de Trypanosoma cruzi para o complemento. Não se observaram diferenças na reatividade específica dos soros chagásicos, antes e depois de inativados, quando os títulos foram determinados por técnicas que não envolvem o complemento, tais como a imunofluorescência e a hemaglutinação.

Modulation by BCG and/or cyclophosphamide of sensitization of mice with flagellar fraction (a tubulin-enriched fraction) prevented death of mice challenged with T. cruzi CL strain trypomastigotes recovered from Vero cells. A methodology was ceveloped to assay specific antigens and to determine optimal doses for sensitization and elicitation of DTH in mice. CL strain is predominantly myotropic strain which does not produce important parasitism of mononuclear phagocyte cells; these cells appear to control infection when activated in vivo. Maximum protection was seen in this study when BCG and cyclophosphamide were associated, but protection was observed also when cyclophosphamide, that prevents supressor T cells, was applied 2 days before flagellar fraction sensitization in normal mice. These experiments suggested that the macrophage may have an important role in the early phases of infection particularly when nonspecific stimulation is associated with specific sensitization. A correlation betwen delayed hypersensitivity to parasite antigens and protection was observed.

In search of a suitable vector species for xenodiagnosis of humans and animals with chronic Chagas' disease we first investigated the reactions of different vector species to acute infection with Trypanosoma cruzi. Vector species utilized in this study were: Triatoma infestans, Rhodnius prolixus and Triatoma dimidiata, all well adapted to human habitats; Triatoma rubrovaria and Rhodnius neglectus both considered totally wild species; Panstrongylus megistus, Triatoma sordida, Triatoma pseudomaculata and Triatoma brasiliensis, all essentially sylvatic but some with domiciliary tendencies and others restricted to peridomestic biotopes with incipient colonization of human houses after successful eradication of T. infestans. Results summarized in Table IV suggest the following order of infectivity among the 9 studied vector species: P. megistus with 97.8% of infected bugs, T. rubrovaria with 95% of positive bugs a close second followed by T. Pseudomaculata with 94.3% and R. neglectus with 93.8% of infected bugs, almost identical thirds. R. prolixus, T. infestans and T. dimidiata exhibited low infection rates of 53.1%, 51.6% and 38.2% respectively, coupled with sharp decreases occuring with aging of infection (Fig. 1). The situation was intermediate in T. brasiliensis and T. sordida infection rates being 76.9% and 80% respectively. Results also point to the existence of a close correlation between prevalence and intensity of infection in that, species with high infection rates ranging from 93.8% to 97.8% exhibited relatively large proportions of insects (27.3% - 33.5%) harbouring very dense populations of T. cruzi. In species with low infection rates ranging from 38.2% to 53.1% the proportion of bugs demonstrating comparable parasite densities was at most 6%. Os extratos solúveis das cepas Y, São Felipe e Colombiana do Trypanosoma cruzi foram analisados contra seus antissoros homólogos e heterólogos por imunoeletroforese bidimensional e imunoeletroforese bidimensional com gel intermediário. Os resultados revelaram a existência de pelo menos 35 linhas de precipitação na cepa Y (32 de migração anódica e três de migração catódica), 24 linhas de precipitação anódica na cepa São Felipe e 22 na cepa Colombiana. Estas duas últimas cepas não apresentaram antígenos de migração catódica. Estes antígenos de migração catódica foram considerados "particulares" a cepa Y uma vez que quando testados contra antissoros heterólogos não observamos linhas de precipitação. Através do uso da imunoeletroforese bidimensional com gel intermediário foram evidenciados cinco antígenos particulares à cepa Y quando comparada à São Felipe e oito quando comparada à cepa Colombiana. A cepa São Felipe mostrou um único antígeno particular quando comparada à cepa Colombiana porém, não se evidenciou nenhum antígeno particular quando a cepa São Felipe foi analisada contra o antissoro da cepa Y. A cepa Colombiana não demonstrou nenhum antígeno particular nem quando comparada à cepa Y nem à São Felipe. Nossos resultados revelam que cepas pertencentes a diferentes tipos quanto ao comportamento morfobiológico e histopatológico também demonstram uma acentuada diferença quanto a seus componentes antigênicos.

The systematic positon of Trypanosoma rangeli is reconsidered and the creation of a new subgenus. Tejeraia, is proposed to remove this trypanosome from the subgenus Herptosoma of the section Stercoraria. The characteristics described for the proposed subgenus indicate that it must be located in the section Salivaria rather than in the Stercoraria. The evidence supporting this proposition is discussed in the text.

O exame da hemolinfa de triatomíneos (Triatoma infestans e Dipetalogastermaxima) infectados pelo Trypanosoma cruzi há 10, 15, 20 e 30 dias não revelaram a presença do flagelado. Material colhido e lavado da hemocele de D. maxima infectados, também foi negativo. A inoculação de formas sanguíneas do parasita e daquelas obtidas do conteúdo intestinal de triatomíneos na hemocele revelou que os parasitas não foram capazes de manter na hemolinfa uma infecção por mais de 40 dias e que não puderam penetrar no intestino dos triatomíneos. A inoculação de hemolinfa de insetos naturalmente infectados em camundongos recém-nascidos não induziu infecção.
