The nucleotide sequence of a 714 bp BamHI-EcoRI fragment of cucumber chloroplast DNA was determined. The fragment contained a gene for tRNA(Leu) together with its flanking regions. The trnL(CAA) gene sequence is about 99% in similarity to broad bean, cauliflower, maize, spinach and tobacco corresponding genes. The relative expression level of the gene was determined by Northern (tRNA) gel blot and Northern (total cellular RNA) slot-blot analyses using the trnL gene probe in 6-day old etiolated cucumber seedlings and the seedlings that had been kept in the dark (dark-grown), treated with benzyladenine (BA) and kept in the dark (BA-treated dark-grown), illuminated (light-grown), and treated with BA and illuminated (BA-treated light-grown), for additional 4, 8 or 12 hr. The trnL transcripts and tRNA(Leu) levels in BA-treated dark-grown seedlings were 5 and 3 times higher, respectively after 4 hr BA treatment, while in the BA treated light-grown seedlings the level of trnL transcripts was only 3 times higher and had no detectable effect on mature tRNA(Leu) when compared to the time-4 hr dark-grown seedlings. However, the level of mature tRNA(Leu) did not show marked changes in the light-grown seedlings, whereas the level of trnL transcripts increases 3 times after 8 hr illumination of dark-grown seedlings. These data indicate that both light and cytokinin can signal changes in plastid tRNA gene expression. The possible regulatory mechanisms for such changes are discussed.

This research investigates the impacts of agricultural market liberalization on food security in developing countries and it evaluates the supply perspective of food security. This research theme is applied on the agricultural sector in Kenya and in Zambia by studying the role policies played in the maize sub-sector. An evaluation of selected policies introduced at the beginning of the 1980s is made, as well as an assessment of whether those policies influenced maize output. A theoretical model of agricultural production is then formulated to reflect cereal production in a developing country setting. This study begins with a review of the general framework and the aims of the structural adjustment programs and proceeds to their application in the maize sector in Kenya and Zambia. A literature review of the supply and demand synthesis of food security is presented with examples from various developing countries. Contrary to previous studies on food security, this study assesses two countries with divergent economic orientations. Agricultural sector response to economic and institutional policies in different settings is also evaluated. Finally, a dynamic time series econometric model is applied to assess the effects of policy on maize output. The empirical findings suggest a weak policy influence on maize output, but the precipitation and acreage variables stand out as core determinants of maize output. The policy dimension of acreage and how markets influence it is not discussed at length in this study. Due to weak land rights and tenure structures in these countries, the direct impact of policy change on land markets cannot be precisely measured. Recurring government intervention during the structural policy implementation period impeded efficient functioning of input and output markets, particularly in Zambia. Input and output prices of maize and fertilizer responded more strongly in Kenya than in Zambia, where the state often ceded to public pressure by revoking pertinent policy measures. These policy interpretations are based on the response of policy variables which are more responsive in Kenya than in Zambia. According the obtained regression results, agricultural markets in general, and the maize sub-sector in particular, responded more positively to implemented policies in Kenya, than in Zambia, which supported a more socialist economic system. It is observed in these results that in order for policies to be effective, sector and regional dimensions need to be considered. The regional and sector dimensions were not taken into account in the formulation and implementation of structural adjustment policies in the 1980s. It can be noted that countries with vibrant economic structures and institutions fared better than those which had a firm, socially founded system.

Kasvit ovat kautta aikojen levinneet uusille elinpaikoille. Kasvin asettumista uuteen paikkaan voidaan tarkastella prosessina, jossa on erilaisia vaiheita ja eri vaiheissa eri tekijät ovat tärkeitä. Ilmasto ja erityisesti lämpötila vaikuttavat kasvien levinneisyyteen ja leviämiseen uusille paikoille ja siksi ilmaston lämpenemisen ennustetaan siirtävän kasvien levinneisyysalueita kohti pohjoista. On mahdollista, että Suomeenkin leviäisi etelämmästä muiden kasvien ohella haitallisia rikkakasvilajeja, kuten esimerkiksi viherrevonhäntä (Amaranthus retroflexus L.) ja kananhirssi (Echinochloa crus-galli L. Beauv.). Tämän tutkimuksen tarkoituksena oli selvittää, selviävätkö viherrevonhäntä ja kananhirssi Suomessa pelto-olosuhteissa ja pohtia niiden vakiintumisen ja leviämisen mahdollisuuksia. Toinen tarkoitus oli selvittää ilmaston lämpenemisen vaikutusta näiden rikkakasvien kasvuun. Tutkimus suoritettiin kenttä- sekä kasvihuonekokeena. Viherrevonhäntä kasvoi pellolla hyvin huolimatta myöhäisestä itämisestä, mutta kananhirssi iti ja kasvoi pellolla huonosti. Kasvihuoneessa molemmat kasvoivat hyvin. Kilpailu vähensi viherrevonhännän ja kananhirssin vegetatiivista kasvua vain kasvihuoneessa, mutta siementuottoon kilpailu vaikutti sekä kasvihuoneessa että pellolla. Kasvihuoneessa korkeampi lämpötila ei vaikuttanut viherrevonhännän tai kananhirssin vegetatiiviseen kasvuun, mutta viherrevonhännän siementuotto parani lämpimämmässä. Lämpötilalla ei ollut vaikutusta kananhirssin siementuotantoon. Tutkimuksen tuloksista voidaan päätellä, että viherrevonhäntä voi hyvinkin kasvaa Suomessa jo nykyisissä lämpötiloissa, mutta siementuotto ei välttämättä olisi varmaa. Viherrevonhäntä voisi näin ollen hyötyä tulevaisuuden pidemmästä kasvukaudesta. Kananhirssin osalta tulokset olivat ristiriitaiset ja ilmaston lämpenemisen vaikutuksia kananhirssiin on tämän tutkimuksen perusteella hankala arvioida.

After ensilation, the toxic Compositae weed Parthenium hysterophorus was devoid of the toxic principle parthenin. Laboratory-scale ensilation indicated that no parthenin was detectable after 5 wk of anaerobic fermentation. For animal feeding studies, silage was made on a large scale from Parthenium mixed with maize or from Parthenium alone. Crossbred bull and buffalo bull calves were fed diets containing the silages, or control diet without silage, for 12 wk. The animals consumed both silages with relish, and body weight gains of silage-fed calves did not differ from those of the controls. The digestibilities of dry matter, fibre and nitrogen-free extract were greater with the control diet, but the biological value of proteins tended to be greater with the silage-containing diets. Haematological studies indicated no significant differences between experimental and control groups in selected parameters, except for a reduction in blood urea nitrogen in the animals fed silage. The possible causes for these biochemical alterations are discussed. Since the nutritive value of Parthenium silage compares favourably with the standard diet, and Parthenium seeds collected from the silage did not germinate, we suggest that ensilation can be used as an additional method in the containment and eradication of these plants, which grow wild in India.

A complete cDNA encoding a novel hybrid Pro-rich protein (HyPRP) was identified by differentially screening 3x10(4) recombinant plaques of a Cuscuta reflexa cytokinin-induced haustorial cDNA library constructed in lambda gt10. The nucleotide (nt) sequence consists of: (i) a 424-bp 5'-non coding region having five start codons (ATGs) and three upstream open reading frames (uORFs); (ii) an ORF of 987 bp with coding potential for a 329-amino-acid (aa) protein of M(r), 35203 with a hydrophobic N-terminal region including a stretch of nine consecutive Phe followed by a Pro-rich sequence and a Cys-rich hydrophobic C terminus; and (iii) a 178-bp 3'-UTR (untranslated region). Comparison of the predicted aa sequence with the NBRF and SWISSPROT databases and with a recent report of an embryo-specific protein of maize [Jose-Estanyol et al., Plant Cell 4 (1992) 413-423] showed it to be similar to the class of HyPRPs encoded by genes preferentially expressed in young tomato fruits, maize embryos and in vitro-cultured carrot embryos. Northern analysis revealed an approx. 1.8-kb mRNA of this gene expressed in the subapical region of the C. reflexa vine which exhibited maximum sensitivity to cytokinin in haustorial induction.

Estimation of soil parameters by inverse modeling using observations on either surface soil moisture or crop variables has been successfully attempted in many studies, but difficulties to estimate root zone properties arise when heterogeneous layered soils are considered. The objective of this study was to explore the potential of combining observations on surface soil moisture and crop variables - leaf area index (LAI) and above-ground biomass for estimating soil parameters (water holding capacity and soil depth) in a two-layered soil system using inversion of the crop model STICS. This was performed using GLUE method on a synthetic data set on varying soil types and on a data set from a field experiment carried out in two maize plots in South India. The main results were (i) combination of surface soil moisture and above-ground biomass provided consistently good estimates with small uncertainity of soil properties for the two soil layers, for a wide range of soil paramater values, both in the synthetic and the field experiment, (ii) above-ground biomass was found to give relatively better estimates and lower uncertainty than LAI when combined with surface soil moisture, especially for estimation of soil depth, (iii) surface soil moisture data, either alone or combined with crop variables, provided a very good estimate of the water holding capacity of the upper soil layer with very small uncertainty whereas using the surface soil moisture alone gave very poor estimates of the soil properties of the deeper layer, and (iv) using crop variables alone (else above-ground biomass or LAI) provided reasonable estimates of the deeper layer properties depending on the soil type but provided poor estimates of the first layer properties. The robustness of combining observations of the surface soil moisture and the above-ground biomass for estimating two layer soil properties, which was demonstrated using both synthetic and field experiments in this study, needs now to be tested for a broader range of climatic conditions and crop types, to assess its potential for spatial applications. Use of fuel other than woody generally has been limited to rice husk and other residues are rarely tried as a fuel in a gasification system. With the availability of woody biomass in most countries like India, alternates fuels are being explored for sustainable supply of fuel. Use of agro residues has been explored after briquetting. There are few feedstock's like coconut fronts, maize cobs, etc, that might require lesser preprocessing steps compared to briquetting. The paper presents a detailed investigation into using coconut fronds as a fuel in an open top down draft gasification system. The fuel has ash content of 7% and was dried to moisture levels of 12 %. The average bulk density was found to be 230 kg/m3 with a fuel size particle of an average size 40 mm as compared to 350 kg/m3 for a standard wood pieces. A typical dry coconut fronds weighs about 2.5kgs and on an average 6 m long and 90 % of the frond is the petiole which is generally used as a fuel. The focus was also to compare the overall process with respect to operating with a typical woody biomass like subabul whose ash content is 1 %. The open top gasification system consists of a reactor, cooling and cleaning system along with water treatment. The performance parameters studied were the gas composition, tar and particulates in the clean gas, water quality and reactor pressure drop apart from other standard data collection of fuel flow rate, etc. The average gas composition was found to be CO 15 1.0 % H-2 16 +/- 1% CH4 0.5 +/- 0.1 % CO2 12.0 +/- 1.0 % and rest N2 compared to CO 19 +/- 1.0 % H-2 17 +/- 1.0 %, CH4 1 +/- 0.2 %, CO2 12 +/- 1.0 % and rest N2. The tar and particulate content in the clean gas has been found to be about 10 and 12 mg/m3 in both cases. The presence of high ash content material increased the pressure drop with coconut frond compared to woody biomass.

In Nigeria, the culture of fish is gaining importance, but local fish farmers face a set back because of the stoppage on importation of fish feed. Locally available raw materials such as yam, plantain, banana, cowpeas, macuna, maize, cassava, millet, sorghum, groundnut, sunnhemp seed and brewery wastes are considered as potential materials for fish feed. These have been examined on their minimum protein contributions since this is the most expensive part of the fish feed. Alternative sources to animal proteins are also examined. Plant protein from groundnut, melon, mucuna and others compare favourably with bloodmeal mixture and thus can be used to replace the more expensive animal proteins. Pellet feed can be produced on a small scale or commercial basis from the locally available raw materials and the fish farmer is advised to seek assistance from qualified fisheries personnel

This study, though, has as its core objective cost reduction in aquaculture nutrition was equally designed to investigate the value of the peels of cassava (Manihot utillisima) as energy source in the diet of Oreochromis niloticus fry. Three levels of cassava peels diet and a control (100% yellow maize in the carbohydrate mixture) was prepared and tested on O. niloticus fry for ten (10) weeks. The fry with mean weight of 0.32g were grouped fifteen (15) in each of the glass aquaria measuring 60x30x30cm with a maximum capacity of 52 litres of water. The fry were fed twice daily at 10% biomass. Weekly, the fry were weighed to determine the weight increment or otherwise and the quality of feed adjusted accordingly. Water quality parameters like temperature, pH and dissolved oxygen (D.0) were monitored and found to be at desirable level. DT 3 (97 % cassava peels and 3% yellow maize) in the carbohydrate mixture gave the best growth performance. The fry fed, this diet gained mean weight of 1.18g for the period of the experiment. However, the poorest performance in terms of growth was from fry fed the control diet (100%yellow maize in the carbohydrate mixture) fry fed this diet gained mean weight of 0.80 for the duration of the experiment. Analysis of the various growth indices like SGR, PER, FCR and NPU shows that DT3 was the overall best diet with an SGR value of2.40 and FCR of 43.83. However, DT 1 (70% cassava peels and 30% yellow maize) gave the poorest SGR of 1.61 and FCR of 67.58. The difference in weight gain among the fry fed the three levels of cassava peels diet and the control was not statically significant (P>0.05)

The study was designed to investigate the value of the peels of yam (Dioscorea rotundata) as energy source in the diet of Oreochromis niloticus fry and to investigate the level of inclusion of this peels that will give optimum growth performance. Four diets, three levels of yam peels and a control, was prepared and tested on O. niloticus fry (mean weight of 0.27g) for ten weeks. Fifteen (15) O. niloticus fry were grouped in each of the glass aquaria, measuring 60x30x3Ocm and with a maximum capacity of 52 liters of water. The fry were fed twice daily at 10% biomass. The fry were weighed weekly to determine weight increment or otherwise and the quality of feed was adjusted accordingly. DTl (70% yam peels and 30% yellow maize) in the carbohydrate mixture gave the best performance. The fry fed this diet, gained a mean weight of 1.20g for the period of the experiment. The poorest performance in terms of growth was from fry fed the control diet (100% yellow maize in the carbohydrate mixture). Fry fed this diet gained mean weight of 0.80g for the duration of the experiment. Analysis of the various growth indices like SGR, PER, FCR and NPU shows that DTl was the overall best diet with an SGR value of I. 92 and FCR of 54.10. The difference in weight gain by fry fed the three levels of yam peels diet and the control diet (100% yellow maize) was not statistically significant (P>0.05)

Este trabalho pretende contribuir para o entendimento sobre a produtividade na agricultura brasileira, bem como propõe uma reflexão sobre o debate da produtividade total dos fatores. Para isso, buscou-se, antes de tudo, debater as medidas de produtividade, sobretudo as medidas de produtividade total dos fatores e, em seguida, apresentar estimativas da produtividade total na agricultura no Brasil e no mundo. Propõem-se também uma estimativa preliminar dessa metodologia para o cálculo da produtividade total dos fatores na cultura do milho através do índice de Tornqvist. O objetivo é, então, revisar a literatura teórica e empírica sobre a produtividade total dos fatores na agricultura e calcular as produtividades parciais e totais na cultura do milho para os estados brasileiros. Apesar da maior parte dos resultados encontrados no cálculo da produtividade total para a cultura do milho no Brasil não terem sido consistentes, a conclusão geral do levantamento bibliográfico foi que tanto a agricultura brasileira quanto a agricultura mundial, independente da metodologia utilizada, apresentou uma tendência ao crescimento da produtividade total dos fatores.

Essa dissertação trata do medo na relação entre alunos e professores.O medo enquanto emoção humana determina alterações biofísicas que são responsáveis pela manutenção da vida. No entanto, em nível elevado, o medo pode se tornar paralisante ou causador de patologias psíquicas. Mobilizador ou paralisante, o medo pode ser causado por situações reais ou imaginárias. O mesmo medo pode ser causa de angústia para uns e não para outros. Existem aqueles que são só de uns e outros que são de todos. Ao longo da História o medo foi utilizado como forma de disciplinarização de indivíduos e até de populações inteiras. Também foi utilizado na Escola, como forma de controle e exercício do poder do adulto sobre a criança e o jovem. Punições, humilhações, palmatórias, varas, joelhos no milho foram alguns dos instrumentos utilizados por longo tempo na intimidação do aluno. O Século XXI, que já vem sendo conhecido como o Século do Medo, vem trazendo as mudanças nos comportamentos, o aumento da violência do mundo e as mudanças na escola. O professor outrora amedrontador, detentor do poder legitimado socialmente, vem se tornando um amedrontado tanto no exercício profissional quanto cidadão da grande cidade. Analisaremos nesse trabalho o quanto do professor amedrontador e do aluno amedrontado convivem hoje como o professor amedrontado e o aluno amedrontador

本文叙述了两个玉米基因型(小八趟×水白和白17)原生质体培养的植株再生;用基因型(小八趟×水白)为材料研究影响玉米原生质体培养的各种因素,并此基因型的原生质体经超低温保存后获得植株再生;以及用多种基因型玉米幼胚为材料诱导愈伤组织与植株再生。 影响玉米原生质体游离、分裂与植株再生的因素是多方面的。酶液组合0.2% Onozuka RS + 1% Hemicellulase + 0.1% Pectolyase,利用继代8-16天的愈伤组织,所获原生质体的数量与质量最佳。在原生质体植板率方面,结果表明:N6作为基本培养基是理想的;氮源中,NO3-具有明显的促进作用,而NH4+具有明显的抑制作用;有机氮源是不能缺少的,所使用的四种有机氮源中L-脯氨酸效果最明显。2,4-D浓度以1.0 mg/l最佳。原生质体培养后的渗透压浓度降低的时间以培养四星期后为宜。利用三步诱导,成功地获得胚胎发生的植株再生,并且还指出原生质体起始材料的保存年限大大影响原生质体所再生愈伤组织的分化。 采用上述筛选出的最佳游离、培养以及植株再生的方法,成功地培养了基因型(白17)的原生质体,并获得植株再生。原生质体再生细胞培养4-5天后开始一次分裂;培养15天后,植板率为3-4%。一个月后,原生质体所再生的肉眼可见的愈伤组织,分步转至分化培养基。最后,愈伤组织通过胚胎发生获得植株再生,频率约10%。 玉米原生质体,利用5%DMSO与0.55 M葡萄糖作为混合保护剂,经慢速(1 ℃/分钟)降至-40 ℃,停留二小时后直接投入液氮保存。保存3天后,原生质体在40 ℃的温水浴中快速化冻,成活率高达30-40%。成活原生质体培养后生长正常,植板率高达8-10%。培养5-6星期后,再生愈伤组织转至分化培养基;最后获得植株再生,频率为5-10%。 本文最后叙述了玉米七种基因型的幼胚诱导获得愈伤组织,再生植株频率可达70-80%。 上述各方面的研究结果,对玉米的遗传操作和细胞抗寒性研究、生理代谢的研究等都是十分有价值的。
