Molecular biology techniques are of help in genetic improvement since they permit the identification, mapping and analysis of polymorphisms of genes encoding proteins that act on metabolic pathways involved in economically interesting traits. The somatotrophic axis, which essentially consists of growth hormone releasing hormone (GHRH), growth hormone (GH), insulin-like growth factors I and II (IGF-I and IGF-II), and their associated binding proteins and receptors (GHRHR, GHR, IGF-IR and IGF-IIR), plays a key role in the metabolism and physiology of mammalian growth. The objectives of the present study were to estimate the allele and genotype frequencies of the IGF-I/SnaBI, IGF-IR/TaqI and GHRH/HaeIII gene polymorphisms in different genetic groups of beef cattle and to determine associations between these polymorphisms and growth and carcass traits. For this purpose, genotyping was performed on 79 Nellore animals, 30 Canchim (5/8 Charolais+3/8 Zebu) animals and 275 crossbred cattle originating from the crosses of Simmental (n=30) and Angus (n=245) sires with Nellore females. In the association studies, traits of interest were analyzed using the GLM procedure of SAS and least square means of the genotypes were compared by the Tukey test. Associations of IGF-I/SnaBI genotypes with body weight and subcutaneous backfat were significant (p < 0.05), and nearly significant for longissimus dorsi area (p=0.06), with the 1313 genotype being favorable compared to the AB genotype. No significant associations were observed between this polymorphism and weight gain or carcass yield (P > 0.05). The contribution of growth and turnover to the muscle delta C-13 change process was investigated using mathematical models which associate delta C-13 change to time of intake of a new diet or increase in body mass. Two groups of Nile tilapia (Oreochromis niloticus) were fed on diets based on C3 (sigma C-13 = - 25.64 +/- 0.06 parts per thousand) or C4 (delta C-13= -16.01 +/- 0.06 parts per thousand) photosynthetic cycle plants to standardize the muscle delta C-13. After establishing the carbon isotopic equilibrium, fish (mean mass 24.12 +/- 6.79 g) then received the other treatment diet until a new carbon isotopic equilibrium could be established, characterizing T1 (C3-C4) and T2 (C4-C3) treatments. No significant differences were observed in fish productive performance. Good fits were obtained for the models that associated the delta C-13 change to time, resulting in carbon half-life values of 23.33 days for T1 and 25.96 days for T2. Based on values found for the muscle delta C-13 change rate from growth (0.0263 day(-1) and 0.0254 day(-1)) and turnover (0.0034 day(-1) and 0.0013 day(-1)), our results indicate that most of the delta C-13 change could be attributed to growth. The application of model that associated the delta C-13 change to body mass increase seems to produce results with no apparent biological explanation. The delta C-13 change rate could directly reflect the daily ration and growth rate, and consequently the isotopic change rates of carbon and other tissue elements can be properly used to assess different factors that may interfere in nutrient utilization and growth. Seasonal changes in vegetative growth, leaf gas exchanges, carbon isotope discrimination (Delta) and carbohydrate status were monitored in de-fruited coffee trees (Coffea arabica L.) grown in the field, from October 1998 through September 1999, in Vicosa (20degrees45'S, 42degrees15'W, 650 m a.s.l.), southeastern Brazil. of the total growth over the 12-month study period, 78% occurred in the warm, rainy season (October-March), and 22% during the cool, dry season (April-September). Throughout the active growth period, the rate of net carbon assimilation (A) averaged 8.6 mumol m(-2) s(-1), against 3.4 mumol m(-2) s(-1) during the period of reduced growth. In the active period, growth, unlike A or Delta, was strongly negatively correlated with air temperature. In contrast, growth and A were both correlated positively, and Delta correlated negatively, with air temperature during the reduced growth period. However, the depressions of A and growth might have simply run in parallel, without any causal relationship. Changes in A appeared to be largely due to stomatal limitations in the active growing season, with non-stomatal ones prevailing in the slow growth period. Foliar carbohydrates seemed not to have contributed appreciably to changes in growth rates and photosynthesis. (C) 2004 Elsevier B.V. All rights reserved.

Coffee (Coffea arabica L.) plants were grown in small (3-L), medium (10-L) and large (24-L) pots for 115 or 165 d after transplanting (DAT), which allowed different degrees of root restriction. Effects of altered source : sink ratio were evaluated in order to explore possible stomatal and non-stomatal mechanisms of photosynthetic down-regulation. Increasing root restriction brought about large and general reductions in plant growth associated with a rising root : shoot ratio. Treatments did not affect leaf water potential or leaf nutrient status, with the exception of N content, which dropped significantly with increasing root restriction even though an adequate N supply was available. Photosynthesis was severely reduced when plants were grown in small pots; this was largely associated with non-stomatal factors, such as decreased Rubisco activity. At 165DAT contents of hexose, sucrose, and amino acids decreased in plants grown in smaller pots, while those of starch and hexose-P increased in plants grown in smaller pots. Photosynthetic rates were negatively correlated with the ratio of hexose to free amino acids, but not with hexose content. Activities of acid invertase, sucrose synthase, sucrose-P synthase, fructose-1,6- bisphosphatase, ADP-glucose pyrophosphorylase, starch phosphorylase, glyceraldehyde-3-P dehydrogenase, PPi : fructose-6-P 1-phosphotransferase and NADP : glyceraldehyde-3-P dehydrogenase all decreased with severe root restriction. Glycerate-3-P : Pi and glucose-6-P : fructose-6-P ratios decreased accordingly. Photosynthetic down-regulation was unlikely to have been associated directly with an end-product limitation, but rather with decreases in Rubisco. Such a down-regulation was largely a result of N deficiency caused by growing coffee plants in small pots.

Extracellular matrix remodeling occurs during ovarian follicular development, mediated by plasminogen activators (PAs) and PA inhibitors including protease nexin-1 (PN-1). In the present study we measured expression/activity of the PA system in bovine follicles at different stages of development by timed collection of ovaries during the first follicular wave and during the periovulatory period, and in follicles collected from an abattoir. The abundance of mRNA encoding PN-1, tissue-type PA (tPA), urokinase (uPA) and PA inhibitor-1 (PAI-1) were initially upregulated by human chorionic gonadotropin (hCG) in bovine preovulatory follicular wall homogenates. PN-1, PAI-1 and tPA mRNA expression then decreased near the expected time of ovulation, whereas uPA mRNA levels remained high. PN-1 concentration in follicular fluid (FF) decreased and reached the lowest level at the time of ovulation, whereas plasmin activity in FF increased significantly after hCG. Follicles collected from the abattoir were classified as non-atretic, early-atretic or atretic based on FF estradiol and progesterone content: PN-1 protein levels in FF were significantly higher in non-atretic than in atretic follicles, and plasmin activity was correspondingly higher in the atretic follicles. No changes in PN-1 levels in FF were observed during the growth of pre-deviation follicles early in a follicular wave. These results indicate that PN-1 may be involved in the process of atresia in non-ovulatory dominant follicles and the prevention of precocious proteolysis in periovulatory follicles.

O objetivo do presente estudo foi determinar o crescimento relativo e a maturidade sexual morfológica de Menippe nodifrons. As coletas foram realizadas na Praia Grande e Tenório, Ubatuba, São Paulo. Os caranguejos foram separados quanto ao sexo e mensurados na região da largura (LC) e comprimento da carapaça, comprimento e altura dos própodos quelares direito e esquerdo, largura do abdome (LA) nas fêmeas e comprimento do gonopódio (CG) nos machos. Obteve-se 399 indivíduos, sendo 195 machos e 204 fêmeas. Os machos atingiram a maturidade sexual com 29,7mm LC e as fêmeas com 31,6mm LC. Para as fêmeas a melhor relação que indicou a muda da puberdade foi LA vs. LC, sendo que o crescimento foi alométrico negativo na fase jovem e alométrico positivo após a muda da puberdade. Para os machos foi CG vs. LC evidenciando crescimento alométrico positivo na fase jovem e isométrico na fase adulta.

The relative growth of the fiddler crab Uca cumulanta was studied, primarily to determine the size at the onset of sexual maturity for a mangrove population in the estuary of the Patitiba River, Paraty, Rio de Janeiro state, Brazil. The evaluation of the morphological sexual maturity of U. cumulanta was performed using the allometric technique. The relationships that most precisely indicated the size at onset of sexual maturity were carapace length (CL) vs propodus length for males and CL vs abdomen width for females. Males and females are mature at 5.25 and 4.75 mm CL, respectively. The remarkable ontogenetic changes observed in the allometric growth of the male major cheliped and the female abdomen, indicate that growth of these structures is closely connected to the timing of sexual maturity. The relative size at onset maturity obtained for this species was 0.68 and this index was compared to that seen in other species in the genus.

The influence of a population of the understorey woody bamboo Merostachys riedeliana and different flooding regimes on tree community dynamics in a section of tropical semideciduous forest in South-Eastern Brazil was examined. A forest section with an area of 1.6 ha composed of 71 adjacent plots was located on a slope ending at the river margin. The section was divided into five topographical sectors according to the mean duration of river floods. In 1991 and 1998 all trees with a diameter at the base of the trunk greater than or equal to 5 cm were measured, identified and tagged, and all live bamboo culms were counted. Annualised estimates of the rates of tree mortality and recruitment, gain and loss of tree basal area, and change in bamboo density were calculated for each of the 71 plots and five topographical sectors as well as for diameter classes and tree species. To segregate patterns arising from spatially autocorrelated events, geostatistical analyses were used prior to statistical comparisons and correlations. In general, mortality rates were not compensated by recruitment rates but there was a net increase in basal area in all sectors, suggesting that the tree community as a whole was in a building phase. Tree community dynamics of the point bar forest (Depression and Levee sectors) differed from that of the upland forest (Ridgetop, Middle Slope and Lower Slope sectors) in the extremely high rates of gain in basal area. The predominant and specialised species, Inga vera and Salix humboldtiana, are probably favoured by relaxed competition in an environment stressed by long-lasting floods. In the upland forest, mortality rates were highest at the Middle Slope, particularly for smaller trees, while recruitment rates were lowest. As bamboo clumps were concentrated in this sector, the locally higher instability in the tree community probably resulted from the direct interference of bamboos. The density of bamboo culms in the upland forest was negatively correlated with the rates of tree recruitment and gain in basal area, and positively correlated with tree mortality rates. Bamboos therefore seemed to restrict the recruitment, growth and survival of trees.

A toxicidade do alumínio (Al) é um dos fatores mais limitantes para a produtividade. Esta pesquisa foi realizada para avaliar a influência do Al, em solução nutritiva, na altura de plantas, no peso da matéria seca e nas alterações morfoanatômicas de raízes e folhas de milho (Zea mays L.). O experimento foi conduzido em casa de vegetação com tratamentos constituídos de cinco doses de Al (0; 25; 75; 150; e 300 µmol L-1) e seis repetições. As soluções foram constantemente aeradas e o pH foi ajustado a 4,3, inicialmente. A matéria seca da parte aérea e das raízes e a altura das plantas diminuíram significativamente com o aumento da concentração de Al. As raízes de plantas de milho cultivadas em soluções com Al tiveram seu crescimento inibido e apresentaram menos raízes laterais e desenvolvimento do sistema radicular inferior, em comparação com as das plantas-controle. As folhas das plantas crescidas em soluções que continham 75 e 300 µmol L-1 de Al não apresentaram muita diferença anatômica em relação às das plantas-controle. A bainha da folha das plantas exposta ao Al apresentou epiderme uniestratificada revestida por uma fina camada de cutícula e as células da epiderme e do córtex foram as que menos se desenvolveram. No feixe vascular, o metaxilema e protoxilema não tinham paredes secundárias, e o diâmetro de ambos foi muito menor quando comparado com os das plantas-controle.
