[ES] La acuicultura como medio de producción de alimento para consumo humano ha experimentado una gran expasión a lo largo de los últimos cincuenta años, incrementando su producción a razón de un 9-10% anual durante los últimos 10 años, muy por encima del 4-5% del resto de los animales de granja. Entre otros factores, la mejora tecnológica de los piensos y adecuación de los mismos a las necesidades nutricionales de los animales han sido determinantes para el desarrollo del sector. Se mostrará la evolución histórica de los piensos acuícolas, estado actual de los mismos y proyecciones futuras

[ES] Durante el desarrollo de este documento, se hace un recorrido por las competencias profesionales, competencias clave; precursoras de las que conocemos hoy día como básicas y, las competencias que estructuran y articulan los factores que conforman la empleabilidad, presentes en los mejores profesionales, independientemente del nivel de cualificación o puesto que ocupen. La educación basada en competencias, que ha comenzado su recorrido hace, relativamente, poco tiempo en nuestro sistema educativo, ha supuesto un cambio total en los paradigmas del sistema, dando un enfoque mucho más profundo al proceso de enseñanza-aprendizaje y marcando el camino a seguir por la comunidad educativa para satisfacer los fines de la Educación. En este viaje de ida y vuelta por las competencias, se analizan algunas de las más importantes para el mundo laboral que vinculo, bien mediante descriptores o como aspectos de aprendizaje, a las competencias básicas de la ESO y a las personales y sociales de los ciclos formativos. La información contenida en este artículo, así como los resultados cualitativos obtenidos del análisis llevado a cabo, aportan un nuevo punto de vista para trabajar las competencias en la educación secundaria, y en los ciclos formativos, especialmente en los últimos cursos de ambos. También se recogen las implicaciones que, a nivel curricular, derivan del nuevo enfoque, de tal manera que se amplía el ámbito de contextualización del entorno de aprendizaje, se establecen los principios metodológicos y estrategias didácticas para establecer dinámicas y actividades que potencien y desarrollen competencias tanto básicas como de empleabilidad en los alumnos. Así mismo, se esboza una guía que ayude a aquellos profesores que quieran dar un enfoque más práctico a las competencias básicas, bien para atender a la diversidad como para motivar a los alumnos que necesitan un enfoque más práctico de la asignatura. Por supuesto para la enseñanza de las formaciones profesionales ésta debería pautar la rutina básica de trabajo en las aulas y talleres. La creciente incertidumbre en el plano económico y, cada vez más, en el social y laboral, donde los escenarios dejan de ser estables y previsibles para dar paso a entornos sometidos a cambios rapidísimos y de gran calado, hace que necesitemos nuevas y más potentes capacidades para aprender y trabajar en el nuevo entorno en el que tendremos que educar a nuestros adolescentes. Hoy más que nunca debemos girar el poliedro de las competencias básicas para exponerlas, más abiertamente, al contexto del mundo laboral, estableciendo una vinculación directa entre éstas y las habilidades para la empleabilidad, que van cobrando importancia creciente, por encima, incluso, del nivel de cualificación técnico requerido para desempeñar una profesión.

[ES]La Directiva europea 2006/7/CE de gestión de la calidad de las aguas de baño define los criterios para clasificar las playas con arreglo a su Calidad, obtenida a partir del percentil 95 de las concentraciones de Escherichia coli y enterococos intestinales de una serie de muestras, tomadas en cada punto de baño siguiendo un calendario de control dentro de la temporada de baños, que en Canarias tiene un año natural de duración. Durante un largo tiempo (1996-2008) se tomaron muestras de cuatro importantes playas del sur de Gran Canaria cada quince días y se elaboró un informe anual, basado en los preceptos de la legislación vigente en esos años, Directiva 76/160/CEE. Al aplicar los criterios de calidad de la nueva Directiva observamos que las playas consideradas no habrían alcanzado la Calidad Excelente en todos los años de estudio. De hecho, las playas de Maspalomas y de El Inglés incumplirían la normativa la mitad de los años. Los resultados de los análisis bacteriológicos mostraron un comportamiento que se repetía a lo largo de los años. Durante los meses de primavera-verano, se obtenían bajas concentraciones de indicadores bacterianos de contaminación, mientras que en otoño-invierno, coincidiendo con las borrascas de Sur-Suroeste, se alcanzaban valores bacterianos de 102 y 103 /100 ml, suficientes para elevar el percentil 95 por encima de los límites de calidad. El principal factor de contaminación son las aguas de escorrentía que desembocan en las playas de baño mezcladas con las aguas residuales procedentes de la red sanitaria en la que se producen inundaciones y desbordamientos causados por el enorme caudal de pluviales. Se plantea así un problema de difícil solución.
Sviluppo di biosensori: modifiche di superfici elettrodiche e sistemi di immobilizzazione enzimatica

An amperometric glucose biosensor was developed using an anionic clay matrix (LDH) as enzyme support. The enzyme glucose oxidase (GOx) was immobilized on a layered double hydroxide Ni/Al-NO3 LDH during the electrosynthesis, which was followed by crosslinking with glutaraldehyde (GA) vapours or with GA and bovine serum albumin (GABSA) to avoid the enzyme release. The electrochemical reaction was carried out potentiostatically, at -0.9V vs. SCE, using a rotating disc Pt electrode to assure homogeneity of the electrodeposition suspension, containing GOx, Ni(NO3)2 and Al(NO3)3 in 0.3 M KNO3. The mechanism responsible of the LDH electrodeposition involves the precipitation of the LDH due to the increase of pH at the surface of the electrode, following the cathodic reduction of nitrates. The Pt surface modified with the Ni/Al-NO3 LDH shows a much reduced noise, giving rise to a better signal to noise ratio for the currents relative to H2O2 oxidation, and a linear range for H2O2 determination wider than the one observed for bare Pt electrodes. We pointed out the performances of the biosensor in terms of sensitivity to glucose, calculated from the slope of the linear part of the calibration curve for enzimatically produced H2O2; the sensitivity was dependent on parameters related to the electrodeposition in addition to working conditions. In order to optimise the glucose biosensor performances, with a reduced number of experimental runs, we applied an experimental design. A first screening was performed considering the following variables: deposition time (30 - 120 s), enzyme concentration (0.5 - 3.0 mg/mL), Ni/Al molar ratio (3:1 or 2:1) of the electrodeposition solution at a total metals concentration of 0.03 M and pH of the working buffer solution (5.5-7.0). On the basis of the results from this screening, a full factorial design was carried out, taking into account only enzyme concentration and Ni/Al molar ratio of the electrosynthesis solution. A full factorial design was performed to study linear interactions between factors and their quadratic effects and the optimal setup was evaluated by the isoresponse curves. The significant factors were: enzyme concentration (linear and quadratic terms) and the interaction between enzyme concentration and Ni/Al molar ratio. Since the major obstacle for application of amperometric glucose biosensors is the interference signal resulting from other electro-oxidizable species present in the real matrices, such as ascorbate (AA), the use of different permselective membranes on Pt-LDHGOx modified electrode was discussed with the aim of improving biosensor selectivity and stability. Conventional membranes obtained using Nafion, glutaraldehyde (GA) vapours, GA-BSA were tested together with more innovative materials like palladium hexacyanoferrate (PdHCF) and titania hydrogels. Particular attention has been devoted to hydrogels, because they possess some attractive features, which are generally considered to favour biosensor materials biocompatibility and, consequently, the functional enzyme stability. The Pt-LDH-GOx-PdHCF hydrogel biosensor presented an anti-interferant ability so that to be applied for an accurate glucose analysis in blood. To further improve the biosensor selectivity, protective membranes containing horseradish peroxidase (HRP) were also investigated with the aim of oxidising the interferants before they reach the electrode surface. In such a case glucose determination was also accomplished in real matrices with high AA content. Furthermore, the application of a LDH containing nickel in the oxidised state was performed not only as a support for the enzyme, but also as anti-interferant sistem. The result is very promising and it could be the starting point for further applications in the field of amperometric biosensors; the study could be extended to other oxidase enzymes.

The present Thesis studies three alternative solvent groups as sustainable replacement of traditional organic solvents. Some aspects of fluorinated solvents, supercritical fluids and ionic liquids, have been analysed with a critical approach and their effective “greenness” has been evaluated from the points of view of the synthesis, the properties and the applications. In particular, the attention has been put on the environmental and human health issues, evaluating the eco-toxicity, the toxicity and the persistence, to underline that applicability and sustainability are subjects with equal importance. The “green” features of fluorous solvents and supercritical fluids are almost well-established; in particular supercritical carbon dioxide (scCO2) is probably the “greenest” solvent among the alternative solvent systems developed in the last years, enabling to combine numerous advantages both from the point of view of industrial/technological applications and eco-compatibility. In the Thesis the analysis of these two classes of alternative solvents has been mainly focused on their applicability, rather than the evaluation of their environmental impact. Specifically they have been evaluated as alternative media for non-aqueous biocatalysis. For this purpose, the hydrophobic ion pairing (HIP), which allows solubilising enzymes in apolar solvents by an ion pairing between the protein and a surfactant, has been investigated as effective enzymatic derivatisation technique to improve the catalytic activity under homogeneous conditions in non conventional media. The results showed that the complex enzyme-surfactant was much more active both in fluorous solvents and in supercritical carbon dioxide than the native form of the enzyme. Ionic liquids, especially imidazolium salts, have been proposed some years ago as “fully green” alternative solvents; however this epithet does not take into account several “brown” aspects such as their synthesis from petro-chemical starting materials, their considerable eco-toxicity, toxicity and resistance to biodegradation, and the difficulty of clearly outline applications in which ionic liquids are really more advantageous than traditional solvents. For all of these reasons in this Thesis a critical analysis of ionic liquids has been focused on three main topics: i) alternative synthesis by introducing structural moieties which could reduce the toxicity of the most known liquid salts, and by using starting materials from renewable resources; ii) on the evaluation of their environmental impact through eco-toxicological tests (Daphnia magna and Vibrio fischeri acute toxicity tests, and algal growth inhibition), toxicity tests (MTT test, AChE inhibition and LDH release tests) and fate and rate of aerobic biodegradation in soil and water; iii) and on the demonstration of their effectiveness as reaction media in organo-catalysis and as extractive solvents in the recovery of vegetable oil from terrestrial and aquatic biomass. The results about eco-toxicity tests with Daphnia magna, Vibrio fischeri and algae, and toxicity assay using cultured cell lines, clearly indicate that the difference in toxicity between alkyl and oxygenated cations relies in differences of polarity, according to the general trend of decreasing toxicity by decreasing the lipophilicity. Independently by the biological approach in fact, all the results are in agreement, showing a lower toxicity for compounds with oxygenated lateral chains than for those having purely alkyl lateral chains. These findings indicate that an appropriate choice of cation and anion structures is important not only to design the IL with improved and suitable chemico-physical properties but also to obtain safer and eco-friendly ILs. Moreover there is a clear indication that the composition of the abiotic environment has to be taken into account when the toxicity of ILs in various biological test systems is analysed, because, for example, the data reported in the Thesis indicate a significant influence of salinity variations on algal toxicity. Aerobic biodegradation of four imidazolium ionic liquids, two alkylated and two oxygenated, in soil was evaluated for the first time. Alkyl ionic liquids were shown to be biodegradable over the 6 months test period, and in contrast no significant mineralisation was observed with oxygenated derivatives. A different result was observed in the aerobic biodegradation of alkylated and oxygenated pyridinium ionic liquids in water because all the ionic liquids were almost completely degraded after 10 days, independently by the number of oxygen in the lateral chain of the cation. The synthesis of new ionic liquids by using renewable feedstock as starting materials, has been developed through the synthesis of furan-based ion pairs from furfural. The new ammonium salts were synthesised in very good yields, good purity of the products and wide versatility, combining low melting points with high decomposition temperatures and reduced viscosities. Regarding the possible applications as surfactants and biocides, furan-based salts could be a valuable alternative to benzyltributylammonium salts and benzalkonium chloride that are produced from non-renewable resources. A new procedure for the allylation of ketones and aldehydes with tetraallyltin in ionic liquids was developed. The reaction afforded high yields both in sulfonate-containing ILs and in ILs without sulfonate upon addition of a small amount of sulfonic acid. The checked reaction resulted in peculiar chemoselectivity favouring aliphatic substrates towards aromatic ketones and good stereoselectivity in the allylation of levoglucosenone. Finally ILs-based systems could be easily and successfully recycled, making the described procedure environmentally benign. The potential role of switchable polarity solvents as a green technology for the extraction of vegetable oil from terrestrial and aquatic biomass has been investigated. The extraction efficiency of terrestrial biomass rich in triacylglycerols, as soy bean flakes and sunflower seeds, was comparable to those of traditional organic solvents, being the yield of vegetable oils recovery very similar. Switchable polarity solvents as been also exploited for the first time in the extraction of hydrocarbons from the microalga Botryococcus braunii, demonstrating the efficiency of the process for the extraction of both dried microalgal biomass and directly of the aqueous growth medium. The switchable polarity solvents exhibited better extraction efficiency than conventional solvents, both with dried and liquid samples. This is an important issue considering that the harvest and the dewatering of algal biomass have a large impact on overall costs and energy balance.

Il lavoro svolto nel corso del mio dottorato ha avuto per oggetto lo studio dell’ inibizione della glicolisi aerobia (il principale processo metabolico utilizzato dalle cellule neoplastiche per produrre energia) ottenuta mediante il blocco dell’enzima lattato deidrogenasi (LDH). La mia attività si è concentrata sulla possibilità di utilizzare questo approccio allo scopo di migliorare l’efficacia della terapia antitumorale, valutandone gli effetti su colture di carcinoma epatocellulare umano Inizialmente, per valutare gli effetti della inibizione della LDH, è stato usato l’acido ossamico ( OXA). Questo composto è l’unico inibitore noto specifico per LDH ; è una molecola non tossica in vivo, ma attiva a concentrazioni troppo elevate per consentirne un uso terapeutico. Un importante risultato ottenuto è stata la dimostrazione che l’ inibizione della LDH ottenuta con OXA non è solo in grado di innescare una risposta di morte nelle cellule trattate, ma, associata alla somministrazione di sorafenib, aumenta fortemente l’efficacia di questo farmaco, determinando un effetto di sinergismo. Questo forte effetto di potenziamento dell’azione del farmaco è stato spiegato con la dimostrazione che il sorafenib ha la capacità di inibire il consumo di ossigeno delle cellule trattate, rendendole più dipendenti dalla glicolisi. Grazie alla collaborazione con il Dipartimento di Scienze Farmaceutiche il nostro gruppo di ricerca è arrivato alla identificazione di un composto (galloflavina) che inibisce la LDH con una efficienza molto maggiore di OXA. I risultati preliminari ottenuti sulle cellule di epatocarcinoma suggeriscono che la galloflavina potrebbe essere un composto promettente nel campo degli inibitori metabolici tumorali e inducono a una sua valutazione più approfondita come potenziale farmaco antineoplastico.

Ziel der vorliegenden Arbeit war es, Stoffwechseluntersuchungen an Experimentaltumoren von humanen Plattenepithelkarzinomen des Kopf-Hals-Bereiches mit bekannter Strahlenempfindlichkeit durchzuführen. Die Resultate sollten mit dem Genexpressionsniveau glykolyseassoziierter Transportproteine und Enzyme, der Proteinexpression von LDH-A, der Hypoxie und der Strahlenresistenz der Tumoren korreliert werden. Während die Tumorproben und die Daten zum biologischen Strahlenverhalten und zur Charakterisierung der Hypoxie aus Dresden stammen, wurden alle anderen Untersuchungen in Mainz durchgeführt. Ein wichtiges Merkmal der kooperativen Studie bestand darin, dass erstmals die Strahlenresistenz von Experimentaltumoren systematisch in einem klinischen Fraktionierungsschema untersucht wurde. Die lokale Bestimmung der Gewebskonzentrationen der Metabolite ATP, Glukose und Laktat erfolgte mit dem Verfahren der bildgebenden Biolumineszenz. Die Auswertung der Ergebnisse mit Unterstützung von Bildverarbeitungs-Software wurde weiterentwickelt und in wesentlichen Punkten verbessert. Zur Ermittlung des mRNA-Expressionsniveaus der Glykolyseenzyme PFK-L und LDH-A sowie des Glukosetransporters GLUT1 diente die real time RT-PCR-Methode. Ein Kernpunkt des methodischen Teils der vorliegenden Arbeit bildeten die Validierung und Etablierung dieses Verfahrens. Durch die Anwendung dieser Technik war es möglich, eine relative Quantifizierung des Expressionslevels durchzuführen. Die Western Blot-Analyse lieferte Aussagen über den Proteingehalt von LDH-A. Dabei kam ein neues Auswerteverfahren durch Anwendung fluoreszenzmarkierter Antikörper zum Einsatz. Die Ergebnisse zeigten erstmals einen direkten Zusammenhang zwischen dem Laktatgehalt von Tumoren und deren Strahlenresistenz. Es wurde im Vergleich zu früheren klinischen Untersuchungen eine Einstufung in Hoch- und Niedriglaktattumoren vorgenommen und eine signifikante Korrelation innerhalb der Hochlaktattumoren zwischen dem Laktatgehalt und der über Pimonidazol quantifizierten hypoxischen Fraktion festgestellt. Während die PCR Unterschiede in den drei untersuchten Genen auf transkriptioneller Ebene zwischen den sieben untersuchten Tumorlinien erkennen ließ, waren die Western Blot-Ergebnisse nahezu gleich. Da auch die Western Blot-Analysen keine Übereinstimmungen mit dem Laktatgehalt zeigten, kann auch der reine Proteingehalt keine Rolle als aktivitätsbestimmende Größe der Glykolyse spielen. Vielmehr scheinen Aktivierungs- und posttranslationale Prozesse oder auch eine Kombination mehrerer Faktoren eine Rolle zu spielen. Letztlich deuten die Befunde darauf hin, dass die glykolytische Aktivität der untersuchten Tumoren nicht über Transkription und Proteinexpression reguliert wird. Der Zusammenhang zwischen dem Laktatgehalt und der Strahlenresistenz der Tumoren kann von großer klinischer Bedeutung sein, da ein klinisch relevantes Fraktionierungsschema bei der Bestrahlung angewandt wurde. Unsere Ergebnisse bestätigen die Arbeitshypothese, dass ein hoher glykolytischer Flux mit einer hohen Umsatzrate an Metaboliten mit Radikalfängerfunktion, wie Pyruvat, einhergeht, die den Tumoren eine Radioresistenz verleihen. Der Laktatgehalt von Biopsien als Marker für die Strahlenresistenz könnte in Zukunft zu einer der Radiotherapie vorangehenden Patientenselektion herangezogen werden, um die Therapie- und insbesondere Dosisplanung in der Onkologie zu unterstützen.

In this research work the optimization of the electrochemical system of LDHs as catalytic precursors on FeCrAlY foams was carried out. Preliminary sintheses were performed on flat surfaces in order to easily characterize the deposited material. From the study of pH evolution vs time at different cathodic potentials applied to a Pt electrode, the theoretical best working conditions for the synthesis of single hydroxides and LDH compounds was achieved. In order to define the optimal potential for the synthesis of a particular LDH compound, the collected data were compared with the interval of precipitation determined by titration with NaOH. However, the characterization of the deposited material on Pt surfaces did not confirm the deposition of a pure and homogeneous LDH phase during the synthesis. Instead a sequential deposition linked to the pH of precipitation of the involved elements is observed. The same behavior was observed during the synthesis of the RhMgAl LDH on FeCrAlY foam as catalytic precursor. Several parameters were considered in order to optimize the synthesis.. The development of electrochemical cells with different feature, such as the counter electrode dimensions or the contact between the foam and the potentiostat, had been carried out in order to obtain a better coating of the foam. The influence of the initial pH of the electrolyte solution, of the applied potential, of the composition of the electrolytic solution were investigated in order to improve a better coating of the catalyst support. Catalytic tests were performed after the calcination of the deposited foam for the CPO and SR reactions, showing an improve of performances along with optimization of the precursors synthesis conditions.

Esta tesis presenta un breve estado de la cuestión de la sociología y la ética del deporte como nueva disciplina académica, desde su origen hasta el presente. En segundo lugar pretende ver que aporta la perspectiva de la sociologia relacional al estudio del deporte. La primera contribución es la “trans-discipliniariedad” entre disciplinas como la sociología y la ética, evitando caer en una ética “sociologizzata” o en una sociología “eticizzata”. Gracias a la relacionalidad se obtiene la des-mercantilización de bienestar. La lógica relacional busca el desarrollo económico sin olvida que la sociedad está formada por personas, que son lo realmente importantes por encima de los interés económicos. Otra contribución es la distinción entre la “sociedad humana” y la “sociedad de lo humano” que evita muchos de los actuales problemas que trae consigo las nuevas tecnologías. Y finalmente aporta el esquema “AGIL” como “brújula relacional” que ayuda a relaizar los objetivos, medios y reglas éticas dentro de la práctica deportiva.

Questa tesi descrive lo sviluppo di un elettrodo modificato con un polimero isolante per la determinazione indiretta del radicale OH. I polimeri testati sono stati polifenolo, polipirrolo e polipirrolo sovraoossidato ed il primo è risultato quello con le migliori prestazioni. Il film di modificante è stato depositato per elettropolimerizzazione del fenolo in ambiente acido, su un elettrodo di carbone vetroso (GC) ed è risultato isolante e perfettamente adeso al GC, impedendo il trasferimento di carica alle più comuni sonde redox. L’attacco dei radicali OH, generati dalla reazione di Fenton o dalla fotolisi di H2O2, rimuove parzialmente il polimero dal GC, ripristinando parzialmente il comportamento conduttore dell’elettrodo. L’entità della degradazione del film polifenolico è stata valutata sfruttando la corrente relativa alla sonda redox Ru(NH3)63+, che rappresenta il segnale analitico per la determinazione del radicale OH. L’elettrodo è stato impiegato per stimare le prestazioni di foto catalizzatori a base di nanoparticelle di TiO2, ottenendo risultati correlati a quelli ricavati da un metodo HPLC. Inoltre esso è stato usato per sviluppare una nuova procedura per la determinazione della capacità di scavenging verso i radicali OH, che è stata applicata all’analisi di composti puri e campioni reali. I risultati erano confrontabili con quelli determinati con metodiche standardizzate, comunemente impiegate per la determinazione della capacità antiossidante. Inoltre è stato condotto uno studio riguardante la modifica di un elettrodo di platino con un idrossido misto a strati a base di cobalto e alluminio (LDH). In particolare si sono valutati gli effetti di diversi pretrattamenti del Pt sulle caratteristiche e prestazioni elettrocatalitiche del film di LDH nei confronti dell’ossidazione di anilina, fenolo e acido salicilico. Questi composti possono essere impiegati come molecole sonda per la determinazione del radicale OH e rivestono interesse da un punto di vista elettroanalitico perché portano facilmente alla passivazione della superficie di Pt.

The aim of the research project discussed in this thesis was to study the inhibition of aerobic glycolysis, that is the metabolic pathway exploited by cancer cells for the ATP generation. This observation has led to the evaluation of glycolytic inhibitors as potential anticancer agents. Lactate dehydrogenase (LDH) is the only enzyme whose inhibition should allow a blocking of aerobic glycolysis of tumor cells without damaging the normal cells which, in conditions of normal functional activity and sufficient oxygen supply, do not need this enzyme. In preliminar experiments we demonstrated that oxamic acid and tartronic acid, two LDH competitive inhibitors, impaired aerobic glycolysis and replication of cells from human hepatocellular carcinoma. Therefore, we proposed that the depletion of ATP levels in neoplastic cells, could improved the chemotherapeutic index of associated anticancer drugs; in particular, it was studied the association of oxamic acid and multi-targeted kinase inhibitors. A synergistic effect in combination with sorafenib was observed, and we demonstrated that this was related to the capacity of sorafenib to hinder the oxidative phosphorylation, so that cells were more dependent to aerobic glycolysis. These results linked to LDH blockage encouraged us to search for LDH inhibitors more powerful than oxamic acid; thus, in collaboration with the Department of Pharmaceutical Sciences of Bologna University we identified a new molecule, galloflavin, able to inhibit both A and B isoforms of LDH enzyme. The effects of galloflavin were studied on different human cancer cell lines (hepatocellular carcinoma, breast cancer, Burkitt’s lymphoma). Although exhibiting different power on the tested cell lines, galloflavin was constantly found to inhibit lactate and ATP production and to induce cell death, mainly in the form of apoptosis. Finally, as LDH-A is able to bind single stranded DNA, thus stimulating cell transcription, galloflavin effects were also studied on this other LDH function.

The aim of this study was to examine whether a real high speed-short term competition influences clinicopathological data focusing on muscle enzymes, iron profile and Acute Phase Proteins. 30 Thoroughbred racing horses (15 geldings and 15 females) aged between 4-12 years (mean 7 years), were used for the study. All the animals performed a high speed-short term competition for a total distance of 154 m in about 12 seconds, repeated 8 times, within approximately one hour (Niballo Horse Race). Blood samples were obtained 24 hours before and within 30 minutes after the end of the races. On all samples were performed a complete blood count (CBC), biochemical and haemostatic profiles. The post-race concentrations for the single parameter were corrected using an estimation of the plasma volume contraction according to the individual Alb concentration. Data were analysed with descriptive statistics and the percentage of variation from the baseline values were recorded. Pre- and post-race results were compared with non-parametric statistics (Mann Whitney U test). A difference was considered significant at p<0.05. A significant plasma volume contraction after the race was detected (Hct, Alb; p<0.01). Other relevant findings were increased concentrations of muscular enzymes (CK, LDH; p<0.01), Crt (p<0.01), significant increased uric acid (p<0.01), a significant decrease of haptoglobin (p<0.01) associated to an increase of ferritin concentrations (p<0.01), significant decrease of fibrinogen (p<0.05) accompanied by a non-significant increase of D-Dimers concentrations (p=0.08). This competition produced relevant abnormalities on clinical pathology in galloping horses. This study confirms a significant muscular damage, oxidative stress, intravascular haemolysis and subclinical hemostatic alterations. Further studies are needed to better understand the pathogenesis, the medical relevance and the impact on performance of these alterations in equine sport medicine.

In dieser Arbeit wurden zytotoxische Effekte sowie die inflammatorische Reaktionen des distalen respiratorischen Traktes nach Nanopartikelexposition untersucht. Besondere Aufmerksamkeit lag auch auf der Untersuchung unterschiedlicher zellulärer Aufnahmewege von Nanopartikeln wie z.B. Clathrin- oder Caveolae-vermittelte Endozytose oder auch Clathrin- und Caveolae-unabhängige Endozytose (mit möglicher Beteiligung von Flotillinen). Drei unterschiedliche Nanopartikel wurden hierbei gewählt: amorphes Silica (aSNP), Organosiloxan (AmorSil) und Poly(ethyleneimin) (PEI). Alle unterschiedlichen Materialien gewinnen zunehmend an Interesse für biomedizinische Forschungsrichtungen (drug and gene delivery). Insbesondere finden aSNPs auch in der Industrie vermehrt Anwendung, und stellen somit ein ernstzunehmendes Gesundheitsrisiko dar. Dieser wird dadurch zu einem begehrten Angriffsziel für pharmazeutische Verabreichungen von Medikamenten über Nanopartikel als Vehikel aber bietet zugleich auch eine Angriffsfläche für gesundheitsschädliche Nanomaterialien. Aus diesem Grund sollten die gesundheitsschädigenden Risiken, sowie das Schicksal von zellulär aufgenommenen NPs sorgfältig untersucht werden. In vivo Studien an der alveolaren-kapillaren Barriere sind recht umständlich. Aus diesem Grund wurde in dieser Arbeit ein Kokulturmodel benutzt, dass die Alveolar-Kapillare Barrier in vivo nachstellt. Das Model besteht aus dem humanen Lungenepithelzelltyp (z.B. NCI H441) und einem humanen microvasculären Endothelzelltyp (z.B. ISO-HAS-1), die auf entgegengesetzten Seiten eines Transwell-Filters ausgesät werden und eine dichte Barriere ausbilden. Die NP Interaktion mit Zellen in Kokultur wurde mit denen in konventioneller Monokultur verglichen, in der Zellen 24h vor dem Experiment ausgesät werden. Diese Studie zeigt, dass nicht nur die polarisierte Eigenschaft der Zellen in Kokultur sondern auch die unmittelbare Nähe von Epithel und Endothelzelle ausschlaggebend für durch aSNPs verursachte Effekte ist. Im Hinblick auf inflammatorische Marker (sICAM, IL-6, IL8-Ausschüttung), reagiert die Kokultur auf aSNPs empfindlicher als die konventionelle Monokultur, wohingegen die Epithelzellen in der Kokultur auf zytotoxikologischer Ebene (LDH-Ausschüttung) unempfindlicher auf aSNPs reagierten als die Zellen in Monokultur. Aufnahmestudien haben gezeigt, dass die Epithelzellen in Kokultur entschieden weniger NPs aufnehmen. Somit zeigen die H441 in der Kokultur ähnliche epitheliale Eigenschaften einer schützenden Barriere, wie sie auch in vivo zu finden sind. Obwohl eine ausreichende Aufnahme von NPs in H441 in Kokultur erreicht werden konnte, konnte ein Transport von NPs durch die epitheliale Schicht und eine Aufnahme in die endotheliale Schicht mit den gewählten Inkubationszeiten nicht gezeigt werden. Eine Clathrin- oder Caveolae-vermittelte Endozytose von NPs konnte mittels Immunfluoreszenz weder in der Mono- noch in der Kokultur nachgewiesen werden. Jedoch zeigte sich eine Akkumulation von NPs in Flotillin-1 und-2 enthaltende Vesikel in Epithelzellen aus beiden Kultursystemen. Ergebnisse mit Flotillin-inhibierten (siRNA) Epithelzellen, zeigten eine deutlich geringere Aufnahme von aSNPs. Zudem zeigte sich eine eine reduzierte Viabilität (MTS) von aSNP-behandelten Zellen. Dies deutet auf eine Beteiligung von Flotillinen an unbekannten (Clathrin oder Caveolae -unabhängig) Endozytosemechanismen und (oder) endosomaler Speicherung. Zusammenfassend waren die Aufnahmemechanismen für alle untesuchten NPs in konventioneller Monokultur und Kokultur vergleichbar, obwohl sich die Barriereeigenschaften deutlich unterscheiden. Diese Arbeit zeigt deutlich, dass sich die Zellen in Kokultur anders verhalten. Die Zellen erreichen hierbei einen höheren Differenzierungsgrad und eine Zellkommunikation mit anderen relevanten Zelltypen wird ermöglicht. Durch das Einbringen eines dritten relevanten Zelltyps in die Kokultur, des Alveolarmakrophagen (Zelllinie THP-1), welcher die erste Verteidigungsfront im Alveolus bildet, wird diese Aussage weiter bekräftigt. Erste Versuche haben gezeigt, dass die Triplekultur bezüglich ihrer Barriereeigenschaften und IL-8-Ausschüttung sensitiver auf z.B. TNF- oder LPS-Stimulation reagiert als die Kokultur. Verglichen mit konventionellen Monokulturen imitieren gut ausgebildete, multizelluräre Kokulturmodelle viel präziser das zelluläre Zusammenspiel im Körper. Darum liefern Nanopartikelinteraktionen mit dem in vitro-Triplekulturmodel aufschlussreichere Ergebnisse bezüglich umweltbedingter oder pharmazeutischer NP-Exposition in der distalen Lung als es uns bisher möglich war.

Abstract The aim of this study was to assess the effects of a series of different surface coated quantum dots (QDs) (organic, carboxylated [COOH] and amino [NH(2)] polytethylene glycol [PEG]) on J774.A1 macrophage cell viability and to further determine which part of the QDs cause such toxicity. Cytotoxic examination (MTT assay and LDH release) showed organic QDs to induce significant cytotoxicity up to 48 h, even at a low particle concentration (20 nM), whilst both COOH and NH(2) (PEG) QDs caused reduced cell viability and cell membrane permeability after 24 and 48 h exposure at 80 nM. Subsequent analysis of the elements that constitute the QD core, core/shell and (organic QD) surface coating showed that the surface coating drives QD toxicity. Elemental analysis (ICP-AES) after 48 h, however, also observed a release of Cd from organic QDs. In conclusion, both the specific surface coating and core material can have a significant impact on QD toxicity.

This work was motivated by the incomplete characterization of the role of vascular endothelial growth factor-A (VEGF-A) in the stressed heart in consideration of upcoming cancer treatment options challenging the natural VEGF balance in the myocardium. We tested, if the cytotoxic cancer therapy doxorubicin (Doxo) or the anti-angiogenic therapy sunitinib alters viability and VEGF signaling in primary cardiac microvascular endothelial cells (CMEC) and adult rat ventricular myocytes (ARVM). ARVM were isolated and cultured in serum-free medium. CMEC were isolated from the left ventricle and used in the second passage. Viability was measured by LDH-release and by MTT-assay, cellular respiration by high-resolution oxymetry. VEGF-A release was measured using a rat specific VEGF-A ELISA-kit. CMEC were characterized by marker proteins including CD31, von Willebrand factor, smooth muscle actin and desmin. Both Doxo and sunitinib led to a dose-dependent reduction of cell viability. Sunitinib treatment caused a significant reduction of complex I and II-dependent respiration in cardiomyocytes and the loss of mitochondrial membrane potential in CMEC. Endothelial cells up-regulated VEGF-A release after peroxide or Doxo treatment. Doxo induced HIF-1α stabilization and upregulation at clinically relevant concentrations of the cancer therapy. VEGF-A release was abrogated by the inhibition of the Erk1/2 or the MAPKp38 pathway. ARVM did not answer to Doxo-induced stress conditions by the release of VEGF-A as observed in CMEC. VEGF receptor 2 amounts were reduced by Doxo and by sunitinib in a dose-dependent manner in both CMEC and ARVM. In conclusion, these data suggest that cancer therapy with anthracyclines modulates VEGF-A release and its cellular receptors in CMEC and ARVM, and therefore alters paracrine signaling in the myocardium.
