Background:Hepatitis C is a disease spread throughout the world. Hepatitis C virus (HCV), the etiological agent of this disease, is a single-stranded positive RNA virus. Its genome encodes a single precursor protein that yields ten proteins after processing. NS5A, one of the non-structural viral proteins, is most associated with interferon-based therapy response, the approved treatment for hepatitis C in Brazil. HCV has a high mutation rate and therefore high variability, which may be important for evading the immune system and response to therapy. The aim of this study was to analyze the evolution of NS5A quasispecies before, during, and after treatment in patients infected with HCV genotype 3a who presented different therapy responses.Methods:Viral RNA was extracted, cDNA was synthesized, the NS5A region was amplified and cloned, and 15 clones from each time-point were sequenced. The sequences were analyzed for evolutionary history, genetic diversity and selection.Results:This analysis shows that the viral population that persists after treatment for most non-responder patients is present in before-treatment samples, suggesting it is adapted to evade treatment. In contrast, the population found in before treatment samples from most end-of-treatment responder patients either are selected out or appears in low frequency after relapse, therefore changing the population structure. The exceptions illustrate the uniqueness of the evolutionary process, and therefore the treatment resistance process, in each patient.Conclusion:Although evolutionary behavior throughout treatment showed that each patient presented different population dynamics unrelated to therapy outcome, it seems that the viral population from non-responders that resists the treatment already had strains that could evade therapy before it started. © 2013 Bittar et al.

Currently, there has been an increasing demand for operational and trustworthy digital data transmission and storage systems. This demand has been augmented by the appearance of large-scale, high-speed data networks for the exchange, processing and storage of digital information in the different spheres. In this paper, we explore a way to achieve this goal. For given positive integers n,r, we establish that corresponding to a binary cyclic code C0[n,n-r], there is a binary cyclic code C[(n+1)3k-1,(n+1)3k-1-3kr], where k is a nonnegative integer, which plays a role in enhancing code rate and error correction capability. In the given scheme, the new code C is in fact responsible to carry data transmitted by C0. Consequently, a codeword of the code C0 can be encoded by the generator matrix of C and therefore this arrangement for transferring data offers a safe and swift mode. © 2013 SBMAC - Sociedade Brasileira de Matemática Aplicada e Computacional.

Experiments of biomass combustion were performed to determine whether specimen size, tray inclination, or combustion air flow rate was the factor that most affects the emission of carbon dioxide, carbon monoxide, and methane. The chosen biomass was Eucalyptus citriodora, a very abundant species in Brazil, utilized in many industrial applications, including combustion for energy generation. Analyses by gas chromatograph and specific online instruments were used to determine the concentrations of the main emitted gases, and the following figures were found for the emission factors: 1400 ± 101 g kg-1 of CO2, 50 ± 13 g kg-1 of CO, and 3.2 ± 0.5 g kg-1 of CH4, which agree with values published in the literature for biomass from the Amazon rainforest. Statistical analysis of the experiments determined that specimen size most significantly affected the emission of gases, especially CO2 and CO. •Statistical analysis to determine effects on emission factors.•CO2, CO, CH4 emission factors determined for combustion of Eucalyptus.•Laboratory results agreed with data for Amazonian biomass combustion in field tests.•Combustion behavior under flaming and smoldering was analyzed. © 2013 Elsevier Ltd.

The aim of the present study was to analyze the effects of age on cardiorespiratory fitness (CRF), muscle strength and heart rate (HR) response to exercise adaptation in women in response to a long-term twice-weekly combined aerobic and resistance exercise program. 85 sedentary women, divided into young (YG; n=22, 30.3±6.2 years), early middle-aged (EMG; n=28, 44.1±2.5 years), late middle-aged (LMG; n=20, 56.7±3.5 years) and older (OG; n=15, 71.4±6.9 years) groups, had their CRF, muscle strength (1-repetition maximum test) and HR response to exercise (graded exercise test) measured before and after 12 months of combined exercise training. Exercise training improved CRF and muscle strength in all age groups (P<0.05), and no significant differences were observed between groups. Exercise training also improved resting HR and recovery HR in YG and EMG (P<0.05), but not in LMG and OG. Maximal HR did not change in any group. Combined aerobic and resistance training at a frequency of 2 days/week improves CRF and muscle strength throughout the lifespan. However, exercise-induced improvements in the HR recovery response to exercise may be impaired in late middle-aged and older women. © Georg Thieme Verlag KG.

Mammalian natriuretic peptides (NPs) have been extensively investigated for use as therapeutic agents in the treatment of cardiovascular diseases. Here, we describe the isolation, sequencing and tridimensional homology modeling of the first C-type natriuretic peptide isolated from scorpion venom. In addition, its effects on the renal function of rats and on the mRNA expression of natriuretic peptide receptors in the kidneys are delineated. Fractionation of Tityusserrulatus venom using chromatographic techniques yielded a peptide with a molecular mass of 2190.64Da, which exhibited the pattern of disulfide bridges that is characteristic of a C-type NP (TsNP, T. serrulatus Natriuretic Peptide). In the isolated perfused rat kidney assay, treatment with two concentrations of TsNP (0.03 and 0.1μg/mL) increased the perfusion pressure, glomerular filtration rate and urinary flow. After 60min of treatment at both concentrations, the percentages of sodium, potassium and chloride transport were decreased, and the urinary cGMP concentration was elevated. Natriuretic peptide receptor-A (NPR-A) mRNA expression was down regulated in the kidneys treated with both concentrations of TsNP, whereas NPR-B, NPR-C and CG-C mRNAs were up regulated at the 0.1μg/mL concentration. In conclusion, this work describes the isolation and modeling of the first natriuretic peptide isolated from scorpion venom. In addition, examinations of the renal actions of TsNP indicate that its effects may be related to the activation of NPR-B, NPR-C and GC-C. © 2013 Elsevier Ltd.

The need for renewal and a more efficient use of energy resources has provided an increased interest in studies of methane activation processes in the gas phase by transition metal oxides. In this respect, the present work is an effort to assess , by means of a computational standpoint, the reactivity of NbOm n+ and FeOm n+ (m = 1, 2, n = 0, 1, 2) oxides in the activation process of the methane C-H bond, which corresponds to the first rate limiting step in the process of converting methane to methanol. These oxides are chosen, primarily, because the iron oxides are the most experimentally studied, and iron ions are more abundant in biological mediums. The main motive for choosing niobium oxides is the abundance of natural reserves of this mineral in Brazil (98%), especially in Minas Gerais. Initially, a thorough investigation was conducted, using different theoretical methods, to analyze the structural and electronic properties of the investigated oxides. Based on these results, the most reliable methodology was selected to investigate the activation process of the methane C-H bond by the series of iron and niobium oxides, considering all possible reaction mechanisms known to activate the C-H bond of alkanes. It is worth noting that, up to this moment and to our knowledge, there are no papers, in literature , investigating and comparing all the mechanisms considered in this work. I n general, the main results obtained show different catalytic tendencies and behaviors throughout the series of monoxides and dioxides of iron and niobium. An important and common result found in the two studies is that the increase in the load on the metal center and the addition of oxygen atoms to the metal, clearly favor the initial thermodynamics of the reaction, i.e., favor the approach of the metal center to methane, distorting its electron cloud and, thereby, decreasing its inertia. Comparing the two sets of oxides, we conclude that the iron oxides are the most efficient in activating the methane C-H bond. Among the iron oxides investigated, FeO + showed better kinetic and thermodynamic performance in the reaction with methane, while from the niobium oxides and ions NbO 2+ and NbO2 2+, showed better catalytic efficiency in the activation of the methane C-H bond.

A cromoblastomicose e uma infeccao subcutanea cronica, granulomatosa, causada pela implantacao traumatica de diversas especies de fungos demaceos, sendo Fonsecaea pedrosoi o principal agente etiologico. O Brasil possui a segunda maior prevalencia mundial da doenca, sendo o estado do Para a maior area endemica. Histologicamente, a cromomicose e caracterizada pela presenca de celulas gigantes, onde podem ser observadas celulas escleroticas fagocitadas por macrofagos. O objetivo do presente estudo foi analisar os diferentes aspectos da interacao de macrofagos peritoneais de camundongos BALB/c e C57/BL6 com conidios ou celulas escleroticas de F. pedrosoi, determinando os indices de infeccao, fagocitose e fusao celular. Os resultados mostraram indices de fagocitose e infeccao maiores em conidios do que em celulas escleroticas para BALB/c (p<0.05), ocorrendo efeito inverso no indice de fusao, com a formacao de celulas gigantes do tipo Langhans na interacao com celulas escleroticas e celulas gigantes do tipo corpo estranho na interacao com conidios. Os macrofagos de BALB/c em interacao com conidios produziram mais TNF-α que o controle nos tempos de 3 a 72h; e mais IL-10 apos 3h. Macrofagos interagindo com celulas escleroticas produziram mais TNF-α que o controle nos tempos de 1h e 3h; e a quantidade de IL- 10 foi maior apos 72h de interacao. No co-cultivo de macrofagos de C57/BL6 com conidios observou-se a presenca de vacuolos aumentados apos 24h, enquanto na interacao com celulas escleroticas, os macrofagos se desprenderam da laminula nos tempos posteriores a 24 h. A quantidade de TNF-α e maior na interacao de conidios comparado ao controle em 1 e 72 h; e a quantidade de IL-10, no tempo de 48h. Ja na interacao com celulas escleroticas, apenas a quantidade de IL-10 diferiu do controle, sendo maior nos tempos de 1 a 48h. Estes dados sugerem que a resposta e macrofagos ao fungo e diferente entre os camundongos de BALB/c e C57/BL6, diferindo tambem a resposta de um mesmo tipo de macrofago para cada forma fungica, sendo as celulas escleroticas aparentemente mais imunogenicas que os conidios.
