Free radicals are produced during aerobic cellular metabolism and have key roles as regulatory mediators in signaling processes. Oxidative stress reflects an imbalance between production of reactive oxygen species and an adequate antioxidant defense. This adverse condition may lead to cellular and tissue damage of components, and is involved in different physiopathological states, including aging, exercise, inflammatory, cardiovascular and neurodegenerative diseases, and cancer. In particular, the relationship between exercise and oxidative stress is extremely complex, depending on the mode, intensity, and duration of exercise. Regular moderate training appears beneficial for oxidative stress and health. Conversely, acute exercise leads to increased oxidative stress, although this same stimulus is necessary to allow an up-regulation in endogenous antioxidant defenses (hormesis). Supporting endogenous defenses with additional oral antioxidant supplementation may represent a suitable noninvasive tool for preventing or reducing oxidative stress during training. However, excess of exogenous antioxidants may have detrimental effects on health and performance. Whole foods, rather than capsules, contain antioxidants in natural ratios and proportions, which may act in synergy to optimize the antioxidant effect. Thus, an adequate intake of vitamins and minerals through a varied and balanced diet remains the best approach to maintain an optimal antioxidant status. Antioxidant supplementation may be warranted in particular conditions, when athletes are exposed to high oxidative stress or fail to meet dietary antioxidant requirements. Aim of this review is to discuss the evidence on the relationship between exercise and oxidative stress, and the potential effects of dietary strategies in athletes. The differences between diet and exogenous supplementation as well as available tools to estimate effectiveness of antioxidant intake are also reported. Finally, we advocate the need to adopt an individualized diet for each athlete performing a specific sport or in a specific period of training, clinically supervised with inclusion of blood analysis and physiological tests, in a comprehensive nutritional assessment. Naked maghemite nanoparticles, namely, surface active maghemite nanoparticles (SAMNs), characterized by a diameter of about 10 nm, possessing peculiar colloidal stability, surface chemistry, and superparamagnetism, present fundamental requisites for the development of effective magnetic purification processes for biomolecules in complex matrices. Polyphenolic molecules presenting functionalities with different proclivities toward iron chelation were studied as probes for testing SAMN suitability for magnetic purification. Thus, the binding efficiency and reversibility on SAMNs of phenolic compounds of interest in the pharmaceutical and food industries, namely, catechin, tyrosine, hydroxytyrosine, ferulic acid, coumaric acid, rosmarinic acid, naringenin, curcumin, and cyanidin-3-glucoside, were evaluated. Curcumin emerged as an elective compound, suitable for magnetic purification by SAMNs from complex matrices. A combination of curcumin, demethoxycurcumin, and bis-demethoxycurcumin was recovered by a single magnetic purification step from extracts of Curcuma longa rhizomes, with a purity >98% and a purification yield of 45%, curcumin being >80% of the total purified curcuminoids.

This work was based to study the influence of the storage temperature (cold and room temperature) in the quality of inflorescences strelitzia. The scapes were selected, labeled and there were zero problems concerning mechanical damage, disease and/or plagues. Subsequently this period, the scapes were moved randomly to recipients with water, in which two postharvest trials were conducted. In experiment 1, the flower scapes were placed in buckets with water from public supply and sanitation department and taken to a cold room at temperature of 7.5 degrees C and RH of 90%, for a twelve day period. For the experiment 2, were kept under the same conditions but at room temperature for a period of six days. In both experiments, the visual analysis: color, gloss, stains (by assigning notes), opening and drop florets (count) were evaluated at intervals of four days in cold and every 48 hours at ambient temperature conditions. The sepal is the organ that showed greater loss in coloration. The variable gloss showed the same pattern for the two experiments. Incidences of stains on the inflorescences occurred in patches at room temperature. The scapes increased number of florets open in cold. This tendency did not occur at room temperature. No were observed differences in the fall of florets. Conclude that the storage temperature does not contribute to postharvest quality of strelitzia.

Curcumin possesses wide-ranging anti-inflammatory and anti-cancer properties and its biological activity can be correlated to its potent antioxidant capacity. Novel maghemite (gamma-Fe3O4) nanoparticles, characterized by a diameter of about 10 nm and possessing peculiar colloidal properties and surface interactions, called Surface Active Maghemite Nanoparticles (SAMN), were superficially modified with curcumin by simple incubation, due to the presence of under-coordinated Fe(III) atoms on nanoparticle surface. The resulting curcumin-modified SAMNs (SAMN@curcumin) were characterized by transmission electron microscopy (TEM), FTIR, Mossbauer, EPR and UV-Vis spectroscopy. The redox properties of bound curcumin were tested by electrochemistry. Finally, SAMN@curcumin was studied in the presence of different electroactive substances, namely hydroquinone, NADH and ferrocyanide, in order to assess its electrochemical behavior. Moreover, SAMN@curcumin was electrochemically tested in the presence of one of the most diffuse reactive oxygen specie, such as hydrogen peroxide, demonstrating its stability. SAMN@curcumin in which curcumin is firmly bound, but still retaining its redox features represents a feasible adduct: a magnetically drivable nano-bio-conjugate mimicking free Curcumin redox behavior. The proposed nanostructured material could be exploited as magnetic drivable curcumin vehicle for biomedical applications.

O objetivo deste trabalho foi avaliar os efeitos de BAP (6-benzilaminopurina) e NAA (ácido naftaleno acético) na indução, na multiplicação in vitro de gemas, nas brotações de Ananas comosus da cultivar 'IAC Gomo-de-mel' e a correlação desses efeitos com a atividade de peroxidase e o teor de proteína solúvel total. Foram utilizadas gemas axilares retiradas da coroa de frutos sadios, inoculadas em tubo de ensaio contendo meio de cultura MS solidificado com ágar a 5%, pH ajustado para 5,7, contendo os tratamentos que incluíam diferentes concentrações e combinações de BAP (0, 0,5, 1,0 e 1,5mg L-1) e NAA (0, 0,5 e 1,0mg L-1). Nessa fase, aos 65 dias, ocorreu a formação de 2,24 brotações, utilizando-se 1mg L-1 de BAP. Após o desenvolvimento, as gemas foram inoculadas em meio MS líquido associado a dois tratamentos (1,0mg L-1 BAP + 0,5mg L-1 NAA e 1,0mg L-1 BAP + 1,0mg L-1 NAA) e, aos 95 dias, o meio de cultura mais adequado foi aquele que continha 1,0mg L-1 BAP + 0,5mg L-1 NAA, proporcionando 7,42 brotações, menor porcentagem de hiper-hidricidade, maior número de brotações e indução de gemas. As proteínas solúveis apresentaram relação negativa com hiper-hidricidade e comprimento de brotações. A atividade da peroxidase foi maior em plantas com maior número de brotos e com maior porcentagem de hiper-hidricidade.

Este trabalho teve como objetivo estudar o efeito das poliaminas espermidina e espermina no crescimento de calos Hancornia speciosa Gomes. Calos com 0,5 cm de diâmetro foram inoculados em meio Murashige & Skoog (1962) (MS) a 50% + 100 mg L-1 de caseína hidrolisada + 200 mg L-1 de levedura de cerveja, variando os tratamentos:A: 1 mmol de espermina + 2 mg L-1 de 2,4-D (ácido 2,4 diclorofenoxiacético) + 0,5 mg L-1 de NAA (ácido naftalenoacético); B: 1 mmol de espermidina + 2 mg L-1 de 2,4-D + 0,5 mg L-1 de NAA; C: 2 mg L-1 de 2,4-D + 0,5 mg L-1 de NAA. Não houve influência das poliaminas no crescimento dos calos. observou-se, nos calos tratados com espermidina, maior concentração celular de putrescina (582,37 µg g mf-1) aos 60 dias, maior teor de espermidina (502,54 µg g mf-1) e espermina (868,53 µg g mf-1) aos 40 dias de cultivo, quando se aplicou a própria poliamina. Conclui-se que a aplicação exógena de poliaminas em Hancornia speciosa não proporciona aumento no crescimento de calos. A oxidação promovida por longos períodos de cultivo in vitro induz aumento nos níveis de putrescina.

Relationships between endogenous levels of polyamines by thin layer chromatography (TLC) and gas chromatography (GC), nitrate and response to the application of ethylene were established between organic and conventional vegetables (broccoli, collard greens, carrots and beets), both raw and cooked. Responses to ethylene showed that organic plants were less responsive to the growth regulator. The levels of free polyamines obtained by TLC were higher in organic vegetables. Organic broccoli showed higher levels of putrescine (Put), and cooking resulted in lowering the overall content of these amines. Conventional collard green showed the highest level of putrescine in the leaves compared with organic. Tubers of carrots and beets contain the highest levels of Put. These plants also contain high levels of spermine. GC analysis showed the highest polyamines contents compared with those obtained by TLC. Cooking process decreased putrescine and cadaverine content, both in conventionally and organically grown vegetables. Organic beets contain lower NO3(-) compared with its conventional counterpart.

This study examined the influence of different speed increments during treadmill exercise tests on peak treadmill speed (Vpeak) and its relationship with a 1-h treadmill running performance. 18 male recreational and amateur runners (10-km running pace: 10–15 km·h−1) performed, in an alternate order, 3 continuous incremental exercise tests with different speed increments (0.5, 1.0 and 2.0 km·h−1) on a motorized treadmill to determine Vpeak. Thereafter they undertook a 1-h time trial on a treadmill. Vpeak was determined as either (a) the highest speed that could be maintained for a complete minute (Vpeak-60 s), (b) the speed of the last complete stage (Vpeak-C), or (c) the speed of the last complete stage added to the product of the speed increment and the completed fraction of the incomplete stage (Vpeak-P). The Vpeak values were highly influenced by the different speed-incremented rates and the Vpeak-P determined during the protocol comprising speed increments of 1 km·h−1 presented the highest correlation with 1-h time trial performance (r=0.89). The results suggest that a protocol with speed increments comprising 1 km·h−1 and with a 3-min stage duration should be used as standard for the determination of Vpeak to assess aerobic fitness and predict endurance performance in recreational runners. Furthermore, the Vpeak-P should be used for the determination of Vpeak.

Objective The present study aimed to examine the reproducibility of heart rate (HR) and rating of perceived exertion (RPE) values obtained during different incremental treadmill tests. Equipment and methods Twenty male, recreational, endurance-trained runners (10-km running pace: 10–15 km·h−1) performed, in a counterbalanced order, three continuous incremental exercise tests with different speed increments (0.5 km·h−1, 1 km·h−1 and 2 km·h−1). Thereafter, each participant performed the three tests again, maintaining the same order as before. The reproducibility of the HR and RPE values were analyzed for all protocols during submaximal intensities (8, 10, 12, and 14 km·h−1). In addition, it was examined the reproducibility of maximal HR (HRmax) and peak RPE (RPEpeak). Results The variability of both the HR and RPE values showed a tendency to decrease over the stages during the incremental test and was not or slightly influenced by the incremental test design. The HR at 14 km·h−1 and HRmax presented the highest reproducibility (CV < 2%). In contrast, the submaximal RPE values showed higher variability indices (i.e., CV > 5.0%). In conclusion, the HR values were highly reproducible during the stages of the incremental test, in contrast to the RPE values that presented limited reproducibility.
