The fungal pathogen Claviceps purpurea infects ovaries of a broad range of temperate grasses and cereals, including hexaploid wheat, causing a disease commonly known as ergot. Sclerotia produced in place of seed carry a cocktail of harmful alkaloid compounds that result in a range of symptoms in humans and animals, causing ergotism. Following a field assessment of C. purpurea infection in winter wheat, two varieties ‘Robigus’ and ‘Solstice’ were selected which consistently produced the largest differential effect on ergot sclerotia weights. They were crossed to produce a doubled haploid mapping population, and a marker map, consisting of 714 genetic loci and a total length of 2895 cM was produced. Four ergot reducing QTL were identified using both sclerotia weight and size as phenotypic parameters; QCp.niab.2A and QCp.niab.4B being detected in the wheat variety ‘Robigus’, and QCp.niab.6A and QCp.niab.4D in the variety ‘Solstice’. The ergot resistance QTL QCp.niab.4B and QCp.niab.4D peaks mapped to the same markers as the known reduced height (Rht) loci on chromosomes 4B and 4D, Rht-B1 and Rht-D1, respectively. In both cases, the reduction in sclerotia weight and size was associated with the semi-dwarfing alleles, Rht-B1b from ‘Robigus’ and Rht-D1b from ‘Solstice’. Two-dimensional, two-QTL scans identified significant additive interactions between QTL QCp.niab.4B and QCp.niab.4D, and between QCp.niab.2A and QCp.niab.4B when looking at sclerotia size, but not between QCp.niab.2A and QCp.niab.4D. The two plant height QTL, QPh.niab.4B and QPh.niab.4D, which mapped to the same locations as QCp.niab.4B and QCp.niab.4D, also displayed significant genetic interactions.

Since the advent of the postgenomic era, efforts have focused on the development of rapid strategies for annotating plant genes of unknown function. Given its simplicity and rapidity, virus-induced gene silencing (VIGS) has become one of the preeminent approaches for functional analyses. However, several problems remain intrinsic to the use of such a strategy in the study of both metabolic and developmental processes. The most prominent of these is the commonly observed phenomenon of ""sectoring"" the tissue regions that are not effectively targeted by VIGS. To better discriminate these sectors, an effective marker system displaying minimal secondary effects is a prerequisite. Utilizing a VIGS system based on the tobacco rattle virus vector, we here studied the effect of silencing the endogenous phytoene desaturase gene (pds) and the expression and subsequent silencing of the exogenous green fluorescence protein (gfp) on the metabolism of Arabidopsis (Arabidopsis thaliana) leaves and tomato (Solanum lycopersicum) fruits. In leaves, we observed dramatic effects on primary carbon and pigment metabolism associated with the photobleached phenotype following the silencing of the endogenous pds gene. However, relatively few pleiotropic effects on carbon metabolism were observed in tomato fruits when pds expression was inhibited. VIGS coupled to gfp constitutive expression revealed no significant metabolic alterations after triggering of silencing in Arabidopsis leaves and a mild effect in mature green tomato fruits. By contrast, a wider impact on metabolism was observed in ripe fruits. Silencing experiments with an endogenous target gene of interest clearly demonstrated the feasibility of cosilencing in this system; however, carefully constructed control experiments are a prerequisite to prevent erroneous interpretation.

The feeding habits of the maned wolf were studied in southeast Brazil to understand its response towards changes in the environment and in relation to its prey. By occurrence, miscellaneous fruits, small mammals and wolf`s fruit were the most consumed items. Armadillos, small mammals and wolf`s fruit ( Solanum lycocarpum) provided most of the ingested biomass. While wolf`s fruit and small mammals were mainly consumed in the dry season, other miscellaneous fruits were taken mostly in the wet season. There was selectivity in the predation on some small mammal species. The maned wolf`s diet followed patterns similar to those found in more pristine areas.

Vegetables are critical for human health as they are a source of multiple vitamins including vitamin E (VTE). In plants, the synthesis of VTE compounds, tocopherol and tocotrienol, derives from precursors of the shikimate and methylerythritol phosphate pathways. Quantitative trait loci (QTL) for alpha-tocopherol content in ripe fruit have previously been determined in an Solanum pennellii tomato introgression line population. In this work, variations of tocopherol isoforms (alpha, beta, gamma, and delta) in ripe fruits of these lines were studied. In parallel all tomato genes structurally associated with VTE biosynthesis were identified and mapped. Previously identified VTE QTL on chromosomes 6 and 9 were confirmed whilst novel ones were identified on chromosomes 7 and 8. Integrated analysis at the metabolic, genetic and genomic levels allowed us to propose 16 candidate loci putatively affecting tocopherol content in tomato. A comparative analysis revealed polymorphisms at nucleotide and amino acid levels between Solanum lycopersicum and S. pennellii candidate alleles. Moreover, evolutionary analyses showed the presence of codons evolving under both neutral and positive selection, which may explain the phenotypic differences between species. These data represent an important step in understanding the genetic determinants of VTE natural variation in tomato fruit and as such in the ability to improve the content of this important nutriceutical.

We studied the potential contribution of frugivorous bats to the reestablishment of vegetational diversity in a restored area. We analysed the diets of the bat species and the differences between them in the consumption of fruits of autochtonous and allochthonous species. Planted (autochtonous) species were the basis of diets, especially Solanum mauritianum and Cecropia pachystachya, whereas for allochthonous species we found that Piperaceae to be of particular importance. Carollia perspicillata was the main seed disperser for allochthonous species, and potentially the most important bat in the promotion of vegetation diversity in the study area. Our results suggest that frugivorous bats are especially important in the reestablishment of vegetation in disturbed areas, and that restorarion efforts should focus on the planting of different zoochorous species that would guarantee a high year-round fruit production, thereby facilitating natural plant reestablishment by frugivorous bats in regenerating areas. With the aim of determining the genetic basis of metabolic regulation in tomato fruit, we constructed a detailed physical map of genomic regions spanning previously described metabolic quantitative trait loci of a Solanum pennellii introgression line population. Two genomic libraries from S. pennellii were screened with 104 colocated markers from five selected genomic regions, and a total of 614 bacterial artificial chromosome (BAC)/cosmids were identified as seed clones. Integration of sequence data with the genetic and physical maps of Solanum lycopersicum facilitated the anchoring of 374 of these BAC/cosmid clones. The analysis of this information resulted in a genome-wide map of a nondomesticated plant species and covers 10% of the physical distance of the selected regions corresponding to approximately 1% of the wild tomato genome. Comparative analyses revealed that S. pennellii and domesticated tomato genomes can be considered as largely colinear. A total of 1,238,705 bp from both BAC/cosmid ends and nine large insert clones were sequenced, annotated, and functionally categorized. The sequence data allowed the evaluation of the level of polymorphism between the wild and cultivated tomato species. An exhaustive microsynteny analysis allowed us to estimate the divergence date of S. pennellii and S. lycopersicum at 2.7 million years ago. The combined results serve as a reference for comparative studies both at the macrosyntenic and microsyntenic levels. They also provide a valuable tool for fine-mapping of quantitative trait loci in tomato. Furthermore, they will contribute to a deeper understanding of the regulatory factors underpinning metabolism and hence defining crop chemical composition.

In the course of our research program to discover novel antileishmanial agents, a biological screening of natural products against Leishmania major promastigotes allowed the identification of a furoquinoline alkaloid (1) and a furanocoumarin (2) as new hits. Subsequently, an integrated ligand-based virtual screening approach was employed to search for new antileishmanial compounds using these naturally occurring molecules as templates. Fourteen out of 40 compounds selected from a database of about 800,000 compounds (extracted from ZINC, a free database for virtual screening) were experimentally confirmed to possess significant in vitro antileishmanial properties. The application of ligand-based virtual screening as a complementary approach to experimental natural product screening was a useful strategy to facilitate the identification of new promising lead candidates.

Cathepsin V is a lysosomal cysteine peptidase highly expressed in thymus, testis and corneal epithelium. Eleven acridone alkaloids were isolated from Swinglea glutinosa (Bl.) Merr. (Rutaceae), with eight of them being identified as potent and reversible inhibitors of cathepsin V (IC(50) values ranging from 1.2 to 3.9 mu M). Detailed mechanistic characterization of the effects of these compounds on the cathepsin V-catalyzed reaction showed clear competitive inhibition with respect to substrate, with dissociation constants (K(i)) in the low micromolar range (2, K(i) = 1.2 mu M; 6, K(i) = 1.0 mu M; 7, K(i) = 0.2 mu M; and 11, K(i) = 1.7 mu M). Molecular modeling studies provided important insight into the structural basis for binding affinity and enzyme inhibition. Experimental and computational approaches, including biological evaluation, mode of action assessment and modeling studies were successfully employed in the discovery of a small series of acridone alkaloid derivatives as competitive inhibitors of catV. A new route to obtain the polyalkylated indole (+/-)-trans-trikentrin A was developed. The synthesis of this natural alkaloid features a thallium(III)mediated ring contraction reaction to obtain the trans-1,3-disubstituted five-membered ring in a diastereoselective manner. Thallium(III) is chemoselective in this rearrangement, reacting with the olefin without oxidation of the indole moiety. Other key transformations are the Bartoli`s reaction to construct the heterocyclic ring and a Heck coupling to add the carbons atom that will originate the nonaromatic cycle.

Plantas de batata (Solanum tuberosum L.) com sintomas de murcha bacteriana (MB) foram coletadas em 25 lavouras de 10 municípios de quatro regiões produtoras do Rio Grande do Sul (RS), de setembro a dezembro de 1999. Quatrocentos e noventa isolados de Ralstonia solanacearum foram obtidos e 96 e 4% identificados como biovares 1 e 2, respectivamente. A análise dos resultados da PCR-ERIC e BOX de 13 estirpes da biovar 1 e 72 da biovar 2 demonstrou baixa variabilidade genética. Através de RAPD foi possível associar local de origem do isolado com perfil eletroforético. Em outro experimento avaliou-se o comportamento de 14 cultivares e clones de batata cultivados nos períodos das safras de primavera de 1999 e 2000, em uma área naturalmente infestada com a biovar 2, em Caxias do Sul, RS. O número de plantas com sintomas foi registrado semanalmente. Os tubérculos produzidos por plantas assintomáticas foram coletados e submetidos a testes para detectar a presença de R. solanacearum. A área sob a curva de progresso da doença foi utilizada para comparar a resistência dos genótipos de batata à MB e o modelo logístico foi o que melhor se ajustou. A cultivar cruza 148 e o clone MB 03 mostraram-se como os mais resistentes, apresentando, no entanto, tubérculos com infecções latentes. As implicações da incidência de R. solanacearum em lavouras de batata e de sua variabilidade genética, assim como da resistência dos genótipos acompanhada de infecções latentes, no manejo integrado da MB no RS foram discutidas.

Nas últimas décadas o conhecimento florístico e taxonômico sobre a família Solanaceae no Rio Grande do Sul tem recebido incremento significativo de informações. Dos gêneros com espécies nativas, Bouchetia, Calibrachoa, Nicotiana, Nierembergia, Petunia e Solanum já foram alvo de revisões. Visando complementar o estudo desta família no Estado, foram estudados os gêneros Acnistus, Athenaea, Aureliana, Brunfelsia, Capsicum, Cestrum, Dyssochroma, Grabowskia, Lycianthes, Solandra e Vassobia, além das espécies do gênero Solanum seção Pachyphylla (Cyphomandra sensu lato). Foram catalogadas 22 espécies nativas, sendo três destas, novas citações de ocorrência. Para os onze primeiros gêneros foram identificados 19 espécies, dos quais cinco são de ocorrência restrita (Athenaea picta, Dyssochroma longipes, Grabowskia duplicata, Lycianthes rantonnei e Solandra grandiflora). A seção Pachyphylla do gênero Solanum está representada por três espécies. Dessa forma, foram atualizados os dados quali e quantitativos sobre a participação da família Solanaceae na flora sul-riograndense. Para cada gênero e para cada táxon específico foram elaboradas descrições, e acrescentados dados sobre fenologia, hábitat, comentários pertinentes e mapas de ocorrência. Chaves analíticas para identificação das espécies, quando mais de uma, também foram elaboradas. A revisão de doze herbários regionais possibilitou a elaboração de uma sinopse taxonômica e de uma chave analítica para identificação dos gêneros presentes no Estado. Dos 27 gêneros confirmados, 22 têm espécies nativas. O número de espécies nativas é de 114 e o de introduzidas, cultivadas ou assilvestradas, é até o momento 26.

A hidroponia é uma técnica poderosa na utilização eficiente dos recursos agrícolas, pois permite um ganho quer na produtividade, quer na qualidade do fruto. Atualmente os sistemas hidropónicos utilizados em plantas de médio porte não são os mais adequados, sendo esta uma área a explorar. Neste trabalho criou-se um novo sistema hidropónico (Deep Large Flow Technique- DLFT) para aplicação a plantas de médio porte e avaliou-se o seu impacto no crescimento, produtividade, biomassa e qualidade do tomate cereja (Solanum lycopersicum var. Moscatel RZ). O sistema de cultivo tradicional (solo) e semi-hidropónico (suporte com fibra de coco) serviram como base de comparação. Observou-se que após 31 dias as plantas cultivadas no novo sistema hidropónico obtiveram um crescimento acentuado, apresentando 3x mais frutos. Em termos de produtividade, os sistemas hidropónicos foram iguais, sendo o sistema tradicional 4x inferior. O total de biomassa foi significativamente maior no novo sistema hidropónico, com mais 20% e 88% que o sistema semi-hidropónico e tradicional, respetivamente. As plantas produzidas no sistema DLFT apresentaram frutos com qualidade superior, com um rácio de monossacarídeos/acidez de 6,6 mg/g. O conteúdo total de ácidos gordos nos frutos cultivados com este sistema foi 39% e 44% superior aos do semihidropónico e do tradicional. Contrariamente, o conteúdo de flavonóides foi inferior nos frutos cultivados com o novo sistema hidropónico, tendo os frutos do sistema tradicional e do sistema semi-hidropónico 40% e 10% maior teor desta família de compostos. Os resultados obtidos neste estudo demonstram que a utilização do novo sistema hidropónico na produção de plantas de médio porte, aumenta o crescimento, a acumulação de biomassa e a qualidade do fruto. Assim, permite ao produtor reduzir os custos, rentabilizar a produção (menor tempo de produção), valorizar a biomassa da planta e aumentar a qualidade do produto. Também, através dos teores de carotenóides, ácidos gordos e polifenóis poderá inferir-se que ocorreu um menor impato dos stressses abióticos subjacentes aos sistemas nas plantas produzidas pelo sistema DLFT.

Berberine is an alkaloid used as a fluorochrome in the identification of heparin and DNA. Enerback, 1974, described the technique used until today to study granules rich in heparin of vertebrate mast cells. Santos et al., 2003, studied mast cells of the mollusk Anomalocardia brasiliana using biochemical and histological analysis. This work used the fluorescent dye berberine technique to improve characterization of these cells. Mollusk organs (ctenidium and mantle) were processed with routine histological techniques. Tissue sections were treated with berberine 0,02% in redistilled water acidified to pH 4, by the addition of citric acid for 20 minutes. The visualization was made through fluorescence microscopy with ultraviolet region emission. The mast cell fluorescence had a strong yellow color, where cell nuclei appeared more greenish. This result was very similar to the ones reported before. Mast cells are location at the epithelium surface is the same in both organs, mantle and ctenidium. The fluorescence was easily observed in the granules. Therefore, this technique showed to be good and sensitive to study mast cell of invertebrates

Flowering is a fundamental process in the life cycle for plant. This process is marked by vegetative to reproductive apical meristem conversion, due to interactions between several factors, both internal and external to plant. Therefore, eight subtractive libraries were constructed using apical meristem induced or not induced for two contrasting species: Solanum lycopersicum cv. Micro-Tom and Solanum pimpinellifolium. Several cDNAs were identified and among these, were selected two cDNAs: one homologous cDNA to cyclophilin (LeCYP1) and the other to Auxin repressed protein (ARP). It has observed that LeCYP1 and ARP genes are important in the developmental process to plants. In silico analysis, were used several databases with the exclusion criterion E-value <1.0x10-15. As a result, conservation was observed for proteins analyzed by means of multiple alignments and the presence of functional domains. Then, overexpression cassettes were constructed for the ARP cDNA in sense and antisense orientations. For this step, it was used the CaMV35S promoter. The cDNA orientation (sense or antisense) in relation to the promoter was determined by restriction enzymes and sequencing. Then, this cassette was transferred to binary vector pZP211 and these cassettes were transferred into Agrobacterium tumefaciens LBA4404. S. lycopersicum cv. Micro-Tom (MT) and MT-Rg1 plants were transformed. In addition, seedlings were subjected to hormone treatments using a synthetic auxin (- naphthalene acetic acid) and cyclosporin A (cyclophilin inhibitor) treatments and it was found that the hormone treatment there were changes in development of lateral roots pattern, probably related to decreases in auxin signaling caused by reduction of LeCYP1 in MT-dgt plants while cyclosporin A treatments, there was a slight delay in flowering in cv. MT plants. Furthermore, assay with real-time PCR (RT-qPCR) were done for expression level analysis from LeCYP1 and ARP in order to functionally characterize these sequences in tomato plants.

There are strong interests in the evaluation of the biological effects of natural and synthetic products. Blood constituents labeled with technetium-99m (99mTc) are used in nuclear medicine. The aim of this work was to study the effects of Clove (Caryophyllus aromaticas L.) and OZE (preparation used in the Health Sciences) on the labeling blood constituents with 99mTc and on the morphologic red blood cells (RBC) and the action of an extract of tomato (TO) on the labeling of blood constituents Blood samples were incubated with clove or OZE or TO, stannous chloride and 99mTc. Plasma (P), blood cells (BC), insoluble fractions (IF) of plasma and blood cells were separated. The radioactivity was counted and percentage of radioactivity (%ATI) to each blood fraction was calculated. The shape and morphometric parameter (perimeter/area ratio) were evaluated in the studies with clove and OZE. Clove extract and OZE altered significantly (p<0.05) the %ATI of blood constituents and the shape of red blood cells. However, clove extract not altered the red blood cells perimeter/area ratio. The tomato extract used at the highest concentrations reduced significantly (p<0.05) the %ATI in IF-P, although this extract did not modify the radiolabeling on BC, neither the radioactivity fixation on IFBC. The results indicate that these chemical compounds would have oxidative/chelating actions
