Using a high-resolution reverse-phase liquid chromatography method we found that the tissues of the hermatypic coral Pocillopora capitato (collected in Santiago Bay, Mexico) contain a high diversity of primary and secondary mycosporine-like amino acids (MAAs) typical of some reef-building coral species: mycosporine-glycine, shinorine, porphyra-334, mycosporine-methylamine-serine, mycosporine-methylamine-threonine, palythine-serine, palythine and one additional novel predominant MAA, with an absorbance maximum of 320 nm. Here we document the isolation and characterization of this novel MAA from the coral A capitata. Using low multi-stage mass analyses of deuterated and non deuterated compounds, high-resolution mass analyses (Time of Flight, TOF) and other techniques, this novel compound was characterized as palythine-threonine. Palythine-threonine was also present in high concentrations in the corals Pocillopora eydouxi and Stylophora pistillata indicating a wider distribution of this MAA among reef-building corals. From structural considerations we suggest that palythine-threonine is formed by decarboxylation of porphyra-334 followed by demethylation of mycosporine-methylamine-threonine. (C) 2008 Elsevier B.V. There is a need of scientific evidence of claimed nutraceutical effects, but also there is a social movement towards the use of natural products and among them algae are seen as rich resources. Within this scenario, the development of methodology for rapid and reliable assessment of markers of efficiency and security of these extracts is necessary. The rat treated with streptozotocin has been proposed as the most appropriate model of systemic oxidative stress for studying antioxidant therapies. Cystoseira is a brown alga containing fucoxanthin and other carothenes whose pressure-assisted extracts were assayed to discover a possible beneficial effect on complications related to diabetes evolution in an acute but short-term model. Urine was selected as the sample and CE-TOF-MS as the analytical technique to obtain the fingerprints in a non-target metabolomic approach. Multivariate data analysis revealed a good clustering of the groups and permitted the putative assignment of compounds statistically significant in the classification. Interestingly a group of compounds associated to lysine glycation and cleavage from proteins was found to be increased in diabetic animals receiving vehicle as compared to control animals receiving vehicle (N6, N6, N6-trimethyl-L-lysine, N-methylnicotinamide, galactosylhydroxylysine, L-carnitine, N6-acetyl-N6-hydroxylysine, fructose-lysine, pipecolic acid, urocanic acid, amino-isobutanoate, formylisoglutamine. Fructoselysine significantly decreased after the treatment changing from a 24% increase to a 19% decrease. CE-MS fingerprinting of urine has provided a group of compounds different to those detected with other techniques and therefore proves the necessity of a cross-platform analysis to obtain a broad view of biological samples.

Em fontes de nêutrons por spallation para pesquisa de materiais, o comprimento de onda dos nêutrons é geralmente determinado pelos tempos de percurso (TOF) dos nêutrons desde a fonte até o detector. A precisão atingível é limitada pelo fato de o tempo de emissão do pulso característico do sistema alvo/moderador ser diferente de zero (a situação ideal fictícia seria a emissão se ocorresse na forma de um impulso). “Moderadores acoplados” (elementos usados para produzir feixes de alta intensidade com nêutrons de baixa energia) apresentam um decaimento de intensidade em função do tempo muito longo, ao longo de todo o espectro usado nos experimentos. Por este motivo, “moderadores desacoplados”, os quais produzem feixes com intensidade mais reduzida, são freqüentemente usados para instrumentos que requerem alta resolução. Neste trabalho, propusemos e analisamos uma nova técnica de filtragem dinâmica de feixes de nêutrons polarizados de baixa energia para experimentos que utilizam TOF na determinação do comprimento de onda. O dispositivo consiste de um sistema ótico polarizador/analisador e um inversor de spin seletivo em energia, o qual funciona por ressonância espacial do spin. Variando a condição de ressonância em sincronia com a estrutura temporal do pulso de nêutrons (através do controle de campos magnéticos), o filtro pode ajustar a resolução de energia (ou de comprimento de onda) de pulsos de banda larga em tais experimentos, separando os nêutrons com a correta relação “TOF/comprimento de onda” dos demais Um método para o cálculo de desempenho do sistema foi apresentado em um Trabalho Individual (TI) (PARIZZI et al., 2002 - i), do qual se fará aqui uma breve revisão além de amplo uso na otimização dos parâmetros a serem ajustados para o projeto do filtro. Os resultados finais mostram que ganhos consideráveis em resolução podem ser obtidos com a aplicação desta técnica em experimentos de reflectometria por tempo de percurso, sem que para tal seja necessário comprometer a intensidade do feixe usado pelo mesmo instrumento quando operado em um modo de baixa resolução, dando ao usuário do instrumento a opção de escolher a relação ótima entre intensidade e resolução para seu experimento. Como parte da conclusão desta dissertação, é apresentada uma proposta de parâmetros para a construção deste tipo de filtro e previsão de desempenho da configuração proposta, baseada no software de modelamento desenvolvido.

Introdução: A solução de UW (University of Wisconsin) tem sido utilizada como padrão para preservação de enxertos hepáticos para transplante ortotópico de fígado (TOF) desde 1990. Seu custo é alto, e não existem, em nosso meio, estudos clínicos que comparem a sua efetividade com a de outras soluções. A solução de HTK (histidina-triptofano-cetoglutarato) foi desenvolvida inicialmente para cardioplegia, porém estudos experimentais e alguns estudos clínicos retrospectivos demonstraram sua eficácia na preservação hepática, mesmo com tempos de isquemia prolongados. Objetivos: Comparar a efetividade das soluções de preservação de órgãos HTK e UW com relação as variáveis: disfunção primária do enxerto (DPE), tempo de isquemia fria (TIF), complicações de via biliar (CVB), alterações de provas funcionais hepáticas (PFH) e sobrevida do enxerto e dos pacientes. Método: Foram estudados os fígados de doadores de múltiplos órgãos, implantados nos receptores pela técnica de piggyback, segundo ordem cronológica de ingresso em lista de espera no RS no período janeiro de 2003 a agosto de 2004. As soluções de preservação HTK e UW foram utilizadas de forma randomizada em blocos. A perfusão na aorta foi feita com 4 litros de HTK ou 2 litros de UW e a perfusão venosa portal com 1 litro de ambas as soluções, utilizando-se 500 ml para perfusão venosa e arterial adicional ex-situ e armazenagem do enxerto. Realizou-se biópsia hepática em cunha do lobo esquerdo quando a esteatose macroscópica estava presente. A análise bioquímica sérica foi diária na primeira semana e, em 15 e 30 dias pós-operatórios. Resultados: Foram estudados 102 pacientes submetidos ao TOF, sendo 65 no grupo UW (63,7%) e 37 no grupo HTK (36,3%). As frequências de sexo, raça, estado hemodinâmico, o uso de vasopressores e a presença de esteatose nos doadores foram igual nos dois grupos (pα>,05). A idade média dos doadores foi de 38,1 anos (DP +-14,4) no grupo UW e de 44,6 anos (DP +-14,2) no HTK (pα=,036). A distribuição de sexo, raça, idade, etiologia da cirrose, re-transplante, hepatite fulminante, trombose portal e escore de Child-Pugh dos receptores foi igual nos dois grupos (pα>,05). O grupo HTK teve 8 casos (25,8%) de CVB (4 estenose, 2 fístulas e 2 lesões do tipo isquêmica) contra 5 casos (8,6%) do grupo UW (pα=,033) em 89 pacientes que completaram 4 meses de seguimento (OR=2,0; IC 95%=1,2 a 3,5). A média do TIF nos dois grupos foi semelhante (UW= 579,2 min.; HTK= 527,9 min. pα>,05) e não houve diferenças nas incidências de CVB, DPE e óbito com relação a TIF estratificados entre os grupos. Não houve variação nas medianas das PFH (pα>,05 para BT, AST, ALT, FA, GGT, LDH, Fator 5 e TP). A incidência de óbito foi similar em ambos os grupos: UW= 6 (9,4%) e HTK= 4 (11,1%). A incidências de DPE foi de 2,8% no grupo HTK (1 caso) e 9,4% no grupo UW (6 casos) (pα=0,15), dos quais 5 (71,4%) evoluíram para o óbito por não funcionamento primário do enxerto, com ou sem outras morbidades. Conclusão: As soluções de UW e HTK foram igualmente efetivas na preservação dos enxertos hepáticos de doadores cadavéricos na amostra analisada, considerando-se aspectos clínicos, laboratoriais e sobrevida dos pacientes e dos enxertos. A utilização rotineira da solução de HTK poderá diminuir os custos do TOF. A média de idade maior dos doadores e a utilização de um volume reduzido de solução podem ter contribuído para uma incidência maior de CVB no grupo HTK.

The complex of Brookhart Ni(α-diimine)Cl2 (1) (α-diimine = 1,4-bis(2,6- diisopropylphenyl)-acenaphthenediimine) has been characterized after impregnation on silica (S1) and MAO-modified silicas (4.0, 8.0 and 23.0 wts.% Al/SiO2 called S2, S3 and S4, respectively). The treatment of these heterogeneous systems with MAO produces some active catalysts for the polymerization of the ethylene. A high catalytic activity has been gotten while using the system supported 1/S3 (196 kg of PE/mol[Ni].h.atm; toluene, Al/Ni = 1000, 30ºC, 60 min and atmospheric pressure of ethylene). The effects of polymerization conditions have been tested with the catalyst supported in S2 and the best catalytic activity has been gotten with solvent hexane, MAO as cocatalyst, molar ratio Al/Ni of 1000 and to the temperature of 30°C (285 kg of PE/mol[Ni].h.atm). When the reaction has been driven according to the in situ methodology, the activity practically doubled and polymers showed some similar properties. Polymers products by the supported catalysts showed the absence of melting fusion, results similar to those gotten with the homogeneous systems by DSC analysis. But then, polymers gotten with the transplanted system present according to the GPC’s curves the polydispersity (MwD) varies between 1.7 and 7.0. A polyethylene blend (BPE/LPE) was prepared using the complex Ni(α-diimine)Cl2 (1) (α-diimine = 1,4-bis(2,6-diisopropylphenyl)-acenaphthenediimine) and {TpMs*}TiCl3 (2) (TpMs* = hydridobis(3-mesitylpyrazol-1-yl)(5-mesitylpyrazol-1-yl)) supported in situ on MAO-modified silica (4.0 wts. -% Al/SiO2, S2). Reactions of polymerization of ethylene have been executed in the toluene in two different temperatures (0 and 30°C), varying the molars fraction of nickel (xNi), and using MAO as external cocatalyst. To all temperatures, the activities show a linear variation tendency with xNi and indicate the absence of the effect synergic between the species of nickel and the titanium. The maximum of activity have been found at 0°C. The melting temperature for the blends of polyethylene produced at 0 °C decrease whereas xNi increases indicating a good compatibility between phases of the polyethylene gotten with the two catalysts. The melting temperature for the blends of polyethylene showed be depend on the order according to which catalysts have been supported on the MAO-modified silica. The initial immobilization of 1 on the support (2/1/S2) product of polymers with a melting temperature (Tm) lower to the one of the polymer gotten when the titanium has been supported inicially (1/2/S2). The observation of polyethylenes gotten with the two systems (2/1/S2 and 1/2/S2) by scanning electron microscopy (SEM) showed the spherical polymer formation showing that the spherical morphology of the support to been reproduced. Are described the synthesis, the characterization and the catalytic properties for the oligomerization of the ethylene of four organometallics compounds of CrIII with ligands ([bis[2-(3,5-dimethyl-1-pyrazolyl)ethyl]amine] chromium (III) chloride (3a), [bis[2-(3,5- dimethyl-l-pyrazolyl)ethyl]benzylamine] chromium (III) chloride (3b), [bis[2-(3,5-dimethyl-lpyrazolyl) ethyl]ether] chromiun(III)chloride (3c), [bis[2-(3-phenyl-lpyrazolyl) ethyl]ether]chromiun(III)chloride (3d)). In relation of the oligomerization, at exception made of the compounds 3a, all complex of the chromium showed be active after activation with MAO and the TOF gotten have one effect differentiated to those formed with CrCl3(thf)3. The coordination of a tridentate ligand on the metallic center doesn't provoke any considerable changes on the formation of the C4 and C6, but the amount of C8 are decrease and the C10 and C12+ have increased. The Polymers produced by the catalyst 3a to 3 and 20 bar of ethylene have, according to analyses by DSC, the temperatures of fusion of 133,8 and 136ºC respectively. It indicates that in the two cases the production of high density polyethylene. The molar mass, gotten by GPC, is 46647 g/mols with MwD = 2,4 (3 bar). The system 3c/MAO showed values of TOF, activity and selectivity to different α-olefins according to the pressure of ethylene uses. Himself that shown a big sensibility to the concentration of ethylene solubilized.

The genus Saccharum belongs to Poaceae family. Sugarcane has become important monocultures in Brazil due to their products: ethanol and sugar. The production may change between different regions from Brazil. This difference is related to soil, climatic conditions and temperature that promotes oxidative stress that may induce an early flowering. The aim of this work was to identify the effects of oxidative stress. In order to analyse this, sugarcane plants were submitted to oxidative stress using hydrogen peroxide. After this treatment, the oxidative stress were analyzed Then, the plant responses were analyzed under different approaches, using morphophysiological, biochemical and molecular tools. Thus, sugarcane plants were grown under controlled conditions and until two months they were subjected first to a hydroponics condition for 24 hours in order to acclimation. After this period, these plants were submitted to oxidative stresse using 0 mM, 10 mM, 20 mM and 30 mM hydrogen peroxide during 8 hours. The histomorphometric analysis allowed us to verify that both root and leaf tissues had a structural changes as it was observed by the increased in cell volume, lignin accumulation in cell walls. Besides, this observation suggested that there was a change in redox balance. Also, it was analyzed the activity of the SOD, CAT and APX enzymes. It was observed an increase in the SOD activity in roots and it was also observed a lipid peroxidation in leaves and roots. Then, in order to identify proteins that were differently expressed in this conditions it was used the proteomic tool either by bidimensional gel or by direct sequencing using the Q-TOF EZI. The results obtained with this approach identified more than 3.000 proteins with the score ranging from 100-5000 ions. Some of the proteins identified were: light Harvesting; oxygenevolving; Thioredoxin; Ftsh-like protein Pftf precusor; Luminal-binding protein; 2 cys peroxiredoxin e Lipoxygenase. All these proteins are involved in oxidative stress response, photsynthetic pathways, and some were classified hypothetical proteins and/or unknown (30% of total). Thus, our data allows us to propose that this treatment induced an oxidative stress and the plant in response changed its physiological process, it made changes in tissue, changed the redox response in order to survival to this new condition

Biodiesel production has increased over the last decade because of the benefits associated with this fuel, including renewability, domestic feedstock, lower toxicity, and biodegradability. From 2008, the use of beef tallow as a feedstock for biodiesel production in Brazil has increased in significance, representing the second largest source of biodiesel, after soybeans. However, the performance of biodiesel in cold weather conditions is worse than diesel because of deposition of insoluble at low temperatures, accelerating the plugging of fuel filters and injectors of the vehicle engine. Studies have been conducted on beef tallow biodiesel, mostly related to the properties of thermal and oxidative stability. However, few studies have described the nature of the precipitate formed and its influence on product quality. Research suggests that the cause of deposition is related to the nature of saturated esters and monoacylglycerols as inducing agents. This study monitored the levels of mono-, diand triacylglycerols, the oxidation stability and the cold filter plugging point (CFPP) in beef tallow biodiesel samples from two commercial producers in Brazil for a period of twelve months. Filtered precipitates were analyzed by comparative techniques of GCFID, HPLC-UV/VIS, HPLC-MS-IT-TOF and TG to verify the nature, using monopalmitin and monostearin as reference standards. The formation of precipitate reduced the levels of monoacylglycerols in the beef tallow biodiesel. GC-FID and LCMS- IT-TOF results confirmed the nature of the deposit as saturated monoacylglycerols, predominantly monostearin and monopalmitin as the second major component. Moreover the TG analysis of the residue indicated similar thermal decomposition of the reference standards. The precipitate did not affect the oxidation stability of beef tallow biodiesel and the CFPP characteristic of blends up B60. However, the presence of iron reduced significantly the oxidation stability of biodiesel

Four antimicrobial peptides were purified from Royal Jelly of honeybees, by using reverse phase-HPLC and sequenced by using Q-Tof-MS/MS: PFKLSLHL-NH2 (Jelleine-I), TPFKLSLHL-NH2 (Jelleine-II), EPFKLSLHL-NH2 (Jelleine-III), and TPFKLSLH-NH2 (Jelleine-IV). The peptides were synthesized on-solid phase, purified and submitted to different biological assays: antimicrobial activity, mast cell degranulating activity and hemolysis. The Jelleines-I-III presented exclusively antimicrobial activities against yeast, Gram+ and Gram- bacteria; meanwhile, Jelleine-IV was not active in none of the assays performed. These peptides do not present any similarity with the other antimicrobial peptides from the honeybees; they are produced constitutively by the workers and secreted into Royal Jelly.
